1. Liver cytosol samples from juvenile coho salmon exposed over a 14-week period to 70 micrograms/l (1/4 LC50) or 140 micrograms/l (1/2 LC50), were separated into high-molecular-weight and low-molecular-weight fractions by column chromatography on Sephadex G-75, and compared to similar samples obtained from control fish. 2. The levels of copper in the low-molecular-weight function of fish exposed to 70 micrograms/l were not significantly increased over control values for the first 6 weeks but then increased rapidly, while those of fish exposed to 140 micrograms/l increased substantially in the first 2-4 weeks of exposure, then levelled off. 3. In the exposed fish the levels of copper in the high-molecular-weight fraction were not significantly elevated above control levels over the first 8-10 weeks but then increased significantly. 4. The low-molecular-weight fractions of exposed fish were shown to contain increasing levels of metallothionein. The metallothionein was isolated on DEAE-cellulose and characterized by amino acid analyses.  相似文献

Following determination of a concentration of selenate-Se lethal during 24-hr exposures at 22 degrees C, routine oxygen consumption was measured in fathead minnows (Pimephales promelas) exposed to various sublethal concentrations of Se for 24 hr. A median lethal concentration for 24-hr exposures (24-hr LC50) of fathead minnows equalled 82 mg Se/l with 95% fiducial limits of 76-89 mg Se/l. Mean weight-specific oxygen consumption rates of minnows exposed to 0, 15, 40 and 60 mg Se/l for 24-hr ranged from 0.43 to 0.49 mg O2/g-1/hr-1 and were not significantly different. These results indicate that acute exposure to Se does not affect oxygen consumption; however, observations during respirometry trials suggest that minnows at the highest Se concentration may have physiologically compensated to maintain oxygen consumption rates.  相似文献

This study was to determine the median lethal concentration (LC50) of copper to Javenese carp, Puntius gonionotus (Bleeker), and the immune response after the fish were exposed to sublethal levels of copper and challenged with formalin killed Aeromonas hydrophila. The LC50 of copper on P. gonionotus at 24, 48, 72, 96 and 120 h were estimated as 2.17, 0.91, 0.57, 0.53 and 0.42 mg l(-1), respectively. To determine the effect of copper on the immune system, fish were exposed for 66 days to 0.05, 0.10 and 0.15 mg Cu l(-1). After 56 days of initial exposure to copper, fish were challenged with 0.1 ml of 4.5 x 10(5) cfu ml(-1) formalin killed A. hydrophila and maintained in the same concentration of copper. After the challenge, the immune response was monitored for 2 weeks using haematological and serological assays. During the initial phase of exposure to copper, significant changes were noted in the white blood cell, lysozyme, potential killing activity, total plasma protein, total immunoglobulin and haematocrit levels between the control and treated fish. One week after challenge with A. hydrophila, there was a significant increase in the values of white blood cells, total protein and total immunoglobulin compared to the values before the challenge. However, these values were not significantly different (P>0.05) between the control and the treated fish. In contrast, NBT and lysozyme assays exhibited a significant difference (P<0.05) in fish exposed to 0.10 mg Cu l(-1) (0.525 +/- 0.17; 24.42 +/- 3.35 x 10(2) micromg ml(-1)) and 0.15 mg Cu 1(-1) (0.536 +/- 0.19; 21.78 +/- 1.29 x 10(2) micromg ml(-1)) compared to the control (0.746 +/- 0.31; 30.73 +/- 5.42 x 10(2) micromg ml(-1)) after the bacterial challenge (day 61). There was however no significant difference (P>0.05) in NBT and lysozyme levels in fish exposed to lower level of copper (0.05 mg Cu l(-1)), suggesting the absence of immunosuppressive effects at lower level of exposure.  相似文献

The survival of Atlantic salmon smolts on exposure to constant concentrations of cyanide and ammonia, singly and together, has been measured under laboratory conditions at a concentration of 5 mgl-1 of carbon dioxide. The 24-h LC50 values of cyanide and of un-ionised ammonia, in fresh water, were 0·073 mg HCN l-1 and 0·20 mg NH3l-1 respectively at a concentration of dissolved oxygen of 10 mg l-1, and 0·024 mg HCN l-1 and 0·08 mgNH3l-1 respectively at a concentration of dissolved oxygen of 3·5 mg l-1. In 30% sea water the corresponding values were similar for cyanide but markedly higher for ammonioa. In 80% sea water the values were intermediate between those of fresh water and 30% sea water. Prior acclimation of the fish to the respective toxicant increased the resistance of the fish only slightly to cyanide, but with ammonia the 24-h LC50 was increased between 1·4 and 2-fold after acclimation for 1–3 days to between 0·2 and 0·5 of the 24-h LC50 value. Mixtures of cyanide and ammonia were between 0·6 and 1·25 times as toxic as expected, assuming simple additivity of toxicity.  相似文献

A series of experiments were conducted to determine the physiological impact of acute sublethal molybdenum exposure to juvenile kokanee salmon (Oncorhynchus nerka Kennerlyi). Molybdenum was found to be relatively non-toxic to kokanee as the 96 h LC(50) was greater than 2,000 mg Mo l(-1). Exposure to either 25 or 250 mg Mo x l(-1) for 7 days was found to stimulate a significant 1.6- to 1.7-fold increase in ventilation which was later characterized to be dose-dependent between 5 and 250 mg Mo l(-1). Acute sublethal molybdenum exposure was found to have little or no impact on kokanee oxygen consumption at rest or immediately following a bout of forced activity or on physiological indicators of stress such as plasma lactate, sodium and cortisol. Despite these findings, prior exposure to 25 or 250 mg Mo l(-1) resulted in post-exercise loss of equilibrium and exercise-induced delayed mortality that were not observed in controls. Molybdenum accumulation in gill and liver of kokanee was also characterized. The findings of this study suggest that despite the non-toxic nature of molybdenum, acute sublethal exposure to this metal has physiological consequences to those fish exposed even for only a brief period. Further studies are needed to more fully elucidate the metabolism and mode of action of this metal in fish.  相似文献

Although the chronotoxicity of xenobiotics is relatively well known in mammals, the existence of daily rhythms of drug toxicity and effectiveness in fish has been neglected to date. The aim of this research was to investigate the influence of the time (middle of the light phase [ML] versus middle of the dark phase [MD]) of exposure to two anesthetic substances (MS-222 or clove oil) commonly used with fish on the median lethal concentration (LC(50)) and swimming activity of zebrafish (Danio rerio). To this end, adult zebrafish were kept under a 12 h:12 h light-dark (LD) cycle and exposed to different concentrations of the anesthetics for 15 min at ML or MD. LC(50) calculations were performed using the Spearman-Karber program, whereas swimming activity was video-recorded and analyzed with specialized software. Zebrafish exhibited a mostly diurnal activity pattern (77.9% of activity occurring during daytime). The acute toxicity and mortality caused by MS-222 and eugenol varied with the time of exposure. For MS-222, the LC(50) was 170.6 ± 7.4 mg/L in fish exposed at ML and 215.6 ± 3.9 mg/L at MD, whereas for eugenol the LC(50) was 70.3 ± 3.1 mg/L at ML and 104.9 ± 5.4 mg/L at MD. Exposure to sublethal concentrations of MS-222 and eugenol altered the swimming patterns of zebrafish in a different manner depending on the time of exposure. Thus, the time required for decreasing swimming activity during exposure to anesthetics was shorter at ML than at MD, whereas the recovery period was longer during the day. In conclusion, these results revealed that the toxicity and effectiveness of both anesthetic substances is highest during daytime, the active phase of fish, thus suggesting a link between the daily rhythms of behavior and toxicity.  相似文献

Metabolic rates associated with sustained, prolonged and critical swimming speeds were examined in 10 g trout exposed to 5% 96 hr LC50 (0.75 microgram X l-1) and 10% 96 hr LC50 (1.50 micrograms X l-1) at 12 degrees C. Permethrin did not influence the metabolic cost for swimming at sustained and prolonged speeds. Basal metabolic rate increased on initial exposure to permethrin reaching maximum values after 7 days and declined to the control level after 13 days in 5% and after 32 days in 10% 96 hr LC50. Critical swimming speeds were adversely affected in a manner reflective of the effects of permethrin on basal metabolic rate. Elevation in basal metabolic rate in fish exposed to permethrin was a result of increased energy requirements due to physiological stress, detoxication and tissue repair.  相似文献

Juvenile rainbow trout were exposed to 0, 0.1, 1, 3, and 5 micro g/l silver (Ag, as AgNO3) for 23 days. Specific growth rate, cumulative food consumption, food-conversion efficiency, and critical swimming speed (U(crit)) were significantly reduced during 5 micro g/l Ag exposure, demonstrating a physiological cost of silver acclimation. Only the 5 microg/l Ag treatment had significant cumulative mortality (5.2%). Fish were most susceptible to silver on days 5 and 15. Exposure to 5 microg/l Ag significantly lowered plasma Na+ and Cl- on days 5 and 10, but plasma ion concentration recovered thereafter. Unidirectional Na+ uptake and gill Na/K-ATPase activity were significantly inhibited by 3 and 5 microg/l Ag exposure. Na+ uptake was inhibited by 3 micro g/l Ag at day 5 alone, whereas the effects at the highest Ag exposure persisted until day 15. Gill Na/K-ATPase was inhibited on day 5 in both the 3 and 5 microg/l Ag treatments but increased to approx. 1.5 times of control levels by day 23. Only the 3 and 5 microg/l Ag treatments produced toxicological acclimation (at least twofold elevations in 168-h LC50 values in fish subsampled on days 15 and 23). We conclude that physiological acclimation results from compensatory changes in Na+ transport at the gills, and that these changes may eventually lead to toxicological acclimation.  相似文献

The 96 hr LC50 for Callinectes sapidus exposed to fenitrothion at 22 degrees C and a salinity of 34 ppt (parts per thousand) was estimated to be 8.6 micrograms/l with a 95% confidence interval of 7.4-9.9 micrograms/l. Acute toxicity was shown to increase with increasing temperature as well as increasing salinity. Exposure of Callinectes to a simulated tidal cycle of 17 ppt salinity change at 6 hr intervals increased the acute toxicity of fenitrothion to Callinectes. The autotomization response in Callinectes was shown to be affected at subacute exposure levels as low as 0.1 microgram/l.  相似文献

1. Coho salmon, exposed to sublethal levels of aqueous copper (1/4 and 1/2 LC50), lost appetite and ceased growing or showed decreased rates of growth. 2. Recovery of appetite and growth rate was faster in fish exposed to 1/4 of the LC50 than in those exposed to 1/2 of the LC50. 3. Copper levels were elevated in liver gill and kidney of exposed fish with the liver tissue accumulating a much larger amount of the metal than any other tissue. 4. The concentration of liver copper became constant at about the time that growth rate recovered. 5. The exposed fish exhibited much higher resistance to elevated aqueous copper levels than did the controls. 6. The results suggest that coho salmon may become acclimated to higher levels of copper and that acclimated fish are more tolerant to copper than control animals.  相似文献

At concentrations of dissolved oxygen close to the air-saturation value, the 24-h LC50 of un-ionised ammonia is 0.15 mg NH31−1 in fresh water (hardness 264 mg 1−1 as CaCO3) and 0.3 mg NH31−1 in 30% sea water; at concentrations of dissolved oxygen of 3.5 mg 1−1 in fresh water and 3.1 mg 1−1 in 30% sea water, the 24-h LC50 is 0.09 mg NH3 1−1 and 0.12 mg NH3 1−1 respectively; for fish acclimated for 1 day to a concentration of ammonia close to the 24-h median for un-acclimated fish, the median is increased between 38 and 79%, depending on test conditions.  相似文献

The disinfectant effects on Legionella and nontuberculous mycobacteria of hot water, ultraviolet light, silver ions and chlorine, were evaluated. The bacterial strains Legionella pneumophila ATCC33152 and Mycobacterium avium ATCC25291 and strains of L. pneumophila and M. avium which had been isolated from a 24 h bath, were examined for their resistance to treatments. All strains were killed within 3 min on exposure to hot water at 70 degrees C and exposure to ultraviolet light at 90 mW.s/cm2. The strains of L. pneumophila tested were killed within 6 h on exposure to a solution of silver ions at 50 micrograms/l. The number of viable cells of strains of M. avium fell from 10(5) CFU/ml to 10(3) CFU/ml after exposure to an aqueous solution of silver ions at 100 micrograms/l for 24 h. Chlorine effectively killed strains of Legionella which were exposed to an aqueous solution of chlorine at 2 mg/l within 3 min, but strains of Mycobacterium survived exposure to chlorine at 4 mg/l for more than 60 min.  相似文献

本研究评估了亚致死浓度的林丹(1/2的60dLC50,0.935mg/L)和阿特拉津(1/2的60dLC50,3.585mg/L)对海洋性贝类影响的生理学反应。林丹和阿特拉津是已被公认的环境污染物质。56d的实验表明,这两种农药在贝类体内具有积累作用,而且在体内各器官的积累程度与其作用的靶器官相一致。林丹在体内的积累浓度(每克干重372μg)比阿特拉津(每克干重137μg)更高。该贝类在林丹中暴露56d,其氧的消耗比对照组降低10%,而在阿特拉津中则比对照组提高了29%。经林丹和阿特拉津暴露后,增加了氨排泄物。但是,两种农药均使贝类的取食率和吸收率下降。林丹会降低贝类的取食率、氧消耗、氨排泄、食物吸收率以及生长范围。在形式上,阿特拉津对贝类的影响与林丹的影响有不同之处,它会降低贝类的取食率和食物吸收率,但与林丹不同的是,它会提高贝类的氧消耗和氨排泄。总之,阿特拉津会明显地减小贝类繁殖净能。因此,阿特拉津暴露与林丹暴露的毒性症状是一致的。结合组织化学分析,贝类的生理学反应可作为一种理想的环境监测工具。研究结果表明,在二分之一的半致死浓度下暴露两个月,不仅能有效地证明林丹和阿特拉津在贝类组织内具有积累作用,而且对这些积累能产生生理反应。  相似文献

Seabream and turbot juveniles (40-520 g) were exposed to constant exogenous NH5-N concentrations (1.27-4.27 mmol/l; pH, 8.15). In 96 hr acclimated fish, plasma TA-N (total ammonia nitrogen) contents were positively correlated to ambient ammonia concentrations. The LD50 were 2.2-2.5 mmol/l TA-N in both species. There were no marked osmoregulatory disturbances and plasma urea-N, thyroid hormones levels and gill (Na-K)-ATPase activities were only affected at the highest concentrations. Liver GOT, GPT and GIDH activity dose-response were low and species dependent. In cannulated and non-cannulated turbot exposed to half 96 hr LC50 (lethal ambient concentration for 50% of the population), there was a rapid, pronounced and prolonged increase in plasma TA-N, followed by an immediate decline when exogenous ammonia supply was stopped. Maximum loading and unloading were observed within 1-3 hr. Plasma cortisol levels indicated a stressful situation in exposed fish (150 ng/ml) and a quick recovery capacity. In dose and time response experiments, the most relevant physiological indicator of ammonia stress was blood TA-N content. Other parameters tested led either to transient or low amplitude responses except when fish approached death.  相似文献

OBJECTIVE: To assess the relation between tap water lead and maternal blood lead concentrations and assess the exposure of infants to lead in tap water in a water supply area subjected to maximal water treatment to reduce plumbosolvency. DESIGN: Postal questionnaire survey and collection of kettle water from a representative sample of mothers; blood and further water samples were collected in a random sample of households and households with raised water lead concentrations. SETTING: Loch Katrine water supply area, Glasgow. SUBJECTS: 1812 mothers with a live infant born between October 1991 and September 1992. Blood lead concentrations were measured in 342 mothers. MAIN OUTCOME MEASURES: Mean geometric blood lead concentrations and the prevalence of raised tap water lead concentration. RESULTS: 17% of households had water lead concentration of 10 micrograms/l (48.3 nmol/l) or more in 1993 compared with 49% of households in 1981. Tap water lead remained the main correlate or raised maternal blood lead concentrations and accounted for 62% and 76% of cases of maternal blood lead concentrations above 5 and 10 micrograms/dl (0.24 and 0.48 mumol/l) respectively. The geometric mean maternal blood lead concentration was 3.65 micrograms/dl (0.18 mumol/l) in a random sample of mothers and 3.16 micrograms/dl (0.15 mumol/l) in mothers whose tap water lead concentrations were consistently below 2 micrograms/l (9.7 nmol/l). No mother in the study had a blood lead concentration above 25 micrograms/dl (1.21 mumol/l). An estimated 13% of infants were exposed via bottle feeds to tap water lead concentrations exceeding the World Health Organisation''s guideline of 10 micrograms/l (48.3 nmol/l). CONCLUSIONS: Tap water lead and maternal blood led concentrations in the Loch Katrine water supply area have fallen substantially since the early 1980s. Maternal blood lead concentrations are well within limits currently considered safe for human health. Tap water lead is still a public health problem in relation to the lead exposure of bottle fed infants.  相似文献

Due to prolonged use of silver in many applications, it enters into the freshwater and affects the aquatic organisms. Fingerlings of Cirrhinus mrigala were exposed to acute and sublethal concentrations of silver nitrate and the alterations of gill Na(+)/K(+)-ATPase, plasma electrolytes and biochemical parameters were assessed. The median lethal concentration of silver nitrate to the fish C. mrigala for 96 h was found to be 0.107 mg/l (with 95% confidence limits). 1/10th of LC 50 96 h value (0.0107 mg/l) was selected for sublethal study. During acute treatment branchial Na(+)/K(+)-ATPase activity was inhibited approximately 44.34% after 96 h of exposure. In sublethal treatment, silver nitrate could not produce a significant change in the activity of the enzyme at the end of 7th day. However, after 14th day, significant (p < 0.05) decrease was noted showing 22.52%-49.11% in rest of the study period. Silver intoxication resulted hyponatremia, hypokalemia, hypochloremia, and hypoproteinemia in both the treatments. Despite the decrease in these parameters, plasma glucose level was found to be increased in both the treatments to endure the silver toxicity. We suggest that the alterations in branchial Na(+)/K(+)-ATPase activity, plasma electrolytes, and biochemical parameters of fish may be useful in environmental biomonitoring and to assess the health of fish in freshwater habitat contaminated with silver.  相似文献

In vivo toxicity of monocrotophos on key metabolites and enzymes of the protein metabolism was investigated in important tissues of the freshwater fish Clarias batrachus. Fish were exposed to 1/10 and 1/20 of LC50 concentration for 28 days. After 28 days of exposure, some fish were transferred to monocrotophos-free water and kept in the same for 21 days (recovery period) in order to study the recovery response. Total protein, amino acid, and ammonia contents were decreased in gill, kidney, liver, and muscle tissues, and recovery was slight at the end of 21 days of transfer of fish into freshwater. Urea and glutamine levels were elevated, except in kidneys, and recovered at the end of the recovery period. The activities of protease, transaminase, and phosphatase enzymes were elevated in all tissues during 28 days of exposure and at both concentrations. Recovery of the activity of enzymes was more significant at the lower concentration as compared to the higher concentration.  相似文献

Bluegill were exposed to copper (2–0 mg/1−1 unfiltered) for 24, 48, 72 or 96 h at 24° C. This concentration approximated the 96 h LC20. Water content of liver increased 4–8% after 24 h exposure and muscle 4–6% by 48 h. These changes persisted throughout the 96 h experiments. Copper exposed fish exhibited decreases in muscle ATP, ADP, total adenylates, and energy charge by 48 h. There was a trend toward recovery of ATP with further exposure. Liver ATP of exposed fish was lower than controls at all intervals with the greatest difference evident at 48 h. No significant changes in brain adenylates were observed. Muscle and liver lactic acid was unchanged at 48 h exposure, therefore tissue hypoxia was not the cause of the adenylate changes. It was concluded that copper causes decreases in muscle and liver ATP several days before probable death of copper exposed fish and the changes seen are the result of dilution by increased tissue water, and the copper acting on certain cellular enzymes involved in detoxification and energy metabolism.  相似文献

The aim of the present study was to evaluate the induced genotoxicity (DNA damage) due to organophosphate pesticide profenofos (PFF) in gill cells of freshwater fish Channa punctatus using single cell gel electrophoresis (SCGE)/Comet assay. The 96h LC(50) value of PFF (50% EC) was estimated for the fish species in a semistatic system and then three sub-lethal of LC(50) concentrations viz the sub-lethal 1, sub-lethal 2 and sub-lethal 3 concentrations were determined as 0.58ppb, 1.16ppb and 1.74ppb, respectively. The fish specimens were exposed to these concentrations of the pesticide and the gill tissue samplings were done on 24h, 48h, 72h and 96h post exposure for assessment of DNA damage in terms of percentage of DNA in comet tails. In general, a concentration dependent response was observed in the gill cells with induction of maximum DNA damage at the highest concentration of PFF. The results of the present investigation indicated that PFF could potentially induce genotoxic effect in fish, even in sub-lethal concentrations and SCGE as a sensitive and reliable tool for in vivo assessment of DNA damage caused by the genotoxic agents.  相似文献

Recently, residual pharmaceuticals are generally recognized as relevant sources of aquatic environmental pollutants. However, the toxicological effects of these contaminants have not been adequately researched. In this study, the chronic toxic effect of carbamazepine (CBZ), an anticonvulsant drug commonly present in surface and ground water, on hepatic antioxidant status and hematological parameters of rainbow trout were investigated. Fish were exposed at sublethal concentrations of CBZ (1.0 μg/l, 0.2 mg/l and 2.0 mg/l) for 7, 21 and 42 days. Compared to the control group, fish exposed at higher concentration (0.2 mg/l or 2.0 mg/l) of CBZ showed significantly higher levels of hemoglobin, ammonia and glucose, and significantly higher plasma enzymes activities. During the exposure duration, erythrocyte count, hematocrit, mean erythrocyte hemoglobin, mean erythrocyte volume, mean color concentration and total protein content in all groups were not significantly different. At the highest test concentration (2.0 mg/l) of CBZ, oxidative stress was apparent as reflected by the significant higher lipid peroxidation and protein carbonyl levels in liver after 42 days exposure, associated with an inability to induce antioxidant enzymes activities including superoxide dismutase, glutathione peroxidase and glutathione reductase. After 42 days exposure, reduced glutathione level was significantly decreased in the fish exposed at 0.2 mg/l CBZ, compared with the control. In short, CBZ-induced physiological and biochemical responses in fish were reflected in the oxidant stress indices and hematological parameters. These results suggest that hepatic antioxidant responses and hematological parameter could be used as potential biomarkers for monitoring residual pharmaceuticals present in aquatic environment.  相似文献
