Sex change in the coral-dwelling goby Gobiodon histrio was induced by placing two adult fish of the same sex on a coral colony. The sex change of individual fish was confirmed using histology, and whole-body concentrations of the gonadal steroids testosterone (T), 11-ketotestosterone (11-KT), and 17β-oestradiol (E2) were examined. The results show that T, 11-KT and E2 occurred in both female and male G. histrio . E2 concentration in females was twice that in males, while concentrations of T did not differ between the sexes. Contrary to predictions, concentrations of T and E2 did not differ between fish that changed sex and those that did not. Most samples had 11-KT concentrations below minimum levels of detection (  i.e. <0·15 ng ml−1) and were therefore not analysed statistically. The results suggest that: (i) specific activation or de-activation of the T–E2 (aromatase) pathway is a probable candidate for mediating serial adult sex change in G. histrio , and (ii) low levels of 11-KT may be important in allowing serial adult sex change in G. histrio .  相似文献

Levels of serum sex steroids (estradiol-17beta, E2; testosterone, T; 11-ketotestosterone, 11-KT) in male, female and natural sex-reversing red-spotted grouper (Epinephelus akaara), and aromatase activity of gonad and brain in both male and female were investigated throughout an annually reproductive cycle. In females, serum E2 and T peaked during vitellogenesis, but in males and natural sex-reversing fish, 11-KT, T and E2 reached peak during spermatogenesis. In addition, in females, serum 11-KT levels (monthly means: 0.32 +/- 0.03 ng/ml) which were very low did not significantly fluctuate during the annual reproductive cycle. In breeding season, females displayed higher E2 levels than males and sex-reversing fish, while males and sex-reversing fish showed higher 11-KT levels and, to a lesser extent, higher T levels than females. Furthermore, the changing pattern of sex steroids in males was similar to that in natural sex-reversing fish, and a second peak of serum androgens 11-KT and T appeared in December both in male and natural sex-reversing fish; significantly higher serum 11-KT levels were observed in natural sex-reversing fish than that in females from December to April. In females, but not in males, aromatase activity of brain and gonad demonstrated significantly seasonal changes (exhibiting a peak in breeding season); moreover, aromatase activity in females was higher than that in males. Furthermore, significantly lower aromatase activity in testis was observed in breeding season, in contrast to that in ovary. Taken together, the present findings indicated that changes of serum sex steroids levels and aromatase activity in red-spotted grouper were closely associated with sex inversion. In addition, the present results also suggested that sex inversion in red-spotted grouper peaked mainly from December to March.  相似文献

Sexually mature male and female spiny damselfish, Acanthochromis polyacanthus (Pomacentridae) were introduced to resident fish of both sexes in paired encounter trials and the effect on activity, agonistic behaviour and plasma levels of testosterone (T) and 11-ketotestosterone (11-KT) in males, and T and 17β-estradiol (E2) in females, and cortisol in both sexes was determined. Introduced males showed suppressed levels of T in the presence of both resident males and females, whereas resident males showed elevated T in response to introduced males but not females. Plasma 11-KT levels were unchanged in either resident or introduced males. Resident males showed increased activity in the presence of males but not females. Resident females showed an increase in activity when paired with introduced females and greater aggressive behaviour towards females than males. There were no differences in plasma hormone levels in either resident or introduced female fish. Plasma cortisol levels were low in all fish and stress effects did not appear to account for differences in T levels in males. The results of the present study show that elevations in plasma T levels in resident male spiny damselfish are associated only with male presence but that suppression of T in introduced fish occurs irrespective of the sex of the resident. In contrast resident females, which show stronger evidence of aggressive behaviour do not show changes in plasma androgen levels. This suggests that aggressive behaviour in male and female fishes may be mediated by different endocrine pathways.  相似文献

Spermatogenesis in male Atlantic halibut (Hippoglossus hippoglossus L.) was investigated by sampling blood plasma and testicular tissue from 15-39-month-old fish. The experiment covered a period in which all fish reached puberty and completed sexual maturation at least once. The germinal compartment in Atlantic halibut testis appears to be organized in branching lobules of the unrestricted spermatogonial type, because spermatocysts with spermatogonia were found throughout the testis. Spermatogenesis was characterized histologically, and staged according to the most advanced type of germ cell present: spermatogonia (Stage I), spermatogonia and spermatocytes (Stage II), spermatogonia, spermatocytes and spermatids (Stage III), spermatogonia, spermatocytes, spermatids and spermatozoa (Stage IV), and regressing testis (Stage V). Three phases could be distinguished: first, an initial phase with low levels of circulating testosterone (T; quantified by RIA) and 11-ketotestosterone (11-KT; quantified by ELISA), spermatogonial proliferation, and subsequently the initiation of meiosis marked by the formation of spermatocytes (Stage I and II). Secondly, a phase with increasing T and 11-KT levels and with haploid germ cells including spermatozoa present in the testis (Stage III and IV). Thirdly, a phase with low T and 11-KT levels and a regressing testis with Sertoli cells displaying signs of phagocytotic activity (Stage V). Circulating levels of 11-KT were at least four-fold higher than those of T during all stages of spermatogenesis. Increasing plasma levels of T and 11-KT were associated with increasing testicular mass throughout the reproductive cycle. The absolute level of, or the relation between, testis growth and circulating androgens were not significantly different in first time spawners compared to fish that underwent their second spawning season. These results provide reference levels for Atlantic halibut spermatogenesis.  相似文献

Labile maturation and sex determination should be advantageous where the probability of finding a mating partner is unpredictable. Here we tested the hypothesis that the presence of a potential mating partner induces maturation and sex determination in a coral-dwelling fish, Gobiodon erythrospilus. In natural populations at Lizard Island (Great Barrier Reef), single individuals were less likely to be mature than paired individuals and they matured at a larger size, indicating plasticity in the timing of maturation. By manipulating group structure we demonstrated that both the timing of maturation and the sex of maturing individuals are socially controlled. Single juveniles did not mature, but maturation was rapidly induced by the presence of an adult partner. In addition, sex determination was found to be labile, with juveniles maturing into the opposite sex of the partner encountered. To our knowledge, this is the first experimental demonstration of social induction of maturation in conjunction with labile sex determination at maturation in vertebrates. This flexibility enables individuals to maximize their reproductive success in an environment where the timing of mate acquisition and the sex of their future partner are unpredictable.  相似文献

The Challenge Hypothesis postulates that androgen levels are a function of the social environment in which the individual is living. Thus, it is predicted that in polygynous males that engage in social interactions, androgen levels should be higher than in monogamous animals that engage in parental care. In this study, we tested this hypothesis at the intra-specific level using a teleost species, Sarotherodon galilaeus, which exhibits a wide variation in its mating system. Experimental groups of individually marked fish were formed in large ponds with different operational sex-ratios (OSR) to study the effects of partner availability on blood plasma levels of sex steroids [11-ketotestosterone (11-KT), testosterone (T), and 17,20beta-dihydroxy-4-pregnen-3-one (17,20beta-P)] and gonadosomatic index (GSI). Polygyny mostly occurred in the female biased OSR groups. 17,20beta-P and gonadosomatic index did not differ among OSR groups. However, 11-KT was high in male biased OSR and positively correlated with aggressive challenges, thereby supporting the central postulate of the Challenge Hypothesis. The results of T were the inverse of those of 11-KT, probably because 11-KT is metabolized from T. 11-KT levels of polygynous males did not differ neither from those of monogamous males, nor from those of males that participated in parental care. These results do not support the expected relationships between polygyny, parental care, and androgen levels. The differences from expectations for 11-KT may be related to the fact that in S. galilaeus, the mating and the parenting phase are not clearly separated and thus, males may still fight and court while they are brooding.  相似文献

Under constant short photoperiod, the spawning time of 2-year-old sea bass Dicentrarchus labrax was advanced as compared to controls, whereas spawnings were delayed under constant long photoperiod. High plasma levels of 17β-oestradiol (E2/) and testosterone (T) in females were coincident with the appearance of vitellogenic oocytes in the ovary, while high levels of 11-ketotestosterone (11-KT) and T in males were coincident with the presence of spermiating males. Although plasma levels of E2 in females and 11-KT in males were low during the remainder of the cycle, levels of T were always >1 ng ml−1 in both sexes, suggesting that T could play an important role during the initial stages of gonadal development. The profiles of E2 and T in females and 11-KT and T in males exposed to constant short days were similar to those in the control group, but fish which were maintained under constant long photoperiods showed a bimodal pattern of these steroids. The results obtained from fish exposed to constant photoperiod regimes provide further evidence that an endogenous process could be operating to control the reproduction of sea bass.  相似文献

Immature 1-year-old Atlantic salmon Salmo salar parr were implanted with Silastic capsules of different sizes filled with testosterone (T). Testosterone had both positive and negative effects on testicular weights, spermatogenesis and steroidogenesis. The positive effects: higher incidence of males with enlarged gonads, spermiation, and high plasma levels of 11-ketotestosterone (11-KT) and 17,20β-dihydroxy-4-pregnen-3-one (17,20β-P), were most pronounced in males treated with small T capsules. The negative effects: suppression of gonadal development and depressed plasma levels of 11-KT and 17,20β-P compared with mature controls, were most evident in fish treated with large T capsules.  相似文献

11-ketotestosterone (11-KT), a potent male-specific androgen in fish, has important roles on spermatogenesis, male behavior, and nuptial coloration. The site of 11-KT synthesis and its role on male germ cell development during protogynous sex change is not clearly understood. We examined the dynamics of steroidogenic enzymes immunolocalization, viz cholesterol side-chain cleavage (P450scc), biomarker of steroids and cytochrome P45011beta-hydroxylase (P45011beta), downstream to 11-KT production, throughout the process of sex change in honeycomb grouper (Epinephelus merra). In female, P450scc immunoreactivity (-ir) was observed in the theca layer and tunica near blood vessels (BV). During the onset of sex change, P450scc reactive cells were observed in the remaining follicle layer of degenerated oocyte of the ovo-testis in early transitional (ET) and late transitional (LT). In male, P450scc-ir was localized in the interstitial Legdig cells of testis. P45011beta reactive cells were observed in the tunica near BV in female but not in theca layer. In ET and LT phases gonads, P45011beta localized in remaining follicle layer of degenerated oocyte and tunica near BV. On the other hand, in male, both interstices and tunica near BV showed strong signals against P45011beta. Moreover, in vivo and in vitro levels of 11-KT related with the changes in the nuclei diameter of P45011beta-positive cells in both tunica near BV and remaining follicle layer of degenerated oocyte to interstices during the progress of sex change. The present results suggest that 11-KT produced in the tunica near BV may provide the stimulus for female to degenerate oocytes and initiate sex change. However, 11-KT produced both in tunica near BV and remaining follicle layer of degenerated oocyte possibly plays critical role during testicular differentiation as well as gonadal restructuring at mid to late phases (ET to LT) of sex change in honeycomb grouper.  相似文献

Male and female Carassius auratus were exposed to safe (SC; 0.01 ppm) and sublethal (SL; 0·1 ppm) concentrations of an organochlorine pesticide γ-hexachlorocyclohexane (γ-HCH) for 4 weeks during the pre-spawning phase (June) of the annual reproductive cycle. Gonadosomatic index and gonadotropin levels were significantly lower after exposure to both γ-HCH concentrations than in control fish. After 4 weeks exposure, gonadal tissue from control, SC and SL exposed fish was incubated with carp hypophyseal homogenate (chh). The chh stimulated production of testosterone, 17,20β-dihydroxy-4-pregnen-3-one (17,20βP), 11-deoxycortisol, and 11-ketotesterone (11-KT) was estimated in the unconjugated (free) and conjugated (glucuronide) fractions by radioimmunoassay. In both sexes, testosterone production was greatly decreased in γ-HCH exposed fish compared to controls. 17,20βP production was low in all fish and was unaffected by γ-HCH. Free 11-deoxycortisol production by testicular fragments was higher in SL and SC compared with controls, while conjugated 11-deoxycortisol was increased only higher in the SL. In ovarian fragments from exposed fish, free 11-deoxycortisol decreased while glucuronide concentrations increased compared with controls. 11-KT production was significantly decreased in testicular fragments of exposed fish. The results indicate that γ-HCH inhibits gonadal recrudescence by decreasing both gonadotropin secretion and its potential for stimulation of steroidogenesis.  相似文献

The metabolic peptide hormone nesfatin-1 has been linked to the reproductive axis in fishes. The purpose of this study was to determine how energy availability after spawning affects plasma levels of nesfatin-1, the metabolic peptide hormone ghrelin, and sex steroid hormones in rematuring female rainbow trout (Oncorhynchus mykiss). To limit reproductive maturation, a group of female trout was food-restricted after spawning and compared with a control group that was fed a standard broodstock ration. The experiment was conducted twice, once using two-year-old trout (second-time spawners) and once using three-year-old trout (third-time spawners). During monthly sampling, blood was collected from all fish, and a subset of fish from each treatment was sacrificed for pituitaries. Pituitary follicle-stimulating hormone-beta (fsh-β) mRNA expression was analyzed with q-RT-PCR; plasma hormone levels were quantified by radioimmunoassay (17β-estradiol and ghrelin) and enzyme-linked immunosorbent assay (11-keto-testosterone and nesfatin-1). Although plasma nesfatin-1 levels increased significantly in the months immediately after spawning within both feeding treatments, plasma nesfatin-1 did not differ significantly between the two treatments at any point. Similarly, plasma ghrelin levels did not differ significantly between the two treatments at any point. Food restriction arrested ovarian development by 15–20 weeks after spawning, shown by significantly lower plasma E2 levels among restricted-ration fish. Pituitary fsh-β mRNA levels were higher among control-ration fish than restricted-ration fish starting at 20 weeks, but did not differ significantly between treatment groups until 30 weeks after spawning. Within both treatment groups, plasma 11-KT was elevated immediately after spawning and rapidly decreased to and persisted at low levels; starting between 20 and 25 weeks after spawning, plasma 11-KT was higher among control-ration fish than restricted-ration fish. The results from these experiments do not provide support for plasma nesfatin-1 as a signal for the initiation of reproductive development in rematuring female rainbow trout.  相似文献

It is widely accepted that the hypothalamic-pituitary-gonadal axis is involved in gonadal sex change in socially controlled sex-changing fish. However, the specific secretion profiles of pituitary gonadotropins (GtHs) in this type of fish are not known. To address this fundamental question, we demonstrated that the diurnal secretion patterns of GtHs differ distinctly between males and females in a socially controlled sex-changing fish. We analyzed the pituitary mRNA levels of glycoprotein hormone subunits (i.e., the common alpha-subunit and specific beta-subunits follicle-stimulating hormone beta, luteinizing hormone beta, and thyroid-stimulating hormone beta) in the wrasse Pseudolabrus sieboldi, which is a model fish that exhibits accurate diurnal rhythms of gametogenesis in both males and females. Northern blots clearly showed that each subunit gene exhibits a diurnal rhythm of expression in the pituitary and that the expression patterns differ distinctly between the sexes. Our results suggest that oogenesis and spermatogenesis in this hermaphroditic fish are regulated differentially through the distinct secretion patterns of pituitary glycoprotein hormones. This study also provides direct evidence of the sexual plasticity of pituitary GtH secretion in a socially controlled sex-changing fish.  相似文献

Experiments were conducted to identify factors involved in sex change in the protogynous black sea bass Centropristis striata . Black sea bass maintained in the ratio of 8 females (F):0 males (M) for 9 months reversed sex while those kept at the ratios of 6F:2M or 4F:4M did not. Female black sea bass implanted with 1·0 mg 11-ketotestosterone (11-KT) or 10 mg fadrozole (FAD) changed sex and began spermiating while those implanted with 0·1 mg 11-KT or 1·0 mg FAD underwent incomplete sex reversal. One fish implanted with 1·0 mg FAD initiated sex change but was not spermiating at the end of the study. One fish in the control group, the largest fish in the study, initiated sex change. These results suggest that the presence of males may restrict sex reversal in black sea bass and that high 17β-oestradiol:11-KT is required for maintaining ovarian function.  相似文献

Steroids both in seminal fluid (SF) and blood serum (BS) as well as the milt quality (sperm motility and sperm production) were investigated during final maturation of Persian sturgeon. The BS levels of testosterone (T), 11-Ketotestosterone (11-KT), progesterone (P), 17α,20β,21-trihydroxy-4-pregnen-3-one (20βS), cortisol (C) and 17α,hydroxyprogesterone (OHP) elevated after pituitary preparation (PP) treatment and then decreased during stripping period for spermiating males. Such elevations did not occur for non-spermiating individuals and steroids remained in basal levels after PP treatment until the end of stripping period. For both groups (spermiating and non-spermiating fish), the BS levels of 17α,20β-dihydroxy-4-pregnen-3-one (DHP) did not show significant changes during experiment. During stripping period, the values of all tested steroids were significantly lower in SF than in BS of spermiating males. SF levels of 20βS and 11-KT showed a decreasing trend and the other steroids were unchanged during this period. Significant positive correlations were found between the values of 20βs and 11-KT in BS with their levels in SF. Also, BS and SF levels of 20βs and 11-KT were positively correlated with sperm motility characteristics (percentage and duration of motility) and sperm production (sperm density and milt volume), respectively. The results showed the probable involvement of 20βs, P, OHP, T, 11-KT and C in final maturation of Persian sturgeon, especially 20βs and 11-KT had good correlations with qualitative parameters of milt. The lower levels of steroids in SF than those in BS might also be essential for viability of Persian sturgeon spermatozoa. Probably, there are mechanisms that stabilize the concentrations of a number of hormones in the SF.  相似文献

The effects of administration of gonadotropin-releasing hormone agonist (GnRHa) on proliferation and apoptosis of male germ cells were evaluated on Atlantic bluefin tuna (Thunnus thynnus L.) reared in captivity. Fish (n = 19) were treated with a sustained-release delivery system loaded with GnRHa during the natural spawning season of 2004 and 2005 (June–July). Untreated Control fish (n = 17) and adult wild spawners were used for comparison. Fish were sacrificed 2–8 d after GnRHa implantation and body weight and gonad weight were recorded, and gonads and blood were taken. Germ cell proliferation and apoptosis were evaluated through the immunohistochemical detection of proliferating cell nuclear antigen (PCNA) and the terminal deoxynucleotidyl transferase-mediated d’UTP nick end labelling (TUNEL) method, respectively. Plasma 11 ketotestosterone (11-KT) levels were measured using an ELISA method. Mean gonado-somatic index and seminiferous lobule diameter did not differ between GnRHa-treated and Control fish, and were significantly lower in captive-reared individuals than in wild spawners. Significant increases in 11-KT plasma levels and spermatogonial mitosis, along with a reduction of germ cell apoptosis were demonstrated in GnRHa-treated fish compared to Controls. The results suggest that GnRHa administration was effective in enhancing germ cell proliferation and reducing apoptosis in captive males through the stimulation of luteinizing hormone (LH) release and testicular 11-KT production.  相似文献

The weakly electric fish from the main channel of the Amazon river, Sternarchogiton nattereri, offers a striking case of morphological variation. Females and most males are toothless, or present only few minute teeth on the mandible, whereas some males exhibit exaggerated, spike-like teeth that project externally from the snout and chin. Androgens are known to influence the expression of sexually dimorphic traits, and might be involved in tooth emergence. In this study we assess the relationship in S. nattereri between morphological variation, 11 ketotestosterone (11-KT) and testosterone (T). We also examine relationships of morphology and androgen levels with electric organ discharge (EOD) frequency, reproductive condition, and seasonality. Our main finding is that male morph categories differed significantly in plasma concentrations of 11-KT, with toothed males showing higher levels of 11-KT than toothless males. By contrast, we did not detect statistical differences in T levels among male morph categories. Reproductive condition, as measured by gonadosomatic indexes (GSI), differed across two sample years, increased as the season progressed, and was higher in toothed males than in non-toothed males. EOD frequency was higher in toothed males than in either toothless males or females. Taken together, our findings suggest that S. nattereri male sexual characters are regulated by 11-KT levels, and that both morphology and androgens interact with reproductive condition and EOD frequency in ways that vary within and across reproductive seasons.  相似文献

Androgens are known to be involved in reproductive behaviours including courtship and aggression. According to the Challenge Hypothesis, androgen activity upregulates male reproductive behaviour seasonally and also modulates short term adaptation of these behaviours in response to social context. In the weakly electric fish, Apteronotus leptorhynchus, 11-ketotestosterone (11-KT) has been previously implicated in the regulation of electrocommunication behaviours that are believed to have roles in both aggression and courtship. Changes in male 11-KT levels were quantified using a non-invasive measurement technique alongside changes in electrocommunication behaviour following environmental cues that simulated the onset of the breeding season. Males showed an increase in mean electric organ discharge frequency (EODf), which is consistent with earlier results showing a female preference for high EODf. A subset of males with high initial EODfs showed increases in both 11-KT and EODf, which provides support for an EODf-based dominance hierarchy in this species. Males housed in social conditions and exposed to breeding conditioning also showed higher overall electric organ discharge frequencies and 11-KT compared to males housed in isolation. Evidence is presented that another type of electrocommunication signal previously implicated in courtship may also serve as an inter-male signal of submission. Our results are consistent with earlier observations that electrocommunication signals produced during inter-male aggression serve in deterring attacks, and their pattern of production further suggested the formation of a dominance hierarchy.  相似文献

When 4 mg of testosterone (T) per kg food was given to 1-year-old protandrous male black porgy Acanthopagrus schlegeli for 7 months, gonadosomatic index was significantly higher than when the dose was 0.5 mg kg−1 food. Both doses of T prolonged the spawning season, and increased the number of spermiating fish and milt volume. Sperm concentrations were similar in spermiating black porgy from the treated and control groups. Low levels of oestradiol-17β were observed during the experimental period while elevated levels of plasma T were observed only in March in both control and T-treated groups. Significantly higher levels of plasma 11-ketotestosterone (11-KT) were observed in the 0.5- and 4.0-mg T-treated groups during the spawning season as compared to the control group. The present data suggest that both 0.5- and 4·0-mg T doses stimulate testicular weight, increase numbers of spermiating males and milt volume without affecting the sperm concentrations. Plasma 11-KT concentrations were elevated during T treatment and closely correlated with testicular development and spermiation.  相似文献

Sex steroid hormones are important for reproduction in all vertebrates, but few studies examine inter-individual, temporal, and population-level variations, as well as environmental influences on circulating steroid levels within the same species. In this study we analyzed plasma 11-ketotoestosterone (11-KT) and 17β-estradiol (E2) levels in the oyster toadfish to test for 1) individual and temporal variations by serially sampling the same individuals during the reproductive and post-reproductive period, 2) variations in steroid levels among toadfish obtained from different sources or maintained under different holding conditions, and 3) correlations with environmental parameters. Results from serial sampling showed marked inter-individual variations in male 11-KT levels in two separate groups of toadfish, but no temporal differences from June to September. Females also showed inter-individual variations in E2 concentrations, but most had elevated levels late in the reproductive season coincident with oocyte growth prior to winter quiescence. E2 concentration, but not 11-KT, was positively correlated with water temperature, and negatively correlated with daylength and lunar phase. Maricultured toadfish held under constant conditions had elevated levels of E2 and 11-KT that should be considered when using these fish for experimentation. This study provides important comparative information on the relationship between individual variations in steroid levels, and how they relate to physiological and environmental correlates in a model marine teleost.  相似文献
