Summary Aliphatic alcohols are found to stimulate the transmembrane fluxes of a hydrophobic cation (tetraphenylarsonium, TPA) and anion (AN-12) 5–20 times in red blood cells. The results are analyzed using the Born-Parsegian equation (Parsegian, A., 1969,Nature (London) 221:844–846), together with the Clausius-Mossotti equation to calculate membrane dielectric energy barriers. Using established literature values of membrane thickness, native membrane dielectric constant, TPA ionic radius, and alcohol properties (partition coefficient, molar volume, dielectric constant), the TPA permeability data is predicted remarkably well by theory. If the radius of AN-12 is taken as 1.9 Å, its permeability in the presence of butanol is also described by our analysis. Further, the theory quantitatively accounts for the data of Gutknecht and Tosteson (Gutknecht, J., Tosteson, D.C., 1970,J. Gen. Physiol. 55:359–374) covering alcohol-induced conductivity changes of 3 orders of magnitude in artificial bilayers. Other explanations including perturbations of membrane fluidity, surface charge, membrane thickness, and dipole potential are discussed. However, the large magnitude of the stimulation, the more pronounced effect on smaller ions, and the acceleration of both anions and cations suggest membrane dielectric constant change as the primary basis of alcohol effects.  相似文献

An approximation method using a sine function is used to solve the second degree growth equation for the case in which an organism may simultaneously become dispersed throughout a uniform region. The resulting expression for a special case is compared with the expression obtained by R. Barakat (1959,Bull. Math. Biophysics,21, 141–51), giving the first two terms, by an iterative, procedure. Estimation for an island model where mutation maintains ak-allele neutral polymorphism at a single locus on each island is considered. The likelihood of an observed sample type configuration is obtained by applying a computational algorithm analogous to Griffiths and Tavaré (Theor. Popul. Biol.46(1994), 131–159). This allows the computation of sampling distributions in an island model and investigation of their properties. Given a sample type configuration, the maximum likelihood estimate of the migration parameter is obtained by simulating independently the likelihood at a grid of points and, also, using a surface simulation method. The latter method generates the whole likelihood trajectory in a single application of the simulation program. An estimate of variance of the estimate of the migration parameter is obtained using the likelihood trajectory. A comparison of the maximum likelihood estimates of the gene flow between subpopulations is made with those obtained by using Wright'sFSTstatistic.  相似文献

【目的】副溶血性弧菌是一种重要的人畜共患病原菌,脂蛋白定位系统(Localization of lipoprotein system,Lol)负责该菌脂蛋白的转运与定位,与其致病力及耐药性密切相关,对Lol系统转运蛋白进行系统的生物信息学分析,有助于推动副溶血性弧菌致病与耐药机理的进一步研究。【方法】本文通过生物信息学分析技术,结合ExPASy在线工具、SignalP 4.0 Server、TMHMM-2.0、STRING、SWISS-MODEL等软件,分析了副溶血性弧菌Lol系统转运蛋白LolA-E及LolCD_2E的基本性质、蛋白互作关系及三级结构。【结果】LolA和LolB为酸性亲水蛋白,含信号肽位点,无跨膜区域。LolC和LolE为碱性疏水膜蛋白,LolCD_2E为中性疏水膜蛋白,LolC-E及LolCD_2E均无显著的信号肽位点。蛋白相互作用网络显示,LolA–E五个蛋白的编码基因均共表达,负责脂蛋白的合成与转运,并与BamA、Pal、MacB、CmeC等外膜蛋白具有密切的互作关系。三级结构同源建模发现,副溶血性弧菌与大肠杆菌拥有相似的LolA和LolB结构,LolC-E含有MacB蛋白的同源结构,赋予了该系统消耗ATP运输脂蛋白的重要功能。此外,本研究还首次发现了副溶血性弧菌LolC和LolE中存在一段保守的Hook结构,是LolCD_2E复合物与LolA结合并转运脂蛋白的关键区域。【结论】本研究为副溶血性弧菌Lol系统转运蛋白的表达纯化、结构与功能的研究提供了重要的数据基础,为后续抗菌药物的研发提供了新型作用靶点。  相似文献

[目的]研究在体外情况下和厚朴酚对白色念珠菌的抑制作用及其可能机制。[方法]采用微量稀释法测定和厚朴酚对白色念珠菌的最低抑菌浓度(MIC80)和最低杀菌浓度(MFC);用透射电镜观察不同浓度和厚朴酚对白色念珠菌超微结构的影响;采用Annexin V-FITC/PI染色法分析不同浓度和厚朴酚对白色念珠菌细胞凋亡的影响;用DCFH-DA染色法测定不同浓度和厚朴酚对白色念珠菌细胞内活性氧积累的影响;用JC-1染色法分析不同浓度和厚朴酚对白色念珠菌线粒体膜电位的影响;用碘化丙啶染色、考马斯亮蓝G-250染色检测和厚朴酚对白色念珠菌细胞膜通透性的影响;通过测定加入麦角甾醇后,和厚朴酚对白色念珠菌的抑制作用的变化,检测和厚朴酚对白色念珠菌细胞膜的影响。[结果]和厚朴酚对白色念珠菌具有很强的抑制作用,MIC和MFC分别为16 μg/mL和32 μg/mL。对白色念珠菌细胞壁、细胞膜和胞浆均有明显的影响。和厚朴酚是通过增加活性氧的产生和破坏线粒体功能来诱导白念珠菌的细胞凋亡和坏死。它也影响细胞膜的通透性,这可能和细胞壁的破坏和与麦角固醇的结合有关。[结论]和厚朴酚通过产生活性氧并伴随着一系列的细胞损伤这种复杂的机制从而对白色念珠菌产生抑制作用,使和厚朴酚成为一种潜在的抗真菌药物。  相似文献

Measurements of the dimensions and membrane rotational frequency of individual erythrocytes steadily tank-treading in a Rheoscope are used to deduce the surface shear viscosity (eta m) and the shear elastic modulus (mu m) of the membrane. Previously published algorithms (Trans-Son-Tay et al., Biophys. J. 46: 65, 1984, and 51: 915, 1987) plus an assumed area-conserving membrane velocity field (Secomb and Skalak, Q. J. Mech. Appl. Math. XXXV 2: 233, 1982) are applied to calculate eta m as a function of the second invariant of the surface strain rate and mu m as a function of the second invariant of membrane strain. The results indicate density-related increases in membrane stiffness and viscosity, shear-thinning viscous behavior, and strain-stiffening elastic behavior.  相似文献

目的:克隆获得合浦珠母贝PU3基因的序列,并研究其在生物矿化中的功能。方法:使用RACE获得PU3基因的全长;利用实时荧光定量PCR的方法检测PU3基因在不同组织中的表达分布;利用实时荧光定量PCR的方法检测贝壳损伤修复过程中PU3基因的表达量的变化;通过RNAi实验,抑制PU3基因的表达,之后用扫描电子显微镜观察合浦珠母贝贝壳表面的变化。结果:合浦珠母贝PU3基因的cDNA全长为2361bp,编码618个氨基酸。氨基酸序列的功能结构域分析表明其含有4个FN3结构域。该基因在外套膜中高表达,且在外套膜边缘区的表达量高于外套膜中心区。在贝壳损伤修复的过程中,该基因的表达水平呈现上升的趋势。利用RNAi技术抑制PU3基因的表达后,贝壳的棱柱层结构发生了变化,缝隙变宽,且出现空洞。结论:PU3基因所表达的蛋白作为正调控因子参与生物矿化的过程,并主要作用于贝壳的棱柱层,抑制其表达会影响棱柱层的框架结构。  相似文献

Abstract

Candida rugosa lipase was immobilized on the activated nylon-6 membrane using 0.5% glutaraldehyde as cross-linking agent. The immobilized lipase had an improved temperature and pH stability and exhibited good reusability. The adsorption process followed pseudo-second order rate equation and the biosorption isotherms correlated well with the Langmuir isotherm model. Negative ΔG indicated the adsorption process was spontaneous and the positive ΔH indicated that the adsorption was endothermic. The standard entropy ΔS value was also positive, which suggested an increase in the freedom of the system.  相似文献

Critical to epithelial cell viability is prompt and direct recovery, following a perturbation of cellular conditions. Although a number of transporters are known to be activated by changes in cell volume, cell pH, or cell membrane potential, their importance to cellular homeostasis has been difficult to establish. Moreover, the coordination among such regulated transporters to enhance recovery has received no attention in mathematical models of cellular function. In this paper, a previously developed model of proximal tubule (Weinstein, 1992, Am. J. Physiol. 263, F784–F798), has been approximated by its linearization about a reference condition. This yields a system of differential equations and auxiliary linear equations, which estimate cell volume and composition and transcellular fluxes in response to changes in bath conditions or membrane transport coefficients. Using the singular value decomposition, this system is reduced to a linear dynamical system, which is stable and reproduces the full model behavior in a useful neighborhood of the reference. Cost functions on trajectories formulated in the model variables (e.g., time for cell volume recovery) are translated into cost functions for the dynamical system. When the model is extended by the inclusion of linear dependence of membrane transport coefficients on model variables, the impact of each such controller on the recovery cost can be estimated with the solution of a Lyapunov matrix equation. Alternatively, solution of an algebraic Riccati equation provides the ensemble of controllers that constitute optimal state feedback for the dynamical system. When translated back into the physiological variables, the optimal controller contains some expected components, as well as unanticipated controllers of uncertain significance. This approach provides a means of relating cellular homeostasis to optimization of a dynamical system.  相似文献

【背景】鱼类诺卡氏菌病潜伏期和病程较长,感染率和死亡率较高,给水产养殖业带来较大的经济损失,其病原鰤诺卡氏菌(Nocardia seriolae)是胞内寄生菌,侵入细胞后引起慢性感染是主要的致病机制。【目的】构建鰤诺卡氏菌侵染大口黑鲈(Micropterus salmoides)头肾巨噬细胞体外模型,观察鰤诺卡氏菌侵染巨噬细胞的过程并探究鰤诺卡氏菌对巨噬细胞的凋亡作用。【方法】采用密度梯度离心法分离巨噬细胞,通过特异性染色和PCR扩增巨噬细胞表达基因mpeg1对细胞进行鉴定,并通过CCK-8法和氧呼吸暴发活性测定检测巨噬细胞的活性;通过倒置荧光显微镜和流式细胞术观察侵染过程中细菌与细胞的形态与数量变化;通过双荧光流式细胞术检测、乳酸脱氢酶(lactate dehydrogenase, LDH)释放试验及线粒体膜电位检测,探究鰤诺卡氏菌对巨噬细胞的凋亡作用。【结果】从大口黑鲈头肾分离获得纯度高的巨噬细胞,经染色和PCR法鉴定为巨噬细胞;筛选出最优的体外培养条件为1640培养基+1%青霉素链霉素+1%胎牛血清。在脂多糖刺激后,巨噬细胞的氧呼吸暴发能力显著提高(P<0.05)。GFP-鰤诺卡氏菌侵染细胞2 h后细菌被细胞吞噬,4 h细胞变圆且贴壁率降低,6 h细菌大量繁殖并包围细胞,8 h后细胞大量死亡。凋亡相关实验结果表明,侵染初期巨噬细胞凋亡率增加,LDH释放增加,线粒体膜电位下降;随着侵染时间延长,细胞凋亡率下降、LDH释放量及线粒体膜电位下降减少,说明鰤诺卡氏菌对巨噬细胞起先促进后抑制凋亡的作用。【结论】通过密度梯度离心法成功分离大口黑鲈头肾巨噬细胞,并通过鰤诺卡氏菌侵染细胞后初步摸清鰤诺卡细菌在细胞水平的致病机理,建立了鰤诺卡氏菌侵染大口黑鲈头肾巨噬细胞的体外模型;证实了鰤诺卡氏菌可侵染巨噬细胞并抑制细胞凋亡,从而达到在巨噬细胞内存活,为进一步开展鰤诺卡氏菌与巨噬细胞相互作用并阐明鰤诺卡氏菌的致病机制奠定了研究基础。  相似文献

【背景】小肠结肠炎耶尔森菌(Yersinia enterocolitica)是重要的人畜共患食源性病原菌。由于其生存环境与传染性生活方式,小肠结肠炎耶尔森菌暴露在各种生存压力中,而胞膜压力应答能力对维持其环境耐受性和毒力发挥着重要作用。【目的】探究小肠结肠炎耶尔森菌在胞膜压力应答中的调节机制。【方法】通过使用多粘菌素B破坏小肠结肠炎耶尔森菌细胞膜的稳定性,并从生长能力、运动能力、生物被膜形成能力以及相关基因表达的变化探讨Rcs (Regulator of Capsule Synthesis)系统对多粘菌素B产生的胞膜压力的应答。【结果】多粘菌素B引起的细胞胞膜压力抑制了小肠结肠炎耶尔森菌的运动和生物被膜形成能力;而阻断Rcs信号途径后,小肠结肠炎耶尔森菌的运动和生物被膜形成能力有所恢复。对flhC、hmsS、hmsT等关键下游表型基因的表达水平的分析结果表明Rcs双组分系统对由多粘菌素B诱导的胞膜压力作出响应,通过感知胞膜胁迫向胞内传递信号,积极地调控细菌增强对抗菌肽的抗性。【结论】明确了Rcs双组分系统在响应多粘菌素B压力胁迫中的特异性调控作用,加深了对小肠结肠炎耶尔森菌环境应答机制的认识。  相似文献

Abstrac A study is made of the nonlocal nature of the electron energy distribution function in the positive column of a glow discharge in a tube filled with pure oxygen. The distribution function and the axial (E z) and radial (E r) electric fields as functions of radius are measured using an array of mobile probes. The experimentally obtained spatial profiles of the distribution function are used to test the applicability of the two-term approximation to the distribution function of the electrons with a nonlocal energy spectrum. The distribution function in a specified electric field E=E z +E r (where E zE r) is calculated by solving the coordinate-dependent Boltzmann equation in the two-term approximation and by directly integrating the equations of electron motion using the Monte Carlo method. A comparison between the experimental data and the results of simulations carried out for a broad parameter range shows that, in the case of a highly nonlocal electron energy spectrum, the two-term approximation makes it possible to calculate the electron distribution function with a fairly good accuracy, in which case, however, in imposing the boundary conditions, the electron losses at the plasma surface should be treated in the kinetic approximation. It is shown that using the reflection coefficient of the plasma surface for electrons instead of the loss cone in space makes it possible to accurately calculate the electron energy distribution function over the entire parameter range under consideration, including the transient region in which the electron-energy relaxation length is comparable to the characteristic plasma dimension. __________ Translated from Fizika Plazmy, Vol. 26, No. 11, 2000, pp. 1038–1045. 目的 研究严重急性呼吸综合征冠状病毒2(SARS-CoV-2)膜蛋白对宿主细胞mRNA前体(pre-mRNA)3"非翻译区(UTR)加工的影响。方法 本研究以人肺上皮细胞系A549为模型,利用瞬时转染在细胞内过表达SARS-CoV-2膜蛋白;利用RNA-Seq测序技术及生物信息学分析方法,系统性描绘宿主细胞选择性多聚腺苷酸化(alternative polyadenylation,APA)事件;Metascape数据库对发生显著APA变化的基因进行功能富集分析;RT-qPCR验证靶基因3"UTR长度变化;蛋白质免疫印迹(Western blot)检测目的蛋白表达水平。结果 SARS-CoV-2膜蛋白外源表达后宿主细胞内共813个基因发生显著APA变化。GO和KEGG分析显示,差异APA基因广泛参与有丝分裂细胞周期、调节细胞应激等生物过程,涉及病毒感染和蛋白质加工等。从中进一步筛选出AKT1基因,在IGV软件中显示3"UTR延长;RT-qPCR验证AKT1基因的3"UTR长度变化趋势;Western blot结果显示AKT1蛋白磷酸化水平增加。结论 SARS-CoV-2膜蛋白潜在影响宿主pre-mRNA的3"UTR加工,其中参与多种病毒性生物过程的AKT1基因 3"UTR延长,且其编码的蛋白质功能在细胞内被激活。  相似文献

Abstract

Isokinetic molecular dynamics simulations have been performed for 13,500 soft-spheres interacting through the inverse-power potential, ε([sgrave]/r)n , near and below the freezing temperature. The bridge function for the integral equation of the theory of liquids is extracted from the pair distribution function (PDF) obtained by the computer simulations for n = 6 and 12. The result is compared with that of approximate theories, i.e., the Rogers-Young (RY) approximation and a modified hypernetted-chain approximation for supercooled soft-sphere fluids (MHNCS approximation). Below the freezing temperature, the bridge function obtained by the computer simulation begins to oscillate around zero at intermediate distances where the second peak of the PDF appears. Such oscillatory behavior of the bridge function is well reproduced by the MHNCS approximation which includes correlations given by the leading elementary diagram, in remarkable contrast to that of the RY approximation. The present result suggests that the split second peak of the PDF for highly supercooled liquids is essentially dominated by the intermediate-distance-range correlation of the leading elementary diagram.  相似文献

Abstract

Lck is a non-receptor tyrosine kinase of the Src family that is essential for T cell activation. Dual N-terminal acylation of Lck with myristate (N-acylation) and palmitate (S-acylation) is essential for its membrane association and function. Reversible S-acylation of Lck is observed in vivo and may function as a control mechanism. Here we identify the DHHC family protein S-acyltransferase DHHC2 as an enzyme capable of palmitoylating of Lck in T cells. Reducing the DHHC2 level in Jurkat T cells using siRNA causes decreased Lck S-acylation and partial dislocation from membranes, and conversely overexpression of DHHC2 increases S-acylation of an Lck surrogate, LckN10-GFP. DHHC2 localizes primarily to the endoplasmic reticulum and Golgi apparatus suggesting that it is involved in S-acylation of newly-synthesized or recycling Lck involved in T cell signalling.  相似文献

In this paper the characteristics of a membrane having properties similar to those of frog skin are considered in relation to renal function. Such a membrane, as discussed by H. H. Ussing and K. Zerahn (Acta. Physiol. Scand.,23, 110, 1951), is capable of acting as a sodium pump actively transferring sodium ions through the membrane. Water transfer is regarded as dependent upon osmotic and hydrostatic pressures and on the permeability of the membrane to water. It is shown that for a membrane having these properties the rate of transfer of sodium is a function of the rate of transfer of water andvice versa. In this case a change in, say, the flux of water across the membrane may lead to a similarly directed change in the sodium flux across the membrane. If the E.M.F. developed by the sodium pump is controlled by a sodium retaining hormone (SRH.) and the permeability of the membrane to water by antidiuretic hormone (ADH.), it follows that there is an interaction between the two hormones which is a property of the membrane upon which they act. It is suggested that the mechanism of action of the renal tubules with regard to sodium and water excretion may be not entirely dissimilar to that discussed for frog skin, and on this supposition the characteristics of the renal response to certain physiological stimuli are deduced and found to be in excellent agreement with observation. In particular, the excretion of a urine initially hypotonic in response to an isotonic saline infusion is explained as is also the inability of, physiological doses of, ADH to inhibit isotonic or hypertonic saline diuresis. Summary. We investigated the behavior of migration of Amoeba proteus in an isotropic environment. We found that the trajectory in the migration of A. proteus is smooth in the observation time of 500-1000 s, but its migration every second (the cell velocity) on the trajectory randomly changes. Stochastic analysis of the cell velocity and the turn angle of the trajectory has shown that the histograms of the both variables well fit to Gaussian curves. Supposing a simple model equation for the cell motion, we have estimated the motive force of the migrating cell, which is of the order of piconewton. Furthermore, we have found that the cell velocity and the turn angle have a negative cross-correlation coefficient, which suggests that the amoeba explores better environment by changing frequently its migrating direction at a low speed and it moves rectilinearly to the best environment at a high speed. On the other hand, the model equation has simulated the negative correlation between the cell velocity and the turn angle. This indicates that the apparently rational behavior comes from intrinsic characteristics in the dynamical system where the motive force is not torquelike.  相似文献

We present a new formulation of the steady state, isothermal, nonlinear reaction-diffusion problem involving nth order reaction kinetics for slab geometry. This results in tractable expressions for the effectiveness factor as a function of the Thiele modulus, the Thiele modulus as a function of the centerline concentration, and the concentration profiles in the slab. The expressions are valid asymptotically in the limit of large orders n. We compare these results with the exact numerical solutions obtained by transforming the nonlinear differential equation into an integral form, using Green's function methods, and solving by successive approximations. The formulation for a membrane is also given, and the nature of the asymmetrical solution discussed. The analysis is facilitated through the introduction of pseudo-reaction orders. A comparison of the asymptotic Thiele modulus obtained herein with a previously given expression shows the present theory to be an improvement.  相似文献
