We studied effects of alternating magnetic fields on the embryonic and fetal development of rats. Mated females of the Han:Wistar-strain were sham exposed or exposed continuously to a 50-Hz field or to a 20,000 pulse-per-second (pps) sawtooth magnetic field from day 0 to day 20 of pregnancy for 24 h/day until necropsied on day 20. The respective peak-to-peak intensities of the fields were 35.6 μT (sinewave) and 15.0 μT (sawtooth). Each treatment group contained 72 bred females. Control animals were kept under the same conditions without the magnetic field. No adverse effects were seen in the dams. The mean numbers of implantations and living fetuses per litter were statistically significantly increased in the 50-Hz group. There were, however, three total resorptions of litters in dams of the control group, which contributed to the difference in the number of living fetuses. The corrected body-mass gains (gains without uterine content) of dams were similar in all groups. Pregnancy rates, incidences of resorptions. late fetal deaths, and fetal body masses were similar in all groups. The incidence of fetuses with minor skeletal anomalies was statistically significantly increased in both exposed groups. Only one serious malformation (anophthalmia, sawtooth-exposed group) and a few minor visceral malformations were found. In conclusion, the magnetic fields used in this study did not increase the incidence of major malformations or resorptions in Wistar rats. The increased number of skeletal anomalies and implantations we observed indicates, however, that some developmental effects in rats may attend exposure to time-varying magnetic fields. © 1993 Wiley-Liss. Effects of alternating magnetic fields (MFs) on the embryonic and fetal development in CBA/Ca mice were studied. Mated females were exposed continuously to a sinusoidal 50 Hz (13 μT or 0.13 mT root mean square) or a sawtooth 20 kHz (15 μT peak-to-peak) MF from day 0 to day 18 of pregnancy for 24 h/day until necropsied on day 18. Control animals were kept under the same conditions without the MF. MFs did not cause maternal toxicity. No adverse effects were seen in maternal hematology and the frequency of micronuclei in maternal bone marrow erythrocytes did not change. The MFs did not increase the number of resorptions or fetuses with major or minor malformations in any exposure group. The mean number of implantations and living fetuses per litter were similar in all groups. The corrected weight gain (weight gain without uterine content) of dams, pregnancy rates, incidences of resorptions and late fetal deaths, and fetal body weights were similar in all groups. There was, however, a statistically significant increase in the incidence of fetuses with at least three skeletal variations in all groups exposed to MFs. In conclusion, the 50 Hz or 20 kHz MFs did not increase incidences of malformations or resorptions in CBA/Ca mice, but increased skeletal variations consistently in all exposure groups. A 10-mG, 50 to 60-Hz magnetic field is in the intensity and frequency range that people worldwide are often exposed to in homes and in the workplace. Studies about the effects of 50- to 100-Hz electromagnetic fields on various species of animal embryos (fish, chick, fly, sea urchin, rat, and mouse) indicate that early stages of embryonic development are responsive to fluctuating magnetic fields. Chick, sea urchin, and mouse embryos are responsive to magnetic field intensities of 10–100 mG. Results from studies on sea urchin embryos indicate that exposure to conditions of rotating 60-Hz magnetic fields, e.g., similar to those in our environment, interferes with cell proliferation at the morula stage in a manner dependent on field intensity. The cleavage stages, prior to the 64-cell stage, were not delayed by this rotating 60-Hz magnetic field suggesting that the ionic surges, DNA replication, and translational events essential for early cleavage stages were not significantly altered. Studies of histone synthesis in early sea urchin embryos indicated that the rotating 60-Hz magnetic field decreased zygotic expression of “early” histone genes at the morula stage and suggests that this decrease in early histone production was limiting to cell proliferation. Whether these comparative observations from animal development studies will be paralleled by results from studies of human embryogenesis, as suggested by some epidemiology studies, has yet to be established.  相似文献

Rats with skin-wounds surgically created on their backs were exposed immediately after surgery and every 12 h thereafter to pulsed, extremely-low-frequency magnetic fields. The shape of the pulse was a positive triangle (50 Hz, 8 mT peak). The rate of healing of skin wounds was evaluated macroscopically and by light and electron microscopy at 6, 12, 21, and 42 days after the operation. A significant increase in the rate of wound contraction was found in rats treated with magnetic fields. Forty-two days after surgery all treated animals show fully closed wounds, while control rats at the same time intervals still lacked a final 6% of the wound surface to be covered. Treated rats showed earlier cellular organization, collagen formation and maturation, and a very early appearance of newly formed vascular network.  相似文献

The influence of low repetition rate pulsed magnetic fields (LRMF) on the evoked potential (population spike) recorded from mouse hippocampal slices was investigated. LRMF were applied according to two protocols. In protocol A, LRMF applied with a constant strength (15 mT) and frequency ranging from 0.03 to 0.5 Hz resulted in an amplification of the potential. Although the frequency of 0.16 Hz was the most effective, enhancing the population spike by over 280%, it also caused an increase in spontaneous activity, seizures, and cessation of neuronal activity in 50% of the slices. In protocol B, LRMF were applied with a variable intensity (9-15 mT) and in cycles of different duration ranging from 5 to 20 min. While an increase in the amplitude of the population spike was observed in all slices exposed to LRMF applied according to protocol B, the longest exposure was the most effective. Neither seizures nor an increase in the spontaneous activity were observed in this group of the slices. These results support and extend our previous data and characterize further the relation between the pattern of applied magnetic fields and their influence on the nervous system.  相似文献

Effects of low-amplitude pulsed magnetic fields on cellular ion transport   总被引:1,自引:0,他引:1  
Pulsed magnetic fields (PMFs) are widely used to treat difficult fractures of bone and other disorders of connective tissue. It is not clear how they interact with tissue metabolism, although it has been proposed that induced currents or electric fields impinging on cell membranes may modify their ion transport function. This hypothesis was tested by treating in vitro models for ion transport processes with short-term exposure to PMFs. No change occurred in active transport of potassium or calcium in human red cells or in calcium transport through an epithelial membrane. We considered less direct action on red cell membranes, that their permeability might be modified after PMF treatment, and also that PMFs might alter the extracellular ionic activity within connective tissue by interacting with its Donnan potential. Each of these studies proved negative, and we conclude that the PMF waveforms used here do not exert a general short-term effect on cellular ion transport.  相似文献

The aim of this study was to explore the role of expectancies and beliefs about the potential effects of electromagnetic fields (EMFs) (what the subject thought the effect was going to be) and the effects of 50 Hz magnetic fields (400 microT(rms)) acute exposure on cognitive performance, the reporting of physical symptoms and some psychological and physiological parameters. Seventy-four healthy male volunteers aged between 40 and 60 years of age were randomly assigned to one of five groups, which differed in (1) the type of information they were given concerning the expected magnetic field effect on performance in cognitive tests (positive = enhancement of the performance; negative = impairment of the performance; neutral) and (2) the type of exposure (real or sham). Three groups were sham exposed with positive (group+), negative (group-) and neutral information (group+/-); one group was really exposed with neutral information (group expo) and one group was not exposed, though they wore the helmet, and did not receive any field-related information (control group). All the volunteers, except the control group, were led to believe that they would be exposed to a magnetic field of 400 microT(rms). The experimental design respected a double blind procedure and the experimental session involved three steps (pre-testing, exposure, and post-testing). Various measurements were taken, including cognitive performance, psychological parameters such as mood, vigilance, and reporting of symptoms. Physiological parameters such as blood pressure and pulse rate were also recorded. The information given did not significantly modify beliefs. No significant difference was found among the five groups depending on the type of information and the type of exposure in cognitive performance, psychological and physiological parameters. In the context of the study, with our population, the type of information given failed to induce expected changes in parameters measured. Our results do not support the hypothesis that an acute exposure to extremely low frequency magnetic fields (50 Hz, 400 microT(rms)) affects the parameters measured.  相似文献

We have previously reported that exposing the vegetative plasmodia stage of Physarum polycephalum to either individual or simultaneously applied electric and magnetic fields (45-75 Hz, 0.14-2.0 G, and 0.035-0.7 V/m) lengthens their mitotic cycle, depresses their rate of reversible shuttle streaming, and lowers their respiration rate. In this article we report the effects of simultaneously applied electromagnetic fields (60 Hz, 1.0 G, 1.0 V/m), electric fields only (60 Hz, 1.0 V/m), magnetic fields only (60 Hz, 1.0 G) on the haploid amoeba of Physarum exposed for 120-180 days. Statistically significant depressions (about 8-11%) in ATP levels were observed with all field conditions; however, respiration was significantly decreased only when amoebae were subjected to either combined fields or electric fields alone. Magnetic fields alone failed to induce a significant decrease in respiration.  相似文献

We have previously shown that neurite outgrowth from 6-day chick embryo dorsal root ganglia (DRG) in vitro was stimulated when nerve growth factor (NGF) and pulsed magnetic fields (PMF) are used in combination. 392 DRGs were studied in a field excited by a commercial PMF generator. We have now analyzed an additional 416 DRGs exposed to very similar PMF's produced by an arbitrary wave from generator and power amplifier. We reproduced our previous findings that combination of NGF and bursts of asymmetric, 220 μs-wide, 4.0 mT-peak pulses induced significantly (p<0.05) greater outgrowth than NGF alone, that fields without NGF do not significantly alter outgrowth, and that, unlike NGF alone, 4.0 mT fields and NGF can induce asymmetric outgrowth. The asymmetry does not seem to have a preferred orientation with respect to the induced electric field. Analysis of the data for the entire 808 DRGs confirms these findings. Initial changes in the fine morphology of apparently normal mouse zygotes and embryos were studied in serial sections of mouse oviducts that had been fixed in situ. These changes included extra sperm in the perivitelline space, nuclear budding, the presence of large nucleoli, perinuclear vesicles, the concentration of cytoplasmic organelles, and the extrusion of cytoplasmic ground substance. Initial changes in the nucleus and cytoplasm were undetectable when the zygotes and embryos were examined with the light microscope. It was concluded that serious abnormalities in zygotes and embryos, which may not be identified at the light microscope level, may be detected if they are examined in the electron microscope. Therefore, zygotes and embryos should be critically evaluated before being rated as normal.  相似文献

Fertilized eggs of Gallus domesticus were exposed continuously during their 21-day incubation period to either 50- or 60-Hz sinusoidal electric fields at an average intensity of 10 Vrms/m. The exposure apparatus was housed in an environmental room maintained at 37 degrees C and 55-60% relative humidity (RH). Within 1.5 days after hatching, the chickens were removed from the apparatus and tested. The test consisted of examining the effect of 50- or 60-Hz electromagnetic fields at 15.9 Vrms/m and 73 nTrms (in a local geomagnetic field of 38 microT, 85 degrees N) on efflux of calcium ions from the chicken brain. For eggs exposed to 60-Hz electric fields during incubation, the chicken brains demonstrated a significant response to 50-Hz fields but not to 60-Hz fields, in agreement with the results from commercially incubated eggs [Blackman et al., 1985a]. In contrast, the brains from chicks exposed during incubation to 50-Hz fields were not affected by either 50- or 60-Hz fields. These results demonstrate that exposure of a developing organism to ambient power-line-frequency electric fields at levels typically found inside buildings can alter the response of brain tissue to field-induced calcium-ion efflux. The physiological significance of this finding has yet to be established.  相似文献

Flow cytometry (FCM) was performed to monitor the cellular effects of extremely-low-frequency magnetic field on mouse spermatogenesis. Groups of five male hybrid F1 mice aged 8–10 weeks were exposed to 50 Hz magnetic field. The strength of the magnetic field was 1.7 mT. Exposure times of 2 and 4 h were chosen. FCM measurements were performed 7, 14, 21, 28, 35, and 42 days after treatment. For each experimental point, a sham-treated group was used as a control. The possible effects were studied by analyzing the DNA content distribution of the different cell types involved in spermatogenesis and using the elongated spermatids as the reference population. The relative frequencies of the various testicular cell types were calculated using specific software. In groups exposed for 2 h, no effects were observed. In groups exposed for 4 h, a statistically significant (P < 0.001) decrease in elongated spermatids was observed at 28 days after treatment. This change suggests a possible cytotoxic and/or cytostatic effect on differentiating spermatogonia. However, further studies are being carried out to investigate the effects of longer exposure times. © 1995 Wiley-Liss, Inc.  相似文献

Adair [Bioelectromagnetics 14:1–4, 1993] writes that “the effects of 60 Hz magnetic fields of 5 μT (50 mG) or less on biological structures holding magnetite (Fe3O4) are shown to be much smaller than those from thermal agitation; hence such interactions cannot be expected to be biologically significant.” This conclusion is questioned, because it appears to be based on a model that probably has very limited validity for pertinent biological systems. Furthermore, biologically plausible parameters can be selected to show that even this particular model does not exclude biologically significant effects of 60 Hz magnetic fields below 5 μT. This study examines childhood cancer risk in relation to certain factors likely to indicate magnetic field exposure from ground currents in the home. Substantial ground currents are most often found in homes having conductive plumbing, in which an uninterrupted metallic path in the water pipes and water main connects the grounding systems of neighboring houses. Information on plumbing conductivity was obtained from water suppliers for the homes of 347 cases and 277 controls identified in an earlier study of magnetic field exposure and childhood cancer in the Denver area. An increased cancer risk was observed for children in homes with conductive plumbing: The matched odds ratio was 1.72 (1.03–2.88) and increased to 3.00 (1.33–6.76) when analysis was limited to cases and controls who were residentially stable from the reference date to the study date. A measurement metric likely to indicate active ground currents (measurements having above-median intensity and a nonvertical orientation of <55° from the horizontal) was identified. In contrast to measured field intensity alone, for which only modest associations with cancer have been reported, this metric shows a high and significant cancer risk [matched O.R. = 4.0 (1.6–10.0)] consistent over a range of intensity and angle cutpoints. Such elevated nonvertical fields were also associated with cancer in an independent data set, which was gathered to study adult nonlymphocytic leukemia in the Seattle area. The associations of cancer with conductive plumbing and with this exposure metric both suggest that cancer risk is increased among persons with elevated magnetic field exposure from residential ground currents. © 1995 Wiley-Liss, Inc.  相似文献

Mesenchymal stem cells (MSCs) are capable of self-renew and multipotent differatiation which allows them to be sensitive to microenvironment is altered. Pulsed electromagnetic fields (PEMF) can affect cellular physiology of some types of cells. This study was undertaken to investigate the effects of PEMF on the growth and cell cycle arrest of MSCs expanded in vitro. To achieve this, cultured of normal rat MSCs, the treatment groups were respectively irradiated by 50 Hz PEMF at 10 mT of flux densities for 3 or 6 h. The effects of PEMF on cell proliferation, cell cycle arrest, and cell surface antigen phenotype were investigated. Our results showed that exposed MSCs had a significant proliferative capacity (P < 0.05) but the effect of PEMF for 3 and 6 h on cell growth was not different (P>0.05) at an earlier phase after PEMF treatment. Exposure to PEMF had a significant increase the percentage of MSCs in G1 phase compare with the control group, with a higher percentage of cells in G1 phase exposed for 6 h then that for 3 h. At the 16th hour after treatment, PEMF had no significant effect on cell proliferation and cell cycle (P>0.05). These results suggested that PEMF enhanced MSCs proliferation with time-independent and increased the percentage of cells at the G1 phase of the cell cycle in a time-dependent manner, and the effect of PEMF on the cell proliferation and cell cycle arrest of MSCs was temporal after PEMF treatment.  相似文献

The effects of pulsed electric fields of low frequency (50 Hz) on DNA of human lymphocytes were investigated. The influence of additional external factors, such as hydrogen peroxide (H2O2) and γ-irradiation, as well as the repair efficiency in these lymphocytes, was also evaluated. The comet assay, a very sensitive and rapid method for detecting DNA damage at the single cells level was the method used. A significant amount of damage was observed after exposure to the electric fields, compared to the controls. After 2 h incubation at 37°C, a proportion of damage was repaired. H2O2 and γ-irradiation increased the damage to lymphocytes exposed to pulsed electric fields according to the dose used, while the amount of the repair was proportional to the damage.  相似文献

The effect of extremely-low-frequency pulsed magnetic fields (PMF) on the response of human peripheral blood mononuclear cells to mitogenic stimulation is reported. We investigated 25 healthy control subjects. Mitogen-stimulated mononuclear cells were exposed to PMF for 72 h and an inhibition of 3H-thymidine uptake was observed in all but one subject. The degree of inhibition of 3H-thymidine uptake was as much as 60%. There was no significant difference between the blastogenic responses of mononuclear cells exposed to PMF for 12 h and control cultures. This study establishes an inhibitory effect of PMF on an in vitro measure of immune function.  相似文献

Astrocytoma risk related to job exposure to electric and magnetic fields   总被引:1,自引:0,他引:1  
To investigate the association between occupational exposure to low-frequency electric and magnetic (EM) fields and risk of brain tumors, a study was performed in Los Angeles County on 272 male adults with primary intracranial gliomas or meningiomas and 272 neighborhood controls. Complete occupational histories were collected. Risk associated with employment for more than 10 years in jobs that are presumed to entail exposure to EM fields was computed for various histological groupings. A nonsignificantly elevated risk of 1.7 was found for gliomas (all types pooled: 95% confidence interval 0.7-4.4), and a nonsignificantly reduced risk of 0.3 (95% confidence interval 0.03-3.2) was found for meningiomas. For astrocytomas, which form a subtype of the gliomas, a significantly elevated risk of 10.3 (95% confidence interval 1.3-80.8) was found; a significant upward trend (P = .01) of tumor incidence with increasing length of employment was observed. Most astrocytoma patients who worked in occupations involving exposure to EM fields were electricians or electrical engineers.  相似文献
