An indigenous betabaculovirus (PapyGV) of the Pandemis leafroller, Pandemis pyrusana (Kearfott), was studied in the laboratory and greenhouse to determine how the virus affected leafroller mortality and foliar damage. Probability of mortality increased with virus concentration as observed after 7 and 10 days of feeding on virus treated diet in neonates and second instar larvae. LC50 estimates for neonates at 7 and 10 days was 2743 and 389 occlusion bodies (OBs)/mm2. For second instars, LC50 was 139,487 and 813 OBs/mm2 at 7 and 10 days. There was no biologically significant mortality response to increasing virus concentrations by fourth instar larvae; however, when fourth instar larvae were infected with virus on diet and then fed apple leaves, the leaf area consumed declined up to 50% with higher virus concentrations. In a greenhouse study, neonate larvae that fed on seedlings treated with water showed >90% survival and 80% pupation rate of larvae after being transferred to diet. In contrast, larvae that fed on apple seedlings sprayed with 3×106 OBs/ml showed poor survival when transferred to diet after acquiring the virus and failed to reach the pupal stage. This virus shows promise for population regulation and can produce reduction in feeding damage.  相似文献

The effects of a transgenic Bacillus thuringiensis (Bt)-cotton cultivar (DPL 32) on three instars of the soybean looper, Pseudoplusia includens (Walker), were determined in laboratory studies. First, third, and fifth instars were fed field collected Bt-cotton leaves for 1, 2, four and 7 d or until pupation, and then transferred to artificial diet. Mortality during the larval stage increased linearly in response to an increase in the length of feeding time on Bt-cotton by first and third instars. The maximum mortality of about two out of three larvae occurred for first instars fed on Bt-cotton until pupation. For the fifth instar, there was no significant response to feeding time; however, most of these larvae reached pupation before 4 d of feeding on Bt-cotton. The length of the larval developmental period also increased linearly with an increase in feeding time on Bt-cotton in first and third instars; again, there was no significant response in the fifth instars. For both mortality and larval developmental time, the linear trend lines for the first and third instars were quite similar. Pupal weight declined linearly in the first and fifth instars in response to feeding time on Bt-cotton. Although pupal weight also declined for third instars, the response was not linear. The effect of Bt-cotton appears not to extend past pupation in that there were no significant responses in mortality and developmental time of pupae during the pupal stage. These data indicate that larvae surviving Bt-cotton are adversely affected in several ways, which should be considered in evaluating Bt-cotton suppression of soybean looper infestations.  相似文献

Feeding performance of larvae of the obliquebanded leafroller, Choristoneura rosaceana (Harris), was estimated on apple and three alternative hosts in the Okanagan Valley. The obliquebanded leafroller is native to the study area and apple was introduced in the valley at the beginning of the century. Components of insect fitness were measured to help formulate predictions about the evolution of host preference in this generalist herbivore. In the summer, females attained a higher pupal weight on apple than on trembling aspen, wild rose and snowberry. Males performed best on apple and trembling aspen. In the field, females arising from caterpillars fed on apple leaves had a 26% greater mean fecundity than if fed on alternative hosts. The diet also affected development time; females initiated pupation later on trembling aspen than on the other hosts. Finally, no differences were found in larval survival nor in the proportion of larvae diapausing when fed on different hosts. These components of fitness indicate apple as the most suitable host in the summer.In the fall, diet influenced larval propensity to initiate diapause in the second or third instar, the time elapsed from hatching to diapause initiation and the weight of the instars initiating diapause. However, no difference in overwintering survival among larvae fed on different hosts was detected. The rank order of water and nitrogen contents of leaves of the host plants did not match the rank order of larval feeding performance in either season. A high suitability of apple and its abundance in the Okanagan valley are factors that would favor the evolution of an increased utilization of this host by the obliquebanded leafroller.  相似文献

【目的】本研究探讨使用人工饲料部分替代桑叶饲育后家蚕Bombyx mori幼虫肠道细菌群落的变化情况,并分析人工饲料替代桑叶的不同饲育模式下肠道关键细菌与蛹重和茧层量的相关性,为人工饲料部分替代法饲育家蚕的实际应用提供理论依据。【方法】采用全龄桑叶育(Mul1-5)、1-2龄人工饲料育+3-5龄桑叶育(Art1-2)、1-3龄人工饲料育+4-5龄桑叶育(Art1-3)、1-4龄人工饲料育+5龄桑叶育(Art1-4)和全龄人工饲料育(Art1-5) 5种饲养模式饲养家蚕幼虫,统计5种饲养模式下家蚕的全茧量、茧层量及蛹重;收集不同饲养模式下家蚕5龄幼虫肠道样品,通过高通量测序技术分析其肠道菌群的组成和多样性差异,采用Spearman相关性热图分析肠道细菌与蛹重和茧层量的相关性。【结果】人工饲料与桑叶不同搭配的饲育模式对家蚕茧质有显著的影响,Mul1-5和Art1-5的全茧量最高,二者无显著性差异,Art1-2其次; Mul1-5茧层量最高,Art1-2其次,Art1-5茧层量最低。在不同饲育模式下,家蚕肠道细菌多样性和组成显著不同:在门水平上,Art1-2和Mul1-5的细菌组成最相近,优势菌主要是变形菌门(Proteobacteria)和厚壁菌门(Firmicutes),而Art1-3, Art1-4和Art1-5的细菌组成最相近,变形菌门丰度随着人工饲料饲养时间的增加逐渐增加,厚壁菌门的丰度则呈现相反的趋势;在属水平上,家蚕肠道优势菌属有假单胞菌属Pseudomonas、肠球菌属Enterococcus、鞘脂单胞菌科未分类菌属Sphingomonadaceae unclassified及罗尔斯通菌属Ralstonia等。不同饲育模式下家蚕肠道菌属丰度差异最大的是肠球菌属,Art1-2组中肠球菌属丰度最高(40.9%),随着人工饲料饲养时间的延长,丰度逐渐降低,在Art1-5中仅为0.02%。家蚕肠道菌群大部分关键物种均与茧质呈一定的相关性:柯克斯体属Coxiella与蛹重呈显著正相关,与茧层量呈显著负相关;葡萄球菌属Staphylococcus与蛹重呈显著负相关;肠球菌属与茧层量呈显著正相关。【结论】饲育模式Art1-2最接近于Mul1-5,可为人工饲料部分替代桑叶饲育法饲育家蚕提供参考。人工饲料部分替代桑叶后家蚕肠道菌群结构发生了显著变化,肠球菌属丰度的显著降低及假单胞菌属的显著增加可能与全龄人工饲料养家蚕体质弱有关;一些关键细菌与家蚕蛹重和茧层量存在显著相关性,其影响机制需要进一步研究。  相似文献

2010年6月至7月,在室内研究了美国白蛾Hyphantria cunea(Drury)第1代幼虫对洋白蜡、欧美107号杨、法桐和白榆的取食量及不同的食料种类对美国白蛾生长发育和繁殖的影响。结果表明,美国白蛾对受试的各树种的取食量以洋白蜡为最多,完成一代平均每头可以取食2.21 g,其次为欧美107号杨1.95 g,再次为法桐和白榆,分别为1.85 g/头和1.68 g/头。3龄后幼虫对不同的寄主呈现出一定的偏好性,特别是5龄和6龄幼虫对不同的寄主的取食量表现出显著差异。不同的食料植物对美国白蛾幼虫和蛹的发育历期、存活率、蛹重、成虫寿命、产卵量等有显著影响。取食法桐对其生长发育和繁殖表现出明显的不利性,主要表现为幼虫期延长、蛹重减轻、单雌产卵量降低。  相似文献

The lethal and sublethal effects of tebufenozide on the survival, development, and reproduction of a field strain of tufted apple bud moth, Platynota idaeusalis (Walker) (Lepidoptera: Tortricidae), were assessed by feeding first and third instars tebufenozide-treated diet until pupation. Larval mortality was 27.4 and 44.7% at 0.1 and 0.2 ppm for first instars and 21.9 and 57.8% at 0.2 and 0.4 ppm for third instars, respectively. Treated larvae exhibited higher pupal mortalities, lower pupal weights, and generally more deformed adults than untreated larvae. Larval development was not affected by tebufenozide when neonates were exposed, but development was accelerated slightly at 0.4 ppm for both males and females when third instars were exposed. All treatments produced sex ratios biased toward males. When paired with either treated or untreated males, females resulting from neonates treated at 0.2 ppm and from third instars treated at both 0.2 and 0.4 ppm laid from 37 to 65% fewer eggs. A reduction in fertility was only found when third instars were treated at the higher 0.4 ppm rate. These results suggest that tebufenozide can exhibit a significant effect on the population dynamics of the tufted apple bud moth.  相似文献

昆虫全纯人工饲料配方是精细研究昆虫营养生理的前提。本研究以棉铃虫Helicoverpa armigera幼虫为供试对象, 以本研究室前期发展的多因素多水平配方优化实验设计与分析方法UD-SVR为指导, 以略加修改的苹浅褐卷蛾Epiphyas postvittana幼虫全纯人工饲料为初始配方, 先优化20种天然氨基酸, 再优化蔗糖等其他8种非氨基酸营养组分, 经4轮97个处理组合, 获得一个棉铃虫幼虫全纯人工饲料满意配方, 平均蛹重、 化蛹率分别由初始配方的0.160 g和10.4%高效增至满意配方的0.255 g和97.9%。该全纯人工饲料满意配方的获得, 不但再次证实了UD-SVR方法的有效性, 同时为进一步深入研究棉铃虫营养需求与营养代谢奠定了基础。  相似文献

Crude methanolic extracts made from the twigs of 39 plant samples from six species of Trichilia collected in Costa Rica, were incorporated into artificial diet and fed to neonate Spodoptera litura larvae. All six plant species tested significantly reduced larval growth after 7 and 10 days. The most active species was T. americana, reducing growth, on average, to 3.9% of control at 1000ppm fresh weight. The least active, on average, was T. glabra. A twig extract of T. americana proved to be more active than wood, bark or leaf extracts, with the twig extract reducing growth of S. litura larvae by 50% (EC(50)) at a dietary concentration of 17.2ppm. When T. americana wood extract was incorporated into artificial diet (10, 25, 50 and 75ppm) and fed to S. litura larvae throughout larval development, growth was slowed and the final weight of pupae and adults was reduced. At higher extract concentrations (50 and 75ppm) larvae entered one or two supernumerary instars before pupation occurred. This was shown to be due to both starvation and to post-ingestive activity of the extract.  相似文献

We have examined the effects of dietary selenium (Se) supplementation on larval growth and immunocompetence of the lepidopteran pest, the cabbage looper, Trichoplusia ni. Supplementation of the diet of T. ni larvae with 10–20 ppm Se resulted in a 1 day delay in pupation. The effects of the addition and/or removal of dietary Se on total Se bioaccumulation and sequestration were determined by neutron activation analysis of pupae. Early penultimate instar larvae moved from selenium containing diet to basal diet lost total pupal Se content down to the level of those fed basal diet. Conversely, larvae moved from basal diet to diet containing additional Se rapidly attained pupal Se levels comparable to larvae fed Se throughout larval development. Therefore, dietary Se is rapidly accumulated or lost during larval development, but significant amounts are sequestered from diet into pupae. Larvae were reared on diet supplemented with 5 or 10 ppm Se until the onset of the penultimate instar then infected per os with increasing concentrations of the fatal baculovirus Autographa californica multiple nucleopolyhedrovirus (AcMNPV). Larvae fed Se in the penultimate and ultimate instars were more resistant to viral infection than larvae not fed Se in the final instars. This study indicates that dietary Se levels rapidly impact Se assimilation and sequestration and that tissue Se levels are an important factor in resistance to AcMNPV infection in larval T. ni.  相似文献

Abstract: The effect of crude methanolic extracts of Adhatoda vasica leaves on the feeding and performance of Spodoptera littoralis larvae was investigated in the laboratory. Feeding on fresh leaves resulted in 100% mortality of larvae after 26 days of unsubstantial growth. The extract exhibited strong antifeedant and toxic activity against the larvae when applied either on leaf discs or incorporated into artificial diet. Under choice conditions the antifeedant index calculated over 72 h for neonate larvae increased significantly (from 71.5 ± 3.2 to 92.1 ± 4.2) as the concentration of extract in the treated diet increased from 200 to 1000 ppm. Consumption by the sixth instar larvae of leaf discs dipped in 0.01, 0.1 and 0.2% extract solutions was significantly lower than consumption of control discs in both choice and no-choice tests. The latter two concentrations deterred feeding by 63.4 and 90.4%, respectively, under choice conditions, while only the 0.2% extract solution deterred feeding by 56.8% in the no-choice test. Toxicity of the extracts was manifested by a high mortality, reduced growth rates, and low weight gain by larvae fed on diets containing 200–2000 ppm of the extract. No larvae survived to pupation under the latter concentration. The time to pupation increased from 15.8 ± 0.4 to 37.9 ± 4.1 days as the extract concentration in diet increased from 0 to 1000 ppm. When fed to the fifth instar larvae, the crude extract significantly reduced consumption, growth, utilization of ingested and digested food, and approximate digestibility. The consumption-dependent growth efficiency of animals fed on extract-free diet was significantly higher than the growth efficiency of animals fed on extract-containing diets, suggesting both antifeedant and toxic activities of the extract.  相似文献

The effects of diet and temperature on instar numbers and head capsule width in Spodoptera litura F. were compared among individuals reared on an artificial diet, lettuce and perilla leaves at 25 and 30 °C. The number of instars that the insect completed varied as diet and temperature were changed. All the larvae developed through seven instars at 25 °C regardless of diet, but at 30 °C, the number of instars varied depending on the diet. All larvae fed on lettuce leaves had six instars, while larvae fed on the artificial diet passed through seven instars. On perilla, 52% of larval individuals had six instars, and the rest had seven instars. Head capsule width could be used effectively to determine the developmental stage of individual larvae. The frequency distribution of head capsule width showed six or seven distinct peaks, depending on diet and temperature conditions. The relationship between mean head capsule width and weight of larvae was described using an exponential model.  相似文献

The effect of ingestion of transgenic tomato leaves expressing the plant lectin Galanthus nivalis agglutinin (GNA) on development of larvae of Lacanobia oleracea (Linnaeus) was studied under laboratory conditions. When L. oleracea larvae were fed on tomato line 14.1H, expressing approximately 2.0% GNA, significant increases in the mean larval weight and in the amount of food consumed were found. This resulted in an overall reduction in the mean development time to the pupal stage of approximately 7 days. A significant increase in the percentage survival to the adult moth was also recorded when newly hatched larvae were reared on transgenic tomato leaves (72%) compared to larvae reared on untransformed leaves (40%). The effects of ingestion of GNA by L. oleracea larvae, via artificial diet or the leaves of transgenic tomato or potato plants, on the subsequent development of its solitary endoparasitoid Meteorus gyrator (Thunberg) was also studied. No significant effects on the life cycle parameters of M. gyrator developing in L. oleracea fed on GNA-containing diets were observed. Experiments with transgenic potato plants indicated that the stadium of the host larvae at parasitism had a greater influence on M. gyrator development than the presence of GNA. Potential GNA-binding glycoproteins were detected in the gut and body tissues of larval M. gyrator. Despite detection in host tissues, GNA could not be detected in adult M. gyrator and therefore it is likely that at the time of pupation M. gyrator are able to void the GNA in the meconial pellet.  相似文献

First-, third-, and fifth-instar Epiphyas postvittana (Walker) were exposed to a range of lufenuron concentrations (0-200 ppm) incorporated into synthetic diet and their subsequent development and mortality responses were determined. For all instars the greatest change in mortality response occurred over lufenuron concentrations < or = 3 ppm. However, third and fifth instars displayed an increase in mortality earlier than first instars, and were more sensitive to the lower lufenuron concentrations in this range. Only first and third instars subjected to < or = 2.5 ppm lufenuron survived the 26-d exposure trial. No larvae first exposed to lufenuron as first or third instars survived to pupation if ingesting concentrations of > or = 1 and > or = 3 ppm, respectively. Consumption of lower lufenuron concentrations by these larvae delayed pupation and resulted in pupal deformity. In contrast, fifth instars subjected to 100 ppm were capable of surviving the 26-d trial period and displayed a slower progressive reduction in survival to pupation with increase in lufenuron concentration. Also in contrast to more immature stages, fifth instars exposed to lufenuron developed more rapidly to pupation than larvae not exposed to the insect growth regulator (IGR), and all resulting pupae were normal. Third instars were exposed to sublethal lufenuron concentrations (0-3 ppm) for 4 d and the fourth-instar survivors subjected to a controlled atmosphere cold storage treatment (2% O2, 2% CO2, 0.6 degree C). Larvae ingesting diet containing 0.5 ppm (and to a lesser extent 1 ppm) lufenuron required longer exposure to the postharvest treatment to achieve > or = 95% mortality than larvae not ingesting the IGR. However, the analogous mortality response of larvae exposed to 3 ppm lufenuron was comparable to the control.  相似文献

Abstract. 1. We determined the phenology of the shrub Spiraea latifolia Ait. Bork. (Rosaceae), which has indeterminate shoot growth, and the effects of phenological changes in leaf quality on growth rate of the early-spring feeding buckmoth caterpillars, Hemileuca lucina Hy. Edw. (Saturniidae).
2. Leaves, regardless of whether they were newly expanded or several weeks old, were tougher later in the growth season (mid-June) than similarly aged leaves collected earlier; correspondingly, water and nitrogen content for leaves of all ages declined through the larval period. By July, newly expanded leaves had no more nitrogen than mature leaves.
3. Relative growth rate of third instar larvae fed new leaves or a mixture of new and mature leaves in early June was higher than that of those fed mature leaves, and efficiency of conversion of digested food to biomass was higher for larvae fed new leaves than for those fed mature leaves or a mixture.
4. In another experiment, larvae were reared on new leaves through the fourth instar and then fed a diet of new, mature or a combination of new and mature leaves, a regimen that was similar to the phenologies of both plants and caterpillars in the field. There was no difference in time to pupation or pupal weights among these treatments.  相似文献

Beauveria bassiana endophytically colonises corn (Zea mays) reducing tunneling from European corn borer (Ostrinia nubilalis). Endophytic colonisation of other plants by B. bassiana has been reported, and potentially, may reduce insect feeding on these plants. We evaluated the effects on larval growth and development, and mortality of different rates of dried, ground mycelia and water-soluble metabolites from fermentation broth culture of different isolates of B. bassiana incorporated into a synthetic diet and fed to neonate bollworm, Helicoverpa zea larvae. Development was delayed, weights of larvae were lower, and mortality was high for larvae fed the highest rates (1.0 and 5.0%, w/v) of mycelia incorporated diet compared to control. Insects fed diets containing mycelia of B. bassiana isolate 11-98 had the greatest mortality. Mortality was 100% for larvae fed 5% (w/v) mycelia incorporated diet of isolate 11-98, and 61% for isolate 3-00. For insects fed low rates (0.1 to 0.5%, w/v) of mycelia incorporated diet, mortality was lower, approximately 5% for isolate 11-98, and 5 to 14% for isolate 3-00. At the 0.1% (w/v) rate of mycelia incorporated diet, development occurred at an accelerated rate, compared to fungus-free controls, indicating increased nutrition in the lowest rate fungal diet. Mortality was low for all larvae fed diets containing spent fermentation broth of B. bassiana; however, development was delayed. Insects fed the highest rate (0.5%, v/v) of spent fermentation broth-amended diet had lower pupal weights, and a greater number of days to pupation than insects fed the lowest (0.1%, v/v) rate. Insects fed the 5% (v/v) rate of spent fermentation broth of isolates 11-98 and 3-00 had the longest days to pupation.  相似文献

Abstract

The juvenile hormone analogue ZR-619—ethyl 11-methoxy-3,7,11-trimethyl-(2E,4E-dodecadienethiolate) —produced morphological and physiological effects when fed via artificial diet to larvae of Epiphyas postvittana (Walker). Morphological effects included changes in larval head and antennal structures after instar V (supernumerary instars); deformation of pupal and adult structures, particularly in genitalia and wings; and development of individuals with varying mixtures of larval and pupal structures. Physiological effects included prolongation of larval life, increase in larval weight, increase in larval instars, and decrease in fecundity. Effects were directly related to dosage concentrations.  相似文献

Urea concentration and urease activity in the midgut content were compared between larvae of the silkworm, Bombyx mori fed an artificial diet and those fed fresh mulberry leaves. A considerable amount of urea was found in the midgut content of the both larvae, however it was significantly lower in the larvae fed fresh mulberry leaves than in the larvae fed the artificial diet; average urea concentrations in the midgut content of the larvae fed fresh mulberry leaves and the artificial diet were 2.9 and 4.6 &mgr;mol/g, respectively. Urea in the midgut content seems to be secreted from the insect itself since the amount of urea in both diets were negligibly small. Urease activity was detected only in the midgut content of the larvae fed fresh mulberry leaves but not in other tissues of the larvae. On the other hand, no urease activity was detected in the midgut content of the larvae fed the artificial diet. Subsequently, to elucidate the role of mulberry leaf urease in the midgut lumen, larvae that had been reared on the artificial diet were switched to fresh mulberry leaves. The diet switch caused a rapid decrease in urea concentration in the midgut content and an increase in ammonia concentration in the midgut content, suggesting that secreted urea could be hydrolyzed to ammonia by mulberry leaf urease in the midgut lumen. Furthermore, to investigate the physiological significance of mulberry leaf urease on urea metabolism of the silkworm, (15)N-urea was injected into the hemocoel, and after 12 h the larvae were dissected for (15)N analysis. A considerable amount of (15)N was found to be incorporated into the silk-protein of the larvae fed fresh mulberry leaves, but there was little incorporation of (15)N into the silk-protein of the larvae fed the artificial diet. These data indicate that urea is converted into ammonia by the action of mulberry leaf urease in the midgut lumen and used as a nitrogen source in larvae fed mulberry leaves.  相似文献

The susceptible species Cedrela odorata and Swietenia macrophylla to attack by Hypsipyla grandella (Zeller) larvae were grafted onto the resistant species Khaya senegalensis and Toona ciliata. Six-month-old grafted plants were then compared to their reciprocal grafts and to both intact (non-grafted) and autografted plants for damage due to H. grandella larvae and for their effects on larval performance. Two experiments were conducted: one in which the apical bud of the main plant shoot was inoculated with H. grandella eggs, and the other in which the bud was inoculated with third instars. Damage in each experiment was assessed by the number of frass piles, number and length of tunnels, number of damaged leaves, and damage to the apical bud. Larval performance was evaluated in terms of time to reach pupation and pupal weight and length. In both experiments, plant damage differed significantly among treatments (P < 0.03). Resistant rootstocks conferred resistance to susceptible scions. In both experiments, grafting by itself, regardless of the rootstock and scion combination, also reduced damage caused by H. grandella larvae. Scions of autografted susceptible species had similar resistance to susceptible scions grafted on resistant rootstocks. Few larvae reached pupation, and their pupal weight and length were similar.  相似文献

The effects of the lipase inhibitor, tetrahydrolipstatin (THL), on neonate Epiphyas postvittana (Walker) (Lepidoptera, Tortricidae) larvae were investigated by feeding on control artificial diets (with and without 2% ethanol) and diets containing 2% ethanol and one of three concentrations of THL (0.011%, 0.037% and 0.11%). Small but significant reductions in growth rate, percent pupation and time to pupation were observed for larvae feeding on 2% ethanol control diet compared with standard control diet, but larger reductions in all parameters occurred with increasing THL concentration. Third instar larvae fed 0.011% THL in the diet had 40% of the midgut lipase activity in the relevant control larvae and showed up-regulation of gene expression of the gastric lipase-like family but not the pancreatic lipase-like family of midgut lipases.  相似文献

This study was conducted to determine the effects of Bt cotton leaves (Bollgard II), non-Bt cotton leaves, and a mixture of Bt+non-Bt cotton leaves on larval orientation behavior, survival and development of Trichoplusia ni in the laboratory. Results indicate that in a no-choice test, more first and fifth instars remained on Bt leaves than the third instars. All larvae that remained on the leaves gradually moved to leaf edge. In the choice between a Bt and a non-Bt leaf, more first instars moved to non-Bt leaves, whereas the third and fifth instars did not show significant difference in the first 8 h, but eventually more moved to non-Bt leaves. More first instars fed non-Bt leaves than third instars and fifth instars. When larvae fed Bt leaves, 100% of first instars, 92.7% of third instars and 51.1% of fifth instars died in 108 h. Once larvae pupated, >90% developed to adults. First and third instars that fed Bt leaves developed slower but their pupae developed faster than those on Bt+non-Bt leaves, whereas fifth instars developed similar on the three types of leaves. First and third instars that fed Bt+non-Bt leaves resulted in less heavy pupae than those fed non-Bt leaves; whereas the fifth instars that survived on Bt leaves produced lighter pupae.  相似文献
