1.Organic acids, such as citric, -ketoglutaric, succinic, fumaricand L-malic acids, inhibit the IAA-induced growth of Avena coleoptilesections. An analysis of metabolism by measurement of respiratory quotient values indicates that reduced substances, such as lipids and/or amino acids, are the primary respiratory substrates of dormant Dictyostelium discoideum spores. The spores appear to consume both reduced substances and carbohydrates during the swelling stage of germination. The respiration of emerged myxamoebae is again dominated by the consumption of reduced substances. The pool of trehalose remains largely intact during heat-induced activation and also during postactivation lag. The initiation of spore swelling is accompanied by a decrease in the trehalose pool; the majority of trehalose is consumed before late spore swelling. Upon placing heat-activated spores under restrictive environmental conditions, swelling and trehalose hydrolysis are both prevented. Release from these conditions results in rapid swelling and hydrolysis of trehalose. Trehalase, the enzyme responsible for trehalose breakdown, is present in dormant spores at basal levels. This preformed enzyme is responsible for the hydrolysis of trehalose even though there is a significant increase in trehalase activity with the emergence of myxamoebae. RNA and protein synthesis inhibitors do not prevent trehalose hydrolysis or spore swelling. It is concluded that oxidation of reduced substances occurs in dormant, activated, and swollen spores, as well as in emerged myxamoebae of D. discoideum. Carbohydrate utilization dominates over the oxidation of reduced substances only during the swelling stage of germination.  相似文献

No exact estimation of the amount of radioactive free aminoacids in the cells of the tissue with large size of apparentfree space was possible, since the exact size of the apparentfree space cannot be measured. Furthermore, estimation of thesize of the protein precursor pool, using the method of Hollemanand Key, was not possible in hypocotyl sections of mung bean(Phaseolus mungo L. cv. Black), because of the great differenceover the length of a section in the rate of the incorporationof leucine-14C into protein. Also, most of the radioactivityin the active pool disappeared within 10 min of the chase periodin the presence or absence of IAA, before the effect of IAAon protein synthesis was shown. Thus, neither can the pulse-chaseexperiment be used to study auxin-induced protein synthesis. IAA stimulated neither the formation of amino acids from acetate-14C,nor the incorporation of the newly formed amino acids into protein.However, IAA did stimulate both the uptake of sucrose-14C andpyruvate-14C into tissue and/or the formation of amino acidsfrom these substances, which resulted in stimulation of theincorporation of these radioactive amino acids into proteins.Enhancement effects of IAA on the rates of amino acid formationand the incorporation of amino acids into protein were of thesame magnitude. These results indicate that radioactive amino acids are spontaneouslyincorporated into proteins without any positive effect by IAA.Furthermore, IAA protects the degradation of some protein fractions.All diis evidence raises questions as to the validity of thehypothesis that auxin promotes protein synthesis. -Amanitin and cordycepin at various concentrations were testedfor their inhibitory effect on the fresh weight increase ofVigna unguiculata embryonic axes after the onset of imbibitionand on the incorporation rate of 3H-labeled leucine into proteinin axes of the 36–38 h stage. -Amanitin at 0.5–5µ/Kg/ml clearly exerted an inhibitory effect on both thefresh weight increase and the protein synthesis. This drug at1 µg/ml, however, showed no significant effect on theprotein synthesis at an early stage of imbibition (4 h), whereascycloheximide was a very potent inhibitor. By experiments inwhich ‘dry’ axes were allowed to imbibe 3H-lebeledadenosine solution for 4 and 12 h in the presence of -amanitin,it was found that poly A+RNA was newly synthesized to some extentin axes as early as 4 h after the onset of imbibition and thatthe drug effectively inhibited the poly A+RNA synthesis. Pattern of 3H-uridine incorporation into RNA of spores of Onocleasensibilis imbibed in complete darkness (non-germinating conditions)and induced to germinate in red light was followed by oligo-dTcellulose chromatography, gel electrophoresis coupled with fluorographyand autoradiography. In dark-imbibed spores, RNA synthesis wasinitiated about 24 h after sowing, with most of the label accumulatingin the high mol. wt. poly(A)RNA fraction. There was noincorporation of the label into poly(A) + RNA until 48 h aftersowing. In contrast, photo-induced spores began to synthesizeall fractions of RNA within 12 h after sowing and by 24 h, incorporationof 3H-uridine into RNA of irradiated spores was nearly 70-foldhigher than that into dark-imbibed spores. Protein synthesis,as monitored by 3H-arginine incorporation into the acid-insolublefraction and by autoradiography, was initiated in spores within1–2 h after sowing under both conditions. Autoradiographicexperiments also showed that the onset of protein synthesisin the cytoplasm of the germinating spore is independent ofthe transport of newly synthesized nuclear RNA. One-dimensionalsodium dodecyl sulphate-polyacrylamide gel electrophoresis of35S-methionine-labelled proteins revealed a good correspondencebetween proteins synthesized in a cell-free translation systemdirected by poly(A) +RNA of dormant spores and those synthesizedin vivo by dark-imbibed and photo-induced spores. These resultsindicate that stored mRNAs of O. sensibilis spores are functionallycompetent and provide templates for the synthesis of proteinsduring dark-imbibition and germination. Effects of -hydroxy-2-pyridinemethanesulfonate (-HPMS), 2,3-epoxypropionate(glycidate), and cyanide on the photosynthetic activities ofChromatiumwere studied. -HPMS stimulated photosynthetic CO2fixation in the bacterial cells in both N2 and O2 environments.The formation and subsequent excretion of both glycolate andglycine in the O2 atmosphere were markedly enhanced by -HPMS.In contrast to a recent report by Zelitch [Arch. Biochem. Biophys.163: 367–377 (1974) ] that glycidate specifically inhibitsglycolate formation in tobacco leaf disks, we found that ithad no influence on CO2 fixation by Chromatium in either N2or O2 atmosphere, and that the synthesis and extracellular excretionof glycolate were markedly stimulated by glycidate treatment.Cyanide (0.01–1 mM) exerted a marked inhibitory effecton photosynthetic CO2 fixation in N2. In O2 atmosphere, photosynthesiswas stimulated by 0.01 mM cyanide, and inhibited by it abovethis level.   1. The role of sulfur in the cell division of Chlorella was studiedby following the fate of the sulfur supplied to the sulfur-deficientcells using 35S as a tracer.
  2. The sulfur-deficient cells whichwere unable to perform celldivision were made capable of divisionby the provision of 36S-labeledsulfate under non-photosynthesizingconditions. Soon after theprovision of sulfate the labeledsulfur went rapidly into thecold perchloric acid (PCA)-solublefraction of algal cells,almost entirely in the form of sulfateand/or some other inorganicsulfur substance (s). With the lapseof time, more or less remarkablechanges occurred in the patternof 35S-distribution in differentfractions of cell material.It was noticed that, at the onsetof cell division, a sulfur-containingpeptide-nucleotide compound(s)(SPN), which has been reportedearlier, appeared in a largequantity in the cold PCA-solublefraction, and that its quantitydecreased gradually during thesubsequent process of cell division,suggesting that the compoundwas transformed into some othersubstance (s), presumably withits nucleotide moiety going intonucleic acids and the peptidemoiety going into some essentialproteins.
  3. Another noteworthyphenomenon observed during the process ofcell division wasthe incorporation of 36S in a group of hotPCA-soluble substances.These sulfur substances were revealedto be sulfur-containingnucleotidic compounds, which might possiblybe some essentialcomponents of, or substances in close relationto, deoxypentosenucleic acid (DNA).

Sugar starvation-induced synthesis and extracellular liberationof -amylase molecules in suspension-cultured cells of rice (Oryzasativa L.) required Ca2+, although the level of translatable-amylase mRNA was not affected in the presence of Ca2+. Sugardepletion markedly stimulated Ca2+ uptake by rice cells andsucrose supplementation reduced it. Immunohistochemical andelectron probe microanalyzer studies indicated an apparent resemblancebetween the distribution pattern of Ca2+ and that of -amylasemolecules induced in the sugar-depleted cells. Ca2+ uptake wasreduced by sucrose, maltose, fructose, and glucose similarlyat more than 5 mM, but was unaffected by mannitol (88 mM), 6-deoxy-D-glucose(10 mM), and 3-O-methyl-D-glucose (10 mM). Furthermore, an effectiveCa2+ channel blocker, La3+ significantly inhibited the Ca2+uptake and the synthesis and extracellular liberation of -amylasemolecules in the absence of sucrose, while a general P-typeATPase inhibitor, vanadate greatly stimulated both in the presenceof sucrose. We concluded that, by controlling the Ca2+ uptake,metabolic sugars regulate the protein synthesis and posttranslationalsecretory processes of -amylase molecules in rice cells. 4 Invited research fellow of the Japan Society for the Promotionof Science. The development of taurine uptake into the unicellular greenalga Chlorella fusca 211-8b was characterized as a specificresponse to either nitrate or sulphate limitation. Taurine transportunder nitrogen starvation was stimulated by low pH and showeda biphasic kinetics with Km-values of 1.1 x 10–3 mol dm–3and 1.0 x 10–2 mol dm–3. Uptake was substantiallyinhibited by all - and ß-amino acids tested, whereassulphonate analogues failed to diminish taurine accumulation.Thus, uptake seemed to be mediated by a ‘general aminoacid permease’, unable to discriminate between carboxyland sulphonyl groups. However, Chlorella fusca could not catabolizethis unusual ß-amino acid and mobilize the amino-boundnitrogen for growth. Only a small group of -amino acids supportedthe growth of Chlorella fusca as an efficient nitrogen source. Nitrite reductase was extracted from the red alga Porphyra yezoensisUeda and purified through precipitation with ammonium sulfate,column chromatographies, and polyacrylamide gel disk electrophoresis.The enzyme preparation thus obtained showed a single band ondisk electrophoresis. The absorption spectrum had three maxima at 385 nm (Soret band),580 nm (-band), and 278 nm; the ratio of absorbance of the Soretband to the -band was 4.3. The molecular weight and the numberof amino acid residues were estimated to be 63,000 and 601,respectively. The enzyme activity was optimal at around pH 7.5, and its activitywas heat labile as indicated by reduction of activity by about70% when heated at 37°C for 10 min. The enzyme used ferredoxin and methyl viologen, but not NADP+or NAD+, as the electron carriers. Moreover, reduced forms ofthe latter two showed no effect on its activity. Km values ofthis enzyme for NO2, Fd, and MV were 8.1 x 10–4M, 4.3 x 10–8 M, and 3.7 x 10–4 M, respectively.Almost half of its activity was lost when potassium cyanidewas added at a concentration as low as 10–5 M, and theKi value was 1.8 x 10–5 M. Thus, the nitrite reductaseof Porphyra must be systematically grouped in EC 1.7.7.1 [EC] . A Ras-related NTP-binding protein was partially purified froma membrane fraction derived from the mycelia of Neurospora crassa.[-32P]ATP and [-32P]GTP were incubated with mem brane and solublefractions which were then irradiated with UV light to inducecrosslinking of tightly bound nucleotides. After SDS-polyacrylamidegel electrophoresis, blotting onto a nitrocellulose filter andautoradiography it was apparent that most of the proteins thatbound [-32P]-GTP also bound [-32P]ATP. Pretreatment of the membranefraction with Ras-specific antibody effectively blocked thebinding of [-32P]ATP and [-32P]GTP to several ATP-GTP-bindingproteins. The band of a protein with a molecular weight of 26kDa on the SDS-polyacrylamide gel cross-reacted strongly withthe Ras-specific antibody. The protein was extracted from thegel and further purified by repeated gel electrophoresis. Thepurified protein bound [-32P]ATP, [-32P]-GTP, [-32P]CTP and[-32P]UTP at 1.6x10 M and was autophosphorylated in thepresence of [-32P]ATP and [-32P]GTP at 1.7x10 M. Pretreatmentof the protein with Ras-specific antibody partially blockedthe autophosphorylation in the presence of these nucleotides.The binding of [-32P]ATP to the NTP-binding protein was blockedby addition of ATP at 10–4–10–3 M. ATP ata concentration of 10–4 M prevented the binding of [-32P]to a greater extent than did GTP at the same concentration.Binding of [-32P]CTP and [-32P]UTP to the protein was also observed. Amino acid composition of the free amino acid pool and the TCA-insolubleprotein fraction were investigated in root tips of pea and Tamarixtetragyna plants grown at various levels of NaCl salinity. Salinitystress induced an increase of proline content, mainly in thefree amino acid pool in both plants, and of proline or hydroxyprolinecontent in the protein. Externally-supplied proline was absorbedand incorporated into protein, by pea roots, more effectivelythan by Tamarix roots. Salinity stress, apparently, stimulatedthe metabolism of externally-supplied labelled proline. Pearoots have a very large pool of free glutamic acid; however,70 per cent of the 14C from externally-supplied 14C-U-glutamicacid was released as CO2. Very small amounts of it were incorporatedinto protein. No measurable amount of radioactivity could bedetected in any one of the individual amino acids, either ofprotein hydrolysate or the free amino acid pool. Proline very effectively counteracted the inhibitory effectof NaCl on pea seed germination and root growth. A similar effectbut to a lesser degree was achieved with phenylalanine and asparticacid. The feasibility of proline being a cytoplasmic osmoticumis discussed.  相似文献

The compositions and positional distributions of fatty acidsin the major leaf phospholipids of phosphatidylglycerol, phosphatidylcholineand phosphatidylethanolamine were analyzed by gas-liquid chromatographyand enzymic hydrolysis, and chilling-sensitive and chilling-resistantplants were comparcd with respect to the relative contents ofpalmitic and trans-3-hexadecenoic acids in the separated phospholipids.A distinct difference between these plants was found in thefatty acid compositions of phosphatidylglycerol, in which thesum of palmitic and trans-3-hexadecenoic acids ranged from 60to 78% of the total fatty acids in 8 species of chilling-sensitiveplants, and from 50 to 57% in 11 species of chilling-resistantplants. The only exception among the chilling sensitive plantsin this respect was the tomato, in which the sum of palmiticand trans-3-hexadecenic acids in phosphatidylglycerol amountedto 54%. The fatty acid compositions and the positional distributionsof fatty acids in phosphatidylglycerol suggest that the occurrenceof high proportions of dipalmitoyl and 1-palmitoyl-2-(trans-3-hexadecenoyl)species in this lipid is correlated with the susceptibilityto chilling of the leaves of higher plants. All three races of Neotricula aperta, an epilithic, schistosometransmitting, snail of the Mekong and Mul rivers of NortheastThailand and southern Laos, were found to take up acetate froma dilute solution. After 48 h incubation the mean specific netuptake rates (µmol–1 g–1 h–1), from750 µM acetate, were: 1.86, -race; 1.39, ß-raceand 3.25, y-race. Over 48 h the snails were able to achievereductions in the ambient acetate concentration of up to 60%.Incubations under bacteriostatis suggested that bacteria arenot involved in the uptake of acetate by N. aperta. The uptakecharacteristics conform to the Michaelis-Menten model. The transportconstants, Jmax (µmol–1 g–1 h–1) andKt (µM) were 1.10 and 102 respectively (-race). Racialdifferences in uptake characteristics are discussed in relationto micro-habitat differences. HPLC indicated concentrations of acetate in y-N. aperta microhabitatsof around 325 µM. This suggests a pool size sufficientto satisfy only 6% of the snail's basal metabolic rate (BMR).Levels within the epilithic aufwuchs, however, are probablyhigh enough to provide for more than 50% of the BMR. The possible role of acetate in the energy metabolism of N.aperta is discussed. Short-chain carboxylic acids (such as acetate),arising from the decomposition of the aufwuchs, could representsources of fermentable organics that may be taken up by N. apertasnails and used to supplement their nutrition during times offood shortage. However, further investigations involving 14C-labellingtechniques are required. The findings of this investigationhave implications for the chemical ecology and life-cycle ofN. aperta. Fatty acid synthesis from [14C]acetate was studied in the brownalgae, Fucus serratus and Fucus vesiculosus, which had beencollected from sites polluted by run-off from old copper mines.Algae collected from areas exposed to copper in situ showedchanges in the pattern of fatty acids labelled in vitro withan increase in oleate labelling and a decrease in palmitatelabelling in both species. The endogenous fatty acid patternsfor algae from polluted sites also differed from those fromunpolluted sites. Algae from sites polluted by copper containedmore oleate than samples from non-polluted sites. In addition,there were decreases in the polyunsaturated fatty acids -linolenate,octadecatetraenoate and docosapentaenoate for copper-exposedalgae. These differences are discussed with regard to the possiblemetabolic site of copper's action and to the relative toleranceof Fucus spp. to high environmental dissolved copper levels. We screened rat brain cDNA libraries and used 5'rapid amplification of cDNA ends to clone two electrogenicNa+-HCO3 cotransporter(NBC) isoforms from rat brain (rb1NBC and rb2NBC). At the amino acidlevel, one clone (rb1NBC) is 96% identical to human pancreas NBC. Theother clone (rb2NBC) is identical to rb1NBC except for 61 uniqueCOOH-terminal amino acids, the result of a 97-bp deletion near the3' end of the open-reading frame. Using RT-PCR, we confirmed thatmRNA from rat brain contains this 97-bp deletion. Furthermore, wegenerated rabbit polyclonal antibodies that distinguish between theunique COOH-termini of rb1NBC (rb1NBC) and rb2NBC (rb2NBC).rb1NBC labels an ~130-kDa protein predominantly from kidney, andrb2NBC labels an ~130-kDa protein predominantly from brain.rb2NBC labels a protein that is more highly expressed in corticalneurons than astrocytes cultured from rat brain; rb1NBC exhibits theopposite pattern. In expression studies, applying 1.5%CO2/10 mM HCO3 toXenopus oocytes injected with rb2NBC cRNA causes 1)pHi to recover from the initial CO2-inducedacidification and 2) the cell to hyperpolarize. Subsequently,removing external Na+ reverses the pHi increaseand elicits a rapid depolarization. In the presence of 450 µM DIDS,removing external Na+ has no effect on pHi andelicits a small hyperpolarization. The rate of the pHidecrease elicited by removing Na+ is insensitive toremoving external Cl. Thus rb2NBC is aDIDS-sensitive, electrogenic NBC that is predominantly expressed inbrain of at least rat.

The metabolic transformation of glycolate to glycine occurringin photosynthesizing cells of Chromatium was investigated bythe radioisotopic technique and by amino acid analysis. By analyzingthe distribution of radiocarbon upon feeding [1-14C] glycolate,[2-14C] glyoxylate and [1-14C] glycine to bacterial cells, itwas demonstrated that glycolate is converted to glycinc viaglyoxylate, and both glycolate and glycine are excreted extracellularly.Although the formation of serine was barely detected by theabove two techniques in both N2 and O2 atmospheres, it was foundthat 14CO2 is evolved quite markedly from both [1-14C] glycolateand [1-14C] glycine fed to the Chromatium cells. Analyticalresults of transient changes in amino acid compositions underatmospheric changes of N2O2 and by the addition of exogenousglycolate in N2 confirm the notion that glycolate is convertedto glycine. Acidic amino acids (glutamic acid and aspartic acid)appear to take part in glycine formation as amino donors. We investigated theeffects of epidermal growth factor (EGF) on activeNa+ absorption by alveolarepithelium. Rat alveolar epithelial cells (AEC) were isolated andcultivated in serum-free medium on tissue culture-treated polycarbonatefilters. mRNA for rat epithelial Na+ channel (rENaC) -, -,and -subunits and Na+ pump1- and1-subunits were detected inday 4 monolayers by Northern analysisand were unchanged in abundance in day5 monolayers in the absence of EGF. Monolayerscultivated in the presence of EGF (20 ng/ml) for 24 h fromday 4 to day5 showed an increase in both1 and1Na+ pump subunit mRNA but noincrease in rENaC subunit mRNA. EGF-treated monolayers showed parallelincreases in Na+ pump1- and1-subunit protein by immunoblotrelative to untreated monolayers. Fixed AEC monolayers demonstratedpredominantly membrane-associated immunofluorescent labeling withanti-Na+ pump1- and1-subunit antibodies, withincreased intensity of cell labeling for both subunits seen at 24 hfollowing exposure to EGF. These changes inNa+ pump mRNA and protein precededa delayed (>12 h) increase in short-current circuit (measure ofactive transepithelial Na+transport) across monolayers treated with EGF compared with untreated monolayers. We conclude that EGF increases activeNa+ resorption across AECmonolayers primarily via direct effects onNa+ pump subunit mRNA expressionand protein synthesis, leading to increased numbers of functionalNa+ pumps in the basolateralmembranes.

The nature of amino acid-sugar linkages in cell walls was investigatedin a monocotyledonous tissue, rice coleoptiles. The molar ratiosof aspartic acid, threonine, and serine in cell walls were decreasedby hydrazinolysis in coleoptiles grown both on and under water.The molar ratios of threonine and serine were decreased alsoby a NaOHNaBH4 treatment, while the alanine content was increased,and -aminobutyric acid was not formed. The cell walls were treated with NaOH in the presence of NaB3H4,hydrolyzed, then divided into amino acid and sugar fractions.Two distinct radioactive peaks were detected in the thin-layerchromatography of the amino acid fractions. One was identifiedas alanine derived from glycosylated serine; the other was confirmedto be an oxidation product of glucosaminitol. There was justone 3H-labeled product in the sugar fractions, galactitol. Theseresults suggest the presence of serine-O-galactose and asparagine-N-N-acetylglucosamine linkages in rice coleoptile cell walls. The existence of glucosamine linked to amino acids was furthersupported by the incorporation of 14C-glucosamine into cellwalls. These linkages were also detected in the cell walls ofa dicotyledonous tissue, Vicia epicotyls. (Received April 2, 1981; Accepted June 24, 1981)  相似文献
