Rifamycin B production by isolated Amycolatopsis sp. RSP 3 was investigated under solid state fermentation (SSF) using agro-industrial waste materials. Corn husk was the most suitable substrate/support material with 4-fold higher production than wheat bran and corn cobs. A two-level (conventional and statistical) methodology was used to optimize fermentation parameters belong to physiological (pH, temperature and aeration), nutritional (carbon and nitrogen sources) and microbial (inoculum level and incubation time). Conventional optimization significantly improved (450%) the rifamycin B production of which two-third was associated with carbon and nitrogen sources. Starch as carbon source showed negative impact. Statistical optimization of suggested potassium nitrate (at individual level), soya bean meal and barbital (at interactive level) were observed to be the most noticeable variables in the maximization of production. At optimized conditions, inorganic nitrogen source played vital role (>59%) compared to all other factors. Overall, more than 920% increase in rifamycin B production was achieved at optimized environment.  相似文献

Effect of various fermentation media, carbon sources, nitrogen sources, phosphate concentration and culture requirements includes inoculum levels and age were determined on gentamicin production and biomass dry weight production for Micromonospora echinospora, a gentamicin producing strain. Of the substrates tested, starch as a sole carbon source promoted maximal gentamicin production, while maltose promoted maximal growth. Yeast extract as a sole nitrogen source promoted maximal growth, while soyabean meal for gentamicin production. Increasing phosphate concentration enhanced gentamicin production and observed optimum production at 1.2 g/1 (6% v/v) of phosphate having 72 h old inoculum in the medium. Highest gentamicin production was obtained after cultivation with shaking for 120 h in a medium containing starch 0.75% (w/v), soyabean meal 0.5%, K2HPO4 0.12%, CaCO3 0.4%, FeSO4 0.003% and CoCl2 0.0001%. The gentamicin production was 1.2-fold in this medium as compared to basal medium.  相似文献

Sequential methodology based on the application of three types of experimental designs was used to optimize the astaxanthin production of the mutant strain 25-2 of Phaffia rhodozyma in shake flask cultures. The first design employed was a factorial design 2(5), where the factors studied were: pH, temperature, percent of inoculum, carbon and nitrogen concentrations, each one at two levels. This design was performed in two medium types: rich YM medium and minimal medium, based on date juice (Yucca medium). With this first design the most important factors were determined (carbon concentration and temperature) that were used in the second experimental strategy: the method of steepest ascent was applied in order to rapidly approach the optimum. Finally, a second-order response surface design was applied using temperature and carbon concentration as factors. The optimal conditions stimulating the highest astaxanthin production were: 19.7 degrees C temperature; 11.25 g l(-1) carbon concentration; 6.0 pH; 5% inoculum and 0.5 g l(-1) nitrogen concentration. Under these conditions the astaxanthin production was 8100 microg l(-1), 92% higher than the production under the initial conditions.  相似文献

AIMS: Evaluation of fermentation process parameter interactions for the production of l-asparaginase by isolated Staphylococcus sp. - 6A. METHODS AND RESULTS: Fractional factorial design of experimentation (L18 orthogonal array of Taguchi methodology) was adopted to optimize nutritional (carbon and nitrogen sources), physiological (incubation temperature, medium pH, aeration and agitation) and microbial (inoculum level) fermentation factors. The experimental results and software predicted enzyme production values were comparable. CONCLUSION: Incubation temperature, inoculum level and medium pH, among all fermentation factors, were major influential parameters at their individual level, and contributed to more than 60% of total l-asparaginase production. Interaction data of selected fermentation parameters could be classified as least and most significant at individual and interactive levels. Aeration and agitation were most significant at interactive level, but least significant at individual level, and showed maximum severity index and vice versa at enzyme production. SIGNIFICANCE AND IMPACT OF THE STUDY: All selected factors showed impact on l-asparaginase enzyme production by this isolated microbial strain either at the individual or interactive level. Incubation temperature, inoculum concentration, pH of the medium and nutritional source (glucose and ammonium chloride) had impact at individual level, while aeration, agitation and incubation time showed influence at interactive level. Significant improvement (ninefold increase) in enzyme production by this microbial isolate was noted under optimized environment.  相似文献

Summary Various medium components (carbon and nitrogen sources, iron, inoculum size) and environmental factors (initial pH and the agitation speed) were evaluated for their effects on the rate and the yield of hydrogen production by Clostridium saccharoperbutylacetonicum. Among the carbon sources assessed, cells grown on disaccharides (lactose, sucrose and maltose) produced on the average more than twice (2.81 mol-H2/mol sugar) as much hydrogen as monosaccharides (1.29 mol-H2/mol sugar), but there was no correlation between the carbon source and the production rate. The highest yield (2.83 mol/mol) was obtained in lactose and sucrose but the highest production rate (1.75 mmol/h) in sucrose. Using glucose as carbon source, yeast extract was the best nitrogen source. A parallel increase between the production rate and the yield was obtained by increasing glucose concentration up to 40 g/l (1.76 mol-H2/mol, 3.39 mmol/h), total nitrogen as yeast extract up to 0.1% (1.41 mol/mol, 1.91 mmol/h) and agitation up to 100 rev/min (1.66 mol-H2/mol, 1.86 mmol/h). On the other hand, higher production rates were favoured in preference to the yield at a neutral initial pH 7 (2.27 mmol/h), 1000 mg iron/l or more (1.99 mmol/h), and a larger inoculum size, 10%, (2.36 mmol/h) whereas an initial alkaline pH of 8.5 (1.72 mol/mol), a lower iron concentration of 25 mg/l (1.74 mol/mol) and smaller inoculum size, 1%, (1.85 mol/mol) promoted higher yield over production rate.  相似文献

Calcium-alginate immobilization method for the production of alkaline protease by Bacillus licheniformis NCIM-2042 was optimized statistically. Four variables, such as sodium-alginate concentration, calcium chloride concentration, inoculum size and agitation speed were optimized by 2(4) full factorial central composite design and subsequent analysis and model validation by a second-order regression equation. Eleven carbon, 11 organic nitrogen and seven inorganic nitrogen sources were screened by two-level Plackett-Burman design for maximum alkaline protease production by using optimized immobilized conditions. The levels of four variables, such as Na-alginate 2.78%; CaCl(2), 2.15%; inoculum size, 8.10% and agitation, 139 rpm were found to be optimum for maximal production of protease. Glucose, soybean meal and ammonium sulfate were resulted in maximum protease production at 644 U/ml, 720 U/ml, and 806 U/ml when screened for carbon, organic nitrogen and inorganic nitrogen sources, respectively, using optimized immobilization conditions. Repeated fed batch mode of operation, using optimized immobilized conditions, resulted in continuous operation for 12 cycles without disintegration of beads. Cross-sectional scanning electron microscope images have shown the growth pattern of B. licheniformis in Ca-alginate immobilized beads.  相似文献

Optimizing nutritional requirements for mass production of microbial inoculants in shortened time has relevance for their economical field application. Therefore, the present study aimed at selecting suitable growth medium, optimizing its components, and up-scaling inoculum production for plant growth-promoting Pseudomonas trivialis BIHB 745. Of the different media tested, the culture exhibited maximal viable colony count in trypticase soya broth with 17.6 % increased biomass on optimizing levels of carbon source, nitrogen source, and NaCl using response surface methodology. A twofold higher biomass with 9 h shorter incubation period was obtained in optimized medium in a bioreactor in comparison to shake flasks.  相似文献

研究了三种碳源Na2CO3、NaHCO3、葡萄糖对眼点拟微绿球藻生长密度和油旨含量的影响,实验结果表明相对于葡萄糖,无机碳源NaHCO3更利于眼点拟微绿球藻的生长.以NaHCO3为碳源,研究了在不同的接种密度、NaNO3浓度下,C/N对眼点拟微绿球藻生长密度和油脂含量的影响.实验结果表明,C/N对眼点拟微绿球藻生长密度的影响与接种密度和NaNO3浓度有关,在高的NaNO3浓度时,C/N对眼点拟微绿球藻生长密度的影响很小;在低的NaNO3浓度时,随着C/N比的增加,微绿球藻的生长密度先增加后下降,存在最佳的C/N比.最佳的C/N比随接种密度而变化,在接种密度为OD440=0.10时,最佳C/N比为3,当接种密度提高到OD440=0.70时,最佳C/N比增加到5.NaNO3浓度和C/N对微藻油脂含量均有较大影响,在不同的接种密度和NaNO3浓度下都表现为C/N=1时最利于微藻油脂的积累,这与卡尔文循环过程中核酮糖-1,5-二磷酸羧化酶/加氧酶的活性有关.本实验的最佳产油培养条件为以NaHCO3为碳源,初始接种密度为OD440=0.70,C/N=1∶1,CNaNO3=0.225g/L,此时油脂产率为56.7 mg/(L·d),EPA产率为6.5 mg/(L·d).  相似文献

为高效利用水稻秸秆中的纤维素和半纤维素产油脂,采用稀酸预处理和酶水解两步法对水稻秸秆进行水解,然后以水解液为碳源,培养发酵性丝孢酵母Trichosporon fermentans HWZ004产微生物油脂。结果表明,经简单overliming法脱毒后水稻秸秆水解液中乙酸、糠醛和5-羟甲基糠醛的浓度分别为0.4 g/L、0.1 g/L和0.05 g/L。只需添加少量氮源和微量CuSO4?5H2O,该水解液即可满足T. fermentans HWZ004发酵产油脂的要求。发酵最适接种量、初始pH和温度分别是5.0%、7.0和25 ℃。T. fermentans HWZ004在优化条件下培养7 d的生物量、油脂含量和油脂产量分别是26.4 g/L,52.2%和13.8 g/L;油脂得率系数为17.0,大大高于驯化前菌株T. fermentans CICC 1368在脱毒水稻秸秆半纤维素水解液中的对应值 (11.9)。所产油脂的脂肪酸组成与植物油相似,不饱和脂肪酸含量达70%以上,宜作为生物柴油的生产原料。  相似文献

Alkaline protease production under solid-state fermentation was investigated using isolated alkalophilic Bacillus sp. Among all agro-industrial waste material evaluated, green gram husk supported maximum protease production. Solid material particle size regulated the enzyme production and yield was improved with the supplementation of carbon and nitrogen sources to the solid medium. Optimum enzyme production was achieved with 1.5% maltose and 2.0% yeast extract with 371% increase than control. Glucose did not repressed enzyme production but inorganic nitrogen sources showed little negative impact. The physiological fermentation factors such as pH of the medium (pH 9.0), moisture content (140%), incubation time (60 h) and inoculum level played a vital role in alkaline protease production. The enzyme production was found to be associated with the growth of the bacterial culture.  相似文献

Effects of the root-knot nematode (Meloidogyne incognita) on lentil (Lens culinaris) were studied under greenhouse conditions. The plants were inoculated with 250, 500, 1000, 2000 and 4000 J2 per plant. Plant growth, yield, nodulation, seed weight, chlorophyll, nitrogen, phosphorus and potassium, (NPK) contents, as compared to control, were found decreased in all the nematode infected plants. The extent of reduction increased with an increase in inoculum levels. The reductions were significant at 500 J2 and at higher inoculum levels, i.e. 1000, 2000 and 4000 J2 per pot over the control. An increase in inoculum level caused enhancement in galling, egg mass production and nematode population. At higher inoculum levels, the population of the nematode in the root as well as in the soil increased to a greater magnitude than at lower inoculum levels. On the contrary, reproduction factor (RF) and rate of population increase (RPI) decreased with increasing inoculum levels.  相似文献

High yields (1939 U/ml) of an alkaline protease were obtained in batch fermentation of a Bacillus sp. using a response surface methodology. The interaction of four variables, viz., starch, peptone, incubation time, and inoculum density, suggested inoculum density to be an insignificant variable. However, incubation time had a profound effect on protease yields at all the concentrations of carbon and nitrogen used. The response surface raised and flattened with increase in time of incubation, and maximum protease production up to 1939 U/ml was obtained after 96 h of incubation. The model equation obtained was validated experimentally at maximum starch (15 mg/ml) and peptone (7.5 mg/ml) concentration with increased incubation time up to 144 h in the presence of minimum inoculum density (1%). An overall 2.6-fold increase in protease production was obtained as compared with mean observed response (750 U/ml) at zero level of all variables. Fumonisins are a group of structurally related mycotoxins produced by Gibberella fujikuroi. The fungus produced fumonisin B1 (FB1) as early as 18 hour in a defined medium containing 1.25 mM or 2.5 mM ammonium phosphate, whereas fumonisin B1 production was repressed for 75 hour and 125 hour when mycelia were resuspended in media containing ammonium phosphate at 10 mM or 20 mM, respectively. Although total fumonisin B1 production was greater in resuspension cultures grown in higher concentrations of ammonium phosphate, the accumulation was independent of the inoculum size and carbon/nitrogen ratio. The addition of ammonium phosphate to cracked corn cultures also repressed fumonisin B1 production by 97%, and persisted for at least three weeks. Thus, biosynthesis of fumonisin B1 is regulated by a mechanism involving nitrogen metabolite repression, suggesting that control strategies that target the regulatory elements of nitrogen metabolism may be effective at reducing the risk of fumonisin contamination in food.  相似文献

Due to the increasing demand for sustainable biofuels, microbial oils as feedstock for the transesterification into biodiesel have gained scientific and commercial interest. Also, microbial carotenoids have a considerable market potential as natural colorants. The carbon to nitrogen (C/N) ratio of the respective cultivation media is one of the most important parameters that influence the production of microbial lipids and carotenoids. Thus, in the present experiment, the influence of different C/N ratios, initial glucose loadings, and ammonium concentrations of the cultivation medium on microbial cell growth and lipid and carotenoid production by the oleaginous red yeast Rhodotorula glutinis has been assessed. As a general trend, both lipid and carotenoid production increased at high C/N ratios. It was shown that not only the final C/N ratio but also the respectively applied initial carbon and nitrogen contents influenced the observed parameters. The lipid yield was not affected by different ammonium contents, while the carotenoid production significantly decreased both at low and high levels of ammonium supply. A glucose-based increase from C/N 70 to 120 did not lead to an increased lipid production, while carotenoid synthesis was positively affected. Generally, it can be asserted that lipid and carotenoid synthesis are stimulated at higher C/N ratios.  相似文献

Sandpack columns filled with heavy oil and water, with or without a nitrate-reducing inoculum, were used to determine if nitrate injections could lead to enhanced oil recovery. Production of oil and water into vials filled with helium or argon was monitored during repeated anaerobic incubations at 30 °C. Regardless of the presence of inoculum, columns containing nitrate consistently produced more oil than those without nitrate during incubations. Microbial reduction of nitrate to nitrogen with production of carbon dioxide might contribute to the establishment of gas drive. The presence of nitrate could also lead to increased production of biomass and/or biosurfactants, reducing oil-water interfacial tension. Counter-diffusion of nitrogen and carbon dioxide dissolved in column liquids and of helium or argon present in the vial gas phase likely contributed to the establishment of gas drive in all columns. Heptane was present in consistently lower concentrations in columns with nitrate than in those without nitrate at the end of incubations, suggesting that it was an important source of carbon and energy for microbial growth. Azoarcus spp. dominated the consortium when the inoculum was grown with heptane as the sole source of carbon and energy, indicating that this bacterium might contribute to the observed oil production.  相似文献

The culture parameters required for optimum production of extracellular α-amylase by the thermophilicBacillus coagulans are described. The optimum pH, temperature and incubation period for amylase production were 7, 50°C and 48 h, respectively. Age of inoculum (48 h) and its level, (2%) were critical for maximum amylase yield. The enzyme secretion was high in rice starch and beef extract as compared to other carbon and nitrogen sources tested. The addition of mustard oil cake (1%) and agitation at 1.7 Hz resulted in an enhancement of α-amylase secretion.  相似文献

Jasmonic acid is a native plant growth regulator produced by algae, microorganisms and higher plants. This regulator is involved in the activation of defence mechanisms against pathogens and wounding in plants. Studies concerning the effects of carbon: nitrogen ratio (C/Nr: 17, 35 and 70), type of inoculum (spores or mycelium) and the yeast extract addition in the media on jasmonic acid production by Botryodiplodia theobromae were evaluated. Jasmonic acid production was stimulated at the carbon: nitrogen ratio of 17. Jasmonic acid productivity was higher in the media inoculated with mycelium and in the media with yeast extract 1.7 and 1.3 times, respectively.  相似文献

ABSTRACT: BACKGROUND: Microbial lipids are a potential source of bio- or renewable diesel and the red yeast Rhodosporidium toruloides is interesting not only because it can accumulate over 50% of its dry biomass as lipid, but also because it utilises both five and six carbon carbohydrates, which are present in plant biomass hydrolysates. METHODS: R. toruloides was grown in batch and fed-batch cultures in 0.5 l bioreactors at pH 4 in chemically defined, nitrogen restricted (C/N 40 to 100) media containing glucose, xylose, arabinose, or all three carbohydrates as carbon source. Lipid was extracted from the biomass using chloroform-methanol, measured gravimetrically and analysed by GC. RESULTS: R. toruloides was grown on glucose, xylose, arabinose or mixtures of these carbohydrates in batch and fed-batch, nitrogen restricted conditions. Lipid production was most efficient with glucose (up to 25 g lipid L1, 48 to 75% lipid in the biomass, at up to 0.21 g lipid L1h1) as the sole carbon source, but high lipid concentrations were also produced from xylose (36 to 45% lipid in biomass). Lipid production was low (15-19% lipid in biomass) with arabinose as sole carbon source and was lower than expected (30% lipid in biomass) when glucose, xylose and arabinose were provided simultaneously. The presence of arabinose and/or xylose in the medium increased the proportion of palmitic and linoleic acid and reduced the proportion of oleic acid in the fatty acids, compared to glucose-grown cells. High cell densities were obtained in both batch (37 g L1, with 49% lipid in the biomass) and fed-batch (35 to 47 g L1, with 50 to 75% lipid in the biomass) cultures. The highest proportion of lipid in the biomass was observed in cultures given nitrogen during the batch phase but none with the feed. However, carbohydrate consumption was incomplete when the feed did not contain nitrogen and the highest total lipid and best substrate consumption were observed in cultures which received a constant low nitrogen supply. CONCLUSIONS: Lipid production in R. toruloides was lower from arabinose and mixed carbohydrates than from glucose or xylose. Although high biomass and lipid production were achieved in both batch and fed-batch cultures with glucose as carbon source, for lipid production from mixtures of carbohydrates fed-batch cultivation was preferable. Constant feeding was better than intermittent feeding. The feeding strategy did not affect the relative proportion of different fatty acids in the lipid, but the presence of C5 sugars did.  相似文献

碱性木聚糖酶在碱性条件下催化水解木聚糖,广泛应用于造纸、纺织等领域.着重对短小芽胞杆菌M-11产碱性木聚糖酶的发酵条件进行初步的探索.研究了菌株的生长曲线、确定最佳接种龄为16 h、最佳接种量为1%;确定最适碳源浓度为7%、最适单一氮源为氯化铵、其浓度为1.0%、最适无机盐为氯化铁、其浓度为3 mmol/L;在此基础之上进行6因素3水平的正交试验,确定最适产酶培养基组成:麸皮5%,接种量3%,氯化铵1.2%,氯化铁3.5 mmol/L,硫酸镁0.03%,氯化钠5 mmol/L,磷酸氢二钾0.4%;最适培养条件:接种龄16 h,初始pH 8.0,温度37℃,300 mL摇瓶装液量50 mL,摇床转速220 r/min,发酵周期48 h.通过对发酵条件的优化使发酵液酶活达613 IU/mL.无机氮源为其最适氮源,因此短小芽胞杆菌M-11在碱性木聚糖酶的产品开发上优于短小芽胞杆菌M -26.  相似文献

土壤氮水平对喜旱莲子草原产地和引入地基因型生长和防御的影响 同种植物生长在资源丰富生境中的个体,其防御水平被认为低于生长在资源匮乏生境中的个体。然而,生境的养分水平如何影响植物的诱导抗性和耐受性,以及这种影响在入侵植物的原产地和引入 地种群间是否存在差异,目前均知之甚少。本研究以入侵植物喜旱莲子草(Alternanthera philoxeroides)的原产地阿根廷和引入地美国的基因型为研究对象设计同质园实验,以探究土壤氮水平对植物的生长、组成和诱导性[莲草直胸跳甲(Agasicles hygrophila)取食诱导]化学防御以及耐受性的影响。实验中,我们测定了植物总生物量、伸长速率(生长速率的表征)以及叶片和根系中总碳、总氮和三萜皂苷(化学防御物质)的含量。研究结果显示,植物在低土壤氮水平下表现出较高的组成抗性(植物在低土壤氮水平下的叶片三萜皂苷含量高于其在高土壤氮水平的33%)和耐受性[植物被取食后总生物量下降的程度更低(植物在高土壤氮水平和低土壤氮水平下被取食后总生物量分别下降了24%和15%)],而在高土壤氮水平下表现出较高的诱导抗性(在高土壤氮水平下的植物被取食后叶片三萜皂苷含量与空白对照的植物相比升高了24%)。植物的组成抗性和耐受性与生长速率存在权衡,但诱导抗性与生长速率存在显著的正相关性。此外,引入地基因型在低土壤氮水平下叶片碳含量显著低于原产地基因型(-6%),但这种差异在高土壤氮水平下消失。这些结果表明,土壤氮水平 影响植物对不同防御策略的选择偏好,并且在决定引入地基因型的表现时与植食作用存在交互作用。  相似文献
