Summary The paper deals with some physiological studies of two typical strains ofPhytophthora palmivora Butl. andP. parasitica Dast. var.macrospora Ashby causing severe fruit rots ofAchras sapota L. andAnona squamosa L. respectively. The effect of temperature on the germination of sporangia had a marked effect. Maximum percentage of indirect germination (by formation of zoospores) occurred at 20° C, whereas direct germination of sporangia (by formation of germ tube) was at 30° C, which further continued upto 37° C. Maximum indirect germination at 20° C shows the favourable cool temperature for the epidemic of the diseases (Fruit rots) in nature. The best substrate (medium) for germination of sporangia was found to be tap water. Next to this was the host decoction or extract. Glucose solution also accelerated sporangial germination. The effect of two dyes viz., Malachite green and crystal violet was also studied in relation to growth and sporulation ofPhytophthora isolates. Their addition to the medium in various fractional dilutions had a profound influence in the rate of growth and sporulation. An interesting observation noted was that growth of the isolates was inversely porportional to the various concentrations of the dyes, under study. An attempt was also made to study the influence of various vitamins. In all, six vitamins were included in the study. Out of these, thiamine and riboflavin were found to be the best sources promoting good growth and sporulation of both the species ofPhytophthora under study.Forms a part of Senior author's M. Sc. Among the various culture media studied, oat meal, corn meal, lima bean and rice meal agars were the best ones, with abundant growth and sporulation of the organism. Lactose, galactose, dextrin and glycogen were the best sources of carbon. Among the various nitrogenous compounds studied, ammonium phosphate (monobasic), ammonium sulphate, ammonium tartrate, ammonium lactate, asparagin, glycine and glutamic acid were the best ones utilized by the organism. The sulphur nutrition of three isolates ofAlternaria tenuis Auct., isolated from the diseased leaves ofMangifera indica L.,Musa paradisiaca L. andPsidium guajava L., was studied. They were grown on the medium devoid of sulphur as well as on media containing various sources of sulphur viz., ammonium sulphate, sodium hyposulphite, sodium thiosulphate, magnesium sulphate, potassium sulphate, potassium metabisulphite, zinc sulphate and thiourea. Sodium hyposulphite, sodium thiosulphate, magnesium sulphate, potassium sulphate and zinc sulphate were generally found to be satisfactory sources for the growth of all the isolates under study. Poor growth of the different isolates was observed on the medium devoid of sulphur.  相似文献

The paper deals with the influence of temperature on the growth and sporulation of two species ofPhytophthora, viz.,P. palmivora Butl. andP. parasitica Dast. var.macrospora Ashby, the causal agents of fruit rots ofAchras sapota L. andAnona squamosa L. respectively. Germination of sporangia at different temperatures were also undertaken. There was marked variation in growth and sporulation of these two organisms. Isolate C (Phytophthora palmivora) showed no growth at 5° and 35°C, scanty growth at 10° and 32.5° with an optimum temperature between 26–28°C. On the other hand, Isolate S (Phytophthora parasitica var.macroscora) showed no growth at 10°C, but slight growth even at 37°C. Eight days exposure at 37°C completely stopped the growth of this Isolate. It showed best growth at 30°C and hence this was its optimum temperature. In general, Isolate C sporulated abundantly at all temperatures tested but reached its maximum at 25°C. On the other hand Isolate S showed best growth but failed to sporulate at any of the temperatures in 98 hours growth, although it sporulated freely when the incubation period extended up to two weeks. On the basis of temperature toleration the twoPhytophthora isolates are distinguished from each other as two different species. This confirms the earlier observations and nomenclature criterion as emphasized and formulated byTucker (1931). In the germination studies, it was observed that the indirect germination with the formation of abundant zoospores started from 5° and continued even up to 35°C, reaching maximum at 20°C. High temperature was not favourable for indirect germination. As the temperature proceeded increasing, the percentage of direct germination by formation of germ tubes also increased. Direct germination was observed from 10° which continued up to 37°C, with a maximum reach at 30°C. This confirms the epidemic of fruit rots in nature during monsoon season which is prevalent with the persistence of high humidity and rainfall.Taken from a thesis submitted by the author for the degree of Master of Science in the Faculty of Agriculture, Poona University, India.  相似文献

Three isolates ofA. tenuis isolated from the diseased leaves ofMangifera indica l. Musa paradisiaca l. andPsidium guajava l. were investigated. They were grown on different sources of nitrogen viz., potassium nitrate, sodium nitrate, calcium nitrate, ammonium nitrate, sodium nitrite, ammonium sulphate, ammonium chloride, glycine, DL-valine, L-glutamic acid, urea, thiourea, L-asparagine and peptone. They were also grown on the medium lacking nitrogen. A wide variation was observed in the growth and reproduction of the different isolates. The growth of all of them was good on potassium nitrate, calcium nitrate, glycine, DL-valine, L-glutamic acid, L-asparagine and peptone but the sporulation was satisfactory on calcium nitrate only. Sodium nitrite supported moderate growth of banana leaf isolate whereas there was no growth of the other two isolates. None of the organisms could grow on the medium lacking nitrogen as well as on thiourea. The results obtained with the isolates under study have been compared with those of earlier investigators and it has been clearly established that the different isolates ofA. tenuis could show marked differences in their nitrogen requirements.  相似文献

Summary This paper gives an account of some cultural and physiological studies of an isolate ofPhytophthora parasitica Dast. var.macrospora Ashby, the causal agent of fruit rot ofAnona squamosa L. Among the various culture media studied, non-synthetic ones like oat meal agar, corn meal agar, lima-bean agar, carrot extract agar, soya-bean extract agar and steamed rice agar were the best, on which the organism showed marked growth and sporulation. Non-synthetic types were poor in this direction. Regarding the effect of various carbon sources, sucrose, lactose, maltose, raffinose, inulin, dextrin, dulcitol, glycogen, rhamnose and xylose supported growth and sporulation of the organism. Sodium nitrate, ammonium nitrate, magnesium nitrate, potassium nitrate, ammonium lactate and asparagin were the best sources of nitrogen. 6.5 was found to be the best pH for the growth and sporulation of the organism.A portion of Senior Author's M.Sc. Summary Nitrogen and sulphur requirements ofColletotrichum inamdarii Lal isolated from the leaves ofCarissa carandas L. have been studied. DL-serine, L-asparagine and L-phenylalanine have been found to be of good nitrogen source followed by potassium nitrate, calcium nitrate, magnesium nitrate, DL-alanine, ammonium nitrate, glutamic acid, ammonium sulphate, DL-valine, aspartic acid, ammonium chloride, ammonium hydrogencarbonate, L-histidine and potassium nitrite. There was no growth in the absence of nitrogen.Sporulation was excellent on calcium nitrate and sodium nitrate, Very good on DL-serine, potassium nitrate, and magnesium nitrate. Good on L-asparagine, L-phenylalanine and ammonium oxalate. Fair on DL-alanine, DL-leucine, ammonium sulphate, DL-valine, ammonium chloride and L-histidine whereas poor on glutamic acid, aspartic acid, ammonium tartarate and ammonium nitrate. Few spores were observed on ammonium hydrogencarbonate but potassium nitrite did not show any sporulation.Amongst the sulphur compounds sodium bisulphate gave the best growth and good sporulation, followed by sodium thiosulphate, magnesium sulphate, ammonium sulphate and potassium sulphate. Thiourea gave negligible growth whereas it failed to grow on zinc sulphate and potassium persulphate.  相似文献

Nine inorganic nitrogenous compounds and sixteen amino acids were used to study the growth and sporulation of highly and weakly virulent isolates ofColletotrichum falcatum Went. Potassium nitrate, sodium nitrate and calcium nitrate supported good growth, while ammonium phosphate, potassium nitrate and sodium nitrate supported good sporulation of both isolates. Among the amino acids tested, glycine and DL-alanine were best for growth, and DL-threonine, L-leucine, DL-tyrosine, DL-serine, DL-phenylalanine and L-cystine for sporulation. No correlation existed between virulence and growth but the highly virulent isolate sporulated significantly more than the weakly virulent isolate.This work is a part of the author's Ph. D. The effect of different sulphur and phosphorus compounds on the growth and sporulation ofCurvularia pallescens Boed. has been studied. Nine different sulphur sources were tried but among them only magnesium sulphate yielded the best dry weight of the fungus. Zinc sulphate, sodium sulphate, sodium thiosulphate, potassium sulphate and calcium sulphate supported good growth. Poor growth was recorded on sodium bisulphite, ammonium sulphate, sodium sulphide and control. Sporulation was excellent on magnesium sulphate. It was good on zinc sulphate, sodium sulphate and potassium sulphate. On sodium thiosulphate, calcium sulphate, sodium bisulphite and control it was fair. Sodium sulphide and ammonium sulphate had inhibitory effect as sporulation was poor and nil on these two compounds respectively.Six phosphorus compounds were studied. Tripotassium phosphate gave best growth and excellent sporulation. Good growth and excellent sporulation was recorded on monobasic potassium phosphate and magnesium phosphate. Growth and sporulation were good on dibasic potassium phosphate and sodium dihydrogen phosphate. Ammonium phosphate was poorly utilized.  相似文献

Summary The growth ofChlorella pyrenoidosa in a special medium based on the critical concentrations of nutrients for autotrophic growth has been shown to be stimulated more by chloride with the sodium salt than with the potassium salt, more by sulphate in the presence of sodium than in the presence of potassium, and to be inhibited by sodium dihydrogen phosphate and not by potassium dihydrogen phosphate. Furthermore, it has been found that calcium reversed the growth inhibition produced by sodium dihydrogen phosphate, and that strontium only partially substituted for calcium.  相似文献

Summary Anacystis nidulans and Anabaena variabilis contain sufficient sulphur reserves to enable them to perform only one round of growth cycle in the non-sulphur growth medium. Sulphate, sulphite, l-methionine and d-methionine, each can act as a suitable sulphur source, but they differ in respect of their growth promoting action; sulphate uptake seems to be a light driven phenomenon and the sulphate metabolizing enzymes are inducible in nature. Methionine appears to act as a repressor of sulphate-metabolizing enzymes.  相似文献

Summary Four isolates ofCytospora cincta Fr. and 2 ofC. leucostoma Fr. were cultured on media containing 7 different nitrogenous compounds. Maltose served as a constant source of carbon. All experiments were carried out at 25° C.Total growth as determined by mycelial weights, and degree of sporulation as determined by an arbitrary system, revealed that 1) potassium nitrate was most satisfactory as a source of nitrogen, 2) response of the isolates tended to segregate them along species lines, and 3) the pH of the medium appeared to be a factor in the degree of response.Growth-habit experiments emphasized 1) extreme variation in colony characteristics and 2) the need for standardization of laboratory environments for comparative studies of the fungi.Approved by the Director of the Idaho Agricultural Experiment Station as Research Paper No. 518.  相似文献

Summary The paper deals with some detailed pathological investigations with two species ofPhytophthora, viz.,Phytophthora palmivora andP. parasitica var.macrospora causing severe fruit rots ofAchras sapota andAnona squamosa respectively. Both the isolates were found pathogenic to a variety of fruits including papaya, tomato, chillis, banana, fig, apple, cucumber, guava, orange, onion bulb and potato tuber. They were, however, non-pathogenic to carrot, zinger, turmeric andColocasia corms. In the seedling inoculation experiment they showed pathogenic nature to castor bean, brinjal, cotton, sesamum, pea, French bean,Cactus sp. andBryophyllum leaves. Growth of the fungusMicrosporum gypseum and utilization of cystine during this growth was studied in a glucose-arginine medium containing either sodium sulphate, andL-cystine orDL-cystine. Replacement of sulphate withL-cystine brought about no significant changes in the growth of the microorganism. Utilization ofL-cystine as a source of carbon and nitrogen was rapid and complete and excess sulphur was excreted into the medium in the form of sulphate. Similarly excreted were also minute amounts of sulphite which immediately reacted with the remaining cystine to formS. sulphocysteine. Growth ofM. gypseum in a medium withDL-cystine was slow. Although this substance was not utilized as readily asL-cystine, its utilization was still complete and excess sulphur was similarly excreted in the form of sulphate and sulphite. The initial step in the utilization of theD-isomer is probably its extracellular deamination.  相似文献

Seven clinical isolates and the ATCC strain 35405 ofTreponema denticola, obtained from human periodontal pockets, were studied for peptidase activity with several chromogenic compounds as substrates. The cell sonicates of all strains hydrolyzed phenylazobenzyloxycarbonyl-l-prolyl-l-leucyl-glycyl-l-prolyl-d-arginine (a collagenase substrate), azocasein, and the 2-naphthylamines ofl-proline,l-hydroxyproline,l-pyrrolidine, and benzoyl-l-arginine, but the rates of hydrolysis varied considerably from strain to strain. Fast protein liquid chromatography on gel and anion exchange columns revealed further biochemical differences between the strains. The ATCC strain consistently produced several proline iminopeptidases, whereas four of the clinical isolates yielded high and three yielded low iminopeptidase activity. The ATCC strain and six clinical isolates displayed high benzoylarginine peptidase activity. The use ofN-l-prolyl-2-naphthylamine as substrate revealed more differences between the strains than other substrates. The substrate specificity of the enzymes discovered suggests that they may be important for the nutrition of the organism or in the protection of the organism against chemical defense factors present in the gingival pocket.  相似文献

Mycelial growth of an isolate ofT. bakamatsutake was tested in media with C/N ratio ranging from 0 to 50 and with 32 carbon and 12 nitrogen sources. The isolate grew best at the C/N ratio of 30. It utilized the monosaccharidesd-glucose,d-mannose, andd-fructose, the disaccharide trehalose, and polysaccharide pectin among the carbon sources; and yeast extract,l-glutamic acid, and ammonium compounds among the nitrogen sources. The growth of ten isolates and secretion of gluconic and oxalic acids were compared ind-glucose, trehalose, and pectin media. The utilization ofd-glucose, trehalose, and pectin differed among the ten isolates, but all the isolates secreted gluconic acid in thed-glucose media and oxalic acid in the pectin media.  相似文献

Summary The cytoplasmic organization of the long-lived, thick walled resting stage of the sporangium ofSynchytrium endobioticum (Schilb.) Perc. is described. The cytoplasm of the resting sporangium contains a large number of closely packed lipid bodies and irregular electron dense bodies, which are interspaced with fine channels of cytoplasm. These ultrastructural observations are discussed in relation to the hypothesis ofBally (1912) andCurtis (1921) that zoospore primordia are already present during the resting stage. It is shown that the zoospore primordium is actually a lipid body and an osmiophilic body and the strands postulated to connect the individual zoospore primordia are actually the fine channels of cytoplasm.A new inner wall layer is laid down prior to the start of the germination. It is this wall layer which will protrude to form the vesicle in which sporogenesis takes place. The germination process observed, protrusion of a vesicle through a crack in the sporangial wall, the migration of the sporangial content into the vesicle, and the formation of a single, membrane-bound sporangium within this vesicle, is in full agreement with the recent light microscopic studies ofSharma andCammack (1976). These observations support the transfer ofS. endobioticum from the subgenusMesochytrium to the subgenusMicrosynchytrium (bothsensu Karling 1964).A major objective of the study, to obtain ultrastructural evidence for the location of the meiotic divisions in the life cycle, was not fulfilled.Three different fungi were observed to parasitize the resting sporangium ofS. endobioticum. These infections are discussed in relation to other mycoparasites of plant pathogenic fungi. The possibility of using a mycoparasite for the biological control of potato wart disease is considered to be without practical relevance.  相似文献
