Abstract: Pseudomonas aeruginosa, P. stutzeri and Azospirillum brasilense showed highest NO production rates and NO consumption rate constants when anaerobically grown cells were tested under anaerobic conditions. Aerobic assay conditions resulted in 20–75-fold lower NO production rates. NO consumption rate constants, however, decreased by less than a factor of four. NO consumption activity was observed even in aerobically grown P. aeruginosa , provided the assay was done under anaerobic conditions. Obviously, NO consumption was less O2-sensitive than NO production so that compensation between production and consumption occurred at lower NO mixing ratios under aerobic than under anaerobic conditions.  相似文献

Oxidative consumption of nitric oxide by heterotrophic bacteria in soil   总被引:1,自引:0,他引:1  
Abstract: Uptake rate constants for nitric oxide were measured in a neutral calcic cambisol (KBE) and an acidic luvisol (PBE). The NO uptake was higher under oxic than under anoxic incubation conditions by a factor of about three. Gassing the soils with air containing 10 ppmv NO resulted in the accumulation of nitrate which accounted for 57–94% of the NO consumed. Aerobic heterotrophic bacteria were isolated on glucose-yeast extract medium from soil dilutions corresponding to a most probable number of 108–109 bacteria per gram dry weight soil. One of the isolates (strain PS88, a Pseudomonas sp.) exhibited NO consumption activity that was much higher under oxic than anoxic incubation conditions. When sterile KBE amended with strain PS88 was gassed with air containing 10 ppmv NO, 88% of the consumed NO was recovered as nitrate and nitrite. A screening of various bacteria obtained from culture collections showed a widespread ability for consumption of low NO concentrations. Our results indicate that NO consumption in soil is not only possible by reductive denitrification, but also by oxidation due to aerobic heterotrophic bacteria such as strain PS88.  相似文献

Methanotrophs can oxidize methane to carbon dioxide through sequential reactions catalyzed by a series of enzymes including methane monooxygenase, methanol dehydrogenase, formaldehyde dehydrogenase, and formate dehydrogenase. When suspensions of methanotrophic bacteria of Methylosinus trichosporium IMV 3011 were incubated at 32°C with methane and oxygen, there was an extracellular accumulation of methanol from methane oxidation in response to carbon dioxide addition. Maximal accumulation of methanol was achieved with 40% carbon dioxide in the mixed reaction gases. A continuous experiment was performed in a continuous ultrafiltration reactor. The optimum gas mixture containing 20% (v v-1) methane, 20% oxygen, 20% nitrogen and 40% carbon dioxide was used to provide substrates and to maintain the transmembrane pressure. The product (methanol) was removed in the eluate buffer. The initial methanol concentration in the eluate buffer was 8.22 μmol L-1. The bioreactor was operated continuously for 198 h without obvious loss of productivity.  相似文献

Abstract Stone samples from corroding historical buildings in Germany released NO in significant amounts during oxidation of ammonium to nitrate. Nitrite intermediately accumulated to low concentrations. Nitrification and NO release were detectable only in stones with neutral pH. Addition of nitrapyrin inhibited both ammonium oxidation and NO production. NO was produced in stone samples containing Nitrosomonas or Nitrosovibrio , but was not produced in samples containing only Nitrobacter . Addition of nitrite stimulated NO production in autoclaved stone samples significantly only if dried to < 2% water content, or in acidic stone samples. In moist neutral stones, NO production was only slightly stimulated by nitrite giving less than about 8% of the NO production observed under nitrifying conditions. Most of the NO production by corroding building stones was apparently due to ammonium-oxidizing nitrifiers.  相似文献

Bacteriophages of methanotrophic bacteria have been found in 16 out of 88 studied samples (underground waters, pond water, soil, gas and oil installation waters, fermentor cultural fluids, bacterial paste, and rumen of cattle) taken in different geographic zones of the Soviet Union. Altogether, 23 phage strains were isolated: 10 strains that specifically lysed only Methylosinus sporium strains, 2 strains that each lysed 1 of 5 Methylosinus trichosporium strains studied, and 11 strains that lysed Flavobacterium gasotypicum and, at the same time, 1 M. sporium strain. By fine structure, the phages were divided into two types (with very short or long noncontractile tails); by host range and serological properties, they fell into three types. One-step growth characteristics of the phages differed only slightly; the latent period varied from 6 to 8 h, the rise period varied from 4 to 6 h, and the average burst size was 100. All phages had guanine- and cytosine-rich double-stranded deoxyribonucleic acid consisting of common nitrogen bases. The molecular mass of the deoxyribonucleic acid as determined by restriction endonuclease analysis was 29.4 X 10(6) for M. sporium phages and 44 X 10(6) for F. gasotypicum phages. By all of the above-mentioned properties, all phages within each of the groups were completely identical to one another, but differed from phages of other groups. Bacteriophages lysing M. sporium and M. trichosporium GB2 were identical to phages M1 and M4, respectively, which were isolated earlier in the German Democratic Republic on the same methanotrophic species.  相似文献

Seven bacteria representing seven genera of enteric bacteria, in addition to Escherichia coli, were shown to reduce nitrite to NO under anaerobic conditions when the cells were grown as nitrate respirers. NO production was inhibited by nitrate and azide and was self limiting, just as was found to be the case previously with E. coli and its nitrate reductase. Structural and functional characteristics of the regular glycoprotein layers in prokaryotes are analyzed with a special emphasis on aerobic methanotrophic bacteria. S-Layers are present at the surfaces of Methylococcus, Methylothermus, and Methylomicrobium cells. Different Methylomicrobium species either synthesize S-layers with planar (p2, p4) symmetry or form cup-shaped or conical structures with hexagonal (p6) symmetry. A unique, copper-binding polypeptide ‘CorA’/MopE (27/45 kDa), which is coexpressed with the diheme periplasmic cytochrome c peroxidase ‘CorB’/Mca (80 kDa) was found in Methylomicrobium album BG8, Methylomicrobium alcaliphilum 20Z, and Methylococcus capsulatus Bath. This tandem of the surface proteins is functionally analogous to a new siderophore: methanobactin. Importantly, no ‘CorA’/MopE homologue was found in methanotrophs not forming S-layers. The role of surface proteins in copper metabolism and initial methane oxidation is discussed.  相似文献

Peroxidases catalyze the oxidation of nitrite to nitrate in the presence of hydrogen peroxide. Two pathways may occur: one entailing the intermediate formation of NO(2) and the other implying the generation of peroxynitrite. The products of nitrite (NO(2) (-) ) oxidation by salivary peroxidase (SPO) and commercial bovine lactoperoxidase (LPO) are studied by utilizing an electrochemical assay that allows the direct, continuous monitoring of NO and/or NO(2) and by HPLC to assess nitrates at the end of the reaction. Dialyzed saliva and LPO, in the presence of H(2) O(2) , convert nitrite into nitrate and form some NO, with a molar ratio of 10(3) . In our experimental conditions, no NO(2) was detectable among the products of nitrite oxidation. SCN(-) inhibits NO formation and so does I(-) , although at higher concentrations. No effects are observed with Cl(-) or Br(-) . We conclude that SPO and LPO transform NO(2) (-) into nitrate-forming small amounts of NO in the presence of H(2) O(2) as an intermediate or a by-product, synthesized through the peroxynitrite pathway.  相似文献

Methanobactin (Mb), a 1217-Da copper chelator produced by the methanotroph Methylosinus trichosporium OB3b, is hypothesized to mediate copper acquisition from the environment, particularly from insoluble copper mineral sources. Although indirect evidence suggests that Mb provides copper for the regulation and activity of methane monooxygenase enzymes, experimental data for direct uptake of copper loaded Mb (Cu-Mb) are lacking. Uptake of intact Cu-Mb by M. trichosporium OB3b was demonstrated by isotopic and fluorescent labeling experiments. Confocal microscopy data indicate that Cu-Mb is localized in the cytoplasm. Both Cu-Mb and unchelated Cu are taken up by M. trichosporium OB3b, but by different mechanisms. Uptake of unchelated Cu is inhibited by spermine, suggesting a porin-dependent passive transport process. By contrast, uptake of Cu-Mb is inhibited by the uncoupling agents carbonyl cyanide m-chlorophenylhydrazone and methylamine, but not by spermine, consistent with an active transport process. Cu-Mb from M. trichosporium OB3b can also be internalized by other strains of methanotroph, but not by Escherichia coli, suggesting that Cu-Mb uptake is specific to methanotrophic bacteria. These findings are consistent with a key role for Cu-Mb in copper acquisition by methanotrophs and have important implications for further investigation of the copper uptake machinery.  相似文献

Two denitrifying bacteria, Pseudomonas stutzeri and Achromobacter cycloclastes, were incubated with Na15NO2 and NaN3 under conditions that allowed catalysis of nitrosyl transfer from nitrite to azide. This transfer, which is presumed to be mediated by the heme- and copper-containing nitrite reductase of P. stutzeri and A. cycloclastes, respectively, leads to formation of isotopically mixed 14,15N2O, whereas denitrification leads to 15N2O. The conditions that emphasized nitrosyl transfer also partially inhibited the nitric oxide reductase system and led to accumulation of 15NO. Absorption of NO from the gas phase by acidic CrSO4 in a sidewell largely abolished nitrosyl transfer to azide. With these two organisms, which are thought to be representative of denitrifiers generally, catalysis of nitrosyl transfer seemed to depend on NO.  相似文献

Many methane-oxidizing bacteria (MOB) have been shown to aerobically oxidize ammonia and hydroxylamine (NH(2)OH) to produce nitrite and nitrous oxide (N(2)O). Genome sequences of alphaproteobacterial, gammaproteobacterial, and verrucomicrobial methanotrophs revealed the presence of haoAB, cytL, cytS, nirS or nirK, and norCB genes that may be responsible for N(2)O production, and additional haoAB genes were sequenced from two strains of Methylomicrobium album. The haoAB genes of M. album ATCC 33003 were inducible by ammonia and NH(2)OH, similar to haoAB induction by ammonia in Methylococcus capsulatus Bath. Increased expression of genes encoding nitric oxide reductase (cNOR; norCB) was measured upon exposure of M. capsulatus Bath to NaNO(2) and NO-releasing sodium nitroprusside. Only incubations of M. capsulatus Bath with methane, ammonia, and nitrite produced N(2)O. The data suggest a possible pathway of nitrite reduction to NO by reversely operating NH(2)OH oxidoreductase and NO reduction to N(2)O by cNOR independently or in conjunction with ammonia-induced enzymes (i.e. HAO or cytochrome c'-β). Results of this study show that MOB likely have diverse mechanisms for nitrogen oxide metabolism and detoxification of NH(2)OH that involve conventional and unconventional enzymes.  相似文献

Nitrous oxide, a potent greenhouse gas and ozone-depleting molecule, continues to accumulate in the atmosphere as a product of anthropogenic activities and land-use change. Nitrogen oxides are intermediates of nitrification and denitrification and are released as terminal products under conditions such as high nitrogen load and low oxygen tension among other factors. The rapid completion and public availability of microbial genome sequences has revealed a high level of enzymatic redundancy in pathways terminating in nitrogen oxide metabolites, with few enzymes involved in returning nitrogen oxides to dinitrogen. The aerobic methanotrophic bacteria are particularly useful for discovering and analysing diverse mechanisms for nitrogen oxide production, as these microbes both nitrify (oxidize ammonia to nitrite) and denitrify (reduce nitrate/nitrite to nitrous oxide via nitric oxide), and yet do not rely on these pathways for growth. The fact that methanotrophs have a rich inventory for nitrogen oxide metabolism is, in part, a consequence of their evolutionary relatedness to ammonia-oxidizing bacteria. Furthermore, the ability of individual methanotrophic taxa to resist toxic intermediates of nitrogen metabolism affects the relative abundance of nitrogen oxides released into the environment, the composition of their community, and the balance between nitrogen and methane cycling.  相似文献

Abstract Production and consumption of NO was measured under anaerobic conditions in a slightly alkaline and an acidic soil as well as in pure cultures of denitrifying Pseudomonas aeruginosa, P. stutzeri, P. fluorescens, Paracoccus denitrificans, Azospirillum brasilense , and A. lipoferum . Growing bacterial cultures reduced nitrate and intermediately accumulated nitrite, NO, N2O, but not NO2. Addition of formaldehyde inhibited NO production and NO consumption. In the presence of acetylene NO was reduced to N2O. Net NO release rates in denitrifying bacterial suspensions and in soil samples decreased hyperbolically with increasing NO up to mixing ratios of about 5 ppmv NO. This behaviour could be modelled by assuming a constant rate of NO production simultaneously with a NO consumption activity that increased with NO until V max was reached. The data allowed calculation of the gross rates ( P ) of NO production, of the rate constants ( k ), V max and K m of NO consumption, and of the NO compensation mixing ratio ( m c). In soil, P was larger than V max resulting in net NO release even at high NO mixing ratios unless P was selectively inhibited by chlorate + chlorite or by aerobic incubation conditions. In bacteria, V max was somewhat larger than P resulting in net NO uptake at high NO mixing ratios. Both P and V max were dependent on the supply of electron donor (e.g. glucose). Both in soil (aerobic or anaerobic) and in pure culture, the K m values of NO consumption were in a similar low range of about 0.5–6.0 nM. Anaerobic soil and denitrifying bacteria exhibited m c values of 1.6–2.1 ppmv NO and 0.2–4.0 ppmv NO, respectively.  相似文献

Excised leaves of kidney bean (Phaseolus vulgaris) were used to investigate the mechanism of NO generation under UV-B stress. We showed that two signaling molecules, NO and H2O2, were produced in the irradiated leaves. NO release was blocked by LNNA, an inhibitor of NOS. Application of CAT (EC 1.11.1.6) not only effectively eliminated H2O2 in the leaves, but also inhibited the activity of NOS and the emission of NO. In contrast, treatment with exogenous H2O2 increased both of those events. Therefore, we suggest that, under UV-B stress, NO production is mediated by H2O2 through greater NOS activity.  相似文献

Abstract A mass spectrometer with membrane inlet was used to measure methane and oxygen utilization rates at various methane concentrations in Methylosinus trichosporium and a locally isolated strain of a methane-oxidizing coccus (OU-4-1). The apparent K m for methane was found to be 2 μM for M. trichosporium and 0.8 μM for strain OU-4-1. These K m-values are 10–30 times lower than most previously reported values. The ratio of oxygen to methane utilization rates was 1.7 for M. trichosporium and 1.5 for strain OU-4-1 corresponding to a growth yield of 0.38 and 0.63 g dry weight/g methane, respectively.  相似文献
