By site-directed mutagenesis near the axial ligand, Cys456, of cytochrome P-450d (P-450d), we obtained five stable P-450d mutants, i.e. mutant (B), Gly450Ser; mutant (E), Lys453Glu; mutant (G), Arg455Gly; mutant (L), Glu459Ala and mutant (M), Ile460Ser. Bindings of aniline and fluoroaniline to the wild type P-450d and these five P-450d mutants were studied with difference absorption spectra at the Soret region. The following results were obtained: (i) binding constants (Kb) of fluoroaniline to wild P-450d were higher than those to cytochrome P-450scc by more than one order; (ii) Kb value of para-fluoroaniline to the wild P-450d was higher than those of aniline, ortho- or meta-fluoroaniline by two orders; (iii) Kb values of para-fluoroaniline to five mutants were lower than that to the wild type by more than one order; (iv) Kb values of para-fluoroaniline to the mutants (E) and (G) were the lowest among those to the five mutants and were lower than that to the wild type by two orders; (v) Kb values of aniline, ortho- and meta-fluoroaniline to mutant (B) were one order higher than those to the wild and other four mutants. These results indicate that the binding of the external axial ligand to the heme iron of P-450d is remarkably influenced by mutations at the proximal site possibly due to change in the polarity of the distal site and/or alteration of the secondary protein structure at the distal site.  相似文献

Increasing areas of Amazonian forest are coming under flood stress due to dam construction and greater variability in river flood levels due to climate change. The physiological responses of Amazonian trees subjected to flooding are important to understand the consequences of these changes. Irradiance response curves for photosynthesis obtained from ten tropical tree species growing in flooded areas were used to fit three empirical models. The study was done in floodplains along the Uatumã River, both upstream and downstream of the Balbina Hydroelectric Dam in Brazil’s state of Amazonas (01°55′S; 59°28′W). Ten species were studied. Models compared were: non-rectangular hyperbola, rectangular hyperbola, and exponential. All models were quantitatively adequate for fitting the response of measured data on photosynthesis to irradiance for all ten species in the non-flooding and flooding periods. Considerable variation was found among the model estimates of maximum photosynthesis (P nmax), dark respiration (R d) and apparent quantum yield of photosynthesis (α). For photosynthesis, the two hyperbolas overestimated P nmax while EXP presented more realistic values. For estimating R d, RH presented the most realistic values. To avoid unrealistic value estimates of R d, we recommend adding measured R d values to the regressions. The results suggest that the EXP model presented the most realistic P nmax and α values, and, in spite of less accuracy in fitting photosynthetic irradiance curves than the RH model, it can be recommended for accessing the information used in photosynthetic irradiance curves for the leaves of tropical trees growing in Amazonian floodplains or in areas that are artificially flooded by dams.  相似文献

A partition chromatographic procedure utilizing a cationic exchange resin column in the Li+ form and 90% ethanol as the mobile phase was employed to quantify 3-deoxy-d-manno-octulosonic acid (KDO) and l-glycero-d-manno-heptose in the lipopolysaccharides (LPS) of Re and RdP? rough mutants of Salmonella minnesota. In a standard mixture of monosaccharides, KDO eluted shortly after the void volume and heptose eluted after the neutral hexoses. Mild acid treatment of either the Re or RdP? LPS with 0.16 n methanesulfonic acid in the presence of Dowex 50-X8 resin (H+ form) released more than 80% of the KDO residues within 15 min. The heptose of the RdP? LPS, first detected after 90 min of hydrolysis, increased gradually to a maximum level at 12 h. A secondary gradual increase in KDO became apparent during the heptose release. The weight contents of these two monosaccharides based upon aheir maximum values detected during hydrolysis were 20.3 ± 0.6% KDO, for the Re LPS, and 13.8 ± 0.4% KDO and 12.0 ± 0.4% heptose, for the RdP? LPS. The relationship between the kinetics of release of KDO and heptose and the nature of the linkages involving these two monosaccharides are discussed.  相似文献

A minor form of hepatic microsomal cytochrome P-450 has been purified to apparent homogeneity from rats treated with the polychlorinated biphenyl mixture, Aroclor 1254. This newly isolated hemoprotein, cytochrome P-450e, is inducible in rat liver by Aroclor 1254 and phenobarbital, but not by 3-methylcholanthrene. Two other hemoproteins, cytochromes P-450b and P-450c, have also been highly purified during the isolation of cytochrome P-450e based on chromatographic differences among these proteins. By Ouchterlony double-diffusion analysis with antibody to cytochrome P-450b, highly purified cytochrome P-450e is immunochemically identical to cytochrome P-450b but does not cross-react with antibodies prepared against other rat liver cytochromes P-450 (P-450a, P-450c, P-450d) or epoxide hydrolase. Purified cytochrome P-450e is a single protein-staining band in sodium dodecyl sulfate-polyacrylamide gels with a minimum molecular weight (52,500) slightly greater than cytochromes P-450b or P-450d (52,000) but clearly distinct from cytochromes P-450a (48,000) and P-450c (56,000). The carbon monoxide-reduced difference spectral peak of cytochrome P-450e is at 450.6 nm, whereas the peak of cytochrome P-450b is at 450 nm. Ethyl isocyanide binds to ferrous cytochromes P-450e and P-450b to yield two spectral maxima at 455 and 430 nm. At pH 7.4, the 455:430 ratio is 0.7 and 1.4 for cytochromes P-450b and P-450e, respectively. Metyrapone binds to reduced cytochromes P-450e and P-450b (absorption maximum at 445–446 nm) but not cytochromes P-450a, P-450c, or P-450d. Metabolism of several substrates catalyzed by cytochrome P-450e or P-450b reconstituted with NADPH-cytochrome c reductase and dilauroylphosphatidylcholine was compared. The substrate specificity of cytochrome P-450e usually paralleled that of cytochrome P-450b except that the rate of metabolism of benzphetamine, benzo[a]pyrene, 7-ethoxycoumarin, hexobarbital, and testosterone at the 16α-position catalyzed by cytochrome P-450e was only 15–25% that of cytochrome P-450b. In contrast, cytochrome P-450e catalyzed the 2-hydroxylation of estradiol-17β more efficiently (threefold) than cytochrome P-450b. Cytochrome P-450d, however, catalyzed the metabolism of estradiol-17β at the greatest rate compared to cytochromes P-450a, P-450b, P-450c, or P-450e. The peptide fragments of cytochromes P-450e and P-450b, generated by either proteolytic or chemical digestion of the hemoproteins, were very similar but not identical, indicating that these two proteins show minor structural differences.  相似文献

The Ah locus regulates the induction of cytochrome P1-450 by foreign chemicals such as 3-methyl-cholanthrene and 2,3,7,8-tetrachlorodibenzo-p-dioxin. The induction process is controlled by the cytosolic Ah receptor. The cytosolic and nuclear Ah receptors were studied in the liver from inbred C57BL/6N (Ahb/Ahb) mice, inbred DBA/2N (Ahd/Ahd) mice and heterozygotes (Ahb/Ahd) and homozygotes (Ahd/Ahd) derived from the (C57BL/6N × DBA/2N)F1 × DBA/2N backcross. After [3H-1,6]-2,3,7,8-tetrachlorodibenzo-p-dioxin (3H-TCDD) is given in vivo, the receptor in Ahb/Ahb and Ahb/Ahd mice is detectable in the cytoplasm and nucleus; in Ahd/Ahd mice the receptor is not measurable in the cytosol, but is found in the nucleus at levels one fourth to one fifth of those in Ahb/Ahb mice. P1-450 (23S) mRNA content was estimated by Northern hybridization and by Rot analysis with a mouse P1-450 cloned cDNA. An excellent dose-response relationship (r = 0.99) was found between the amount of 3H-TCDD-Ah receptor complex appearing in the nucleus and the quantity of P1-450 mRNA induced in mice with all three possible Ah genotypes.  相似文献

The paper considers the properties of individual life history corresponding to the Leslie model of age-structured population. The life history is modelled as a finite Markov chain with absorption at a death state of individual. In this model, individual longevity, average number of offspring R L (produced by an individual over the entire life), and some other known characteristics of the life history have been derived using simple probability methods that do not involve matrix calculus and their individual components have been interpreted. In the linear Leslie population model (derived by simple modification of a Markov chain), R L determines the growth or decline of a population. Individuals with higher R L values have evolutionary advantages in the population due to accelerated growth in their number. The selection of fertility as a factor of the increase in R L is considered. In the Leslie model, fertility is a set of correlated quantitative traits, where the age-specific fertility components are determined both by multiple loci and the environment. According to the genetic theory of quantitative trait selection, they evolve towards an increase in R L . Taking into account the limited resources for reproduction, selection optimizes the fertility distribution according to age. Optimal distribution corresponds to the attainment of the maximum R L . This complies with the maximization of the rate of population growth (r-selection), which is characteristic of linear population models. The search for the R L maximum and optimal distribution of fertility belongs to the field of linear programming.  相似文献

At evolutionary equilibrium, ecological factors will determine the optimal combination of life-history trait values of an organism. This optimum can be assessed by assuming that the species maximizes some criterion of fitness such as the Malthusian coefficient or lifetime reproductive success depending on the degree of density-dependence. We investigated the impact of the amount of resources and habitat stability on a plant's age at maturity and life span by using an evolutionary optimization model in combination with empirical data. We conducted this study on sea beet, Beta vulgaris subsp. maritima, because of its large variation in life span and age at first reproduction along a latitudinal gradient including considerable ecological variation. We also compared the consequence in our evolutionary model of maximizing either the Malthusian coefficient or the lifetime reproductive success. Both the data analysis and the results of evolutionary modeling pointed to habitat disturbance and resources like length of the growing season as factors negatively related to life span and age at maturity in sea beet. Resource availability had a negative theoretical influence with the Malthusian coefficient as the chosen optimality criterion, while there was no influence in the case of lifetime reproductive success. As suggested by previous theoretical work the final conclusion on what criterion is more adequate depends on the assumptions of how in reality density-dependence restrains population growth. In our case of sea beet data R0 seems to be less appropriate than λ.  相似文献

Several reproductive triats in plants were studied in more than 200 populations of 61 wild species from diverse ecological conditions. As a result, it was found that there occur three distinct types of plants in the energy allocation patterns to reproductive structures (RA) and the propagule output per plant (PN), i.e. (1) the number of propagules per plant increases in response to the increase in RA (Type I), (2) the number of propagules decreases in response to the increase in RA (Type II), and (3) the RA remains constant despite the great differences in the propagule number per plant. A conspicuous trade-off relationship was also discovered to occur between the RA to a single propagule (RA) and the propagule output per plant (PN), such that log RA=logC−blot PN, or RA=C/PN b =CPN b , where C is a constant. The three different ranges ofb-values were recognized, i.e.b<1.0,b>1.0, andb=1.0, which correspond to Type I, Type II, and Type III, respectively. Related problems to the concept ofr- andK-strategy are also discussed.  相似文献

Numerous species of diatoms liberate oxylipins including polyunsaturated aldehydes (PUAs) in response to cellular damage such as may occur during grazing. PUAs are cyto- and genotoxic and negatively disrupt reproductive processes in copepods, their principal grazers, although experimental evidence would suggest that the grazer response may be species specific. The reproduction of the benthic harpacticoid copepod Tisbe holothuriae was compared over two generations. Copepods were reared using four diet treatments: PUA-producing diatom strains Skeletonema marinoi (Adriatic Sea Isolate FE6) and Melosira nummuloides (CCAP 1048/6); and non-PUA-producing diatom strains Phaeodactylum tricornutum (CCAP 1052/A) and S. marinoi (Seasalter (Walney) Ltd). Life tables were generated for each treatment using measured reproductive parameters and the net reproductive rate (R0) calculated. No significant differences were observed between the individual reproductive parameters of T. holothuriae fed PUA-producing diatoms compared to those fed non-PUA-producing diatoms although diets of P. tricornutum resulted in some decreases in individual reproductive parameters in the second generation. There were no significant differences in the R0 values between the four tested diets. These observations indicate that T. holothuriae exhibits a tolerance of known PUA-producing diatom diets that has not been similarly demonstrated in pelagic calanoid copepods. Harpacticoid copepods may have a greater capacity to detoxify diatom oxylipins than their planktonic calanoid counterparts.  相似文献

The capacity for an ectothermic reptile to thermoregulate has implications for many components of its life history. Over two years, we studied thermoregulation in a population of Midland painted turtles (Chrysemys picta marginata) in a shallow, thermally variable wetland during summer in Northern Michigan. Mean body temperature (Tb) of free-ranging turtles was greater in 2008 (25.8 °C) than in 2010 (19.7 °C). Laboratory determined thermoregulatory set point (Tset) ranged from 25 °C (Tset-min) to 31 °C (Tset-max) and was lower during the fall (17–26 °C). Deviations of Tb distributions from field measured operative temperatures (Te) and indices of thermoregulation indicated that C. picta marginata were capable of a limited degree of thermoregulation. Operative temperatures and thermal quality (de=|Tset-minTe| and |TeTset-max|) cycled daily with maximal thermal quality occurring during late morning and late afternoon. The accuracy of thermoregulation (db=|Tset-minTb| and |TbTset-max|) was maximal (db values were minimal) as Tb declined and traversed Tset during the late afternoon–early evening hours and was higher on cloudy days than on sunny days because relatively low Te values decreased the number of Tb values that were above Tset. Our index of thermal exploitation (Ex=frequency of Tb observations within Tset) was 36%, slightly lower than that reported for an Ontario population of C. picta marginata. Regression of db (thermal accuracy) on de (thermal quality) indicated that turtles invested more in thermoregulation when thermal quality was low and when water levels were high than when they were low. There were no intersexual differences in mean Tb throughout the year but females had relatively high laboratory determined Tb values in the fall, perhaps reflecting the importance of maintaining ovarian development prior to winter.  相似文献

Complex formation between the phenobarbital-inducible form of rabbit liver microsomal cytochrome P-450 incorporated into phosphatidylcholine and detergent-solubilized cytochrome b5 is associated with a low-to-high spin transition of the former pigment. It is concluded that the proteins combine in a 1:1 molar ratio. CD spectral analysis in the far uv region reveals that interaction of the cytochromes results in a conformational change of one or both hemoproteins. Such a cytochrome b5-induced structural alteration of the reconstituted enzyme system is accompanied by an increase in affinity of 4-chloroaniline for cytochrome P-450, as measured in terms of cumene hydroperoxide-supported N-oxidation of the arylamine; the maximum velocity of the catalytic process remains unchanged. Similarly, incorporation into the assay media of cytochrome b5 decreases the apparent Kd values of both the amine substrate and the oxygen donor, as determined by optical titration. Stopped-flow spectrophotometric studies on the influence of cytochrome b5 on the kinetics of binding to cytochrome P-450 of 4-chloroaniline and/or cumene hydroperoxide show that the rates of formation and decay of the adducts change as the molar ratio of cytochrome b5 to cytochrome P-450 varies. Moreover, cytochrome b5 modifies the activation energies required for production of the substrate-bound oxy complex. These findings suggest that cytochrome b5, apart from its well-known role as an electron carrier, might exert an effector function in the cytochrome P-450 system.  相似文献

Cytochrome bd is a terminal quinol:O2 oxidoreductase of respiratory chains of many bacteria. It contains three hemes, b558, b595, and d. The role of heme b595 remains obscure. A CO photolysis/recombination study of the membranes of Escherichia coli containing either wild type cytochrome bd or inactive E445A mutant was performed using nanosecond absorption spectroscopy. We compared photoinduced changes of heme d-CO complex in one-electron-reduced, two-electron-reduced, and fully reduced states of cytochromes bd. The line shape of spectra of photodissociation of one-electron-reduced and two-electron-reduced enzymes is strikingly different from that of the fully reduced enzyme. The difference demonstrates that in the fully reduced enzyme photolysis of CO from heme d perturbs ferrous heme b595 causing loss of an absorption band centered at 435 nm, thus supporting interactions between heme b595 and heme d in the di-heme oxygen-reducing site, in agreement with previous works. Photolyzed CO recombines with the fully reduced enzyme monoexponentially with τ ∼ 12 μs, whereas recombination of CO with one-electron-reduced cytochrome bd shows three kinetic phases, with τ ∼ 14 ns, 14 μs, and 280 μs. The spectra of the absorption changes associated with these components are different in line shape. The 14 ns phase, absent in the fully reduced enzyme, reflects geminate recombination of CO with part of heme d. The 14-μs component reflects bimolecular recombination of CO with heme d and electron backflow from heme d to hemes b in ∼ 4% of the enzyme population. The final, 280-μs component, reflects return of the electron from hemes b to heme d and bimolecular recombination of CO in that population. The fact that even in the two-electron-reduced enzyme, a nanosecond geminate recombination is observed, suggests that namely the redox state of heme b595, and not that of heme b558, controls the pathway(s) by which CO migrates between heme d and the medium.  相似文献

Soybean plants (Glycine max [L.] Merr. cv Clark) carrying nuclear and cytoplasmic “stay-green” mutations, which affect senescence, were examined. Normally, the levels of chlorophyll (Chl) a and b decline during seedfill and the Chl a/b ratio decreases during late pod development in cv Clark. Plants homozygous for both the d1 and d2 recessive alleles, at two different nuclear loci, respectively, retained most (64%) of their Chl a and b and exhibited no change in their Chl a/b ratio. Combination of G (a dominant nuclear allele in a third locus causing only the seed coat to stay green during senescence) with d1d2 further inhibited the loss of Chl in the leaf. Whereas the thylakoid proteins seem to be degraded in normal Clark leaves during late pod development, they were not substantially diminished in d1d2 and Gd1d2 leaves. In plants carrying a cytoplasmic mutation, cytG, Chl declined in parallel with normal cv Clark; however, the cytG leaves had a much higher level of Chl b, and somewhat more Chl a, remaining at abscission, enough to color the leaves green. In cytG, most thylakoid proteins were degraded, but the Chl a/b-binding polypeptides of the light-harvesting complex in photosystem II (LHCII), and their associated Chl a and b molecules, were not. Thus, the combination of d1 and d2 causes broad preservation of the thylakoid proteins, whereas cytG appears to selectively preserve LHCII. The cytG mutation may be useful in elucidating the sequence of events involved in the degradation of LHCII proteins and their associated pigments during senescence.  相似文献

Annual reproductive cycle and spawning ecology of Kelee shad, Hilsa kelee were studied. The sex ratios did not exhibit as 1: 1; whereas 69.19% were females. On the basis of histological examination, the maturity of female gonads of H. kelee was categorized in eight developmental stages: immature, developing, fully developed, hydrated, running ripe, partially spent, spent and post spawning. Whereas males of this species were categorized in five developmental stages: immature, developing, ripe, partially spent and spent. This fish is repeatedly spawner and spawns twice per year: in April to June and August to October. The values of exponent b in the length weight regressions (W = al b ) slightly varied in males b = 3.133, R 2 = 0.96 and P < 0.01 and females b = 3.143, R 2 = 0.95 and P < 0.001. The linear regressions of length-weight relationships were significant for both males and females.  相似文献

The structural elucidation of the minor carotenoid sulfates from the marine sponge lanthella basta is discussed in context with the structure assigned to the major sulfate bastaxanthin (c; 3,19,17′-trihydroxy-7,8-didehydro-β-κ-carotene-3′,6′-dione 3-sulfate. Plausible structures are assigned to other bastaxanthins (b,b2, c2, d, e and f) on the basis of electroic, IR, 1H NMR, mass and CD spectra, electrophoretic behaviour, chemical derivatization and enzymatic or acid-catalysed hydrolysis. The minor sulfates represent structural variation in the cylopentane end group with different oxidation levels. Bastaxanthol b (desulfated bastaxanthin b) was a minor carotenoid constituent of l. basta. Including tentative chiralities, the structures favoured for the bastaxanthins are: c2, (3R,3′R, 5′R)-3,19,3′-trihydroxy-7,8-didehydro-β,κ-caroten-6′-one 3-sulfate; b2, (3R,3′R,5′R)-3, 19-dihydroxy-7,8-didehydro-β,κ- dione 3-sulfate; b, (3R,1′R, 5′R)-3, 19-dihydroxy 3′,6′-dioxo-7,8-didehydro-β,κ-caroten-17′-al 3-sulfate; d. (3R,1′R,3′R,5′R)-3, 19,3′,17′-tetrahydroxy 7,8 didehydro-β,κ-caroten-6′-one 3-sulfate; e. hydrogen (3R,1′R,5′R)-3, 19-dihydroxy-3′,6′-dioxo-7,8-didehydro-β,κ-caroten-17′-oate 3 sulfate (?); and f, hydrogen (3R.1′R,3′R,5′R)-3,19,3′-trihydroxy-7,8-didehydro-6′-oxo-β,κ-caroten-17′-oate 3-sulfate; for bastaxanthol b(3R.1′R.5′R)-3, 19-dihydroxy-3′,6′-dioxo-7,8-didehydro-β,κ-caroten-17′-al. The bastaxanthins are considered as metabolic products of l. basta, diadinoxanthin of phytoplankton origin representing a plausiable precursor.  相似文献

The carbon balances of whole, 21-d old French bean plants (Phaseolus vulgaris L.) grown in standard nutrient solution (1K) and its modifications without (OK) or surplus (2K) potassium were calculated from the daily photosynthetic carbon inputs of individual leaves, and the daily respiratory carbon losses by individual leaves, stalks and petioles, and roots. Under the three K concentrations, maximum net photosynthetic rates (Pn) were found in the 2nd or in the 3rd trifoliate leaves, maximum respiratory rates (Rd) in the youngest, 4th trifoliate leaves; the Pn/Rd ratio decreased with leaf age. In all leaves of 2K plants, leaf dry masses and thicknesses, Pn, Pn/Pd ratios, and stomatal and intracellular conductances were lower than in OK and IK plants. Daily whole-plant net carbon gain was highest in IK plants, whereas in OK and 2K plants it was 98.0 and 81.3 % of IK, respectively. Similar values were found in the parameters of growth analysis, namely in net assimilation rates and relative growth rates. No differences were found in water potential (Ψ w ) or water saturation deficit (Wsat) in the OK, 1K and 2K plants sufficiently supplied with water or during wilting and resaturation. The decrease in Ψw to −0.97 MPa was associated with a 19.9 %, 31.4 % and 23.4 % decrease in Pn of OK, 1K and 2K plants, respectively, but no effect on Rd was found. In the three variants, the short-time effect of mild water stress was fully reversible.  相似文献

Theory predicts that sexual reproduction can increase population viability relative to asexual reproduction by allowing sexual selection in males to remove deleterious mutations from the population without large demographic costs. This requires that selection acts more strongly in males than females and that mutations affecting male reproductive success have pleiotropic effects on population productivity, but empirical support for these assumptions is mixed. We used the seed beetle Callosobruchus maculatus to implement a three‐generation breeding design where we induced mutations via ionizing radiation (IR) in the F0 generation and measured mutational effects (relative to nonirradiated controls) on an estimate of population productivity in the F1 and effects on sex‐specific competitive lifetime reproductive success (LRS) in the F2. Regardless of whether mutations were induced via F0 males or females, they had strong negative effects on male LRS, but a nonsignificant influence on female LRS, suggesting that selection is more efficient in removing deleterious alleles in males. Moreover, mutations had seemingly shared effects on population productivity and competitive LRS in both sexes. Thus, our results lend support to the hypothesis that strong sexual selection on males can act to remove the mutation load on population viability, thereby offering a benefit to sexual reproduction.  相似文献

In this study, on the basis of empirical data, we have simulated the foraging tunnel patterns of two subterranean termites, Coptotermes formosanus Shiraki and Reticulitermes flavipes (Kollar), using a two-dimensional model. We have defined a territory as a convex polygon containing a tunnel pattern and explored the effects of competition among termite territory colonies on the territory size distribution in the steady state that was attained after a sufficient simulation time. In the model, territorial competition was characterized by a blocking probability Pblock that quantitatively describes the ease with which a tunnel stops its advancement when it meets another tunnel; higher Pblock values imply easier termination. In the beginning of the simulation run, N=10, 20,…,100 territory seeds, representing the founding pair, were randomly distributed on a square area. When the territory density was less (N=20), the differences in the territory size distributions for different Pblock values were small because the territories had sufficient space to grow without strong competitions. Further, when the territory density was higher (N>20), the territory sizes increased in accordance with the combinational effect of Pblock and N. In order to understand these effects better, we introduced an interference coefficient γ. We mathematically derived γ as a function of Pblock and N: γ(N,Pblock)=a(N)Pblock/(Pblock+b(N)). a(N) and b(N) are functions of N/(N+c) and d/(N+c), respectively, and c and d are constants characterizing territorial competition. The γ function is applicable to characterize the territoriality of various species and increases with both the Pblock values and N; higher γ values imply higher limitations of the network growth. We used the γ function, fitted the simulation results, and determined the c and d values. In addition, we have briefly discussed the predictability of the present model by comparing it with our previous lattice model that had been used to explain the territory size distributions of mangrove termites on the Atlantic coast of Panama.  相似文献

Cytochrome bd is a terminal component of the respiratory chain of Escherichia coli catalyzing reduction of molecular oxygen to water. It contains three hemes, b558, b595, and d. The detailed spectroelectrochemical redox titration and numerical modeling of the data reveal significant redox interaction between the low-spin heme b558 and high-spin heme b595, whereas the interaction between heme d and either hemes b appears to be rather weak. However, the presence of heme d itself decreases much larger interaction between the two hemes b. Fitting the titration data with a model where redox interaction between the hemes is explicitly included makes it possible to extract individual absorption spectra of all hemes. The α- and β-band reduced-minus-oxidized difference spectra agree with the data published earlier ([22] J.G. Koland, M.J. Miller, R.B. The Soret band spectra show λmax = 429.5 nm, λmin ≈ 413 nm (heme b558), λmax = 439 nm, λmin ≈ 400 ± 1 nm (heme b595), and λmax = 430 nm, λmin = 405 nm (heme d). The spectral contribution of heme d to the complex Soret band is much smaller than those of either hemes b; the Soret/α (ΔA430A629) ratio for heme d is 1.6.  相似文献
