Silver crucian carp (Carassius auratus gibelio) is a unique triploid bisexual species that can reproduce by gynogenesis. As all other gynogenetic animals, it keeps its chromosome integrity by inhibiting the first meiosis division (no extrusion of the first pole body). To understand the molecular events governing this reproduction mode, suppression subtractive hybridization was used to identify the genes differentially expressed in fully-grown oocytes of the gynogenetic and gonochoristic crucian carp (gyno-carp and gono-carp). From two specific subtractive cDNA libraries, the clones screened out by dot blots and virtual Northern blots were chosen to clone full-length cDNA by RACE. Four differentially expressed genes were obtained. Two are novel genes and are expressed specifically in the oocytes. The gyno-carp stores much more mRNA of cyclin A2, a new member of the fish A-type cyclin gene, in its fully-grown oocyte than in the gono-carp. The last gene is histone H2A. The histone H2As of these two closely related crucian carps are quite different in the C-terminus. Preliminary characterization of the four genes has been analyzed by nucleotide and deduced amino acid sequence and Northern analysis.  相似文献

The spindle behavior and MPF activity changes in the progression of oocyte maturation were investigated and compared with cytological observation and kinase assay between gynogenetic silver crucian carp and amphimictic colored crucian carp.MPF activity was measured by using histone H1 as phosphorylation substrate.There were two similar oscillatory MPF kinase activity changes during oocyte maturation in two kinds of fishes with different reproductive modes,but there existed some subtle difference between them.The subtle difference was that the first peak of MPF kinase activity was kept to a longerlasting time in the gynogenetic silver crucian carp than in the amphimictic colored crucian carp.It was suggested that the difference may be related to the spindle behavior changes,such as tripolar spindle formation and spindle rearrangement in the gynogenetic crucian carp.  相似文献

银鲫与彩鲫卵母细胞cDNA文库构建及周期蛋白A1的cDNA克隆   总被引:12,自引:3,他引:12  
分别取行天然雌核发育繁殖的银鲫和两性生殖的彩鲫的卵母细胞为材料,提取总RNA,分离mRNA,进而反转录合成cDNA并定向插入λgtll Sfi-Not克隆载体,经体外包装构建了银鲫与彩鲫卵母细胞的表达型cDNA文库。测试结果表明库容量分别达到3.1×106(银鲫)和1.6×106(彩鲫)。进一步人工合成Cyclin A1保守引物,采用PCR扩增文库的方法,克隆了银鲫(1616bp)与彩鲫(1626bp)的Cyclin A1全长cDNA。序列分析结果表明:两种鱼编码区长度均为1173bp,起始于一个包含在脊椎动物起始密码子ANNATG基元内ATG的单一开放读码框,编码391个氨基酸;5'-端非编码区长度也同为70bp,3-'端非编码区长度略有不同,银鲫为373bp,而彩鲫则为383bp;二者3'-端均带有AATAAA的Poly(A)加尾信号以及24bp(银鲫)和27bp(彩鲫)的Poly(A)尾巴。比较银鲫、彩鲫和金鱼与人、爪蟾Cyclin A1氨基酸序列同源性的结果表明,Cyclin A1在人、爪蟾与鱼类之间具有较高同源性;而在银鲫、彩鲫和金鱼之间,Cyclin A1仅在周期蛋白框外存在5个氨基酸的差异,且这些差异均是由个别碱基的变异造成的。    相似文献

以雌核发育银鲫和两性生殖彩鲫的成熟卵为材料,分离卵壳,经处理得到卵壳可溶性蛋白组分。SDS-PAGE梯度凝胶电泳分析在雌核发育银鲫中揭示出3条较明显的差异蛋白带。同时,采用相同处理方法对受精前后卵壳蛋白组分进行比较分析后发现,这些差异蛋白带在受精后发生了变化,其带纹表现为减弱或消失,表明这些差异蛋白可能与受精过程相关。 Abstracts:Egg chorions were isolated from unfertilized and fertilized eggs of gynogenetic silver crucian carp and gonochoristic color crucian carp by homogenization and further purification techniques.Then,soluble proteins were extracted from the isolated egg chorions,and were analyzed by gradient SDS-PAGE.Three differential protein bands were revealed between the gynogenetic silver crucian carp and gonochoristic color crucian carp.Furthermore,these differential proteins were demonstrated to undergo obvious changes during fertilization.It was suggested that these differential proteins should be related to the special fertilization process in gynogenetic silver crucian carp.  相似文献

对雌核发育银鲫和两性融合发育红鲤的卵母细胞成熟过程,进行了详细的细胞学比较研究。在银鲫卵母细胞成熟过程中,绝大部分卵母细胞的核物质,在胚泡破裂(GVBD)后的第一次成熟分裂时期,出现明显的不同于红鲤卵母细胞核的行为,其染色体逐渐清晰地分为三群,这三群染色体的发育既彼此独立又相互联系,最终形成一首尾相接的三极纺锤体。随后,三极纺锤体扭转、重叠、合并成为一个正常的中期纺锤体。但极少数银鲫卵母细胞也出现了类似于红鲤卵母细胞的成熟单星光,并进而发育成两极纺锤体,形态类似红鲤第一次成熟分裂中期纺锤体。在银鲫上述两类卵母细胞的成熟过程中均未观察到“第一极体”外排的现象。由此,我们确认,银鲫卵是通过第一次成熟分裂异常,卵核染色体不减数来维持染色体倍性的;并且,根据上述特殊现象,我们对银鲫卵子第一次成熟分裂异常的机制进行了初步分析。    相似文献

采用实验细胞学技术,比较研究了红鲤精核在雌核发育鱼类(银鲫)和两性融合发育鱼类(红鲤、彩鲫)卵母细胞成熟各阶段中的发育状况,结果表明:在两性融合发育鱼类中,当卵母细胞发育到胚泡破裂期吋,精核开始解凝发育,及至第一次成熟分裂中期后阶段,高度解凝的精核才开始原核化,相应地,在雌核发育银鲫中,精核也只有在与胚泡物质接触后,才能开始解凝,并且在三极纺锤体形成后期初步原核化,但与两性融合发育鱼类相比,精核的原核化程度明显降低。这些实验结果初步揭示,在两性融合和雌核发育鱼类卵中均可产生促精核解凝和原核化的两类因子(SNDFs和SPDFs),其中SNDFs的活性表达起始于卵母细胞胚泡破裂时期,而SPDFs活性则产生于第一次成熟分裂中期后或三极纺锤体形成后阶段,并且两性融合发育鱼类卵中的SPDFs活性远远高于雌核发育鱼类卵母细胞。  相似文献

Sex evolution has been a debating focus in evolutionary genetics. In lower vertebrates of reptiles, amphibians, and fish, a species or a bioform reproduces either sexually or asexually but never both. A few species were found to consist of all females in fish. These all-female species can propagate by asexual reproduction modes, such as gynogenesis and hybridogenesis. However, the coexistence of sexuality and asexuality in a single species was recently noted only in a cyprinid fish silver crucian carp, Carassius auratus gibelio. This fish had been demonstrated to be capable of gynogenesis stimulated by sperm from other related species. Surprisingly, natural populations of this fish consist of a minor but significant portion (approx. 20%) of males. As different clones with specific phenotypic and genetic characteristics have been found, and RAPD markers specific to each clone have recently been identified, this fish offers many advantages for analyzing whether or not genetic recombination occurs between different clones. In this study, artificial propagation was performed in clone F and clone D. Ovulated eggs from clone F were divided into two parts and respectively inseminated with sperm from a clone D male and from a red common carp (Cyprinus carpio) male. The control clone D individuals were selected from gynogenetic offspring of clone D activated by sperm of red common carp. The phenotype and sex ratio in the experimental groups were also observed. Using RAPD molecular markers, which allow for reliable discrimination and genetic analysis of different clones, we have revealed direct molecular evidence for gonochoristic reproduction in the gynogenetic silver crucian carp and confirmed a previous hypothesis that the silver crucian carp might reproduce both gynogenetically and gonochoristically. Therefore, we conclude that the silver crucian carp possesses two reproductive modes, i.e., gynogenetic and gonochoristic reproduction. The response mechanism of two reproductive development modes may be the first discovery in vertebrates. Additionally, we discuss the evolutionary implication between gynogenetic and gonochoristic reproduction modes and the contribution of the minor proportion of males to genetic flexibility in the gynogenetic silver crucian carp. 银鲫是天然雌核发育的三倍体两性型种群, 因其遗传背景和生殖方式的特殊性, 已经成为研究单性和多倍体脊椎动物进化遗传学的理想模式鱼类。利用33个微卫星序列对雌核发育银鲫的突变速率和突变模式进行研究, 结果表明: (1) 22个子代个体中, 检测到1尾个体在15个微卫星位点具有18个突变等位基因; (2) 每个微卫星位点每代总体平均突变率是1.16×10-2, 95%置信区间是6.87×10-3~1.83×10-2, 与其他鱼类相比, 雌核发育银鲫的突变率明显偏高, 这与天然雌核发育鱼类处在单性生殖和两性生殖的过度阶段有密切关系; (3) 具有突变等位基因的13个位点的重复单元数目都在10次以上, 11个复合型微卫星位点的突变率(1.31×10-2)与21个完美型位点(1.00×10-2)的突变率没有明显差异(P = 0.67), 微卫星突变率受到重复单元数目的影响, 然而与重复结构类型和侧翼序列GC碱基含量无相关性; (4) 序列分析表明, 雌核发育银鲫的微卫星突变模式并不严格遵守逐步突变模型。  相似文献

萍乡肉红鲫(Pingxiang red-transparent crucian carp,Carassius auratus L.)是在江西省萍乡地区分布的天然三倍体鲫突变体经人工选育后获得的遗传性状基本稳定的后代,具有两性生殖和雌核生殖两种生殖方式.研究以F5代萍乡肉红鲫为材料,自孵化后每满1个月开始取性腺,观察了其卵巢1周年性成熟和精巢的发育过程,结果表明萍乡肉红鲫的性腺为1年成熟类型.卵巢发育进程町以分为6个时期,卵母细胞发育相应可分为6个时相.统计了卵巢成熟系数周年变化,体重为95 g左右的雌性萍乡肉红鲫,其成熟卵巢的成熟系数约为(11.73±2.8)%,成熟的卵母细胞内充满卵黄,相对怀卵量为(3018±310)粒/g.萍乡肉红鲫精巢属于小叶型,在精小叶中可观察到不同发育阶段的生殖细胞.由精原细胞分裂而来的仞级精母细胞经分裂增殖,产生次级精母细胞并最终发育成为精子.萍乡肉红鲫的精巢发育程序与普通鲫鱼和鲤鱼相似,卵巢和精巢的发育过程基本同步,孵化后50日龄内性腺分化不明显,到70日龄左右开始出现雌雄分化,3月龄发育为第1期,4-5月龄发育为第2期,6-7月龄发育至第3期,7-10月龄可见第4期卵巢,1年即可成熟产卵,精巢可排出精液.结果表明,该鲫鱼突变体的性腺发育与普通二倍体鲤(鲫)鱼的性腺发育方式类似.  相似文献

The histone H3.3 chaperone HIRA is essential for chromatin assembly during male pronucleus formation in Drosophila. However, the role of HIRA during fertilization in vertebrates remains unclear. The gibel carp (Carassius auratus gibelio) is a unique gynogenetic crucian carp (gyno-carp). Heterologous sperm nuclei cannot decondense when incorporated in the egg, thus the eggs produce a clonal lineage of all females by typical gynogenesis. In contrast, after entering the egg, homologous sperm can undergo decondensation and sexual reproduction is activated, which may produce both female and male offspring. Therefore, this fish is a useful model for studying the mechanisms of fertilization. Herein, we first compared HIRA expression during embryogenesis between gyno-carp and the gonochoristic color crucian carp (Carassius auratus; gono-carp). In gono-carp, a dramatic reduction of HIRA protein occurs shortly after fertilization, whereas HIRA protein is consistently expressed during embryogenesis of gyno-carp. Next, we used immunodepletion and an in vitro sperm decondensation system, and found that complete removal of HIRA inhibited sperm decondensation in both of the fish. Immunofluorescence localization showed that in the condensed sperm nuclei of gono-carp incubated in gyno-carp egg extracts, HIRA was detected, but neither the histone H2A variant H2af1o nor acetylated histone H4 was observed. These results suggest that HIRA may be a critical factor required for sperm nucleus decondensation, while the defect in deposition of some maternal histones in the sperm nucleus could be one reason why heterologous sperm cannot decondense in the gibel carp egg.  相似文献

Variation of two mtDNA fragments amplified in polymerase chain reaction was compared by the RFLP method in a population of the sympatric bisexual (diploid) and gynogenetic (triploid) forms of silver crucian carp. The mtDNA haplotypes of all individuals differed in at least 2.5% of nucleotide substitutions and fall into two phylogroups. All gynogenetic individuals have haplotypes of a single phylogroup, whereas haplotypes of diploid fish belonged to both mtDNA phylogroups. This testifies to a possibility of transformation of gynogenetic individuals into a bisexual form, whereas the reverse process is either extremely rare or impossible.  相似文献

Isozyme zymograms of esterase (EST), lactate dehydrogenase (LDH), malate dehydrogenase (MDH) and superoxide dismutase (SOD) were analysed by polyacrylamide gradient gel electrophoresis at different developmental stages of embryogenesis in 4 types of various ploidy crucian carp embryos, including haploids, diploids, natural triploids, and multiple tetraploids, and 2 types of haploid and diploid common carp embryos. Haploid embryos of crucian carp (Carassius auratus) and common carp (Cyprinus carpio) were produced by treating eggs with UV-irradiated milt from blunt snout bream (Megalobrama amblycephala). Natural triploid embryos were obtained from the eggs of gynogenetic silver crucian carp (Carassius auratus gibelio) inseminated with milt from red common carp. Multiple tetraploid embryos were also produced by gynogenesis from eggs of the newly discovered multiple tetraploid females inseminated with milt from red common carp. Gradient gel electrophoresis indicated that the band types and staining intensity of 4 isozymes expressed in haploid embryos of crucian carp and red common carp were similar to that in the correlative diploid embryos. In natural triploid silver crucian carp embryos, the zymograms of MDH and SOD isozymes were identical with that of diploid crucian carp embryos, but the EST and LDH isozymes manifested more new enzyme bands in comparison with diploid embryos. The corresponding expressed products of some bands in the triploid embryos, such as EST5 and EST6, could be observed also in red common carp embryos, which provided evidence for hybrid origin about the gynogenetic fish. The multiple tetraploids incorporated one foreign genome of red common carp, therefore, the effects of genes from the foreign genome could be observed in the multiple tetraploid embryos. Gene expression of the isozymes in the tetraploid embryos was somewhat similar to that in hybrids. Owing to interaction of triploid silver crucian carp genomes and common carp haploid genome, some isozyme bands, such as EST5 and EST6, changed in quantity, and some bands increased, such as s-SOD1, s-SOD2, s-SOD3 and s-SOD4 in the tetraploid embryos. Moreover, the heterogeneity was revealed among embryos developed from gynogenetic eggs of 3 different multiple tetraploid individuals.  相似文献

Massive production of all-female diploids and triploids in the crucian carp   总被引:3,自引:0,他引:3  
In many species of aquaculture importance, all-female and sterile populations possess superior productivity due to faster growth and a relatively homogenous size of individuals. However, the production of all-female and sterile fish in a large scale for aquaculture is a challenge in practice, because treatments necessary for gynogenesis induction usually cause massive embryonic and larval mortality, and the number of induced gynogens is too small for their direct use in aquaculture. Here we report the massive production of all-female triploid crucian carp by combining artificial gynogenesis, sex reversal and diploid-tetraploid hybridization. Previously, we have obtained an allotetraploid carp population (4n = 200) by hybridization between red crucian carp (Carassius auratus red var; ♀) and common carp (Cyprinus carpio; ♂). We induced all-female diploid gynogens of the Japanese crucian carp (Carassius cuvieri; 2n = 100). We also generated male diploid gynogens of the same species treated gynogenetic fry with 17-α-methyltestosterone, leading to the production of sex-revered gynogenetic males. Finally, these males were used to cross with the female diploid Japanese crucian carp gynogens and the allotetraploid females, resulting in the production of fertile all-female diploid Japanese crucian carp (2n=100) and sterile all-female triploid hybrids (3n = 150), respectively. Therefore, diploid crucian carp gynogenetic females and sex-reversed male together with an allotetraploid line provide an opportunity to produce all-female triploid populations in a large scale to meet demands in aquaculture industry.  相似文献

用银鲫克隆D,克隆A和鲤鱼的精子分别与银鲫克隆F的卵子受精产生了三种繁殖组合FD,FA和FL;再用转铁蛋白和同工酶标记对这三种组合的遗传模式进行比较研究。结果发现,FL组合的子代具有其母本完全相同的体形和电泳图谱,表现出雌核生殖银鲫的克隆品性。而在FD组合中则出现了体形的分化和酶谱的多态性;在一些个体的蛋白座位上同时检测到了父本和母本各自特有的谱带,证明FD生殖过程中有性重组的发生。同时,在所研究的蛋白的不同座位上存在着极端的连锁不平衡现象,可以推断在FD的重组过程中多个基因座位组成的连锁群(甚至是染色体组)可能作为一个整体参与基因的传递。此外,不同的蛋白座位上都未观察到重组的纯合表型,暗示在不同的基因连锁群之间还可能存在一种平衡致死的机制。FA组合的F1代具有类似母本克隆F的体形和蛋白表型,FA组合俱本近交产生的F2代却同时出现了克隆F和克隆A的体形和表型。即父本和母本的染色体组都能够通过有性生殖传递给FA子代,然而可能由于父母本基因的不相容性而使F1代父本的基因表达受到抑制。相对于雌核生殖的克隆遗传 ,本研究的揭示出来的有性生殖特性及伴随的有性质组能够使银鲫在一定程度上卸去积累的遗传负荷并从其它种群获取新的基因型,以维持其遗传多样性。多种可选择的生殖方式可能对于银鲫在自然条件下的生态适应有着重要意义,对于银鲫的遗传选育和养殖管理也有一定的参考价值。  相似文献

雌核发育个体的基因型基本上完全与母本相同,这是源于卵子发生过程中没有经过减数分裂.父本的遗传物质是在随机水平、亚基因组水平或基因组水平参与到子代的遗传重组过程,从而对长期突变积累的雌核发育生物基因组进行补偿,一直是遗传学家关注的问题.本文对雌核发育银鲫特异个体及父母本5个微卫星位点的扩增条带进行了克隆测序,相似性比对结果显示,特异个体所表现的父咎匾霥NA条带,序列结果与父本完全相同或相似(SCM4、SCM9、YJ5),并在某些位点上保留了母本的特异条带(YJ5),而个体本身特异的DNA条带与父母本的相似性均较高(SCM13).同时,所检测到的个体经越冬后查验为雌性个体,进一步进行同源繁育,研究变异条带在繁殖中的命运.连续2代的繁育检测结果表明,融合了父本特异性条带的银鲫个体在繁殖过程中仍行雌核发育的生殖方式,变异来的条带能够传递给子代,进一步证实同源雌核发育银鲫通过小概率两性融合事件丰富银鲫种群的遗传多样性[动物学报 53(3):537-544,2007].  相似文献

PCR-RFLP analysis of the ND3/ND4L/ND4 and 12S/16S rRNA regions and nucleotide sequence variation of the cytochrome b gene were used to study the mtDNA divergence in species of the family Cyprinidae, to examine the phylogenetic relationships of the species, and to identify their taxonomic status. The results indicated that an ancestral form diverged into silver crucian carp and crucian carp after its separation from the common carp lineage. The divergence of continental Carassius auratus gibelio and Japanese C. auratus cuvieri occurred more recently. Two well distinguishable mtDNA phylogroups, suggesting divergent evolution, were observed in continental C. auratus gibelio populations. The divergence was possibly related to the formation of two silver crucian carp groups with different types of reproduction, triploid gynogenetic and diploid gonochoric. At the same time, the results supported the high probability of current genetic exchange between the forms. In view of these findings and high morphological similarity of the two forms, they were not considered to be separate species.  相似文献

新疆伊犁河鲫鱼遗传多样性初步研究   总被引:1,自引:0,他引:1  
为了解我国西北地区野生鲫鱼的遗传多样性,鲫鱼样品从新疆伊梨河中采用小型拖网捕获。通过解剖观察性腺鉴定性别;采用碘化丙啶(PI)染色,经流式细胞仪测定血细胞核DNA含量;采用非变性聚丙烯酰胺凝胶电泳,考马斯亮蓝染色分析血清转铁蛋白多态性。结果表明:新疆伊梨河鲫鱼群体为具雌、雄的两性个体的群体;伊犁河鲫鱼血细胞核DNA平均含量为(5.315±0.215) pg/个细胞核,血细胞核DNA含量约为二倍体彩鲫DNA含量的1.53倍,与银鲫血细胞核DNA含量相当;在所采集的鲫鱼样品中共发现9种转铁蛋白表现型, 由7个等位基因调控,等位基因频率依次为Tf a0.063、Tf b0.063、Tf f0.095、Tf g0.169、Tf c0.174、Tf e0.175和Tf d0.270。基因型频率分别为Tf cd0.043、Tf cdf0.043、Tf abdg0.043、Tf def 0.087、Tf ce0.13、Tf de0.13、Tf df0.13、Tf abeg0. 13和Tf cdg0.26。综合分析,伊犁河现存野生鲫鱼为银鲫,且具有两性融合生殖能力。伊犁河鲫鱼表现出的遗传特性可能与该特定水环境有关。  相似文献

目的了解鲫成体透明的原因,探讨该性状的应用特性,为透明鲫作为水生实验动物材料系统开发提供基础。方法对透明鲫进行繁殖,并观察其后代性状,了解透明性状的遗传规律;体视镜观察透明鲫色素细胞的种类与分布,并与鲫比较;组织切片和压片确认微孢子虫对透明鲫的感染,并观察感染症状的变化。结果鲫的透明性状可以遗传,大部分后代表现为通体透明,心、肝、肾、肠、鳔、鳃、脊椎等组织器官肉眼清晰可见。与正常鲫比较,透明鲫的主要色素细胞为黄色素细胞,并未发现虹彩色素细胞,黑色素细胞的数量也大为减少。微孢子虫对鱼体的感染过程可直观观察,病原的扩散和空间分布能实时获得,具普通鱼类无法比拟的应用优势。结论虹彩色素细胞的缺失是鲫透明突变的结构基础。由于透明鲫内部器官可直接观测,无需依靠解剖或复杂仪器系统,在同一动物身上可能获得一系列的动态试验数据,或可作为模型材料广泛应用于生命科学不同领域。  相似文献

This study investigated the gynogenetic cytobiological behavior of the third gynogenetic generation (G3), which was generated from the diploid eggs produced by the second gynogenetic generation (G2)of red crucian carp × common carp, and determined the chromosomal numbers of G3, G2×scatter scale carp and G2×allotetraploid hybrids of red crucian carp × common carp. The results showed that the diploid eggs of G2 with 100 chromosomes, activated by UV-irradiated sperm from scatter scale carp and without the treatment for doubling the chromosomes, could develop into G3 with 100 chromosomes.Similar to the first and second gynogenetic generations (G1 and G2), G3 was also diploid (2n=100) and presented the hybrid traits. The triploids (3n=150) and tetraploids (4n=200) were produced by crossing G2 with scatter scale carp, and crossing G2 with allotetraploids, respectively. The extrusion of the second polar body in the eggs of G2 ruled out the possibility that the retention of the second polar body led to the formation of the diploid eggs. In addition, we discussed the mechanism of the formation of the diploid eggs generated by G2. The establishment of the diploid gynogenesis clonal line (G1, G2 and G3) provided the evidence that the diploid eggs were able to develop into a new diploid hybrid clonal line by gynogenesis. By producing the diploid eggs as a unique reproductive way, the diploid gynogenetic progeny of allotetrapioid hybrids of red crucian carp × common carp had important significances in both biological evolution and production application.  相似文献
