Cactoblastis cactorum's unintended arrival to Florida and its expansion in North America represent a threat to Opuntia-based agriculture and natural ecosystems in the United States and Mexico. Apanteles opuntiarum attacks C. cactorum and is a potential biocontrol agent due to its specificity, wide distribution and occurrence. Laboratory rearing methods using excised cladodes for C. cactorum as host larvae of A. opuntiarum were developed, but require a continuous supply of cactus with a risk of microorganisms compromising the rearing. Host cues —including odour of host metabolic subproducts like faeces and chemicals emitted by the attacked plant— are the most important signals that help a parasitoid locate a host. Little attention has been paid to behavioural differences of parasitoids in the presence of hosts reared on artificial diet. Thus, the aims of the present work were to determine the effect of meridic diet for C. cactorum on parasitoid behaviour and to determine whether prior experience (previous exposure to stimuli) influences the response of A. opuntiarum towards hosts. Parasitism rates were assessed using cladodes or meridic diet as larvae food source. Behavioural experiments also evaluated the effect of prior experience (larvae or frass from larvae fed on cactus or diet) on host searching, encounter and attack of different stimulus (larvae or frass from larvae fed on cactus or diet). Parasitism behaviour of A. opuntiarum was negatively affected by the use of meridic diet to feed host larvae. Presenting parasitoids with larvae rather than their frass influenced excitatory behavioural responses in terms of contact, probing and stinging in A. opuntiarum females, whereas the effect of prior experience on the behaviours was not quite consistent. For laboratory mass-rearing procedures of A. opuntiarum, we recommend previous contact of females with frass from cactus-fed larvae and a piece of cactus, which showed enhanced parasitism rates.  相似文献

Sensory organs that detect CO2 are common in herbivorous moths and butterflies, but their function has been unclear until now. As the CO2 gradients in the vicinity of a host plant depend on its physiological condition, CO2 could provide a sensory cue for the suitability of the plant as a larval food source. This study investigated whether changing the atmospheric CO2 concentration affected oviposition by Cactoblastis cactorum on its host, the cactus Opuntia stricta. On host plants exposed to rapid fluctuations in CO2 concentration, the frequency of oviposition was reduced by a factor of 3.2 compared to the control. As the fluctuations mask the much smaller CO2 signals generated by the plants, this suggests that those signals constitute an important component of the host identification process. On host plants exposed to a constant background of doubled CO2, oviposition was also reduced, by a factor of 1.8. An increased background reduces host signal detectability, partially as a consequence of a general principle of sensory physiology (Weber-Fechner's law), and partially due to other factors specific to CO2-receptor neurons. The moth Cactoblastis cactorum (Berg), the poster child of weed biological control in Australia, has recently invaded the United States and threatens native cacti. Concern is greatest for the endangered semaphore cactus, Opuntia corallicola, of which only two known populations exist in the wild. We made three separate outplantings of O. corallicola, designed to bolster the number of extant cacti and to test the effectiveness of three different treatments to protect the cacti from Cactoblastis. In one outplanting, we tested the associational susceptibility hypothesis and found that cacti planted more than 20 m away from the common prickly pear cactus, Opuntia stricta, which act as a reservoir of Cactoblastis, were just as frequently attacked and killed by Cactoblastis as cacti planted within 5 m. In addition, Cactoblastis attack was greater in the shade than in the sun. In the second outplanting, we minimized the attack from Cactoblastis by using protective cages planted at least 500 m from O. stricta in areas not inhabited by cacti. Cages attracted the attention of local animals, which destroyed the cages and trampled the cacti inside to death. Crown rot caused high mortality in this outplanting. In the third outplanting, again conducted at least 500 m away from O. stricta, fertilization did not reduce crown rot mortality. We suggest that increasing populations of O. corallicola in Florida, by means of outplantings, will remain a challenge because of death from Cactoblastis when planted in areas where cacti normally grow and because of death from crown rot in areas where they do not. Because Cactoblastis is moving rapidly northward and westward and has already reached Charleston, South Carolina, rare cacti in the rest of the U.S. Southeast may be in danger. Eventually, many cactus species in the U.S. South, Southwest, and Mexico will likely be threatened by this moth.  相似文献

The morphological sensillum types on the antennae of male and female Cactoblastis cactorum were visualized by scanning electron microscopy. Electrophysiological recordings were performed for the first time on single olfactory sensilla of C. cactorum. The male sensilla trichodea house a receptor cell responding to the putative pheromone component (9Z,12E)-tetradecadienyl acetate. The sensilla trichodea of the females were much shorter than those of the males and contained specialized receptor cells responding to certain terpenoids, the most frequent being the nerolidol-sensitive cell. The sensilla auricillica and sensilla basiconica of both sexes contained cells responding less specifically to terpenoid compounds as well as to green leaf volatiles. Cells of the sensilla coeloconica responded to aliphatic aldehydes and acids. Eight volatile organic compounds emitted by Opuntia stricta, a host plant of C. cactorum, were identified using gas chromatography-mass spectrometry, beta-caryophyllene being the major compound. Five compounds identified by gas chromatography in the headspace of O. stricta elicited responses in olfactory receptor cells of C. cactorum, nonanal being the most active compound and therefore a candidate attractant of C. cactorum.  相似文献

The interaction between the moth, Cactoblastis cactorum, and the cactus, Opuntia stricta, is used as a model to examine the question of whether the CO2 sense of a herbivorous insect can detect the CO2 gradients associated with a plant's metabolic activity. Both the anatomical and the electrophysiological characteristics of CO2-sensitive receptor neurons in C. cactorum indicate an adaptation to the detection of small fluctuations around the atmospheric background. Evidence is provided that further rises in background will impair the function of the sensory organ. In the habitat of the plant, during the diurnal window of the moth's activity, two types of CO2 gradients occur that are detectable by the moth's sensors. The first gradient, associated with soil respiration, is vertical and extends from the soil surface to an altitude of approximately 1 m. Its magnitude is well above the detectability limit of the sensors. The notion that this gradient provides, to a flying insect, a cue for the maintenance of a flight altitude favourable for host detection is supported by field observations of behaviour. The second gradient, associated with CO2 fixation by the plant, extends from the surfaces of photosynthetic organs (cladodes) over a boundary layer distance of approximately 5 mm. Again, its magnitude is well above the detectability limit. The notion that this gradient provides, to a walking insect, a cue to the physiological condition of the plant is supported by the observation that females of C. cactorum, prior to oviposition, actively probe the plant surface with their CO2 sensors. In a simulation of probing, pronounced responses of the sensors to the CO2-fixing capacity of O. stricta are observed. We propose that by probing the boundary layer, females of C. cactorum can detect the healthiest, most active O. stricta cladodes, accounting for earlier observations that the most vigorous plants attract the greatest density of egg sticks.  相似文献

The unintentional introduction of the cactus moth, Cactoblastis cactorum, a successful biological control agent formerly employed in the control of invasive prickly pear cactus species (Opuntia spp.) in Australia, Hawaii, South Africa, and various Caribbean islands, has posed great concern as to the possible threat to native, endangered species of cactus in the southeastern USA as well as with the potential to cause a major infestation of commercial and agricultural cactus crops in Mexico. A number of control measures have been investigated with varying degrees of success including, field exploration for cactus moth-specific parasitoids, insecticides, fungal, bacterial, and nematode agents. Current tactics used by the USA-Mexico binational program to eradicate cactus moth from Mexico and mitigate its westward movement in the USA include host plant removal, the manual removal and destruction of egg sticks and infected cacti stems, and the Sterile Insect Technique. One other approach not taken until now is the development of a cactus moth cell line as a tool to facilitate the investigation of baculoviruses as an alternative biocontrol method for the cactus moth. Consequently, we established C. cactorum cell lines derived from adult ovarian tissue designated as BCIRL-Cc-AM and BCIRL-Cc-JG. The mean cell population doubling time was 204.3 and 112 h for BCIRL-Cc-AM and BCIRL-Cc-JG, respectively, with weekly medium change, while the doubling time was 176.6 and 192.6 h for BCIRL-Cc-AM and BCIRL-Cc-JG, respectively, with a daily change of medium. In addition, the daily versus weekly change in medium was reflected in the percentage viability with both cell lines showing higher levels with a daily medium change. Of the three baculoviruses tested, only the recombinant AcMNPV-hsp70Red and GmMNPV at a multiplicity of infection (MOI) of 1.0 were able to demonstrate significant production of extracellular virus (ECV) in each of the cell lines, whereas both cell lines were refractive to an HzSNPV challenge at an MOI of 10. In this study, we have demonstrated both the successful development of a C. cactorum cell line and its ability to support a complete baculovirus infection. The potential is also there to pursue further investigations to determine the susceptibility of the cactus moth cell line to other viruses. Additionally, the availability of a cactus moth cell line will facilitate the analysis of viruses prior to using the more expensive bioassay test. Finally, it is hoped with the knowledge presented here that baculoviruses may also be considered as an alternative biocontrol method for the cactus moth.  相似文献

Abstract. The cactus moth, Cactoblastis cactorum (Berg) (Phycitidae) is native to South America. It was released as a biological control agent against alien Opuntia- cacti in Australia in the 1920s, then in southern Africa, and latterly on several islands, including those in the Caribbean. In 1989, the cactus moth was discovered in Florida, in the United States of America, where it is now threatening the survival of indigenous Opuntia species. In this paper we identify some of the attributes that have contributed to the success of C. cactorum as a weed biological control agent. Many of these same qualities account for the problems that C. cactorum has caused in Florida and predispose it as a major threat to the speciose, native Opuntia- floras of Central and North America. An estimated 79 platyopuntia (prickly pear) species are at risk: 51 species endemic to Mexico; nine species endemic to the United States; and 19 species common to both countries. Many cultivated and wild Opuntia species, that are used in various ways, are also vulnerable to attack by C. cactorum , including at least 25 species in Mexico and three species in the United States, particularly the widely exploited and culturally important cultivars of O. ficus-indica . Some control strategies are suggested that may minimize the risk and consequences of invasion by the cactus moth. The wider implications of this threat to the practice of weed biological control and to conservation are discussed.  相似文献

Recent severe hurricanes in the Caribbean and south-east United States have had devastating socio-economic effects, and there is a pressing need to learn how animals are impacted by such events. We serendipitously deployed a multi-channel data logger onto a hawksbill turtle (Eretmochelys imbricata) during the breeding season in 1998 and logged various aspects of her behaviour before, during and after passage of hurricane Georges. As Georges passed by, the turtle made shorter dives, became more active and spent less time at the surface between dives compared with its baseline, non-storm activity. However, after passage of the hurricane the turtle quickly resumed its pre-hurricane behaviour and nested successfully a few days later. These results show that, in this case, the hurricane had a minor impact on the submerged animal presumably because of the dampening effect of depth on high winds over water.  相似文献

A 10-year-old boy with juvenile metachromatic leukodystrophy (MLD) presented with the 459 + 1G→A arylsulfatase A (ASA) mutation on one allele. To detect his complete genotype, the other ASA allele was sequenced and a T-to-C transition at nucleotide 376 in exon 2 was identified. This missense mutation results in a substitution of leucine 76 by proline. Of 20 MLD unrelated controls, 18 carried the L/P76 mutation either in the homozygous (n = 6) or heterozygous (n = 12) state. The presence or absence of L/P76 did not influence leukocyte ASA activity or urinary sulfatide excretion. Apparently, the substitution of leucine 76 by proline is a common ASA polymorphism, neither being related to MLD nor creating ASA pseudodeficiency. However, because of its frequency and location, L/P76 may be of particular importance in genetic studies requiring the differentiation of the ASA alleles within a kindred. The immunoglobulin heavy-chain genes are located in a cluster on chromosome 14. The simultaneous absence of the human IgG1, IgG2, IgG4, and IgA1 subclasses was previously reported in a healthy Tunisian Berber and was later shown to be due to a multigene deletion. We now describe a serological and molecular study of a different deletion observed in a healthy Tunisian. Blot hybridization analysis of the proband's DNA using gamma, epsilon, alpha, and mu switch probes showed that the deletion involves a large region of the immunoglobulin heavy-chain gene cluster: C psi epsilon, C alpha 1, C psi gamma, C gamma 2, and C gamma 4. Incidentally, we showed that the restriction enzyme EcoRI alone can be used with the alpha probe to differentiate A2m types. The deletion described, present in a person homozygous for GM-Am haplotypes (Gm1,17;..;5,14,11,13,10 A2m2), is consistent with previous location, by association analysis, of C psi gamma between C alpha 1 and C gamma 2. There is evidence to suggest that deletions may be more common in the Mediterranean region than in North American Caucasians.  相似文献

The productivity and biomass of pristine coral reef ecosystems is poorly understood, particularly in the Caribbean where communities have been impacted by overfishing and multiple other stressors over centuries. Using historical data on the spatial distribution and abundance of the extinct Caribbean monk seal (Monachus tropicalis), this study reconstructs the population size, structure and ecological role of this once common predator within coral reef communities, and provides evidence that historical reefs supported biomasses of fishes and invertebrates up to six times greater than those found on typical modern Caribbean reefs. An estimated 233,000-338,000 monk seals were distributed among 13 colonies across the Caribbean. The biomass of reef fishes and invertebrates required to support historical seal populations was 732-1018 gm(-2) of reefs, which exceeds that found on any Caribbean reef today and is comparable with those measured in remote Pacific reefs. Quantitative estimates of historically dense monk seal colonies and their consumption rates on pristine reefs provide concrete data on the magnitude of decline in animal biomass on Caribbean coral reefs. Realistic reconstruction of these past ecosystems is critical to understanding the profound and long-lasting effect of human hunting on the functioning of coral reef ecosystems.  相似文献

Adventitious shoot formation was studied with leaf, stem and axillary bud explants of carnation (Dianthus caryophyllus L.). The shoot regeneration procedures were applicable for a wide range of cultivars and shoot regeneration percentages were high for all explant types. Using axillary bud explants, shoot regeneration efficiency was independent of the size of the bud and of its original position in the plant. In contrast, shoot regeneration from stem and leaf explants was strongly dependent on their original position on the plant. The most distal explants (just below the apex) showed the highest level of shoot regeneration. The adventitious shoot primordia developed at the periphery of the stem segment and at the base of leaf explants. In axillary bud, stem and leaf explants, shoot regeneration originated from node cells, located at the transition area between leaf and stem tissue. Neisseria meningitidis is a human-specific pathogen with capacity to cause septic shock and meningitis. It has been hypothesized that invasion of the central nervous system (CNS) is a complication of a bacteremic condition. In this study, we aimed to characterize the invasion route of N. meningitidis to the CNS. Using an intranasally challenged mouse disease model, we found that twenty percent of the mice developed lethal meningitis even though no bacteria could be detected in blood. Upon bacterial infection, epithelial lesions and redistribution of intracellular junction protein N-cadherin were observed at the nasal epithelial mucosa, especially at the olfactory epithelium, which is functionally and anatomically connected to the CNS. Bacteria were detected in the submucosa of the olfactory epithelium, along olfactory nerves in the cribriform plate, at the olfactory bulb and subsequently at the meninges and subarachnoid space. Furthermore, our data suggest that a threshold level of bacteremia is required for the development of meningococcal sepsis. Taken together, N. meningitidis is able to pass directly from nasopharynx to meninges through the olfactory nerve system. This study enhances our understanding how N. meningitidis invades the meninges. The nasal olfactory nerve system may be a novel target for disease prevention that can improve outcome and survival.  相似文献

Most research on the impacts of plant invasion focuses on native plant performance, community structure, and ecosystem functioning. Some non-native species can also pose a risk to human health. One such risk is the allergenic nature of the pollen of some introduced plants. We examined whether patterns of airborne pollen differed between non-native and native taxa by summarizing data from seven Spanish Mediterranean localities monitored over 13 yr. The pollen spectra contained 27 native pollen taxa and 18 non-native taxa. Even though pollen from native taxa were more diverse and were present longer in the atmosphere than the non-native, in some years neither the prevalence of the two nor their weekly maximum pollen values differed significantly. However, maximum values for non-native taxa were found earlier in the season than for native pollen. A small percentage of non-native pollen includes pollen from introduced taxa that have not invaded natural habitats (e.g., ornamental plants). Non-native pollen has a larger proportion of allergenic pollen than native pollen. Therefore, the results reveal that the presence of non-native airborne pollen from naturalized and non-naturalized plant species increases the total amount of airborne allergenic pollen grains and the period of allergenic susceptibility.  相似文献

Ecologists are often required to estimate the number of species in a region or designated area. A number of diversity indices are available for this purpose and are based on sampling the area using quadrats or other means, and estimating the total number of species from these samples. In this paper, a novel theory and method for estimating the number of species is developed. The theory involves the use of the Laplace method for approximating asymptotic integrals. The method is shown to be successful by testing random simulated datasets. In addition, several real survey datasets are tested, including forests that contain a large number (tens to hundreds) of tree species, and an aquatic system with a large number of fish species. The method is shown to give accurate results, and in almost all cases found to be superior to existing tools for estimating diversity.  相似文献

European round gobies (Neogobius melanostomus) are displacing several important native North American fish species. Controlling their invasion is contingent on understanding their swimming inclination and potential. We assessed goby swimming inclination by recording activity in a 2 m flume over a ~24 h period, and swimming potential using a critical swimming (U crit) test, as well as burst tests in still and flowing water. When given the choice to move, gobies covered as much as 14 m/h, with a slight bias towards nocturnal activity and an overall upstream preference. When confined and coerced to perform a U crit test, they burst-and-held to achieve 35.5 ± 1.1 cm/s. Thirty minutes following U crit, they were able to burst-and-coast in a sprint test to almost twice this speed. In still water, they exhibited startle bursts of up to 163 cm/s. We provide a swimming endurance model that indicates flow rates would need to be >125 cm/s to prevent upstream movement, and free of refuge areas in which to recover. The current study shows that the round goby is a surprisingly powerful swimmer with the capacity to continue its invasion should hydrologic control be absent.  相似文献

Activation of blood coagulation factor V (FV) is a key reaction of hemostasis. FV circulates in plasma as an inactive procofactor, and proteolytic removal of a large central B-domain converts it to an active cofactor (FVa) for factor Xa (FXa). Here we show that two short evolutionary conserved segments of the B-domain, together termed the procofactor regulatory region, serve an essential autoinhibitory function. This newly identified motif consists of a basic (963-1008) and an acidic (1493-1537) region and defines the minimal sequence requirements to maintain FV as a procofactor. Our data suggest that dismantling this autoinhibitory region via deletion or proteolysis is the driving force to unveil a high affinity binding site(s) for FXa. These findings document an unexpected sequence-specific role for the B-domain by negatively regulating FV function and preventing activity of the procofactor. These new mechanistic insights point to new ways in which the FV procofactor to cofactor transition could be modulated to alter hemostasis.  相似文献
