Degeneration of biogenic superparamagnetic magnetite   总被引:2,自引:0,他引:2  
Magnetite crystals precipitated as a consequence of Fe(III) reduction by Shewanella algae BrY after 265 h incubation and 5-year anaerobic storage were investigated with transmission electron microscopy, Mössbauer spectroscopy and X-ray diffraction. The magnetite crystals were typically superparamagnetic with an approximate size of 13 nm. The lattice constants of the 265 h and 5-year crystals are 8.4164Å and 8.3774Å, respectively. The Mössbauer spectra indicated that the 265 h magnetite had excess Fe(II) in its crystal-chemistry (Fe3+1.990Fe2+1.015O4) but the 5-year magnetite was Fe(II)-deficient in stoichiometry (Fe3+2.388Fe2+0.419O4). Such crystal-chemical changes may be indicative of the degeneration of superparamagnetic magnetite through the aqueous oxidization of Fe(II) anaerobically, and the concomitant oxidation of the organic phases (fatty acid methyl esters) that were present during the initial formation of the magnetite. The observation of a corona structure on the aged magnetite corroborates the anaerobic oxidation of Fe(II) on the outer layers of magnetite crystals. These results suggest that there may be a possible link between the enzymatic activity of the bacteria and the stability of Fe(II)-excess magnetite, which may help explain why stable nano-magnetite grains are seldom preserved in natural environments.  相似文献

Anaeromyxobacter dehalogenans strain 2CP-C reduces U(VI) and Tc(VII) to U(IV)O2(s) (uraninite) and Tc(IV)O2(S) respectively. Kinetic studies with resting cells revealed that U(VI) or Tc(VII) reduction rates using H2 as electron donor exceeded those observed in acetate-amended incubations. The reduction of U(VI) by A. dehalogenans 2CP-C resulted in extracellular accumulation of ∼5 nm uraninite nanoparticles in association with a lectin-binding extracellular polymeric substance (EPS). The electron donor did not affect UO2(S) nanoparticle size or association with EPS, but the utilization of acetate as the source of reducing equivalents resulted in distinct UO2(S) nanoparticle aggregates that were ∼50 nm in diameter. In contrast, reduction of Tc(VII) by A. dehalogenans 2CP-C cell suspensions produced dense clusters of TcO2 particles, which were localized within the cell periplasm and on the outside of the outer membrane. In addition to direct reduction, A. dehalogenans 2CP-C cell suspensions reduced Tc(VII) indirectly via an Fe(II)-mediated mechanism. Fe(II) produced by strain 2CP-C from either ferrihydrite or Hanford Site sediment rapidly removed 99Tc(VII)O4 from solution. These findings expand our knowledge of the radionuclide reduction processes catalysed by Anaeromyxobacter spp. that may influence the fate and transport of radionuclide contaminants in the subsurface.  相似文献

The relationship between the instantaneous mortality rate ( Z ) and the instantaneous change in length‐frequency distribution of organisms per unit of animal size (μ L ) takes the following form: Z  = μ L k ( L  −  L ) +  k , where k and L are coefficients of the von Bertalanffy equation, and L is organism size (length). Z and μL change coherently when they are measured for a specific size or age class. Therefore, observations of μ L can provide information on the relative changes in mortality. This is useful when no precise information about animal growth is available and growth curve is assumed to be invariable. This method was tested on a heavily exploited population of St Peter's fish (mango tilapia) Sarotherodon galilaeus in Lake Kinneret, Israel, where large fluctuations in the size structure of the catch have occurred over the past few years. Analysis of the changes in the length‐frequency distributions showed that the changes in μ L over multiple years estimated for fully exploited fish reflected the respective changes in Z .  相似文献

By analysis of gas exchange and chlorophyll fluorescence, the effects of NaCl treatment and supplemental CaCl2 on photosynthesis, photosystem II (PSII) photochemistry and photoinhibition were investigated in Rumex leaves. Photosynthesis in Rumex leaves was strongly inhibited by 200 m M NaCl treatment. Such inhibition of photosynthesis was ameliorated by CaCl2 supplement. Neither NaCl treatment nor CaCl2 supplement had any significant effects on the PSII primary photochemical reaction in dark-adapted leaves. In light-adapted leaves, however, 200 m M NaCl treatment significantly decreased photochemical quenching (qp), efficiency of excitation energy capture by open PSII reaction centers (FV'/FM') and quantum yield of PSII electron transport (ΦPSII). These decreases in qp, FV'/FM' and ΦPSII were mitigated by CaCl2 supplement with the maximum of its effect appearing at a concentration of 8 m M CaCl2. A similar mitigating effect was shown in 200 m M NaCl-treated Rumex leaves when susceptibility of PSII to photoinhibition was determined under high irradiance. It is suggested that the mitigation of photoinhibition in NaCl-treated leaves is because of the amelioration of inhibition of photosynthesis.  相似文献

ABSTRACT Endosymbiotic methanogenic bacteria of three species of anaerobic ciliates (Plagiopyla frontata, Metopus conforms , and M. palaeformis) were inactivated with the specific methanogen inhibitor 2-bromoethanesulfonic acid. the absence of endosymbiont methanogens reduced growth rate and growth yield by about 30% in P. frontata and M. contortus , while no significant change in fitness was observed in M. palaeformis. In Plagiopyla the growth rate constant is not affected by an artificially increased pH2 neither in normal nor in methanogen-free ciliates. the energetic advantage conferred by endosymbiont methanogens in Plagiopyla and in Metopus contortus probably is due to excretion of organic material from the bacteria at the expense of bacterial reproduction. It is unlikely that the maintenance of a low pH2 within the cells due to H2-consumption by the bacteria is important to the ciliates.  相似文献

SYNOPSIS. Very similar results are obtained by the addition of 300 μg/ml chloramphenicol or of 15 μg/ml ethidium bromide to cultures of Tetrahymena pyriformis strain ST. In such cultures the exponential growth rates and the yields are reduced. Unlike the untreated ciliates, which retain the pyriform shape throughout the exponential and stationary phases, those exposed to either of the drugs become more spherical. In the control organisms progressively less surface area/unit of cell volume is exposed to the environment during the growth cycle; in the drug-treated ciliates this pattern is reversed. Increased endogenous respiration, reduced specific activities of states 3 and 4, and reduction in the levels of cytochromes a, a3, b , and c + c 1 are found in mitochondria from ciliates exposed to chloramphenicol and ethidium bromide.  相似文献

Elevation of intracellular Ca2+ concentration ([Ca2+]i) triggers exocytosis of secretory granules in pancreatic duct epithelia. In this study, we find that the signal also controls granule movement. Motions of fluorescently labeled granules stopped abruptly after a [Ca2+]i increase, kinetically coincident with formation of filamentous actin (F-actin) in the whole cytoplasm. At high resolution, the new F-actin meshwork was so dense that cellular structures of granule size appeared physically trapped in it. Depolymerization of F-actin with latrunculin B blocked both the F-actin formation and the arrest of granules. Interestingly, when monitored with total internal reflection fluorescence microscopy, the immobilized granules still moved slowly and concertedly toward the plasma membrane. This group translocation was abolished by blockers of myosin. Exocytosis measured by microamperometry suggested that formation of a dense F-actin meshwork inhibited exocytosis at small Ca2+ rises <1 μ m . Larger [Ca2+]i rises increased exocytosis because of the co-ordinate translocation of granules and fusion to the membrane. We propose that the Ca2+-dependent freezing of granules filters out weak inputs but allows exocytosis under stronger inputs by controlling granule movements.  相似文献

Routine oxygen consumption ( M o 2) was 35% higher in 1 day starved and 21% higher in 4 day starved adult transgenic coho salmon Oncorhynchus kisutch relative to end of migration ocean-ranched coho salmon. Critical swimming speed ( U crit) and M o 2 at U crit ( M o 2max) were significantly lower in 4 day starved transgenic coho salmon (1·25 BL s−1; 8·79 mg O2 kg−1 min−1) compared to ocean-ranched coho salmon (1·60 BL s−1; 9·87 mg O2 kg−1 min−1). Transgenic fish swam energetically less efficiently than ocean-ranched fish, as indicated by a poorer swimming economy at U crit ( M o 2max     ). Although M o 2max was lower in transgenic coho salmon, the excess post-exercise oxygen consumption (EPOC) measured during the first 20 min of recovery was significantly larger in transgenic coho salmon (44·1 mg O2 kg−1) compared with ocean-ranched coho salmon (34·2 mg O2 kg−1), which had a faster rate of recovery.  相似文献

The underground vole Microtus thomasi , a Balkan endemic, displays remarkable variability in sex chromosome size and morphology. In the present study, we demonstrate this variability in two of its chromosomal races with 2 n  = 44 (i.e. 'thomasi' and 'atticus') with the use of C-banding on a sample of 189 individuals from 50 localities of Greece. In 'thomasi', five different, acrocentric X chromosome variants (X0–X4) are described, which differ significantly in size, due to heterochromatin addition. Also, three Y chromosome variants are described (Y0–Y2), ranging in size from very small (Y0) to large (Y2). The 'atticus' race displays three subtelocentric variants of the X chromosome (Xst0–Xst2), which differ in arm length ratio and heterochromatin content. In Peloponnesus, males of this race exhibit Y0 and Y1, whereas, in Attiki (south-east Sterea Ellada), males carry the small metacentric, Ym. Overall, there is a trend towards sex chromosome size increase in a south to north direction. We propose that the last glaciation must have restricted M. thomasi to a refugium in southern Peloponnesus. During post-glacial colonization, limited northward expansion of its distribution area must have been accompanied by consecutive heterochromatin addition, which is proven today by comparatively larger sex chromosomes in the northern populations of 'thomasi' and 'atticus' in Greece than in their southern populations.  © 2009 The Linnean Society of London, Biological Journal of the Linnean Society , 2009, 96 , 685–695.  相似文献

Aim:  To investigate the plasma membrane damage of chlorine dioxide (ClO2) to Candida albicans ATCC10231 at or below the minimal fungicidal concentration (MFC).
Methods and Results:  ClO2 at MFC or below was adopted to treat the cell suspensions of C. albicans ATCC10231. Using transmission electron microscopy, no visible physiological alteration of cell shape and plasma membrane occurred. Potassium (K+) leakages were significant; likewise, it showed time- and dose-dependent increases. However, adenosine triphosphate (ATP) leakages were very slight. Research shows that when 99% of the cells were inactivated, the leakage was measured at 0·04% of total ATP. Compared with the mortality-specific fluorescent dye of DiBAC4(3), majority of the inactivated cells were poorly stained by propidium iodide, another mortality-specific fluorescent dye which can be traced by flow cytometry.
Conclusion:  At or below MFC, ClO2 damages the plasma membranes of C. albicans mainly by permeabilization, rather than by the disruption of their integrity. K+ leakage and the concomitant depolarization of the cell membrane are some of the critical events.
Significance and Impact of the Study:  These insights into membrane damages are helpful in understanding the action mode of ClO2.  相似文献

In the present study, glass eels Anguilla anguilla in the Minho River estuary (41·5° N, 8·5° W) decreased in size (standard length, L S and mass, M ) from the beginning (autumn) to the end of the sampling season (summer). On the other hand elvers increased in L S and M from spring to summer and were significantly larger than glass eels in paired comparisons. Branchial Na+/K+-ATPase and vacuolar (V-type) proton ATPase ( in vitro activities), two important ion transporting pumps, did not show significant seasonal changes in either glass eels or elvers although in glass eels Na+/K+-ATPase (activity) expression was significantly higher than in elvers. In a single month comparison Na+/K+-ATPase branchial mRNA expression was also higher in glass eels as was the protein level expression of both Na+/K+-ATPase and NKCC (Na+:K+:2Cl co-transporter). Immunofluorescence microscopy indicated apical CFTR Cl channel labelling in Na+/K+-ATPase positive chloride cell in glass eels which was absent in elvers. Whole body sodium concentration and percentage water did not show significant seasonal differences in either glass eels or elvers although there were significant differences between these two groups during some months.  相似文献

Cells of Geobacter metallireducens , Magnetospirillum strain AMB-1, Magnetospirillum magnetotacticum and Magnetospirillum gryphiswaldense showed N2-dependent growth, the first anaerobically with Fe(III) as the electron acceptor, and the latter three species microaerobically in semi-solid oxygen gradient cultures. Cells of the Magnetospirillum species grown with N2 under microaerobic conditions were magnetotactic and therefore produced magnetosomes. Cells of Geobacter metallireducens reduced acetylene to ethylene (11.5 ± 5.9 nmol C2H4 produced min−1 mg−1 cell protein) while growing with Fe(III) as the electron acceptor in anaerobic growth medium lacking a fixed nitrogen source. Cells of the Magnetospirillum species, grown in a semi-solid oxygen gradient medium, also reduced acetylene at comparable rates. Uncut chromosomal and fragments from endonuclease-digested chromosomal DNA from these species, as well as Geobacter sulphurreducens organisms, hybridized with a nifHDK probe from Rhodospirillum rubrum , indicating the presence of these nitrogenase structural genes in these organisms. The evidence presented here shows that members of the metal-metabolizing genera, Geobacter and Magnetospirillum , fix atmospheric dinitrogen.  相似文献

Membrane-bound [NiFe]-hydrogenase from Hydrogenophaga sp. AH-24 was purified to homogeneity. The molecular weight was estimated as 100±10 kDa, consisting of two different subunits (62 and 37 kDa). The optimal pH values for H2 oxidation and evolution were 8.0 and 4.0, respectively, and the activity ratio (H2 oxidation/H2 evolution) was 1.61 × 102 at pH 7.0. The optimal temperature was 75 °C. The enzyme was quite stable under air atmosphere (the half-life of activity was c . 48 h at 4 °C), which should be important to function in the aerobic habitat of the strain. The enzyme showed high thermal stability under anaerobic conditions, which retained full activity for over 5 h at 50 °C. The activity increased up to 2.5-fold during incubation at 50 °C under H2. Using methylene blue as an electron acceptor, the kinetic constants of the purified membrane-bound homogenase (MBH) were V max=336 U mg−1, k cat=560 s−1, and k cat/ K m=2.24 × 107 M−1 s−1. The MBH exhibited prominent electron paramagnetic resonance signals originating from [3Fe–4S]+ and [4Fe–4S]+ clusters. On the other hand, signals originating from Ni of the active center were very weak, as observed in other oxygen-stable hydrogenases from aerobic H2-oxidizing bacteria. This is the first report of catalytic and biochemical characterization of the respiratory MBH from Hydrogenophaga .  相似文献

The mean rate of oxygen consumption (routine respiration rate, R R, mg O2 fish−1 h−1), measured for individual or small groups of haddock Melanogrammus aeglefinus (3–12 cm standard length, L S) maintained for 5 days within flow‐through respiratory chambers at four different temperatures, increased with increasing dry mass ( M D). The relationship between R R and M D was allometric ( R R = α  M b ) with b values of 0·631, 0·606, 0·655 and 0·650 at 5·0, 8·0, 12·0 and 15·0° C, respectively. The effect of temperature ( T ) and M D on mean R R was described by     indicating a Q 10 of 2·27 between 5 and 15° C. Juvenile haddock routine metabolic scope, calculated as the ratio of the mean of highest and lowest deciles of R R measured in each chamber, significantly decreased with temperature such that the routine scope at 15° C was half that at 5° C. The cost of feeding ( R SDA) was c . 3% of consumed food energy, a value half that found for larger gadoid juveniles and adults.  相似文献

ABSTRACT. Strombidium purpureum Kahl, is an anaerobic oligotrichous ciliate with endosymbiotic phototrophic bacteria. Like other anaerobic ciliates, S. purpureum reacts to O2-pressure. In the light, the ciliates avoid even traces of O2 (< 1% atmospheric saturation). In the dark, however, the ciliates accumulate in water with a pO2 of 1–4% atmospheric saturation. Experiments show that ciliates which have accumulated in the dark under microaerophilic conditions react when the light is turned on, and their ability to escape is enhanced by steep O2-gradients. The ciliates orient themselves in O2-gradients by a series of phobic responses. They tumble whenever they swim towards a higher O2-tension and thus they eventually all swim in the direction of lower pO2. The ecological implications of such behaviour are discussed.  相似文献

Di- n -butyl phthalate (DBF) is widely used as a plasticizer and has been found in all types of ecosystems. It inhibits growth and photosynthesis of green algae ( Chlorella emersonii CCAP strain 211/8 h and Selenastrum capricornutum CCAP strain 278/4) at concentrations higher than 10-5 M . The IC50 value for CO2-dependent oxygen evolution in algae was 3 × 10-4M. The CO2-reduction in isolated protoplasts prepared from barley ( Hordeum vulgare L. cv. Simba) was also inhibited by phthalate. The IC50 value was 2 × 10-4 M . The electron transport in isolated thylakoids prepared from spinach was inhibited with an IC50 value of 3 × 10-4 M . The IC50 value for uncoupled electron transport extrapolated to zero chlorophyll concentration was 2.5 × 10-5 M . The effect of di-n-butyl phthalate was localized to reactions in photosystem II. Di-n-butyl phthalate could thus be a pollutant which affects growth and photosynthesis of plants. The reported IC50 values may be underestimated since di- n -butyl phthalate can attach to surfaces. The results are discussed in relation to observed effects of di- n -butyl phthalate on other organisms.  相似文献

Age and total length ( L T) data from a 11 year monitoring of the Anguilla anguilla eel population of the Camargue lagoons (Rhône delta, southern France) were collected for glass, yellow and silver eels. Three distinct models were calibrated to describe the growth process of undifferentiated eels, females and males, respectively. Uncertainty of parameter estimates was evaluated by bootstrapping. Females were characterized by larger asymptotic body size ( L T) than males (580 ± 50 v . 388 ± 13 mm) and faster growth, whilst the Brody growth coefficient was larger for males than for females (means ±  s . d . 3·00 10−3 ± 1·68 10−3 v . 1·73 10−3 ± 0·50 10−3). Sexual differentiation was estimated to begin at 204 ± 38 mm mean ±  s . d ., i.e . at the end of the second year in the lagoons, well before the L T at which macroscopic differentiation became possible ( c . 300 mm). Males probably leave the lagoon or die (due to either natural or fishing mortality) within the first 3 years, whilst females can remain up to 5 years. Sexual differentiation and maturation have a major role in shaping the L T structure of the population. The L T and mass ( M ) data were fitted by allometric curves     . The calibration of distinct curves for data from different years indicated that the allometric coefficient a was subject to wider interannual fluctuations than the allometric exponent b . A negative correlation linked the average L T and the allometric exponent ( r  = −0·58, P  < 0·01).  相似文献

Aims:  This paper investigates a selection-based acclimation strategy for improving the performance and stability of aerobic granules at a high chloroanilines loading.
Methods and Results:  The experiments were conducted in a sequencing airlift bioreactor (SABR) to develop aerobic granules fed with chloroanilines (ClA). The evolution of aerobic granulation was monitored using image analysis and scanning electron microscopy, and PCR–DGGE analysis of microbial community was performed. The sludge granulation was apparently developed by decreased settling time and gradual increased ClA loading to 0·8 kg m−3 day−1. A steady-state performance of the granular SABR was reached at last, as evidenced by biomass concentration of 6·3 g l−1 and constant ClA removal efficiency of 99·9%. The mature granules had a mean size of 1·55 mm, minimal settling velocity of 68·4 m h−1, specific ClA degradation rate of 0·181 g gVSS−1 day−1. Phylogenetic analysis of aerobic ClA-degrading granules confirmed the dominance of β - , γ -Proteobacteria and Flavobacteria.
Conclusions:  The chosen operating strategy involving step increase in ClA loading and enhancement of major selection pressures was successful in cultivating the aerobic ClA-degrading granules.
Significance and Impact of the Study:  This research could be helpful for improving the stability of aerobic granules via optimizing operating conditions and developing economic feasible full-scale granular bioreactor.  相似文献

α-Conotoxins interact with nicotinic acetylcholine receptors (nAChRs) and acetylcholine-binding proteins (AChBPs) at the sites for agonists/competitive antagonists. α-Conotoxins blocking muscle-type or α7 nAChRs compete with α-bungarotoxin. However, α-conotoxin ImII, a close homolog of the α7 nAChR-targeting α-conotoxin ImI, blocked α7 and muscle nAChRs without displacing α-bungarotoxin ( Ellison et al. 2003, 2004 ), suggesting binding at a different site. We synthesized α-conotoxin ImII, its ribbon isomer (ImII iso ), 'mutant' ImII(W10Y) and found similar potencies in blocking human α7 and muscle nAChRs in Xenopus oocytes. Both isomers displaced [125I]-α-bungarotoxin from human α7 nAChRs in the cell line GH4C1 (IC50 17 and 23 μM, respectively) and from Lymnaea stagnalis and Aplysia californica AChBPs (IC50 2.0–9.0 μM). According to SPR measurements, both isomers bound to immobilized AChBPs and competed with AChBP for immobilized α-bungarotoxin ( K d and IC50 2.5–8.2 μM). On Torpedo nAChR, α-conotoxin [125I]-ImII(W10Y) revealed specific binding ( K d 1.5–6.1 μM) and could be displaced by α-conotoxin ImII, ImII iso and ImII(W10Y) with IC50 2.7, 2.2 and 3.1 μM, respectively. As α-cobratoxin and α-conotoxin ImI displaced [125I]-ImII(W10Y) only at higher concentrations (IC50≥ 90 μM), our results indicate that α-conotoxin ImII and its congeners have an additional binding site on Torpedo nAChR distinct from the site for agonists/competitive antagonists.  相似文献

Foliar uptake pathways for hydrophilic solutes were studied by the analysis of co-uptake of 15N-labelled urea, NH4+ or NO3 and 13C-labelled sucrose across leaf surfaces of various plant species. Uptake of N (y) and sucrose (x) were strongly correlated. Curvilinear regression revealed significantly positive intercepts with the y-axis indicating the involvement of a sucrose-excluding pathway consisting of small pores with radii <0.5 nm. Depending on plant species, N source, leaf side and aperture of stomata, these small pores accounted for 6–62% of total N uptake. Regression analysis revealed that in stomatous leaf surfaces of Vicia faba L., Coffea arabica L. and Prunus cerasus L., the remaining N uptake occurred via another pathway with an estimated average pore radius (rP) greater than 20 nm. This is two orders of magnitude greater than previous estimations of cuticular rP, indicating that this pathway, which was only found in stomatous leaf surfaces, was probably not located in the cuticle but at the surfaces of the stomatal pores. In astomatous leaf surfaces of C. arabica and Populus × canadensis Moench, average rP was 2.0 and 2.4 nm, respectively, which is four to eight times larger than previous estimations of cuticular rP. These results indicate that for polar solutes, the size exclusion limits of plant surfaces can be considerably larger than previously estimated. The far-reaching implications of these findings are discussed.  相似文献
