The 3561 m Vostok ice core sample originating from the subglacial Lake Vostok accretion (frozen lake water) ice with sediment inclusions was thoroughly studied by various means to confirm the presence of the thermophile bacterium Hydrogenophilus thermoluteolus reported earlier in the 3607 m accretion ice sample. PCR and molecular-phylogenetic analyses performed in two independent laboratories were made using different 16S rRNA gene (rrs) targeted primers. As a result, rrs-targeted PCR permitted to recover several very closely related clones with a small genetic distance to Hydrogenophilus thermoluteolus (< 1%). In addition, RubisCO (cbbL or rbcL) and NiFe-Hydrogenase (hoxV or hupL) targeted PCR have also allowed to recover sequences highly related to Hydrogenophilus thermoluteolus. All these results point to the presence of thermophilic chemoautotrophic microorganisms in Lake Vostok accretion ice. They presumably originate from deep faults in the bedrock cavity containing the lake in which episodes of seismotectonic activity would release debris along with microbial cells.  相似文献

Isolation of bacteria and 16S rDNAs from Lake Vostok accretion ice   总被引:6,自引:0,他引:6  
Lake Vostok, the largest subglacial lake in Antarctica, is separated from the surface by ≈ 4 km of glacial ice. It has been isolated from direct surface input for at least 420 000 years, and the possibility of a novel environment and ecosystem therefore exists. Lake Vostok water has not been sampled, but an ice core has been recovered that extends into the ice accreted below glacial ice by freezing of Lake Vostok water. Here, we report the recovery of bacterial isolates belonging to the Brachybacteria , Methylobacterium , Paenibacillus and Sphingomonas lineages from a sample of melt water from this accretion ice that originated 3593 m below the surface. We have also amplified small-subunit ribosomal RNA-encoding DNA molecules (16S rDNAs) directly from this melt water that originated from α- and β-proteobacteria, low- and high-G+C Gram-positive bacteria and a member of the Cytophaga / Flavobacterium / Bacteroides lineage.  相似文献

A bacterial ice-binding protein from the Vostok ice core   总被引:1,自引:0,他引:1  
Bacterial and yeast isolates recovered from a deep Antarctic ice core were screened for proteins with ice-binding activity, an indicator of adaptation to icy environments. A bacterial strain recovered from glacial ice at a depth of 3,519 m, just above the accreted ice from Subglacial Lake Vostok, was found to produce a 54 kDa ice-binding protein (GenBank EU694412) that is similar to ice-binding proteins previously found in sea ice diatoms, a snow mold, and a sea ice bacterium. The protein has the ability to inhibit the recrystallization of ice, a phenotype that has clear advantages for survival in ice.  相似文献

A general list of the flora of Vostok Bay (Sea of Japan) compiled on the basis of original and literature data includes 161 species, among them 25 green algae, 42 brown algae, 90 red algae, and 4 species of seagrass. In the period from 2000 to 2004, 114 species of macrophytes (19 green algae, 29 brown algae, 63 red algae, and 3 species of seagrass) were found in Vostok Bay; 19 species of algae were first records for this bay. On the whole, the flora of Vostok Bay is dominated by warm-water species, among which Asian low-boreal species occupy the leading position. The flora of Vostok Bay is changing under the influence of global climatic and local anthropogenic factors. In recent years, there is a tendency for the proportion of warm-water species to increase. The local anthropogenic influence on the flora of Vostok Bay has caused changes in the species composition of plant communities.  相似文献

The year 2012 began with reports on the completion of drilling over the largest subglacial lake of Antarctica, Lake Vostok. It has been sealed from free exchange with the atmosphere for ~420,000 years, making it a potentially rich and largely unexplored storehouse of genetic information for solving evolutionary queries. Indirect evidence suggests that many impediments exist in the subglacial environment, such as high oxygen tension, low inorganic and organic nutrient concentrations, etc., whereas analysis of glacial and accretion ice from Vostok also implies that the elemental requirements for microbial growth could be satisfied in the lake by many possible electron donors (like H2, Fe2+ and NO 2 ? ). Recent reports on the recovery of the longest ever ice core of ~3,769 m open up a new window for answering many questions about microbiological life in extremes, evolution, and adaptations. This review attempts to present an updated understanding on the potential significance of subglacial environments in unraveling the mystery of evolution of life.  相似文献

All of the accreted ice samples, taken from depths between 3541 and 3611 m, were found to contain pro- and eukaryotic microorganisms, whose number and diversity varied in different ice horizons and correlated, to a certain degree, with the occurrence of organic and inorganic impurities in a given horizon. Some biological objects found in the accreted lake ice, including bacteria, microalgae, and the pollen of higher plants, were morphologically similar to those found earlier in the glacier ice bulk. The others were not. Quantitative samples of the macrobenthos were collected in May, July, and October–November from 2000 to 2002 in Vostok Bay, Sea of Japan. The main characteristics of the subtidal macrobenthos communities are described. The present study shows that the rocky bottom communities of Vostok Bay have changed little over the past 30 years. Changes have occurred in the structure of soft bottom communities and phytocenoses. Analysis of the macrophytes species composition and the value of the floristic coefficient suggest a currently moderate anthropogenic load on Vostok Bay.Original Russian Text Copyright © 2004 by Biologiya Morya, Galysheva.  相似文献

The qualitative and quantitative composition of free-living marine nematodes have been studied in Vostok Bay (Peter the Great Bay, Sea of Japan). It is found that the population density of nematodes in the bottom sediments of Vostok Bay shows an uneven distribution. The mean population density equaled 56800 ± 23400 specimens/m2. A correlation has been revealed between the population density of nematodes and the substrate type. Altogether, 85 species of nematodes have been found; they were dominated by Sabatieria palmaris, Rhabdodemania orientalis, Araeolaimus parvibulbosus, Oncholaimium paraolium, Doryolaimopsis peculiaris, and Metachromadora itoi. Six taxocenoses of nematodes were distinguished, taking into account the species dominating in the population density and using cluster analysis of the obtained data. The dominating trophic assemblage of nematodes was “scrapers.” In general, the species composition of nematodes in Vostok Bay is characterized by the relatively great similarity with that in other areas of Peter the Great Bay (Sea of Japan).  相似文献

We summarize the results of a study of the taxonomic composition and seasonal variations of ichthyoplankton in Vostok Bay. Samples were collected during 2003–2008 at a stationary light station located on the mooring module of the experimental biological station Vostok (Institute of Marine Biology). Within the observation period, larval and juvenile fish of 67 species belonging to 11 orders and 30 families were found in 412 samples. The seasonal dynamics of species composition of ichthyoplankton were examined over the period from March to November. The number of species in the samples changed from 40–36 in April–May, respectively, to 6 species in October. The contribution of resident species and southern migrants to the ichthyoplankton samples was estimated. The light station sampling has proven to be most effective in studying the species diversity of the ichthyofauna in Vostok Bay. Using light station sampling, 18 fish species were found that were not recorded using other sampling techniques.  相似文献

Fauna and systematics of sipunculans inhabiting Vostok Bay, Sea of Japan, are discussed. A revision of the local species of sipunculans is performed for the first time. On the basis of study of outer and inner morphology, some species are reduced to synonyms. Extended diagnoses are proposed for all registered species. Genotypic variability at six allozyme loci and six morphological shell characters was examined in 450 individuals (four samples) of mollusk Nucella heysana from the Vrangel' Bight (Nakhodka Bay) and the Vostok Bay (Peter the Great Gulf, Sea of Japan). An analysis of variation in allele frequencies showed that each of the two localities (Vostok and Nakhodka bays) in the analyzed region is inhabited by a single, albeit genetically heterogeneous, population. A canonical analysis and an analysis of variance of individual heterozygosity (Ho) and morphological variation indicate an association between Ho and morphological variation depending on habitat (interaction) in settlements in the Vrangel' Bight and, to a lesser extent, in the Vostok Bay. These results indicate that, in extreme environmental conditions, allozyme phenotypes may be selected either themselves or as markers of genes linked with them.  相似文献

The occurrence of the solitary ascidian Ciona savignyi Herdman, 1882 in Vostok Bay (Peter the Great Bay, Sea of Japan) was first documented in 2004. Adult specimens occurred in fouling communities of floating docks in Gaidamak Bay and on different anthropogenic substrates. The introduction of this ascidian into Vostok Bay is attributable to fishing ships which regularly frequent ports of Japan and to favorable environmental conditions (temperature and salinity of seawater).  相似文献

The taxonomic composition of marine mycelial fungi was determined in the interstitial habitats of the Vostok Bay littoral (Peter the Great Bay, the Sea of Japan). A total of 39 species of ascomycetes and anamorphic fungi were detected and identified. The predominant species of the intertidal zone were Corollospora maritima, C. lacera, Carbosphaerella leptosphaerioides, Arenariomyces trifurcatus (Ascomycota), Alternaria alternata, Scolecobasidium arenarium, and Zalerion maritimum (anamorphic fungi). Comparative biotesting was performed using embryos and larvae of the sea urchin Scaphechinus mirabilis and the microalga Dunaliella salina. These two species were taken as test organisms for estimating water quality in areas of various anthropogenic loads. Seawater from Nakhodka and Vostok bays, as well as from the southwestern part of Peter the Great Bay (Sea of Japan) near the Tumen River mouth, was tested. Water from Vostok and Nakhodka bays had a harmful effect on embryonic and larval development of the sea urchin. The algal culture kept in the water of Vostok Bay was depressed throughout the experiment, while development of the alga in the water from Nakhodka Bay hardly differed from the control but was unstable. Water from the southwestern part of Peter the Great Bay did not have any significant harmful effect on both test organisms. Embryos and larvae of the sea urchin S. mirabilis were several orders more sensitive to salinity changes and content of toxic agents; they responded more readily to changes in water quality than D. salina cells. Correspondingly, embryos and larvae of the sea urchin S. mirabilis were found to be a preferable test organism for assessment of pollution in various marine environments.  相似文献

Subglacial environments, particularly those that lie beneath polar ice sheets, are beginning to be recognized as an important part of Earth's biosphere. However, except for indirect indications of microbial assemblages in subglacial Lake Vostok, Antarctica, no sub-ice sheet environments have been shown to support microbial ecosystems. Here we report 16S rRNA gene and isolate diversity in sediments collected from beneath the Kamb Ice Stream, West Antarctic Ice Sheet and stored for 15 months at 4°C. This is the first report of microbes in samples from the sediment environment beneath the Antarctic Ice Sheet. The cells were abundant (∼107 cells g−1) but displayed low diversity (only five phylotypes), likely as a result of enrichment during storage. Isolates were cold tolerant and the 16S rRNA gene diversity was a simplified version of that found in subglacial alpine and Arctic sediments and water. Although in situ cell abundance and the extent of wet sediments beneath the Antarctic ice sheet can only be roughly extrapolated on the basis of this sample, it is clear that the subglacial ecosystem contains a significant and previously unrecognized pool of microbial cells and associated organic carbon that could potentially have significant implications for global geochemical processes.  相似文献

For the first time, year round and long-term seasonal research on the composition, phenology, and population dynamics of pelagic larvae of bottom invertebrates were conducted in the Vostok Bay of the Sea of Japan. Larvae of 98 taxa of various ranks were found. The larvae of bivalves and polychaetes were the most diverse. The larvae of bottom invertebrates occurred in plankton year-round. Their density was at its minimum in January (less than 100 ind./m3), and its maximum from June to November (more than 1000 ind./m3). The larvae of polychaetes dominated in the autumn and winter seasons, whereas bivalve and gastropod larvae dominated in the spring and summer seasons. It was shown that the proportion of pollution tolerant meroplankton larvae of the Polydora and Pseudopolydora genera was high in the Vostok Bay, and could indirectly indicate increasing anthropogenic pressure in the area.  相似文献
