铁是大多数生物包括细菌生存的必需营养元素.对于感染宿主的致病细菌,血红素(heme/haem)可作为一种主要的铁来源.血红素转运系统在革兰氏阴性菌和革兰氏阳性菌中均有发现和鉴定,其转运机制在革兰氏阴性菌中有较为深入研究.革兰氏阴性菌血红素转运系统主要由分泌于细胞外的血红素载体(hemophore)、血红素受体、TonB ExbB ExbD复合物、ABC转运体、血红素降解蛋白和调控蛋白等结构单元组成.对参与该系统的各个蛋白结构特点以及它们之间的相互作用机制的讨论,有助于对病原菌致病机制的深入研究和抗菌新药的研发.  相似文献

A number of inhibitor systems which have been reported to allow selection of Gram-negative bacteria were tested against Gram-positive and Gram-negative isolates of dairy origin. No one system worked perfectly. A mixture of crystal violet-penicillin-nisin or monensin had least inhibitory effect on Gram-negative isolates whereas Selectocult (a commercially available mixture of Benzalkon A 50% and crystal violet) and sodium deoxycholate were the most effective inhibitors of Gram-positive bacteria. Cetrimide-fucidin-cephaloridine solutions, which have been reported as allowing selective growth of pseudomonads, were not so specific when applied to milk systems.  相似文献

Abstract A wide variety of proteins are exported or secreted by a range of morphologically distinct bacteria. The processes of protein export are most extensively characterised in Escherichia coli , where recent advances have been made in the identification of genes involved in forming the export machinery. Both Gram-positive and Gram-negative bacteria secrete proteins into the medium. Gram-negative bacteria have adopted a variety of approaches in order to overcome the additional permeability of the outer membrane.  相似文献

Ribosomes from Gram-positive and Gram-negative bacteria have been analysed for the presence of ribosomal protein S1 by three methods, sodium dodecyl sulfate polyacrylamide gel electrophoresis, immunoreaction with E. coli anti-S1 and chromatography on poly (U)-Sepharose. We observed that protein S1 is predominantly present in Gram-negative bacteria in comparison with Gram-positive bacteria. Exceptions are noted in both species.  相似文献

Glutathione transferases (GSTs) have been widely studied in Gram-negative bacteria and the structure and function of several representatives have been elucidated. Conversely, limited information is available about the occurrence, classification and functional features of GSTs both in Gram-positive bacteria and in Archaea. An analysis of 305 fully-sequenced Gram-positive genomes highlights the presence of 49 putative GST genes in the genera of both Firmicutes and Actinobacteria phyla. We also performed an analysis on 81 complete genomes of the Archaea domain. Eleven hits were found in the Halobacteriaceae family of the Euryarchaeota phylum and only one in the Crenarchaeota phylum. A comparison of the identified sequences with well-characterized GSTs belonging to both Gram-negative and eukaryotic GSTs sheds light on their putative function and the evolutionary relationships within the large GST superfamily. This analysis suggests that the identified sequences mainly cluster in the new Xi class, while Beta class GSTs, widely distributed in Gram-negative bacteria, are under-represented in Gram-positive bacteria and absent in Archaea.  相似文献

革兰氏阴性菌在生长繁殖过程中需要从外界摄取营养物质。一些小分子营养物质可以自由地通过革兰氏阴性菌的细胞膜,而一些大分子营养物质的转运需要特异性的TonB复合物依赖性的外膜受体进行转运。TonB复合物由TonB、ExbB、ExbD构成,是革兰氏阴性菌对外界营养物质主动转运过程的能量提供单位,在革兰氏阴性菌分布广泛。近年来,对TonB-ExbB-ExbD复合物的功能、结构及作用机制取得了重大研究进展,然而此复合物在不同的细菌也存在功能及作用机制上的差异。基于此背景,本文综述了TonB复合物的功能和结构研究进展,并分析了TonB复合物在革兰氏阴性菌中的分布、进化,比较了不同革兰氏阴性菌此复合物的差异,有助于进一步发现和揭示TonB复合物的新功能。  相似文献

Lipopolysaccharide (LPS) is the major surface molecule of Gram-negative bacteria and consists of three distinct structural domains: O-antigen, core, and lipid A. The lipid A (endotoxin) domain of LPS is a unique, glucosamine-based phospholipid that serves as the hydrophobic anchor of LPS and is the bioactive component of the molecule that is associated with Gram-negative septic shock. The structural genes encoding the enzymes required for the biosynthesis of Escherchia coli lipid A have been identified and characterized. Lipid A is often viewed as a constitutively synthesized structural molecule. However, determination of the exact chemical structures of lipid A from diverse Gram-negative bacteria shows that the molecule can be further modified in response to environmental stimuli. These modifications have been implicated in virulence of pathogenic Gram-negative bacteria and represent one of the molecular mechanisms of microbial surface remodeling used by bacteria to help evade the innate immune response. The intent of this review is to discuss the enzymatic machinery involved in the biosynthesis of lipid A, transport of the molecule, and finally, those enzymes involved in the modification of its structure in response to environmental stimuli.  相似文献

The influence of Gram-negative bacteria on the migratory and adhesive activity of polymorphonuclear leukocytes (PMNL) in the peripheral blood of clinically normal donors has been studied by the specially developed method with the use of Boyden chambers. Pseudomonas and enterobacteria have been found to produce complex and various effects on the above-mentioned properties of PMNL. When incubated in fresh serum, Gram-negative bacteria are capable of enhancing the migratory activity of PMNL, this property being least pronounced in P. aeruginosa. The incubation of live bacteria from the authors' collection in the patients' sera or in sera obtained from normal donors and inactivated by heating induces no hemotaxis of PMNL, and P. aeruginosa strains even suppress it under such conditions. The isolated Gram-negative bacteria under study increase the number of highly adhesive PMNL in the population used in this investigation, but P. aeruginosa cultures do not produce such effect.  相似文献

Many different bacteria secrete proteases into the culture medium. Extracellular proteases produced by Gram-positive bacteria are secreted by a signal-peptide-dependent pathway and have a propeptide located between the signal peptide and the mature protein. Many extracellular proteases synthesized by Gram-negative bacteria are also produced as precursors with a signal peptide. However, at least two species of Gram-negative bacteria secrete one or more proteases via a novel signal-peptide-independent route. Most proteases secreted by Gram-negative bacteria also have a propeptide whose length and location vary according to the protease. Specific features of protease secretion pathways and the mechanisms of protease activation are discussed with particular reference to some of the best-characterized extracellular proteases produced by Gram-positive and Gram-negative bacteria.  相似文献

In 44 isolated cultures of Gram-negative bacteria, besides commonly known pathogenicity factors, their adhesive activity towards the cells of the buccal epithelium and their interrelations with the representatives of normal microflora which determine natural resistance to colonization have been studied. The artificial adhesion of target cells is accompanied by the inhibition on the natural colonization of epithelial cells by Streptococcus salivarius; it is, therefore, evident that adhesiveness is one of the factors which determine the behavior of microorganisms in cenoses. The circulation of adhesive strains of Gram-negative bacteria has been noted in the burn ward.  相似文献

Synthesis and antibacterial activity of metronidazole–triazole conjugates are reported. Total 21 hybrid compounds have been synthesized with different substitution pattern on the triazole ring in order to study their influence on the antibacterial activity. These compounds demonstrated potent to weak antibacterial activity against Gram-positive, and Gram-negative bacteria. Six compounds have shown equal or better antibacterial activity against Gram-negative strains than the reference compound.  相似文献

Some proteins extracted from the cell wall of Gram-positive and Gram-negative bacteria that affect the time for which meat can be stored are antigens produced by the strains from our laboratory collection or from field samples. 血红素作为宿主体内最丰富的铁离子来源,是致病菌营养竞争的主要目标,尤其对于血红素自身合成途径部分丧失的细菌。革兰氏阴性菌血红素转运系统由血红素载体蛋白(Hemophore)、外膜血红素受体、TonB-ExbB-ExbD复合物、ABC转运体等组成。Hemophore是存在于细菌细胞膜上或分泌到胞外环境中的一种蛋白。它能从宿主血红素结合蛋白中捕获血红素并将其传递给外膜受体。目前,在不同革兰氏阴性菌中已发现3种类型的Hemophore,分别是HasA、HxuA和HmuY型。本文将详细描述这3种Hemophore捕获血红素及与外膜受体相互作用的机制,以期为进一步研究其他细菌血红素载体蛋白的功能及作用机制奠定基础。  相似文献

Mammals have four peptidoglycan recognition proteins (PGRPs or PGLYRPs), which are secreted innate immunity pattern recognition molecules with effector functions. In this study, we demonstrate that human PGLYRP-1, PGLYRP-3, PGLYRP-4, and PGLYRP-3:4 have Zn(2+)-dependent bactericidal activity against both Gram-positive and Gram-negative bacteria at physiologic Zn(2+) concentrations found in serum, sweat, saliva, and other body fluids. The requirement for Zn(2+) can only be partially replaced by Ca(2+) for killing of Gram-positive bacteria but not for killing of Gram-negative bacteria. The bactericidal activity of PGLYRPs is salt insensitive and requires N-glycosylation of PGLYRPs. The LD(99) of PGLYRPs for Gram-positive and Gram-negative bacteria is 0.3-1.7 muM, and killing of bacteria by PGLYRPs, in contrast to killing by antibacterial peptides, does not involve permeabilization of cytoplasmic membrane. PGLYRPs and antibacterial peptides (phospholipase A(2), alpha- and beta-defensins, and bactericidal permeability-increasing protein), at subbactericidal concentrations, synergistically kill Gram-positive and Gram-negative bacteria. These results demonstrate that PGLYRPs are a novel class of recognition and effector molecules with broad Zn(2+)-dependent bactericidal activity against both Gram-positive and Gram-negative bacteria that are synergistic with antibacterial peptides.  相似文献

The function of the S-layer, a regularly arranged structure on the outside of numerous bacteria, appears to be different for bacteria living in different environments. Almost no similarity exists between the primary sequences of S-proteins, although their amino acid composition is comparable. S-protein production is directed by single or multiple promoters in front of the S-protein gene, yielding stable mRNAs. Most bacteria secrete S-proteins via the general secretory pathway (GSP). Translocation of S-protein across the outer membrane of Gram-negative bacteria sometimes occurs by S-protein-specific branches of the GSP. O-polysaccharide side-chains of the lipopolysaccharide component of the cell wall of Gram-negative bacteria appear to function as receptors for attachment of the S-layer. Silent S-protein genes have been found in Campylobacter fetus and Lactobacillus acidophilus. These silent genes are placed in the expression site in a fraction of the bacterial population via inversion of a chromosomal segment.  相似文献
