To construct a linkage map enriched with tapping panel dryness (TPD)-related markers, we firstly utilized rubber tree ESTs associated with TPD to develop intron length polymorphism (ILP) markers. In this study, 52 new ILP markers were further developed. Together with the ILP markers previously reported, 102 ILP markers developed from TPD-related ESTs were analyzed within 39 Hevea germplasm in detail. The PCR success rate and polymorphism rate of ILP markers was 97.06 and 61.62 %, respectively. The results based on PCR amplification and sequence analyses provided the evidences on cross-species/genera transferability of rubber tree ILP markers. The average polymorphic information content (PIC) values of 39 Hevea germplasm were about 0.1719, indicating that the genetic base of Hevea germplasm selected in this study was very narrow. Among 39 Hevea germplasm, the PIC value of wild rubber tree accessions was the highest, followed by Hevea species and cultivated rubber tree clones. Based on the similarity coefficient of ILP markers, 39 Hevea germplasm were divided into three groups including cultivated clones, wild accessions and Hevea species, suggesting that the classification was generally related to the characterization of Hevea germplasm. The ILP markers developed in this study further enriches the number of molecular marker in rubber tree, and the ILP markers will have a wide application in DNA fingerprinting, genetic diversity, marker-assisted selection and genetic mapping, etc. Moreover, the ILP markers transferred cross-euphorbiaceae family might be utilized in cassava, castor bean and physic nut.  相似文献

Trees ofDyera, a small genus of the Apocynaceae, are characteristic of the rain forests in western Malaysia. The raw product prepared from the coagulated latex was formerly used in Malaya and elsewhere as an admixture with guttapercha(Palaquium spp.) and later as a substitute forHevea rubber for purposes not requiring elasticity. During the past 50 years, trees of the 2 species of this genus hare been the principal source in southeast Asia of a gutta-gum, exported in appreciable quantities to Europe and particularly to the United States.  相似文献

ABSTRACT

Natural rubber, obtained almost exclusively from the Para rubber tree (Hevea brasiliensis), is a unique biopolymer of strategic importance that, in many of its most significant applications, cannot be replaced by synthetic rubber alternatives. Several pressing motives lead to the search for alternative sources of natural rubber. These include increased evidence of allergenic reactions to Hevea rubber, the danger that the fungal pathogen Microcyclus ulei, causative agent of South American Leaf Blight (SALB), might spread to Southeast Asia, which would severely disrupt rubber production, potential shortages of supply due to increasing demand and changes in land use, and a general trend towards the replacement of petroleum-derived chemicals with renewables. Two plant species have received considerable attention as potential alternative sources of natural rubber: the Mexican shrub Guayule (Parthenium argentatum Gray) and the Russian dandelion (Taraxacum koksaghyz). This review will summarize the current production methods and applications of natural rubber (dry rubber and latex), the threats to the production of natural rubber from the rubber tree, and describe the current knowledge of the production of natural rubber from guayule and Russian dandelion.  相似文献

Using cultivars which are genetically resistant to South American leaf blight (SALB) caused by the fungus Microcyclus ulei is the only way to plant rubber trees in disease-affected areas. Numerous field observations led to the hypothesis that the resistance of the cultivar Fx2784 to SALB is likely to be monogenic. In this study, we investigated this hypothesis by examining the distribution of the trait in a cross between the resistant cultivar and a susceptible one. The individuals resulting from this cross were planted in field trials in French Guiana and Brazil. The resistance of all the trees was assessed by field observations. Bulk segregant analysis (BSA) using microsatellite markers was performed in French Guiana to determine which markers were genetically linked to resistance, and the results were validated by field observations in Brazil. In both locations, a 1:1 segregation of the resistance trait was observed, thus reinforcing the monogenic hypothesis. BSA showed tight linkage between resistance and the microsatellite markers located in linkage group 2 in the Hevea genome and enabled to pinpoint the resistance locus. The location was confirmed by observations on the trees planted in Brazil. This result should facilitate the use of Fx2784 resistance in future breeding programs for SALB resistance. This is the third major locus conferring resistance to SALB identified in rubber tree (Hevea spp.). These three loci are genetically independent, a favorable situation for genetic improvement of SALB resistance.  相似文献

Rigidoporus microporus (Polyporales, Basidiomycota) syn. Rigidoporus lignosus is the most destructive root pathogen of rubber plantations distributed in tropical and sub-tropical regions. Our primary objective was to characterize Nigerian isolates from rubber tree and compare them with other West African, Southeast Asian and American isolates. To characterize the 20 isolates from Nigeria, we used sequence data of the nuclear ribosomal DNA ITS and LSU, β-tubulin and translation elongation factor 1-α (tef1) gene sequences. Altogether, 40 isolates of R. microporus were included in the analyses. Isolates from Africa, Asia and South/Central America formed three distinctive clades corresponding to at least three species. No phylogeographic pattern was detected among R. microporus collected from West and Central African rubber plantations suggesting continuous gene flow among these populations. Our molecular phylogenetic analysis suggests the presence of two distinctive species associated with the white rot disease. Phylogenetic analyses placed R. microporus in the Hymenochaetales in the vicinity of Oxyporus. This is the first study to characterize R. microporus isolates from Nigeria through molecular phylogenetic techniques, and also the first to compare isolates from rubber plantations in Africa and Asia.  相似文献

Selective logging systems have been used to prevent the rapid decline of forest resources in Southeast Asia, but little is known about the impacts of selective logging on the genetic diversity of Southeast Asian rainforests. We evaluated the effects of silvicultural systems with differing cutting rotations and enrichment planting regimes on the genetic diversity of Shorea parvifolia, an abundant and ecologically important tree in Southeast Asian rainforests. Our result showed that in most respects the genetic diversity is not significantly different between primary forest and the other silvicultural systems; however, the proportion of private alleles is significantly different between them. Intensive second-rotation (L3) harvesting of individuals >40 cm in diameter at breast height (dbh) resulted in a sizable reduction in the number of reproductive trees and a dramatic decrease in the numbers of rare and private alleles, suggesting a negative impact on the genetic diversity of the remaining tree population. Enrichment planting with S. parvifolia in the logged forest improved some genetic parameters, significantly increasing the number of rare alleles in L3 in particular. We conclude that the genetic diversity of logged tropical forests gradually decreases depending on logging rotation times, especially with respect to sensitive genetic parameters such as the numbers of rare and private alleles, and that enrichment planting with native dipterocarps can maintain or even increase the genetic diversity of logged tropical forests in Southeast Asia.  相似文献

Hevea brasiliensis Muell. Arg (Para rubber tree) is a tropical tree species of Amazonian origin widely cultivated in several parts of the world for natural rubber, a highly priced commodity inevitable for the world rubber industry. Large, tree to tree variation in growth and latex yield among individual plants of high yielding Hevea clones is a common phenomenon observed in mature rubber plantations. The genetic heterogeneity of the seedlings which are used as rootstocks for propagation through budgrafting is considered as a major factor  responsible for this variation. In order to minimize this variation, attempts were made to develop highly uniform rootstock material via an in vitro technique by inducing zygotic polyembryony in Hevea. Immature open pollinated fruits of a high yielding clone RRII 105 were cultured by half ovulo embryo culture technique. Multiple embryos were induced from the 8–10-week-old zygote with a novel combination of gibberellic acid (GA3), kinetin, and zeatin. Plantlets were successfully generated from the multiple embryos and raised in the field post hardening. Screening using genetic and epigenetic molecular markers revealed that the multiple seedlings developed are highly uniform and are of single zygotic origin. Development of plants having genetic and epigenetic uniformity suggests that this technique is ideal for raising uniform rootstock material in Hevea which may significantly reduce intraclonal variations. Moreover, these plants could serve as ideal material for physiological and molecular investigations towards the understanding of  stock–scion interaction process in rubber.  相似文献

It has been shown that the disappearance of, or drastic changes in, ancestral and indigenous (or native) endosymbiotic microbiota can lead to many adverse health consequences. However, the effects of changes in beneficial endosymbionts in plants are poorly known (except for mycorrhizal and rhizobial associations). We sampled and compared endophytes from hundreds of trees belonging to the economically important genus Hevea, the source of natural rubber, in their native range in the Amazon basin and in plantations. We also conducted antagonism tests to determine the potential effects that some of these endophytes may have on selected plant pathogenic fungi. The natural and indigenous endosymbiotic mycota of the rubber tree (Hevea) contains a high diversity of beneficial fungi that may protect against pathogens (protective mutualism). In contrast, plantation trees have a reduced and different diversity of these beneficial fungi. We propose that abundance, and not just presence, of competitive fungal strains and species (i.e., Trichoderma and Tolypocladium) create a protective effect against pathogens in wild trees. This study provides support for the importance of mutualistic endosymbionts in plant health and ecosystem resilience, and calls for awareness of their potential loss by human-related activities.  相似文献

Originally from Asia, Rubus niveus has become one of the most widespread invasive plant species in the Galapagos Islands. It has invaded open vegetation, shrubland and forest alike. It forms dense thickets up to 4 m high, appearing to displace native vegetation, and threaten the integrity of several native communities. This study used correlation analysis between a R. niveus cover gradient and a number of biotic (vascular plant species richness, cover and vegetation structure) and abiotic (light and soil properties) parameters to help understand possible impacts in one of the last remaining fragments of the Scalesia forest in Santa Cruz Island, Galapagos. Higher cover of R. niveus was associated with significantly lower native species richness and cover, and a different forest structure. Results illustrated that 60% R. niveus cover could be considered a threshold for these impacts. We suggest that a maximum of 40% R. niveus cover could be a suitable management target.  相似文献

Spirocerca lupi is a common parasitic nematode associated with esophageal cancer of canids. Recent surveys have revealed an increasing number of canids infected with Spirocerca spp. in Africa, the Americas, Europe and Western Asia, and described a new species, Spirocerca vulpis, from red foxes (Vulpes vulpes). However, in Southeast Asia, research on Spirocerca spp. is scarce. Therefore, the aim of this study is to explore Spirocerca infection in domestic dogs in Vietnam and to identify the Spirocerca species by analyzing their morphometric and molecular data. We found Spirocerca spp. specimens in 51 (17.7%) out of 287 dogs examined with the intensity of infection ranging from one to 29 nematodes per dog. These nematodes were morphologically and molecularly identified as S. lupi. For morphology, the presence/absence of teeth, the ratio of glandular to muscular esophagus lengths, and the position of the vulva opening of S. lupi and S. vulpis varied between reports, suggesting caution in identification of Spirocerca species based exclusively on morphological characteristics. The molecular analysis based on a partial cox1 sequence revealed that S. lupi from Vietnam is genetically close to those from India and China, but far different from those of Israel, South Africa, Peru and Hungary. Given high genetic and morphological variations, more extensive surveys on Spirocerca spp. from various mammalian hosts at a greater scale are necessary to elucidate the divergence of this nematode.  相似文献

Nearly all of the species diversity in Piperaceae is encompassed within Piper and Peperomia. Both genera are pan-tropical with areas of diversification in the Neotropics and Southeast Asia. Piperaceae are less diverse in Africa with only two native species of Piper. This study examines the distribution of both Piper and Peperomia with representative samples from the Neotropics, Asia, Pacific Islands, and Africa. Molecular dating is used to place an age for the crown clades of Piper and Peperomia as well as ages for diversification within the clades. Both genera have origins in the late Cretaceous, but species level diversification occurred much later in the Tertiary. Biogeography of both genera are correlated with paleoclimate evidence to better explain the distribution and diversification of these large genera.  相似文献

Background

Snake envenomation is a serious public health threat in the rural areas of Asian and African countries. To date, the only proven treatment for snake envenomation is antivenom therapy. Cross-neutralization of heterologous venoms by antivenom raised against venoms of closely related species has been reported. The present study examined the cross neutralizing potential of a newly developed polyvalent antivenom, termed Neuro Polyvalent Snake Antivenom (NPAV). NPAV was produced by immunization against 4 Thai elapid venoms.

Principal Findings

In vitro neutralization study using mice showed that NPAV was able to neutralize effectively the lethality of venoms of most common Asiatic cobras (Naja spp.), Ophiophagus hannah and kraits (Bungarus spp.) from Southeast Asia, but only moderately to weakly effective against venoms of Naja from India subcontinent and Africa. Studies with several venoms showed that the in vivo neutralization potency of the NPAV was comparable to the in vitro neutralization potency. NPAV could also fully protect against N. sputatrix venom-induced cardio-respiratory depressant and neuromuscular blocking effects in anesthetized rats, demonstrating that the NPAV could neutralize most of the major lethal toxins in the Naja venom.

Conclusions/Significance

The newly developed polyvalent antivenom NPAV may find potential application in the treatment of elapid bites in Southeast Asia, especially Malaysia, a neighboring nation of Thailand. Nevertheless, the applicability of NPAV in the treatment of cobra and krait envenomations in Southeast Asian victims needs to be confirmed by clinical trials. The cross-neutralization results may contribute to the design of broad-spectrum polyvalent antivenom.  相似文献

Supplying 98% of the world's natural rubber,Hevea brasiliensis has transformed life around the world in only a century. This species-one of 10 in its genus-gives the best natural rubber, but much more remains to be studied concerning the numerous localised strains of the species. The British introduced to the Orient seeds from the lower Amazon-representatives of a not superior ecotype of the species but, at that time a century ago, the only available germ plasm. The time for study of the intraspecific variants ofH. brasiliensis and their use in programmes of genetic manipulation of the genus is long overdue. But basic to this desired programme is a clear taxonomic understanding of infraspecific variants, still awaiting evaluation. Although numerous taxonomic treatments of subspecific variants have been offered, there is little available to give genetic programmes concrete information.  相似文献

Peat swamp forest is an important refuge for biodiversity in Southeast Asia and is now becoming a target of exploitation. The scarcity of information on avifauna and ecology of birds in peat swamp forests prevents understanding of the effects of land use change on avifauna. In this study, we describe the bird assemblages in habitats with different land uses by comparing species richness, community composition, and feeding guild patterns in Bukit Batu, Indonesia. Bird assemblages in natural peat swamp forests (NPF), high-maintenance industrial acacia plantations (planted acacia forest, PAF), low-maintenance rubber plantations (jungle rubber forest, JRF), and village areas (VIL) were studied using a fixed-radius point-count method. Of the 95 species observed, 45, 20, 35, and 48 species were observed in NPF, PAF, JRF, and VIL, respectively. Estimated species richness was the highest in NPF, followed by VIL, JRF, and PAF. NPF had the highest species diversity and β-diversity, more endangered species, and a distinctive species composition characterized by fly-catching insectivores. The relative conservation value of PAF was notably low, particularly compared with JRF. The avifauna in VIL was characterized by more generalists that favor open spaces and therefore is not considered an important habitat for forest-dependent birds that are of conservation concern. Our results indicate that NPF has irreplaceable value for bird diversity conservation, but low-maintenance rubber plantations were home to several forest-dependent species and partially supported bird diversity, particularly compared with high-maintenance acacia plantations.  相似文献

Myxosporean parasites Kudoa spp. have been reported in several marine fish species worldwide. However, little is known about the contamination of these parasites in raw fish in Southeast Asia, where the consumption demand of uncooked fish is increasing. In 2019, the occurrence of several cases of raw yellowfin tuna (Thunnus albacares) obtained from retail shops with the presence of unknown white, nodular cysts within the musculature have raised public health concerns for the consumption of raw marine fish in Vietnam. Microscopic examination revealed numerous myxospores with the quadratic shape of the Kudoidae. Morphologically, stained spores detected in this study are suspected to Kudoa thunni. To confirm the suspected Kudoa species, further examination of the 18S small-subunit (SSU) was conducted and the results of nucleotide sequence analysis obtained from nodular cysts revealed 99.18–100% identity to that of Kudoa thunni sequences available in GenBank. Detection of K. thunni infection in tuna in Southeast Asia highlights the need for appropriate surveillance and control measures to ensure high quality standards and safety on raw fish production and consumption.  相似文献

Glochidion (Phyllanthaceae; leafflower trees) is a genus of trees which is widely reported to be pollinated by leafflower moths (Gracillariidae: Epicephala) in temperate and subtropical Asia, Australia, and the Pacific islands. However, the pollination ecology of Glochidion is not well described from tropical Asia, the region where it is most species-rich at both local (≤9 spp.) and regional (~200 spp.) scales. Here we report investigations of pollination biology and species-specificity of five Glochidion species in tropical Southeast Asia (Cambodia). Through nocturnal observations and fruit dissections, we find that at least three and likely five Glochidion species in Cambodia are pollinated by seed-parasitic leafflower moths. We find no evidence that any of these leafflower moths are non-mutualistic parasites, despite known examples of such parasites of this mutualism elsewhere in Asia. While the presence of a single larva in a fruit results in only a fraction of seeds being consumed, the presence of more than one larva per fruit—a frequent occurrence in some species—can result in almost all seeds within the fruit being infested. Multilocus phylogenetic analysis indicates that there are five different minimally monophyletic leafflower moth clades, each of which pollinates a unique Glochidion host species. Our results indicate that in its center of diversity in tropical Asia this system is an obligate pollination mutualism as previously described at the global margins of its distribution. These findings provide insights into the processes that generate and maintain biodiversity and maintain mutualism stability in plant–insect interactions in this biodiversity hotspot.  相似文献

Genotyping-by-sequencing (GBS) provides the marker density required for genomic predictions (GP). However, GBS gives a high proportion of missing SNP data which, for species without a chromosome-level genome assembly, must be imputed without knowing the SNP physical positions. Here, we compared GP accuracy with seven map-independent and two map-dependent imputation approaches, and when using all SNPs against the subset of genetically mapped SNPs. We used two rubber tree (Hevea brasiliensis) datasets with three traits. The results showed that the best imputation approaches were LinkImputeR, Beagle and FImpute. Using the genetically mapped SNPs increased GP accuracy by 4.3%. Using LinkImputeR on all the markers allowed avoiding genetic mapping, with a slight decrease in GP accuracy. LinkImputeR gave the highest level of correctly imputed genotypes and its performances were further improved by its ability to define a subset of SNPs imputed optimally. These results will contribute to the efficient implementation of genomic selection with GBS. For Hevea, GBS is promising for rubber yield improvement, with GP accuracies reaching 0.52.  相似文献
