The development and fine structure of articulated anastomosinglaticifers in Papaver somniferum were studied. Laticifers arenot present in the embryos but differentiate soon after germinationand are found in the phloem areas 18–30 h after the seedis sown. Laticifers and sieve elements are generally separatedby at least one cell layer in the roots, but in cotyledons,stems, and leaves they usually occur adjacent to each other. As the laticifers differentiate an abundance of vesicles formsin the cytoplasm. This process appears to involve the endoplasmicreticulum and it is suggested that the vesicles may be a specializedform of vacuole. Substances present in the vesicles react stronglywith iodine-potassium iodide. Laticifer-cytoplasm persists peripherallyand between the vesicles. It contains the usual cell organelles,the presence of which substantiates an active metabolic rolefor the laticifer contents.  相似文献

Summary. In this paper, a system of laticifers in Camptotheca acuminata Decne (Nyssaceae) is described. Laticifers were already present in the leaf primordia of the shoot apex. In the mature leaves, laticifers were found in the midrib and in the larger veins, both in the parenchymatic region delimited by vascular bundles and in the cortex just external to the phloem. In the stem, laticifers were present in both the primary and secondary body, running parallel to the longitudinal axis. They were located in the pith and in the cortex proximal to the phloem. No laticifers were found in the roots. The histochemical analyses indicated that the main compounds accumulated in laticifers were phenols. Neutral lipids and fatty acids were also present. Ultrastructural observations showed osmiophilic globules both in the vacuoles and in the peripheral regions of the cytoplasm of the laticifer cells. Plastids were present, although altered, with some parallel membranes and lacking starch grains. The discovery in C. acuminata of a laticifer system, which had never been described for the order Cornales, could be of taxonomic value, also considering that this order has traditionally represented one of the most problematic groups of flowering plants. The downy mildew pathogen of basil (Ocimum spp.) has caused considerable damage throughout the past five years, and an end to the epidemics is not in sight. The downy mildew of coleus (Solenostemon spp.) is just emerging and here we report that it was very recently introduced into Germany. Although it has been recognised that these pathogens are a major threat, the identity of the pathogens is still unresolved, and so it is difficult to devise quarantine measures against them. Using morphological comparison and molecular phylogenetic reconstructions we confirmed in this study that the downy mildews of basil and coleus are unrelated to Peronospora lamii, which is a common pathogen of the weed Lamium purpureum. In addition, we conclude by the investigation of the type specimen of P. swingleii and downy mildew specimens on Salvia officinalis that the newly occurring pathogens are not identical to P. swingleii on Salvia reflexa. The taxonomy of the downy mildew pathogens of hosts from the Lamiaceae and, in particular, from the tribes Mentheae and Elsholtzieae, is discussed, and a new species is described to accommodate the downy mildew pathogen of basil and coleus, which is the first downy mildew pathogen known to be parasitic to hosts of the tribe Ocimeae.  相似文献

Laticifers of Papaver bracteatum Lindl., population Arya II, seedlings were examined by electron microscopy. Laticifers were first differentiated in procambium of the radicle associated with phloem about 72 hr after seeds were sown. Proliferation of membrane-bound vesicles of apparent endoplasmic reticulum origin distinguished laticifers from adjacent cells. Vesicles developed electron-dense caps from the internal condensation of small particles. Laticifer initials possessed the usual complement of organelles that became obscured in mature cells by the large, closely packed vesicles. Plastids contained an electron-dense, membrane-bound inclusion, but never developed lamellae or starch grains. Articulation and anastomoses between laticifer elements resulted from gradual removal of wall materials by both cells on opposite sides of the common walls at a perforation site. Differentiation of the laticifer initials and the micromorphology of the protoplast of P. bracteatum is similar to that reported for P. somniferum.  相似文献

Latex from the opium poppy, Papaver somniferum L., was analyzed by polyacrylamide gel electrophoresis (PAGE). Two latex-specific bands were identified in protein samples of poppy latex using one-dimensional native PAGE. Second dimension analysis with SDS-PAGE indicates that these proteins have a relative molecular weight of approximately 20 kilodaltons. We have termed these polypeptides the major latex proteins (MLPs). Polyclonal antibodies prepared against the MLPs were used to probe protein gel blots of latex and poppy tissues known to lack laticifers. Laticifer-free tissues showed no reaction with anti-MLP immunoglobulin G indicating that MLPs are found only in poppy latex. MLP distribution was also examined in mature opium poppy tissues by immunocytochemistry. Laticifers were differentially labeled by fluorescein isothiocyanate secondary labeling of anti-MLP immunoglobulin G and could easily be identified in both transverse and longitudinal section. Fractionation studies of isolated latex showed that MLPs are concentrated in the latex cytosol and not in alkaloidal vesicles. Analysis of latex proteins by conventional two-dimensional electrophoresis indicates that the two MLP bands are composed of several distinct polypeptides with similar relative molecular weights. The pIs of these molecules range from 6.0 to 3.5. The role(s) of MLPs in laticifer metabolism has not been determined.  相似文献

Two known sesquiterpene lactones, antheindurolides A and B, together with five new related lactones with the same unusual skeleton were isolated from the aerial parts of Anthemis arvensis growing in Serbia. The originally proposed structure of antheindurolide B was revised. This (antheindurolide) type of lactones, detected so far only in the genus Anthemis (Anthemis indurata, Anthemis pseudocotula and Anthemis cotula) could be of chemotaxonomic significance.  相似文献

Hyaloscyphaceae is the largest family in Helotiales, Leotiomycetes. It is mainly characterized by minute apothecia with well-differentiated hairs, but its taxonomic delimitation and infrafamilial classification remain ambiguous. This study performed molecular phylogenetic analyses using multiple genes including the ITS-5.8S rDNA, the D1–D2 region of large subunit of rDNA, RNA polymerase II subunit 2, and the mitochondrial small subunit. The primary objective was to evaluate the phylogenetic utility of morphological characters traditionally used in the taxonomy of Hyaloscyphaceae through reassessment of the monophyly of this family and its genera. The phylogenetic analyses inferred Hyaloscyphaceae as being a heterogeneous assemblage of a diverse group of fungi and not supported as monophyletic. Among the three tribes of Hyaloscyphaceae only Lachneae formed a monophyletic lineage. The presence of hairs is rejected as a synapomorphy, since morphologically diversified hairs have originated independently during the evolution of Helotiales. The true- and false-subiculum in Arachnopezizeae are hypothesized to have evolved through different evolutionary processes; the true-subiculum is likely the product of a single evolutionary origin, while the false-subiculum is hypothesized to have originated multiple times. Since Hyaloscyphaceae sensu lato was not resolved as monophyletic, Hyaloscyphaceae sensu stricto is redefined and only applied to the genus Hyaloscypha.  相似文献

The kipunji, a recently discovered primate endemic to Tanzania's Southern Highlands and Udzungwa Mountains, was initially referred to the mangabey genus Lophocebus (Cercopithecinae: Papionini), but subsequent molecular analyses showed it to be more closely related to Papio. Its consequent referral to a new genus, Rungwecebus, has met with skepticism among papionin researchers, who have questioned both the robustness of the phylogenetic results and the kipunji's morphological distinctiveness. This circumstance has been exacerbated by the immaturity of the single available specimen (FMNH 187122), an M1-stage juvenile. Therefore, a geometric morphometric analysis of juvenile papionin cranial shape was used to explore the kipunji's phenetic affinities and evaluate morphological support for its separation from Lophocebus. Three-dimensional craniometric landmarks and semi-landmarks were collected on a sample of 124 subadult (dp4-M2 stage) cercopithecid crania. Traditional interlandmark distances were compared and a variety of multivariate statistical shape analyses were performed for the zygomaxillary region (diagnostic in mangabeys) and the cranium as a whole. Raw and size-adjusted interlandmark distances show the kipunji to have a relatively taller, shorter neurocranium and broader face and cranial base than is seen in M1-stage Lophocebus. Principal components and cluster analyses consistently unite the two Lophocebus species but group the kipunji with Cercocebus and/or Macaca. Morphological distances (Mahalanobis D2) between the kipunji and Lophocebus species are comparable to distances between recognized papionin genera. Discriminant function analyses suggest phenetic affinities between the kipunji and Cercocebus/Macaca and do not support the kipunji's classification to Lophocebus or to any other papionin taxon. In canonical plots, the kipunji occupies a region intermediate between macaques and African papionins or groups with Cercocebus, suggesting that it retains basal papionin shape characteristics. In shape comparisons among M1-stage papionins, the kipunji cranium is distinguished from Lophocebus by its relatively unrestricted suborbital fossa, more parasagittally oriented zygomatic arches, and longer auditory tube and from all papionins by its relatively tall, short neurocranium, broad face and cranial base, short nasals, dished nasal profile, and dorsally oriented rostrum. The kipunji is thus a cranially diagnosable phenon with a unique combination of cranial traits that cannot be accommodated within Lophocebus as currently defined. Based upon these results, Rungwecebus appears to be a valid and useful nomen that accurately reflects the morphological diversity of African papionins.  相似文献

Natural rubber was identified for the first time in the latex of Ficus benghalensis, and the rubber biosynthetic activity in latex and rubber particles was investigated. 13C NMR analysis of samples prepared by successive extractions with acetone and benzene confirmed that the benzene-soluble residues were natural rubber, cis-1,4-polyisoprene. The rubber content in the latex of F. benghalensis was approximately 17 %. Gel permeation chromatography revealed that the molecular mass of the natural rubber from F. benghalensis was approximately 1 500 kDa. The high rubber content and large molecular size suggest that F. benghalensis is a good candidate for an alternative rubber source. Examination of latex serum from F. benghalensis by SDS-polyacrylamide gel electrophoresis revealed a small number of proteins with major proteins of 31 and 55 kDa in size. The 31-kDa protein was predominant in catalytically-active rubber particles. Determination of metal ion concentration in latex and a comparison of the effect of ethylenediamine-tetraacetic acid on in vitro rubber biosynthesis in F. benghalensis, F. carica and Hevea brasiliensis suggest that the divalent metal ion present in latex serum is an important physiological factor controlling the rubber biosynthetic activities in these plant species. Microscopic examination revealed that the rubber in F. benghalensis occurred in a series of laticifer cells located in concentric zones in the inner bark of stems and branches.  相似文献

Laticifers are highly specialized cells present in over 20 plant families. They are well defined in planta. In vitro development of laticifers was also observed in some plants, but uncertain in the callus cultures of rubber tree, one of the most economically important latex producing plants. In the present study, we provide evidence that laticifer cells present in the callus cultures of rubber tree by histochemical and immunohistochemical studies. They present in the callus mainly as separate non-elongated form, a novel morphology different from the morphology of laticifer cells in planta, excluding their origin from explants. The occurring frequency of laticifer cells in the callus was genotype-dependent and negatively correlated with the somatic embryogenetic ability, suggesting that the presence of laticifer cells in the callus inhibit somatic embryogenesis in tissue culture of rubber tree. The genotypes PR107, RRIM600, Reyan8-79, and Reyan7-33-97 with lower embryogenetic ability compared to Haiken 2 had more laticifer cells, and laticifer clusters were only observed in these genotypes.  相似文献

外源钙离子对植物响应外界环境胁迫具有重要作用.寄生植物对入侵植物具有显著的抑制作用.以入侵植物喜旱莲子草为研究对象,采用完全随机区组的实验设计分析不同钙离子浓度与南方菟丝子寄生对喜旱莲子草茎形态结构的影响,探讨外源钙离子在寄主植物响应寄生植物胁迫中的作用.研究结果表明南方菟丝子寄生可以显著改变喜旱莲子草茎的形态结构,如显著降低喜旱莲子草茎的总长、分枝数、分节数、茎直径和髓腔直径,显著增加茎的厚角厚度与皮层厚度.外源钙离子对喜旱莲子草茎总长、分枝数、分节数、茎直径、髓腔直径和维管束直径没有显著影响,但可增加喜旱莲子草茎的维管束数目,降低茎的节间长、厚角厚度与皮层厚度.外源钙离子与南方菟丝子寄生相互作用可以显著增加喜旱莲子草茎的厚角厚度与皮层厚度,表明南方菟丝子寄生与高浓度的钙离子对喜旱莲子草茎的厚角厚度和皮层厚度具有显著的拮抗的交互作用.这种交互作用可以提高寄主植物的防御能力,减少寄生植物对寄主植物的损伤.  相似文献

In our screening program for new agrochemicals from local wild plants, essential oil of Artemisia vestita Wall (Asteraceae) was found to possess strong insecticidal activity against maize weevil, Sitophilus zeamais Motsch. Essential oil of aerial parts of A. vestita was obtained from hydrodistillation and was investigated by GC and GC–MS. The main components of essential oil were grandisol (40.29%), 1,8-cineol (14.88%) and camphor (11.37%). The essential oil of A. vestita possessed strong fumigant toxicity against S. zeamais adults with a LC50 value of 13.42 mg/L air. The essential oil of A. vestita also showed contact toxicity against S. zeamais adults with a LD50 value of 50.62 mg/adult.  相似文献

This paper deals with Neocalamites tubulatus nov. sp., an equisetophyte belonging to the Equisetaceae family. The material studied originates from the Upper Permian deposits of the northern part of European Russia. N. tubulatus nov. sp. is characterized by both sterile and fertile organs belonging to the same parent plant. It has long, flexible stems, rarely branched, with alternating nodes and prolonged ribs on the stem surface corresponding to vascular strands of the stem, and compact lateral strobili, in general aspect similar to the strobili of extant species of Equisetum. N. tubulatus nov. sp. is one of the most ancient representatives of the Equisetaceae family, which shows a distinct morphological pattern of features, among which the lateral position of strobili, characteristic of primitive equisetophytes, is most prominent in this phase of their evolution.  相似文献

Hispaniola Island was the first stopover in the travels of Columbus between America and Spain, and played a crucial role in the exchange of Phaseolus vulgaris seeds and their endosymbionts. The analysis of recA and atpD genes from strains nodulating this legume in coastal and inner regions of Hispaniola Island showed that they were almost identical to those of the American strains CIAT 652, Ch24-10 and CNPAF512, which were initially named as Rhizobium etli and have been recently reclassified into Rhizobium phaseoli after the analysis of their genomes. Therefore, the species R. phaseoli is more abundant in America than previously thought, and since the proposal of the American origin of R. etli was based on the analysis of several strains that are currently known to be R. phaseoli, it can be concluded that both species have an American origin coevolving with their host in its distribution centres. The analysis of the symbiovar phaseoli nodC gene alleles carried by different species isolated in American and European countries suggested a Mesoamerican origin of the α allele and an Andean origin of the γ allele, which is supported by the dominance of this latter allele in Europe where mostly Andean cultivars of common beans have been traditionally cultivated.  相似文献

The geographical distribution and analysis of the essential oils of species from three sections of Hypericum L. (Guttiferae/Clusiaceae/Hypericaceae) from Portugal are presented. Hypericum perfoliatum (section Drosocarpium) grows wild in the centre and south of Portugal; Hypericum humifusum and Hypericum linarifolium are both from section Oligostema, the former occurring throughout the country, while the second is distributed mainly in the north and centre; Hypericum pulchrum (section Taeniocarpium) is confined to the littoral north of Portugal. The essential oils were obtained by distillation–extraction, hydrodistillation and distillation in a modified Marcusson apparatus from the dried aerial parts of the different populations and were analysed by GC and GC–MS. Monoterpene hydrocarbons constituted the main fraction in all oils (43–69%, 53–85%, 28–45% and 48–65% for H. perfoliatum, H. humifusum, H. linarifolium and H. pulchrum, respectively). Sesquiterpene hydrocarbons (2–13%, 6–18%, 21–27% and 16–18%, respectively) and a third fraction of non-terpenic compounds (20–29%, 3–16%, 2–14% and 5–11%, respectively) from the four species attained relatively high amounts in all oils. Within each species, no major differences were detected in the essential oil composition, despite the fact that different locations, phenological phases and extraction methodologies were used. Notwithstanding the dominance of α-pinene in all four species' oils, cluster and principal components analysis on the identified components showed that the range of α-pinene, β-pinene and n-nonane supported a separation of the four species. The essential oil composition of the four species showed some qualitative resemblances, which correlate well with the taxonomical classification based on morphological characters.  相似文献
