为分析中国莲Cot-1DNA在其中期染色体上的分布,从中国莲基因组DNA中分离出Cot-1DNA,将基因组和所分离的Cot-1DNA用生物素标记后作探针,对中国莲染色体进行原位杂交。杂交结果用耦联有荧光素Cy3的生物素抗体检测,发现在每对染色体上均显示出特定的荧光原位杂交带。同时分析了FISH和GISH信号分布的异同。基于Cot-1DNA荧光原位杂交带型及染色体型,构建了中国莲核型。  相似文献

以早熟白菜苔为实验材料,从其基因组DNA中分离出C0t-1 DNA并用生物素标记作探针,25S rDNA用地高辛标记作探针,对有丝分裂中期相染色体进行双色荧光原位杂交。每对染色体上均显示出了特定的C0t-1 DNA荧光原位杂交带型,5对染色体上显示出了25S rDNA荧光原位杂交带型。双色荧光原位杂交证实了C0t-1 DNA与25S rDNA二者具有一致的染色体位置特征,表明基于rDNA及C0t-1 DNA的荧光原位杂交核型分析技术,优于目前普遍采用的只基于rDNA的荧光原位杂交核型分析方法。结合C0t-1 DNA与25S rDNA的荧光原位杂交带型和传统的染色体的形态学标记分析方法及白菜已公布的基于rDNA分布的核型分析结果,创建了一个精确的白菜核型。  相似文献

荧光原位杂交(FISH)是在染色体、间期细胞核和DNA纤维上进行DNA序列定位的一种有效手段。近年来,围绕提高检测的分辨率和灵敏性,不断将免疫染色、量子点和微流控芯片等物理化学技术引入到荧光原位杂交中,促进了它的快速发展。本文主要综述了荧光原位杂交的基本原理和发展历程,重点介绍了免疫染色-荧光原位杂交(immuno-FISH)、量子点-荧光原位杂交(QD-FISH)和微流控芯片-荧光原位杂交(FISH on microchip)等多种新技术及其检测特点,如快速、灵敏、动态、多样化等。随着荧光原位杂交技术的不断完善与发展,将在细胞遗传学、表观遗传学及分子生物学等领域发挥更加重要的作用。  相似文献

目的建立利用流式荧光原位杂交法检测细胞端粒长度的技术方法。方法以端粒酶敲除的G3小鼠和同龄野生型小鼠为检测对象,分离其外周血中的单个核细胞后与肽核酸荧光探针杂交,用流式细胞仪采集和分析其端粒长度,分别用荧光原位杂交方法和SYBR Green荧光定量PCR方法验证其准确性。结果流式荧光原位杂交法测定G3小鼠细胞端粒相对长度与C57BJ/6野生型小鼠相比为0.5345,荧光定量PCR测定端粒相对长度为0.5717,结果基本一致。结论流式细胞术与原位杂交方法结合起来检测细胞端粒的平均长度可靠易行,对单个核细胞端粒平均长度的检测有较高的实用性。  相似文献

为了解栽培种甘薯(徐薯18,Ipomoea batatas cv.XushuNo.18)的染色体结构,文章利用45SrDNA荧光原位杂交、自身基因组荧光原位杂交和银染技术对栽培种甘薯进行分子细胞遗传学研究。银染结果显示,徐薯18间期核有6对、8对和9对银染点;45SrDNA荧光原位杂交结果显示,徐薯18染色体上有8对或9对强弱不一的45SrDNA信号;自身基因组荧光原位杂交结果表明,所有染色体的全长分布强烈而密集的杂交信号,着丝粒区、近着丝粒区和端粒区有增强的信号带。  相似文献

荧光原位杂交技术是一种能够同时对微生物进行定性、定量和研究微生物群落空间分布情况的有力工具。简要介绍了荧光原位杂交技术的方法,并对其在人为创制环境和自然环境中特征性微生物种群及群落生态学中的应用研究进行了讨论,指出了该种技术在应用中存在的问题与缺陷,最后对荧光原位杂交技术在堆肥微生物生态中的应用及与其他方法的组合应用进行了展望。  相似文献

The lotus (Nelumbo nucifera Gaertn.) is an aquatic plant that grows in shallow water and has long been cultivated in South China. It can improve the incomes of farmers and plays an important role in alleviating poverty in rural China. However, a modern method is required to accurately estimate the area of lotus fields. Lotus has spectral characteristics similar to those of rice, grassland, and shrubs. The features surrounding areas where it is grown are complex, small, and fragmented. Few studies have examined the remote sensing extraction of lotus fields, and automatic extraction and mapping are still challenging methods. Here, we compared the spectral characteristics of lotus fields and other ground objects and devised a remote sensing method for the rapid extraction of lotus fields. Using this method, the extraction accuracy of lotus was 96.3%. The Kappa coefficient was 0.926, which is higher than those of the unsupervised K-means classification, Mahalanobis distance, and support vector machine supervised classification, and demonstrates the potential of this method for extracting and mapping lotus fields by remote sensing.  相似文献

莲(Nelumbo nucifera Gaertn.)不仅是重要的水生蔬菜作物之一,而且是进行基础研究的好材料。本文采用4种蛋白质提取方法(新型TCA/丙酮法、传统TCA/丙酮法、改良的Tris-HCl法、Tris-饱和酚法)并结合双向电泳技术,对莲子蛋白质提取方法进行筛选与优化。双向电泳实验结果显示,所得蛋白质图谱与莲种子蛋白质组成分布特点一致。通过PDQuest软件分析表明,新型TCA/丙酮法适用于莲子叶和胚芽组织的双向电泳蛋白质提取,而传统TCA/丙酮法则适用于莲胚轴组织双向电泳的蛋白质提取。研究结果为进一步利用质谱进行莲子蛋白质组研究奠定了基础。  相似文献

Sacred lotus (Nelumbo nucifera or lotus) is an important aquatic plant in horticulture and ecosystems. As a foundation for exploring genomic variation and evolution among different germplasms, we re‐sequenced 19 individuals from three cultivated temperate lotus subgroups (rhizome, seed and flower lotus), one wild temperate lotus subgroup (wild lotus), one tropical lotus group (Thai lotus) and an outgroup (Nelumbo lutea). Through genetic diversity and polymorphism analysis by non‐missing SNP sites widely distributed in the whole genome, we confirmed that wild and Thai lotus exhibited greater differentiation with a higher genomic diversity compared to cultivated lotus. Rhizome lotus had the lowest genomic diversity and a closer relationship to wild lotus, whereas the genomes of seed and flower lotus were admixed. Genes in energy metabolism process and plant immunity evolved rapidly in lotus, reflecting local adaptation. We established that candidate genes in genomic regions with significant differentiation associated with temperate and tropical lotus divergence always exhibited highly divergent expression pattern. Together, this study comprehensive and credible interpretates important patterns of genetic diversity and relationships, gene evolution, and genomic signature from ecotypic differentiation of sacred lotus.  相似文献

It is well known that no chlorophyll synthesis and photosystem biogenesis have been detected in dark-grown angiosperm seedlings. However, in this report, we showed that both PSⅡ and PSⅠ could be formed in dark-grown lotus ( Nelumbo nucifera Gaertn.) seedlings. Lots of evidence were given: First, during the dark-grown period, the single fluorescence emission peak at 679 nm in lotus embryo red-shifted and transformed into the normal PSⅡ fluorescence emission; Simultaneously, PSⅠ fluorescence emission at 730 nm appeared and increased obviously; Second, with partial denaturing SDS-PAGE method, PSⅠ chlorophyll-protein complex could be clearly separated from 10 days dark-grown lotus seedlings; Third, the existence of Lhca1 was also proved by Western blots. Moreover, measurements of electron transfer rate demonstrated that both PSⅡand PSⅠ core in dark-grown lotus seedlings were photochemically active.  相似文献

Nelumbo nucifera (lotus) is a perennial aquatic crop of substantial economical and ecological importance. Currently, the evaluation of the genetic variation of lotus germplasm accessions using codominant simple sequence repeat (SSR) markers is significant, and it is essential for understanding the population structure of N. nucifera. Here we report the genetic diversity and differentiation of 92 N. nucifera accessions (82 cultivated varieties and 10 wild lotus) using 50 polymorphic SSR markers. A total of 195 alleles were detected, with an average of 3.9 alleles/locus. The mean polymorphic information content (PIC) and the mean expected heterozygosity were 0.43 and 0.50, respectively. The genetic relationships among accessions were estimated using an unweighted pair‐group method with arithmetic average (UPGMA) cluster and principal coordinate analysis (PCA). Both methods revealed that the lotus accessions from China and those from its adjacent Asian countries formed a single cluster, respectively. The cultivated varieties were correlated with their major characteristics in cultivation (the seed, rhizome and flower type) rather than their geographic distribution. On the basis of the Bayesian model‐based analyses, two genetically distinct groups (the seed lotus group and the rhizome lotus group) were generated, with a strong differentiation between them (FST = 0.57). The seed lotus group exhibited higher genetic diversity than did the rhizome lotus group. The results herein indicated that the current levels of genetic diversity and differentiation between the lotuses have been greatly influenced by artificial selection.  相似文献

研究了莲的非可食用部分荷叶、莲子心、莲房、藕节、莲子壳和藕皮中生物碱的组分及含量。采用乙醇水溶液从非可食用部分提取生物碱,然后用高效液相色谱法进行测定。结果表明莲子心提取物中含有5种主要的生物碱,其中4种为莲心碱、异莲心碱、荷叶碱和甲基莲心碱。除莲子心外的其他部分的提取物组分较相似,都含有6~8种主要的生物碱。荷叶碱在荷叶中的含量最高,可达149.64μg/g;莲心碱、异莲心碱和甲基莲心碱在莲子心中的含量最高,分别为457.76、6155.85、1420.90μg/g。莲的六个非可食用部分中都含有5~8种主要的生物碱,有很好的开发利用价值。  相似文献

利用分子标记SRAP技术对国内广泛栽培以及新近育成的39个莲品种进行了DNA多态性分析。选取5对引物扩增基因组DNA,共获得101条带,其中88条为多态性条带,每对引物平均提供20个标记信息。由UPMGA方法得到的聚类分析结果表明了39个品种间的遗传关系,结果表明:花莲、籽莲和藕莲三大类群有明显的界限,花莲和籽莲的遗传距离较近,藕莲与它们的遗传距离较远。分子方差分析结果表明:莲品种间和品种内均存在遗传变异,藕莲品种内的遗传变异略低于品种间的遗传变异,而诱变籽莲、诱变花莲和常规籽莲品种内遗传变异均大于品种间的遗传变异,尤其是诱变籽莲、诱变花莲品种内遗传变异占总变异的分量分别超过了70%和60%,这种品种内的遗传变异,一方面对于莲的高产稳产具有重要的意义,另一方面也说明了从这些国内广泛栽培的品种以及新近育成的莲品种中可以直接进一步选育出更优良的新品种。  相似文献

采取超临界CO2萃取和水蒸气蒸馏提取荷叶挥发油,利用GC-MS对它们进行了定性、定量分析。结果表明这两种方法提取荷叶挥发油的化学成分及含量皆有很大差别。超临界CO2萃取的荷叶挥发油更具天然性,超临界CO2萃取法为提取荷叶挥发油的理想方法。  相似文献

利用ISSR技术对 1 2个莲藕品种进行了DNA多态性分析。从 6 5个随机引物中筛选出 7个引物扩增基因组DNA ,共获得 91条带 ,其中 75条为多态性标记 ,每个引物平均提供 1 3个标记信息。由UPMGA方法得到的聚类分析结果表明了1 2个品种间的亲缘关系 ,聚类结果与它们的系谱关系非常吻合  相似文献
