During a four and a half year period a group of cattle was grazed continuously with a group of sheep and observed for Toxoplasma antibodies. Initially, there were 25 cattle and 31 sheep, but these were reduced to 19 and 18, respectively, by the end of the observation. Only transient antibody responses were noted in 2 cattle, whereas persistent titres developed in 12 sheep. Interpreted in the light of available information, these results were taken to indicate that cattle do not readily acquire persistent T. gondii infections. 相似文献

A serological survey of Toxoplasma gondii infection among slaughtered pigs in the largest slaughterhouse located in Taoyuan County of northwestern Taiwan was conducted using the latex agglutination (LA) test during 1998. The overall seroprevalence of T. gondii infection was 28.8% (32/111) with LA titers of 1:32 (6, 18.8%), 1:64 (10, 31.2%), 1:128 (9, 28.1%), 1:256 (6, 18.8%), and 1:512 (1, 3.1%). No significant difference (P > 0.05) in seroprevalence between male (28.6%, 20/70) and female (29.7%, 12/41) slaughtered pigs was observed. A decreasing trend in the seroprevalence among slaughtered pigs examined in the same slaughterhouse was observed because of a lower seroprevalence (P < 0.05) than that (44.4%, 128/288) previously reported about 10 yr ago using the LA test. Nevertheless, it is important to avoid eating raw or undercooked pork in order to prevent the acquisition of T. gondii infection among people in Taiwan. 相似文献

Neospora caninum is now recognized as a major cause of abortion in cattle worldwide, but there is no report of N. caninum infection in cattle in India. Serum samples from 427 dairy cattle and 32 dairy water buffaloes from 7 organized dairy farms located in Punjab, India, were tested for N. caninum antibodies using a commercial monoclonal antibody-based competitive enzyme-linked immunosorbent assay (ELISA). Antibodies to N. caninum were found in 35 of 427 cattle from 6 of the 7 farms; 9.6% of cows, 5.1% of heifers, and 5.0% of calves were seropositive, suggesting postnatal transmission of N. caninum on the farm. Antibodies to N. caninum were found in 16 of 32 buffaloes tested from 2 dairy farms. In total, 64 cattle and 16 buffalo sera already tested by ELISA were also evaluated by an indirect fluorescent antibody test (IFAT) to verify ELISA results. Of the 64 cattle samples, 29 sera were negative by both tests and of the 35 ELISA-positive sera, 12 had IFAT titers of 1:100 or higher (1 had IFAT titer of 100, 2 had IFAT titer of 200, and 9 had IFAT titers of 400 or higher). Of the 16 buffalo sera positive by ELISA, 1 had an IFAT titer of 1:400. Thus, antibodies to N. caninum were demonstrated in cattle sera by 2 serologic methods. Antibodies to Toxoplasma gondii were assayed by ELISA in 22 experimentally inoculated domestic ducks. In addition, a serological assay was carried out at Obihiro, Hokkaido, Japan, in 2004 and 2005, on 221 wild ducks of 5 species: Anas platyrhynchus (n = 111); A. poecilorhyncha (n = 27); A. acuta (n = 58); A. penelope (n = 16); and A. crecca (n = 9). Assays were also conducted using sera from 197 wild geese of 2 species, i.e., Anser albifrons (n = 162) and Ans. fabalis (n = 35). Birds were collected between 2003 and 2005 from 3 different areas: Lake Miyajima-numa, Hokkaido, Japan, regions around Anadyr city of Chukotka autonomous okrug, and Lake Makobetukoe, Kamchatka oblast, Russia. The ELISA cutoff value (OD) was > or =0.395 based on results from uninfected ducks; the final dilution ratio recognized as positive was represented by the end titer. The end titer in the experimentally infected ducks ranged from 1:400 to 1:3,200. Antibodies to T. gondii were found in 49 of the 221 wild duck samples from Japan: A. platyrhynchus (22/74); A. poecilorhyncha (2/15); A. penelope (3/16); A. acuta (4/58); and A. crecca (0/9), all in 2004. In 2005, T. gondii was found in A. platyrhynchus (13/37); and A. poecilorhyncha (5/12). Thirty-two of 197 wild goose samples were seropositive, i.e., Ans. albifrons (7/51) in 2004 and (11/72) in 2005 in Miyajima-numa, Japan and 9/39 in Chukotka, Russia as well as in Ans. fabalis (5/35) in Kamchatka, for which the end titer ranged from 1:100 to 1:3,200. In immunoblotting, the A. platyrhynchus samples showed specific IgG antibody binding to several antigens in the T. gondii lane, i.e., at 30 and 43 kDa, but not in the Neospora caninum lane. No specific bands were noted in samples for which antibody activity was not detected. These results suggest that wild waterfowl inhabiting Hokkaido, Chukotka, and Kamchatka may be exposed to T. gondii. 相似文献

Using murine chronic toxoplasmosis as an experimental model, we examined the utility of immunoenzymatic methods in recognizing reinfection in chronically infected individuals. Primary infection with avirulent Toxoplasma gondii DX strain (genotype II) induced strong immunity protecting the mice from mortality after inoculation with LD(100) of virulent BK strain (genotype I) and triggered highly expressed antibody production, within one new isotype detected by comparative immunoblots. The parasites multiplying at the site of reinfection were of BK origin as found by RAPD-PCR. The results revealed that the immunoblot assay seems to be a useful and reliable method for the monitoring of specific antibody profile in chronically infected individuals. In our opinion ELISA combined with immunoblot could enable the recognition of reinfection cases in humans, but earlier our experimental data should be verified in clinical laboratory. 相似文献

Toxoplasma gondii and Neospora caninum are two closely related protozoan parasites which can cause abortion and significant economic losses in sheep and goats. However, it is yet to know whether black-bone sheep and goats are infected with T. gondii and N. caninum in China. In the present investigation, the seroprevalence and risk factors of T. gondii and N. caninum infections in black-boned sheep and goats were investigated in Yunnan Province, subtropical southwest China between July and August of 2017. A total of 481 serum samples were tested for T. gondii antibodies using the Modified Agglutination Test (MAT), and 468 serum samples were examined for N. caninum antibodies by indirect Enzyme-Linked Immunosorbent Assay (iELISA). The overall seroprevalence of T. gondii in black-boned sheep and goats was 36.80% (177/481, 95% CI 32.49–41.11), and 40 out of 468 serum samples were N. caninum-seropositive (8.55%, 95% CI 6.02–11.08). There was significant difference in the seroprevalence of T. gondii infection in different regions (χ2 = 19.869, df = 2, P<0.01). As for the seroprevalence of N. caninum infection, region (χ2 = 8.558, df = 2, P<0.05), age (χ2 = 16.631, df = 3, P < 0.01), gender (χ2 = 11.219, df = 1, P < 0.01) and species (χ2 = 8.673, df = 1, P < 0.01) were the risk factors. In addition, the seroprevalence of coinfection of T. gondii and N. caninum in black-boned sheep and goats was 3.63% (17/468, 95% CI 1.94–5.32). To our knowledge, this is the first report of T. gondii and N. caninum seroprevalence in black-boned sheep and goats in China, which provided base-line data for the execution of control strategies and measures against T. gondii and N. caninum infection in black-boned sheep and goats. 相似文献

Prevalence of Toxoplasma gondii infection in pregnant women in Grenada is considered high. Little is known of the epidemiology of T. gondii infection in Caribbean Islands. Serum samples of 750 food animals in Grenada and Carriacou were tested for antibodies to T. gondii by the modified agglutination test (MAT). Antibodies to T. gondii (MAT, 1∶25 or higher) were found in 23.1% of 247 pigs, 44.1% of 204 sheep, 42.8% of 180 goats, and 8.4% of 119 cattle. Seroprevalence increased with age, indicating postnatal acquisition of T. gondii. Antibody titers of 1∶200 or higher were present in 65 of 90 seropositive sheep, 61 of 77 seropositive goats, and 23 of 57 seropositive pigs. However, none of the cattle had a MAT titer of 1∶200, suggesting that bovines are a poor host for T. gondii. Results indicate that pigs, sheep, and goats could be important sources of T. gondii infection if their meat is consumed undercooked. 相似文献

This article reports the first national serological prevalence of Toxoplasma gondii in India. In total, 23,094 serum samples were tested for T. gondii IgG and IgM antibodies with the use of a solid-phase immunocapture ELISA. Antibodies (IgG) were found in 24.3%; IgM antibodies were detected in 2% of the samples. The lowest seroprevalences were in the northern parts of India, with the highest in the south. These data probably reflect the effects of significantly drier conditions and, therefore, a negative impact on the survivability of T. gondii oocysts. 相似文献

The prevalence of antibodies to Toxoplasma gondii was determined in blood and tissue exudates recovered from the spleens of 41 wolverines (Gulo gulo) collected in Nunavut, Canada, using a modified agglutination test (MAT). Antibodies to T. gondii were found in 17 (41.5%) of the 41 wolverines with MAT titers of 1:25 in 1, 1:50 in 4, 1:100 in 5, 1:200 in 6, and 1:400 in 1. To investigate the prevalence of Toxoplasma gondii infection in free-ranging Eurasian lynx (Lynx lynx) in Sweden, serosanguinous fluids and feces were collected from 207 carcasses of lynx killed or found dead from 1996 to 1998. Sera were tested for antibodies against T. gondii by the direct agglutination test, and 156 (75.4%) of the sera tested positive at antibody titers>or=40. Antibody prevalence was significantly lower in lynx originating from the northern parts of Sweden than in lynx from the more southern regions that are more densely populated by humans. Age-related differences also were found, with a significantly lower prevalence (55%) in juvenile (<1-yr-old) than in subadult and adult animals (82%). There was no significant difference in seroprevalence between males and females. Oocysts typical of T. gondii were not detected in any of the fecal samples. 相似文献

Toxoplasma gondii and Neospora caninum are closely related cyst-forming parasites identified as important causes of reproductive failures in ruminants. While these parasites have been reported worldwide, seroprevalence and associated risk factors for cattle infections have not been determined in India. A total of 576 serum samples of cattle were analyzed for antibodies to T. gondii and N. caninum using enzyme-linked immunosorbent assay (ELISA), modified/Neospora agglutination test (MAT/NAT), and an indirect fluorescent antibody test (IFAT-tachyzoite and bradyzoite). Additionally, general information about cattle, movement of cats and dogs, the menace of rodents, management, and reproductive disorders were assessed to identify the potential risk factors. Overall, 32.9% (190/576) serum samples reacted positively to T. gondii and 24.8% (143/576) to N. caninum. The performance of the diagnostic tests showed excellent agreement between IFAT and ELISA (kappa [κ] = 0.98) and between MAT/NAT and ELISA (κ = 0.97). Combining both infections on avidity test, 94% sera had high-IgG avidity, and 3% had low-IgG avidity antibodies, indicating chronic infection in the majority of the cases. The identified risk factors (p < 0.05) for exposure to T. gondii were: increasing age (Odds Ratio [OR]: 2.02), movement of cat (OR: 4.8) and rodents (OR: 1.57) in the farm; and for N. caninum: increasing age (OR: 1.6), movement of dogs in the farm (OR: 2.07), drinking pond water (OR: 1.64) and abortion (OR: 1.82). These findings revealed that T. gondii and N. caninum infections are widespread in the study area and suggest conducting nationwide epidemiological studies owing to their economic importance. 相似文献

In the present investigation, the seroprevalence of Toxoplasma gondii antibodies in 455 Tibetan sheep in Tibet, China, was examined using an indirect hemagglutination test. Of these, 26 (5.7%) Tibetan sheep were seropositive at the cut-off of 1:64 serum dilution. The seroprevalence ranged from 2.2% to 8.9% among Tibetan sheep of <1-yr-old, 1-3-yr-old, and >3-yr-old, but the differences among the age groups were not significant (P > 0.05). The prevalence in male Tibetan sheep (2.8%) was lower than that in female Tibetan sheep (6.6%), but the difference was not statistically significant (P > 0.05). The results of this survey indicated the presence of T. gondii infection in Tibetan sheep, which may cause economic losses to the local livestock industry and which poses a potential threat to human health in this area. 相似文献

Genotypes of Toxoplasma gondii in naturally infected tissues from 13 ovine abortions from 10 farms in the United Kingdom, from 2 wild rodents captured on 1 farm, and from 2 isolates in laboratory mouse brains (made from the hearts of healthy, infected lambs) all conformed to the type II lineage determined by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) analysis of the SAG2 gene. PCR was applied directly to tissue extracts without parasite isolation or DNA extraction. The aim of the present study was to determine the seroprevalence of Toxoplasma gondii in horses used for sporting purposes in the Province of Riyadh, Saudi Arabia. In total, 266 serum samples from clinically healthy horses were analyzed for anti- T. gondii antibodies using the Sabin-Feldman dye test. Antibodies to T. gondii were found in 84 (31.6%) horses, with specific titers of 1∶16 (78 with a prevalence of 29.3%), 1∶64 (4 with a prevalence of 1.5%), and 1∶256 (2 with a prevalence of 0.8%). The number of seropositive horses in Shaqra (43.7%) was considerably higher than in other regions, with the lowest occurring in Al-Majmaah (11.0%). However, the differences in seroprevalence between the 6 locations were not statistically significant (P > 0.05). In contrast, the overall seroprevalence in Riyadh province was higher than that reported in other countries. Horses are not a direct source of infection for human toxoplasmosis in this region. Usually human infection is facilitated via cats, which are the reservoir hosts for this parasite. 相似文献

Prevalence of antibodies to Toxoplasma gondii was investigated in 673 domestic dogs from northeastern Portugal, using the modified agglutination test (MAT) with 1 : 20 as cutoff for seropositivity; antibodies were found in 256 dogs (38.0%). Differences between seroprevalence levels in males (36.7%) and females (41.8%) and between pure-breed (42.1%) and mixed-breed dogs (35.2%) were not statistically significant. Multiple logistic regression analysis identified age above 12 mo (odds ratio [OR] = 4.0), chance of eating birds or small mammals (OR = 4.0), housing exclusively outdoors (OR = 1.5), home-cooked meals (OR = 3.0), and eating raw meat or viscera (OR = 7.7) as risk factors for the canine T. gondii infection. Twelve species of free-living African mammals from Kenya, Tanzania, Uganda and Zambia were tested for antibodies to Toxoplasma gondii using the indirect hemagglutination test. Of 157 animals sampled, 20 (13%) were seropositive. T. gondii antibodies were detected in Burchell's zebra, (Equus burchelli), hippopotamus (Hippopotamus amphibius), African elephant (Loxodonta africana), defassa waterbuck (Kobus defassa), lion (Panthera leo), and rock hyrax (Procavia capensis), The highest titers were found in elephants, two having titers of 1:4096 and one of 1:8192. Ground-feeding birds are considered important in the epidemiology of Toxoplasma gondii because they serve as indicators of soil contamination by oocysts, and birds of prey are indicators of T. gondii prevalence in rodents and other small mammals. Cats excrete environmentally resistant oocysts after consuming tissues of T. gondii -infected birds. In the present study, sera and tissues from 382 wild birds from Colorado were tested for T. gondii infection. Antibodies to T. gondii were found in 38 birds with the use of the modified agglutination test (MAT, 1∶25 titer). Tissues (brains, hearts) of 84 birds were bioassayed in mice. Viable T. gondii was isolated from 1 of 1 barn owl (Tyto alba), 1 of 5 American kestrels (Falco sparverius), 1 of 7 ferruginous hawks (Buteo regalis), 1 of 4 rough-legged hawks (Buteo lagopus), 2 of 13 Swainson's hawks (Buteo swainsoni), and 1 of 25 red-tailed hawks (Buteo jamaicensis). The aim of this study was to estimate the prevalence of Toxoplasma gondii infection in dogs in Shanghai, China. A total of 1,736 sera (1,178 from the city center and 558 from the outskirts) was collected from healthy dogs and tested for T. gondii infection by indirect hemagglutination; 56 sera (3.2%) were considered positive, with titers > 1∶64. The seroprevalence in dogs from the outskirts of the city (town dogs, 6.0%; countryside dogs, 9.8%) was significantly higher (P > 0.05) than that in city center dogs (1.3%). The age of the dog has an apparent association with T. gondii infection; that is, the seroprevalence ranged from 1.9% (in dogs ≤ 1 year old) to 3.6% (in dogs > 5 years old). There was no significant difference in gender (P ≥ 0.05), that is, 1.4% versus 1.1% for male and female dogs in the city center, 6.2% versus 5.9% in town dogs, and 8.4% versus 11.5% in country dogs, respectively. These results suggest that T. gondii infections are common in dogs from the city center and outskirts of Shanghai, but the T. gondii seroprevalence in dogs is considerably lower than in other regions in PR China. The presence of T. gondii DNA was investigated by nested PCR on 110 blood samples from city center dogs, but no positive samples were found, which may suggest that there were no acute infections of T. gondii in the city center samples. Our results indicate that the control and treatment of toxoplasmosis in Shanghai has been effective. However, it is still essential to further implement integrated strategies to prevent and control T. gondii infection in dogs in both the city center and the outskirts. 相似文献
