Spherical, coated microconidia-aggregates as survival and dispersal units of Botrytis cinerea Pers. As a reaction to longer lasting unfavourable growth conditions Botrytis cinerea is able to form spherical microconidia-aggregates enclosed in a protective coat. Genesis, structure and occurrence of these so far unknown microconidia-aggregates are described and their function as survival and dispersal units of the fungus is discussed.  相似文献

In vivo-investigations on the formation and function of chlamydospores of Botrytis cinerea Pers. in the host-parasite-system Fuchsia hybridaB. cinerea On naturally with Botrytis cinerea Pers. infected and artificially inoculated outdoor- and greenhouse-plants of Fuchsia hybrida the extra- and intramatrical formation of the B. cinerea- chlamydospores was investigated. The chlamydospores served 1. as structures of survival, which were tested with regard to their tolerance of drought, nutrient- and oxygen-deficiency, attack by bacteria and pH-requirements. 2. The chlamydospores represented dispersal units, which were capable of germination. 3. The chlamydospores could function as structures of infection, because after chlamydospore germination the outgrowing mycelium – either directly or after production of macroconidia – could serve as secondary inoculum and start new infections.  相似文献

Resistance of different cultivated and wild tomato plants (Lycopersicon spp.) to Botrytis cinerea Pers. 20 provenances of different cultivated and wild tomato plants (Lycopersicon spp.) were screened for resistance to Botrytis cinerea Pers. using an in vitro-leaf necrosis test. The Botrytis resistance decreased with increasing age of the leaves corresponding to their insertion height (relative youth resistance respectively senescence susceptibility). The 6 B. cinerea-isolates used for inoculation differed significantly in virulence. With increasing inoculum age a virulence reduction of the various B. cinerea-isolates occurred. Within the investigated test plant collection 2 wild species –L. columbianum and L. hirsutum– proved to be resistant in each stage of development to all B. cinerea-isolates and additionally showed field resistance.  相似文献

Fungicidal activity of N-cetyl N,N,N-trimethyl ammonium chloride to Botrytis cinerea Pers. Grey mould of grapes caused by Botrytis cinerea is the most important phytopatho-logical problem in swiss vineyards, since the appearance of resistant strains to dicarboxi-mides. Research for new chemicals is necessary. One compound, a cationic surfactant, the N-cetyl N,N,N-trimethyl ammonium chloride (CTAC), is very active in vitro against Botrytis cinerea. On the genesis and germination of the chlamydospores of Botrytis cinerea Pers. The chlamydospores of Botrytis cinerea are hyaline single cells of extremely variable form and size. They are formed under conditions unfavourable for growth as terminal or intercalary cells by transformation of vegetative mycelium parts and are liberated by hyphal disintegration. The chlamydospore genesis in vitro in aging malt agar cultures began about after one month. But the chlamydospore formation could also be initiated earlier by different conditions of culture. The chlamydospores germinated either with hypha or by microconidia — a herewith first described mode of germination. Intermediates of these both modes of chlamydospore germination could be regulated very differentiatedly by transferring the chlamydospores into malt solution (2%)and/or destilled water and by changing the duration of stay in the individual media. Under adverse external conditions no germination occurred. The three Botrytis cinerea-isolates did not show any differences in habitus, genesis and germination of their chlamydospores. Also in vivo on outdoor- and greenhouse-tomatoplants the occurrence of chlamydospores was no rarity. Since the chlamydospores are produced under very different adverse conditions of growth but cannot survive a period of drought lasting longer than three months without damage, they do not represent long-termed resistant perennating structures, but temporary stages of the fungus for intervening periods.  相似文献

Endo- and Exochitinase Activity in Kiwifruit Infected with Botrytis cinerea   总被引:1,自引:0,他引:1  
Endo- and exochitinase activities were determined in autoclaved leaves, live leaves, and fruit of kiwifruit (Actinidia deliciosa var. deliciosa cv. Hayward [A. Chev.]Liang and Ferguson) following infection with Botrytis cinerea (Pers.). Appreciable endochitinase activity was found in healthy and diseased leaves and fruit at harvest and after cool-storage, but was absent in plant tissue killed by autoclaving. Later harvested fruit produced more endochitinase activity in storage than did early harvested fruit. Exochitinase activity was only associated with diseased tissue, and was present in diseased autoclaved leaves. The results suggest that endochitinase activity originates from the plant whilst exochitinase is associated with the pathogen. The significance of these findings is discussed.  相似文献

Laboratory experiments were conducted to develop an assay for evaluation of carrot (Daucus carota L.) leaf reaction to Botrytis cinerea Pers. ex Pers. The detached carrot leaflets were inoculated with the colonized agar plugs attached to the cut surface of the midrib or by spraying conidial suspension. The estimation of the infected leaflet area expressed as the area under the disease progress curve enabled the discrimination between carrot genotypes differing in their susceptibility to the pathogen. Evaluation based on the measurement of the length of the midrib lesion did not allow such differentiation. The inoculation of leaf surface with suspension of conidia was less reliable and the development of the disease symptoms was not reproducible. The established assay using colonized plugs enables a fast assessment of carrot leaf susceptibility to grey mould and can be particularly useful for a non‐destructive, preliminary evaluation of precious and limited source materials.  相似文献

Grey mould, caused by the fungus Botrytis cinerea Pers ex Fr., is a very destructive and important disease worldwide. Fluazinam is a phenylpyridinamine fungicide with broad‐spectrum activities. The baseline sensitivity of B. cinerea to fluazinam is yet to be established in Henan Province, China. In this study, a total of 117 field isolates of B. cinerea were collected from 49 commercial greenhouses in different locations of Henan Province, in 2016, and the sensitivities of these isolates to fluazinam were determined based on mycelial growth. The effective concentration for 50% (EC50) values ranged from 0.0038 to 0.0441 μg/ml, and the mean EC50 value was 0.0201 ± 0.0081 μg/ml for mycelial growth. The frequency distribution range presented a unimodal curve. To define the cross‐resistance relationships, the linear correlation coefficients of the EC50 values between fluazinam and carbendazim, procymidone, pyrimethanil or boscalid were analysed. The results showed that no correlation was observed between fluazinam and the other tested fungicides. These results provide important information to growers for the prevention and control of grey mould.  相似文献

Summary The progression of infection caused by a fungal pathogen,Botrytis cinerea Pers.: Fr., in a fruit of red raspberry (Rubus idaeus L.) was followed by nuclear magnetic resonance (NMR) microscopic imaging over a 4 day period. It was found that a standard gradient echo sequence discriminated clearly between infected and healthy tissue as mycelium spread across the fruit from a single woundinoculated drupelet.  相似文献

In dual cultures Fusarium lateritium Nees ex Link coiled, penetrated and destroyed young actively growing hyphae of Botrytis cinerea Pers., Cytospora rubescens Fr., Eutypa armeniacae Hansf. et Carter, Sphaeropsis malorum Peck, Rhizoctonia solani Kühn, Trichoderma koningii Oud. (Tk-1) and Trichoderma hamatum (Bon.) Bain. (Tha-2), and conidiophores and conidia of Botrytis cinerea Pers. The isolates of Trichoderma koningii Oud. (Tk-3) and Trichoderma pseudokoningii Rifai (Tp-1) parasited and destroyed the hyphae of Fusarium lateritium. There was no hyphal interaction between Fusarium lateritium and an isolate of Trichoderma longibrachiatum Rifai. Hyphal, interactions occurred mostly on the aerial hyphae of test fungi which grew over the inhibition zone formed between the colonies of Fusarium lateritium and the test fungus, and contacted the hyphae of Fusarium lateritium. Grey mould, caused by Botrytis cinerea Pers ex Fr., is one of the most common diseases of tomato worldwide. Fludioxonil belongs to the phenylpyrrole fungicides, which have high activity against B. cinerea. The sensitivity of fludioxonil was evaluated on the basis of the level of inhibition of mycelium growth in 274 B. cinerea isolates collected from different locations (untreated with this fungicide) in Henan Province, China. The EC50 values for fludioxonil ranged from 0.0033 to 0.0415 mg/l, and the average EC50 values were 0.0156 ± 0.0078 mg/l. Three fludioxonil‐resistant mutants were obtained by subculturing fludioxonil‐sensitive wild‐type isolates on continuously increasing fludioxonil concentrations. For the cross‐resistance assay, fludioxonil revealed positive cross‐resistance with procymidone but did not reveal cross‐resistance with pyrimethanil, boscalid and trifloxystrobin. Mycelial growth, conidial production, hyphal dry weight and pathogenicity were diminished significantly between the fludioxonil‐resistant mutants and their sensitive wild‐type parental isolates. This study shows for the first time that fludioxonil‐resistant isolates of B. cinerea are still not present in Henan Province because this fungicide is an attractive and effective fungicide for chemical control. Recommendations can be made to growers to use fludioxonil to control grey mould and to consider the potential moderate resistance risk of using this fungicide.  相似文献

The ability of a cell-free secretion from germinating conidia(germination fluid) and heat-killed conidia of Botrytis cinereaPers. ex Pers. to induce resistance in exposed carrot root tissueswas unaffected during freezing to –25 °C and rapidthawing, and by heating to 50 °C for 10 min. Activity waslost by boiling for 10 min or at room temperature after 24 h. There was less cell death (Evans blue and neutral red staining)in the surface tissue of slices pretreated with heat-killedconidia and subsequently exposed to infection than in similartissue of untreated, inoculated slices. Both inducing preparationsalone caused maceration and cell death in one or two cell layersof the slice surface. Healthy tissue killed by freezing to –25°Cfor 2 h and rapid thawing, released a factor which, when similarlytested on slices, resulted in germ-tube inhibition and reducedvisual symptoms following inoculation with live spores. Thepossibility that cell death triggered the release of a hostelicitor which caused enhanced accumulation of phytoalexinsand suberin deposition is discussed. Botrytis cinerea is an economically important fungal pathogen with a host range of over 200 plant species. Unfortunately, gray mold disease caused by B. cinerea has not been effectively controlled because of its high risk for fungicide resistance development. As a part of our ongoing efforts to develop novel sulfonamides as agricultural fungicides against Botrytis cinerea, we introduced 2-aminoethanesulfonic acid (taurine) substructure, designed and synthesized a series of novel 2-substituted acylaminoethylsulfonamides. The newly synthesized sulfonamides were evaluated in vitro and in vivo for their fungicidal activity against Botrytis cinerea, of which the 2-ethoxyacetylamide derivative (V-A-12, EC50 = 0.66 mg·L−1) exhibited the highest potency in vitro and superior fungicidal activity compared with procymidone (EC50 = 1.06 mg·L−1). In vivo bioassay indicated that compound V-A-12 could be effective for the control of tomato gray mold. Moreover, the structure-activity relationship of these sulfonamides was analyzed by establishing a three-dimensional quantitative structure-activity relationship (3D-QSAR) model, which can provide guidance for the development of sulfonamides as fungicides. Finally, the effeicacy of sulfonamide derivatives was again verified in the activity evaluation against resistant Botrytis cinerea strains. These results further enhance the development value of 2-substituted acylaminoethylsulfonamides to control the tomato gray mold.  相似文献

[背景] 灰葡萄孢(Botrytis cinerea)是引起葡萄采后病害的主要病原菌之一,严重影响葡萄的贮期和品质,给葡萄产业带来极大损失。利用拮抗微生物抑制采后病原菌生长已逐渐成为防治葡萄采后灰霉病的重要手段。[目的] 利用昆虫病原线虫共生细菌广谱高效的抑菌特性,从现有共生细菌资源中筛选对灰葡萄孢具有高拮抗作用的菌株,为葡萄采后灰霉病的抑制提供新的材料和研究方向。[方法] 通过平板对峙培养法和菌丝生长速率法分离筛选拮抗共生细菌,并对优选的高效拮抗共生细菌进行16S rRNA基因序列进化分析,采用扫描电镜观察其对灰葡萄孢菌丝生长的影响,利用损伤接种法对红地球葡萄防治效果进行验证。[结果] 初步分离筛选共获得9株拮抗菌,复筛与复测得到一株抑菌效果显著的共生细菌(命名为ALL),经进化分析其为嗜线虫致病杆菌(Xenorhabdus nematophila),其16S rRNA基因序列的Genbank登录号为MW488402,与菌株Xenorhabdus nematophi la NC116聚于同一分支,相似性达99.79%。扫描电镜观察该菌株导致灰葡萄孢菌丝扭曲变形、表面皱缩、失水塌陷,该菌株发酵(36 h)上清液浓度为1%时对灰葡萄孢菌丝抑制率达44.5%。在葡萄常温防效实验中,与对照组比较,ALL菌株发酵上清液对灰霉菌防治效果较好,3 d后防效为63.50%。[结论] 本研究应用昆虫病原线虫共生细菌生物防治葡萄贮期灰霉病,筛选出一株高效拮抗灰葡萄孢的昆虫病原线虫共生细菌,而且其上清液对灰葡萄孢具有良好的抑制效果,为生物防治贮期葡萄灰霉病提供了新的生物材料和相关研究基础。  相似文献

The ethanolic and aqueous extracts from in vitro shoots of Quillaja saponaria Mol. (Quillay) were studied for their antifungal activity against the phytopathogenic fungus Botrytis cinerea Pers. These extracts reduced conidial germination and mycelial growth of B. cinerea, ethanolic extracts being more active than aqueous extracts. In addition, the damage areas produced by this fungus on tomato leaves and strawberry fruits pre-treated with quillay extracts were diminished. The fungitoxic effect of in vitro-grown quillay extract was similar to those obtained with commercial fungicides of both natural (BC-1000) and synthetic (iprodione–dicarboximide) origin. On the other hand, the antifungal action of quillay extracts obtained from adult trees naturally grown was only slightly superior to the fungitoxic activity of the extract from in vitro plants. HPLC analysis of the extract showed that it contained saponins and some phenolic compounds such as chlorogenic, caffeic, vanillic, and salicylic acids, and scopoletin, which have been identified as antifungal agents on phytopathogenic fungi. The results obtained in this work, suggests that extracts of in vitro-grown quillay have an important protective effect against B. cinerea and support the use of an in vitro culture system as a biotechnological alternative to obtain environmental safe antifungal quillay extracts to control B. cinerea, contributing to the preservation of this indigenous Chilean species.  相似文献

Grey mould has been detected on Salicornia bigelovii Torr plants in nursery and in the wild in north‐west Mexico. Sampling of the grey mould was performed in the state of Sonora, Mexico, of wild as well as cultivated S. bigelovii plants. The samples were isolated, and based on morphology, the species was identified as Botrytis cinerea Pers. Koch's postulates were fulfilled by pathogenicity tests carried out in plated petri dishes on branches from the 3‐month‐old potted S. bigelovii plants. To our knowledge, this is the first report of the isolation and identification of the fungal pathogen B. cinerea from S. bigelovii in the north‐west Mexico.  相似文献

【目的】利用荧光显微镜和激光共聚焦扫描显微镜技术初步探讨解淀粉芽孢杆菌(Bacillus amyloquefaciens)B15菌株发酵液中的抑菌混合物质伊枯草菌素A(iturin A)和芬芥素(fengycin)对葡萄灰霉病病原菌灰葡萄孢(Botrytis cinerea)的抑菌机理。【方法】采用琼脂稀释法讨论解淀粉芽孢杆菌B15发酵液对灰葡萄孢的抑菌活性。利用台盼蓝(trypan blue)染色、4′,6-二脒基-2-苯基吲哚(DAPI)、双氢罗丹明123(DHR123)、钙离子探针fluo-3/am和Annexin V-PI探针染色来观察解淀粉芽孢杆菌B15发酵液对灰葡萄孢细胞膜和菌丝形态、细胞核、活性氧、钙离子和磷脂酰丝氨酸层的影响。【结果】抑菌活性实验发现解淀粉芽孢杆菌B15发酵液对灰葡萄孢具有良好抑菌效果。荧光显微镜台盼蓝染色观察发现,经B15发酵液处理过的灰葡萄孢出现菌丝畸形、菌丝体粗大、尖端肿胀并被染成蓝色和明显的液泡化现象。同时未在处理组中观察到细胞内容物泄漏,说明处理组菌丝细胞膜未发生破损。该结果表明在此次试验中,B15发酵液中的抑菌有效物质不以破损细胞膜的方式直接导致灰葡萄孢的死亡。激光共聚焦显微镜观察结果发现,处理组的灰葡萄孢菌丝出现典型的细胞凋亡现象、染色质固缩、细胞核裂解、磷脂酰丝氨酸层外翻、活性氧和钙离子积累。【结论】该实验表明解淀粉芽孢杆菌B15发酵液以诱导细胞凋亡的形式来抑制灰葡萄孢菌丝的生长。  相似文献

Six selected antioxidants (catechin, quercetin-3-galactoside, cyanidin-3-glucoside, pelargonidin-3-glucoside, ellagic and gallic acids) were evaluated in vitro for their antifungal activities and mode of action on Botrytis cinerea Pers., one of the most important pathogens of strawberries. Inhibitory effects were found for all the tested antioxidants, but varied at different fungal developmental stages. Catechin and quercetin-3-galactoside showed linear inhibitory effects on germ tube elongation, with the highest suppression ratios of 54.8% and 58.8% respectively. No significant effect was found on spore germination between treatments and control. Gallic acid showed very strong and linear inhibition on spore germination (r = ?0.95), but the effect diminished after spore germination. Cyanidin-3-glucoside and pelargonidin-3-glucoside provided effective control on the fungi as concentrations increased. The arresting effect of ellagic acid on development of B. cinerea was quadratic. Ellagic acid inhibited germ tube elongation and mycelial growth at its highest and lowest concentrations, while no effects were observed at its medium concentration used in this study.  相似文献

Botrytis cinerea (teleomorph Botryotinia fuckeliana) is a necrotrophic plant pathogenic fungus that causes grey mould and enormous economic losses worldwide in different crops. Control of B. cinerea is difficult due to the appearance of fungicide‐resistant isolates, and the diversity in virulence due to genetic variability and, perhaps, the infection with mycoviruses or fungal viruses. The discovery of mycoviruses and their possible application as biocontrol agents, as well as their use as tools to study the plant–pathogen interaction, has encouraged their study in B. cinerea. Herein, we have analysed the occurrence of mycoviruses in Spanish B. cinerea isolates to approach a better understanding of the interactions among viruses, fungi and plants in this pathosystem. Fifty‐five percent of the B. cinerea isolates analysed contained double‐stranded RNA (dsRNA) elements, and the number of dsRNA elements, their relative concentration and size were variable among isolates. Some of these dsRNAs were related to the presence of virus like rod or isometric particles, and to cellular degeneration and malformed mitochondria. We have also demonstrated that a 3 kb dsRNA present in 55% of the isolates having dsRNA elements was a mycovirus genome. Partial sequence of that mycovirus presented high identity in nucleotide and amino acid sequence with Botrytis cinerea mitovirus 1 (BcMV1). Analysis of the genetic distance within Spanish BcMV1 sequences showed the existence of different isolates of this mitovirus inside the Spanish B. cinerea population analysed. This is the first report of the variability of dsRNA elements and the partial genome sequence of a mitovirus associated with Spanish B. cinerea isolates and the genetic diversity within Spanish isolates of BcMV1.  相似文献

Thirty-six phytohormone-affected mutants of Arabidopsis thaliana (L.) Heynh. and their parental ecotypes were tested for resistance/susceptibility to Botrytis cinerea Pers.; Fr. and ability to develop Trichoderma-mediated induced systemic resistance (ISR). Ecotype Colombia-0 (Col-0) was relatively resistant to B. cinerea, and Trichoderma harzianum Rifai T39 application at sites spatially separated (roots) from the B. cinerea inoculation (leaves) resulted in reduction of grey mold symptoms. Ecotypes Wassilewskija-4, Nossen-0 and Landsberg-0 had low levels of basal resistance to B. cinerea and were unable to express ISR. Mutants derived from ISR-non-inducible ecotypes displayed ISR-non-inducible phenotypes, whereas the ISR inducibility of mutants derived from the ISR-inducible genotype Col-0 varied according to the type of mutant. Thus, salicylic acid (SA)-impaired mutants derived from Col-0 were ISR-inducible, while ethylene/jasmonic acid (ethylene/JA)-impaired mutants of the same origin were ISR-non-inducible. SA-impaired mutants retained basal level of resistance to B. cinerea, while most ethylene/JA-impaired mutants were highly susceptible. Abscisic acid- and gibberellin-impaired mutants were highly susceptible to B. cinerea and showed ISR-non-inducible phenotypes irrespective of their lines of origin. Auxin-resistant mutants derived from Col-0 were ISR-inducible; mutant originating from Landsberg-0 and mutants which were resistant to both auxin and ethylene were ISR-non-inducible. Most of the arabidopsis genotypes which were unable to express Trichoderma-mediated ISR against B. cinerea exhibited enhanced susceptibility to this pathogen. T. harzianum treatments enhanced the growth of arabidopsis plants regardless of genotype or ISR inducibility.  相似文献
