Infection of cells by picornaviruses of the rhinovirus, aphthovirus, and enterovirus groups results in the shutoff of host protein synthesis but allows viral protein synthesis to proceed. Although considerable evidence suggests that this shutoff is mediated by the cleavage of eukaryotic translation initiation factor eIF4G by sequence-specific viral proteases (2A protease in the case of coxsackievirus), several experimental observations are at variance with this view. Thus, the cleavage of other cellular proteins could contribute to the shutoff of host protein synthesis and stimulation of viral protein synthesis. Recent evidence indicates that the highly conserved 70-kDa cytoplasmic poly(A)-binding protein (PABP) participates directly in translation initiation. We have now found that PABP is also proteolytically cleaved during coxsackievirus infection of HeLa cells. The cleavage of PABP correlated better over time with the host translational shutoff and onset of viral protein synthesis than did the cleavage of eIF4G. In vitro experiments with purified rabbit PABP and recombinant human PABP as well as in vivo experiments with Xenopus oocytes and recombinant Xenopus PABP demonstrate that the cleavage is catalyzed by 2A protease directly. N- and C-terminal sequencing indicates that cleavage occurs uniquely in human PABP at 482VANTSTQTM↓GPRPAAAAAA500, separating the four N-terminal RNA recognition motifs (80%) from the C-terminal homodimerization domain (20%). The N-terminal cleavage product of PABP is less efficient than full-length PABP in restoring translation to a PABP-dependent rabbit reticulocyte lysate translation system. These results suggest that the cleavage of PABP may be another mechanism by which picornaviruses alter the rate and spectrum of protein synthesis.  相似文献

Endophytic actinomycetes isolated from Datura stramonium L. was evaluated for its effects against in vitro α-glucosidase inhibition, antioxidant, and free radical scavenging activities. Based on microbial cultural characteristic and 16S rRNA sequencing, it was identified as Streptomyces sp. loyola UGC. The methanolic extract of endophytic actinomycetes (MeEA) shows remarkable inhibition of α-glucosidase (IC50 730.21 ± 1.33 μg/ml), scavenging activity on 2,2-diphenyl-picrylhydrazyl (DPPH) (IC50 435.31 ± 1.79 μg/ml), hydroxyl radical (IC50 350.21 ± 1.02 μg/ml), nitric oxide scavenging (IC50 800.12 ± 1.05 μg/ml), superoxide anion radical (IC50 220.31 ± 1.47 μg/ml), as well as a high and dose-dependent reducing power. The MeEA also showed a strong suppressive effect on rat liver lipid peroxidation. Antioxidants of β-carotene linoleate model system revels significantly lower than BHA. The total phenolic content of the extract was 176 mg of catechol equivalents/gram extract. Perusal of this study indicates MeEA can be used as natural resource of α-glucosidase inhibitor and antioxidants.  相似文献

The activity of 6-(D-α-sulfoaminophenylacetamido)-penicillanic acid was determined against 357 clinical isolates of gram-negative bacilli by use of the tube-dilution technique. The majority of the isolates of Pseudomonas species were inhibited by 200 μg/ml or less of this antibiotic. Most of the isolates of Escherichia coli had a minimal inhibitory concentration of 50 μg/ml or less. Seventy-three per cent of the isolates of P. mirabilis, 40% of the isolates of P. morganii, and 45% of the isolates of Enterobacter species were inhibited by 12.5 μg/ml or less, whereas most of the isolates of Klebsiella species and Serratia species were resistant. The activity of this semisynthetic penicillin was affected by the size of the inoculum. The drug was bactericidal against all isolates of E. coli and Proteus species that were sensitive to it, but it was bactericidal against only 32% of the sensitive isolates of Pseudomonas species.  相似文献

Conclusion There is a class of scholars who seek to write about and perpetuate the culture of all those who do not write about and perpetuate their own culture. Do these scholars write about and perpetuate their own culture?Paul Ricouer and others have put forward the text metaphor for social science production — for what we produce and the way we work with it. It is one of many metaphors that we anthropologists have available to enable us to grasp better our challenging and so often inchoate enterprise. I have expressed misgivings about this metaphor because it takes us back into a world, the academic one, where we do not really belong — with its concerns for distance, invariance, truth value — and away from a world — the variable and voluble world of discourse — where anthropology should be at work if not at home. It is also a metaphor that can beguile us into ignoring the pervasiveness of self-reference and self-replication in our (the intellectual's) creation of worlds and surely in our anthropological creation of other people's worlds. As the pronouns are the fundamental entities of the world of discourse, we have focused on what we can learn from them about the dynamics of self-reference and reference and particularly about the parlous shift from talking to persons — as with the first two persons of discourse — to talking about persons — which is to say the third or absent person of discourse. The pronouns also teach us about turn-taking. Here also one has misgivings about the text metaphor for it takes us into altogether too sui generis a world of the scholar alone with his writings and his imagined or self-created audience for those writings. It is the world of the I perpetually talking and the other perpetually listening. Anthropology in contrast works on the other side of turn-taking where we emphasize the other as an I talking and we ourselves as a you listening. In a world where the powerful military-industrial centers of things do most of the talking, it is altogether appropriate that the peripheries should have their turn through the work of anthropology. That those voices should be heard is a fundamental anthropological task.We should not pretend in the conclusion, however, that the channel for hearing those voices is absolutely clear and unimpeded by self-reference — from the insertion, as it were, of the anthropologist's own voice. We do not escape the need to negotiate these various voices. And for that reason I have placed as an epigraph to the conclusion a rephrasing for the anthropologist of Epimenides' paradox. It is a rephrasing that evokes the paradox involved in giving voice to the voices of others. but to recognize the problem at the very center of our enterprise does not justify the abandonment of that enterprise nor the admirable and unique efforts at turn-taking that characterize the calling of anthropologists.James W. Summary Nothapodytes foetida (Wight) is a small evergreen tree and the extract from this tree is used to make the antileukaemia and antitumoral compound camptothecin. Due to exploitation of this resource, efficient methods for rapid propagation of N. foetida are highly desirable. Multiple shoots were induced on hypocotyl segments of 20–25-d-old seedlings of N. foetida cultured on Murashige and Skoog (MS) medium supplemented with different concentrations and combinations of cytokinins. The highest shoot multiplication was achieved on MS medium containing thidiazuron (TDZ) at the concentration of 2.2 μM. Inhibition of shoot elongation by TDZ was overcome by transferring shoot cultures to medium containing 2.2 μM benzylaminopurine which produced healthy shoots after three additional subcultures. The production of shoots was further promoted by repeated subculturing of original explants on fresh multiplication medium after each harvesting of the newly formed shoots. In vitro rooting was best induced (87%) in shoots excised from proliferated shoot cultures on one-fourth MS medium augmented with 5.7 μM indole-3-acetic acid and 2.4 μM indolebutryic acid (IBA). In vitro-developed shoots were also rooted ex vitro by dipping in 49 μM IBA for 10 min. In vitro- and ex vitro-rooted plants were successfully acclimatized and established in greenhouse conditions.  相似文献

Background

Studies have shown that metabolic syndrome and its consequent biochemical derangements in the various phases of diabetes may contribute to carcinogenesis. A part of this carcinogenic effect could be attributed to hyperinsulinism. High levels of insulin decrease the production of IGF-1 binding proteins and hence increase levels of free IGF-1. It is well established that bioactivity of free insulin growth factor 1 (IGF-1) increases tumor turnover rate. The objective is to investigate the role of insulin resistance/sensitivity in carcinogenesis by studying the relation between insulin resistance/sensitivity and IGF-1 levels in cancer patients. We postulate that hyperinsulinaemia which prevails during initial phases of insulin resistance (condition prior to overt diabetes) increases bioactivity of free IGF-1, which may contribute to process of carcinogenesis.

Methods/Design

Based on our pilot study results and power analysis of the same, we have designed a two group case-control study. 800 proven untreated cancer patients (solid epithelial cell tumors) under age of 50 shall be recruited with 200 healthy subjects serving as controls. Insulin resistance/sensitivity and free IGF-1 levels shall be determined in all subjects. Association between the two parameters shall be tested using suitable statistical methods.

Discussion

Well controlled studies in humans are essential to study the link between insulin resistance, hyperinsulinaemia, IGF-1 and carcinogenesis. This study could provide insights to the role of insulin, insulin resistance, IGF-1 in carcinogenesis although a precise role and the extent of influence cannot be determined. In future, cancer prevention and treatment strategies could revolve around insulin and insulin resistance.  相似文献

Subspecific classification of Asian rice (Oryza sativa L) into indica and japonica has always been a subject of interest althrough for rice breeders and geneticists. The present study aims at identifying subspecies specific microsatellite markers in six genotypes, each of indica and japonica using 372 microsatellite primers covering the entire genome. Only 36 primers gave clear polymorphism on 3% agarose gel and these can be used as diagnostic markers for routine and easy identification of the subspecies.  相似文献

The efficacy of a new bispyridinium oxime 1-(4-hydroxyiminomethylpyridinium)-4-(4-carbamoylpyridinium)butane dibromide, called K048, and currently used oximes (pralidoxime, obidoxime, the oxime HI-6) to reactivate acetylcholinesterase inhibited by various nerve agents (sarin, tabun, cyclosarin, VX) was tested by in vitro methods. The new oxime K048 was found to be a more efficacious reactivator of nerve agent-inhibited acetylcholinesterase than pralidoxime (in the case of VX, tabun and cyclosarin), obidoxime (cyclosarin and tabun) and HI-6 (tabun) but it did not reach the efficacy of currently used oximes for the reactivation of acetylcholinesterase inhibited by sarin. Thus, the oxime K048 seems to be a relatively efficacious broad spectrum acetylcholinesterase reactivator and, therefore, it could be useful for the treatment of a nerve agent-exposed population if information about detection of the type of nerve agent is not available.  相似文献

The aim of this study was to investigate the neuroprotective effects of (2S)-5, 2′, 5′-trihydroxy-7-methoxyflavanone (TMF), a natural product from Abacopteris penangiana (Hook.) Ching, in oxidative stress-induced neurodegeneration models in vitro and in vivo. In PC12 cells, preincubation of TMF (3–20 μM) for 24 h decreased the dopamine-induced toxicity and attenuated the redox imbalance in PC12 cells through regulating the ratio of reduced glutathione/oxidized glutathione (GSH/GSSG), which is a sensitive marker of oxidative stress. Additionally, long-term intraperitoneal (i.p.) injection of TMF (4 or 8 mg/kg/day) for 2 weeks significantly improved the behavioral performance of d-galactose (d-gal) treated mice in a Morris water maze test. Biochemical analysis revealed that TMF inhibited the activation of AP-1 (activator protein-1) and upregulated the level of BDNF (brain derived neurophic factor) as well as the ratio of GSH/GSSG in the hippocampus of d-gal treated mice. Furthermore, western blotting analysis indicated that TMF increased phosphorylation of cAMP-response element-binding protein (CREB). Therefore, the natural product TMF possessed a potential for the treatment of neurodegenerative diseases.  相似文献

α-Tropomyosin (α-TM) has a conserved, charged Asp-137 residue located in the hydrophobic core of its coiled-coil structure, which is unusual in that the residue is found at a position typically occupied by a hydrophobic residue. Asp-137 is thought to destabilize the coiled-coil and so impart structural flexibility to the molecule, which is believed to be crucial for its function in the heart. A previous in vitro study indicated that the conversion of Asp-137 to a more typical canonical Leu alters flexibility of TM and affects its in vitro regulatory functions. However, the physiological importance of the residue Asp-137 and altered TM flexibility is unknown. In this study, we further analyzed structural properties of the α-TM-D137L variant and addressed the physiological importance of TM flexibility in cardiac function in studies with a novel transgenic mouse model expressing α-TM-D137L in the heart. Our NMR spectroscopy data indicated that the presence of D137L introduced long range rearrangements in TM structure. Differential scanning calorimetry measurements demonstrated that α-TM-D137L has higher thermal stability compared with α-TM, which correlated with decreased flexibility. Hearts of transgenic mice expressing α-TM-D137L showed systolic and diastolic dysfunction with decreased myofilament Ca2+ sensitivity and cardiomyocyte contractility without changes in intracellular Ca2+ transients or post-translational modifications of major myofilament proteins. We conclude that conversion of the highly conserved Asp-137 to Leu results in loss of flexibility of TM that is important for its regulatory functions in mouse hearts. Thus, our results provide insight into the link between flexibility of TM and its function in ejecting hearts.  相似文献

Background, aim and scope  

The environmental impact of building products made from heavy metals has been a topic of discussion for some years. This was fuelled by results of life cycle assessments (LCAs), where the emission of heavy metals strongly effected the results. An issue was that the characterisation factors of the Centre for Environmental Studies (CML) 2000 life cycle impact assessment (LCIA) methodology put too much emphasis on the impact of metal emissions. We adjusted Zn characterisation factors according to the most recent insights in the ecotoxicity of zinc and applied them in an LCA using zinc gutters and downpipes as an example.  相似文献

The study examines the occurrence of aflatoxin and ochratoxin A in the !988 dried figs crop. Mycotoxin content, moisture, and aw (water activity) were analyzed in a total of 103 fig samples collected from various orchards and different stages of fig processing. Aflatoxins (B1, B2, G1, and G2) were present in 29% of the samples examined at 0.5–63.0, 0.5–37.7, 0.5–78.3, and 0.5–12.5μ/kg, respectively. Ochratoxin A was detected in only 3% of the samples at 5.2–8.3 μ/kg. The moisture (and aw) values of the fruits were found suitable for mycotoxin formation in firm ripened and shrivelled figs.  相似文献
