Extracellular polymeric substances (EPS) of microbial origin are a complex mixture of biopolymers comprising polysaccharides, proteins, nucleic acids, uronic acids, humic substances, lipids, etc. Bacterial secretions, shedding of cell surface materials, cell lysates and adsorption of organic constituents from the environment result in EPS formation in a wide variety of free-living bacteria as well as microbial aggregates like biofilms, bioflocs and biogranules. Irrespective of origin, EPS may be loosely attached to the cell surface or bacteria may be embedded in EPS. Compositional variation exists amongst EPS extracted from pure bacterial cultures and heterogeneous microbial communities which are regulated by the organic and inorganic constituents of the microenvironment. Functionally, EPS aid in cell-to-cell aggregation, adhesion to substratum, formation of flocs, protection from dessication and resistance to harmful exogenous materials. In addition, exopolymers serve as biosorbing agents by accumulating nutrients from the surrounding environment and also play a crucial role in biosorption of heavy metals. Being polyanionic in nature, EPS forms complexes with metal cations resulting in metal immobilization within the exopolymeric matrix. These complexes generally result from electrostatic interactions between the metal ligands and negatively charged components of biopolymers. Moreover, enzymatic activities in EPS also assist detoxification of heavy metals by transformation and subsequent precipitation in the polymeric mass. Although the core mechanism for metal binding and / or transformation using microbial exopolymer remains identical, the existence and complexity of EPS from pure bacterial cultures, biofilms, biogranules and activated sludge systems differ significantly, which in turn affects the EPS-metal interactions. 高效、低耗、环境友好的重金属废水处理方法是当前的研究热点之一,微生物吸附法因具有优良的吸附性能、不产生二次污染、环境友好性等优点,在重金属废水处理中有巨大的应用潜力。细菌、真菌、藻类等微生物可通过静电吸附、络合作用等将重金属结合到细胞壁表面。但未经处理的微生物往往吸附效果不佳,通过对微生物进行物理、化学等方法的改性处理,能显著增加微生物与重金属离子结合的活性位点,提高去除效果。本文对国内外微生物的改性方法以及改性微生物吸附剂对废水中重金属的吸附能力和影响因素进行阐述,讨论微生物吸附剂存在的相关问题,并对其未来的研究方向做简要展望。  相似文献

Unlike organic pollutants, heavy metals cannot be degraded and can constitute a persistent environmental hazard. Here, we investigated the success of different remediation strategies in promoting microbial diversity and function with depth in an acidic soil heavily contaminated with Cu, Pb and Zn. Remediation involved the incorporation of either a high- or a low-quality compost or inorganic fertilizer into the topsoil and monitoring of microbial activity and diversity with soil depth over a 4-month period. While changes in topsoil microbial activity were expected, the possible effects on the subsurface microbial community due to the downward movement of metals, nutrients and/or soluble organic matter have not been examined previously. The results showed that both compost additions, especially the low-quality compost, resulted in significantly increased bacterial and fungal diversity (as assessed by terminal restriction fragment length polymorphism) and activity compared with the inorganic and control treatments in the topsoil. Although phospholipid fatty acid profiling indicated that compost addition had promoted enhanced microbial diversity in the subsoil, no concomitant increase in subsoil microbial activity was observed, suggesting that amelioration of the heavy metals remained localized in the topsoil. We conclude that although composts can successfully immobilize heavy metals and promote ecosystem diversity/function, surface incorporation had little remedial effect below the surface layer over the course of our short-term trial.  相似文献

Cell surface display of heavy metal-binding proteins has been used to enhance the adsorption capacity of heavy metals and the engineered microbial cells can be potentially used for the bioremediation of heavy metals. In this study, the proteins PbrR, PbrR691, and PbrD from the Cupriavidus metallidurans strain CH34 were displayed on the extracellular membrane of Escherichia coli BL21 cells, with the N-domain of ice-nucleation protein as the anchor protein to achieve specific adsorption of lead ions (Pb2+) and bioremediation of lead in the soil. The localization of fusion proteins was confirmed by western blot analysis. We investigated the effects of fusion pattern, expression level, heavy metal concentration, and the presence of other heavy metal ions on the adsorption of Pb2+ by these engineered bacteria, and the optimal linker peptide (flexible linker) and inducer concentration (0.5 mM) were obtained. The engineered bacteria showed specific selectivity and strong adsorption capacity for Pb2+. The maximum Pb2+ adsorption capacity of strains displaying the three proteins (PbrR, PbrR691, and PbrD) were 942.1-, 754.3-, and 864.8-μmol/g cell dry weight, respectively, which was the highest reported to date. The engineered E. coli bacteria were also applied to Pb2+-contaminated soil and the detoxification effects were observed via the seed germination test and the growth of Nicotiana benthamiana in comparison with the control BL21, which provides the proof-of-concept for in situ remediations of Pb2+-contaminated water or soil.  相似文献

Results obtained in studies of the biology of Pseudomonas plasmids are presented here as a mini-review. These data indicate that plasmids are ubiquitous in Pseudomonas, but the frequency of their occurrence varies greatly in particular species, or groups of species and in different microbial habitats. Some species of Pseudomonas, for instance P. aeruginosa, possess great diversity of plasmids both from the viewpoint of their incompatibility properties and their ability to endow bacteria with additional features such as resistance to antibiotics or heavy metals, degradation of xenobiotics or inhibition of phage development.  相似文献

In contrast to thermophilic or psychrophilic organisms, heavy metal-resistant bacteria do not supply enzymes that are active under harsh conditions, but are themselves tools for the evaluation and remediation of heavy metal-contaminated environments. Ralstonia sp. CH34 is a gram-negative bacterium with a remarkable set of resistance determinants, allowing this bacterium to live in extreme environments that are heavily contaminated with toxic metal ions. These heavy metal ions are mostly detoxified by inducible ion efflux systems that reduce the intracellular concentration of a given ion by active export. Because all metal resistance determinants in this bacterium are inducible, their regulatory systems can be used to develop biosensors that measure the biologically important concentrations of heavy metals in an environment. Resistance based on metal ion efflux detoxifies only the cytoplasm of the respective cell. Therefore, this resistance mechanism cannot be used directly to develop biotechnological procedures; however, metal ion efflux can protect a cell in a metal-contaminated environment. Thus, the cell can be enabled to mediate biochemical reactions such as precipitation of heavy metals with the carbon dioxide produced during growth or degradation of xenobiotics. ABC transporters from the multidrug resistance-associated protein (MRP) subfamily are glutathione S-conjugate pumps exhibiting a broad substrate specificity illustrated by numerous xenobiotics, such as anticancer drugs, herbicides, pesticides and heavy metals. The engineering of MRP transporters into plants might be interesting either to reduce the quantity of xenobiotics taken up by the plant in the context of “safe-food” strategies or, conversely, in the development of phytoremediation strategies in which xenobiotics are sequestered in the vacuolar compartment. In this report, we obtained Arabidopsis transgenic plants overexpressing human MRP1. In these plants, expression of MRP1 did not increase plant resistance to antimony salts (Sb(III)), a classical glutathione-conjugate substrate of MRP1. However, the transporter was fully translated in roots and shoots, and targeted to the plasma membrane. In order to investigate the functionality of MRP1 in Arabidopsis, mesophyll cell protoplasts (MCPs) were isolated from transgenic plants and transport activities were measured by using calcein or Sb(III) as substrates. Expression of MRP1 at the plasma membrane was correlated with an increase in the MCPs resistance to Sb(III) and a limitation of the metalloid content in the protoplasts due to an improvement in Sb(III) efflux. Moreover, Sb(III) transport was sensitive to classical inhibitors of the human MRP1, such as MK571 or glibenclamide. These results demonstrate that a human ABC transporter can be functionally introduced in Arabidopsis, which might be useful, with the help of stronger promoters, to reduce the accumulation of xenobiotics in plants, such as heavy metals from multi-contaminated soils.  相似文献

德兴铜矿尾矿重金属污染对土壤中微生物多样性的影响   总被引:12,自引:2,他引:10  
【目的】为更好地了解重金属污染与微生物多样性之间的相互作用关系,以江西德兴铜矿4#尾砂库为研究对象,采集野外实地样品共16件进行分析(包括尾砂样品以及周围农田和菜地土壤样品)。【方法】一方面对样品中可培养异养细菌进行平板计数,一方面采用变性梯度凝胶电泳(Denaturing gradient gel electrophoresis,DGGE)对样品中可培养和不可培养微生物分子生态多样性进行研究;同时采用PCA(Principle component analysis)方法分析样品理化性质、重金属及主要元素与可培养细菌数量及微生物多样性之间的相互关系。【结果】元素分析结果表明该尾矿区样品受到不同程度重金属Cu、Cd、Zn、Ni、Pb和Cr的污染;可培养异养细菌在尾砂样品中数量最少,在菜地和农田土壤样品中有明显增加;多样性指数(Shannon-Weaver index H)计算结果发现H最大值出现在距离尾矿中等距离、重金属浓度在中等程度的样品中。PCA分析结果表明可培养异养菌数量与理化性质如有机碳、有机质、含水率等相关性较大,重金属影响不明显;而多样性指数H除与上述理化性质相关性较大外,还受到重金属Ag、Zn、As、Pb、Ni、Cr等的影响,而在样品中含量普遍比较高的重金属如Cu、Cd等并不成为影响微生物多样性的主要因素。【结论】从这些长期受重金属污染的野外实地样品来看,以上结果说明不同重金属浓度对微生物多样性的影响可能并不是实验室研究的简单的线性关系。  相似文献

Today indiscriminate and uncontrolled discharge of metal contaminated industrial effluents into the environment has become an issue of major concern. Heavy metals, being non-biodegradable and persistent, beyond a permissible concentration form unspecific compounds inside the cells thereby causing cellular toxicity. The only alternative to remove them from the wastewater is by immobilizing them. The conventional methods adopted earlier for this purpose included chemical precipitation, oxidation, reduction, filtration, electrochemical treatment, evaporation, adsorption and ion-exchange resins. These methods require high energy inputs especially when it refers to dilute solutions. Here microbial biomass offers an economical option for removing heavy metals by the phenomenon of biosorption. Non-living or dead biomass sequester metal(s) on their cell surface due to certain reactive groups available like carboxyl, amine, imidazole, phosphate, sulphydryl, sulfate and hydroxyl. The process can be made economical by procuring spent biomass from industry or naturally available bulk biomass. A batch or a continuous process of removal of heavy metals directly from effluents can be developed in a fixed bed reactor using the immobilized biomass. Further biosorption potential of the biomass can be improved by various physical and chemical treatments. The availability of variety of microbial biomass and their metal binding potential makes it a economical and sustainable option for developing effluent treatment process for removal and recovery of heavy metals.  相似文献

The metalloregulatory protein MerR, which exhibits high affinity and selectivity toward mercury, was exploited for the construction of microbial biosorbents specific for mercury removal. Whole-cell sorbents were constructed with MerR genetically engineered onto the surface of Escherichia coli cells by using an ice nucleation protein anchor. The presence of surface-exposed MerR on the engineered strains enabled sixfold-higher Hg(2+) biosorption than that found in the wild-type JM109 cells. Hg(2+) binding via MerR was very specific, with no observable decline even in the presence of 100-fold excess Cd(2+) and Zn(2+). The Hg(2+) binding property of the whole-cell sorbents was also insensitive to different ionic strengths, pHs, and the presence of metal chelators. Since metalloregulatory proteins are currently available for a wide variety of toxic heavy metals, our results suggest that microbial biosorbents overexpressing metalloregulatory proteins may be used similarly for the cleanup of other important heavy metals.  相似文献

酸性矿山废水污染的水稻田土壤中重金属的微生物学效应   总被引:20,自引:1,他引:20  
采样调查了广东大宝山地区受酸性采矿废水长期污染的亚热带水稻田的土壤理化性质 ,重金属 Cu、Pb、Zn、Cd的全量及其 DTPA浸提量 ,以及微生物生物量及其呼吸活性等指标。利用主成分和逐步回归分析了影响土壤重金属的有效性及其微生物学效应的因素。结果表明 :土壤高含硫 ,强酸性 ,有机碳、全氮较低 ,4种金属的全量普遍超标。DTPA可提取态金属含量较高 ,不仅与其全量呈显著正相关 ,而且与土壤酸度和粘粒含量正相关 ,和 Mn含量负相关。过量的金属显著降低了土壤微生物生物量 C、N、微生物商、生物量 N/全 N比 ,并抑制了微生物呼吸强度和对有机碳的矿化率 ,导致了土壤 C/N比的升高。同时 ,金属对微生物群落及生理代谢指标 ,如微生物生物量 C/N比和代谢商的影响不显著。 DTPA可提取态金属 ,特别是 DTPA- Cu是导致微生物生物量和活性指标变化的主要因素。以有机碳 (或全氮 )为基数的复合微生物指标降低了土壤性质差异造成的干扰 ,较单一指标更能准确指示微生物对金属胁迫的反应。土壤硫没有对金属有效性和微生物指标产生明显影响 ,但其氧化过程可能引起酸化和金属离子的释放  相似文献

Phytoremediation is an eco friendly approach for remediation of contaminated soil and water using plants. Phytoremediation is comprised of two components, one by the root colonizing microbes and the other by plants themselves, which degrade the toxic compounds to further non-toxic metabolites. Various compounds, viz. organic compounds, xenobiotics, pesticides and heavy metals, are among the contaminants that can be effectively remediated by plants. Plant cell cultures, hairy roots and algae have been studied for their ability to degrade a number of contaminants. They exhibit various enzymatic activities for degradation of xenobiotics, viz. dehalogenation, denitrification leading to breakdown of complex compounds to simple and non-toxic products. Plants and algae also have the ability to hyper accumulate various heavy metals by the action of phytochelatins and metallothioneins forming complexes with heavy metals and translocate them into vacuoles. Molecular cloning and expression of heavy metal accumulator genes and xenobiotic degrading enzyme coding genes resulted in enhanced remediation rates, which will be helpful in making the process for large-scale application to remediate vast areas of contaminated soils. A few companies worldwide are also working on this aspect of bioremediation, mainly by transgenic plants to replace expensive physical or chemical remediation techniques. Selection and testing multiple hyperaccumulator plants, protein engineering ofphytochelatin and membrane transporter genes and their expression would enhance the rate of phytoremediation, making this process a successful one for bioremediation of environmental contamination. Recent years have seen major investments in the R&D, which have also resulted in competition of filing patents by several companies for economic gains. The details of science & technology related to phytoremediation have been discussed with a focus on future trends and prospects of global relevance.  相似文献

This review provides an analysis of recent data on the mechanisms of degradation of lignocellulosic materials and xenobiotics by basidiomycetes. Special attention is given to the analysis of the current state of research of ligninolytic enzymes and their involvement in the degradation of xenobiotics. Data on the practical use of basidiomycetes for bioconversion of industrial wastes are systematized. The most promising areas of bioconversion technologies are considered, such as contaminated water purification (including wastewater), cleanup of soils contaminated with heavy metals and xenobiotics, and degradation of difficult-to-degrade substrates (lignin and lignocellulose wastes, low-energy coal, and synthetic polymers).  相似文献

The lamB gene was inserted at with DNA fragments encoding N-terminal beta- and C-terminal alpha-domains of human metallothionein 1A (HMT1A). The hybrid LamB proteins were expressed as full-length products. Virtually whole pool of hybrid LamB proteins was found localized in the outer membrane of E. coli to and cells expressing LamB variants retained sensitivity to lambda phage, indicating their correct folding. Expression of hybrid LamB proteins increased natural ability of E. coli accumulate bivalent heavy metals ions with the highest efficiency observed for cadmium. The order of amount of cadmium accumulated is alpha-domain of HMT1A > HMT1A > beta-domain of HMT1A. This correlates with affinity for cadmium and stability of metallothionein and its individual domains. This confirms suitability of LamB vehicle for surface display of various bioactive molecules and suggests possibility of engineering of cell surface for bioremediation of heavy metals.  相似文献

Glutathione (GSH) plays an important role in protecting plants from environmental stresses like oxidative stress and xenobiotics. Glutathione-derived peptides are involved in heavy metal detoxification in plants and fungi. Terrestrial and aquatic bryophytes were investigated for their biochemical response to heavy metals. The GSH pool increased significantly in the first two days after supply of 100 μmol/L Cd(II). PCs were not detected. Cd(II) also induced the enhancement of the GSH pool in the water moss Fontinalis antipyretica. Cysteine and γ-glutamyl-cysteine also increased during Cd(II) treatment, but remained on a lower level. Uptake experiments with Cd(II) showed a fast regulation of equilibrium between the Cd(II) content of the medium and the plant surface, followed by a slow migration of Cd(II) to intracellular sites. The main storage compartment of heavy metals in Fontinalis are the vacuoles, where they are precipitated as phosphates. In the cytoplasm, the S-content increased during Cd(II) exposition. EEL-spectra indicate that in the cytoplasm, Cd(II) is chelated by SH-groups. All findings support the idea that in the investigated moss species, GSH plays an essential role in heavy metal detoxification during the transport of the metals through the cytoplasm.  相似文献

调查了沈阳张士灌区长期污水灌溉造成的原位农田土壤重金属污染状况,从土壤微生物生物量、微生物活性和微生物种群数量的角度评价了长期重金属污染对农田土壤生态系统的影响.结果表明,张士灌区土壤存在严重的Cd污染,土壤Cd含量达1.75~3.89 mg·kg -1,部分区域还伴有Cu、Zn复合污染.在目前污染程度下,土壤微生物生物量碳(Cmic)、微生物商(qM)、土壤脱氢酶活性以及自生固氮菌数量随土壤重金属含量增加呈下降趋势,代谢商(qCO2)随土壤重金属含量增加显著升高,而底物诱导呼吸强度(SIR)、纤维素酶活性以及细菌、放线菌和真菌数量无明显变化.相关性分析表明,土壤Cd含量变化是影响微生物参数变化的主要因素,在微生物参数中微生物商和代谢商对重金属污染最敏感.  相似文献
