Airlift bioreactors were programmed for continuous and temporary immersion culture to investigate factors that affect the rhizome proliferation, shoot formation, and plantlet regeneration of Cymbidium sinense. During rhizome proliferation, the continuous immersion bioreactor system was used to explore the effects of activated charcoal (AC) in the culture medium, inoculation density, and air volume on rhizome differentiation and growth. The optimum conditions for obtaining massive health rhizomes were 0.3 g l?1 AC in the culture medium, 7.5 g l?1 inoculation density, and 150 ml min?1 air. In addition, the temporary immersion bioreactor system was used for both shoot formation and plantlet regeneration. Supplementing 4 mg l?1 6-benzylaminopurine and 0.2 mg l?1 naphthalene acetic acid (NAA) to the culture medium promoted shoot induction from the rhizome. Cutting the rhizome explants into 1 cm segments was better for massive shoot formation than cutting into 0.25 and 0.5 cm explant segments. NAA promoted plantlet regeneration and the rooting rate (94.7 %), with whole plantlets growing well in culture medium containing 1.0 mg l?1 NAA. Therefore, applying bioreactors in C. sinense micropropagation is an efficient way for scaling up the production of propagules and whole plantlets for the industrial production of high-quality seedlings.  相似文献

The study assessed the influence of sugar concentration (10, 20, 30, 50, 70, 100, 120 g l?1) on growth and ginsenoside biosynthesis in Panax quinquefolium hairy roots cultivated in shake flasks and a nutrient sprinkle bioreactor. The highest growth rate was achieved in medium containing 3–5 % sucrose. More than 70 g l?1 or less than 20 g l?1 sugar content in the medium induces significant inhibition of root growth when cultivated in shake flasks. The saponin content was determined using HPLC. The maximum yield (above 9 mg g?1 d.w.) of the sum of six examined ginsenosides (Rb1, Rb2, Rc, Rd, Re and Rg1) in hairy roots cultivated in shake flasks was obtained with 30 g l?1 sucrose in the medium. The sucrose concentration in the medium was found to correlate with saponin content in bioreactor-cultured specimens. A higher level of protopanaxadiol derivatives was found for lower (20 and 30 g l?1) sucrose concentrations; higher sucrose concentrations (50 and 70 g l?1) in the medium stimulated a higher level of Rg group saponins.  相似文献

Caralluma tuberculata (C. tuberculata) is a very important medicinal plant with a range of anti-diabetic and weight reduction properties. This high-valued medicinal plant is nowadays considered as endangered due to its unsustainable elimination from wild habitats. There is lack of research efforts on its propagation to overcome escalating demand. In this research study, an effort has been made to optimize protocol for large-scale mass propagation and production of natural antioxidants. Highest callogenic response (87.2 %) was observed from shoot tip explants on Murashige and Skoog (MS) medium containing 30 g l?1 sucrose and combination of 2, 4-D (2.0 mg l?1) and BA (1.0 mg l?1). During shoot morphogenesis, 50 g l?1 sucrose along with BA (2.0 mg l?1) and GA3 (1.0 mg l?1) enhanced shoot regeneration (91.3 %), mean shoot length (2.6 cm) and shoots per explant (24.5) as compared to control. The combination of IBA and IAA (2.0 mg l?1) was found optimum for root induction (74.98 %), mean root length (4.1 cm) and roots per shoot (6.9) as compared to control. The plantlets were successfully acclimatized in plastic cups and various tissues were investigated for accumulation of antioxidant secondary metabolites including phenolics, flavonoids, stress enzymes and antioxidant activities. The superoxide dismutase enzyme was higher in shoots; protein content was higher in callus cultures; phenolics, DPPH and protease activity were higher in plantlets, while flavonoids, peroxidase, reducing power and total antioxidant activities were higher in wild plants. This simple protocol is very useful for commercial production of consistent plantlets and metabolites of interest.  相似文献

Isomaltulose is a structural isomer of sucrose commercially used in food industries. In this work, recombinant Escherichia coli producing sucrose isomerase (SIase) was used to convert sucrose into isomaltulose. To develop an economical industrial medium, untreated cane molasses (10.63 g l?1), yeast extract (25.93 g l?1), and corn steep liquor (10.45 g l?1) were used as main culture compositions for SIase production. The relatively high SIase activity (14.50 ± 0.11 U mg DCW?1) was obtained by the recombinant cells. To the best of our knowledge, this is the first investigation on SIase production by engineered E. coli using untreated cane molasses. The recombinant E. coli cells expressing the SIase gene were immobilized in calcium alginate gel in order to improve the efficiency of recycling. The immobilization was most effective with 2 % (w/v) sodium alginate and 3 % (w/v) calcium chloride. The optimal initial biomass for immobilization was 20 % (w/v, wet wt.), with a hardening time of 8 h for cell immobilization. The immobilized E. coli cells exhibited good stability for 30 batches with the productivity of 0.45 g isomaltulose g pellet?1 h?1. A continuous isomaltulose formation process using a column reactor remained stable for 40 days with 83 ± 2 % isomaltulose yield, which would be beneficial for economical production of isomaltulose.  相似文献

The persistent edaphic stress on microbial succession due to dynamic changes during composting was explored for selection of multi-stress tolerant microbe(s) desirable for ethanol production. A total of 23 strains were isolated from mango compost using four successive enrichments in YP broth (g l?1): glucose, 100; 150; 250 with ethanol (40) and cycloheximide (0.4) at 40 °C, pH 6.0. Based on multi-gene ribotyping, 14 yeasts (61 %) of Saccharomycetaceae, 2 filamentous fungi (8.6 %) and 7 bacteria (30.4 %) were obtained. Phenetic and phylogenetic analysis of the 14 yeasts revealed 64.3 % tolerant to 500 g l?1 glucose, growth at 45 °C and resemblance to Candida sp. (14.3 %), Kluyveromyces marxianus (35.7 %), Pichia kudriavzevii (21.4 %) and Saccharomyces cerevisiae (28.6 %). Assessment of the 14 yeasts in glucose fermentation medium (pH 4.5 at 40 °C) showed ethanol productivity of ≥92 % by 12 yeasts with theoretical yields of 90–97 %. Fermentation of molasses (150 g l?1 glucose equivalent) by P. kudriavzevii D1C at 40 °C resulted in 73.70 ± 0.02 g l?1 ethanol and productivity of 4.91 ± 0.01 g l?1 h?1. Assessment of P. kudriavzevii D1C revealed multi-stress tolerance towards 5-hydroxymethyl furfural, ethanol (20 %, v/v), high gravity and H2O2 (0.3 M) indicating suitability for ethanol production using high gravity molasses and pre-treated lignocellulosic biomass fermentation.  相似文献

Among several fatty acids tested, oleic acid was selected as the most efficient inducer for the production of 4-hydroxydodecanoic acid, a metabolite of β-oxidation, by Waltomyces lipofer. Cells were induced by incubation for 12 h in a medium containing 10 g l?1 yeast extract, 10 g l?1 peptone, 5 g l?1 oleic acid, 1 g l?1 glucose, and 0.05 % (w/v) Tween 80. The optimal reaction conditions for the production of γ-lactones by induced cells were pH 6.5, 35 °C, 200 rpm, 0.71 M Tris, 60 g l?1 hydroxy fatty acid, and 20 g l?1 cells. Non-induced cells produced 38 g l?1 γ-dodecalactone from 60 g l?1 10-hydroxystearic acid after 30 h, with a conversion yield of 63 % (w/w) and a productivity of 1.3 g l?1 h?1 under the optimized conditions, whereas induced cells produced 51 g l?1 γ-dodecalactone from 60 g l?1 10-hydroxystearic acid after 30 h, with a conversion yield of 85 % (w/w) and a productivity of 1.7 g l?1 h?1. The conversion yield and productivity of induced cells were 22 % and 1.3-fold higher, respectively, than those of non-induced cells. Induced cells also produced 28 g l?1 γ-decalactone and 12 g l?1 γ-butyrolactone from 60 g l?1 12-hydroxystearic acid and 60 g l?1 10-hydroxydecanoic acid, respectively, after 30 h. The concentration, conversion yield, and productivity of γ-dodecalactone and γ-decalactone are the highest reported thus far. This is the first study on the biotechnological production of γ-butyrolactone.  相似文献

We developed an efficient and simple system for inducing somatic embryogenesis and regenerating plantlets from petal explant of Chrysanthemum (Chrysanthemum morifolium) cv. ‘Baeksun’. Somatic embryogenesis was induced from petal explants on the Murashige and Skoog (MS) medium supplemented with 1.0 mg l?1 2,4-dichlorophenoxyacetic acid (2,4-D) and 3.0 mg l?1 6-benzyladenine (BA), yielding the highest mean number of embryos (56.3) per explant after 5 weeks of culture. We evaluated the effects of basal medium and various concentrations of sucrose on the proliferation of secondary somatic embryos. MS medium was observed to be more effective in promoting the proliferation of somatic embryos than half-strength Murashige and Skoog (1/2MS). In addition, 1 % sucrose was also found to be the best in induction of secondary embryogenesis. The highest germination rate (70 %) of the somatic embryos was observed on the MS medium containing 0.2 mg l?1 α-naphthalene acetic acid and 1 g l?1 activated charcoal (AC). Shoots elongated rapidly and roots developed well on hormone-free MS medium with 1 g l?1 AC and successfully acclimated in the greenhouse. Flow cytometric analysis of the primary somatic embryos, secondary somatic embryos, and the somatic embryo-obtained plants along with the parent grown in the greenhouse showed that they all had same identical peaks, indicating that there was no variation of ploidy level during the regeneration process. We expect that our report would be useful for micropropagation and Agrobacterium-mediated genetic transformation studies of this cultivar.  相似文献

In vitro plantlets of sugarcane cultivar NCo310 were maintained in slow growth conditions at both 18 and 24°C and on four semi-solid media: SG1—Murashige and Skoog (MS) salts and vitamins with 20 g L?1 sucrose, SG2—½ MS with 10 g L?1 sucrose, SG3—MS with 20 g L?1 sucrose and 1 mg L?1 abscisic acid (ABA), and SG4—½ MS with 10 g L?1 sucrose and 1 mg L?1 ABA. After 8, 12, 24, 36, and 48 mo shoot multiplication rates were recorded, shoots were removed from storage and subcultured every 2 wk on SG1 with 0.015 mg L?1 kinetin and 0.1 mg L?1 benzyl aminopurine for 2 mo. At 18°C, all media supported storage for 48 mo with subculturing every 12 mo. Shoot multiplication post-retrieval was significantly higher on the SG2 medium compared with the non-stored control (362 ± 84 and 126 ± 26 shoots per recovered shoot after 2 mo, respectively). In addition, shoots could be maintained for 48 mo on SG2 medium with one subculture without compromising post-storage multiplication ability. At 24°C, storage on all four media supported recovery and multiplication of shoots for 8 mo and only SG2 medium facilitated survival for 12 mo. There was no advantage to incorporating ABA into the storage media, regardless of the temperature and storage time. Cryopreservation of cultivar NCo376 in vitro-derived shoot meristems using the V-cryo-plate method demonstrated that the sucrose concentration in the loading solution (0.8–1.8 M) had no significant effect on survival of the meristems, which ranged from 41.7 ± 4.8 to 69.4 ± 10%.  相似文献

A new xylose fermenting yeast was isolated from over-ripe banana by enrichment in xylose-containing medium. The phylogenetic analysis of ITS1-5.8S-ITS2 region sequences of ribosomal RNA of isolate BY2 revealed that it shows affiliation to genus Pichia and clades with Pichia caribbica. In batch fermentation, Pichia strain BY2 fermented xylose, producing 15 g l?1 ethanol from 30 g l?1 xylose under shaking conditions at 28°C, with ethanol yield of 0.5 g g?1 and volumetric productivity of 0.31 g l?1 h?1. The optimum pH range for ethanol production from xylose by Pichia strain BY2 was 5–7. Pichia strain BY2 also produced 6.08 g l?1 ethanol from 30 g l?1 arabinose. Pichia strain BY2 can utilize sugarcane bagasse hemicellulose acid hydrolysate for alcohol production, efficiency of fermentation was improved by neutralization, and sequential use of activated charcoal adsorption method. Percent total sugar utilized and ethanol yield for the untreated hydrolysate was 17.14% w/v and 0.33 g g?1, respectively, compared with 66.79% w/v and 0.45 g g?1, respectively, for treated hemicellulose acid hydrolysate. This new yeast isolate showed ethanol yield of 0.45 g g?1 and volumetric productivity of 0.33 g l?1 h?1 from sugarcane bagasse hemicellulose hydrolysate detoxified by neutralization and activated charcoal treatment, and has potential application in practical process of ethanol production from lignocellulosic hydrolysate.  相似文献

Asymbiotic seed germination of Coelogyne flaccida varied with the capsule stage and the culture medium used for germinating seeds. The capsules were harvested at two different stages of development. The seeds were cultured on three asymbiotic orchid seed germination defined and undefined media, i.e. Mitra (M) medium, Murashige and Skoog (MS) medium and potato dextrose agar (PDA) medium. The seeds obtained from undehisced green capsules germinated with a maximum germination percentage (84.50 ± 0.33%) on M medium followed by MS and PDA medium. The effect of cytokinins, such as 6-benzylaminopurine and furfurylaminopurine and the synthetic auxin α-naphthalene acetic acid, on seed germination was also assessed. Simultaneously, in vitro multiplication using protocorms as explants was also studied. The effect of organic growth supplements, such as banana homogenate (BH, 25, 50, 75 g l? 1) and peptone (P, 1.0, 1.5, 2.0 g l? 1), was tested on the de novo formation of protocorm-like bodies (PLBs), development of the maximum number of shoots and early formation of plantlets using the M medium. Among the treatments, the highest regeneration frequency (87.50 ± 0.20%) and the highest number of PLBs per explant (10.25 ± 0.50) were obtained in P (1.5 g l? 1)-supplemented cultures, and the plantlets were formed within 18 weeks of culture. BH favoured the development of healthy plantlets, with a maximum fresh weight of 1.02 ± 0.04 g per plantlet.  相似文献

γ-Aminobutyric acid (GABA), a non-protein amino acid, is a bioactive component in the food, feed and pharmaceutical fields. To establish an effective single-step production system for GABA, a recombinant Corynebacterium glutamicum strain co-expressing two glutamate decarboxylase (GAD) genes (gadB1 and gadB2) derived from Lactobacillus brevis Lb85 was constructed. Compared with the GABA production of the gadB1 or gadB2 single-expressing strains, GABA production by the gadB1gadB2 co-expressing strain increased more than twofold. By optimising urea supplementation, the total production of l-glutamate and GABA increased from 22.57 ± 1.24 to 30.18 ± 1.33 g L?1, and GABA production increased from 4.02 ± 0.95 to 18.66 ± 2.11 g L?1 after 84-h cultivation. Under optimal urea supplementation, l-glutamate continued to be consumed, GABA continued to accumulate after 36 h of fermentation, and the pH level fluctuated. GABA production increased to a maximum level of 27.13 ± 0.54 g L?1 after 120-h flask cultivation and 26.32 g L?1 after 60-h fed-batch fermentation. The conversion ratio of l-glutamate to GABA reached 0.60–0.74 mol mol?1. By co-expressing gadB1 and gadB2 and optimising the urea addition method, C. glutamicum was genetically improved for de novo biosynthesis of GABA from its own accumulated l-glutamate.  相似文献

Mutants of Candida magnoliae NCIM 3470 were generated by treatment of ultra-violet radiations, ethyl methyl sulphonate and N-methyl-N′-nitro-N-nitrosoguanidine. Mutants with higher reductase activity were screened by means of 2,3,5-triphenyl tetrazolium chloride agar plate assay. Among the screened mutants, the mutant R9 produced maximum mannitol (i.e. 46 g l?1) in liquid fermentation medium containing 250 g l?1 glucose and hence was selected for further experiments. Preliminary optimization studies were carried out on shake-flask level which increased the mannitol production to 60 g l?1 in liquid fermentation medium containing 300 g l?1 glucose. A two-stage fermentation process comprising of growth phase and production phase was employed. During the growth phase, glucose was supplemented and aerobic conditions were maintained. Thereafter, the production phase was initiated by supplementing fructose and switching to anaerobic conditions by discontinuing aeration and decreasing the speed of agitation. The strategy of two-stage fermentation significantly enhanced the production of mannitol up to 240 g l?1, which is the highest among all fermentative production processes and corresponds to 81 % yield and 4 g l?1 h?1 productivity without formation of any by-product.  相似文献

An effective protocol was developed for in vitro propagation of Psoralea corylifolia via somatic embryogenesis in cell suspension culture. Embryogenic callus was obtained on Murashige and Skoog (MS) medium supplemented with 6 μM naphthaleneacetic acid (NAA) and 30 μM glutamine from transverse TCLs from 10-day-old hypocotyl explants with a 96.4% frequency. Embryogenic callus produced a higher number of somatic embryos (123.7 ± 1.24 per gram fresh weight callus) on MS medium containing 30 g l?1 sucrose, 1 μM NAA, 4 μM benzyladenine (BA), 15 μM glutamine and 2 μM abscisic acid (ABA) after 4 weeks of culture. Somatic embryos successfully germinated (97.6%) on ½ MS medium containing 20 g l?1 sucrose, 8 g l?1 agar and supplemented with 2 μM BA, 1 μM ABA and 2 μM gibberellic acid (GA3) within 2 weeks of culture. Somatic embryos developed into normal plants, which hardened with 100% efficiency in soil in a growth chamber. Plants were successfully transferred to greenhouse and subsequently established in the field. Plant survival percentage in the field differed with seasonal variations. Average psoralen content of 12.9 μg g?1 DW was measured in different stages of somatic embryo development by high-performance liquid chromatography (HPLC). This protocol will be helpful for efficient propagation of elite clones on a mass scale, conservation efforts of this species and for secondary metabolites production studies.  相似文献

Manipulation of culture strategies was adopted to study the influence of nutrient stress, pH stress and precursor feeding on the biosynthesis of capsaicin in suspension and immobilized cell cultures of C. chinense. Cells cultured in the absence of one of the four nutrients (ammonium and potassium nitrate for nitrate and potassium stress, potassium dihydrogen orthophosphate for phosphorus stress, and sucrose for sugar stress) influenced the accumulation of capsaicin. Among the stress factors studied, nitrate stress showed maximal capsaicin production on day 20 (505.9 ± 2.8 μg g?1 f.wt) in immobilized cell, whereas in suspension cultures the maximum accumulation (345.5 ± 2.9 μg g?1 f.wt) was obtained on day 10. Different pH affected capsaicin accumulation; enhanced accumulation of capsaicin (261.6 ± 3.4 μg g?1 f.wt) was observed in suspension cultures at pH 6 on day 15, whereas in case of immobilized cultures the highest capsaicin content (433.3 ± 3.3 μg g?1 f.wt) was obtained at pH 5 on day 10. Addition of capsaicin precursors and intermediates significantly enhanced the biosynthesis of capsaicin, incorporation of vanillin at 100 μM in both suspension and immobilized cell cultures resulted in maximum capsaicin content with 499.1 ± 5.5 μg g?1 f.wt on day 20 and 1,315.3 ± 10 μg g?1 f.wt on day 10, respectively. Among the different culture strategies adopted to enhance capsaicin biosynthesis in cell cultures of C. chinense, cells fed with vanillin resulted in the maximum capsaicin accumulation. The rate of capsaicin production was significantly higher in immobilized cells as compared to freely suspended cells.  相似文献

To assess the potential of different genotypes of Brazilian oil palm (Elaeis guineensis Jacq.) to somatic embryogenesis and somatic embryo proliferation, mature zygotic embryos of nine commercial genotypes of E. guineensis (BRSC2001, BRSC2328, BRSC2301, BRSC3701, BRSCM1115, BRSC7201, BRSC2528, BRSC2501, and BRSCN1637) were used. Explants were incubated on Murashige and Skoog (MS) supplemented with 450 μM picloram, 3.0 % sucrose, 500 mg l?1 glutamine, and 2.5 g l?1 activated charcoal, and gelled with 2.5 g l?1 Phytagel. After induction, for differentiation and maturation, the embryogenic calli (ECs) were transferred into fresh medium supplemented with 0.6 μM naphthaleneacetic acid (NAA) and 12.30 μM 2-isopentenyladenine (2iP) or 40 μM picloram in combination with 0.3 g l?1 activated charcoal, and 500 mg l?1 glutamine. Somatic embryos were converted into plants on MS medium with macro- and micro-nutrients at half strength, 2 % sucrose, and 2.5 g l?1 activated charcoal, and gelled with 2.5 g l?1 Phytagel. In general, zygotic embryos swelled after 14 days. Primary calli, which were observed in all the genotypes after 45–60 days of culture, eventually progressed to ECs at 90 days. At this time, scanning electron microscopy (SEM) analysis showed cellular differences between compact and friable calli. After 150 days in the induction phase, the ECs with proembryos that were transferred to the medium for differentiation and maturation, differentiated asynchronically into somatic embryos at globular and torpedo stages. The results showed that BRSC2328 and BRSCM1115 had the highest potential for EC formation (90–100 %) and somatic embryo differentiation (40.7 and 52.5 somatic embryos per callus, respectively) when compared to other genotypes. After approximately 90 days of culture on MS basal medium without growth regulators, protrusion of the leaf primordia was observed, characterizing the onset of germination of the somatic embryos into plants.  相似文献

Biomass and lipid productivities of Isochrysis galbana were optimized using nutrients of molasses (4, 8, 12 g l?1), glucose (4, 8, 12 g l?1), glycerol (4, 8, 12 g l?1) and yeast extract (2 g l?1). Combinations of carbon sources at different ratios were evaluated in which the alga was grown at three different light intensities (50, 100 and 150 μmol m?2 s?1) under the influence of three different photoperiod cycles (12/12, 18/6 and 24/0 h light/dark). A maximum cell density of 8.35 g l?1 with 32 % (w/w) lipid was achieved for mixotrophic growth at 100 μmol m?2 s?1 and 18/6 h light/dark with molasses/glucose (20:80 w/w). Mixotrophic cultivation using molasses, glucose and glycerol was thus effective for the cultivation of I. galbana.  相似文献

An efficient and simple system for inducing somatic embryogenesis and regenerating plantlets from immature zygotic embryos of açaí palm (Euterpe oleracea) has been developed. Embryogenic calli (ECs) were induced from immature zygotic embryos of açaí palm on Murashige and Skoog (MS) modified medium with 2,4-dichlorophenoxyacetic acid and picloram. Embryogenic frequency was dependent on auxin type and concentration. The optimal concentration of picloram for the high-frequency induction of embryogenic calli (72%) was 225 μM. ECs were then subcultured on a differentiation and maturation medium composed of MS modified medium with 2-isopentenyladenine and naphthaleneacetic acid with subcultures at 4-week intervals. SEs were converted to plants on MS modified medium with half-strength macro- and micronutrients, 20 g l?1 sucrose, and 2.5 g l?1 activated charcoal and gelled with 2.5 g l?1 Phytagel. Detailed morpho anatomical changes during the different stages of somatic embryogenesis were characterized. The development of SEs was asynchronous, and ontogenic studies confirmed that the initial cell divisions occur in the epidermal and subepidermal regions of the zygotic embryos. Broad base attachment of SEs to the epidermis indicates the presence of a suspensor.  相似文献

The effect of organic and inorganic nitrogen sources on Trichoderma reesei Rut-C30 cellulase production was investigated in submerged cultivations. Stirred tank bioreactors and shake flasks, with and without pH control, respectively, were employed. The experimental design involved the addition of individual organic nitrogen sources (soy peptone, glutamate, glycine and alanine) within a basal medium containing Avicel (i.e. micro crystalline cellulose) and ammonium sulphate. It was found that in the shake flask experiments, the highest cellulase activities (~0.1 ± 0.02 FPU ml?1) were obtained with media containing soy peptone (3–6 g l?1) and glutamate (3.6 g l?1). However, these improvements in the cellulase titers in the presence of the organic nitrogen sources appeared to be related to smaller changes in the pH of the medium. This was confirmed using stirred tank bioreactors with pH control. No significant differences were observed in the highest cellulase titers and the protein pattern (according to the SDS-PAGE) of supernatants from pH controlled stirred tank bioreactor cultivations, when different nitrogen sources were used in the medium. Here the cellulase activities (~1.0 ± 0.2 FPU ml?1) were also much greater (8–150 times) than in shake flask cultivation. This paper describes the in situ bioconversion of glycyrrhizin of Glycyrrhiza glabra root to 18-β glycyrrhetinic acid by solid state fermentation. Fermentation was carried out with two different fungal strains, Penicillium chrysogenum and Rhizopus oryzae. The solid state fermentation was carried out under stationary state and under rotating state. Penicillium chrysogenum is a better producer of 18-β glycyrrhetinic acid than Rhizopus oryzae. The induced P. chrysogenum seed culture produces higher 18-β glycyrrhetinic acid with 2.955 mg g?1 and maximum β-glucuronidase activity of 3,583.8 U ml?1 under stationary solid state fermentation. The mycelium growth and bioconversion rate is highest at pH of 5.5 and 4.5, respectively. G. glabra root supplemented with a solution of dextrose 9 g l?1, MnSO4?·?H2O 3 g l?1 and (NH4)2SO4 0.540 g l?1 produces 48.580 mg of 18-β glycyrrhetinic acid per gram of G. glabra root, i.e. 86.74 % bioconversion by P. chrysogenum in 96 h under stationary state solid state fermentation.  相似文献

Objective

To produce δ-decalactone from linoleic acid by one-pot reaction using linoleate 13-hydratase with supplementation with whole Yarrowia lipolytica cells.

Results

Whole Y. lipolytica cells at 25 g l?1 produced1.9 g l?1 δ-decalactone from 7.5 g 13-hydroxy-9(Z)-octadecenoic acid l?1 at pH 7.5 and 30 °C for 21 h. Linoleate 13-hydratase from Lactobacillus acidophilus at 3.5 g l?1 with supplementation with 25 g Y. lipolytica cells l?1 in one pot at 3 h produced 1.9 g l?1 δ-decalactone from 10 g linoleic acid l?1 via 13-hydroxy-9(Z)-octadecenoic acid intermediate at pH 7.5 and 30°C after 18 h, with a molar conversion yield of 31 % and productivity of 106 mg l?1 h?1.

Conclusion

To the best of our knowledge, this is the first production of δ-decalactone using unsaturated fatty acid.
