There has been an interest in using hyperaccumulating plants for the removal of heavy metals and metalloids. High selenium (Se) concentrations in the environment are detrimental to animals, humans, and sustainable agriculture, yet selenium is also an essential nutrient for humans. This experiment was conducted to screen fern plants for their potential to accumulate selenium. Eleven fern species, Pteris vittata, P. quadriaurita, P. dentata, P. ensiformis, P. cretica, Dryopteris erythrosora, Didymochlaena truncatula, Adiantum hispidulum, Actiniopteris radiata, Davallia griffithiana, and Cyrtomium fulcatum, were grown under hydroponic conditions for one week at 20 mg L(-1) selenate or selenite. Root Se concentrations reached 245-731 and 516-1082 mg kg(-1) when treated with selenate and selenite, respectively. The corresponding numbers in the fronds were 153-745 and 74-1,028 mg kg(-1) with no visible toxicity symptoms. Only three fern species were able to accumulate more Se in the fronds than the roots, which were D. griffithiana when treated with selenate, P. vittata when treated with selenite, and A. radiata regardless of the forms of Se. A. radiata was the best species overall for Se accumulation. More research is needed to further determine the potential of the fern species identified in this study for phytoremediation of the Se contaminated soils and water.  相似文献

The influence of increasing concentrations of copper, zinc, lead, nickel, chromium and cadmium on 14-day-old seedlings of wheat (Triticum aestivum L. cv. Vergina) was studied. Plants were grown in 1/10 strength Rorison’s nutrient solution with increasing concentrations of each of the metals added separately. The toxicity of metals depressed shoot growth but the most evident symptoms were on roots. The concentration of each metal which caused inhibition of root growth was chosen to study the influence of metals on isoperoxidases of wheat shoots. The concentrations employed did not alter the number of peroxidase bands but almost in all cases enhanced the intensities of bands of pH 4.0-4.2 and 5.0-5.4, while they decreased the intensities of bands of pH 4.2-4.6 and 5.4-6.5. The similar effects of the different heavy metals employed may suggest similarity in metal action on wheat isoperoxidases. The increased intensities of peroxidase bands may be considered as an indication of enhanced senescence caused by the heavy metal treatments. Generally, our results suggest that the heavy metals employed have caused complex changes on the multiple forms of peroxidases.  相似文献

After 3 h, selenate uptake by roots of Phaseolus vulgaris L.cv. Contender resulted in more than 50% of the Se absorbed beingconveyed to the aerial organs. This distribution was sensitiveto respiratory inhibitors and when roots were soaked in a solutionsupplied with hydroxylamine, the level of Se decreased by about80% in the whole plant, suggesting that selenate uptake requiresenergy. Addition of glucose to the nutrient medium resultedin slightly decreased uptake and distribution. Under the same growth conditions and 3 h incubation with selenite,a major part of the Se had accumulated in the roots, while asmall fraction was conveyed towards the aerial organs. Thispercentage was decreased by about 20% when plants were transferredto a solution supplied with hydroxylamine, suggesting that partof the selenite entered the roots passively. Addition of glucoseto the nutrient solution, resulted in enhanced levels of Sein the whole plant. Application of plant growth substances affected Se transport.When roots were incubated in abscisic acid (ABA), selenate uptakewas affected, while foliar spraying of gibberellin A3 (GA3)enhanced selenite uptake and translocation. Selenium (Se) is an essential trace element for humans and animals. A hydroponic experiment was performed to study the effects of sulphur (S) on Se uptake, translocation, and assimilation in wheat (Triticum aestivum L.) seedlings. Sulphur starvation had a positive effect on selenate uptake and the form of Se supplied greatly influenced Se speciation in plants. Compared with the control plants, Se uptake by the S-starved plants was enhanced by 4.81-fold in the selenate treatment, and selenate was readily transported from roots to shoots. By contrast, S starvation had no significant effect on selenite uptake, and selenite taken up by roots was rapidly converted to organic forms and tended to accumulate in roots. X-ray absorption near edge spectroscopy (XANES) analysis showed that organic forms of selenium, including selenocystine, Se-methyl-selenocysteine (MeSeCys), and selenomethionine-Se-oxide, were dominant in the plants exposed to selenite and accounted for approximately 90 % of the total Se. Whereas selenate remained as the dominant species in the roots and shoots exposed to selenate, with little selenate converted to selenite and MeSeCys. Besides, sulphur starvation increased the proportion of inorganic Se species in the selenate-supplied plants, but had no significant effects on Se speciation in plants exposed to selenite. The present study provides important knowledge to understand the associated mechanism of Se uptake and metabolism in plants.  相似文献

An experiment was undertaken to evaluate the effect of liquid manure amendment on heavy metal accumulation in wheat and barley. For this purpose, both kinds of seedlings were grown simultaneously in a Petri dish, while wheat seedlings were also grown in pots containing unpolluted agricultural soil. All of the seedlings were irrigated with one of the three prepared solutions: artificial rainwater solution, heavy metal solution and liquid manure solution containing NH4NO3, H3PO4 and KOH along with equal amounts of heavy metals as in the second solution. Twenty days later, 1 g of plant tissue was digested with the mixture of HNO3 and H2O2 for ICP-OES/HG-ICP-OES analysis. The results showed that the uptake of arsenic and mercury was highest for both plants grown in a Petri dish. Furthermore, the wheat grown in a Petri dish also had a high content of nickel, cadmium and copper, while the pot-grown wheat contained high amounts of iron and manganese, probably due to the adsorption of nickel, cadmium, copper and mercury on soil phases. The lower uptake of all heavy metals was observed after the amendment of liquid manure, with the exception of manganese in wheat and mercury in all plants.  相似文献

APR2 is the dominant APR (adenosine 5'-phosphosulfate reductase) in the model plant Arabidopsis thaliana, and converts activated sulfate to sulfite, a key reaction in the sulfate reduction pathway. To determine whether APR2 has a role in selenium tolerance and metabolism, a mutant Arabidopsis line (apr2-1) was studied. apr2-1 plants had decreased selenate tolerance and photosynthetic efficiency. Sulfur metabolism was perturbed in apr2-1 plants grown on selenate, as observed by an increase in total sulfur and sulfate, and a 2-fold decrease in glutathione concentration. The altered sulfur metabolism in apr2-1 grown on selenate did not reflect typical sulfate starvation, as cysteine and methionine levels were increased. Knockout of APR2 also increased the accumulation of total selenium and selenate. However, the accumulation of selenite and selenium incorporation in protein was lower in apr2-1 mutants. Decreased incorporation of selenium in protein is typically associated with increased selenium tolerance in plants. However, because the apr2-1 mutant exhibited decreased tolerance to selenate, we propose that selenium toxicity can also be caused by selenate's disruption of glutathione biosynthesis leading to enhanced levels of damaging ROS (reactive oxygen species).  相似文献

Effects of selenium compounds on the induction of heme oxygenase in human cells exposed to sodium arsenite or cadmium chloride have been investigated by an immunoblotting technique. Exposure of HeLa cells to arsenite or cadmium ions caused a marked increase in the synthesis of heme oxygenase, and the presence of sodium selenite suppressed the induction. DL-Selenocystine was an effective suppressor, and sodium selenate was less effective. DL-Selenomethionine had no effect. Northern blot analysis showed that selenite abolished the induction of heme oxygenase mRNA in the cells exposed to arsenite or cadmium ions. These results indicated that selenium antagonizes the induction of heme oxygenase by heavy metals ions.  相似文献

Selenite can be a dominant form of selenium (Se) in aerobic soils; however, unlike selenate, the mechanism of selenite uptake by plants remains unclear. Uptake, translocation and Se speciation in wheat (Triticum aestivum) supplied with selenate or selenite, or both, were investigated in hydroponic experiments. The kinetics of selenite influx was determined in short-term (30 min) experiments. Selenium speciation in the water-extractable fraction of roots and shoots was determined by HPLC-ICPMS. Plants absorbed similar amounts of Se within 1 d when supplied with selenite or selenate. Selenate and selenite uptake were enhanced in sulphur-starved and phosphorus-starved plants, respectively. Phosphate markedly increased K(m) of the selenite influx. Selenate and selenite uptake were both metabolically dependent. Selenite was rapidly converted to organic forms in roots, with limited translocation to shoots. Selenomethionine, selenomethionine Se-oxide, Se-methyl-selenocysteine and several other unidentified Se species were detected in the root extracts and xylem sap from selenite-treated plants. Selenate was highly mobile in xylem transport, but little was assimilated to organic forms in 1 d. The presence of selenite decreased selenate uptake and xylem transport. Selenite uptake is an active process likely mediated, at least partly, by phosphate transporters. Selenite and selenate differ greatly in the ease of assimilation and xylem transport.  相似文献

Background and aims

Selenium is an essential micro-nutrient for animals, humans and microorganisms; it mainly enters food chains through plants. This study proposes to explore effect of inorganic Se forms on its uptake and accumulation in Zea mays.

Methods

Zea mays was grown in a controlled-atmosphere chamber for 2 weeks in a hydroponic solution of low-concentration selenium (10 μg/L (i.e.0.12 μM) or 50 μg/L (i.e. 0.63 μM) of Se). For each concentration, four treatments were defined: control (without selenium), selenite alone, selenate alone and selenite and selenate mixed.

Results

At low concentrations, selenium did not affect the biomass production of Zea mays. However, for both concentrations, Se accumulation following a selenite-only treatment was always higher than with selenate-only. Moreover, in the selenate-only treatment, Se mainly accumulated in shoots whereas in the selenite-only treatment, Se was stocked more in the roots. Interactions between selenate and selenite were observed only at the higher concentration (0.63 μM of selenium in the nutrient solution).

Conclusions

Se form and concentration in the nutrient solution strongly influenced the absorption, allocation and metabolism of Se in Zea mays. Selenate seems to inhibit selenite absorption by the roots.  相似文献

Heavy metal contaminated soils often show increased levels of more than one metal, e.g. copper (Cu), cadmium (Cd), zinc (Zn), lead (Pb) or nickel (Ni). In case such soils are used for crop production, prediction of yield reduction or quality decline due to heavy metals in the soil is inadequate when based only on chemical soil analysis. The use of biomarkers such as phytochelatins (PC), non-protein thiols specifically induced in plants upon exposure to heavy metals, may be an additional tool or diagnostic criterion in heavy metal research and in practice. In the present work, Cu and Cd uptake and induction of PC synthesis are studied with hydroponically grown maize and wheat plants exposed to mixtures of the two metals. We observed a close positive relationship between the concentrations of Cd and PC in the plant shoot material. A decreased shoot concentration of Cd after addition of Cu, due to metal competition at common root absorption sites, coincided with lower shoot PC levels. Also differences in metal uptake and xylary metal transport among the two plant species were reflected in corresponding differences in PC concentration. The observed direct relationship between shoot PC concentration and the degree of metal-induced growth inhibition makes the use of PC promising for the purpose tested for.  相似文献

为了进一步研究镉、锌、铅 3种重金属元素间的相互作用以及对植物吸收重金属能力的影响 ,在模拟单一重金属污染试验研究的基础上 ,采用正交回归设计方案 ,研究了 Cd、Zn、Pb复合污染情况下紫花苜蓿和披碱草两种敏感性植物对 3种重金属的吸收效应。结果表明 ,在单一污染条件下 ,镉元素对紫花苜蓿生长的影响大于锌、铅 ,铅元素对披碱草生长的影响大于锌、镉 ;紫花苜蓿对于镉的吸收累积显著高于披碱草 ,植物内镉元素浓度最高达到 1 0 88.5 mg/kg,而披碱草对于铅元素的吸收则高于紫花苜蓿 ,植物内铅元素浓度最高达到 1 3 4 5 .5 mg/kg。在复合污染条件下 ,两种植物对铅、锌和铅、镉的吸收在不同浓度范围内分别存在存在着协同效应和拮抗效应 ;同时两种植物对锌、镉元素在实验涉及浓度范围内都存在着拮抗效应。这对于深入研究复合污染条件下重金属的土壤环境化学行为 ,对植物的综合毒性以及不同植物对重金属的吸收累积效应等 ,具有一定的参考意义  相似文献

Summary The influence of heavy metal additions on availability and uptake of cadmium, lead, zinc, copper, manganese and iron by oat was studied. The experiments were carried out as pot experiments using sandy loam, sandy soil and organic soil. Selective extractants were used to remove metals held in different soil fractions.Lead and copper were preferently bound by organics and oxides, zinc by oxides and inorganics, and cadmium by inorganics and organics.Addition of cadmium to the soils resulted in higher cadmium concentrations in all plant parts but lower concentrations of lead, zinc, copper, manganese and iron, and the accumulation indexes of these metals were also lower when cadmium was added to the soil.Addition of cadmium plus lead, zinc and copper resulted in higher cadmium concentrations in leaves and straw of plants grown in sandy loam and sandy soil, but lower concentrations when plants were grown in organic soil as compared with the results when cadmium was added separately. The transfer of cadmium, lead, zinc and copper from soil to plant was greatest from sandy soil, and zinc and cadmium were more mobile in the plant than were lead and copper.Cadmium concentrations in leaves correlated significantly with CaCl2 and CH3COOH extractions in sandy loam and sandy soil and with CH3COOH extractions in organic soil.Generally, the total metal uptake was lowest from organic soil.  相似文献

The relative plant availability of selenate versus selenite depends on the concentrations of competing ions, specifically sulfate and phosphate, respectively. In solution culture, the concentration of phosphate is typically 100- to 1000-fold greater than in soil solution, an artifact that could lead to underestimation of the phytoavailability of selenite. A nutrient solution study was conducted to compare the availability of selenite and selenate to perennial ryegrass (Lolium perenne L. cv. Evening Shade) and strawberry clover (Trifolium fragiferrum L. cv. O'Conner) at basal concentrations of SO4 (0.5 mM) and PO4 (2 μM) similar to those found in soil solution. Concentrations up to 5 mM SO4 and 200 μM PO4 allowed quantitative comparison of the efficacy of the competing ions. In both species, selenite was more phytotoxic than selenate, especially for shoot growth. Selenate was less toxic, and tended to preferentially inhibit root growth. Translocation percentages were much higher with selenate (≥84%) than with selenite (≤47%). A 10-fold increase in sulfate decreased uptake from selenate by >90% in both species. In ryegrass, 10-fold increases in phosphate caused 30% to 50% decreases in Se accumulation from selenite, but in clover such decreases only occurred in the roots. Sulfate-selenate antagonisms were thus stronger than phosphate-selenite antagonisms. Nonetheless, conventional nutrient solutions with millimolar phosphate will significantly underestimate Se availability from selenite, and moderate levels of sulfate salinity can inhibit selenate uptake sufficiently to reverse the apparent relative availability of the two forms of Se. Summary Rape, cucumber, wheat, oats and tomato were grown for one to two weeks in nutrient solutions with heavy metals added. Of the metal ions tested (Cr3+, Cu2+, Co2+, CrO4 2-, Ni2+, Cd2+, Pb2+, Mn2+, Zn2+ and Ag+), manganese, nickel and lead exhibited the greatest mobility in cucumber plants, which resulted in the highest shoot/root concentration ratio. Silver was not translocated to the shoots of cucumber plants in measurable amounts.When the plants were grown with 1.0, 10 and 100 M cadmium or nickel in the solution, the shoot and root concentration increased 5–10 times if the metal ion concentration of the solution was increased 10 times.The plants showed great differences in cadmium and nickel uptake. In the shoot, the cadmium concentration increased in the order: oats = wheat < cucumber = rape < tomato, and in the root in the order: oats = wheat < cucumber = rape < tomato. The great uptake of cadmium and nickel by tomato is notable and agrees with other reports.The nickel, and especially the cadmium, concentration in roots and shoots increases with the age of the plant.The results are discussed and related to other investigations. The effects of different concentrations of selenite (2–30 μM) and selenate (2–60 μM) on biomass production, leaf area, and concentrations of photosynthetic pigments in lettuce plants were investigated. On the basis of the obtained results, the threshold of toxicity for the selenite and selenate has been designated. The toxicity thresholds for selenite and selenate were determined at concentrations of 15 and 20 μM, respectively. Next, four selenium (Se) concentrations (2, 4, 6 or 15 μM), below or near the toxicity boundary, have been selected for the lettuce biofortification experiment. In the biofortified plants, the oxidant status (levels of lipid peroxidation and H2O2 concentrations), as well as Se and sulphur (S) accumulation were analysed. In the edible parts of the lettuce, the Se concentration was higher for selenate presence compared to selenite; however, this difference was not as obvious as it was noted in the case of the roots, where selenite application caused the high accumulation of Se. An application of 15 μM Se as selenite caused a decline in the biomass and an intensification of prooxidative processes in the plant’s tissues and as toxic should be excluded from further biofortification experiments. These results indicate that an application of either selenate or selenite to the nutrient solution at concentrations below 15 μM can be used for biofortification of lettuce with Se, evoking better plant growth and not inducing significant changes in the oxidant status, the concentration of assimilation pigments and S accumulation.  相似文献

Pot culture experiments containing sand and soil, at two levels of mercury (2 and 5 g mL-1) added through irrigation with increasing supplementation of selenium (selenite and selenate) led to a decrease in the uptake of mercury by tomato (Lycopersicum esculentum) plant. Both the forms of selenium (selenite and selenate) were found to be equally effective in reducing the mercury accumulation by plants. The observed reduction pattern of mercury accumulation in plant tissues has been discussed on the basis of the formation of insoluble HgSe complex in soil-root environment.  相似文献

The high levels of selenium (selenate, selenite) in agricultural drainage water in the San Joaquin Valley of California, which have led to environmental problems, might be lowered if the selenate/selenite could be reduced to elemental insoluble selenium. Two organisms have been newly isolated which do this in anaerobic coculture. One, a strictly anaerobic, Gram-positive rod, reduces selenite to elemental selenium. The other, a Pseudomonas species, was shown to respire selenate to selenite. Cells grown anaerobically in Minimal Medium on acetate plus selenate oxidized 14C-acetate to 14CO2 with concomitant reduction of selenate to selenite and small amounts of elemental selenium.  相似文献
