Summary Using antibodies against mammalian LH-RH, the double antibody immunofluorescence technique has been applied to serial cross sections of the brains of adult Rana esculenta. Immunoreactive material was found in perikarya of an unpaired nucleus in front of the preoptic recess. The axons of these perikarya also contain fluorescing material. They form a single bundle which passes under the preoptic recess, than splits into two tracts, one on either side of the optic chiasm. The two tracts reunite just before entering the median eminence. The axons end near the capillaries in the outer zone of the median eminence. Summary For the study of the hypothalamo-hypophysial system of Xenopus laevis tadpoles, hypothalamic lesions were made by means of the electrocoagulation technique. Lesioning of the ventral region of the preoptic nucleus resulted in a decrease of the number of ACTH cells in the pars distalis of the pituitary gland and in a diminution of the PAS-positive reaction of these cells. In addition, regeneration of the neurosecretory cells of the ventral region of the preoptic nucleus observed 6 weeks after lesioning was accompanied by the reappearance of normal PAS-positive ACTH cells in the pars distalis. It is suggested that the neurosecretory cells of the ventral region of the preoptic nucleus of Xenopus laevis tadpoles are related to the ACTH synthesizing cells, probably by producing CRF.Dedicated to Prof. Dr. med. Summary The influence of a long daily photoperiod and continuous darkness was studied on the hypothalamo-hypophysial system of Rana temporaria. After six weeks of treatment (December-January 1968/1969) the secretory activity of the hypothalamo-hypophysial system of the light-treated animals was enhanced.The amount of aldehyde-fuchsin positive material in the preoptic nucleus increased as a result of the light treatment, and decreased in the median eminence, whereas in the posterior lobe no difference could be observed between the light- and dark-treated animals.The karyometric results showed an increase of the nuclear volume of the cells of the preoptic nucleus after light treatment; small differences were noticed between distinct areas of the preoptic nucleus.Autoradiographic data especially showed that light influenced the hypothalamo-hypophysial system to a high degree. Half an hour after the injection of 35S-cysteine, the incorporation of the isotope in the preoptic nucleus of the light-treated animals was five times as much as in the dark-treated animals. In the median eminence and in the posterior lobe, the autoradiographic results confirmed the results found with regard to the amount of aldehyde-fuchsin positive material. Summary The distribution of monoamine oxidase (MAO) in the brain of Xenopus laevis tadpoles (stage 52–56) was studied histochemically with a modified Glenner's tryptamine-tetrazolium method. A moderate activity was observed in fibre regions of the striatum and septum (including the medial and lateral forebrain bundles), in the neuropil of the nucleus amygdalae, in the commissura anterior and commissura hippocampi, in the fibre regions of the diencephalon (including the optic chiasma), in the fibre regions of the tectum opticum and the tegmentum of the mesencephalon and in the white substance of the ventral half of the medulla oblongata. A greater MAO activity was found in the neuropil of the entire nucleus praeopticus. In the partes anterior and magnocellularis of this nucleus, MAO positive fibres are present in close contact with the perikarya, indicating a monoaminergic innervation of these neurons. The perikarya themselves did not show MAO activity. In the neurons of the nucleus praeopticus epichiasmaticus, the paraventricular organ (PVO) and nucleus infundibularis dorsalis (NID), only a slight MAO activity has been demonstrated in the perikarya, whereas a strong MAO positivity was found in the intraventricular protrusions and the neuropil. These data indicate the aminergic character of the neurons of these nuclei. From the postoptic fibre region a MAO positive tract was observed towards the developing median eminence and pars intermedia of the hypophysis. The pars nervosa and some cells of the pars distalis also contained MAO. Along the border of the aquaeduct of Silvius and the fourth ventricle, MAO positive liquor-containing neurons are also present.The distribution of acetylcholinesterase (AChE) was investigated in the hypothalamohypophysial region. AChE activity was found in the neuropil of the nucleus praeopticus magnocellularis, in the fibres of the optic chiasma and in the postoptic fibre region. The neurons of the PVO and NID were AChE negative. An AChE positive tract could be traced from the postoptic fibre region to the developing median eminence and pars nervosa. The pars distalis did not show AChE activity. However, in tadpoles reaching the metamorphic climax, ChE activity appeared in certain cells of the pars distalis; this might be related to degenerative phenomena in the acidophilic cells. The absence of AChE activity in the pars intermedia indicates a regulation of MSH release by peptidergic nerves to be unlikely.The stimulating interest and helpful advice of Prof. Dr. P. G. W. Summary After some remarks on ambiguities in the conventions of the Braun-Blanquet approach of phytosociology, a syntaxonomical study is presented of theCalthion palustris (wet grassland vegetation) of the Netherlands and surroundings. A numerical analysis of the non-optimal vegetation yielded a classification well comparable with the one established before. One new association is described. The pattern of variation in this dataset is compared in several ways with the one described elsewhere. Synoptic tables of different associations from several authors were compared by numerical analyses. Summary The development and activity of the basophils in the meso-adenohypophysis of Anopthichthys jordani were studied in relation to the maturation of the gonads. In addition, pituitaries and ovaries of post-spawning females were studied during a period 0–11 days subsequent to spawning. The large and mainly PAS-positive basophilic cells in the meso-adenohypophysis were the only cell type that showed a correlation with the development and maturation of the ovaries and testes. These cells were subjected to a partial or complete degranulation in females following oviposition, when yolk formation in the ovary was at a maximum. It is concluded that the large PAS-positive basophils in the mesoadenohypophysis have a gonadotropic function and that only one gonadotropic cell type is present in the adenohypophysis of Anoptichthys jordani.This study was partly supported by a grant from the Netherlands Organization for the Advancement of Pure Research (Z.W.O.).I wish to express my gratitude to Prof. Dr. P. Summary The ultrastructure of the paraventricular organ ofXenopus laevis andRana esculenta has been investigated. Special attention has been paid to contact regions between nerve cells of the paraventricular organ and other nerve cells.According to the results of the glutaraldehyde-osmium tetroxide fixation as well as the zinc iodide-osmium method, it appears that the perikarya of several nerve cells of the paraventricular organ make synaptic contacts with dendrites of other nerve cells. It is suggested that the presence of the somato-dendritic synapses is a morphological indication for the sensory function of the nerve cells.Thanks are due to Prof. Dr. P. G. W. Summary By use of a new antiserum, raised against synthetic pigment-dispersing hormone (PDH) from Uca pugilator, immunoreactive structures were studied at the light-microscopic level in the eyestalk ganglia of Carcinus maenas and Orconectes limosus. PDH-reactivity was mainly found in two types of neurons that were located between the medulla interna (MI) and the medulla terminalis (MT) in both species. Several additional perikarya were located in the distal part of the MI in O. limosus. In C. maenas, two to three PDH-positive perikarya were found in the region of the X-organ (XO) in the MT. Processes from single and clustered cells could be traced into all medullae of the eyestalk. Axons from the immunoreactive perikarya running between MI and MT form a larger tract that traverses the MT. Fibers from this tract give rise to extensive arborizations and plexuses throughout the proximal MT. A plexus containing very fine fibers is located at the surface of the MT in a position distal to the XO-area of C. maenas only. The proximal plexus also receives PDH-positive fibers through the optic nerve. PDH-perikarya in the cerebral ganglion may also project into the more distal regions of the eyestalk. Distal projections of the perikarya between the MI and MT consist of several branches. Most of these are directed toward the MI and ME (medulla externa) wherein they form highly organized, layered plexuses. One branch was traced into the principal neurohemal organ, the sinus gland (SG). In the SG, the tract gives off arborizations and neurosecretory terminals. It then proceeds in a proximal direction out of the SG, adjacent to the MT. Its further course could not be elucidated. The lamina ganglionaris (LG) receives PDH-fibers from the ME and fine processes from small perikarya located in close association with the LG in the distal part of the first optic chiasma. The architecture of PDH-positive elements was similar in both C. maenas and O. limosus. The distribution of these structures suggests that PDH is not only a neurohormone but may, in addition, have a role as a neurotransmitter or modulator. Immunostaining of successive sections with an FMRF-amide antiserum revealed co-localization of FMRFamideand PDH-immunoreactivities in most, but not all PDH-containing perikarya and fibers. The axonal branch leading to the SG and the SG proper were devoid of FMRFamide immunoreactivity.  相似文献

Summary The distribution of an immunoreactive endothelin-1-like peptide was investigated in the nereid, Neanthes diversicolor, using an antiserum raised against synthetic endothelin-1. Immunoreactive perikarya were localized in the brain, and nerve fibers containing endothelin-1-like material were found in the neuropil occupying the central portion of the brain. No immunostained fiber elements were traced in the circumesophageal connectives. Immunoreactive perikarya occurred in the subesophageal ganglion. From this ganglion, specifically stained fibers run posteriorly toward the ventral nerve cord. In each segmental ganglion, immunoreactive neurons were observed in medio-ventral and latero-ventral regions, and one or two marked fibers extended to the parapodium. In the parapodium, small immunoreactive perikarya and fiber elements were visible. Immunolabeled fibers occurred in the stomatogastric nerves, in the wall of the buccal cavity, and in the pharynx, esophagus, intestine and its anal region. Immunoreactive perikarya and nerve fibers were visualized between the circular muscle layer and epithelial cell layer in the esophagus and intestine. The endothelin-1-like substance shown to occur in N. diversicolor appears to function as a neurotransmitter or neuromodulator.  相似文献

Summary The ultrastructure of the slit diaphragms between the pedicels of the podocytes of the prosobranch Viviparus viviparus and between the cytoplasmic tongues of the haemocyanin producing pore cells of the pulmonate Lymnaea stagnalis was investigated. In both cell types 2 diaphragms are present in the slits. They form a 3-dimensional sieve structure with holes of respectively 90 × 110 Å (podocyte) and 200 × 220 Å (pore cell). Injection experiments showed that the size of the holes of the pore cell sieve matches that of particles which can be ingested by this cell type. The substructure of the sieves of the molluscs is compared to that of the 2-dimensional sieve of the podocytes of the mouse and the rat.The authors thank Mrs. J.E. Summary In several coronet cells of the saccus vasculosus of Scylliorhinus large quantities of glycogen occur, as shown by light and electron microscopy. Summary Following exposure to tritiated 5-HTP, silver grains were observed over the perikarya of the GSCs (Giant serotonin cells) of Helix pomatia and other known serotonin-containing neurones in light and electron microscope autoradiograms. There was no indication that the 5-HTP was taken up by nerve endings or by non-nervous structures. The distribution of radioactivity was completely different in autoradiograms of tissue exposed to tritiated serotonin. Silver grains, often in very high concentrations, were observed only over certain fine axon branches and processes thought to be nerve endings. Electron microscope autoradiography showed that these processes contained small dense-cored vesicles, morphologically identical to those thought to sequester serotonin in the perikarya of the GSCs. The accumulation of tritiated tryptophan was less specific; all the neurone perikarya showed an accumulation of radioactivity after exposure to this substance.We are grateful to Professor J. F. Summary In Xenopus laevis tadpoles the relation between a paired nucleus of bio-amine producing neurons in the caudal hypothalamus and the pars intermedia of the hypophysis was studied.Treatment of the animals (stage 49 to 50 of Nieuwkoop and Faber's normal table) with reserpine caused aggregation of the skin melanophores within one hour, followed by redispersion five to six hours after the beginning of the experiment. This was at exactly the same time as the bio-amines in the caudal hypothalamus disappeared. However, the drug was ineffective if the nuclei had been removed. This indicates that reserpine acts via these nuclei and does not influence the skin melanophores directly.It was concluded that the initial aggregation of the melanophores may be the result of a reduced extrusion of MSH from the pars intermedia, caused by an increased output of a MIF by the bio-amine producing nuclei. The redispersion was explained by assuming that the bio-amines were depleted and the nuclei stopped with the extrusion of the MIF. This does not mean that the production of a MIF is the only function of the paired bio-amine producing nucleus in the caudal hypothalamus.The author thanks Prof. Dr. P. G. W. Zusammenfassung In vergleichend autoradiographischen Untersuchungen wurde der Einbau von percutan applizierten 3H-Uridin, 3H-Histidin und 3H-Glucose in die wichtigsten Organsysteme (Epidermis, ZNS, Muskeln, Chorda, Leber, Kiemendarm, Darm) von Branchiostoma lanceolatum (Acranier) und Brachydanio rerio (Teleosteer) nach Inkorporationszeiten von 11 min bis zu 7 Tagen verfolgt. Der Stoffwechsel der markierten Substanzen in den einzelnen morphologisch miteinander zu homologisierenden Organen war bei den beiden Spezies sehr ähnlich, bei Branchiostoma allerdings (mit Ausnahme des ZNS) 4–5mal stärker als bei Brachydanio. Bei dem letzteren wurde außerdem eine zeitliche Verzögerung in der Tracer-Aufnahme (lag-Phase) beobachtet. Insbesondere der ZNS-Stoffwechsel von Acraniern zeigte ähnliche Charakteristika wie der von Vertebraten: Verbleib des Hauptanteils der neusynthetisierten RNS im Perikaryon, axonalen Protein-Transport, Vorwiegen der Glykogensynthese in den Nervenfaserendformationen. Allerdings fanden sich im ZNS von Branchiostoma niedrigere Stoffwechselraten als im ZNS von Brachydanio.
Comparative histophysiological investigations of different organs in Branchiostoma lanceolatum (Cephalochordata) and Brachydanio rerio (Teleostei)
Summary Incorporation of 3H-uridine, 3H-histidine and 3H-glucose into some organs (epidermis, CNS, muscles, spinal cord, notochord, liver, gills, intestine) of Branchiostoma lanceolatum (Acrania) and Brachydanio rerio (Teleostei) was investigated by means of comparative autoradiograms following incorporation periods of 11 min to 7 days. The metabolism of the labeled substances in the various homologous organs examined was quite similar, although it was 4 to 5 times higher in Branchiostoma than in Brachydanio; in the latter there was also a delay of tracer incorporation of about 3 hrs, a so-called lag-phase. In particular the metabolism of the CNS of Branchiostoma showed the same characteristics as the CNS of vertebrates, e. g. storage of neuronally synthesized RNA in the neuronal perikarya, axonal flow of proteins, glycogen synthesis in nerve endings. However, metabolic activity of the CNS was lower in Branchiostoma.

Summary Corticotropin-releasing factor (CRF) was localized in the brains of two passerine species, the European starling (Sturnus vulgaris) and the song sparrow (Melospiza melodia), by means of immunohistochemistry. The hypothalamic distribution of this peptide in these species includes a complex of immunoreactive perikarya observed in the paraventricular nucleus (PVN), in both its medial and lateral divisions. Nerve fibers were also seen running from these areas to the anterior median eminence (AME) where a terminal field is apparent. A wide variety of extra-hypothalamic nuclei containing CRF-immunoreactive cells and fibers were identified. An apparent CRF terminal field can be visualized in the lateral septum. A dense fiber plexus is present in the nucleus accumbens (Ac) and more caudally in the nucleus of the stria terminalis (nST). In colchicinepretreated animals, it was revealed that these areas also contain CRF-stained perikarya. The pattern of CRF immunoreactivity in the Ac-nST complex is continuous, with no distinction apparent between the nuclei. The medial preoptic area (mPOA) and the adjacent diagonal band of Broca contain CRF-fibers, while cells are apparent in the mPOA. In the mesencephalon, cells were visualized in the midbrain central gray; a terminal field and scattered positively stained perikarya were found in areas more ventral to the central grey that are adjacent to the third cranial nerve. Scattered cells were also seen at the border of the nucleus intercollicularis-nucleus mesencephalicus lateralis, pars dorsalis complex. In contrast to mammalian studies, no immunoreactive nerve fibers or perikarya were observed in telencephalic areas homologous to the mammalian neocortex. These studies confirm the presence of a CRF path-way regulating pituitary function and suggest a broad role played by CRF as a neuromodulator or neurotransmitter in autonomic and possibly behavioral activities in these species.  相似文献

Summary The fine structure of the brain of the monogenean Gastrocotyle trachuri (Platyhelminthes) is described. The brain consists of a central neuropile surrounded by a layer of cell bodies. The neuropile is composed of a fine meshwork of naked neurites which contain various types of vesicles and other organelles although microtubules have not been found. Five kinds of vesicles; three clear types and two dense types, were found within the neuropile.Two types of neuronal cell body were identified on the basis of their vesicle contents although it is possible that these two types represent the extremes of a single cell type. A characteristic feature of the neuronal perikarya of Gastrocotyle is the presence of deep infoldings into the cell of the outer membrane. These infoldings often contain fibrous interstitial material and in a number of cases hemidesmosome-like structures have been found in the distended, distal end of the infoldings.  相似文献

Nineteen species of the paper wasp genus Ropalidia are recorded from Vietnam, of which three are new records for Vietnam. A key to all of the species, distribution records and taxonomic remarks are given. New synonymies proposed are: Ropalidia bicolorata parvula van der Vecht, 1962, under R. bicolorata van der Vecht, 1962; R. hongkongensis juncta van der Vecht, 1941, under R. hongkongensis (de Saussure, 1854); Icaria jucunda Cameron, 1898, and R. marginata sundaica van der Vecht, 1941, under R. marginata (Lepeletier, 1836); R. rufocollaris atrata van der Vecht, 1941, under R. rufocollaris (Cameron, 1900); and R. stigma rufa van der Vecht, 1941 and R. stigma nigrolineata van der Vecht, 1962, under R. stigma (Smith, 1858).  相似文献

Summary The peptidergic nature of most of the fibres of the neurosecretory system of Scyliorhinus caniculus has been demonstrated with the oxidation method of Naumann and Sterba (1976). Some of the fibres of this peptidergic system terminate on capillaries in the hypothalamic floor, some on neurointermediate cells. A recent proteomic analysis of the thylakoid lumen of Arabidopsis thaliana revealed the presence of several PsbP-like proteins, and a homologue to this gene family was detected in the genome of the cyanobacterium Synechocystis sp. PCC 6803 (Schubert M, Petersson UA, Haas BJ, Funk C, Schröder WP, Kieselbach T (2002) J Biol Chem 277, 8354–8365). Using a peptide-directed antibody against this cyanobacterial PsbP-like protein (sll1418) we could show that it was localized in the thylakoid membrane and associated with Photosystem II. While salt washes did not remove the PsbP-like protein from the thylakoid membrane, it was partially lost during the detergent-based isolation of PSII membrane fractions. In total cell extracts this protein is present in the same amount as the extrinsic PsbO protein. We did not see any significant functional difference between the wild-type and a PsbP-like insertion mutant.  相似文献

Females of the social wasp, Belonogaster petiolata,rub the secretion of van der Vecht's gland, located on their terminal gastral sternite, onto the nest pedicel. In bioassays, the secretion was repellent to two species of ants, while shortchain acids were effective releasers of rubbing behavior. Rubbing was associated with pedicel enlargement and departure from the nest in preemergence colonies. Its frequency was high where wasps were often exposed to ants and low where ants were rare or absent. Rubbing also decreased significantly from the pre-to the postemergence stage of the colony cycle. In both stages, subordinate foundresses rubbed more often than queens or workers. These observations support the hypothesis that rubbing behavior and the secretion of van der Vecht's gland function in chemical defense of the nest against ant predation. The general morphology of the gland in B. petiolataresembles that of the four other independent-founding polistine wasp genera.  相似文献
