The structure of horse methaemoglobin at 2-0 A resolution   总被引:13,自引:0,他引:13  
The structure of horse methaemoglobin has been redetermined by phase extension and refinement. This has improved our knowledge of the haem geometry and the stereochemistry of the interfaces between the subunits, and confirmed the disorder of the C-terminal residues. Using new four-circle diffractometer data between the limiting spheres of radius 10 and 2.0 Å?1, the co-ordinates determined by Perutz et al. (1968a,b) were subjected to successive cycles of real-space refinement into electron density maps calculated with observed ¦F¦ values and phases derived from the latest refined model, until the reliability index had dropped from an initial value of 0.45 to 0.23. The positions of the iron atoms relative to the planes of the porphyrin rings were refined separately, and checked by Fourier syntheses based on anomalous scattering and by difference Fourier syntheses calculated with coefficients from which the iron contributions had been removed. The general root-mean-squared error in atomic positions is 0.32 Å; the probable error in the displacement of the iron atoms from the porphyrin planes is 0.06 Å. The difference Fourier synthesis, obtained after refinement of the protein was complete, showed 41 bound water molecules per asymmetric unit and also revealed five errors in amino acid sequence, one of which was confirmed chemically.The secondary structures of the subunits are stabilized by hydrogen bonds formed by main-chain NH and CO groups either with each other or with nearby polar side-chains. There are few internal hydrogen bonds linking the various chain segments; many of the external polar side-chains help to stabilize the tertiary structure by forming hydrogen bonds with each other or through bound water molecules. Several of the helical segments are irregular and the terminal residues are disordered. The contacts between the subunits are more polar than the earlier 2.8 Å map had led us to believe, because it had failed to show up the 15 bound water molecules at the α1β1 and the four at the α1β2 contact. Their inclusion has raised the number of hydrogen bonds between neighbouring subunits at α1β1 from five to 17 or possibly 19, and at α1β2 from two to six or possibly seven. The remaining 22 water molecules are distributed over the internal cavity and the molecular surface; most of them make hydrogen bonds with at least two polar groups of the protein. Despite several amino acid differences, the structure of the α1β1 contact, including the bound water, is the same as in human deoxyhaemoglobin (Fermi, 1975).  相似文献

Schizosaccharomyces pombe whole-cell glycoproteins, previously depleted of N-linked glycans by sequential treatment with endo-ss-N-acetylglucosaminidase H and peptide-N4-asparagine amidohydrolase F, were ss-eliminated with 0.1 M NaOH/1 M NaBH4 to release the O-linked oligosaccharides. The saccharide-alditols were separated by gel-exclusion chromatography into pools from Hexitol to Hex4Hexitol in size. Analysis of the Hexitol pool indicated Man to be the only sugar linked to Ser or Thr residues. The Hex1Hexitol pool contained two components, Galalpha1,2Man-ol (2A) and Manalpha1, 2Man-ol (2B). The Hex2Hexitol pool contained two components, Galalpha1,2Manalpha1,2Man-ol (3A) and Manalpha1,2Manalpha1,2Man-ol (3B). The two Hex3Hexitol components were Galalpha1,2(Galalpha1, 3)Manalpha1,2Man-ol (4A) and Manalpha1,2(Galalpha1,3)Manalpha1, 2Man-ol (4B). The Hex4Hexitol component was found to be a single isomer with the composition of Galalpha1,2(Galalpha1,3)Manalpha1, 2Manalpha1,2Man-ol (5AB). Surprisingly, galactobiose was not detected in any of these oligosaccharides. The gma12 (T. G. Chappell and G. Warren (1989) J. Cell Biol., 109, 2693-2707) and gth1 (T. G. Chappell personal communication) alpha1, 2-galactosyltransferase-deficient mutants and the gma12/gth1 double mutant S.pombe strains were similarly examined. The results indicated that gma12p is solely responsible for the addition of terminal alpha1,2-linked Gal in compound 2A, while one or both of gma12p and gth1p are required for the alpha1,2-linked Gal in 4A. Both transferases are largely responsible for terminal Gal in isomer 5AB. Neither gma12 nor gth1 had any discernible effect on the structure of the large N-linked galactomannans as determined by 1H NMR spectroscopy. Thus, while gth1p and gma12p appear responsible for adding alpha1,2-linked Gal to terminal Man, neither adds galactose side chains to the N-linked poly alpha1,6-Man outerchain, nor the O-linked branch-forming alpha1,3-linked Gal. Furthermore, the presence of Hexalpha1,2(Galalpha1,3)Manalpha1,2- structures in the O-linked glycans implies the presence of a novel branch-forming alpha1,3-galactosyltransferase in S.pombe.  相似文献

A bacterial artificial chromosome (BAC) library containing a large genomlc DNA insert is an important tool for genome physical mapping, map-based cloning, and genome sequencing. To Isolate genes via a map-based cloning strategy and to perform physical mapping of the cotton genome, a high-quality BAC library containing large cotton DNA Inserts Is needed. We have developed a BAC library of the restoring line 0-613-2R for Isolating the fertility restorer (Rf1) gene and genomic research in cotton (Gossypium hirsutum L.). The BAC library contains 97 825 clones stored In 255 pieces of a 384-well mlcrotiter plate. Random samples of BACs digested with the Notl enzyme Indicated that the average Insert size Is approximately 130 kb, with a range of 80-275 kb, and 95.7% of the BAC clones in the library have an average insert size larger than 100 kb. Based on a cotton genome size of 2 250 Mb, library coverage is 5.7 × haploid genome equivalents. Four clones were selected randomly from the library to determine the stability of the BAC clones. There were no different fingerprints for 0 and 100 generations of each clone digested with Notl and Hlndiii enzymes. Thus, the atabiiity of a single BAC clone can be sustained at iesat for 100 generations. Eight simple sequence repeat (SSR) markers flanking the Rf; gene were chosen to screen the BAC library by pool using PCR method and 25 positive clones were identified with 3.1 positive clones per SSR marker.  相似文献

本文建立了利用高效液相色谱法定量分析L-山梨糖发酵过程中主产物2-酮基-L-古龙酸含量的方法。色谱柱为Nuclesoil-N(CH_3)_2 5μm;流动相为0.01M KH_2PO_4,用2%H_3PO_4调到pH=4.0,流速为1ml/min,检测器为示差折光仪(RI)。在该条件下色谱峰面积与进样的2-酮基-L-古龙酸含量之间呈线性相关,相关系数R=0.9965,最小检出量为0.1%。检测结果与采用碘量法分析结果一致。经多次使用,证明该方法简单可靠,能准确及时地判断L-山梨糖发酵的终点。  相似文献

1-0-octadecyl-2-0-acetyl-sn-glyceryl-3-phosphorylcholine (C18-AGEPC) stimulated a concentration (10-10-10-6M)-dependent release of granule-associated enzymes from human neutrophils. Cells which were not preincubated with cytochalasin B prior to exposure to C18-AGEPC did not degranulate. C18-AGEPC-elicited enzyme release was significantly reduced, but not abolished, in the absence of extracellular calcium. The lipoxygenase inhibitor, nordihydroguaiaretic acid and the lipoxygenase/cyclo-oxygenase inhibitor, 5,8,11,14-eicosatetraynoic acid, an acetylenic analog of arachidonic acid, caused a concentration-dependent suppression of enzyme discharge from neutrophils exposed to C18-AGEPC. However, the cyclo-oxygenase inhibitors, indomethacin, ibuprofen and flurbiprofen had no effect on C18-AGEPC-induced enzyme extrusion.  相似文献

Abstract

Reaction of the 5′-O-t-butyldimethylsilyl derivatives of thymidine and 2′-deoxyuridine with the Vilsmeier reagent (POCI3/DMF), and removal of the t–butyldimethylsilyl protecting group, afforded 3′-O-formylthymidine (5) and 3′-O-formyl-2′-deoxyuridine (6), respectively. In vitro evaluation, determined as the ability of the test compound to inhibit HIV induced cytopathogenicity in CEM cells, indicated that 5 was moderately active, whereas 6 was inactive.  相似文献

Aneuploid cells are frequently observed in human tumors, suggesting that aneuploidy may play an important role in the development of cancer. In this review, I discuss the processes that may give rise to aneuploid cells in normal tissue and in tumors. Aneuploid cells may arise directly from diploid cells through errors in chromosome segregation, as a consequence of incorrect microtubule-kinetochore attachments, or through failure of the spindle checkpoint. A second route to formation of aneuploid cells is through a tetraploid intermediate, where division of tetraploid cells can yield very high rates of chromosome missegregation as a consequence of multipolar spindle formation. Diploid cells may become tetraploid through a variety of mechanisms, including endoreduplication, cell fusion, and cytokinesis failure. Although aneuploid cells may arise from either diploid or tetraploid cells, the fate of the resulting aneuploid cells may be distinct. It is therefore important to understand the different pathways that can give rise to aneuploid cells, and how the varied origins of these cells affect their subsequent ability to survive or proliferate.  相似文献

Leptin is a hormone affecting the regulation of body composition, energy balance, and meat quality in mammals. The objective of this study was to evaluate the association of novel single nucleotide polymorphisms in coding region for leptin gene with carcass and meat quality traits of Chinese Simmental-cross steers. Two SNPs (E2-169 T > C and E3-299 T > A) were genotyped on 135 crossbred bulls. The 45 traits being measured included dressing percentage, dressed weight, marbling score, muscle color score, backfat thickness, fatty acid content, etc. Statistical analysis revealed that two SNPs in the exon of leptin gene were associated with the carcass and meat quality traits. The C-bearing genotypes (CC or TC) of E2-169 T > C (C57R) showed higher dressed weight, thickness of loin, MCS, FCS, intramuscular fat content, and polyunsaturated fatty acid content (P < 0.05). E3-299  > A(S100T) also showed a significant association with the carcass traits (dressing percentage, living QIB) and fatty acid content in Simmental-cross steers(P < 0.05). Our findings suggested that polymorphisms in leptin might be one of the important genetic factors that influence carcass yield and meat quality in beef cattle, and it may be a useful marker for meat quality traits in future marker-assisted selection programs in beef cattle breeding and production.  相似文献

The binding of 1-O-alkyl-2-acetyl-sn-glycero-3-phosphocholine, a platelet activating factor (PAF), to plasma components was studied. Gel filtration and lipoprotein fractionation revealed the presence in the plasma of PAF-binding fractions corresponding to plasma albumin as well as of low and high density lipoproteins. Incubation of PAF-containing lipoproteins with rabbit platelets resulted in a transfer of PAF to the platelets. PAF bound to plasma albumin is less exchangeable than PAF bound to lipoproteins. The PAF-transferring efficiency of high density lipoproteins (HDL) and of low density lipoproteins (LDL) correlates with the amounts of HDL- and LDL-receptors on the platelet surface. It may thus be assumed that PAF released by various cells interacts with lipoproteins which further transport the bound PAF to target cells carrying lipoprotein receptors.  相似文献

2′-0-Methyladenosine 5′-diphosphate has been chemically synthesized with an overall yield of 48% by adopting the direct phosphorylation of the nucleoside by phosphoryl chloride in step 1 and the morpholidate procedure in step 2 of the following scheme: Am1 - pAm1 (step 1) - ppAm (step 2). The method has been successfully used in the synthesis of 2′-O-methyluridine 5′-diphosphate and 2′-O-methylcytidine 5′-diphosphate and is applicable to the synthesis of nucleoside 5′-diphosphates in general.  相似文献
