Summary A solid state fermentation process is described for the production of citric acid from apple pomace byAspergillus niger NRRL 567. The yields of citric acid varied with the pomace varieties, and were dependent on (1) the amount of methanol present in the pomace, and (2) the fermentation time and temperature. The process yielded as much as 90 g citric acid/kg apple pomace fermented in the presence of 3–4% methanol at 30°C in five days.
Resumen Se describe un proceso de fermentación en estado sólido para la producción de ácido cítrico a partir de residuos de manzana, utilizandoAspergillus niger NRRL 567. La producción de ácido cítrico varió con el tipo de residuo utilizado y dependió de (1) la cantidad de metanol presente en los residuos y (2) del tiempo y la temperatura del proceso. La fermentación puede producir hasta 90 g de ácido cítrico/kg de residuo de manzana, fermentado en presencia de 3–4% de metanol a 30°C de temperatura durante 5 días.

应用基因组改组技术提高,L-谷氨酸生产菌在高温发酵条件下的谷氨酸产量。以天津短杆菌T6—13变异株SW07-1为原始亲株,分别经紫外线(UV)-硫酸二乙酯(DES)和X射线诱变,获得5株耐温性能略有提高的突变菌株。经2轮基因组改组,获得耐高温(能在44℃生长)的L-谷氨酸菌株F2-50。F2—50在38℃下,摇瓶发酵40h,发酵液中L-谷氨酸浓度比原始出发菌株提高了近41%,在41℃高温下,摇瓶发酵40h,L-谷氨酸浓度比原始出发菌株提高了近2倍。 相似文献

Summary The production of gibberellic acid (GA3) was found to be influenced by physical and nutritional factors. The nutritional factors studied were the urea nitrogen and MgSO4 concentrations, whereas the physical factors were moisture content, autoclaving time, inoculum ratio and moist medium to culture flask volume ratio. The yield of GA3 was improved 2.9 times by judicious selection of these parameters. Thus, the increase in GA3 yield in the wheat bran medium reduces the expense on medium and further leads to reduction of the overall cost of production.Offprint requests to: B. K. Lonsane 相似文献

Bacillus sp. DT7 produced very high levels of alkaline and thermotolerant pectinase by solid state fermentation. Production of this enzyme was affected by nature of solid substrate, level of moisture content, presence or absence of carbon, nitrogen, mineral and vitamin supplements. Maximum enzyme production of 8050 U/g dry substrate was obtained in wheat bran supplemented with polygalacturonic acid (PGA; 1%, w/v) and neurobion (a multivitamin additive; 27 micro l/g dry substrate) with distilled water at 75% moisture level, after 36 h of incubation at 37 degrees C. 相似文献

A solid state fermentation (SSF) method was used to produce citric acid by Aspergillus niger DS 1 using sugarcane bagasse as a carrier and sucrose or molasses based medium as a moistening agent. Initially bagasse and wheat bran were compared as carrier. Bagasse was the most suitable carrier, as it did not show agglomeration after moistening with medium, resulting in better heat and mass transfer during fermentation and higher product yield. Different parameters such as moisture content, particle size, sugar level and methanol concentration of the medium were optimised and 75% moisture level, 31.8 g sugar/100 g dry solid, 4% (v/w) methanol and particles of the size between 1.2 and 1.6 mm were found to be optimal. Sucrose and clarified and non-clarified molasses medium were also tested as moistening agents for SSF and under optimised conditions, 20.2, 19.8 and 17.9 g citric acid /100 g of dry solid with yield of 69.6, 64.5 and 62.4% (based on sugar consumed) was obtained in sucrose, clarified and non-clarified molasses medium respectively, after 9 days of fermentation. 相似文献

Gibberellic acid from the fungi has been widely used in agriculture. In this study, more than 20 fungal isolates were screened and Paecilomyces sp. ZB shown to produce more gibberellic acid than other fungal isolates. Cow dung was used as low cost substrate for gibberellic acid production in solid state fermentation (SSF). Carbon, nitrogen and ionic sources stimulated gibberellic acid production in SSF. Lactose emerged as the significant carbon source supporting more gibberellic acid production (731 µg/g). Among the nitrogen sources, glycine appeared to influence the production of more gibberellic acid (803 µg/g). The process parameters were optimized to enhance gibberellic acid production using a two-level full factorial design and response surface methodology. The amount of gibberellic acid production was influenced mainly by moisture and pH of the substrate. Gibberellic acid production was 1312 µg/g under the optimized conditions and the predicted response was 1339 µg/g. The gibberellic acid yield increased twofolds after medium optimization. The extracted gibberellic acid was sprayed on the growing Mung bean plant and it stimulated the growth of the plant effectively. 研究了Rhizopus sp.PW358菌的固态生长和产脂肪酶条件。结果表明:黄豆饼粉为培养基的基本成分,用来生产脂肪酶。培养基中可加入淀粉和蛋白胨作为碳源和源,有利于脂肪酶的合成,培养基的含水量以及金属离子Ca^2 ,Mg^2 的浓度也影响Rhizopus sp.PW358菌和脂肪酶 产生。在优化条件下,12g豆粉中含1.0g淀粉及0.5g蛋白胨、15ml营养盐中Ca^2 ,Mg^2 离子浓度分别为8.0和4.0g/L,培养基含水量为55.6%,在接种后培养48h,酶活力可达最大值320IU/g干培养基。脂肪酶的基本性质研究表明,酶的最适反应温度和PH分别为35℃和7.0,酶的半失活温度为53.5℃,不同的PH环境中,30℃保温1h后酶在PH6.5-8.5范围内较为稳定。 相似文献

Several varieties of heat-sterilized broiler litter with 60% (wet basis, wb) moisture content were substrate in solid-state fermentations to produce biocontrol agents. Litter varieties included litter produced by one flock of broilers from medicated and non-medicated controlled rations, and litter produced by two flocks and four flocks on a single application of bedding material from medicated commercial sources. Litter preparations were inoculated with monocultures of Bacillus thuringiensis serovar japonensis strain Buibui, a pathogen of Japanese beetle larvae (Popillia japonica), or Pseudomonas fluorescens 2-79. B. thuringiensis did not grow in unextracted 1-flock litter nor in water extracted litter, but grew in methanol extracted litter to 5 x 10(10) cell forming units (CFU)/g litter (dry weight, dw) and a spore count of 1 x 10(10) CFU/g litter (dw). B. thuringiensis also grew in unprocessed 2-flock and 4-flock litter, achieving cell counts of 3 x 10(9) and 1 x 10(9) CFU/g litter (dw), respectively, and spore counts of 1 x 10(9) CFU/g litter (dw). P. fluorescens grew in medicated 1-flock litter with no extraction to a cell density greater than 4 x 10(11) CFU/g litter (dw). Bioassays in soil containing over 0.5% (db) litter fermented with B. thuringiensis resulted in over 90% mortality in 21 days for first instars of Japanese beetle when compared to a control treatment using compost without fermented litter. The investigations demonstrate that bacterial biocontrol agents produced via solid substrate fermentations using broiler poultry litter have potential in biocontrol applications in the soil environment. 相似文献

Xylanase production by a newly isolated strain of Burkholderia sp. was studied under solid state fermentation using anaerobically treated distillery spent wash. Response surface methodology (RSM) involving Box-Behnken design was employed for optimizing xylanase production. The interactions between distillery effluent concentration, initial pH, moisture ratio and inoculum size were investigated and modeled. Under optimized conditions, xylanase production was found to be in the range of 5200-5600 U/g. The partially purified enzyme recovered after ammonium sulphate fractionation showed maximum activity at 50 degrees C and pH 8.6. Kinetic parameters like Km and Vmax for xylan were found to be 12.75 mg/ml and 165 micromol/mg/min. In the presence of metal ions such as Ca2+, Co2+, Mn2+, Ba2+, Mg2+ and protein disulphide reducing agents such as beta-mercaptoethanol and dithiotheritol (DTT) the activity of enzyme increased, where as strong inhibition of enzyme activity was observed in the presence of Cu2+, Ag+, Fe2+ and SDS. The effect of the partial pressure of O(2) and CO(2) on the acid protease production in solid state fermentation by Aspergillus niger on wheat bran was studied. A fermentation system was used, which allowed on-line reactor measurements and continuous data acquisition of pH, temperature, gas flow, pressure drop and CO(2) production. Six paired combinations of CO(2) and O(2) concentrations were studied. The results showed a direct relationship between pressure drop, production of CO(2) and temperature increase. The pH evolution patterns were similar in all cases but different if the measurements were made on-line or on a liquid homogenate of the fermented substrate. Acid protease production was increased when the gas had 4% CO(2), (vol/vol), and it reached its highest level, a 43% increase over air, with a mixture of 4% CO(2) and 21% O(2). The protease production was strongly related to the mold metabolic activity as represented by the total CO(2) evolved. 相似文献

Mycophenolic acid (MPA) can be produced in solid state fermentation. An isolate of Penicillium brevi-compactum ATCC 16024 grown on moist wheat bran produced a titre of 425 mg per kg of wheat bran. Central composite rotatable design
and response surface methodology were employed to derive a statistical model for media optimization towards production of
mycophenolic acid. Five levels with a five factorial design were adopted. The correlation coefficient was 0.82, ensuring a
satisfactory adjustment of the model to the experimental values. This statistical design was very effective in improving the
titre of mycophenolic acid up to 3286 mg per kg of wheat bran.

Rice chaff was used as the substrate for the production of fibrinolytic enzyme by Fusarium oxysporum in solid state fermentation.
The optimized moisture content of the medium was 30 - 50% (v/w). An inoculum of 5 - 10% (v/v) and an average particle size
of 400 μm for the substrate were optimum for productivity and enzyme activity of 80 IU per ml filtrate was achieved after
96 hours of fermentation.
