A manometric technique for the determination of turgor pressuresin laticiferous phloem tissues has given reproducible resultsin Hevea brasiliensis and a few other tree species. In Hevea,early morning pressures are in the range 7.9–15.0 atmospheres,falling during the day and recovering at night. These diurnalpressure changes are positively correlated with atmosphericrelative humidity and negatively correlated with changes intemperature, evaporation, leaf water deficit, and stomatal opening.They are not displayed by trees devoid of leaves. Thus the lossin turgor is probably the result of withdrawal of water fromthe phloem tissues under transpirational stress. Pressures at the base of the trunk normally exceed those atthe top, the gradient usually approximating to 1 atmosphere/10metres at night and rising up to six times this figure duringthe day. This increase probably reflects the development oftension gradients in the xylem during transpiration. A generalturgor gradient from base to crown does not preclude mass flowin sieve-tubes in the opposite direction provided that ratesof loading and unloading are such that a sufficient osmoticgradient is maintained in them in the required direction. No marked long-term effects of regular tapping on turgor pressurehave been noted in Hevea trees and there is no evidence forseasonal changes in turgor under our conditions.  相似文献

Treatments which increase latex yield, e. g. bark scraping,latex tapping, and bark application of 2, 4-D or 2-chloroethylphosphonicacid (CEPA) were found to enhance the activity of latex invertase.In previously untapped trees, both the introduction of tappingand the application of 2, 4-D brought about an increase in thelevel of invertase. In regularly tapped trees, the amount oflatex invertase is several times higher than in untapped treesand evidence was obtained that its activity is regulated bythe variation of latex pH. The pH of latex of the clone investigated(PR 107) was shown to vary between 6.3 and 7.1 whereas the activityof invertase, as assayed directly in the latex, has a sharpoptimum at pH 7.5 and falls rapidly with the shift of pH tothe acid side. There was no increase in the content of latexinvertase when trees adapted to regular tapping were treatedwith 2, 4-D. The effect of auxin on actual invertase activitywas essentially mediated through related increase of latex pH.The CEPA and bark scraping were also shown to increase latexpH in tapped trees. The treatment of the bark of tapped trees with CEPA increasedthe level of latex sucrose, as did auxins. Bark scraping alsohad a slight stimulatory effect. The Km of latex invertase asa function of pH was found to change in the same way as Vmax,being highest at pH optimum.  相似文献

Manipulations of production systems in rubber tree which were intended to improve sucrose translocation in tapped bark resulted in an increase of latex sucrose and of latex production and reduced the incidence of nonyielding laticiferous tissue. This was achieved by shortening the tapping cut from full to half spiral, by changing the descending direction into an ascending mode of tapping or by annual change-over of tapping panel allowing for a longer time the regeneration of bark removed above the location of the cut. The increase of latex yield did not result in a significant decrease in the growth of trees over a period of three years. Clonal differences in nonyielding bark appeared to be related to differences in sucrose depletion by tapping. In clone PB 235 which exhibited low latex sucrose, a reduction of tapping frequency resulted in an increase in sucrose level and in a decrease of bark “dryness” tending to an increase in total yield. The tapping manipulations examined did not affect latex flow characteristics such as the plugging index of latex vessels and the bursting index of lutoids. The results stress the importance of photosynthate allocation for the physiology of laticiferous system productivity and indicate the possibilities of improving assimilate economy in rubber trees. Using segments of etiolated hypocotyls of Gossypium, a comparativestudy has been made of the processes which determine the patternsof uptake of a very weak auxin, phenoxyacetic acid (POA), anda very powerful one, 2,4,5-trichlorophenoxyacetic acid (2,4,5-T). When segments are placed in solutions of POA-1-14C, a continuousincrease in the radioactivity of the tissues is accompaniedby the formation of radioactive metabolities which can be separatedfrom POA by techniques of paper chromatography. At the sametime there is a progressive increase in the amount of radio-activitywhich cannot be removed by transferring the tissues to buffer.Uptake is inhibited by low temperature, anaerobiosis, 2,4-dinitrophenol,and iodoacetate. It is concluded that the accumulation of POAinvolves its metabolic conversion to products which do not readilydiffuse out into the external medium. With 2,4,5-T-1-14C the radioactivity of the segments at firstincreases rapidly but this is followed after two hours by aphase of rapid decrease. No radioactive metoabolites can bedetected by paper chromatography and all of the 14C taken upcan be rapidly removed by transfer to buffer. The magnitudeof the decrease in radioctivity during the second phase of uptakeis balanced by a release to the medium of a matched amount ofradioactive 2,4,5-T. Uptake of 2,4,5-T is somewhat less sensitiveto temperature and anaerobiosis than uptake of POA and is bycontrast only slightly inhibited by 2,4-dinitrophenol and iodoacetate. Pretreatment of segments in buffer markedly alters the patternof uptake of 2,4,5-T but not that of POA. It reduces both theamount of 2,4,5-T initially taken up and the amount subsequentlyreleased to the medium. In addition, both net loss of radioactivityduring the course of uptake of 2,4,5-T and the reduction inthe extent of uptake following pretreatment are both arrestedby adding streptomycin, but not by the addition of pencillinor chloramphenicol. It is concluded that the uptake of 2,4,5-T involves reversibleaccumulation by a process whose efficiency decreases with time:the most likely systems are a metabolically linked mechanismfor the active transport across a membrane or reversible adsorptionon specific binding sites.  相似文献

Determination of bark contraction due to tapping made simultaneously at different positions on the trunk ofHevea trees (young PR 107, S/2 ↓) provided information about the flow area and the speed of contration spreading. The relation between truck contraction at the given distance from the cut and the amount of latex tapped was experessed in mathematical terms. A quantitative relation was also established between the yields of latex and the linear dimension of flow areas in longitudinal direction below the middle of tapping cut. From the contraction curves it is deduced that the lateral flow of latex becomes relatively more important with increasing yields.  相似文献

The concentration of phloem solute generally falls from leaves to roots. However, a local increase in latex total solid content (LILTSC) was identified near the tapping cut of rubber trees. To understand the mechanism of ethephon-stimulated latex yield, the formation and ethephon (an ethylene releaser) alleviation of the LILTSC near the tapping cut were examined. It was found that the LILTSC near the tapping cut of a tapped rubber tree was caused by the tapping-accelerated rubber biosynthesis which began following the first tapping and became significant after the fourth tapping. Ethephon stimulation markedly reduced the LILTSC. The latex yield change pattern upon ethephon stimulation was associated with the kinetic change of LILTSC and the decomposition dynamic of ethephon into ethylene. Once the LILTSC was reduced by ethylene release upon ethephon stimulation, the latex yield increased; however, when the ethylene release upon ethephon stimulation receded, the LILTSC was restored and the effect of ethephon stimulation dissipated. The reduction of LILTSC by ethephon stimulation could be ascribed to the translocation property of ethylene in plants and its regulation of aquaporins. Because maximum ethylene release upon tapping-cut-ethephon-application occured close to the tapping cut, the aquaporins were more up-regulated in this region, leading to a reduction of the LILTSC and an increase in latex yield. All these results suggest that the LILTSC near the tapping cut was caused by tapping; the ethephon-induced aquaporin up-regulation and LILTSC reduction are involved in the mechanism of ethephon-promoted latex yield.  相似文献

The effects of coumarin on the translocation of 2,4,5-trichlorophenoxyaceticacid (2,4,5-T) in bean (Phaseolus vulgaris L. cv. StringlessGreenpod) seedlings was determined. 14C-labeled 2,4,5-T wasinjected in the stem tissue at the cotyledonary node along withthe coumarin or the coumarin was added to the nutrient solutionprior to, or at the time of, 2,4,5-T treatment. The amount oftranslocation of radioactive 2,4,5-T to plant parts was thendetermined at various times after treatment. An immediate effectof coumarin was to enhance acropetal 2,4,5-T translocation tothe young shoots. This effect occurred at low 2,4,5-T treatmentlevels and appeared to be specific for 2,4,5-T since sucrosetranslocation was not affected. Prolonged treatment with coumarininhibited acropetal and basipetal 2,4,5-T translocation in amanner similar to prolonged treatment with abscisic acid (ABA).Gibberellin A3 (GA) reversed the inhibitory effects of coumarinand ABA on 2,4,5-T acropetal translocation. 1 Journal article 3244 of the Agricultural Experiment Station,Oklahoma State University. When segments excised from the etiolated hypocotyls of Gossypiumhirsutum are pretreated in buffer, the subsequent uptake ofradioactive 2,4-dichlorophenoxyacetic acid (2,4-D-1-14C) isdepressed and the net loss of radioactivity which normally followsa phase of positive uptake by freshly excised segments doesnot take place. Uptake by fresh segments, in contrast with uptakeafter pretreatment, has a high Q10 and is markedly depressedby both 2,4,5-trichlorophenoxyacetic acid (2,4,5-T) and 3-indolylaceticacid. On these grounds it is proposed that net loss resultsfrom the release of material accumulated by a specific mechanismwhich, with time, becomes inoperative. Additional experimentssuggest that part of the 2,4-D taken up by stem segments ofTriticum vulgare and Avena sativa is accumulated by a similarmechanism. For 1-cm segments, entry is most rapid through the cut ends,and the effects of pretreatment exert their maximal effectsin the tissue near the ends. Therefore very short segments havebeen used to compare the courses of uptake of phenoxyaceticacid (POA) and its 2-, 4-, 2,6-, 2,4- and 2,4,5- chloro- derivatives.The patterns observed are similar to those previously reportedfor 1 -cm segments, although the differences between compoundsare greater. The courses of uptake of 2,4-D and 2,4,5-T, bothterminate in a phase when there is a net loss. POA and the 2-chloro-substitutedacid (2-CPA) are both continuously accumulated. No net lossis found with either the 2,6- (2,6-D) or the 4- chloro (4-CPA)compounds but the rates of uptake progressively decrease toa low level. It is proposed that the processes which determine the patternof uptake of chlorinated phenoxyacetic acids include two typesof accumulation. With Type I accumulation the mechanisms involvedrapidly become disorganized after tissues are excised from theplant. Type 2 accumulation, on the other hand, is stable. Theavailable data indicate that Type I accumulation is peculiarto compounds with marked auxin-like properties.  相似文献

The effects of abscisic acid (ABA) and gibberellic acid (GA)on the translocation of 2,4,5-trichlorophenoxyacetic acid (2,4,5-T)in beans (Phaseolus vulgaris L. cv. Stringless Greenpd) seedlingswere determined. 14C-labeled 2,4,5-T was injected in the stemtissue at the cotyledonary node in 1-µl amounts alongwith the AB or GA. ABA caused an enhancement of 2,4,5-T translocationto the lower hypocotyl and roots and promoted a decrease inthe accumulation of 2,4,5-T in the young expanding shoots andprimary leaves. GA promoted the accumulation of 2,4,5-T in youngshoots. The enhanced basipetal translocation of 2,4,5-T wasmeasurable after a few hours of treatment and was partiallyeffective even in the presence of the protein synthesis inhibitorcychloheximide. An examination has been made of the phase of continuous accumulationof phenoxyacetic acid (POA) and the 2-, 4-, 2,4-, 2,6-, and2,4,5-chloro-derivatives, containing carbon-14 in the carboxylgroup, by segments of the Avena mesocotyl. On the basis of previousfindings to eliminate the initial transient components of uptake(Type I processes) the segments were pretreated for 13 to 18h in buffer or buffer containing the respective non-radioactivecompound. For five of the compounds the relationship between the rateof uptake and the external concentration takes the form of arectangular hyperbola, but for the sixth, 2,4,5-T, this relationshipdoes not hold. The data, except those for 2,4,5-T, have beenevaluated as linear regressions of rate of uptake against thequotient of rate over concentration. From each regression equationtwo constants have been derived: the point ‘B’ wherethe line intersects the rate axis (the theoretically maximumrate) and the slope of the regression ‘K’, whichcan alternatively be expressed as the concentration at whichthe observed rate equals half the value of ‘B’. The calculated values of B and K for POA are approximately twiceas great as the corresponding values for 2-CPA, and about 25times greater than for 4-CPA. The values for 2,4-D are closeto those for 4-CPA, and 2,6-D is intermediate between 2-CPAand 4-CPA. Although the constants for 2,4,5-T could not be calculatedprecisely, the rates of accumulation are about one-fourth ofthose measured for 2,4-D at equivalent concentrations. The uptake of radioactive 2,4-D is slightly depressed in thepresence of nonradioactive POA. Greater reductions are causedby 2-CPA or 2,6-D, and 2,4-D or 2,4,5-T are even more inhibitory.The pattern of inhibition caused by 2,4,5-T indicates competitionfor common sites of uptake, while POA appears not to be competitive.In corresponding experiments with POA, the presence of the otherregulators only caused small inhibitions and the order was different. Earlier work showed that in Avena accumulation is accompaniedby the conversion to a varying degree of the individual substitutedphenoxyacetic acids to conjugated derivatives. It is suggestedthat the variation between compounds in their rates of accumulationis in part due to differences in the stability of the conjugatedderivatives, and that the facility of conversion is a factorin determining physiological activity.  相似文献

The radiotracer technique is described for transport and metabolic studies in latex producing bark. This technique provided further evidence that carbohydrates are supplied to latex vessels as sucrose and that sucrose is readily used for rubber synthesis. Bark treatment with ethylene resulting in latex flow stimulation is shown to activate within a few hours sucrose and water transport into the latex vessels. The available knowledge suggests an implication of an energy-dependent process of proton excretion which could explain the earlier observed ethylene induced alkalization of latex cytosol enhancing invertase and sucrose utilization. An increase of tapping intensity decreases the level of latex sucrose and changes its distribution pattern in drained bark. There is a clonal difference in the early response to exploitation. Bark excision by conventional tapping inhibits downward sucrose translocation, the inhibition being proportional to the surface of consumed bark above the tapping cut. Seasonal variation in the level of latex sucrose is related to variations in sunshine duration and sucrose concentration rises during the process of refoliation. The results stress the importance of sucrose supply to latex vessels for latex producing potentialities and indicate an association of low sucrose availability in latex vessels with premature latex vessel degeneration and bark dryness.  相似文献

The comparative patterns of entry into segments with sealedand open ends, excised from etiolated internodes of Pisum sativum,have been examined for phenoxyacetic acid (POA) and its 2-,4-, 2,4-, 2,6-, 3,5-, 2,4,5- and 2,4, 6-chloro derivatives,each containing 14C in the carboxyl group. Sealing the ends greatly depresses the level of entry, on averagean eight-fold reduction at 9 h. Likewise, the interrelationsbetween the degree of chlorination and uptake potential aredisparate. For segments with exposed cut surfaces the finalcontent is maximal for POA and the 2-chloro compound and minimalfor the 3,5-dichloro derivative (3,5-D) with an eight-fold difference.With sealed ends this difference is reduced to two-fold butwhile 3,5-D accumulates least uptake is now highest for POAand 2,4-D. There are also changes in the order with time. Initially,2,4,5-T penetrates fastest into sealed segments but for segmentswith open ends entry is most rapid for the 4- and 2,4,6-chloroderivatives. Additions of streptomycin and cetyltrimethylammoniumbromide(CTAB) induce differential changes in the patterns of uptake.Where uptake is promoted the enhancement is not restricted toactive auxins. Sealing the ends may alter the nature of theresponse. The likely physico-chemical and metabolic processes concernedin the two routes of entry are discussed and the results comparedwith previous divergent findings on the relationship betweenchemical structure and uptake by Lemna minor and penetrationinto leaves of Phaseolus vulgaris.  相似文献

The molecular basis of the toxic properties of phenoxy herbicides in humans and animals has been insufficiently studied. In this study, damage parameters [levels of reduced glutathione (GSH) and total glutathione; activity of glutathione reductase (GR); activities of catalase (CAT) and superoxide dismutase (SOD); levels of adenine nucleotides and adenine energy charge (AEC)] were measured in human erythrocytes exposed in vitro to 2,4,5-trichlorophenoxyacetic acid (2,4,5-T) and its metabolite 2,4,5-trichlorophenol (2,4,5-TCP). Both 2,4,5-T and 2,4,5-TCP decreased the level of reduced glutathione (GSH) in erythrocytes in comparison to the control, but did not significantly change the total glutathione (2GSH + GSSG). This suggests that GSH concentration decreases concomitantly with an increase in oxidized glutathione (GSSG). 2,4,5-TCP at 100 ppm significantly decreased catalase and SOD activities. 2,4,5-T and 2,4,5-TCP did not significantly change the activity of glutathione reductase. 2,4,5-TCP decreased the level of ATP and increased the content of ADP and AMP, indicating a fall in AEC. 2,4,5-T and 2,4,5-TCP significantly changed the erythrocyte morphology. All these data are evidence of oxidative stress in erythrocytes incubated with 2,4,5-T and 2,4,5-TCP; the stress appears to be more intense in the case of 2,4,5-TCP.  相似文献

SUMMARY: A simple apparatus, designed to eliminate the bacterial infection of Hevea latex which occurs on tapping, is described. It produces two materials, ultraviolet irradiated latex and non-irradiated latex. An attempt has been made to maintain normal tapping conditions on a half-spiral cut. Natural rubber is synthesized in laticifers in the inner liber of the rubber tree (Hevea brasiliensis). Upon bark tapping, the latex is expelled due to liber turgor pressure. The mature laticifers are devoid of plasmodesmata; therefore a corresponding decrease in the total latex solid content is likely to occur due to water influx inside the laticifers. Auxins and ethylene used as efficient yield stimulants in mature untapped rubber trees, but, bark treatments with abscisic acid (ABA) and salicylic acid (SA) could also induce a transient increase latex yield. We recently reported that there are three aquaporin genes, HbPIP2;1, HbTIP1;1 and HbPIP1;1, that are regulated differentially after ethylene bark treatment. HbPIP2;1 was up-regulated in both the laticifers and the inner liber tissues, whereas HbTIP1;1 was up-regulated in the latex cells, but very markedly down-regulated in the inner liber tissues. Conversely, HbPIP1;1 was down-regulated in both tissues. In the present study, HbPIP2;1 and HbTIP1;1 showed a similar expression in response to auxin, ABA and SA, as seen in ethylene stimulation, while HbPIP1;1 was slightly regulated by auxin, but neither by ABA nor SA. The analysis of the HbPIP1;1 promoter region indicated the presence of only ethylene and auxin responsive elements. In addition, the poor efficiency of this HbPIP1;1 in increasing plasmalemma water conductance was confirmed in Xenopus oocytes. Thus, an increase in latex yield in response to all of these hormones was proposed to be the major function of aquaporins, HbPIP2;1 and HbTIP1;1. This study emphasized that the circulation of water between the laticifers and their surrounding tissues that result in latex dilution, as well as the probable maintenance of the liber tissues turgor pressure, favor the prolongation of latex flow.  相似文献

Actin microfilaments of laticiferous cells and bark wounds in Hevea brasiliensis were studied using TRITC-phalloidin fluorescent microscopy. Actin in latex from mature rubber trees was also investigated using SDS-PAGE and western-boltting. TRITC-fluorescent substance plugged the end of laticifers when latex flow stopped. Actin was detected only in C serum among the four latex fractions. Higher actin content was found in the latex collected at the beginning of tapping than in that collected just before latex flow stopped. Lower actin content was detected in the latex from rubber trees with more intensive exploitation. The present study indicated that actin microfilaments might play an important role in regulation of latex flow and plugging of the laticifers wounds.  相似文献
