Isolates of Magnaporthe grisea causing gray leaf spot on rice were collected in Argentina and analyzed for mating distribution and fertility. One hundred and twenty-five isolates of M. grisea were collected from rice plants between 2000 and 2003. Each isolate was tested for mating type through a polymerase chain reaction based assay. All M. grisea isolates from Argentina belonged to a single mating type, MAT1.1. The fertility status of isolates was determined using controlled crosses in vitro, pairing each isolate with GUY11 and KA9 (MAT1.2 standard hermaphroditic testers). Production of perithecia was scarce among isolates of the blast pathogen since a low percentage of them (7.2%) developed perithecia with only one of the fertile tester (KA9); all crosses failed with the other tester strain. Asci and ascospores were not observed. The presence of only one mating type and the absence of female fertile isolates indicate that sexual reproduction is rare or absent in M. grisea populations associated with rice in Argentina.  相似文献

  • 1 The success of mating disruption using synthetic sex pheromones depends not only on preventing mating, but also on delaying mating in the target insect. Using the geometrid pest of Eucalyptus plantations, Mnesampela privata (Guenée), we determined the effect of delaying mating when imposed on males only, females only or on both sexes simultaneously, for 1, 3, 5 and 7 days.
  • 2 Delayed mating had a significant negative impact on reproduction, with a 0.89‐fold decrease in the likelihood of mating and a 0.67‐fold decrease in the likelihood of that mating resulting in fertile eggs for every day that mating is delayed. A mating delay of 7 days reduced the mean number of viable eggs laid to 4–13% of that laid by moths paired immediately after emergence.
  • 3 Male only imposed mating delays had a significantly lower effect on reducing the likelihood of pairs mating than when both sexes were delayed. A delay imposed on one sex only or on both sexes simultaneously, however, had a similar negative impact on the proportion of fertile matings as well as on the total number of fertile eggs laid.
  • 4 Longevity of mated female and male M. privata was significantly different between mating delay treatments, with a significant decline in female longevity when they mated with older males.
  • 5 The underlying mechanisms causing a decline in female reproductive output when a mating delay was imposed on males versus females are discussed in relation to the reproductive biology of M. privata and the potential of using mating disruption strategies to control populations in Eucalyptus plantations.

Sexual reproduction may be cryptic or facultative in fungi and therefore difficult to detect. Magnaporthe oryzae, which causes blast, the most damaging fungal disease of rice, is thought to originate from southeast Asia. It reproduces asexually in all rice‐growing regions. Sexual reproduction has been suspected in limited areas of southeast Asia, but has never been demonstrated in contemporary populations. We characterized several M. oryzae populations worldwide both biologically and genetically, to identify candidate populations for sexual reproduction. The sexual cycle of M. oryzae requires two strains of opposite mating types, at least one of which is female‐fertile, to come into contact. In one Chinese population, the two mating types were found to be present at similar frequencies and almost all strains were female‐fertile. Compatible strains from this population completed the sexual cycle in vitro and produced viable progenies. Genotypic richness and linkage disequilibrium data also supported the existence of sexual reproduction in this population. We resampled this population the following year, and the data obtained confirmed the presence of all the biological and genetic characteristics of sexual reproduction. In particular, a considerable genetic reshuffling of alleles was observed between the 2 years. Computer simulations confirmed that the observed genetic characteristics were unlikely to have arisen in the absence of recombination. We therefore concluded that a contemporary population of M. oryzae, pathogenic on rice, reproduces sexually in natura in southeast Asia. Our findings provide evidence for the loss of sexual reproduction by a fungal plant pathogen outside its centre of origin.  相似文献

Summary The A mating type factor of Coprinus cinereus regulates part of a developmental sequence that leads to the conversion of the asexual monokaryon into the fertile dikaryon. The A42 factor is a complex of seven genes, at least four of which are involved in determining the specificity of mating interactions. In this report we show that the A42 genes are constitutively expressed in both monokaryons and dikaryons. This has important implications with respect to intracellular recognition of a compatible mating, which requires an interaction between proteins already present within the cells of the mating partners, and for the subsequent maintenance of dikaryotic growth.  相似文献

Drosophila suzukii (Matsumura) (Diptera: Drosophilidae) is a widely distributed pest species of soft-skinned fruits. Recent studies suggest the use of sterile insect technique (SIT) as a control method for this species; however, many factors can impact effectiveness of a SIT programme, including the environmental conditions. Environmental condition is critical at the time of the release and in the days afterwards, since it may impact sterile insects’ survival and ability to mate. Thus, we verified the influence of temperature and relative humidity on mating and survival of fertile and sterile D. suzukii, when insects were food provided or deprived. Highest mating rates occurred when sterile or fertile flies provided with food were exposed to 25ºC or 81%–100% relative humidity, while temperatures of 10 and 35ºC and humidity below 60% impaired mating. Overall, mating rate among food-deprived flies was low in all temperatures and humidity levels tested, but fertile insects were more prone to mate when compared to sterile flies. Survival was negatively influenced by high temperatures, low relative humidity and food deprivation. The information present in this study is useful to be considered for release of sterile D. suzukii.  相似文献

Prior work has demonstrated significant phenotypic benefits to female promiscuity in the arctiid moth Utetheisa ornatrix. We were interested in determining whether U. ornatrix females also derive genetic benefits from mating multiply. We specifically tested whether, by mating with several males, females are able to exploit postcopulatory mechanisms that decrease the risk of fertilization by incompatible sperm. We show evidence that U. ornatrix females are not taking multiple mates as fertilization insurance because: (1) females that mate once are as fertile as those that mate three times; and (2) females that take three different mates are no more fertile than those that mate three times with the same male.  相似文献

Crosses were made between three closely related species of the planthopper genus Muellerianella, M. brevipennis, M. extrusa and M. fairmairei. Most hybrids are produced in crosses between M. extrusa and M. fairmairei. Most of the hybrid females in these crosses and some of the hybrid males are fertile and can be successfully backcrossed with the parental species. Crosses of M. brevipennis with either M. fairmairei or M. extrusa yield less progeny, of which all males are sterile. Backcrosses of the hybrid females with the parental species are partly successful. The possible evolutionary consequences of mating readiness and interfertility between the three species under field conditions are discussed. Efforts to resynthesize triploid gynogenetic forms by backcrossing hybrid females of M. fairmairei and M. brevipennis with males of M. fairmairei, as was reported by Drosopoulos (Evolution 32: 916–920, 1978), failed. To produce offspring, the existing gynogenetic forms have to mate with males of one of the three bisexual species.  相似文献

Four isolates of aSaccharomyces species which differed fromS. kluyveri by their ability to use cellobiose were analyzed genetically in relation to the latter species. Isolated single spores had low viability. Spore tetrads segregated mating types 2 2, with sexual agglutination occurring between complementary mating types. All single-spore isolates assimilated cellobiose indicating that these isolates were not naturally occurring hybrids betweenS. kluyveri and a cellobiose assimilatingSaccharomyces species.Two cell types were exhibited by single-spore cultures ofS. kluyveri, one granulated (G-type) and one vacuolated (g-type). G-type cultures formed fertile hybrids with complementary mating types of both G- and g-type cultures. Hybrids between two g-type cultures were sterile. They would, however, give fertile hybrids when mixed with G-type cultures.Sporulating hybrids betweenSaccharomyces sp. andS. kluyveri were produced. However the percentage spore germination was low. Single-spore cultures examined had cell types atypical of either parent. The ability to assimilate cellobiose was dominant and appeared to segregate with mating type and cell type.Weak mating reactions occurred when the (+) and (-) mating types ofSaccharomyces sp. were mixed with (a) and () mating types ofS. cerevisiae, respectively.The species ofSaccharomyces isolated from the Pacific Coast are designated as strains ofS. kluyveri.  相似文献

Pineapple (Ananas comosus) is one the important fruit crops planted in Malaysia, and this study was conducted to determine Fusarium spp. associated with diseases of the fruit crop as Fusarium is prevalent in tropical countries. Our objective was to identify and characterize Fusarium spp. associated with pineapple fruit rot and leaf spot mainly found on the fruits and leaves in Peninsular Malaysia. Fusarium isolates (n = 108) associated with pineapple fruit rot and leaf spot were characterized by morphological, molecular and phylogenetic analyses, a mating study and pathogenicity testing. TEF‐1α sequence analysis identified Fusarium proliferatum, Fusarium verticillioides, Fusarium sacchari and Fusarium sp. Mating was successful only between tester strains of F. proliferatum and F. verticillioides. Sexual crosses with standard tester strains showed that 82 isolates of F. proliferatum produced fertile crosses with mating population D (Gibberella intermedia) and three isolates of F. verticillioides were fertile with the tester strain of mating population A (Gibberella moniliformis). All isolates were pathogenic, causing pineapple fruit rot and leaf spot, thus fulfilling Koch's postulates.  相似文献

Female remating is a widespread behaviour, reported in several insect species. This behaviour can affect the efficiency of sterile insect technique (SIT); however, little is known about the postcopulatory behaviour of some pest species considered as candidates to be controlled by this technique, such as Drosophila suzukii (Matsumura, 1931) (Diptera: Drosophilidae). In this study, we investigated the effects of male and female sterilization on mating and remating behaviour of D. suzukii. First, we tested the occurrence of multiple mating in different combinations between sterile and fertile males and females. Then, we tested the effects of male and female sterility on female propensity to mate and remate. We found an overall low remating rate by D. suzukii females. Male sterility did not influence mating and remating likelihood; however, copula duration of sterile males was shorter compared to fertile males. On the other hand, sterile females were less likely to mate. Our findings encourage further research regarding the use of SIT to control D. suzukii.  相似文献

Sex in fungi is driven by peptide pheromones sensed through seven‐transmembrane pheromone receptors. In Cryptococcus neoformans, sexual reproduction occurs through an outcrossing/heterothallic a ‐ sexual cycle or an inbreeding/homothallic – unisexual mating process. Pheromone receptors encoded by the mating‐type locus ( MAT ) mediate reciprocal pheromone sensing during opposite‐sex mating and contribute to but are not essential for unisexual mating. A pheromone receptor‐like gene, CPR2 , was discovered that is not encoded by MAT and whose expression is induced during a ‐ mating. cpr2 mutants are fertile but have a fusion defect and produce abnormal hyphal structures, whereas CPR2 overexpression elicits unisexual reproduction. When heterologously expressed in Saccharomyces cerevisiae , Cpr2 activates pheromone responses in the absence of any ligand. This constitutive activity results from an unconventional residue, Leu222, in place of a conserved proline in transmembrane domain six; a Cpr2L222P mutant is no longer constitutively active. Cpr2 engages the same G‐protein activated signalling cascade as the Ste3 a /α pheromone receptors, and thereby competes for pathway activation. This study established a new paradigm in which a naturally occurring constitutively active G protein‐coupled receptor governs morphogenesis in fungi.  相似文献

Ageing can reduce the probability that individuals reproduce. The present study investigates whether ageing influences the mating frequency of mass‐reared fertile and sterile Mexican fruit flies Anastrepha ludens (Loew). The ability of males of different ages to inhibit female remating is also determined, and the growth of male reproductive organs is measured as they age. Young males (6 days old) have a lower mating frequency than older males, and also have a lower capacity to inhibit female remating than older males. However, 7‐day‐old males are as likely to inhibit female remating as older males. Young males also have smaller reproductive organs than middle‐aged (21‐day‐old) or senescent males (57‐day‐old). These results have implications for the sterile insect technique because sterilized males of A. ludens are released in the field 6 days after emergence. The highest mating frequency, the lowest mating latency and the largest size of testes are observed at 21 days of age. Older males (57 days old) have more sperm in their seminal vesicles than young males (6 and 9 days old). Accessory glands take longer to grow to their complete size compared with testes, and mating frequency is more closely associated with accessory gland size than testes size. Furthermore, there are more sperm in the seminal vesicles during the afternoon period of peak sexual activity than during the morning when sexual activity is absent. These results indicate that, even at the onset of reproductive senescence, mass‐reared males of A. ludens are still capable of mating, as well as inhibiting remating in females.  相似文献

刺孢小克银汉霉[Cunninghamella echinulata(Thaxt.) Thaxt. ex Blakeslce]是小克银汉霉属的模式种,多年来因不同作者是否将贝尼尔小克银汉霉(C. bainieri Naum.)包含在内而对此种持广义(sensu lato)或狭义(scnsu stricto)的不同理解。我们在国内分离到51株刺孢小克银汉霉(广义)菌种,经与获自css的7株菌(包括刺孢小克银汉霉的模式菌种在内)和IMI的1株菌(原定名称均为刺孢小克银汉霉)一起研究后,根据它们的无性型形态特征有明显不同但在配合试验中又可互相配合形成可育的接合孢子的结果,认为将它们处理为同一个种的两个不同变种较为合理.其中,13株中国的菌和3株CBS及IMI的菌被鉴定为刺孢小克银汉霉原变种(C. echinulata var. echinulata);其余的38株中国的菌和5株CBS的菌被鉴定为轮生刺孢小克银汉霉变种(新组合)[C. echinulata var. verticillata (F. S. Paine.) comb. nov.j.以‘轮生(verticillata)’作为新组合变种的加词是根据与贝尼尔小克银汉霉同物异名的‘轮生布拉小克银汉霉(C. blakesleeana var. verticillata (F. S. Paine) Baijal & B. S. Mchrotra)’而来的.在前人的工作中,对刺孢小克银汉霉持狭义理解的作者往往根据孢子枝的分枝方式、巨型暗色孢子的有无、以及40℃能否生长来区分这两个分类群.对刺孢小克银汉霉持广义理解的作者则认为这些特性毫无意义.我们同意后面两个特性意义不大,因为二者的许多菌系都能形成巨型暗色袍子并在40℃生长,但认为孢子枝分枝方式确实是区别这两个分类群的重要依据.此外,根据产孢构造的种类、孢子枝主枝的直径、孢子枝第一次分枝的数目、分枝长度、孢子枝主枝的顶生泡囊形状以及其直径等性状综合考虑,这两个分类群是不难明确区分的,对12株刺孢小克银汉霉原变种和13株轮生刺孢小克银汉霉相互之间所进行的配合试验结果表明,在具一定性亲和力的(+)、(一)菌系同时存在时,两个变种内和变种之间均可形成可育的接合孢子。对上述三种配合的各六对菌所形成的有性型形态的研究结果则表明,除轮生刺孢小克银汉霉变种内形成的接合孢子的壁往往较厚外,各种配合形成的有性型形态基本上是一致的.我们研究组的其他成员最近曾对小克银汉霉的22株菌进行了rDNA的ITS区的PCR产物长度测定,其中包括了刺孢小克银汉霉原变种和轮生刺孢小克银汉霉变种的各3株菌,它们的平均长度(bp)分别为890±7.5和915±7.6.这个结果也支持了我们在这两个分类群可以互相配合的情况下仍将它们作为两个不同变种的处理.这两个分类群虽然已被发表多次,但它们的描述和绘图均过于简单而未能充分显示出它们的区别性特征,因此我们在本文中再次作了描述并再次提供了详尽的线条图和显微照相.  相似文献

The A mating factor of Coprinus cinereus determines compatibility in mating by regulating part of a developmental sequence that leads to dikaryon formation. The A genes that trigger development encode two different classes of homeodomain proteins, and for a successful mating, a protein of one class, HD 1, must interact with a protein of the other class, HD 2. In this report we show that C. cinereus A genes that encode HD 2 proteins, a2-1 and b2-1, can elicit A-regulated development in the heterologous host C. bilanatus. Transformation rates were very low, suggesting that the genes were poorly transcribed. The fact that the HD 2 genes are functionally expressed implies successful heteromultimeric association of putative DNA-binding proteins coded by the two Coprinus species. This interaction was sufficient to satisfy the need for different A factors in the formation of a fertile C. bilanatus dikaryon, but fertile dikaryons were more readily produced in matings with the a2-1 gene transformants. The C. cinereus A genes, b1-1 and d1-1, which encode HD1 proteins, were either not expressed or their proteins were non-functional in C. bilanatus. These experiments raise some interesting questions regarding HD1–HD2 protein interactions.  相似文献

Mating behaviour is an important component of species’ life histories. Knowledge of natural patterns of mating can lead also to more effective management strategies for populations of conservation concern. Despite a high conservation profile many aspects of the biology of the common dormouse (Muscardinus avellanarius) remain unknown, potentially limiting present conservation efforts. We determine the mating behaviour of M. avellanarius at two woodland sites in the UK: (1) Bontuchel (a natural population in Wales) and (2) Wych (a population in England that was established by reintroducing captive-bred animals) by genotyping mothers and litters at a panel of 10 microsatellite loci. Adult female body weight positively correlates with litter size and no apparent reproductive skew was evident. We found that multiple mating by female dormice is prevalent at both sites, with litters containing three or more offspring sired by multiple fathers; moreover, multiple mating is adopted by released animals even after a period of captive breeding where females are mated singly or as a breeding pair. We also present evidence for low proportion of fathers identified in our samples that probably related to unsampled individuals and/or larger than anticipated population sizes. This first report of mating behaviour in M. avellanarius highlights the role of genetic studies to uncover species’ reproductive behaviours and include these data for conservation management.  相似文献

Moroccan isolates of Pyricularia grisea (Magnaporthe grisea) from Oryza sativa and Stenotaphrum secundatum were crossed with standard fertile isolates (mating type Mat 1.1 and Mat 1.2) by using the three‐points culture method on oatmeal agar. Only Mat 1.1 isolates from rice were identified and considered to have only the ability to function as males, whereas no isolate from S. secundatum mated with Mat 1.1 or Mat 1.2. Cross‐inoculation studies using Pyricularia grisea (M. grisea) isolates from Oryza sativa and S. secundatum show that rice varieties cultivated in Morocco were susceptible to isolates originating from S. secundatum. Similarly, S. secundatum was susceptible to some isolates from O. sativa.  相似文献

The aim of this 14 year study was to elucidate the entire courtship and mating behaviour of manta rays Mobula alfredi and M. birostris using behavioural observations, video and photographic records. From 2003 to 2016, over 11,000 surveys were undertaken at known manta ray aggregation sites in the Maldives to record any observed manta rays reproductive activity. From 47,591 photo‐ID sightings, 4,247 individual M. alfredi were identified and 226 individual M. birostris from 229 photo‐ID sightings, all recorded at 22 atolls across 265 different sites. Courtship activity was observed on 206 surveys at 30 different sites. A total of 229 courtship events were recorded, with 90% (n = 205) of them occurring at cleaning sites. The observed courtship activity was categorised into seven distinct stages which are described in detail: initiation, endurance, evasion, pre‐copulatory positioning, copulation, post‐copulatory holding and separation. Photographs provide the first scientific record of the entirety of manta rays courtship and mating. Both M. alfredi and M. birostris appear to engage in the same elaborate courtship rituals, exhibiting the same behaviours during all stages of the courtship and mating process.  相似文献

Sordaria macrospora is a homothallic ascomycete which is able to form fertile fruiting bodies without a mating partner. To analyze the molecular basis of homothallism and the role of mating products during fruiting body development, we have deleted the mating type gene Smta-1 encoding a high-mobility group domain (HMG) protein. The ΔSmta-1 deletion strain is morphologically wild type during vegetative growth, but it is unable to produce perithecia or ascospores. To identify genes expressed under control of Smta-1, we performed a cross-species microarray analysis using Neurospora crassa cDNA microarrays hybridized with S. macrospora targets. We identified 107 genes that are more than twofold up- or down-regulated in the mutant. Functional classification revealed that 81 genes have homologues with known or putative functions. Comparison of array data from ΔSmta-1 with those from three phenotypically similar mutants revealed that only a limited set of ten genes is deregulated in all mutants. Remarkably, the ppg2 gene encoding a putative lipopeptide pheromone is 500-fold down-regulated in the ΔSmta-1 mutant while in all other sterile mutants this gene is up-regulated. The cross between two sibling species,D. simulans andD. mauritiana, produced F1 fertile females and sterile males. Backcrosses of these females to parental males resulted in the production of a small proportion of fertile males. From this second generation, four lines were propagated by mass mating and the frequency of fertile males was determined up to the 20th generation. In all cases, a linear increase of the proportion of fertile males was observed between the 2d and 8th generations and then the proportion remained stable at a level of 91%. From these observations several conclusions seem possible. (1) Male fertility was determined by several genes existing under different allelic conditions in the two sibling species; (2) the progressive restoration of male fertility was due to a normalizing selection which favored the homozygotes against the heterozygotes; (3) the persistence of a small frequency of sterile males after the 8th generation was the consequence of a balancing selection favoring the heterozygous condition (possibly in the females); (4) introgression of genes from one species into the genome of another species appeared capable of increasing the genetic heterogeneity of the population for a long time.  相似文献

Flatworms generally are simultaneous hermaphrodites that exhibit various kinds of mating behavior. Here we report on the mating behavior and reproductive biology of the planarian Paucumara falcata. We recognized three phases in its mating behavior: a courtship, copulation, and postcopulatory phase. During the last‐mentioned phase, the partners showed a unique and very characteristic behavior in which their bodies intertwined, forming a spiral. Histological study of partners in copula revealed that the sclerotic tip of the musculo‐parenchymatic organ pierces the body wall of the partner and then becomes lodged in its parenchyma, suggesting that this organ may act as an anchor, thus stabilizing the worms during copulation. Similar organs in other species of marine triclad may also perform a stabilizing role during copulation. During copulation in individuals of P. falcata, sperm transfer was reciprocal or only unilateral. Copulation duration ranged 13–35 min (average 20 ± 5 min), irrespective of whether the mating was successful (i.e., resulted in the production of fertile cocoons). The spiraling phase lasted on average 10 min; some worms did not show the postcopulatory spiraling phase during their mating behavior. After successful copulation, an individual worm produced 1–12 fertile cocoons over a period of 1–17 days; from a cocoon hatched either one young (in 70% of the cases), or two young worms.  相似文献
