采用大孔吸附树脂分离纯化人工蛹虫草培养基残基中的虫草素,以HPLC法测定样品中虫草素含量,筛选出了适宜的大孔树脂NKA-II。研究了pH、洗脱剂乙醇体积分数等因素对该树脂吸附性能及解析性能的影响。结果表明,NKA-II型树脂纯化虫草素的最佳吸附条件为pH9,吸附流速2BV/h,该树脂对虫草素的吸附量可达到16.5mg/g。洗脱工艺条件为2.5倍树脂柱体积的50%乙醇,NKA-II大孔树脂对虫草素的解析率可达到95%以上。  相似文献

通过采用大孔吸附树脂对海红果黄酮粗提液的静态吸附和解吸试验,从10种大孔吸附树脂中筛选出海红果黄酮纯化的最优树脂,考察了该树脂对诲红果黄酮的静态、动态吸附与解吸性能并对吸附与洗脱的最佳条件进行了研究.结果表明:NKA-9树脂对海红果黄酮有很好的吸附和解吸性能,其最优的动态吸附工艺条件为:上样液pH值为4.0,浓度5.15 mg/mL,上样量为4 BV,流速控制在2 BV/h.最优的解吸工艺条件为:洗脱剂为80%乙醇溶液,洗脱液用量为3 BV,洗脱流速控制在1 BV/h.在此优化条件下,海红果黄酮的吸附率、解析率、收率、纯度的平均值分别达到为(79.39±0.13)%,(84.14±0.11)%,(68.20±0.15)%和(28.81 ±0.06)% (n=5).  相似文献

研究了大孔吸附树脂对乳清分离蛋白(WPI)酶解液的吸附特性。比较了6种大孔吸附树脂对WPI酶解物的静态吸附率与解吸附率。结果表明,DA201-C大孔吸附树脂最适合WPI酶解物的分离,其对WPI酶解液的动态吸附条件为:上样液浓度:10mg/mL;洗脱剂:75%乙醇溶液;洗脱剂流速:1BV/h。  相似文献

研究大孔吸附树脂吸附链霉菌702抗真菌活性物质的工艺条件。采用5种不同大孔吸附树脂吸附链霉菌702发酵液提取液中抗真菌活性物质,选择吸附效果较佳树脂进行吸附条件优化,以桔青霉为指示菌,纳他霉素为对照抗生素.采用“管碟法”测定抗真菌活性物质含量。结果发现,XAD18树脂吸附效果较好,获得优化吸附条件:上样液pH6,NaCl质量浓度10g/L,上样量22.63mg/g湿树脂,吸附流速2.5mL/min,水洗体积180mL,洗脱流速1.5mL/min,洗脱剂为体积分数10%、50%和90%的甲醇,洗脱方式为梯度洗脱。在确定的工艺条件下XAD18对链霉菌702抗真菌活性物质的吸附率可达90%,洗脱率可达75%,回收率可达80%。  相似文献

采用大孔树脂吸附法富集纯化黄芪总皂苷,以HPLC-ELSD法测定黄芪甲苷的含量作为考察指标,筛选了树脂型号、吸附流速、洗脱溶剂、洗脱流速以及洗脱溶剂用量等工艺条件.结果表明:最佳工艺为选择D101型大孔树脂,吸附流速为2 BV·h-1,洗脱流速为4 BV·h-1,收集5 BV的70%乙醇部分,得到的黄芪总皂苷纯化效果最好,黄芪甲苷的转移率可达93.21%.  相似文献

本文研究了用HZ-841吸附树脂精制银杏叶总黄酮的工艺。用10 BV 70%的乙醇分三次提取脱脂银杏叶粉中的银杏叶总黄酮,其收得率为4.8%,纯度为21.7%;用30BV纯净水、微波解冻提取30min,银杏叶总黄酮的收得率及纯度分别是2.63%和13.4%。HZ-841树脂对银杏叶总黄酮的动态吸附容量在pH=7.0时为0.265g/mL,树脂,动态吸附平衡时间为10min。酸度对HZ-841树脂吸附银杏叶总黄酮有显著影响,当pH=5.0时,其静态吸附量可达到0.322g/mL。吸附了银杏叶总黄酮的HZ-841树脂可用乙醇洗脱,当洗脱液pH=9.0、乙醇浓度为90%、洗脱流速3BV/h时,5BV洗脱液的收得率为1.8%。用无水乙醇洗脱的银杏叶总黄酮经过真空浓缩、干燥,获得的浅黄色粉末中银杏叶总黄酮含量为37.3%,产品收得率为2.41%。  相似文献

本文以板蓝根总木脂素和Clemastanin B的含量为指标探讨D101大孔树脂纯化板蓝根木脂素活性部位的工艺。通过对树脂比容积、吸附容量、吸附流速、泄漏曲线、水洗终点、径高比、梯度解吸、解吸曲线等参数的考察,从而得出富集纯化板蓝根木脂素活性部位的最佳工艺条件为:上样量5 BV,径高比1∶3,吸附流速4 BV/h,1BV水洗,5 BV的50%乙醇洗脱。并证明了所选的大孔树脂纯化工艺稳定、可靠,有效成分损失少,杂质去除率高,值得在生产中推广应用。  相似文献

以甜叶菊渣为原料,采用大孔树脂吸附和溶剂萃取法相结合的方法,得到90%以上纯度的总黄酮.通过对大孔树脂及溶剂萃取法的各影响因素进行研究,确定纯化甜叶菊渣中总黄酮的最佳工艺条件:AB-8型大孔树脂吸附流速为2 mL/min、上样液质量浓度1.5 mg/mL、上样液pH值为3.5、上样量4 BV,解吸液为50%乙醇溶液、解吸量5 BV、解吸流速为1.5 mL/min.优化后的甜叶菊总黄酮平均纯度为50.11%.后经乙酸乙酯在常温条件下萃取5次,得到甜叶菊渣中总黄酮纯度为91.8%.结果表明:通过AB-8型大孔吸附树脂和乙酸乙酯萃取相结合的方法,可以很好地纯化甜叶菊总黄酮.  相似文献

主要研究了从蛹虫草发酵液中提取虫草素的工艺,确定了大孔吸附树脂AB-8及十八烷基键合硅胶反相层析柱对蛹虫草发酵液中虫草素的分离条件;吸附最佳工艺条件为上样液p H6,上样浓度0.4mg/m L,上样量5BV,吸附流速1.0BV/h;解吸最佳工艺条件包括解吸液20%乙醇,解吸体积15BV,解吸流速4BV/h。得到的解吸液进一步以十八烷基键合硅胶分离,条件为上样液p H6,梯度洗脱,流动相为p H6的水和p H6的乙醇。洗脱液通过结晶和重结晶得到精制虫草素。三步得到的虫草素纯度分别达到40%、90%和99%以上。  相似文献

通过比较10种不同大孔树脂吸附蛹虫草深层发酵液中虫草素的性能,筛选出合适的大孔树脂H103,并对大孔树脂H103的吸附和解吸条件进行了考察。实验结果表明,上样吸附前调节pH值为9,上样浓度为0.2mg·mL^-1,吸附流速为2BV·h^-1时,大孔树脂H103对蛹虫草深层发酵液中的虫草素有良好的吸附性能;解吸时先用去离子水洗去部分杂质,再用40%的乙醇对虫草素进行洗脱,可以得到较好的效果。  相似文献

It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. Some correlations between ploidy level, morphological characters and distribution are noted.  相似文献

采用原位分子杂交方法检测HCV RNA及HBV X基因;采用免疫组织化学方法研究HCV核心抗原,非结构区C33c抗原及HBxAg在肝细胞肝癌中的定位及分布.结果表明(1)HCV RNA、HBV X基因在肝细胞肝癌组织检出率分别为40%(55/136)和82%(112/136).HCV RNA定位于癌细胞的胞浆内,阳性细胞呈散在、灶状及弥漫分布三种形式;HBV X基因在肝癌细胞中的分布呈胞浆型、核型及核浆型,阳性细胞也呈上述三种分布形式;(2)HCV C33c抗原、核心抗原在肝细胞肝癌中的阳性率为81%(133/164)及86%(141/164).C33c抗原定位于癌细胞及肝细胞的胞浆内;核心抗原既定位于癌细胞核中,又可定位于胞浆中.C33c抗原阳性细胞以灶状分布为主;而核心抗原阳性细  相似文献

For a plant selection model with frequency-independent viabilities, fertilities and selfing rates, it is shown that apart from global fixation, for certain parameter combinations a protected polymorphism and facultative fixation (either allele may become fixed according to initial frequencies) may both occur. Facultative fixation requires different selling rates for the dominant and recessive type. Protection of the polymorphism requires resource allocation for male and female function. In this connection the problem of purely genetically caused population extinction is discussed.
For general frequency dependence and regular segregation, the chances for establishment of a completely recessive gene are compared to those of a completely dominant gene. It is proven that the process of establishment of the recessive gene, despite a fitness advantage, may be considerably endangered by drift effects if random mating prevails. The recessive gene may reach the same effectivity in establishment as a dominant gene, only if the recessive homozygote mates exclusively with its own type during the period of establishment.  相似文献
