Summary Small intensely fluorescent (SIF) cells appeared singly or, more frequently, in variably-sized clusters in the sacroccygeal 8th and 9th sympathetic ganglia of the bullfrog. Smaller clusters containing only two to nine SIF cells accounted for 61% of 1773 clusters examined. The largest cluster contained 283 cells. The number of cells in individual ganglia also varied from 21 to 3332. SIF cells, solitary as well as in smaller clusters, received no distinct form of the synaptic contact. In contrast, the cells in larger clusters were frequently innervated by nerve endings that were similar in vesicular constitution to the nerve endings on principal ganglion (PG) cells. No synaptic contact was found between SIF cells and PG cells. SIF cells were also characterized by their location in the vicinity of blood capillaries with a continuous endothelium. p]Our observation seems to suggest that larger clusters of SIF cells receiving nerve endings are linked to a paracrine and/or endocrine system. Chemical influence via the blood stream and intraganglionic milieu for non-innervated SIF cells in the solitary or smaller clusters is a subject for speculation. An interneuronal role of SIF cells to relay stimuli to PG cells seems unlikely. The possible functions here assigned to SIF cells could be variable in efficiency depending on their population and density.  相似文献

Summary The ultrastructure of the paracervical (Frankenhäuser) ganglion in the rat was studied after immersion or perfusion fixation with glutaraldehyde followed by post-osmification. This ganglion is located at the uterovaginal junction in the vicinity of arteria uterina and contains three neuronal cell types. (1) Principal neurons have a fine structure mainly similar to the ganglion cells of other autonomic ganglia. (2) Small granule-containing cells occur in clusters often close to fenestrated capillaries. They are divided into two subgroups according to the size of their cytoplasmic granules; those containing only small granulated vesicles of 800 to 1400 Å in diameter and those having also large granulated vesicles of 2000 to 3000 Å in diameter. (3) Vacuolated nerve cells are large cells that resemble the principal neurons in their cytoplasmic components, except that they contain one to ten vacuoles with corpuscles of different size and shape. The possible physiological significance of the small, granule-containing cells in the uterine function is discussed.  相似文献

Mycolic acid patterns of some species of Mycobacterium   总被引:5,自引:0,他引:5  
Representative strains of some species of Mycobacterium were degraded by both acid and alkaline methanolysis. Two-dimensional thin-layer chromatography was used to determine the patterns of mycolic acids and other long-chain components in these methanolysates. Patterns composed of -, methoxy- and ketomycolates were found in Mycobacterium asiaticum, Mycobacterium bovix, Mycobacterium gastri, Mycobacterium gordonae, Mycobacterium kansasii, Mycobacterium marinum and Mycobacterium tuberculosis; a representative of Mycobacterium thermoresistibile also contained lower molecular weight -mycolates in addition to these three acids. In representatives or Mycobacterium avium, Mycobacterium intracellulare, Mycobacterium nonchromogenicum, Mycobacterium novum, Mycobacterium paratuberculosis, Mycobacterium scrofulaceum, Mycobacterium terrae, Mycobacterium xenopi, and Mycobacterium sp. MNC 165 - and ketomycolates were accompanied by -carboxymycolates and 2-eicosanol and homologous alcohols which are derived from wax-ester mycolates. Mycobacterium fortuitum and Mycobacterium giae contained - and epoxymycolates and both serovars of Mycobacterium simiae had a very characteristic pattern of -, - and ketomycolic acids. Summary The distribution of cholinergic neurons in the urinary tract and male genital organs of the cat was studied by a histochemical method for acetylcholinesterase. In addition to cell clusters in autonomic ganglia (intraganglionic cells), isolated extraganglionic cholinergic cells were found within the innervated tissues, usually in association with nerve trunks and blood vessels. Smaller neural cells with multiple axonal processes, identical to Cajal's interstitial cells, were found in the meshes of the terminal nerve plexus in smooth muscle, lamina propria and vascular wall.It is concluded that peripheral cholinergic neurons, like their adrenergic analogues, are arranged as a short intraganglionic, a shorter extraganglionic, and a terminal system of neurons.Supported in part by grants 10465 and 11285 from the USPHS and the Henry C. Summary The localization of l-glutamate decarboxylase (GAD), the GABA-synthesizing enzyme, was studied in the rat major pelvic ganglion and in the coeliac-superior mesenteric ganglion complex by indirect immunofluorescence technique with a specific antiserum raised in rabbits. GAD immunoreactivity was demonstrated in small cells of these ganglia. The GAD-immunoreactive small cells were 10–20 m in diameter and formed clusters or occured as solitary cells. The principal neurons were non-reactive but they were surrounded by immunoreactive processes. Studies on colocalization of GAD with tyrosine hydroxylase (TH), the rate-limiting enzyme of the catecholamine synthesis, in the major pelvic ganglion and in the coeliac-superior mesenteric ganglion complex indicated that all GAD-immunoreactive small cells were also labelled with TH. In the major pelvic ganglion all TH-immunoreactive SIF cells were also immunoreactive for GAD. However, in the coeliac-superior mesenteric ganglion complex there occured TH-immunoreactive small cells which showed no immunoreactivity to GAD. It is suggested that the small GAD-immunoreactive cells represent small intensely fluorescent (SIF) cells.  相似文献

Sixteen genetic lines of peas were screened for their ability to regenerate whole plants from callus cultures. Epicotyl sections from germinating seeds were placed on callus-inducing medium; the resulting callus was subcultured monthly and was tested every other month for its regeneration ability. Six lines were found that would regenerate after 2 months' growth as callus. Four of these continued to regenerate after 4 months and, of these, two after 6 months. Unlike most marine teleosts, the coral reef-dwelling spiny damsel, Acanthochromis polyacanthus, lacks a pelagic larva dispersal phase and represents one of few examples of self recruitment onto a natal reef by a marine teleost immediately after hatching. Benthic eggs are protected by the parents, and upon hatching the young remain under parental care for several months. Visual morphogenesis of spiny damsel embryos and juveniles was examined to evaluate the potential visual capabilities of the young after emergence onto the reef. The optic primordia were visible in the embryo as hollow spheres of undifferentiated neuroblasts 2 days after fertilization (daf). Visual morphogenesis proceeded rapidly thereafter in the embryo such that at hatching (between 10 and 12daf) gross visual morphology was consistent with that reported in the majority of juvenile marine teleosts, reflecting direct development of the retina of the spiny damsel within the egg. At hatching, the outer nuclear layer comprised 2 classes of photoreceptors; cones and rods. Tangential sections of the retina revealed a square cone mosaic in which 4 double cones surrounded a single cone. This arrangement remained unchanged in all later life history intervals examined. Absolute eye size was large compared to larvae of marine pelagic spawners. Eye and lens diameters increased from 0.69 and 0.23mm, respectively, on the day of hatching (12daf), to 3.77 and 1.52mm, respectively, in a fish 131daf. Angular density of cones increased from 0.25 cones 10 visual arc–1 in an embryo 8daf, to 1.14 cones 10 visual arc–1 in a fish 131daf, demonstrating the potential for significant increase in spatial resolution with increasing eye size. Convergence ratios of cones to ganglion cells remained relatively constant from the time of hatching, suggesting that the determinate ganglion cell photopic receptive field was established early in development. The increase in the convergence ratios of rods: ganglion cells from 1.4 in the late stages of embryogenesis (10daf; 2 days prior to hatching) to 4.9 in a fish 103daf, demonstrated increasing scotopic ganglion cell receptive field size, with increasing age. This was a result of rod cell addition with growth. An increase in the angular density of rods from 0.18 rods 10 visual arc–1 in an embryo 8daf, to 4.07 rods 10 visual arc–1 in a fish 131daf, and the increase in mean scotopic light path-length from 13.3±1.1m in an embryo 8dpf, to 55±5.2m a fish 22dpf, collectively indicate the potential for increasing scotopic sensitivity during growth. On the basis of visual morphology it is predicted that newly hatched spiny damsel juveniles have substantially greater visual capabilities than first feeding larvae with a pelagic dispersal phase. In addition, we propose that the developmental trajectory of the spiny damsel is different from that of pelagic dispersing larvae and does not simply reflect displacement along a common developmental continuum by an extended embryonic duration.  相似文献

Summary The distribution, morphology and synaptic connections of the hindgut efferent neurons in the last (sixth) abdominal ganglion of the crayfish, Orconectes limosus, have been investigated using light and electron microscopy in conjunction with retrograde cobalt/nickel and HRP labeling through the intestinal nerve. The hindgut efferent neurons occur singly and in clusters, and are unipolar. Their axonal projections are uniform and consist of a thick primary neurite with typical lateral projections and limited arborization of varicose fibers in the ganglionic neuropil. They also send lower order axon processes to the ganglionic neural sheath, where they arborize profusely, forming a network of varicose fibers. The majority of the efferent neurons project to the anterior part of the hindgut. HRP-labeled axon profiles are found in both pre- and postsynaptic position in the neuropil of the ganglion. HRP-labeled axon profiles also establish pre- and postsynaptic contacts in the intestinal nerve root. All hindgut efferent terminals contain similar synaptic vesicle populations: ovoid agranular vesicles (50–60 nm) and a few large granular vesicles (100–200 nm). It is suggested that the hindgut efferent neurons in the last abdominal ganglion are involved in: (1) innervation of the hindgut; (2) central integrative processes; (3) en route synaptic modification of efferent and afferent signals in the intestinal nerve; (4) neurohumoral modulation of peripheral physiological processes.Fellow of the Alexander von Humboldt Stiftung  相似文献

Summary The ganglionated plexus of the trachea of mice was studied quantitatively with a histochemical method that stains electively the ganglion nerve cells in whole-mount preparations. The plexus lies exclusively over the muscular part of the trachea, dorsal to the muscle itself, and it varies considerably in pattern and extent between individual animals. In young adult mice the plexus contains on average 235 neurons, occurring singly or gathered in packed ganglia. The ganglion neurons are relatively small, the profile area of three quarters of them measuring between 150 and 275 m2 with an average of 251 m2. In ageing mice the average number of ganglion neurons is the same as in young animals; however, cell sizes are markedly increased, the average being 341 m2. Among the ultrastructural features of the ganglia, is a capsule (perineurium) of very regular structure, the presence of collagen, capillaries and myelinated axons inside the ganglia, and the presence of only few and short dendrites, some of which are abutted by synapsing nerve endings.  相似文献

Summary The ontogenesis of small intensely fluorescent (SIF) cells and the adrenergic nerve plexus is described in stretch preparations of the rabbit portal vein. On the 25 to 26th days of gestation there was a predominance of SIF cells (8 to 30 m in diameter), but a few nerve fibres in bundles were also present. Each portal vein preparation contained 6 to 9 groups of cells. The distribution and number of SIF cells and nerve bundles remained constant until the 31st day of gestation at which stage the number of SIF cells had decreased, while the density of the nerve plexus had increased approximately 4-fold. The adult portal vein exhibited a dense adrenergic plexus, but SIF cells were absent from nine out of ten preparations.  相似文献

Summary The pelvic ganglia supply cholinergic and noradrenergic nerve pathways to many organs. Other possible transmitters are also present in these nerves, including peptides. Multiple labelling immunofluorescence techniques were used in this study of the male rat major pelvic ganglion (MPG) to examine: (1) the peptides present in noradrenergic (tyrosine hydroxylase (TH)-positive) and non-noradrenergic (putative cholinergic) neurons, and (2) the types of peptide-containing nerve fibres closely associated with these two groups of neurons. The distribution of the peptide galanin (GAL) within the MPG was also investigated. All of the TH-neurons contained neuropeptide Y (NPY), but none of the other tested peptides. However, many NPY neurons did not contain TH and may have been cholinergic. TH-negative neurons also displayed vasoactive intestinal peptide (VIP), enkephalin (ENK) or GAL. VIP and NPY formed the most common types of putative cholinergic pelvic neurons, but few cells contained both peptides. Many ENK neurons exhibited VIP, NPY or GAL. Varicose nerve terminals surrounding ganglion cells contained ENK, GAL, somatostatin (SOM) and cholecystokinin (CCK). These peptide-immunoreactive fibres were more often associated with the non-noradrenergic (putative cholinergic) than the noradrenergic neurons; two types (SOM and CCK) were preferentially associated with the non-noradrenergic NPY neurons. GAL was distributed throughout the MPG, in small neurons, scattered small, intensely fluorescent (SIF) cells, and both varicose and non-varicose nerve fibres. The nerve fibres were concentrated near the pelvic and penile nerves; most of the varicose fibres formed baskets surrounding individual GAL-negative somata.  相似文献

Sequence alignment and structure prediction are used to locate catalytic -amylase-type (/)8-barrel domains and the positions of their -strands and -helices in isoamylase, pullulanase, neopullulanase, -amylase-pullulanase, dextran glucosidase, branching enzyme, and glycogen branching enzymes—all enzymes involved in hydrolysis or synthesis of -1,6-glucosidic linkages in starch and related polysaccharides. This has allowed identification of the transferase active site of the glycogen debranching enzyme and the locations of loops making up the active sites of all enzymes studied. Activity and specificity of the enzymes are discussed in terms of conserved amino acid residues and loop variations. An evolutionary distance tree of 47 amylolytic and related enzymes is built on 37 residues representing the four best conserved -strands of the barrel. It exhibits clusters of enzymes close in specificity, with the branching and glycogen debranching enzymes being the most distantly related.  相似文献

The mangrove killifish Rivulus marmoratus was reared at 25°±1°C and 17ppt salinity from 0 to 100 days after hatching (DAH), and its early development was described by examining growth and morphometric parameters, meristic characters (vertebral and fin-ray counts), bone-cartilage development, and pigmentation. Growth was isometric for preanal length, head length, snout length, body depth, pectoral-fin length, dorsal-fin length, anal-fin length, and caudal-peduncle depth. Negative allometric growth was observed in eye diameter and gape size. Meristic counts (mean±SD) for vertebrae (34.2±0.4) and dorsal- (8.6±0.5), anal- (11.4±0.5), and caudal-fin rays (30.2±0.8) were complete at 0 DAH (n=5), whereas pectoral-fin rays and pelvic-fin rays were complete by 30 DAH (14.5±0.4, n=5) and 60 DAH (4.2±0.8, n=5). Full ossification of meristic elements proceeded in the following sequence: vertebrae (by 30 DAH), caudal-, dorsal-, and anal-fin rays (by 60 DAH), pectoral-fin rays (between 60 DAH and 100 DAH), and pelvic-fin rays (by 100 DAH). Both morphological characters and meristic counts indicate that this species can be considered to be a juvenile after 9.8mm in standard length (20 DAH).  相似文献

Incubation of synthetic Man\1-4GlcNAc-OMe, GalNAc1-4GlcNAc-OMe, Glc1-4GlcNAc-OMe, and GlcNAc1-4GlcNac-OMe with CMP-Neu5Ac and rat liver Gal1-4GlcNAc (2-6)-sialyltransferase resulted in the formation of Neu5Ac2-6Man1-4GlcNAc-OMe, Neu5Ac2-6GalNAc1-4GlcNAc-OMe, Neu5Ac2-6Glc1-4GlcNAc-OMe and Neu5Ac2-6GlcNAc1-4GlcNAc-OMe, respectively. Under conditions which led to quantitative conversion of Gal1-4GlcNAc-OEt into Neu5Ac2-6Gal1-4GlcNAc-OEt, the aforementioned products were obtained in yields of 4%, 48%, 16% and 8%, respectively. Summary DNA sequences of the human, mouse, and rabbit immunoglobulin kappa-gene (J-C regions) are compared with respect to various DNA patterns, including dyad symmetry pairings, runs of nucleotides, repeat clusters, and repeats that occur with unusually high frequency. The significant dyad symmetry pairings within each of the sequences emphasize the two control-enhancer elements of the J5-C intron. Dyad symmetry pairs between the J-C region and a number of kappa variable (V)-gene domains suggest differences in the affinities between the V and J segments. It is the consensus heptamer rather than the consensus nonamer that embodies the longest V-J dyad symmetry combinations. In the rabbit there are long runs and repeat clusters of the sequences that identify regions of high duplication; these regions are absent in the human and mouse sequences. High-frequency oligonucleotides feature the consensus nonamer 5 to the J segments, especially in the mouse sequence.  相似文献

Summary Immunohistochemistry of peptide-and dopamine--hydroxylase-(DBH)-containing varicose nerve fibres and ganglion cells, respectively, in the guinea pig inferior mesenteric ganglion was investigated followinga) transsection of mesenteric (colonic) branches,b) transsection of central (lumbar splanchnic, intermesenteric and hypogastric) branches, andc) transplantation into the spleen.The findings indicate that pathways of different opioid peptides are not identical Met-enkephalin-and met-enkephalin-arg-phe-(cleavage products from pre-proenkephalin) containing fibres course in central branches to make contact in the inferior mesenteric ganglion. Dynorphin-and -neo-endorphin-(deriving from pre-prodynorphin) containing fibers as well as leu-enkephalin-(included in the dynorphin sequence) fibres appear to rise not only from central and from enteric somata, but also from intraganglionic noradrenergic neurons. Similar pathways seem to be used by VIP-and by neurotensin-immunoreactive cell bodies are rare. Practically all substance P-and most CGRP-immunoreactive fibres enter the ganglion via central branches and, to a large extent, traverse it, but some CGRP-immunoreactive influx appears to come from the intestine. The origin of intraganglionic substance P-and CGRP-immunoreactive fibres after ganglion transplantation remained unidentified. Somatostatin-and neuropeptide Y-immunoreactive fibres predominantly have an intraganglionic origin as have DBH-immunoreactive noradrenergic fibres. The demonstrated alterations in neuropeptide immunoreactivity of intraganglionic and periganglionic nerve fibres following the applied transsection procedures contribute to the present knowledge on origin and destination of peptidergic transmitter segments in the guinea pig inferior mesenteric ganglion. Moreover, the present study provides evidence that intrinsic participitation in intraganglionic fibre supply is more extensive than hitherto believed.Dedicated to Professor Dr. T.H. Zusammenfassung In der Einleitung ist das Ziel der Arbeit in den wesentlichsten Punkten herausgestellt.Die Bursastrukturen (Bursavelum und Rippen bzw. Papillen) der parasitischen Strongylina lassen sich von den entsprechenden Bildungen der freilebenden Rhabditina, vor allem der Gattung Rhabditis, ableiten und in ihren Einzelgliedern homologisieren.Die im Laufe der Phylogenie bei den Strongylina auftretenden strukturellen Transformationen lassen sich auf einige wenige, relativ einfache morphogenetische Grundvorgänge zurückführen, die da sind: Wachstumsallometrien, Materialkompensationen, Organverschmelzungen und Spaltungen (Fissationen), Rudimentationen und ähnliche Vorgänge.Innerhalb der Strongylina Bursa ist ein Gefälle der Wachstumsgradienten feststellbar, das sich vom Zentrum der Bursa sowohl nach distal als auch proximalwärts abschwdcht. Zunehmende Förderung der zentral gelegenen Organe (Rippen) führt zu entsprechender Reduktion der peripheren Bursastrukturen, was vor allem im terminalen Schwanzabschnitt auffällt und zur Ausbildung des oft nur noch als Rudiment vorhandenen Dorsalrippenkomplexes führt. Letzterer entspricht in seiner Gesamtheit der Schwanzspitze der peloderen Rhabditiden mit den Papillen 9 und 10.Die bei Rhabditis moist getrennten Papillen 7 und 8 sind bei allen Strongylina zu einer Rippe (Externodorsal-Rippe) verschmolzen, die jedoch in manchen Aberrationen durch Abspaltung eines akzessorischen Astes ihre wahre Natur (als Verschmelzungsprodukt) zu erkennen gibt (Atavismus).Da dieselben Transformationsvorgänge innerhalb der Strongylina mehrfach unabhängig voneinander wirksam geworden sind, treten bestimmte Strukturformen als Parallelbildungen in verschiedenen phylogenetischen Union auf (polytope Entstehung).Zahlreich untersuchte Bildungsabweichungen (Aberrationen), deren Bedeutung für die Morphologie kurz umrissen wird, erschöpfen sich in den gleichen strukturellen Transformationstypen, die auch bei der Evolution der verschiedenen Union der Strongylina nachweisbar sind. Die Aberrationen führen daher häufig zu Atavismen oder zu Parallelvariationen (homologe Variationen").Die Zahl der Umwandlungsmbglichkeiten (Potenzen) der Bursastrukturen innerhalb der Strongylina ist beschränkt (Paripotenz im Sinne Haeckers). Bestimmte Arten (und Entwicklungshnien) haben jeweils nur bestimmte Potenzen realisiert. Andere können jedoch latent (virtuell) im Kryptotypus vorhanden sein, ohne normalerweise in Erscheinung. zu treten. In bestimmten Aberrationen können sie jedoch plötzlich realisiert werden, so ihr latentes Vorhandensein demonstrierend (Pluripotenz).Wie lange bestimmte Potenzen in einer Gruppe erhalten bleiben konnen, verdeutlichen auch die Schwanzhocker weiblicher Nematoden, als zum Bauplan der Nematoden gehbrende Bildungen. Die Potenz zur Ausbildung dieser Strukturen kommt offensichtlich sehr vielen Nematoden-Arten zu, wird jedoch nur in relativ wenigen Fällen, aber innerhalb der verschiedenen Gruppen bald hier, bald dort (disjunkte Verbreitung), realisiert. Es handelt sich bei den Schwanzhöckern um rudimentäre Organe, die bei keiner Nematoden-Art mehr voll ausgebildet erhalten sind. Ihre Rudimentation beruht zum Teil auf Materialentzug, als Folge von Unkonstruktionen der Schwanzregion, wobei die Adultstadien zuerst betroffen werden (Aphanisie nach Sewertzoff).Bei den in Chiropteren parasitierenden Strongylacanthinae haben sich Schwanzhöcker noch bei allen Arten erhalten, was ein offensichtlich archaisches Merkmal darstellt. Bei anderen Nematoden, denen sie nur im Larvalstadium zukommen, treten sie wohl durch Fötalisation in seltenen Fällen auch bei den adulten Stadien wieder auf.Alle speziellen Bursaformen der Strongylina lassen sich durch relativ wenige und einfache Transformationsvorgänge aus einem durch Abstraktion gewonnenen diagrammatischen Typus ableiten (Prinzip der variablen Proportionen" nach Troll).Die typisierten Umwandlungsvorgänge decken sich weitgehend mit den von Remane allgemein gefaßten strukturellen Typen der Realmutationen. Da sie bei den beobachteten Aberrationen, deren Entstehung auf dem Wege über Realmutationen sehr wahrscheinlich ist, in homologer Weise auftreten, kann das innerhalb der Strongylina zu beobachtende Evolutionsphänomen auf Realmutationen zurückgeführt warden.Obwohl sich die untersuchten strukturellen Transformationen in dem systematisch relativ wait gefaßten Rahmen einer Unterordnung abspielen (transspezifische Evolution nach Rensch), handelt es sich bei der von uns bevorzugten Terminologie (nach Woltereck und Remane), unter Berücksichtigung des Charakters der Umwandlungen, doch nur um Vorgänge, die in den Bereich der Mikroevolution fallen.  相似文献

Zusammenfassung In der Meerschweinchenzirbeldrüse lassen sich elektronenmikroskopisch helle und dunkle Pinealzellen sowie einzelne Gliazellen nachweisen. In den bei weitem überwiegenden hellen Pinealzellen zeichnet sich ein Teil der vesicle-crowned rodlets (VCR) durch lokale Auftreibungen aus. Von VCR deutlich abzugrenzen sind die vesicle-crowned balls (VCB). Erstmalig beschrieben wird das Vorkommen von sog. Zylindern, die als Vorstufen von VCB aufgefaßt werden. In den relativ seltenen dunklen Pinealzellen, die sich durch chromatinreiche Kerne und elektronendichtes Zytoplasma auszeichnen, sind Vesikel, VCR, VCB und Zylinder seltener als in hellen Pinealzellen. Die reichlich vorhandenen marklosen Nervenfasern finden sich vor allem in perivasculären Räumen, seltener im Parenchym. Synapsen zwischen Nerven und Pinealzellen wurden nicht beobachtet. In den Zirbeldrüsen trächtiger Meerschweinchen zeichnen sich in der 2. Hälfte der Tragzeit die hellen Pinealzellen durch stärkere Lappung der Kerne, gehäuftes Auftreten von laktiven Zonen, Vermehrung von Mitochondrien, glattem ER, agranulären Vesikeln, VCR, VCB und Zylindern aus. Die dunklen Pinealzellen nehmen während der Tragzeit an Zahl zu. Post partum bilden sich diese Veränderungen innerhalb einer Woche zurück. Längerer Aufenthalt der Tiere in Dunkelheit führt zu einer Aktivierung der hellen Pinealzellen mit auffallender Vermehrung der VCR und zu einer Zunahme der dunklen Zellen. Unter Dauerbelichtung kommt es in den hellen Zellen zu einer Abnahme fast aller Zellorganellen und zu einer starken Vermehrung der VCR, die nach 70 Tagen auch Formveränderungen aufweisen. Nach Reserpinbehandlung beobachtet man eine Verminderung und degenerative Veränderungen der VCR. Es wird diskutiert, daß die VCR als prae- bzw. postsynaptische Strukturen der Erregungsübertragung von Nerven zu Pinealzellen bzw. von Pinealzellen untereinander dienen könnten.
The fine structure of the pineal gland of normal, pregnant and experimentally affected guinea-pigs
Summary By means of electron microscopy light and dark pinealocytes can be distinguished in the guinea-pig pineal gland. Glial cells are rare. In the light pinealocyte. the most frequent cell type, some vesicle-crowned rodlets (VCR) show circumscribed thickenings. From these structures vesicle-crowned balls (VCB) have to be clearly distinguished. Furthermore cylinders occur, which, it is suggested, are precursors of VCB. Dark pinealocytes characterized by chromatin-rich nuclei and electron-dense cytoplasm are rare and contain fewer vesicles, VCR, VCB and cylinders than light pinealocytes. Numerous non-myelinated nerve fibres are situated within perivascular spaces, a few also in the parenchyma. Synapses between nerve fibres and pinealocytes were not observed. In the pineal gland of pregnant guinea-pigs the following changes can be observed in the second half of gestation. The light cells show many nuclear indentations and an increase of active zones, mitochondria, smooth ER, agranular vesicles, VCR, VCB, and cylinders respectively. The dark cells increase in number. After birth these changes reverse to normal within one week. Constant darkness leads to an activation of the light cells accompanied by an increase of the VCR and to an increase in number of the dark cells. Under constant illumination the light cells show a decrease of their organelles and a strong increase of the VCR. After 70 days the VCR also show a change in shape. Following reserpine treatment the VCR decrease in number and show signs of degeneration. It is discussed that the VCR function as pre- or postsynaptic structures and that they are involved either in transmitting impulses from nerve fibres to pinealocytes or from one pinealocyte to the other.

Electrical stimulation of the periaqueductal gray matter (PGM) bringing about inhibition of high-threshold mouth-opening reflex on somatosensory response of neurons belonging to the pontine caudal reticular nucleus (CN) and the reticular gigantocellular nucleus (GN) were investigated in cats under light chloralose-induced anesthesia. It was found that inhibitory effects on CN and GN neurons adhered to the principle of control over their main synaptic input: in GN neurons and particularly in high-threshold cells, inhibition mainly affected response to nociceptive stimuli, in contrast to CN neurons, and low-threshold cells in particular, where response to non-nociceptive stimulation was inhibited. A defined set of oligosaccharides and glycopeptides containing -linked fucose were used to examine the specificity of the immobilized fucose-binding lectin Lotus tetragonolobus agglutinin (LTA1), also known as lotus lectin. Glycans containing the Lewis x determinant (Lex) Gal1-4[Fuc1-3]GlcNAc1-3-R were significantly retarded in elution from high density LTA-Emphaze columns. The lectin also bound the fucosylated lacdiNAc trisaccharide GalNAc1-4[Fuc1-3]GlcNAc. The lectin did not bind glycans containing either sialylLex or VIM-2 determinants, nor did it bind the isomeric Lea, Gal1-3[Fuc1-4]GlcNAc-R. Although 2-fucosyllactose Fuc1-2Gal1-4Glc) was retarded in elution from the columns, larger glycans containing the H-antigen Fuc1-2Gal1-3(4)GlcNAc-R interacted poorly with immobilized LTA. Our results demonstrate that immobilized LTA is effective in isolating glycans containing the Lex antigen and is useful in analyzing specific fucosylation of glycoconjugates.
