Annetocin is a structurally and functionally oxytocin-related peptide isolated from the earthworm Eisenia foetida. We present the characterization of the annetocin cDNA. Sequence analyses of the deduced precursor polypeptide revealed that the annetocin precursor is composed of three segments: a signal peptide, an annetocin sequence flanked by a Gly C-terminal amidation signal and a Lys-Arg dibasic processing site, and a neurophysin domain, similar to other oxytocin family precursors. The proannetocin showed 37.4-45.8% amino acid homology to other prohormones. In the neurophysin domain, 14 cysteines and amino acid residues essential for association of a neurophysin with a vasopressin/oxytocin superfamily peptide were conserved, suggesting that the Eisenia neurophysin can bind to annetocin. Furthermore, in situ hybridization experiments demonstrated that the annetocin gene is expressed exclusively in neurons of the central nervous system predicted to be involved in regulation of reproductive behavior. These findings confirm that annetocin is a member of the vasopressin/oxytocin superfamily. This is the first identification of the cDNA encoding the precursor of an invertebrate oxytocin-related peptide and also the first report of the identification of an annelid vasopressin/oxytocin-related precursor.  相似文献

We have recently isolated a myoactive peptide, called leech excitatory peptide, belonging to the GGNG peptide family from two species of leeches, Hirudo nipponia and Whitmania pigra. Immunohistochemistry and in situ hybridization were employed to localize leech excitatory peptide-like peptide(s) and its gene expression in the central nervous system of W. pigra. A pair of neuronal somata were stained by both immunohistochemistry and in situ hybridization in the supraesophageal, subesophageal, and segmental ganglia. In addition, several other neurons showed positive signals by either immunohistochemistry or in situ hybridization in these ganglia. An immunoreactive fiber was observed to run in the anterior root of segmental ganglion 6, which is known to send axons to the sexual organs, though we failed to detect immunoreactivity in possible target tissues. Antiserum specificity was established by enzyme-linked immunosorbent assay using different leech excitatory peptide-related peptides. Leech excitatory peptide elicited muscular contraction of isolated preparations of penis and intestine at concentrations of 10(-8 )M. These results suggest that leech excitatory peptide is a neuropeptide modulating neuromuscular transmission in multiple systems, including regulation of reproductive behavior.  相似文献

Annetocin is an egg-laying-inducing oxytocin-related peptide which we have previously isolated from the earthworm, Eisenia foetida. Here we report the results of immunohistochemical and ultrastructural studies on annetocin-secretory cells in the earthworm. Annetocin-immunoreactive (IR) cell-somata were located mainly at the ventro-lateral side of the subesophageal ganglion. Only four annetocin-IR cells were seen in the cerebral ganglion. Some annetocin-IR cells displayed unipolar-like structure with a process directing to the core region (the neuropile) of the ganglion. Annetocin-IR fibers were also observed in the neuropile of the ventral ganglia and the ventral nerve cord between the 4th and the 30th segments including the clitellum, but not in the posterior segments (31-55th). The number of annetocin-IR fibers decreased from the 4th to the 30th segment. The annetocin-secretory cells were identified by the immunogold staining, and filled with gold-labeled vesicles, 200-250 nm in diameter, which included moderately electron dense material. The annetocin-secretory cells possessed a euchromatic nucleus, well-developed rough endoplasmic reticulum and Golgi apparatus. Some of the annetocin-secretory cells were found to form a neurohemal-like structure, where somata or fibers with loose glial investment came in contact with the coelomic space at the ventral side of the subesophageal ganglion. The results suggest that annetocin is a neuropeptide produced and secreted by the neuron in the cerebral and subesophageal ganglia of the earthworm.  相似文献

Leeches maintained either in tap-water or under desiccated conditions show a continuous decline in body-weight. Debrained leeches show a significantly greater loss of weight than unoperated or sham-operated controls. Injections of brain homogenate (0.2 ml/leech = 3 brains/leech) significantly (p<0.01) reduces weight loss in debrained leeches, whereas saline injections are without effect. The results indicate that water (weight) loss is influenced by a neurosecretory hormone secreted by the brain of Poecilobdella viridis.

1. This review covers the pharmacology and physiology of the body wall muscle systems of nematodes and annelids.2. Both acetylcholine and gamma-aminobutyric acid (GABA) play important roles in the control of body wall muscle in both phyla. In annelids and nematodes, acetylcholine is the excitatory neuromuscular transmitter while GABA is the inhibitory neuromuscular transmitter. In addition, 5-hydroxytryptamine (5-HT) has a modulatory role at annelid body wall muscle but little if any effect on nematode body wall muscle.3. The acetylcholine receptor of the body wall muscle can be classified as nicotinic-like in both phyla though the annelid receptor has not been analysed in detail. In nematodes, vertebrate ganglionic nicotinic agonists were the most effective of those so far examined while mecamylamine and benzoquinonium were the most effective antagonists. Both neuronal bungarotoxin and neosurugatoxin were potent antagonists of acetylcholine excitation at the nematode receptor.4. The GABA receptor of the body wall muscle exhibits similarities with the vertebrate GABA-A receptor in both phyla. Picrotoxin is a very weak or inactive antagonist at leech and nematode GABA receptors, while bicuculline methiodide blocks leech GABA receptors but is inactive on nematode GABA receptors. Picrotoxin does block GABA responses of earthworm body wall muscle. All these GABA responses are chloride mediated.5. Neuroactive peptides of the RFamide family occur m both phyla and FMRFamide has been identified in leeches. RFamides probably have an important role in heart regulation in leeches and in modulation of their body wall muscles. RFamides also modulate nematode body wall muscle activity with KNEFIRFamide raising muscle tone while SDPNFLRFamide relaxes the muscle. It is likely that this family and other neuroactivc peptides play an important role in the physiology of body wall muscle throughout both phyla.  相似文献

Blood-feeding invertebrates are emerging model taxa in biodiversity assessments, both as indicators of mammal abundance and also as sources of mammal DNA for identification. Among these, terrestrial leeches arguably offer the greatest promise; they are abundant and widespread in the humid tropics, and their blood meals can be easily assayed to establish diet. Unfortunately, terrestrial leeches are understudied, with little known about their ecology and behavior. Such information is needed to evaluate their utility as ecological indicators and to account for potential sampling biases that might arise from habitat preferences. By combining occupancy modeling and thermal tolerance assays, we determined the factors affecting species occurrence in the related terrestrial brown (Haemadipsa sumatrana) and tiger leech (Haemadipsa picta), both of which are widespread in tropical forests in Southeast Asia. We sampled both species across a degraded forest landscape in Sabah, Borneo, in wet and dry seasons, associating occurrence with habitat-level metrics. We found that, for both species, detection probability increased with canopy height regardless of season. Additionally, increased vegetation heterogeneity had a strong negative influence on brown leech occurrence in the dry season, implying an interaction between vegetation structure and climate. However, we found no difference in physiological thermal tolerance (CTMAX) between the two species. Finally, using a reduced dataset, we found a small improvement in brown leech model fit when including mammal abundance. Our results suggest that the presence of terrestrial leeches may act as useful ecological indicators of habitat quality and potentially mammalian abundance. A member of the GGNG peptide family was isolated from Hirudo nipponia (leech). GGNG peptides had only been isolated previously from earthworms. The C-terminus structure of the leech peptide, LEP (leech excitatory peptide), was –Gly–Gly–Asn–amide, while that of the earthworm peptides, EEP (earthworm excitatory peptide), was –Gly–Gly–Asn–Gly. LEP exerted 1000-fold more potent activities on leech gut than did EEP-2. On the other hand, EEP-2 was 1000-fold more potent than LEP on the crop-gizzard of the earthworm. Analog peptides of LEP and EEP-2 were synthesized, and the myoactive potency of each analog on the leech and earthworm tissues was compared.  相似文献

Parasites are important members of the ecological web within which an animal lives, and can be used as indicators of ecosystem health. However, few baseline parasitological data are available for free-ranging animals, particularly for the black howler monkey (Alouatta pigra). In this study a total of 283 fecal samples were collected from 50 individually identified A. pigra during 2003 and 2004 and examined for parasites. The samples were processed using standard quantitative centrifugation concentration techniques, with sugar and zinc sulfate used as flotation media. The slides were examined using bright-field and phase microscopy. Antigen capture enzyme-linked immunosorbent assays (ELISAs) were used to detect protozoa. Four parasites were detected: 1) Controrchis biliophilus (Dicrocoeliidae), 2) Trypanoxyuris minutus (Oxyuridae), 3) Giardia sp. (Hexamitidae), and 4) Entamoeba sp. (Endamoebidae). Controrchis biliophilus was detected in 80% (wet season) and 81% (dry season) of the A. pigra samples; Trypanoxyuris minutus was detected in 8% (wet season) and 27% (dry season) of samples; and Giardia sp. was detected in 40% (wet season) and 27% (dry season) of samples. For the first time, DNA from Giardia sp.-positive fecal samples was extracted from A. pigra. Alouatta pigra individuals that lived near human settlements in Belize were infected with Giardia duodenalis (syn. G. lamblia, G. intestinalis) Assemblages A and B. These results suggest that G. duodenalis is transmitted from people and/or domestic animals to A. pigra.  相似文献

The seasonal occurrence of medicinal leeches was studied at two lakes in the Black Sea coastland of north-western Turkey. The number of leeches attracted by a collector per hour was taken as an indicator for population density. Leeches were most abundant in May/June, and were almost absent during the cold season (November – March). Leeches with a weight of less than 1 g occurred principally between July and October with a peak in July. It is thought that these individuals represent newly hatched leeches. Although both study areas are close to each other, significant differences were found in respect to mean leech size and the seasonal frequency of leeches (both total number and certain age classes). It is suggested that leech biology is greatly dependent on factors such as microclimate and the availability of hosts, and no general conclusions on phenology can be drawn. There is therefore no scientific basis for restricting commercial leech collecting to certain months of the year.  相似文献

A novel GGNG-related neuropeptide from the polychaete Perinereis vancaurica   总被引:2,自引:0,他引:2  
The GGNG peptides are myoactive peptides so far identified from earthworms and leeches, which are the earthworm excitatory peptides (EEP) and the leech excitatory peptide (LEP), respectively. A novel GGNG peptide was isolated and structurally determined from a marine polychaete, Perinereis vancaurica, using a combination of immunological assay and high performance liquid chromatography (HPLC). The peptide was a pentadecapeptide whose amino acid sequence was similar to that of EEP and LEP, and showed myoactivity on isolated esophagus of P. vancaurica with a threshold concentration of 10(-10)M. The peptide was designated as polychaete excitatory peptide (PEP). Amidation of the alpha-carboxyl group of C-terminal residue occurred in PEP. This is the case for LEP, but not for EEP. The cDNA cloning revealed that the structure of the PEP precursor is more similar to the EEP precursor than to the LEP precursor. Immunohistochemical staining showed the presence of PEP in several neurons of central nervous system (CNS) as somata and neuropile structure, epithelial cells of the pharynx and epidermal cells throughout the body wall. Altogether these results support the physiological significance of PEP in regulation of the CNS neural activity and the peripheral myoactivity.  相似文献

Leaving the water to bask (usually in the sun) is a common behavior for many freshwater turtles, with some species also engaging in “nocturnal basking.” Ectoparasite removal is an obvious hypothesis to explain nocturnal basking and has also been proposed as a key driver of diurnal basking. However, the efficacy of basking, day or night, to remove leeches has not been experimentally tested. Therefore, we examined the number of leeches that were removed from Krefft''s river turtles (Emydura macquarii krefftii) after experimentally making turtles bask at a range of times of day, durations, and temperatures. Turtles had high initial leech loads, with a mean of 32.1 leeches per turtle. Diurnal basking under a heat lamp for 3 hr at ~28°C significantly reduced numbers of leeches relative to controls. In diurnal trials, 90.9% of turtles lost leeches (mean loss of 7.1 leeches per turtle), whereas basking for 30 min under the same conditions was not effective (no turtles lost leeches, and all turtles were still visibly wet). Similarly, “nocturnal basking” at ~23°C for 3 hr was not effective at removing leeches. Only 18% of turtles lost leeches (one turtle lost one leech and another lost four leeches). Diurnal basking outdoors under direct sunlight for 20 min (mean temp = 34.5°C) resulted in a small reduction in leeches, with 50% of turtles losing leeches and an average loss of 0.7 leeches per turtle. These results indicate basking can remove leeches if temperatures are high or basking durations are long. However, it was only effective at unusually long basking durations in this system. Our data showed even the 20‐min period was longer than 70.1% of natural diurnal basking events, many of which took place at cooler temperatures. Therefore, leech removal does not appear to be the purpose of the majority of basking events.  相似文献

Leeches are a unique group of annelids arising from an ancestor that would be characterized as a freshwater oligochaete worm. Comparative biology of the oligochaetes and the leeches reveals that body plan changes in the oligochaete-to-leech transition probably occurred by addition or modification of the terminal steps in embryonic development and that they were likely driven by a change in the feeding behavior in the ancestor of leeches. In this review article, developmental changes that are associated with the evolution of several leech-specific traits are discussed. These include (1) the evolution of suckers, (2) the loss of chaetae, (3) the loss of septa, and (4) a fixed number of segments. An altered developmental fate of the teloblast is further proposed to be a key factor contributing to the fixation of the segment number, and the evolutionary change in teloblast development may also account for the loss of the ability to regenerate the lost body segments in the leech.  相似文献

从表现软体、水肿、出血和硬结等特征的患病宽体金线蛭(Whitmania pigra)中分离到13株细菌分离物。通过培养特征、形态学观察及生化试验,确定了所分离的细菌中有6株为大肠杆菌(占46.15%),4株为变形杆菌(占30.77%),3株为沙门氏菌(占23.08%)。动物回归试验表明,接种后10d内大肠杆菌、沙门氏菌和变形杆菌对水蛭的致死率分别为100%(4/4)、100%(4/4)和25%(1/4);药敏试验结果表明,这三种细菌普通对复合磺胺、氯霉素、丁胺卡那霉素、头孢哌酮、丙氟哌酸、氟哌酸敏感。研究结果表明,本病是由多个病因导致的一种临床综合症。因此,在进行药物治疗时,应选用多种药物联合以达到最佳的治疗效果。  相似文献

The effect of short term influence of different concentrations of Pb(NO3)2 in the soil on the accumulation and localization of lead in the body wall cells of the earthworm Eisenia foetida (Savigny), Oligocheta, was studied histochemically. The experimental animals displayed the presence of lead deposits in the cytoplasm and vacuols of the epithelial gland cells which also showed morphological changes indicative of their increased secretory activity. Furthermore, a considerable accumulation of lead was found in the sarcoplasm and particularly in the sarcoplasmic reticulum of the muscle cells in the body wall. The histochemical data show that the most intense accumulation of lead is found in animals kept in an environment containing 0-05 mg Pb per 1 g of the soil. At this concentration of lead noteworthy morphological changes together with deposits of this metal are found in the mitochondria. This mitochondrial changes occurred together with the above mentioned, lead induced cytological deviations. They did not appear in other groups of experimental animals.  相似文献

The selection of distinct movements involved in various body postures and locomotion is often dependent on higher-order descending neurons. To study how such cells select different actions, we used a nearly-intact leech preparation (Hirudo sp.) in which cephalic projection interneurons were recorded and stimulated while the leech generated overt behaviors. Two long-distance projecting neurons were identified in the sub-packet of the third neuromere (R3b) of the subesophageal ganglion. These interneurons, named R3b2 and R3b3, produced changes in whole-body posture, crawling and swimming. Cell R3b2 reliably caused the body to become turgid, to hyper-elongate, and to thrash cyclically. Such robust activity resembled struggling behavior exhibited by intact leeches when grasped. The neighboring cell R3b3 elicited body elongation accompanied by a static whole-body bend to the left or right. R3b3 activity was context-dependent, oscillated in phase with crawling, reset the crawl rhythm, and terminated swimming. Both neuronal types responded to multi-modal sensory stimulation delivered to various rostral and caudal regions of the body. Our study illustrates the need to study behavioral selection with a neuroethological approach, and provides a cellular substrate for the motor action-selection cluster proposed for the vertebrate brainstem.  相似文献

Using immunocytochemistry techniques, we demonstrated specific immunostaining with antibodies rose against fragment (547-560) and (614-626) of bovine chromogranin B (CGB) at the level of brain and the tegument of the leech Theromyzon tessulatum. The used of these antibodies on leech sections revealed immunopositive labeling in neurons and glial cells of the central nervous system (CNS), and epidermal glandular cells of the tegument. Co-localization between two antibodies rose against different fragment of bovine CGB have been demonstrated in neurons and glial cells of leech CNS like in vertebrates. Finally, the whole of the data showed for the first time the presence in leeches of CGB like derived peptides.  相似文献

Summary Raising the blood volume in leeches by blood transfusion from donor leeches resulted in temporarily increased urinary flow. Displacement of the blood within the leech by massage, produced temporarily increased urinary flow in segments with elevated blood volume and seemed to decrease urinary flow in segments with lowered blood volume. It is suggested that blood volume and body water homeostasis are controlled by a feed-back system.  相似文献

The serotonin (5-hydroxytryptamine, 5-HT) content of tissue compartments in the medicinal leech, Hirudo medicinalis, was measured by means of high-pressure liquid chromatography coupled with electrochemical detection (HPLC-EC). Each segmental ganglion contains 21.3 +/- 2.9 (9) pmol 5-HT [X +/- SEM (N)]. The pharynx contains 7.1 +/- 1.1 (9) pmol 5-HT/mg wet weight; the salivary glands 3.2 +/- 0.9 (10), ventral body wall 2.0 +/- 0.2 (11), and vasofibrous tissue 1.2 +/- 0.2 (11). The blood of hungry leeches contains 8.7 +/- 1.9 (7) nM 5-HT while that of well-fed leeches is 2.2 +/- 0.4 (6) nM. Leeches were injected with the cytotoxic analog of serotonin, 5,7-dihydroxytryptamine (5,7-DHT) producing selective lesions of the peripherally projecting serotonin-containing neurons, and which in turn abolished their feeding behavior. The serotonin content of the pharynx and ganglia of these toxin-treated leeches were lowered significantly. The serotonin levels within the body wall and salivary glands were not altered significantly by the toxin treatment, but the levels within the vasofibrous tissue and blood were elevated substantially.  相似文献

During October 2004 to April 2005, when water temperature ranged between 18 and 22 degrees C, over than 500 specimens of leeches namely Alboglossiphonia polypompholyx, Batracobdelloides tricarinata, Helobdella conifera (Rhynchobdellida), Barbronia assiuti, and Salifa delicata (Arhynchobdellida) were collected from Al-Sont canal near Assiut city. About 80% of these specimens were infected with the peritrich ciliate Epistylis sp. The infection occurred in the field, both rhynchobdellid and arhynchobdellid leeches were infected, but Epistylis can infect rhynchobdellids more than arhynchobdellids. Both light and scanning electron microscopy showed the morphology of this parasite which attached to the surface of the leech body by a stalk. Histopathological studies revealed the damage of cuticle, epidermis, and dermis of the leech body wall at the area of attachment and the leech body seemed to be sloughy. Presence of this parasite in high numbers covering large areas of the body can impede gas exchange leading to leech suffocation and finally its death. Like the other sessile single-celled ciliates Epistylis spp. feed on bacteria and suspended organic debris, which are prevalent in nutrient-rich water, hence, Epistylis is a good indicator of poor water quality (pollution). The present study represents the first report of the effect of Epistylis on leeches collected from Egypt.  相似文献

The leeches Whitmania pigra and Hirudo nipponia live in similar environments but have different feeding habits. At present, there are few studies of the foraging mechanism of leeches with different feeding habits. In this study, we first used maze tests to show that these two species of leeches could locate and distinguish their prey through chemosensory activity without mechanical stimulation. However, the two leech species have different foraging behaviors: Individuals of W. pigra move slowly and repeatedly adjust direction through probing and crawling to detect the location of prey (snails), whereas individuals of H. nipponia move quickly, and after determining the location of food (porcine blood), they quickly swim or crawl to the vicinity of their prey. Scanning electron microscopy (SEM) revealed that there are two types of sensory cilia and pore structures related to mucus secretion in the heads of both leeches. There are two differently sized types of chemoreceptors on the dorsal lip in W. pigra, which may have different functions during foraging, whereas in H. nipponia there is only one type of chemoreceptor, which is small. We detected the chemical components in the natural food of these two leech species by UHPLC–MS. There were 934 metabolites in the body fluid of snails and 751 metabolites in porcine serum; five metabolites unique to the body fluid of snails and to porcine serum were screened as candidate feeding attractants. Of these metabolites, betaine and arginine effectively attracted individuals of W. pigra and H. nipponia, respectively. In summary, leeches with different feeding habits use chemoreceptors to sense external chemical signals when foraging, and there are significant differences between species in foraging behavior, chemoreceptors, and attractants.  相似文献
