Unfertilized Lytechinus variegatus eggs in sea water in their normal physiological state have membrane potentials that approximate ?70 to ?80 mV. This conclusion is based on microelectrode measurements and on computation from the Na+ and K+ fluxes. The ?8 to ?15 mV values for the membrane potential previously reported and which are generally measured are the consequence of depolarization by impalement. The activation potential in inseminated eggs with an initial membrane potential more negative than ?60 mV is a compound event involving sperm-induced as well as voltage dependent conductance changes. The sperm-induced mechanism is a two-phase conductance increase which involves both Na+ and Ca2+ during the first phase, and Na+ alone during the second phase. In addition, the sperm-induced depolarization at the beginning of the first phase activates a voltage dependent Ca2+-conductance mechanism resulting in generation of an action potential.  相似文献

The effect of potassium on the water potential, the osmotic potential and the pressure potential in younger and older leaves of Phaseolus vulgaris grown in hydroponic culture was studied. Inadequate potassium supply resulted in an increase of the osmotic potential. In the older leaves the water potential was raised, in the younger leaves the pressure potential was depressed in the treatment insufficiently supplied with potassium as compared with leaves with an adequate potassium supply. Cell size of the younger leaves was smaller in the treatment with the low K+ supply in comparison with the leaves well supplied with K+. Potassium had a beneficial effect on plant growth, especially on fresh matter production. The water status of leaves (water content, pressure potential, osmotic potential) responded more sensitively to potassium supply than dry matter production. Besides organic N and organic anions, K+ was the most abundant solute found in the press sap of the leaves. From the results it is concluded that K+ is indispensible for attaining an optimum potential (turgor) in young leaves which in turn has an impact on plant growth.  相似文献

Water potential, osmotic potential and turgor measurements obtained by using a cell pressure probe together with a nanoliter osmometer were compared with measurements obtained with an isopiestic psychrometer. Both types of measurements were conducted in the mature region of Tradescantia virginiana L. leaves under non-transpiring conditions in the dark, and gave similar values of all potentials. This finding indicates that the pressure probe and the osmometer provide accurate measurements of turgor, osmotic potentials and water potentials. Because the pressure probe does not require long equilibration times and can measure turgor of single cells in intact plants, the pressure probe together with the osmometer was used to determine in-situ cell water potentials, osmotic potentials and turgor of epidermal and mesophyll cells of transpiring leaves as functions of stomatal aperture and xylem water potential. When the xylem water potential was-0.1 MPa, the stomatal aperture was at its maximum, but turgor of both epidermal and mesophyll cells was relatively low. As the xylem water potential decreased, the stomatal aperture became gradually smaller, whereas turgor of both epidermal and mesophyll cells first increased and afterward decreased. Water potentials of the mesophyll cells were always lower than those of the epidermal cells. These findings indicate that evaporation of water is mainly occurring from mesophyll cells and that peristomatal transpiration could be less important than it has been proposed previously, although peristomatal transpiration may be directly related to regulation of turgor in the guard cells.  相似文献

Calculation of the potential distribution across a uniformly charged ion-penetrable membrane that we developed previously is extended to derive a relationship among the surface potential, Donnan potential and charge density of an ion-penetrable membrane in a mixed solution of 2-1 and 1-1 electrolytes. Ketosis, meaning elevation of D-beta-hydroxybutyrate (R-3hydroxybutyrate) and acetoacetate, has been central to starving man's survival by providing nonglucose substrate to his evolutionarily hypertrophied brain, sparing muscle from destruction for glucose synthesis. Surprisingly, D-beta-hydroxybutyrate (abbreviated "betaOHB") may also provide a more efficient source of energy for brain per unit oxygen, supported by the same phenomenon noted in the isolated working perfused rat heart and in sperm. It has also been shown to decrease cell death in two human neuronal cultures, one a model of Alzheimer's and the other of Parkinson's disease. These observations raise the possibility that a number of neurologic disorders, genetic and acquired, might benefit by ketosis. Other beneficial effects from betaOHB include an increased energy of ATP hydrolysis (deltaG') and its linked ionic gradients. This may be significant in drug-resistant epilepsy and in injury and anoxic states. The ability of betaOHB to oxidize co-enzyme Q and reduce NADP+ may also be important in decreasing free radical damage. Clinical maneuvers for increasing blood levels of betaOHB to 2-5 mmol may require synthetic esters or polymers of betaOHB taken orally, probably 100 to 150 g or more daily. This necessitates advances in food-science technology to provide at least enough orally acceptable synthetic material for animal and possibly subsequent clinical testing. The other major need is to bring the technology for the analysis of multiple metabolic "phenotypes" up to the level of sophistication of the instrumentation used, for example, in gene science or in structural biology. This technical strategy will be critical to the characterization of polygenic disorders by enhancing the knowledge gained from gene analysis and from the subsequent steps and modifications of the protein products themselves.  相似文献

Summary Chloroperoxidase is an extracellular heme glycoprotein produced by the imperfect fungusCaldariomyces fumago. The enzyme can catalyse chlorination reactions as well as act as a catalase or a peroxidase. As a peroxidase, it has a wide substrate specificity and we are interested in some applied aspects of this activity, requiring the production and purification of moderate quantities of the enzyme. High levels of chloroperoxidase are produced in a fructose synthetic medium, and highest enzyme production occurs in a low-shear environment. fungal pellets produce enzyme continuously at low medium replacement rates and at up to 0.6 g enzyme per 1: chloroperoxidase is essentially the only extracellular enzyme produced. Enzyme purification is uncomplicated and gives good yields of high purity. Pure enzyme is stable for weeks at room temperature and under pH control. Chloroperoxidase can be ionically bound to aminopropyl glass, then covalently immobilized by glutaraldehyde crosslinking. Immobilized preparations have been washed and re-used five times, and are most stable at pH 5.5-6. Like many peroxidases, chloroperoxidase will oxidize phenols and phenolics, often causing a precipitate, and can totally remove phenols at low aqueous concentrations. Chloroperoxidase incubation with the petroporphyrin component of crude oil asphaltene (fraction 5) causes a reduction or removal of the Soret band (410 nm) and the -peak (573 nm). This petroporphyrin fraction is enriched with vanadium which poisons the chemical catalyst used in cracking crude oil.  相似文献

Sweet corn is a widely distributed crop that generates agricultural waste without significant commercial value. In this study, we show that sweet corn varieties produce large amounts of residual biomass (10 t ha?1) with high content of soluble sugars (25% of dry matter) in a short growing season (3 months). The potential ethanol production from structural and soluble sugars extracted from sweet corn stover reached up to 4400 l ha?1 in the most productive hybrids, 33% of which (1500 l ha?1) were obtained by direct fermentation of free sugars. We found wide genetic variation for biomass yield and soluble sugars content suggesting that those traits can be included as complementary traits in sweet corn breeding programs. Dual‐purpose sweet corn hybrids can have an added value for the farmers contributing to energy generation without affecting food supply or the environment.  相似文献

Papaver bracteatum, native to Iran and southern Russia, has been grown successfully in many countries. Research in the northwest United States has confirmed the potential for its commercial production as a source of the alkaloid thebaine. Potential for the chemical conversion of thebaine into codeine, one of man’s most widely used alkaloidal medicinal agents, is reviewed. Economic and social advantages of growing this species over opium poppy (Papaver somniferum) are discussed. The value of the seed oil for cooking and industrial use is considered.  相似文献

A synthetic progestin Nestorone® is being developed for female-contraception. This study was conducted to determine the distribution, metabolism, and excretion of tritium-labeled Nestorone (3H Nestorone) in adult female rats. Rats were injected subcutaneously (S.C.) with a single dose of 400 μCi 3H Nestorone/kg BW. Its distribution and concentrations in blood, plasma and other tissues were determined at defined times. The excreta were examined for elimination of 3H Nestorone. Radioactivity in all samples was analyzed by liquid scintillation counter. Metabolite profiling was performed by HPLC and LC/MS analysis of the plasma, urine, and feces samples. Following subcutaneous injection of 3H Nestorone, the mean peak concentrations of radioactivity (Cmax) in the blood and plasma were 58.1 and 95.5 ng equiv. 3H Nestorone/g, respectively, at 2-h postdose (Tmax). Thereafter, the concentration of drug steadily declined through 96-h postdose with a terminal elimination half-life (t1/2) of 15.6 h. 3H Nestorone-derived radioactivity was widely distributed in most tissues by 0.5 h and attained a mean maximal concentration by 2-h postdose. Approximately, 81.4% and 7.62% of the administered dose was excreted via feces and urine, respectively. In vivo metabolism of 3H Nestorone resulted into a total of 19 metabolites. Among them, two metabolites viz., 17α-deacetyl-Nestorone (M9) and 4,5-dihydro-17α-deacetyl-Nestorone (M19) were identified by HPLC and LC/MS analysis. Metabolite profiling of plasma samples showed that most of the circulating radioactivity was associated with unchanged parent drug, and M19. The M19 was a major metabolite in the profiled urine and feces samples. Presence of large proportion of drug/drug-related material in feces suggested that the biliary excretion is a main elimination route of 3H Nestorone. The distribution, metabolism, and excretion profiles of 3H Nestorone obtained in this study provide a fairly good insight about its fate in women.  相似文献

Standard methods of analysing foods for the presence of moulds are inadequate for thedetection of genera such as Chrysosporium which do not grow at the high wateractivities of most mycological media. The use of malt, yeast, 50% glucose agar (MY50G) insealed containers as an enrichment medium allowed time for germination and growth ofheat-stressed spores. Three Chrysosporium spp., C. xerophilum Pitt, C. inops (Carmichael) and C. farinicola (Burnside) Skou, were isolated fromcommercial chocolate bars with a water activity (a w ) of approximately0·28. Chrysosporium inops was isolated from commercial milk crumb and anew Chrysosporium sp. was isolated from Ghanaian cocoa beans. In chocolates madeby coating MY50G agar (aw = 0·89) with chocolate (aw = 0·27) containing C. inops arthroconidia, two types of deterioration were seenafter storage. The first was fat bloom due to recrystallization of the cocoa butter on the outer andinner chocolate surface. The second was growth of C. inops which occurred on theinside chocolate surface adjacent to the MY50G agar filling and on the outside surface afterholding at 92% equilibrium relative humidity (erh) for 12 d. There was some evidence that C. inops could grow on the outside of chocolates held at 5·7% erh after 4months' storage at 25 °C. The appearance of the white fungal growth was not unlikefat bloom to the naked eye but was clearly different with the electron microscope.  相似文献

Evidence is presented to show that pancreatic islets of Langerhans are capable of producing hepoxilins A3 and B3 from endogenous substrates as well as 14C-labeled 12-HPETE. Both hepoxilins are active in stimulating the release of insulin from these cells in the presence of 10 mM glucose. These experiments suggest that the hepoxilins may participate as potential endogenous mediators of insulin release in islets of Langerhans.  相似文献

Although the preparation of cyclodextrin (CD) monoesters with a variety of carboxylic acids has been already described in the literature, the direct regioselective CD acylation has proved to be critical, often requiring to be replaced with a more elaborate synthetic process. In this paper we describe the one-step preparation of several monoacylated CDs from acyclic or aromatic carboxylic acid derivatives. The ability of β-CD to enclose cupric ions in a sandwich-type manner was exploited to lead to high regioselectivity in the acylation of β-CD with benzoyl chloride, cinnamoyl chloride and phenyl acetyl chloride in water. Long chain aliphatic monoesters of α-, β- and γ-CD were best prepared in DMF. The results of our study showed that solvent and general conditions determined an overwhelming regioselectivity of acylation. 1H, 13C and 2D NMR experiments could easily discriminate the position of the ester. Monoacylated CDs were evaluated as a carrier of silibinin, the inclusion complexes were prepared and characterized by thermal analysis.  相似文献

Morphine is an opioid alkaloid commonly used in clinical practice for its analgesic properties. The phenolic hydroxyl group of that phenanthrene derivative is pivotal for binding to opioid receptors but it may also be responsible for the antioxidant behavior of morphine reported in several in vitro experiments. In this study, we assessed the effect of morphine on myeloperoxidase (MPO), a hemic enzyme from azurophilic granules of polymorphonuclear neutrophils involved in the production of cytotoxic and microbicidal reactive oxidants during inflammatory response. Specific immunological extraction followed by enzyme detection (SIEFED) and molecular modeling (docking) were performed to study the potential anti-catalytic action of morphine on MPO in comparison with the inhibitory effects of reference antioxidant molecules quercetin, gallic acid and ascorbic acid. The reducing action of morphine on the MPO peroxidase cycle has been investigated using electron paramagnetic resonance (EPR) and UV-visible absorption spectroscopy. Morphine acted as a reducing substrate in the peroxidase cycle of MPO and therefore protected the enzyme against the suicide action of its natural substrate, hydrogen peroxide. The SIEFED experiments associated with the docking study, further demonstrated a lack of strong and sustained anti-catalytic activity of morphine. In summary, from the results of this study, it appears that morphine acts as a weak and reversible inhibitor of MPO that may nonetheless contribute to immunomodulatory and antioxidant effects of this opioid analgesic in vivo.  相似文献
