The knowledge about a seed??s optical parameters is of great relevance in the seed technology practice. Such parameters provide information about its absorption and reflectance, which in turn is related to its color, quality, and health condition. The objective of the present study was to determine the optical absorption coefficient ?? for maize seeds ( Zea mays L.) by means of the photoacoustic spectroscopy (PAS). Untreated seeds (I) and seeds dyed with methyl red (II) were used in this investigation. In addition, conventional reflectance measurements (obtained with the integrating sphere) were performed to validate PAS absorption measurements. The results show that the absorption spectra and reflection data of the seed samples are complementary. When used with thermally thick and optically opaque seeds, PAS may be considered as a potential diagnostic tool for the characterization of the seeds. 相似文献

目的对广西不同产地番石榴叶中总黄酮含量进行测定。方法应用紫外分光光度法,以芦丁为对照品,于357nm下测定吸光度,计算番石榴叶中总黄酮的含量。结果在1.7~14.3范围内总黄酮含量与吸光度A值线性关系良好,平均回收率=99.28%,RSD=3.69%,R2=0.9993。结论该方法简便可靠、快速、重现性好,适用于番石榴叶总黄酮含量测定。采收地区不同,番石榴叶中总黄酮的含量也不同。 相似文献

Tolerance to photoinhibition was compared between a paraquat-resistant and a sensitive biotype of Conyza bonariensis (L.). Cronq. Photoinhibitory damage was measured as a decrease in oxygen evolution or energy storage using photoacoustic spectroscopy, or as a decrease of 14CO 2-fixation. Prior to exposure to high fluence rates, both biotypes had similar quantum yields of oxygen evolution and energy storage. After exposure to high intensity light, the resistant biotype continued to evolve oxygen and to store energy with a high quantum yield while both energy storage and oxygen evolution were severely reduced in the sensitive biotype. CO 2-fixation was less rapidly inhibited in the resistant biotype compared to the sensitive one. The data show that the paraquat resistant biotype with its high constitutive levels of the chloroplast localized enzymes of the oxygen detoxification pathway, is also partially protected from photoinhibition. This supports the theory that an enhanced radical scavenging system can give temporary protection against photooxidative damage from a variety of sources. 相似文献

Methanogenic activity (MA) tests are commonly carried out to estimate the capability of anaerobic biomass to treat effluents, to evaluate anaerobic activity in bioreactors or natural ecosystems, or to quantify inhibitory effects on methanogenic activity. These activity tests are usually based on the measurement of the volume of biogas produced by volumetric, pressure increase or gas chromatography (GC) methods. In this study, we present an alternative method for non-invasive measurement of methane produced during activity tests in closed vials, based on Infrared Tunable Diode Laser Absorption Spectroscopy (MA-TDLAS). This new method was tested during model acetoclastic and hydrogenotrophic methanogenic activity tests and was compared to a more traditional method based on gas chromatography. From the results obtained, the CH(4) detection limit of the method was estimated to 60ppm and the minimum measurable methane production rate was estimated to 1.09(.)10(-3)mgl(-1)h(-1), which is below CH(4) production rate usually reported in both anaerobic reactors and natural ecosystems. Additionally to sensitivity, the method has several potential interests compared to more traditional methods among which short measurements time allowing the measurement of a large number of MA test vials, non-invasive measurements avoiding leakage or external interferences and similar cost to GC based methods. It is concluded that MA-TDLAS is a promising method that could be of interest not only in the field of anaerobic digestion but also, in the field of environmental ecology where CH(4) production rates are usually very low. 相似文献

In this paper, we propose a scheme for achieving ultrafast coherent control of the selective excitation among three excited states in stimulated Raman scattering process on a tip-enhanced Raman spectroscopy (TERS). The center frequencies of the pump and Stokes laser pulses are 14,000 cm−1 and 12,500 cm−1, and their spectral bandwidths are both 700 cm−1. By properly modulating the spectral phase distribution and cutting the frequency components, the stimulated Raman transition probabilities of two excited state keep maximal, while the other one can be suppressed to zero. The shaped pump and Stokes pulse irradiate obliquely into the TERS nanostructure containing a single layer molecule. The impulse response in temporal and frequency domain is calculated by using finite-difference time-domain (FDTD) simulation followed by Fourier transform. The frequency components and the relative phase are same with the corresponding input pulses, but the intensities are enhanced by more than 10 times. Compared with the case without the TERS nanostructure, the probability of selective excited Raman transition increases by more than 4 orders of magnitude, and the selective depressed Raman peak keeps at 0. 相似文献

Short-term (4 hours) effect of different concentrations of SO 2 fumigation on in vivo photochemical activities of sugar maple ( Acer saccharum Marsh.) leaves was investigated using photoacoustic spectroscopy. The relative quantum yield of O 2 evolution (ratio of O 2 signal to the photothermal signal) and photochemical energy storage are increased by 0.05 microliter per liter of SO 2. This increase is more pronounced in 5 to 7 year old saplings than in 3 month old seedlings. Both oxygen-relative quantum yield and energy storage of seedlings are inhibited by increased concentrations of SO 2 and the inhibition is concentration dependent. The inhibition is greater in seedlings than in saplings at 2 microliters per liter of SO 2, indicating the more susceptible nature of seedlings. The present study indicates a concentration dependent differential effect of SO 2 on photochemical activities of sugar maple leaves. 相似文献

Carotenoids scavenge simple lipid-like alkylperoxyl radicals. However, the rate constant is too low to be determined directly and the mechanism is likewise not known with certainty [Mortensen, A. and Skibsted, L.H. (1998) FEBS Lett . 426 , 392-396]. It is demonstrated that carotenoids react with peroxyl radicals only slightly more reactive than lipidperoxyl radicals neither by electron transfer nor by hydrogen atom donation, but by adduct formation. Benzylperoxyl radicals are scavenged by the carotenoids g -carotene and canthaxanthin with a second-order rate constant of at least 1 u 2 u 10 6 u M m 1 u s m 1 by formation of an adduct which decays in a first-order reaction. 相似文献

Near-infrared reflectance spectroscopy (NIRS) has been used extensively in the forage industry for rapid measurement of forage constituents and could be useful for determining quality of biomass feedstocks at the point of delivery. In previous work, we developed an assay that partitions feedstock carbohydrates based on their availability to be converted to fermentable sugars, including non-structural carbohydrates ( C N), biochemically available carbohydrates ( C B) with an associated first-order availability rate constant ( k B), and unavailable carbohydrates ( C U ). Additional quality parameters measured included neutral detergent lignin (NDL), total available carbohydrates ( C A), and total carbohydrates ( C T). We evaluated the variability of biomass quality parameters in a set of corn stover samples and developed calibration equations for determining parameter values using NIRS. Fifty-two corn stover samples harvested in Iowa and Wisconsin in 2005 and 2006 were analyzed using a high-throughput assay for determining feedstock quality for biochemical conversion. Non-structural carbohydrates ranged from 84 to 155?g?kg ?1 dry matter (DM); C B ranged from 354 to 557?g?kg ?1 DM; k B ranged from 0.199 to 0.330?h ?1; C A ranged from 463 to 699?g?kg ?1 DM, and NDL ranged from 32 to 74?g?kg ?1 DM. Significant differences ( P?<?0.0001) among samples were observed for all parameters, except k B. Near-infrared reflectance spectroscopy calibration equations were developed for C N, C B, C A, C U , C T, and NDL. It was not possible to generate a meaningful calibration equation for k B. There is significant variability within the corn stover population for several key quality-related carbohydrate and lignin constituents which can be predicted reliably using NIRS. 相似文献

Using an optical beating technique, the diffusion coefficients and relative scattered intensity of Escherichia coli 70S, 50S, and 30S ribosomes are measured as a function of temperature and Mg 2+ concentration. For solutions at 10 mM Mg 2+ and between 0°C and about 40°C, the values of D20,w obtained are 1.7, 1.9, and ≈2.1 × 10 -7 cm 2/s, respectively. Preparative procedures drastically affect these values and equivalent hydrodynamic ellipsoids of revolution models give large axial ratios indicating extensive hydration or a deviation from the assumed shape. Calculations also indicate that the subunits expand upon dissociation. Measurements of D20,w vs. temperature indicate that 70S particles undergo a conformational change prior to dissociation and can be heat dissociated at 30-32°C at low concentrations. Treatment of 70S ribosomes with EDTA causes a biphasic dissociation reaction. Addition of Mg 2+ after dissociation with EDTA shows that longer waiting times yield fewer 70S particles and that even short waiting times may yield ribosomes differing from the native conformation. Addition of p-chloromercuribenzoic acid (PCMB) is shown to dissociate 70S particles, but to a lesser extent than ethylenediaminetetraacetic acid (EDTA). 相似文献

Carotenoids are organic pigments that are produced predominantly by photosynthetic organisms and provide antioxidant activity to a wide variety of plants, animals, bacteria, and fungi. The carotenoid biosynthetic pathway is highly conserved in plants and occurs mostly in chromoplasts and chloroplasts. Leaf carotenoids play important photoprotective roles and targeted selection for leaf carotenoids may offer avenues to improve abiotic stress tolerance. A collection of 332 soybean [ Glycine max (L.) Merr.] genotypes was grown in two years and total leaf carotenoid content was determined using three different methods. The first method was based on extraction and spectrophotometric determination of carotenoid content (eCaro) in leaf tissue, whereas the other two methods were derived from high-throughput canopy spectral reflectance measurements using wavelet transformed reflectance spectra (tCaro) and a spectral reflectance index (iCaro). An association mapping approach was employed using 31,253 single nucleotide polymorphisms (SNPs) to identify SNPs associated with total carotenoid content using a mixed linear model based on data from two growing seasons. A total of 28 SNPs showed a significant association with total carotenoid content in at least one of the three approaches. These 28 SNPs likely tagged 14 putative loci for carotenoid content. Six putative loci were identified using eCaro, five loci with tCaro, and nine loci with iCaro. Three of these putative loci were detected by all three carotenoid determination methods. All but four putative loci were located near a known carotenoid-related gene. These results showed that carotenoid markers can be identified in soybean using extract-based as well as by high-throughput canopy spectral reflectance-based approaches, demonstrating the utility of field-based canopy spectral reflectance phenotypes for association mapping. 相似文献

Carotenoids produced by Sporidiobolus pararoseus were studied. It was found that biomass was connected with carbon source, temperature, and pH, but carotenoids proportion was seriously influenced by dissolved oxygen and nitrogen source. Different carotenoids could be obtained by using selected optimum conditions. In the end we established the strategies to produce β-carotene or torulene. Fed-batch fermentation in fermentor was used to prove the authenticity of our conclusions. The cell biomass, β-carotene content, and β-carotene proportion could reach 56.32?g/L, 18.92?mg/L and 60.43%, respectively, by using corn steep liquor at 0-5% of dissolved oxygen saturation. β-Carotene content was 271% higher than before this addition. The cell biomass, torulene content, and torulene proportion could reach 62.47?g/L, 31.74?mg/L, and 70.41%, respectively, by using yeast extract at 30-35% of dissolved oxygen saturation. Torulene content was 152% higher than before this addition. The strategy for enhancing specific carotenoid production by selected fermentation conditions may provide an alternative approach to enhance carotenoid production with other strains. 相似文献

Animals must allocate finite resources amongst competing demands. A suite of such trade-offs is thought to occur in the deployment of carotenoids, being widely responsible for sexual coloration and also important in antioxidant and immune defences. Experimental manipulation of dietary carotenoid availability is a useful approach for elucidating the mechanistic bases of carotenoid allocation trade-offs. Recent work using birds has shown that both sexual display and immune defences can be limited by carotenoid availability, providing support for the hypothesis that males allocating greater amounts of carotenoids to sexual coloration are advertising their superior health. Carotenoid availability has also been shown to limit egg-laying capacity in birds, although it remains to be seen whether carotenoid display in females advertises reproductive potential. More experiments are required to ascertain the importance of direct (material) and indirect (genetic) benefits accruing through choosing to mate with individuals that have greater carotenoid display. 相似文献
