 A polygalacturonase (PG) gene, TPG7 (Lyces;Pga1;8), has been cloned from tomato (Lycopersicon esculentum Mill., cv. Rutgers). RNA blot analysis reveals that TPG7 is highly expressed in pistils (ovary removed) from unopened and fully open flowers. Dissection of mature pistils demonstrated that TPG7 expression is limited to the top third (stigmatic region) of the pistils. This is contrasted with another tomato PG, TAPG4, which is also expressed in the same region of the pistil but only in mature pistils from fully open flowers. Hybridization of the TPG7 probe to anther RNA was nil to none and was barely detectable in RNA from leaf and flower abscission zones. The TPG7 polypeptide shares 39% sequence identity with the tomato fruit PG and between 63% and 73% sequence identities with six other tomato PGs. Successful sexual reproduction relies on gene products delivered by the pistil to create an environment suitable for pollen tube growth. These compounds are either produced before pollination or formed during the interactions between pistil and pollen tubes. Here we describe the pollination-enhanced expression of the cp100 gene in pistils of Solanum tuberosum. Temporal analysis of gene expression revealed an enhanced expression already one hour after pollination and lasts more than 72 h. Increase in expression also occurred after touching the stigma and was not restricted to the site of touch but spread into the style. The predicted CP100 protein shows similarity to leguminous isoflavone reductases (IFRs), but belongs to a family of IFR-like NAD(P)H-dependent oxidoreductases present in various plant species.  相似文献

The progamic phase, the period of pollen tube growth through the pistil, is a period of specific interactions between the male gametophyte and the pistil. Understanding of pollen germination and pollen tube growth are relevant for the study of pollen-pistil interactions and for understanding the function of components specifically accumulated in the transmitting tissue cell walls and intercellular matrix that may interact with pollen tubes. Ubiquitin-conjugated compounds were localized in anthers and pistils of Nicotiana alata by immuno-cytochemistry. In young anthers, antibodies to ubiquitin bound to callose cell walls surrounding pollen mother cells and to organelles in the endothecium. At the freespore stage, antibodies bound to circular-cell cluster cells subtending the stomium and to organelles and cell walls of endothecial cells. Near anther dehiscence, locular material was labeled. In pistils, cell walls of stylar transmitting tissue were labeled in a beaded pattern. Antibodies bound to a thin layer surrounding ovules, to the lining of embryo sacs, to cytoplasm of eggs and synergids, and to starch grains in central cells. Sites of localization were tissue- and time-specific, suggesting a regulatory role for ubiquitin in development of reproductive structures in flowering plants.  相似文献

 This study examines sporophytic and gametophytic vigor in wild and cultivated varieties of Cucurbita pepo L. and their hybrids in order to determine whether hybrid vigor extends to the microgametophyte generation. It also examines the variation in sporophytic and gametophytic vigor to discern the non-genetic influences of pollen provisioning by the sporophyte on pollen performance from the genetic influences of the microgametophyte’s own genotype on pollen performance. A cultivated and a wild C. pepo and their F1 and the F2 generations were grown under field conditions and flower and fruit production were monitored over one summer. We found that the four types of plants differed significantly in the number of male and female flowers and the number of fruits they produced. The F1 plants produced significantly more male flowers and marginally more female flowers and fruits than the parental lines. To estimate gametophytic vigor pollen was germinated in vitro and pollen tube length measured after 30 min. We found that pollen tubes from the F1 plants had significantly greater growth than tubes from the parental lines or the F2 generation, indicating that hybrid vigor extends to the microgametophytic generation. By partitioning the variance of pollen tube growth into ’within’ and ’among’ plant components of variation, we were able to show that the genotype of the microgametophyte influences pollen performance in vitro, but that expression of hybrid vigor in the microgametophyte is likely to be due to an environmental effect related to provisioning of the pollen grains during development. Summary Pollination and pollen tube growth was observed in pistils of fertile and female-sterile plants ofOenothera mut.brevistylis. The stigmas and styles of sterile plants were very irregularly formed and additional ovules often developed in the style. The malformations did not inhibit pollen tube growth but made it slower and less efficient. The pollen tubes grew in the ovary but did not find the ovules. Near their tips, pollen tubes branched or meandered without any clear direction. Lack of tropism caused by malfunction of the ovules is postulated in the female-sterile plant.  相似文献

 Somatic hybrids between three dihaploid Solanum tuberosum (2n=2x=24) genotypes and the frost-tolerant, diploid, relative Solanum commersonii (2n=2x=24) were analysed for variation in morphological traits, fertility and frost tolerance. The somatic hybrids were more vigorous than their parents and in many ways resembled a tetraploid S. tuberosum. All of the hybrids flowered profusely, although the male fertility was largely dependent on the S. tuberosum genotype used. In one hybrid combination all plants were both male- and female-fertile, while in the other two combinations the majority of plants were male-sterile but female-fertile. The somatic hybrids showed an increase in direct frost tolerance when compared with the dihaploid S. tuberosum parents, and to a varying extent had gained the capacity to cold acclimate. Based on the assumptions that pollen tube length is predetermined by provisions in the pollen and that it is a function of pistil length, I hypothesise that species with longer pistils will have larger pollen grains than species with shorter pistils, and that pistil length and pollen size will be positively correlated in a linear manner. To test this hypothesis, the relationship between pollen grain volume and pistil length was compared in 43 Asteraceae species from Argentina. A positive linear correlation was found between pollen volume and pistil length. This correlation remained significant even after potential effects of phylogenetic relatedness were removed. The maintenance of this correlation suggests that in Asteraceae the association between pistil length and pollen volume may reflect a functional rather than a phyletic relationship. In addition, the pistil length: pollen volume ratio (PPR) was analysed in relation to the phylogenetic position of the species. High values of PPR would imply a reduction of the male gametophyte in relation to the minimal volume that a pollen grain must have to grow and fertilise an ovule. Thus, the general pattern of pollen volume reduction in relation to pistil length previously found among many angiosperm families will be also present within a family, i.e., PPR values of derived Asteraceae would be higher than those of basal species. Results indicated that reduction of pollen volume in derived Asteraceae was three times greater than the concomitant shortening of pistil length. Consequently, PPR increased with the phylogenetic position of the taxa. This work supports the correlation between pistil and pollen characters previously found for other plant families and confirms the influence of post-pollination processes on pollen size evolution.  Crossing experiments were conducted to introduce resistance to the root-knot nematodes, Meloidogyne chitwoodi and M. fallax, from various polyploid Central American Solanum spp. into the cultivated potato, S. tuberosum ssp. tuberosum. The most effort was put into producing tetraploid hybrids through inter-EBN (Endosperm Balance Number) crosses. From the crosses of tetraploid S. tuberosum (4 EBN) with tetraploid S. stoloniferum and S. fendleri (both 2 EBN), few seeds were derived that led to viable plants. In vitro culture of immature seeds also yielded several hybrid plants. From crosses of diploid S. tuberosum (2 EBN) with hexaploid S. hougasii (4 EBN) four hybrids were obtained through in vitro culture. Backcrosses were made with selected hybrids and a variable number of seeds was produced depending on the hybrid genotype. The successful introgression of resistance into backcross populations is shown. A scheme is presented for the introgression of traits at a tetraploid level from allotetraploid Solanum species into autotetraploid S. tuberosum through sexual crosses.  Inbreeding depression may affect the performance of consecutive generations of potatoes propagated by true potato seed (TPS). The effect of inbreeding was established using selfed and sib-mated generations of five TPS families. Correlation coefficients were calculated between the level of inbreeding and different traits. Inbreeding depression was expressed mainly by pollen viability, as measured by its stainability (r=−0.912, P<0.01), and tuber yield (r=−0.837, 0.01<P<0.05). The results also indicated that without unavoidable selection inbreeding depression is expected to be more evident. Furthermore, the TPS families responded quite differently to inbreeding depression. They did not show the same amount of depression for yield as they did for the characters concerning fertility.  In 1992, 72 seedlings from each of 198 pair crosses were grown in a glasshouse, and the tubers produced by each plant were visually assessed on a 1–9 scale of increasing preference. Three groups of four progenies with high, medium and low mean scores were chosen to progress, without selection via tuber progenies and four-plant plots at a high-grade seed site, to replicated yield trials in the third clonal generation. The three groups maintained their high, medium and low scores for visual preference over the three clonal generations and also had high, medium and low scores in the second and third clonal generations for yield, size and appearance of tubers, all of which were components of visual preference. The three groups were predicted to have 13.6%, 1.8% and 0.2% of their clones exceeding the mean of 13 control cultivars for visual preference in the replicated trials, and 12.1%, 4.9% and 1.4% for yield, and 56.8%, 37.1% and 14.8% for appearance. The experiment confirmed that selection for visual preference within crosses in the seedling and first clonal generations is very ineffective, but that worthwhile progress can be made from selection in the second clonal generation, with correlated responses for faster emergence, earlier maturity, higher yield and greater regularity of shape (appearance). Combining selection of the high group of progenies with selection in the second clonal generation of the best 34 out of the 120 clones in this group, produced a response in visual preference in the third clonal generation of 1.00 compared with a maximum possible of 1.74. In Drosophila melanogaster, alcohol dehydrogenase (ADH) activity is essential for ethanol tolerance, but its role may not be restricted to alcohol metabolism alone. Here we describe ADH activity and Adh expression level upon selection for increased alcohol tolerance in different life-stages of D. melanogaster lines with two distinct Adh genotypes: Adh(FF) and Adh(SS). We demonstrate a positive within genotype response for increased alcohol tolerance. Life-stage dependent selection was observed in larvae only. A slight constitutive increase in adult ADH activity for all selection regimes and genotypes was observed, that was not paralleled by Adh expression. Larval Adh expression showed a constitutive increase, that was not reflected in ADH activity. Upon exposure to environmental ethanol, sex, selection regime life stage and genotype appear to have differential effects. Increased ADH activity accompanies increased ethanol tolerance in D. melanogaster but this increase is not paralleled by expression of the Adh gene.  相似文献

 Pollen tube growth in the pistil and pollen tube penetration of ovules have both been studied in crosses between cultivars from Tulipa gesneriana L. and 12 tulip species from all eight sections of the genus Tulipa to identify pre-fertilization barriers. Depending on the cross, pollen tubes grew as far as the stigma or the style or continued growing down into the ovary. Pollen tubes penetrated none or only a few percent of the ovules of some crosses, despite the presence of many pollen tubes in the ovary. In other crosses, from which no or only a few hybrids have been obtained after seed maturation on the plant, pollen tube penetration was found in up to 79% of the ovules. A breeding scheme involving ploidy and EBN manipulations was set up to overcome the interspecific barriers existing between the cultivated Solanum tuberosum and the wild species S. commersonii. Three backcross generations were obtained that were analyzed by means of molecular markers in order to verify the occurrence of recombination between homeologous chromosomes, the extent of the wild genome carried in each backcross, and the efficiency of introgressing useful genes. Twenty commersonii-specific RFLPs provided evidence for recombination on 5 out of 12 chromoso-mes; for the others no more than 1 commersonii-specific marker was found. Thirty-four commersonii-specific RAPDs and 61 commersonii-specific AFLPs were used to estimate the wild genome content in the BC1, BC2, and BC3 generations. The mean value of commersonii-specific markers was 93% and 91% in the BC1, 74% and 76% in the BC2, and 31% and 26% in the BC3 using RAPDs or AFLPs, respectively. Efficiency of the breeding scheme was evaluated by monitoring across these progenies the introgression of S. commersonii resistance to tuber soft rot caused by Erwinia carotovora. Eleven and five resistant genotypes were found among BC2 and BC3 hybrids, respectively. The same progenies were also evaluated for chromosome number and tuber traits. For all analyzed traits except stolon length, all BC2 and BC3 hybrids resembled the tuberosum type. In order to choose the best genotypes to obtain the following backcrosses, we performed, in each generation, a negative molecular-assisted selection against the wild genome combined with selection for resistance to Erwinia spp. and other traits. In angiosperms, the female gametophyte has a secluded life; it is protected by several concentric layers that envelop each other. The embryo sac is surrounded by the nucellus, which in turn is wrapped by the integuments forming the ovule, which is nested in the ovary. These wrappings are not hermetic, but contain little ”gates” the pollen tube must traverse on its way towards the embryo sac. Information is emerging that shows that the ovary and ovule provide signals orienting and directimng the pollen tube on the right course. There are three main bodies of evidence supporting this hypothesis. One relates to developmental changes in the female tissues and how they affect pollen tube growth. The second refers to defective ovule mutants, which induce defective pollen tube guidance. And the third relates to the possible molecules involved in this signalling. Here, information gathered along these three main lines of evidence is reviewed. All converge to the conclusion that different checkpoints exist all along the pollen tube pathway. A coherent picture of pollen tube growth is beginning to emerge that couples ion dynamics with biochemical, biophysical and cytological processes in ordered and controlled feedback circuits that define the nature of polarized apical growth. It is a paradox, however, that complete understanding of the mechanical forces that drive cell elongation in this system still remains to be fully achieved. The results of our recent studies to characterize Cl ion dynamics during apical growth in tobacco pollen tubes led us to re-examine this question in the light of a possible force-generating role provided by hydrodynamic flow. Previously we found that oscillatory Cl efflux from the apex is closely coupled to oscillatory growth and the cell volume of the apical domain. Cl influx occurs in a region of the tube that is distal to the clear zone; hence, a vectorial flow of anion traverses the apical domain and fluxes out of the tip with oscillatory dynamics. Because of the effects that this could induce on charge and osmotic potentials, water could potentially flow through the apical domain, linked to the flux of Cl. This conjecture is consistent with studies in other plant cells that demonstrate a pivotal role for flux through anion channels in the control or normalization of osmotic status. In the current report, the relationship between Cl efflux oscillations and the physical characteristics of the apical dome during oscillatory growth is examined in closer detail. Evidence is presented that shows a cyclic deformation of the extreme apex occurs during the growth pulse and is correlated with cyclic Cl efflux. In addition, there is a dramatic increase in the number and density of clear thread-like zones traversing the apical plasma membrane during the process of tip elongation. Possible functional roles of Cl flux and hydrodynamics are discussed in the context of what drives tip elongation during cycles of pollen tube growth. The alcohol dehydrogenase (ADH) family has evolved into at least eight ADH classes during vertebrate evolution. We have characterized three prevertebrate forms of the parent enzyme of this family, including one from an urochordate (Ciona intestinalis) and two from cephalochordates (Branchiostoma floridae and Branchiostoma lanceolatum). An evolutionary analysis of the family was performed gathering data from protein and gene structures, exon–intron distribution, and functional features through chordate lines. Our data strongly support that the ADH family expansion occurred 500 million years ago, after the cephalochordate/vertebrate split, probably in the gnathostome subphylum line of the vertebrates. Evolutionary rates differ between the ancestral, ADH3 (glutathione-dependent formaldehyde dehydrogenase), and the emerging forms, including the classical alcohol dehydrogenase, ADH1, which has an evolutionary rate 3.6-fold that of the ADH3 form. Phylogenetic analysis and chromosomal mapping of the vertebrate Adh gene cluster suggest that family expansion took place by tandem duplications, probably concurrent with the extensive isoform burst observed before the fish/tetrapode split, rather than through the large-scale genome duplications also postulated in early vertebrate evolution. The absence of multifunctionality in lower chordate ADHs and the structures compared argue in favor of the acquisition of new functions in vertebrate ADH classes. Finally, comparison between B. floridae and B. lanceolatum Adhs provides the first estimate for a cephalochordate speciation, 190 million years ago, probably concomitant with the beginning of the drifting of major land masses from the Pangea.
