禽流感病毒分型基因芯片的研制   总被引:11,自引:0,他引:11  
[目的]禽流感病毒是一种全球重要的人和动物呼吸道病病原,快速确定其不同亚型对于全球流感监测具有重要的意义.本研究意在研制一种可同时鉴定禽流感病毒所有亚型的方法.[方法]根据GenBank上已发表的禽流感病毒不同亚型(16个HA亚型和9个NA亚型)的基因序列,设计合成了25对特异性引物和1对通用引物,然后以各亚型病毒的参考株RNA作为模板,建立扩增不同亚型的多重RT-PCR方法.参考各亚型病毒靶cDNAs区域的保守序列设计了52条亚型特异的探针,进而利用扩增的各亚型病毒的靶cDNAs对其特异性进行评价.在此基础上,将设计好的探针点制到处理好的玻片上,制备了禽流感病毒分型鉴定基因芯片,结合所建立的扩增不同亚型的多重RT-PCR方法,开发了禽流感病毒亚型鉴定基因芯片试剂.利用收集自49个地区的2653份标本对其特异性和敏感性进行了初步评价.[结果]用于评价的各亚型参考毒株均出现良好的特异性杂交信号,检测的敏感度可达2.47 PFU/mL或2.5 ng靶DNA片段,而且与禽类常见的IBV、NDV等6种病毒均无交叉反应.[结论]证明该病毒分型基因芯片具有良好的特异性、敏感性.  相似文献

目的检测全省六个实验兔场的兔子所携带的兔出血症病毒(RHDV)情况,调查实验兔RHDV抗体水平,评价不同疫苗的免疫效果,比较HAI与ELISA两种方法的符合率。方法采用HAI、ELISA方法对1168份实验兔RHDV抗体进行了检测,并与RT-PCR方法的检测结果进行对比分析。结果我省实验兔免疫情况较好,不同饲养场的实验兔免疫合格率虽有不同,但未发生疫情。通过比较发现ELISA法检测的抗体合格率明显高于HAI法。结论LISA、HAI和RT-PCR方法均适合实验兔RHDV的检测。  相似文献

[目的]信号淋巴激活分子(SLAM又称CD150)为犬瘟热病毒(Canine distemper virus,CDV)的细胞受体.本研究目的在于建立稳定表达犬瘟热易感动物貉SLAM基因的Vero细胞系,用于犬瘟热诊断及CDV强毒株的快速分离.[方法]从重组质粒pMD-18-T-rSLAM扩增rSLAM基因编码区,通过分子克隆技术构建真核表达质粒pIRES2-EGFP-rSLAMhis.将该质粒转染Vero细胞后,经EGFP荧光观察、G418抗性压力和单细胞克隆化及RT-PCR筛选表达rSLAM阳性细胞系.应用获得细胞系对临床犬瘟热病例进行病毒分离,对分离得到CDV进行RT-PCR鉴定及动物回归试验.[结果]经过RT-PCR和免疫组化试验证实,本实验筛选获得一株能稳定表达rSLAM的Vero细胞系——Vero-rSLAM.该细胞系接种CDV阳性样品36-48h即可产生典型CDV致细胞融合病变,而其亲本Vero细胞接种至6d均无可见细胞病变出现.应用Vero-rSLAM细胞系从狐狸、貉犬瘟热阳性病料中分离获得了3株犬瘟热病毒.其中犬瘟热病毒LN(10) f1株对易感狐狸、貉均可产生致死性感染.[结论]成功建立了稳定表达rSLAM的Vero细胞系,用它分离的CDV对本动物保持较强的毒力.  相似文献

[目的]建立狂犬病毒的反向遗传系统,为研制不含狂犬病病毒致病性的新型安全高效的狂犬疫苗提供技术依据.[方法]本研究采用反向遗传学方法和分子克隆技术,建立了狂犬病病毒Evelyn-R0kitnieki-Abelseth(ERA)疫苗株的cMV/T7、T7启动子病毒拯救系统,构建了表达N、P、L蛋白的辅助质粒.[结果]成功拯救出野生型病毒rERA-VC,在Vero细胞上的生长动力学特性与父母本ERA相同,第三代可在Vero细胞上可获得很高的生长滴度.[结论]建立了狂犬病病毒的反向遗传系统,拯救出的野生型病毒生物学特性与父母本相同.  相似文献

[目的]探讨登革病毒(dengue virus,DEN)3′非编码区(untranslated region,UTR)RNA元件(VR、RCS2、CS2、CS1和SL)对基因组翻译的影响.[方法]首先构建登革2′型病毒中国株(DEN2-43)UTR与萤火虫荧光素酶基因(LUC)组成的病毒诱导报告基因(virus induced reportergene,VIRG),在此基础上分别构建包含DEN2-43 3′UTR不同RNA元件的VIRG,并通过LUC检测、实时RT-PCR和Western blot方法分析含有不同元件VIRG对翻译效率的影响.[结果]发现完整的3′UTR缺失可显著抑制翻译,含有病毒VR元件VIRG的翻译水平与完整3′UTR的VIRG相似,RCS2或CS2元件可提高VIRG的翻译效率,CS1或SL元件可降低VIRG的翻译效率.[结论]DEN 2-43病毒基因组3′UTR参与了报告基因的翻译,其不同元件具有可上调和下调报告基因翻译效率的作用.  相似文献

[目的]对从酸马奶中分离出来的10株乳酸菌进行鉴定和生理生化特性研究,为工业生产筛选特性优良的菌种.[方法]通过形态学观察、生理生化特性、分子生物学特性及其对致病菌抑制作用的研究对其进行鉴定,并筛选特性优良菌株.[结果]10株乳酸菌分别为2株Lactobacillus plantarum、2株Enterococcus villorum、2株Enterococcus dispar、3株Enterococcus durans和1株Enterococcus raffinosus;其对Staphylococcus aureus、Escherichia coli和Enteritidis bacillus有不同程度的抑制作用.[结论]菌株HZ24、HZ25具有良好的生物学特性和益生功能,可以应用到食品发酵工业生产中.  相似文献

极其多样的病毒广泛存在于我们周围的环境和动物体内,其中很多病毒是人类所未知的,而发现未知病毒常常受制于病毒常规检测技术的局限性.[目的]构建未知病毒检测技术平台.[方法]应用病毒宏基因组学的理念,结合新型分子诊断技术,首先利用过滤和核酸酶处理去除样品宿主核酸干扰,然后随机PCR扩增潜在的病毒宏基因组,最后通过大规模测序及序列分析获取病毒核酸信息.[结果]利用此技术我们对猪瘟病毒(CSFV)细胞培养物和猪圆环病毒2型(PCV2)感染猪病料进行了分析,分别检测到序列总长度1680 bp,占基因组13.7%的CSFV序列和序列总长度834 bp,占基因组47.2%的PCV2序列;利用此检测技术平台研究一未知病原细胞培养物,通过测序和序列分析,结果显示56条序列中有26条为副流感病毒5型(PIV5)同源序列,覆盖了其基因组全长的16.4%;此外,应用本研究建立的方法结合新一代高通量测序,我们在混合的7份病原未知的病猪组织样品中检测到了1.1%的病毒序列,包括CSFV、PCV2、猪细环病毒(TTSuV)、猪bocavirus (PBoV)和人腺病毒6型(Ad6)等的部分基因序列.[结论]本研究建立的基于病毒宏基因组学的未知病毒的检测方法突破了传统病毒研究方法的缺陷,对于猪群样本中病毒的检测具有较高的敏感性,有望为新发、突发感染性疾病的诊断和监测提供技术支持.  相似文献

[目的]为了探讨高致病性禽流感病毒对水禽致病性差异的分子致病机理.[方法]我们对从野鸭分离到的H5N1亚型禽流感病毒的生物学特性进行鉴定,其中A/mallard/Huadong/Y/2003(Y)是对麻鸭无致病性病毒,而 A/mallard/Huadong/S/2005(S)是对麻鸭高致病性病毒.利用反向遗传技术构建一系列单个和多个基因组合替换基因重排病毒,并验证重排病毒在麻鸭上的致病力.[结果]研究表明,PB2, PB1, PA(3P), HA单基因以及3P基因组合替换的使S病毒对麻鸭的毒力完全致弱,但相应的基因替换后仅使Y病毒对麻鸭的毒力略有上升.两病毒的其它基因对毒力影响较小.[结论]H5N1亚型禽流感病毒对麻鸭的致病力受多基因调控,且这种调控作用在不同病毒骨架上的影响不一致,强毒受影响程度远比弱毒的大.  相似文献

为探索四溴甘脲消毒剂杀灭细菌的机理,采用透射电镜技术对四溴甘脲消毒剂处理过的枯草杆菌黑色变种芽胞的超微结构进行了分析和比较.结果显示,以含有效溴274mg/L的四溴甘脲消毒剂作用30min,可使枯草杆菌黑色变种芽胞杀灭率达到100%.在透射电镜下观察到,经该消毒剂作用的枯草杆菌黑色变种芽胞壳质破损断裂明显,壳内结构模糊,核心溶解,有的芽胞近似空壳.结果显示,四溴甘脲消毒剂杀灭芽胞效果优于普通含氯消毒剂,对细菌芽胞超微结构破坏明显.  相似文献

目的:研究多药耐药大肠杆菌对不同消毒剂的抗力水平,为临床上合理选择消毒剂提供依据。方法:采用营养肉汤稀释法及悬液定量杀菌实验分别测定临床分离出的具有多重耐药性的大肠杆菌对临床常用消毒剂的最低抑茵浓度(minimum inhibitory concentration,MIC)及不同作用时间消毒剂对菌株的杀灭率,并与大肠杆菌标准株的测定结果进行比较。结果:不同消毒剂对多药耐药大肠杆菌的MIC值不同,洁芙柔消毒凝胶的MIC值为2.8±1.9,绿莎新爱尔施牌消毒片的MIC值为2583.4±1107.2,爱护佳免洗手消毒液的MIC值为144.63:86.9,安尔碘II型皮肤消毒剂的MIC值为16.1±7.7。4种常用消毒剂对多要耐药大肠杆菌的MIC值均高于标准菌,绿莎新爱尔施牌消毒片及爱护佳免洗手消毒液的MIC值与标准大肠杆菌较为接近。另外,4种常用消毒剂在常用浓度下对标准大肠杆菌和多药耐药大肠杆菌作用5min的杀灭率均能打到100%,标准大肠杆菌的被杀灭率略高于多药耐药大肠杆菌。结论:多药耐药菌株对消毒剂的抗性日益受到关注,使用时调整浓度、加强消毒剂科学管理对控制医院感染具有重要意义。  相似文献

In this study, efficacy of seven different commercial disinfectant preparations was investigated against characteristic bacteria of a poultry slaughterhouse in Ankara (Turkey) by using paper disc-agar diffusion method and surface effectiveness test. According to paper disc-agar diffusion method, some disinfectants had wide efficacy against the test bacteria. The disinfectant effectiveness generally increased by the increasing of disinfectant concentration. However, some disinfectants were ineffective even though at their highest concentrations. The most effective disinfectant was B which contains QAC as active agent.Staphylococcus spp.,Staphyloccus aureus andEscherichia coli were the most sensitive bacteria to the disinfectants. Some pathogenic isolates, especiallySalmonella spp. andCampylobacter spp., were the most resistant ones to many of the disinfectants tested. The results of paper disc-agar diffusion method indicated the importance of characteristic bacterial strains of food plants as the test bacteria for disinfectant efficacy tests. Conversely of paper disc-agar diffusion method, all disinfectants were effective against the isolates by surface effectiveness test depending on exposure time. The disinfectants, except A and F, produced at least 3 log unit reduction during 5 min of exposure. However, all disinfectants at their lowest concentrations were effective against all tested bacteria during 15 and 30 min by this test.  相似文献

A number of chemical disinfectants were evaluated for their bactericidal and virucidal effectiveness in holding-tank sewage. It was found that the disinfection efficiencies of formaldehyde, benzalkonium chloride, cetylpyridinium chloride, and methylene blue were markedly improved if the pH of the sewage was raised from 8.0 to 10.5. When formaldehyde, benzalkonium chloride, and methylene blue were tested with either 2-week holding times with no sewage additions or 10-day holding times with daily sewage additions, disinfection effectiveness was maintained as long as the sewage pH was kept at 10.5 and the disinfectant concentration was kept at 100 mg/liter or more. Calcium hypochlorite, zinc sulfate, and phenol were found to be relatively ineffective disinfectants for holding-tank sewage.  相似文献

Chytridiomycosis, a disease contributing to amphibian declines worldwide, is caused by the fungus Batrachochytrium dendrobatidis. Identifying efficient and practical disinfectants effective against B. dendrobatidis is important to reduce the spread of the disease both in the wild and captivity. Previous studies identified a range of suitable disinfectant strategies. We evaluated the suitability of 3 additional disinfectants: two of these (TriGene Virucidal Disinfectant Cleaner and F10 Super Concentrate Disinfectant) are mixtures of chemicals and one (Betadine Antiseptic Liquid) contains a single active ingredient, povidone iodine. The disinfectants were tested using a range of concentrations for 1,5 and 10 min to determine their ability to kill B. dendrobatidis in vitro. The measure of effectiveness was 100% kill of zoosporangia grown in multiwell plates. All disinfectants had a 100% efficacy at concentrations recommended by the manufacturers. The lowest concentrations capable of 100% kill after exposure for 1 min were 0.1 ml l(-1) for TriGene, 0.33 ml l(-1) for F10 and 100 ml l(-1) for Betadine. TriGene is the most effective disinfectant yet to be found, and both TriGene and F10 are more effective than various disinfectants tested in previous studies. TriGene and F10 are considered suitable for use in the field, as only small amounts of concentrate are needed.  相似文献

Minute virus of mice (MVM), a single-stranded deoxyribonucleic acid Parvovirus, was subjected to various inactivation procedures, including chemical disinfectants, heat, and ultraviolet radiation. MVM was found to be less stable than has been reported for other Parvoviruses. This virus was readily inactivated by a variety of chemical disinfectants, including alcohols, formaldehyde, glutaraldehyde, and chloroform. MVM was more sensitive to ultraviolet radiation than was Kilham's rat virus. MVM was more sensitive to heating at temperatures of 35 to 100 C than has been reported for other Parvoviruses. More than 95% of MVM infectivity was inactivated by heating (45, 60, or 100 C) for 60 min, acid (pH 2.0) treatment, or ultraviolet radiation treatment, although a small percentage (less than 2%) of the virus preparation was found to be resistant to these treatments. In addition, more than 99% of the infectivity of MVM was lost after storage at 4C for 10 weeks, although the virus was stable on storage in liquid nitrogen.  相似文献

消毒剂是一种可杀灭物体表面、器材设备、皮肤、空气和水源等传播媒介上携带的病原微生物的有机分子。它在体外能杀灭病原微生物,切断其传播途径,进而达到控制污染的目的,在生命安全防控中起着重要的作用。但是不合理地使用消毒剂导致细菌对消毒剂产生耐药。消毒剂耐药基因在不同种属间的水平转移加剧其传播风险,使消毒剂耐药情况进一步恶化。更令人担忧的是,细菌对消毒剂的耐药可能会导致对抗生素产生共耐药,给公共安全带来巨大的威胁。但目前为止,对消毒剂耐药以及共耐药的认识还不够全面。本文总结了关于细菌对消毒剂耐药的研究报道,对消毒剂的作用机制、细菌对消毒剂的耐药机制进行了论述,另外针对消毒剂耐药基因的传播以及细菌对消毒剂和抗生素的共耐药进行了综述,为减少消毒剂耐药性的产生和制定合理的消毒剂使用规范奠定基础。  相似文献

Touching of contaminated objects and surfaces is a well-known method of virus transmission. Once they are attached to the hands, viruses can easily get adsorbed and initiate infection. Hence, disinfection of frequently touched surfaces is of major importance to prevent virus spreading. Here we studied the antiviral activity of a glucoprotamin-containing disinfectant against influenza A virus and the model virus vaccinia virus (VACV) dried on inanimate surfaces. The efficacy of the surface disinfectant on stainless steel, polyvinyl chloride, and glass coupons was investigated in a quantitative carrier test. Vacuum-dried viruses were exposed to 0.25%, 0.5%, and 1% disinfectant for 5 min, 15 min, and 30 min without agitation, and residual infectivity was determined by endpoint titration. Although glucoprotamin was highly active against both viruses in suspension, limited antiviral activity against the surface-dried viruses was detected. Even after 30 min of exposure to 1% disinfectant, VACV was not completely inactivated. Furthermore, influenza A virus inactivation was strongly affected by the surface composition during the 5-min and 15-min treatments with 0.25% and 0.5% disinfectant. The results presented in this study highlight the relevance of practical tests to assess the antiviral activity of surface disinfectants. High virucidal activity in solution is not necessarily indicative of high antiviral activity against surface-dried viruses. In addition, we want to emphasize that the mere exposure of surfaces to disinfectants might not be sufficient for virus inactivation and mechanical action should be applied to bring attached viruses into contact with virucidal compounds.  相似文献

The efficacy of a number of disinfection treatments was tested on in vitro cultures of the fungus Batrachochytrium dendrobatidis, the causative agent of chytridiomycosis in amphibians. The aim was to evaluate the fungicidal effects of chemical disinfectants, sterilising ultraviolet (UV) light, heat and desiccation, using methods that were feasible for either disinfection in the field, in amphibian husbandry or in the laboratory. The chemical disinfectants tested were: sodium chloride, household bleach (active ingredient: sodium hypochlorite), potassium permanganate, formaldehyde solution, Path-X agricultural disinfectant (active ingredient: didecyl dimethyl ammonium chloride, DDAC), quaternary ammonium compound 128 (DDAC), Dithane, Virkon, ethanol and benzalkonium chloride. In 2 series of experiments using separate isolates of B. dendrobatidis, the fungicidal effect was evaluated for various time periods and at a range of chemical concentrations. The end point measured was death of 100% of zoospores and zoosporangia. Nearly all chemical disinfectants resulted in 100%, mortality for at least one of the concentrations tested. However, concentration and time of exposure was critical for most chemicals. Exposure to 70% ethanol, 1 mg Virkon ml(-1) or 1 mg benzalkonium chloride ml(-1) resulted in death of all zoosporangia after 20 s. The most effective products for field use were Path-X and the quaternary ammonium compound 128, which can be used at dilutions containing low levels (e.g. 0.012 or 0.008%, respectively) of the active compound didecyl dimethyl ammonium chloride. Bleach, containing the active ingredient sodium hypochlorite, was effective at concentrations of 1% sodium hypochlorite and above. Cultures did not survive complete drying, which occurred after <3 h at room temperature. B. dendrobatidis was sensitive to heating, and within 4 h at 37 degrees C, 30 min at 47 degrees C and 5 min at 60 degrees C, 100% mortality occurred. UV light (at 1000 mW m(-2) with a wavelength of 254 nm) was ineffective at killing B. dendrobatidis in culture.  相似文献
