报道一例我国典型的HIV感染家庭,其中对新生儿通过垂直传播感染的早期诊断为我国首例报道。父亲通过静脉吸毒感染HIV,经性途径传播给妻子,并通过母婴途径传播至婴儿,导致婴儿发病。通过血清学、PCR检测和病毒培养确认为整个家庭的感染。序列分析结果强烈支持杂来自同一毒株,流行背景主实了上述感染的传播途径。  相似文献

研究了栗疫病菌[Cryphonectria parasitica(Murr.)Barr]的营养体亲和性基因及dsRNA病毒对菌株间病毒特征传播的影响。试验选用已知4个VC基因座位的15个Vc基因型菌株和3种dsRNA病毒,通过含病毒菌株与野生型菌株的配对培养,将病毒逐个转入不同Vc基因型菌株。将不同Vc基因型的含病毒菌株与具特定VC基因差异的野生型菌株配对培养,根据培养两周后野生型菌株培养性状的改变与否,统计菌株间的病毒传播率。结果表明,各个VC基因对菌株间病毒传播的影响不同;存在2个VC基因差异的菌株间病毒传播率低于相差1个VC基因的:病毒在相差1个VC基因的菌株间的传播多具单向性:不同类型的病毒在菌株间的传播率也有差异。  相似文献

RNAi是指通过双链RNA介导特异性降解靶mRNA,导致转录后水平基因沉默的现象。其作用途径有RdRP依赖的RNAi的途径与非RdRP依赖的RNAi途径2种。利用RNAi的基因敲除技术在dsRNA序列选择、质粒或病毒为载体的dsRNA体内合成、发夹样siRNA的转录、dsRNA的导入方法等方面取得了很大进展,在研究人类或其他生物基因组中未知基因及蛋白质的功能等领域具有诱人的应用前景。  相似文献

将国内青霉素产生菌(Penicillium chrysogenum)的黄孢子系统及绿孢子(包括淡绿,灰绿)系统的十多个菌株,经过病毒提取、电镜观察、奥氏免疫双扩散、凝胶电泳及放射免疫测定,证明黄孢子系统的菌株含有不同滴定度的、直径40nm的球形病毒,而绿孢子系统中检查不出病毒。从营养要求、孢子颜色不同的带病毒和无病毒菌体中分离原生质体,进行不同组合的原生质体的融合杂交,获得营养互补融合的异核体。异核体1中,病毒通过胞质融合转移到原来无病毒的灰绿孢子菌株及细胞核融合后的杂合二倍体中。灰绿孢子的病毒量接近二倍体的1/3。二倍体菌落生长稳定,低温保存二年后经0.01—0.02M对氟苯丙氨酸(PFA)诱发和分离,产生亲本类型的分离子,分离子及二倍体仍然含有病毒。异核体2作亲本性分离,黄孢子仍有病毒,淡绿孢子及细胞核融合后产生的二倍体均无病毒,表明非感染性为显性。此种淡绿孢子的突变体中存在非感病菌系,它不支持病毒的复制。提取各杂交组二倍体内的病毒所特有的dsRNA时,可看出dsRNA的存在和病毒的存在一致。多数杂合二倍体的青霉素产量比亲本高。  相似文献

在室内观察了小菜蛾颗粒体病毒PlutellaxylostellaGranulosisVirus(RxGV)在小菜蛾种群水平上可能的垂直传播途径。结果表明 :(1 )上代小菜蛾Plutellaxylostella(L .)幼虫期饲毒不能将小菜蛾颗粒体病垂直传递到下代 ;(2 )取食病毒的雌蛾产的卵所孵化的幼虫中 ,没病毒感染出现 ,说明病毒不能经小菜蛾成虫体内传播到下一代。 (3 )用小菜蛾颗粒体病毒处理卵卡时 ,其初孵幼虫有少量被病毒感染死亡 ,浓度高时感染率也较高 ,但感染率均在 1 0 %以下 ,说明小菜蛾颗粒体病毒能经卵表垂直传递给下代 ,但传递效率不高。但当用病毒处理叶片上的卵时 ,初孵幼虫死亡率高达 77 73 % ,而且孵化出的幼虫大多数染病且死于 3龄以前 ,因此田间病毒的施用时间可提早到卵高峰期。  相似文献

以两种浓度的病毒液感染甜菜夜蛾4龄、5龄幼虫,以研究甜菜夜蛾核型多角体病毒从亲代到子代垂直传递的方式和效率。实验结果表明,该病毒可通过母体介导(卵内、卵表)和父体介导等3种方式传递给子代,造成子代感染。幼虫经口感染后母体介导传递实验表明,病毒垂直传播可引起20%~48%的子代幼虫病死,并能使子代成虫产卵量下降;母体介导传播实验中,以5%福尔马林进行卵表面消毒证实仍有18.8%的幼虫感染病毒死亡,表明了该病毒可经卵内传播,并同时存在着附卵传播;母体介导传递实验还表明病毒对宿主有弱化作用,亲代免于病死的蛹重量减轻,羽化所得雌成虫的产卵量下降,产卵期延长。以交互配对将感染与未感染的雌雄成虫彼此交配,表明雄成虫精子也能垂直传递病毒(父体介导),引起20%~40%子代幼虫感病死亡。成虫经口感染,对所产卵块表面消毒后,仍导致28.6%的子代幼虫死于病毒感染,也表明存在卵内传播。  相似文献

为明确棉花粉蚧Phenacoccus solenopsis对扶桑Hibiscus rosa-sinensis感染的木尔坦棉花曲叶病毒(CLCuMV)的传播风险,本文应用PCR方法研究了棉花粉蚧获取、存留和传播CLCuMV的规律和能力。结果表明棉花粉蚧各取食危害虫期均可获取、携带CLCuMV,获取病毒所需时间为2-48 h,虫体内病毒最多可存留312 h;但是,粉蚧体内病毒无法侵染健康扶桑,亦不会经卵或者交配进行垂直和水平传播。本研究证明了棉花粉蚧不能传播CLCuMV,为防治该虫、该病毒病提供了依据。  相似文献

Transmissions of plant viruses between individuals of their vector insects through mating are rare events. Recently, three begomoviruses were found to be transmitted between males and females of the whitefly Bemisia tabaci through mating, and two viruses were shown to be transmitted transovarially to progeny. However, results between reports were not consistent. Here we examined the horizontal and vertical transmission of Tomato yellow leaf curl virus (TYLCV) and Tomato yellow leaf curl China virus (TYLCCNV) by the B and Q biotypes of B. tabaci, using virus isolates and whitefly colonies established recently in China. Both TYLCV DNA and TYLCCNV DNA were shown to be transmitted horizontally and vertically by each of the two biotypes of the whitefly, but frequency of transmission was usually low. In transovarial transmission, virus DNA was detected in eggs and nymphs but not in the adults of the first generation progeny, except in the combination of TYLCV and Q biotype whitefly where 2–3% of the offspring adults contained the virus DNA. We also showed that the first generation adults, which developed from eggs of viruliferous whiteflies, were not infective to plants. These results demonstrated that for the viruses and whiteflies tested here low frequency of horizontal and vertical transmission can be expected but these two modes of transmission are unlikely to have much epidemiological relevance in the field.  相似文献

The microsporidian parasite Edhazardia aedis is capable of vertical or horizontal transmission among individuals of its host, the mosquito Aedes aegypti, and either mode of transmission may follow the other. We show that following the horizontal infection of host larvae, the parasite's subsequent mode of transmission largely depends on host life history traits and their responses to different environmental conditions. In two experiments the intensity of larval exposure to infection and the amount of food available to them were simultaneously manipulated. One experiment followed the dynamics of host development and the parasite's production of spores while the other estimated the outcome of their relationship. Host life history traits varied widely across treatment conditions while those of the parasite did not. Of particular importance was the host's larval growth rate. Horizontal rather than vertical transmission by the parasite was more likely as low food and high dose conditions favoured slower larval growth rates. This pattern of transmission behaviour with host growth rate can be considered in terms of reproductive value: the potential vertical transmission success that female mosquitoes offer the parasite decreases as larval growth rates slow and makes them more attractive to exploitation for horizontal transmission (requiring host mortality). However, the lack of variation in the parasite's life history traits gave rise in some conditions to low estimates for both its vertical and horizontal transmission success. We suggest that the unresponsive behaviour of the parasite's life history traits reflects a bet-hedging strategy to reduce variance in its overall transmission success in the unpredictable environmental conditions and host larval growth rates that this parasite encounters in nature.  相似文献

褐飞虱Nilaparvata lugens Stl是我国重要的迁飞性水稻害虫,本文研究了金龟子绿僵菌Metarhizium anisopliae及其与dsRNA混合使用对褐飞虱的防治效果。绿僵菌悬浮液1.6×108孢子/m L至8×106孢子/m L对褐飞虱2龄、4龄和成虫进行喷药,发现1.6×107孢子/m L对各个虫态虫龄均有良好致死效果,并且成虫和4龄若虫均好于2龄若虫。在交配行为上来看,绿僵菌处理过的褐飞虱成虫活跃度非常低,从配对开始一直到交配结束的各个阶段都受到明显影响,处理组3 h的交配率只有3.70%,而对照组的交配率为24.44%。还把褐飞虱几丁质合成酶基因A的dsRNA与绿僵菌混合使用防治褐飞虱2龄和4龄若虫,结果表明0.5μg/μL ds CHSA与绿僵菌混合使用的防治效果最好,2龄若虫的死亡率为89.63%,4龄若虫的死亡率达到93.94%。而0.2μg/μL ds CHSA与绿僵菌的混合,对2龄和4龄若虫的致死率为65.56%-76.52%。研究结果为褐飞虱的生物防治提供了新的思路。  相似文献

Honeybees (Apis mellifera) can be attacked by many eukaryotic parasites, and bacterial as well as viral pathogens. Especially in combination with the ectoparasitic mite Varroa destructor, viral honeybee diseases are becoming a major problem in apiculture, causing economic losses worldwide. Several horizontal transmission routes are described for some honeybee viruses. Here, we report for the first time the detection of viral sequences in semen of honeybee drones suggesting mating as another horizontal and/or vertical route of virus transmission. Since artificial insemination and controlled mating is widely used in honeybee breeding, the impact of our findings for disease transmission is discussed.  相似文献

Various instars of Choristoneura occidentalis were fed with a range of doses of Nosema fumiferanae and reared at 20, 24 and 28 degrees C to determine the influence of temperature and dose on the time to spore egestion and the number of spores egested in the frass. When larvae were fed in the third stadium, as few as 10(2) spores per larva initiated infection, and both onset of spore egestion and the number of spores egested were affected by a complex relationship between temperature and inoculation dose. Onset of spore egestion varied from 11 to 15 days postinoculation. At 20 degrees C, the onset was delayed and spore production decreased with increasing inoculation dose whereas at higher temperatures spores were first egested at the lowest dose and spore production increased with dose. When larvae were fed spores in the fifth and sixth stadium, no spores were egested because pupation occurred before completion of the incubation period. To assess the effect of temperature on horizontal transmission, Choristoneura fumiferana larvae fed with 10(4) N. fumiferanae spores per larva were reared with uninfected larvae at 15, 20 and 25 degrees C. At 15 degrees C, we observed the highest degree of horizontal transmission, defined by the largest change in N. fumiferanae prevalence, even though the density of spores available for horizontal transmission was the lowest. Infected adults eclosed later than uninfected adults and the time to eclosion was also dependent on sex and temperature. We relate our experimental findings to consequences for horizontal and vertical transmission of N. fumiferanae in spruce budworm populations.  相似文献

【目的】烟粉虱 Bemisia tabaci 是番茄黄曲叶病毒(Tomato yellow leaf curl virus, TYLCV)在自然界的唯一传播媒介,除了可以直接取食获取TYLCV,烟粉虱还可以通过交配获取此植物病毒。虽然前人研究证明了烟粉虱Middle East-Asia Minor 1 (MEAM1)和Mediterranean (MED)隐种都可以通过交配在两性个体之间水平传播TYLCV,但有关MEAM1与MED隐种水平传播TYLCV的能力是否存在差异,不同研究的结果却并不一致。另外,目前尚无关于烟粉虱的水平传播行为是否有助于TYLCV在田间扩散的相关研究。【方法】从浙江、广东、云南和河南4个省份的田间采集MEAM1和MED隐种烟粉虱种群,在室内应用分子标记对各种群所属隐种鉴定后分别建立7个供试种群,然后观察每个种群内带毒成虫与不带毒成虫通过交配在不同性别个体之间水平传播TYLCV的能力,并选用采自广东的MEAM1隐种烟粉虱模拟群体交配实验探究通过交配获毒的烟粉虱个体是否具有传播TYLCV致健康番茄植株发病的能力。【结果】4个省内的MEAM1和MED隐种烟粉虱都可通过交配对TYLCV进行水平传播,但传播频率一般在10%以下。不同省份的MEAM1隐种种群之间在水平传播TYLCV的能力上无显著差异,不同省份的MED隐种种群之间也不存在显著差异。另外,同一省份的MEAM1隐种与MED隐种之间在水平传播TYLCV的能力上也不存在显著差异,而且在两个隐种中,带毒雄虫将病毒水平传播给不带毒雌虫与带毒雌虫将病毒水平传播给不带毒雄虫的频率没有显著差异。研究还表明,采自广东的MEAM1隐种烟粉虱个体通过交配水平获毒后不能致健康番茄植株发病。【结论】综合以上结果,我们推测TYLCV在中国境内两个入侵烟粉虱隐种MEAM1和MED各自种群内个体之间的水平传播概率较低,对该病毒在田间的扩散可能没有作用或作用不大。  相似文献

The most crucial stage in the dynamics of virus infections is the mode of virus transmission. In general, transmission of viruses can occur through two pathways: horizontal and vertical transmission. In horizontal transmission, viruses are transmitted among individuals of the same generation, while vertical transmission occurs from mothers to their offspring. Because of its highly organized social structure and crowded population density, the honey bee colony represents a risky environment for the spread of disease infection. Like other plant and animal viruses, bee viruses use different survival strategies, including utilization of both horizontal and vertical routes, to transmit and maintain levels in a host population. In this review, we explore the current knowledge about the honey bee viruses and transmission routes of bee viruses. In addition, different transmission strategies on the persistence and dynamics of host-pathogen interactions are also discussed.  相似文献

The idea that vertical transmission of parasites selects for lower virulence is widely accepted. However, little theoretical work has considered the evolution of virulence for parasites with mixed horizontal plus vertical transmission. Many human, animal, and plant parasites are transmitted both vertically and horizontally, and some horizontal transmission is generally necessary to maintain parasites at all. We present a population-dynamical model for the evolution of virulence when both vertical and horizontal transmission are present. In the simplest such model, up to two infectious strains can coexist within one host population. Virulent, vertically transmitted pathogens can persist in a population when they provide protection against more virulent, horizontally transmitted strains. When virulence is maintained by a correlation with horizontal transmission rates, increased levels of vertical transmission always lower the evolutionarily stable (ESS) level of virulence. Contrary to existing theory, however, increases in opportunities for horizontal transmission also lower the ESS level of virulence. We explain these findings in light of earlier work and confirm them in simulations including imperfect vertical transmission. We describe further simulations, in which both vertical and horizontal transmission rates are allowed to evolve. The outcome of these simulations depends on whether high levels of vertical transmission are possible with low virulence. Finally, we argue against the notion of a virulence-avirulence continuum between horizontal and vertical transmission, and discuss our results in relation to empirical studies of transmission and virulence.  相似文献

It has been hypothesized that there is a fundamental conflict between horizontal (infectious) and vertical (intergenerational) modes of parasite transmission. Activities of a parasite that increase its rate of infectious transmission are presumed to reduce its host's fitness. This reduction in host fitness impedes vertical transmission of the parasite and causes a tradeoff between horizontal and vertical transmission. Given this tradeoff, and assuming no multiple infections (no within-host competition among parasites), a simple model predicts that the density of uninfected hosts in the environment should determine the optimum balance between modes of parasite transmission. When susceptible hosts are abundant, selection should favor increased rates of horizontal transfer, even at the expense of reduced vertical transmission. Conversely, when hosts are rare, selection should favor increased vertical transmission even at the expense of lower horizontal transfer. We tested the tradeoff hypothesis and these evolutionary predictions using conjugative plasmids and the bacteria that they infect. Plasmids were allowed to evolve for 500 generations in environments with different densities of susceptible hosts. The plasmid's rate of horizontal transfer by conjugation increased at the expense of host fitness, indicating a tradeoff between horizontal and vertical transmission. Also, reductions in conjugation rate repeatedly coincided with the loss of a particular plasmid-encoded antibiotic resistance gene. However, susceptible host density had no significant effect on the evolution of horizontal versus vertical modes of plasmid transmission. We consider several possible explanations for the failure to observe such an effect.  相似文献

A very restricted amount of high-molecular-weight double-stranded RNA (dsRNA) has been found in healthy japonica rice plants. We discriminated dsRNA-carrying rice plants from noncarriers. The endogenous dsRNA was localized in the cytoplasm (about 100 copies per cell) and was transmissible to progeny plants by mating. In crosses between carriers and noncarriers, the RNA was transmitted efficiently to F1 plants via both egg and pollen. The rice dsRNA was maintained at an almost constant level by host plant cells from generation to generation. The high-efficiency transmission of the endogenous dsRNA to progeny plants appears to depend on the autonomously controlled replication of the dsRNA localized in cytoplasmic vesicles. However, an increase in copy number (about 10-fold) of the dsRNA was observed during the suspension culture of host cells. The number of copies of dsRNA returned to the original low value in regenerated plants, suggesting that the copy number is stringently and developmentally regulated in rice cells.  相似文献
