The sustained speed of kill against Dermacentor reticulatus of two topical combinations, one containing fipronil/amitraz/(S)-methoprene and the other, imidacloprid/permethrin, was evaluated in dogs. Two treated groups and one untreated control group of eight adult Beagle dogs each were randomly formed based on pre-infestation rates and bodyweight. Each treatment was administered topically once on Day 0, according to the recommended label dose and instructions for use. All dogs were infested with 50 adult unfed D. reticulatus starting on Day 1, then weekly, for a total of five weeks. While most studies determine tick efficacy at 48 hours (h), in this study, all remaining ticks were counted and categorized 24 h following each infestation. The numbers of ticks (living or dead) that remained attached on treated dogs were compared to those on the control animals. The percent reduction of attached ticks (disruption of attachment) at 24 h on dogs treated with fipronil/amitraz/(S)-methoprene remained above 92% for four weeks. The reduction of attached ticks at 24 h on dogs treated with imidacloprid/permethrin did not reach 80% during the entire study. The number of ticks attached at 24 h was significantly (p<0.05) lower in the fipronil/amitraz/(S)-methoprene group than in the imidacloprid/permethrin group in assessments on Days 2, 15, 22, 29 and 36. When assessing efficacy based upon live ticks on treated versus control dogs, fipronil/amitraz/(S)-methoprene 24 h efficacy was above 95% for four weeks, decreasing to 77.8% at Day 36. The 24 h efficacy of imidacloprid/permethrin ranged from 56.2% to 86.7% through Day 29, never achieving 90% throughout the study. The 24-hour efficacy of fipronil/amitraz/(S)-methoprene was significantly (p<0.05) higher than imidacloprid/permethrin at all time points, including Day 36.  相似文献

Flea infestations of pets continue to persist due to the lack of knowledge of flea biology and ecology. It is not unusual that pet owners believe regular hygiene, such as shampooing their dogs can replace regular insecticidal treatment. The objective of this study was to compare in a flea simulated environment, modelling exposure similar to that found in a home, that the use of regular physiological shampoo does not control fleas adequately when compared to a long acting topical formulation. Three groups of six dogs were formed: one untreated control group, one group treated monthly with the topical formulation of fipronil/(S)-methoprene, and a third group treated weekly with a hygiene shampoo. All dogs were infested with adult unfed Ctenocephalides felis fleas (200±5) on Days -28 and -21. Each animal's sleeping box was fitted with a plastic cup mounted to the inside roof of the box. The sleeping bench of each animal was covered with a carpet to accommodate flea development. The dogs were maintained in their kennels throughout the study. In order to maintain the environmental flea challenge, C. felis pupae (100±5) were placed in the plastic cup in each animal's sleeping box on Days -14, -7, 0, 7, 14, 21, 28, 35 and 42. The dogs were combed and fleas counted weekly on Days -1, 3, 10, 17, 24, 31, 38, 45, and 51. The fleas were placed immediately back on the dogs. On Day 60, fleas were counted and removed. Flea infestations in the untreated control group at each count averaged between 46.2 and 74.2 fleas throughout the study. The average number of fleas infesting dogs was significantly different (p<0.05) between the untreated and the two treatment groups and between the two treatment groups at all counts throughout the two months study (Days 3, 10, 17, 24, 31, 38, 45, 51 and 60). The efficacy was never below 99.1% in the fipronil/(S)-methoprene group, and efficacy in the shampoo group was never above 79.2%. Weekly shampooing in treatment group 3 was intentionally delayed after Day 42, to evaluate wether missing a weekly bath would affect the flea population. The Day 48 data indicate that forgetting or delaying a single weekly shampooing resulted in a clear increase in flea numbers and a significant decrease in efficacy from 68.2% to 34.8%. The fipronil(S)/methoprene treatment allowed a continuous control as demonstrated by the high efficacy against fleas, and also the number of flea-free dogs on seven of the nine weekly assessments, in spite of what was essentially a continuous flea challenge model.  相似文献

The acaricidal efficacy against Dermacentor reticulatus in dogs of the commercial topical combinations fipronil/(S)-methoprene (FRONTLINE Combo spot-on dog), imidacloprid/permethrin (Advantix) and metaflumizone/amitraz (ProMeris Duo) was evaluated and compared. Three treatment groups and one untreated control group of six adult Beagle dogs each were randomly formed. Each treatment was administered topically once on Day-0, according to the recommended label dose and instructions for use. All dogs were infested weekly with approximately 50 adult unfed D. reticulatus over a period of seven weeks. Ticks were removed and counted approximately 48 hours after each infestation. The percent reduction in numbers of ticks for fipronil/(S)-methoprene was > or = 97% compared to untreated controls for all seven weekly infestations. The percent reductions for imidacloprid/permethrin and metaflumizone/amitraz were satisfactory initially but fell and stayed below 90 % after three weeks. From the third week onwards, fipronil/(S)-methoprene treated dogs had significantly fewer ticks than imidacloprid/permethrin or metaflumizone/amitraz treated dogs (p < 0.05).  相似文献

Despite the increasing number of ectoparasiticides for pets and their use, flea infestations of cats and dogs are still widespread in Europe. It is therefore important to assess the maintenance of efficacy of the ectoparasiticides for cats and dogs. The present studies aimed to evaluate the efficacy of monthly treatments using a fipronil/(S)-methoprene combination spot-on (Frontline Combo) on dogs and cats from private veterinary clinics located in seven European countries. The survey was conducted for three months during the flea season 2009. A total of 233 dogs and 180 cats were included. Each animal was treated at Days 0 (Day 0), 30 (D30) and 60 (D60) at the vet clinics. For each animal, at least three flea counts were performed at D0, D30 and/or D60 and/or day 90 (Day 90) in order to evaluate the prevalence of flea infestation and the efficacy of control. At the beginning of the study the prevalence of infested animals was 41.63% (97/233) in dogs and 47.22% (85/180) in cats. At D90, the number of dogs remaining infested fell to 8/211 therefore 91.75% became flea-free. The number of infested cats fell from 85 to 9/173 at D90 therefore 89.41% were cured. All animals still infested at Day 90 were living under epidemiological conditions that favour heavy flea burdens. These results are similar or better to previous studies, indicating the continuous high level of efficacy for fipronil 10 years after launch.  相似文献

The efficacy of the phosphonate fertilizers, Calphos® (a.i. calcium phosphonate), Magphos® (a.i. magnesium phosphonate and potassium phosphonate) and Phosphoros® (a.i. potassium phosphonate) against two species of root knot nematodes (RKN), Meloidogyne javanica and M. incognita is evaluated. Laboratory experiments showed that Calphos®, Magphos® and their main components inhibited egg hatching and caused 100% mortality of the second stage juveniles (J2s) of the two RKN species; the hatching inhibition effects persisted after transferring the egg masses of both species to water. However, Phosphoros® (0.5%) did not suppress egg hatching or the survival of J2s of both RKN species. No hatching occurred when egg masses were treated for one week with the nematicide Vydate L® (2 ml/l), however, J2s hatched when the Vydate L® treated egg masses were moved to water. The glasshouse study indicated that Magphos®, Calphos® and Phosphoros® reduced root galling caused by M. javanica by 98, 66 and 47%, respectively, in comparison to the untreated controls. Magphos® resulted in the lowest number of root galls formed by M. incognita, the reduction was 84%. In contrast, Calphos® and Phosphoros® reduced galling by 47 and 39%, respectively. The Magphos® treatment resulted in the lowest numbers of egg masses and the lowest reproductive factor (RF) of both nematode species. However, plants treated with Phosphoros® resulted in higher foliage weights compared with the application of the other two fertilizers and the untreated plants.  相似文献

Nodulisporic acid A (NSA) is a novel natural product from a new structural class that was shown previously to have insecticidal activity against blowfly larvae. To determine if there was useful systemic efficacy against fleas (Ctenocephalides felis). NSA was evaluated in an artificial membrane flea feeding device and in dogs. In the artificial membrane flea feeding device, adult C. felis were allowed to feed on bovine blood containing various concentrations of NSA through a Parafilm membrane. NSA killed the fleas with a 50% lethal concentration of 0.68 microg/ml and was approximately 10-fold more potent than the systemic insecticide ivermectin. In the initial probe dog test, a single beagle was challenged with 100 C. felis before oral dosing with 15 mg/kg of NSA. Flea counts conducted at 72 hr postdosing showed an 88% reduction relative to control. Re-challenge of the same dog at 5 days postdosing showed 50% reduction of fleas at day 7, demonstrating some residual flea activity. In a confirmatory study, 8 dogs were challenged with 100 fleas just before oral dosing with 15 mg/kg of NSA (4 dogs) or vehicle (4 dogs). There was 99% reduction of fleas at 48 hr postdosing in the NSA-treated dogs relative to control. Additional challenges with 100 fleas were performed on these 8 dogs at 48-hr intervals to determine the duration of efficacy, and there was 97, 51, and 0% reduction of fleas relative to control on days 4, 6, and 8, respectively. No adverse effects were observed in the dogs in these studies. These data show that NSA has potent oral activity in the dog for the control of fleas, while lacking overt mammalian toxicity.  相似文献

Bubonic plague (Yersinia pestis) is a deadly zoonosis with black‐tailed prairie dogs (Cynomys ludovicianus) as a reservoir host in the United States. Systemic insecticides are a promising means of controlling the vectors, Oropsylla spp. fleas, infesting these prairie dogs, subsequently disrupting the Y. pestis cycle. The objective of this study was to conduct a field trial evaluating the efficacy of a grain rodent bait containing fipronil (0.005%) against fleas infesting prairie dogs. The study was performed in Larimer County, CO, where bait was applied to a treatment area containing a dense prairie dog population, three times over a three‐week period. Prairie dogs were captured and combed for fleas during four study periods (pre‐, mid‐, 1st post‐, and 2nd post‐treatment). Results indicated the use of bait containing fipronil significantly reduced flea burden. The bait containing fipronil was determined to reduce the mean number of fleas per prairie dog >95% for a minimum of 52 days post‐initial treatment application and 31 days post‐final treatment application. These results suggest the potential for this form of treatment to reduce flea population density on prairie dogs, and subsequently plague transmission, among mammalian hosts across the United States and beyond.  相似文献

The safety of two vaccines available on the French market against canine babesiosis – Nobivac Piro® (NP) and Pirodog® (P) – have been evaluated. Their local, general and biochemical impacts have been compared in a controlled experimental study. Three groups were used: a control group (T) and two groups vaccinated twice at 21 days interval. All dogs presented moderate local reaction. However, either clinical and biological parameters showed that the NP group presented a significantly more intense reaction at the injection site compared to the P group. No statistical difference has been revealed between the groups P and T evolutions.  相似文献

Medically important arthropods, including fleas, play an important role in causing clinical disorders and disease in man and domestic animals. This study was conducted to determine the seasonal flea infestations for domestic dogs from different geographic regions of Iran. A total of 407 fleas, belonging to 5 different species, were recovered from 83 domestic dogs from 3 regions. There was a distinctive pattern of species distribution and infestations with the highest infestation rates observed in a temperate climate and higher rainfall. Additionally, fleas were observed over all seasons, except February and March, with the highest infestation rate observed in August (24.7%) and the lowest rate in January (1.7%). They also parasitize dogs with a different spectrum of species. The cat flea, Ctenocephalides felis (67.5%), exhibited the highest prevalence among all flea species found on dogs. Thus, climatic conditions and seasonal patterns impact on flea infestation and must be considered in developing control programs.  相似文献

The aim of this study was to evaluate the effects of adding different concentrations of edible bird’s nest (EBN) which is secreted by swiftlet birds (Aerodramus fuciphagus), into EquiPlus® and E-Z Mixin® extenders on the quality of chilled Arabian stallion semen at various storage times (0, 24 and 48 h). Ten ejaculates were collected from five stallions, and diluted using the two extenders containing 0% (control), 0.12%, 0.24% and 0.24% of EBN + seminal plasma (SP). All the diluted semen samples were then cooled and stored at 5 °C, and examined at 0, 24 and 48 h. Sperm kinetic parameters were assessed using computer assisted sperm analysis (CASA) and viability were assessed using Hoechst33342/PI stain. In both extenders, total motility (TM) and progressive motility (PM) were significantly higher at 0.12% and 0.24% compared to 0.24% + SP at 24 and 48 h. At 0.12%, E-Z mixin® treated semen had significantly higher TM and PM than EquiPlus® at 24 and 48 h. At 0.12% and 0.24%, average path velocity (VAP), straight-line velocity (VSL) and curvilinear velocity (VCL) were significantly higher in E-Z mixin® treated semen compared to EquiPlus® at 24 and 48 h. Comparisons between the two extender types at different concentrations of EBN showed no significant difference in lateral head amplitude (ALH), linearity (LIN), straightness (STR), beat cross frequency (BCF) and viability, irrespective of the storage time. The percentage of viable was significantly higher in E-Z mixin® than EquiPlus® at 0 and 48 h in control and 0.12%. Supplementation of the E-Z mixin® extender with 0.12% and 0.24% EBN concentrations in the absence of SP provided better CASA parameters such as TM, PM, VAP, VSL, and VCL at 24 and 48 h storage time. In conclusion, the results of this study indicated that chilled semen from Arabian stallion that was extended using E-Z mixin® and supplemented with 0.12% and 0.24% EBN concentrations performed better and yielded superior results in sperm kinetic parameters and % viable compared to EquiPlus® at 24 and 48 h storage time.  相似文献

Sylvatic plague (Yersinia pestis) was introduced into North America over 100 years ago. The disease causes high mortality and extirpations in black-tailed prairie dogs (Cynomys ludovicianus), which is of conservation concern because prairie dogs provide habitat for the critically endangered black-footed ferret (Mustela nigripes). Our goal was to help elucidate the mechanism Y. pestis uses to persist in prairie ecosystems during enzootic and epizootic phases. We used a nested PCR protocol to assay for plague genomes in fleas collected from prairie dog burrows potentially exposed to plague in 1999 and 2000. No active plague epizootic was apparent in the 55 prairie dog colonies sampled in 2002 and 2003. However, 63% of the colonies contained plague-positive burrows in 2002, and 57% contained plague-positive burrows in 2003. Within plague-positive colonies, 23% of sampled burrows contained plague-positive fleas in 2002, and 26% contained plague-positive fleas in 2003. Of 15 intensively sampled colonies, there was no relationship between change in colony area and percentage of plague-positive burrows over the two years of the study. Some seasonality in plague prevalence was apparent because the highest percentages of plague-positive colonies were recorded in May and June. The surprisingly high prevalence of plague on study area colonies without any obvious epizootic suggested that the pathogen existed in an enzootic state in black-tailed prairie dogs. These findings have important implications for the management of prairie dogs and other species that are purported to be enzootic reservoir species.  相似文献

Toxicity of spinosad to immature stages of Chrysoperla carnea (Stephens) (Neuroptera: Chrysopidae) and its effect on the reproduction and survival of adult stages after direct spray and ingestion treatments were evaluated. Spinosad was harmless to C. carnea eggs and pupae irrespective of concentrations or method of treatments. Direct spray of spinosad to first instar caused significant reduction in rate of pupation and L1-adult survival, but did not affect the rate of adult emergence. Third-instar bioassay revealed significant difference in L3-adult survival. When C. carnea first instars were fed upon spinosad-treated-Brevicoryne brassicae (L.), significant differences in investigated parameters were not observed among different treatments. The weight of third instar and pupae were comparable regardless of tested spinosad concentrations. Also, feeding on spinosad treated-B. brassicae had no negative impact on fecundity and fertility. Furthermore, ingestion of spinosad contaminated-B. brassicae significantly prolonged larval and larval + pupal periods. The larval feeding capacity of C. carnea did not differ significantly between larvae fed on spinosad-treated and untreated aphids with the exception of first instar. When C. carnea adults were allowed to oviposit on spinosad treated-substrate, total number of eggs laid, percent of eggs laid on the treated substrate and egg hatching did not differ from those of control. However, when adults were fed on spinosad-treated artificial diet, negative effects on adult survival and fecundity were observed.

Eggs and rhabditiform juveniles of the ruminant parasite Trichostrongylus colubriformis developed normally in Caenorhabditis briggsae Maintenance Medium. A toxin from a crystal-enriched preparation of the bacterium Bacillus thuringiensis israelensis was lethal to nematode eggs and juveniles within 24 hours and to eggs and juveniles after 24 hours of development. Treated eggs had refractive granules and development was arrested, whereas nontreated eggs developed normally. Eggs treated after 24 hours of development contained juveniles that were granulated, had esophageal derangements, and were moribund or dead. The ovicidal toxin from B. t. israelensis may facilitate microbial control of parasitic nematodes.  相似文献

The aim of this study was to evaluate the efficacy of a polymerase chain reaction (PCR)-based method to detect Schistosoma mansoni DNA in stool samples from individuals living in a low-endemicity area in Brazil. Of the 125 initial stool samples, 80 were ELISA reactive and eggs were identified in 19 of the samples by parasitological examination. For the PCR evaluations, 56 stool samples were selected and divided into five groups. Groups I-IV were scored negative for S. mansoni eggs by parasitological examination. Groups I and II were ELISA reactive, whereas Groups III and IV were ELISA nonreactive. Groups II and III were positive for other intestinal parasites. PCR testing scored eight samples as positive from these four groups. Group V represented the S. mansoni -positive group and it included ELISA-reactive samples that were scored positive for S. mansoni by one or more parasitological examinations (6/19 were positive by Kato-Katz method, 9/17 by saline gradient and 10/13 by Helmintex®). PCR scored 13 of these 19 samples as positive for S. mansoni . We conclude that while none of these methods yielded 100% sensitivity, a combination of techniques should be effective for improving the detection of S. mansoni infection in low-endemicity areas.  相似文献

Abstract

A conceptual model was developed to describe the dynamics of the pet dog population. The model synthesizes existing data collected for a variety of purposes to estimate the size of the various components of the pet dog population in Washington and Iowa during 1991. The total population mortality rate was estimated as 12.4% per year. Animal shelters in Washington and Iowa handled 7.6% and euthanized 4.0% of the dog population in those two states. When these estimates were extrapolated to the entire U.S. dog population, the model predicted that the total annual turnover in owned dogs was 14.7%, or 7.71 million dogs, that 4 million dogs were handled by animal shelters, and that 2.1 million were euthanized. It was also estimated that 79% of all female dogs were spayed, that household breeding could be attributed to less than one-fifth (18.7%) of the female dogs in the reproductive pool, and that the number of owners contributing to total dog population turnover through failure to retain their dog (103,453) was approximately three times the number of owners who allowed their female dogs to be bred (32,513).  相似文献

Extracts from the plants Plantago lanceolata and P. rugelii were evaluated for toxicity to the root-knot nematode Meloidogyne incognita, the beneficial microbes Enterobacter cloacae, Pseudomonas fluorescens and Trichoderma virens, and the plant-pathogenic fungi Fusarium oxysporum f. sp. gladioli, Phytophthora capsici, Pythium ultimum, and Rhizoctonia solani. Wild plants were collected, roots were excised from shoots, and the plant parts were dried and ground to a powder. One set of extracts (10% w/v) was prepared in water and another in methanol. Treatments included extract concentrations of 25%, 50%, 75% and 100%, and water controls. Meloidogyne incognita egg hatch was recorded after 7-day exposure to the treatments, and second-stage juvenile (J2) activity after 48 hours. All extracts were toxic to eggs and J2, with P. lanceolata shoot extract tending to have the most activity against M. incognita. Numbers of active J2 remained the same or decreased in a 24-hour water rinse following the 48-hour extract treatment, indicating that the extracts were lethal. When data from water- and methanol-extracted roots and shoots of both plant species were combined for analysis, J2 tended to be more sensitive than eggs to the toxic compounds at lower concentrations, while the higher concentrations (75% and 100%) were equally toxic to both life stages. The effective concentrations causing 50% reduction (EC50) in egg hatch and in J2 viability were 44.4% and 43.7%, respectively. No extract was toxic to any of the bacteria or fungi in our assays.  相似文献

Developing methods to label viruses with fluorescent moieties has its merits in elucidating viral infection mechanisms and exploring novel antiviral therapeutics. Fluorescent quantum dots (QDs), an emerging probe for biological imaging and medical diagnostics, were employed in this study to tag retrovirus encoding enhanced green fluorescent protein (EGFP) genes. Electrostatic repulsion forces generated from both negatively charged retrovirus and QDs were neutralized by cationic Polybrene, forming colloidal complexes of QDs-virus. By examining the level of EGFP expression in 3T3 fibroblast cells treated with QDs-tagged retroviruses for 24 hours, the infectivity of retrovirus incorporated with QDs was shown to be only slightly decreased. Moreover, the imaging of QDs can be detected in the cellular milieu. In summary, the mild method developed here makes QDs-tagged virus a potential imaging probe for direct tracking the infection process and monitoring distribution of viral particles in infected cells.  相似文献

Contact toxicity of a commercial bacterial fermentation insecticide, spinosad, to adults of eight stored-product beetles was evaluated on four different surfaces. Aqueous spinosad suspension was sprayed with an airbrush to 30.5-cm2 surfaces of concrete, galvanized steel, unwaxed floor tile, or waxed floor tile to obtain deposits of 0.05 or 0.1 mg (AI)/cm2. Control surfaces were sprayed with distilled water. Approximately 24 h after distilled water or spinosad application, 30 adult beetles were confined, by species, to each untreated and spinosad-treated surface. Insects on surfaces were exposed for 24 h to assess knockdown at 25 +/- 1 degree C and 50 +/- 10% RH, and then were held on food for an additional 24 h to assess mortality. Knockdown and mortality of each insect species on all four surfaces were significantly greater on spinosad-treated surfaces than on distilled water-treated surfaces. Knockdown and mortality of all species on all surfaces was similar at the two spinosad deposit levels. Except for Tribolium spp., mortality of all other species exposed to spinosad was 99-100%. Tribolium spp. were highly susceptible to spinosad on concrete (98-100% mortality); however, on unwaxed floor tile, steel, and waxed floor tile recovery on food after knockdown resulted in only 72-92% mortality. Our results suggest that spinosad has excellent contact activity against adults of stored-product insects, especially on concrete, and has potential for use as a general surface, spot, or crack/crevice spray to control insects in empty bins, warehouses, food-processing facilities, and retail stores.  相似文献

We collected Oropsylla montana from rock squirrels, Spermophilus varigatus, and infected a subset of collected fleas with Yersinia pestis, the etiological agent of plague. We used bar-tagged DNA pyrosequencing to characterize bacterial communities of wild, uninfected controls and infected fleas. Bacterial communities within Y. pestis-infected fleas were substantially more similar to one another than communities within wild or control fleas, suggesting that infection alters the bacterial community in a directed manner such that specific bacterial lineages are severely reduced in abundance or entirely eliminated from the community. Laboratory conditions also significantly altered flea-associated bacterial communities relative to wild communities, but much less so than Y. pestis infection. The abundance of Firmicutes decreased considerably in infected fleas, and Bacteroidetes were almost completely eliminated from both the control and infected fleas. Bartonella and Wolbachia were unaffected or responded positively to Y. pestis infection.  相似文献
