The effects of carbon, nitrogen, phosphate, and copper on cell growth and production of the isoflavone puerarin by suspension cultures of Pueraria tuberosa (Roxb. ex. Willd.) DC were investigated. Among the various sugars evaluated (glucose, galactose, fructose, maltose, and sucrose), use of sucrose in the medium led to the maximum accumulation of puerarin. A sucrose-feeding strategy in which additional sucrose was added to the flasks 15?d into the culture cycle stimulated both cell biomass and puerarin production. The maximum production of puerarin was obtained when a concentration balance of 20:60?mM NH 4 + /NO 3 ? was used as the nitrogen source. Alteration in the concentration balance of nitrogen components (NH 4 + /NO 3 ? 60:20?mM) or the use of either NH 4 + or NO 3 ? alone decreased biomass production and puerarin accumulation compared with the control culture (NH 4 + /NO 3 ? 20:20?mM). High amounts of phosphate (2.5 and 5?mM) in the medium inhibited puerarin production whereas 0.625?mM phosphate promoted puerarin production (68.3???g/g DW on day?25). An increase in Cu2+ concentration from 0.025 to 0.05?mg/l in the P. tuberosa cell culture medium resulted in a 2.2-fold increase in puerarin production (up to 141???g/g DW on day?25) but reduced cell culture biomass.  相似文献

Withania somnifera is an important medicinal plant that contains withanolides as bioactive compounds. We have investigated the effects of macroelements and nitrogen source in hairy roots of W. somnifera with the aim of optimizing the production of biomass and withanolide A content. The effects of the macroelements NH4NO3, KNO3, CaCl2, MgSO4 and KH2PO4 at concentrations of 0, 0.5, 1.0, 1.5 and 2.0× strengths and of nitrogen source [NH4 +/NO3 ? (0.00/18.80, 7.19/18.80, 14.38/18.80, 21.57/18.80, 28.75/18.80, 14.38/0.00, 14.38/9.40, 14.38/18.80, 14.38/28.20 and 14.38/37.60 mM)] in Murashige and Skoog medium were evaluated for biomass and withanolide A production. The highest accumulation of biomass (139.42 g l?1 FW and 13.11 g l?1 DW) was recorded in the medium with 2.0× concentration of KH2PO4, and the highest production of withanolide A was recorded with 2.0× KNO3 (15.27 mg g?1 DW). The NH4 +/NO3 ? ratio also influenced root growth and withanolide A production, with both parameters being larger when the NO3 ? concentration was higher than that of NH4 +. Maximum biomass growth (148.17 g l?1 FW and 14.79 g l?1 DW) was achieved at NH4 +/NO3 ? ratio of 14.38/37.60 mM, while withanolide A production was greatest (14.68 mg g?1 DW) when the NH4 +/NO3 ? ratio was 0.00/18.80 mM. The results are useful for the large scale cultivation of Withania hairy root culture for the production of withanolide A.  相似文献

The optimal culture medium for the production of flavonoid compounds from Orostachys cartilaginea V. N. Boriss. calluses was studied. In callus cultures of O. cartilaginea, the flavonoid monomer content, in decreasing order was kaempferol-3-O-rutinoside (Kp-3-rut), quercetin 3-O-glucoside (Qc-3-glc), epicatechin gallate (Ecg), kaempferide (Ke), and quercetin (Qc). The results of the uniform design experiment indicated that the production of Qc, Ke, Qc-3-glc, Kp-3-rut, and total flavonoids were satisfactory in callus grown on full salt strength (1×) of Murashige and Skoog (MS) medium supplemented with 3.5 mg L?1 6-benzylaminopurine (BA) and 0.1 mg L?1 1-naphthalene acetic acid (NAA). By contrast, only Ecg was found in callus grown on 0.75× MS medium supplemented with 1.5 mg L?1 BA and 0.3 mg L?1 NAA. A phosphate concentration of 1.25 mM in the MS medium favored the production of Qc and Ke, whereas 0.75 mM phosphate was optimal for the production of Ecg, Qc-3-glc, Kp-3-rup, and total flavonoids. The NH4 +/NO3 ? ratios of 30/30 mM in the MS medium promoted Ke, Ecg, Qc-3-glc, Kp-3-rup, and total flavonoid production. However, a NH4 +/NO3 ? ratio of 20/40 mM enhanced Qc production. The effect of sucrose concentrations on the accumulation of different flavonoid monomers was comparatively more regular. The flavonoid content increased as the sucrose concentration increased from 20 to 40 g L?1, peaked at 40 g L?1, and decreased at concentrations greater than 40 g L?1. Therefore, 40 g L?1 sucrose was optimal for the production of the five flavonoid monomers and total flavonoids. The present findings demonstrate the possibility of producing flavonoid compounds from O. cartilaginea callus.  相似文献

The present work deals with optimization of adventitious shoot culture of Bacopa monnieri for the production of biomass and bacoside A and has investigated the effects of macro elements (NH4NO3, KNO3, CaCl2, MgSO4 and KH2PO4) and nitrogen source [NH4 +/NO3 ] of Murashige and Skoog (Physiol Plant 15:473–497, 1962) medium (MS) on accumulation of biomass and bacoside A content. Optimum number of adventitious shoots (99.33 shoots explant−1), fresh weight (1.841 g) and dry weight (0.150 g) were obtained in the medium with 2.0× strength of NH4NO3. The highest production of bacoside A content was also recorded in the medium of 2.0× NH4NO3, which produced 17.935 mg g−1 DW. The number of adventitious shoot biomass and bacoside A content were optimum when the NO3 concentration was higher than that of NH4 +. Maximum number of shoots (70.00 shoots explant−1), biomass (fresh weight 1.137 g and dry weight 0.080 g) and also bacoside A content (27.106 mg g−1 DW) were obtained at NH4 +/NO3 ratio of 14.38/37.60 mM. Overall, MS medium supplemented with 2.0× NH4NO3 is recommended for most efficient bacoside A production.  相似文献

Withania somnifera is an important medicinal plant that contains withanolides and withaferins, both bioactive compounds. The effects of the macroelements NH4NO3, KNO3, CaCl2, MgSO4 and KH2PO4 at concentrations of 0.0, 0.5, 1.0, 1.5 and 2.0× strength and of the nitrogen source [NH4 +/NO3 (mM/mM) ratio of: 0.00/18.80, 7.19/18.80, 14.38/18.80, 21.57/18.80, 28.75/18.80, 14.38/0.00, 14.38/9.40, 14.38/18.80, 14.38/28.20, and 14.38/37.60 (mM)] in Murashige and Skoog medium were tested for biomass and withanolide A production. The highest accumulation of biomass [147.81 g l−1 fresh weight (FW) and 14.02 g l−1 (dry weight (DW)] was recorded in the medium containing a 0.5× concentration of NH4NO3, and the highest production of withanolide A content was recorded in the medium with 2.0× KNO3 (4.36 mg g−1 DW). The NH4 +/NO3 ratio also influenced cell growth and withanolide A production, with both parameters being larger when the NO3 concentration was higher than that of NH4 +. Maximum biomass growth (110.45 g l−1 FW and 9.29 g l−1 DW) was achieved at an NH4 +/NO3 ratio of 7.19/18.80, while withanolide A production was greatest (3.96 mg g−1 DW) when the NH4 +/NO3 ratio was 14.38/37.60 mM.  相似文献

An in vitro system was established for the characterisation of inorganic nitrogen uptake by sugarcane plantlets of variety NCo376. After multiplication and rooting, plantlets (0.27–0.3 g fresh mass) were placed on N-free medium for 4 days, and then supplied with 2–20 mM N as NO3 ?-N only, NH4 +-N only or NO3 ?-N + NH4 +-N (as 1:1). With few exceptions, on all the tested N media, the in vitro plants always had a higher Vmax for NH4 +-N (28.69–66.51 μmol g?1 h?1) than for NO3 ?-N uptake (10.24–30.19 μmol g?1 h?1) and the Km indicated a higher affinity for NO3 ?-N (0.02–7.38 mM) than for NH4 +-N (0.06–9.15 mM). When N was applied as 4 and 20 mM to varieties N12, N19 and N36, the interaction between variety, N form and concentration resulted in differences in the Vmax and Km. The high N-use efficient varieties (N12 and N19), as determined in previous pot and field trials, behaved similarly under all tested conditions and displayed a lower Vmax and Km than the low N-use efficient ones (NCo376 and N36). Based on this finding, it was suggested that the N-use efficient designation (from pot and field trials) may not be ascribed solely to N uptake. Assessment of the relative preference index (RPI) for NO3 ?-N and NH4 +-N uptake revealed that, at present, the RPI has no application in sugarcane due to its preferential uptake of NH4 +-N.  相似文献

An efficient and reliable transformation system for a very important medicinal plant Papaver bracteatum was developed through optimization of several factors that affect the rate of effective A. rhizogenes-mediated transformation and growth rate of hairy root. Five bacterial strains, A4, ATCC15834, LBA9402, MSU440 and A13, and three explants types, hypocotyls, leaves and excised shoots were examined. The highest frequency of transformation was achieved using LBA9402 strain in the excised shoots. Several inoculation and co-cultivation media and different concentration of arginine were evaluated using LBA9402 strain and the excised shoots as explant. Interestingly, a drastic increase in the frequency of transformation (47.3 %) was observed when Murashige and Skoog medium containing 1 mM arginine and lacking NH4NO3 KH2PO4, KNO3 and CaCl2 was used. The effect of sucrose concentration and the ratio of NH4 +: NO3 ? on hairy root biomass was examined. Maximum biomass was obtained in 30 g/l sucrose and 20:10 mM ratio of NH4 + to NO3 ? on MS medium. Transgenic hairy root lines were confirmed by polymerase chain reaction (PCR) and Southern hybridization.  相似文献

A protocol has been standardized for establishment and characterization of cell suspension cultures of Stevia rebaudiana in shake flasks, as a strategy to obtain an in vitro stevioside producing cell line. The effect of growth regulators, inoculum density and various concentrations of macro salts have been analyzed, to optimize the biomass growth. Dynamics of stevioside production has been investigated with culture growth in liquid suspensions. The callus used for this purpose was obtained from leaves of 15-day-old in vitro propagated plantlets, on MS medium fortified with benzyl aminopurine (8.9 μM) and naphthalene acetic acid (10.7 μM). The optimal conditions for biomass growth in suspension cultures were found to be 10 g l?1 of inoculum density on fresh weight basis in full strength MS liquid basal medium of initial pH 5.8, augmented with 2,4-dichlorophenoxy acetic acid (0.27 μM), benzyl aminopurine (0.27 μM) and ascorbic acid (0.06 μM), 1.0× NH4NO3 (24.7 mM), 3.0× KNO3 (56.4 mM), 3.0× MgSO4 (4.5 mM) and 3.0× KH2PO4 (3.75 mM), in 150 ml Erlenmeyer flask with 50 ml media and incubated in dark at 110 rpm. The growth kinetics of the cell suspension culture has shown a maximum specific cell growth rate of 3.26 day?1, doubling time of 26.35 h and cell viability of 75 %, respectively. Stevioside content in cell suspension was high during exponential growth phase and decreased subsequently at the stationary phase. The results of present study are useful to scale-up process and augment the S. rebaudiana biological research.  相似文献

We investigated the effects of medium salt strength and ammonia/nitrate ratio on biomass production and metabolites accumulation of adventitious root. The medium with full-strength Murashige and Skoog (MS) reached the highest growth rate (16.77), and the contents of saponin and polysaccharide reached the peak (i.e., 0.65 and 24.85 %) at 3/4 MS and 1 MS, respectively. In case of ammonia/nitrate ratio, a NH4 +/NO3 ? ratio of 20:40 was optimal for the production of biomass and polysaccharide (23.27 %). In contrast, the content of saponin achieved the optimum (0.74 %) at a NH4 +/NO3 ? ratio of 30:30. In 5-L balloon-type bubble bioreactor (BTBB) cultures, an approximately 23-fold increase in biomass was recorded. The fresh weight (FW) and dry weight (DW) were 72.78 and 6.79 g per bioreactor with the contents of saponin (0.62 %) and polysaccharide (17.32 %), respectively. It indicated potential application to produce adventitious roots of pseudostellaria heterophylla with bioreactors on large scale in commercial.  相似文献

Nitrogen is a major driver of plant growth and the nitrogen source can be critical to good growth in vitro. A response surface methodology mixture-component design and a data mining algorithm were applied to nitrogen (N) nutrition for improving the micropropagation of Prunus armeniaca Lam. Data taken on shoot cultures included a subjective quality rating, shoot number, shoot length, leaf characteristics and physiological disorders. Data were analyzed using the Classification and Regression Tree data mining algorithm. The best overall shoot quality as well as leaf color were on medium with NO3??>?25 mM and NH4+/Ca+ >?0.8. Improving shoot length to15 mm required 25?<?NO3? ≤?35 mM with NH4+/Ca2+ ≤?2.33. The most shoots (11.6) were produced with NO3? >?25 mM and NH4+/Ca2+ ≤ 0.8, but there were 5–10 shoots at other NO3? concentrations regardless of NH4+/Ca2+ proportion. Leaves increased in size with higher NO3? concentrations (>?55 mM). Physiological disorders were also influenced by the nitrogen components. Shoot tip necrosis was rarely present with NO3? > 45 mM. Callus production decreased somewhat with NH4+/Ca2+ >?2.33. Suggested concentrations for an improved medium considering all of these growth characteristics would be 25?<?NO3? ≤?35 mM and NH4+/Ca+ ≤ 0.8. Validation experiments comparing WPM and three trial media showed improvements in several shoot growth parameters on medium with optimized mesos and optimized nitrogen components.  相似文献

We have established cell-suspension cultures of mountain ginseng (Panax ginseng G A. Mayer), and have attempted to increase the yield of saponin by manipulating our processing method and culturing factors (e.g., media strengths; the presence of plant growth regulators or sucrose; ratios of NO+ 3/ NH- 4). Maximum biomass yield was obtained in media containing 2,4-D. However, saponin productivity was much higher in a medium comprising either IBA or NAA; 7.0 mg/L IBA was optimal for promoting both cell growth (10.0 g/L dry weight) and saponin production (7.29 mg/g DW total ginsenoside). Although the addition of cytokinins (BA and kinetin) did not affect cell growth, the level of saponin (particularly in the Rb group) was enhanced when the media were supplemented with either 0.5 mg/L BA or 0.5 mg/L kinetin. Half- and full-strength MS media were equally suitable for inducing both biomass as well as saponin production. We also investigated the effect of various concentrations of sucrose and nitrogen, and found that 30 g/L sucrose enhanced biomass yield as well as saponin content However, further increases (i.e., up to 70 g/L) led to a decrease in saponin accumulation and biomass production. Maximum growth and saponin productivity were reported from treatments with an initial nitrogen concentration of 30 mM. In general, the amount of saponin increased when the test media had high NO+ 3/ NH- 4 ratios; in fact, saponin production was greatest when nitrate was the sole nitrogen source.  相似文献

An effective protocol was developed for in vitro propagation of Psoralea corylifolia via somatic embryogenesis in cell suspension culture. Embryogenic callus was obtained on Murashige and Skoog (MS) medium supplemented with 6 μM naphthaleneacetic acid (NAA) and 30 μM glutamine from transverse TCLs from 10-day-old hypocotyl explants with a 96.4% frequency. Embryogenic callus produced a higher number of somatic embryos (123.7 ± 1.24 per gram fresh weight callus) on MS medium containing 30 g l?1 sucrose, 1 μM NAA, 4 μM benzyladenine (BA), 15 μM glutamine and 2 μM abscisic acid (ABA) after 4 weeks of culture. Somatic embryos successfully germinated (97.6%) on ½ MS medium containing 20 g l?1 sucrose, 8 g l?1 agar and supplemented with 2 μM BA, 1 μM ABA and 2 μM gibberellic acid (GA3) within 2 weeks of culture. Somatic embryos developed into normal plants, which hardened with 100% efficiency in soil in a growth chamber. Plants were successfully transferred to greenhouse and subsequently established in the field. Plant survival percentage in the field differed with seasonal variations. Average psoralen content of 12.9 μg g?1 DW was measured in different stages of somatic embryo development by high-performance liquid chromatography (HPLC). This protocol will be helpful for efficient propagation of elite clones on a mass scale, conservation efforts of this species and for secondary metabolites production studies.  相似文献

Cotyledon explants of Panax ginseng produced somatic embryos directly on solid hormone-free MS medium containing 3% (w/v) sucrose while high concentration of NH4NO3 (60 mM) induced embryogenic callus. Ten subcultures of the embryogenic callus on hormone-free MS medium with 40 mM NH4NO3 gave hormone-independent proliferation of callus, which exhibited proliferation potential even on MS medium with a standard level of NH4NO3 (20 mM). Pulse treatment of callus with exogenous auxin or cytokinin (1.0 mg 1–1 2,4-D, 1.0 mg 1–1 kinetin) resulted in the loss of the hormone-independent characteristic and caused the callus to brown. For the suspension culture, embryogenic callus was transferred to MS liquid medium containing 3% (w/v) sucrose in an 500 ml Erlenmyer flask. Embryogenic cell clumps in full-strength MS liquid medium discharged toxic substances, resulting in strong suppression of cell growth. In 1/3-strength MS medium, exudation of toxic material did not occur. Embryogenic cell clumps were mass-grown on a large-scale in a bioreactor (20-1), showing a 7.1 increase of fresh weight in 1/3-strength MS medium with 3% (w/ v) sucrose after 5 weeks of culture. Total ginsenoside content of cultured embryogenic cell clumps was low and 6 times below naturally-cultivated ginseng roots.  相似文献

In an attempt to scale-up of adventitious root cultures of Echinacea angustifolia for the production of biomass and caffeic acid derivatives, i.e. echinacoside, chlorogenic acid, cichoric acid, caftaric acid, and cynarin, the effects of Murashige and Skoog (MS) medium dilutions, and initial sucrose concentrations were investigated in a 5-L airlift bioreactor. In addition, the kinetics of adventitious root growth and accumulation of secondary metabolites were also studied. The greatest root dry weight (6.50 g L?l) and accumulation of total phenolics [22.06 mg g?1 DW (dry weight)], total flavonoids (5.77 mg g?1 DW) and total caffeic acid derivatives (10.63 mg g?1 DW) were obtained at quarter-strength MS medium. Of the various gradients of sucrose tested, 5 % sucrose supplementation was regarded as an optimal concentration for enhancing productivity of biomass and bioactive compounds. Neither higher salt strength (3/4–2 MS) nor sucrose concentrations (7 and 9 %) showed promotive effect on root growth and metabolite production. The kinetic studies revealed that 4 weeks of culture period is the optimal time to achieve highest productivity of metabolites. Based on these results, a large-scale (20 L) and a pilot-scale (500 L) adventitious root culture system was established. In the pilot-scale bioreactor, adventitious roots were elicitor-treated with 100 μM methyl jasmonate (MJ) on day 28. After 1 week of elicitation, 1.75 kg dry root biomass was harvested containing 60.41 mg g?1 DW of total phenolics, 16.45 mg g?1 DW of total flavonoids, and 33.44 mg g?1 DW of total caffeic acid derivatives. Among the caffeic acid derivatives, the accumulation of echinacoside (the major bioactive compound) in MJ-treated adventitious roots grown in the 500-L bioreactor was the highest (12.3 mg g?1 DW), which is approximately threefold more than the non-MJ-treated roots cultured in 5- and 20-L bioreactors.  相似文献

Excess of ammonia generates oxidative and osmotic stress, and results in an accumulation of compatible solutes. The aim of this study was to investigate the physiological significance of excess ammonium-induced proline and sucrose accumulation on antioxidative activity and osmotic adjustment. The detached leaves of white clover (Trifolium repense L.) were fed with 0, 10, 50, 100, and 200 mM NH4Cl, and the contribution of proline and sucrose to osmotic adjustment and their relationship with antioxidative enzymes activity were assessed. A gradual decline of relative water content and osmotic potential (Ψπ) with increasing NH4Cl feeding level was accompanied by an increase in ammonia concentration. Significant accumulation of proline and sucrose was observed when NH4Cl was fed over 100 mM compared with control (0 mM NH4Cl). The increase in enzyme activity was significant only at 200 mM for ascorbate peroxidase (APOD) and over 100 mM NH4Cl for guaiacol peroxidase (GPOD) and catalase (CAT). The contribution of proline and sucrose to osmotic adjustment over 100 mM, where proline and sucrose accumulation was more important, maintained at control levels or significantly decreased. The content of proline and sucrose as affected by NH4Cl feeding level was positively related with the activity of APOD, GPOD, and CAT. These results suggest that proline and sucrose accumulation induced by the excess of ammonium has a more influential role in antioxidative activity rather than osmotic adjustment.  相似文献

We investigated the effects on ginseng adventitious root growth and ginsenoside production when macro-element concentrations and nitrogen source were manipulated in the culture media. Biomass growth was greatest in the medium supplemented with 0.5-strength NH4PO3, whereas ginsenoside accumulation was highest (9.90 mg g-1 DW) in the absence of NH4PO3. At levels of 1.0-strength KNO3, root growth was maximum, but a 2.0 strength of KNO3 led to the greatest ginsenoside content (9.85 mg g-l). High concentrations of MgSO4 were most favorable for both root growth and ginsenoside accumulation (up to 8.89 mg g-1 DW). Root growth and ginsenoside content also increased in proportion to the concentration of CaCI2 in the medium, with the greatest accumulation of ginsenoside (8.91 mg g-1 DW) occurring at a 2.0 strength. The NH4/NO3 -- ratio also influenced adventitious root growth and ginsenoside production; both parameters were greater when the NO3 - concentration was higher than that of NH4 +. Maximum root growth was achieved at an NH4 +/NO3 - ratio of 7.19/18.50, while ginsenoside production was greatest (83.37 mg L-1) when NO3 - was used as the sole N source.  相似文献

The influence of cytokinins and culture conditions including medium volume, harvest time and elicitation with abiotic elicitors (SA/MeJ) have been studied for the optimal production of biomass and withanolides in the multiple shoot culture of Withania somnifera. Elicitation of shoot inoculum mass (2 g l?l FW) with SA at 100 μM in the presence of 0.6 mg l?l BA and 20 mg l?l spermidine for 4 h exposure time at the 4th week in 20 ml liquid medium recorded higher withanolides production (withanolides A [8.48 mg g?l DW], withanolides B [15.47 mg g?l DW], withaferin A [29.55 mg g?l DW] and withanone [23.44 mg g?l DW]), which were 1.14 to 1.18-fold higher than elicitation with MeJ at 100 μM after 5 weeks of culture. SA-elicited cultures did not exhibit much variation in biomass accumulation when compared to control. This cytokinin induces and SA-elicited multiple shoot culture protocol provides a potential alternative for the optimal production of biomass and withanolides utilizing liquid culture.  相似文献

Embryogenic avocado cultures derived from ‘Hass’ protoplasts were genetically transformed with the plant defensin gene (pdf1.2) driven by the CaMV 35S promoter in pGPTV with uidA as a reporter gene and bar, the gene for resistance to phosphinothricin, the active ingredient of the herbicide Finale® (Basta) (Bayer Environmental Science, Research Triangle Park, Durham, NC ). Transformation was mediated by Agrobacterium tumefaciens strain EHA105. Transformed cultures were selected in the presence of 3.0 mg l?1 phosphinothricin in liquid maintenance medium for 3–4 mo. Liquid maintenance medium consisted of modified MS medium containing (per liter) 12 mg NH4NO3 and 30.3 mg KNO3 and supplemented with 0.1 mg l?1 thiamine HCl, 100 mg l?1 myo-inositol, 30 g l?1 sucrose, 3.0 mg l?1 phosphinothricin, and 0.41 μM picloram. Somatic embryo development from transformed cultures was initiated on MS medium supplemented with 45 g l?1 sucrose, 4 mg l?1 thiamine HCl, 100 mg l?1 myo-inositol, 10% (v/v) filter-sterilized coconut water, 3.0 mg l?1 phosphinothricin, and 6.0 g l?1 gellan gum. Limited plant recovery occurred from somatic embryos on semi-solid MS medium supplemented with 3.0 mg l?1 phosphinothricin, 4.44 μM 6-benzylaminopurine (BA), and 2.89 μM GA3; transformed shoots were micrografted on in vitro-grown seedling rootstocks. Approximately 1 yr after acclimatization in the greenhouse, transformed shoots were air-layered to recover transformed roots. In this study, we investigated the induction of Pseudostellaria heterophylla adventitious root and the effects of sucrose concentration and phosphate source on biomass increase and metabolites accumulation. These roots were initially cultured in Murashige and Skoog medium for 4 weeks. IBA 3.0 mg L?1 proved to be the best auxin for inducing adventitious roots and the frequency of adventitious roots induced from roots (100 %) was higher than that from leaves (78 %) and stems (27 %). The medium with 4 % sucrose resulted in the optimum biomass i.e. 1.04 g/flask DW, and the content of saponin and polysaccharides reached the peak i.e. 0.676 and 24.4 %, respectively. With regards to phosphate source, 1.25 mM phosphate concentration was more favorable for biomass of roots (0.87 g/flask of DW), whereas the optimum saponin (0.74 %) and polysaccharides (22.09 %) were achieved with 2.5 mM phosphate. However, the saponin content at 2.5 mM phosphate did not show significant difference from the saponin content at 0.625 mM (0.69 %) or 3.75 mM phosphate (0.69 %).  相似文献
