The origin of spelt and free-threshing hexaploid wheat   总被引:1,自引:0,他引:1  
It is widely believed that hexaploid wheat originated via hybridization of hulled tetraploid emmer with Aegilops tauschii (genomes DD) and that the nascent hexaploid was spelt, from which free-threshing wheat evolved by mutations. To reassess the role of spelt in the evolution of Triticum aestivum, 4 disomic substitution lines of Ae. tauschii chromosome 2D in Chinese Spring wheat were developed and one of them was used to map the Tg locus, which controls glume tenacity in Ae. tauschii, relative to simple sequence repeat (SSR) and expressed sequence tag loci on wheat chromosome 2D. The segregation of SSR markers was used to assess the presence of Tg alleles in 11 accessions of spelt, both from Europe and from Asia. Ten of them had an inactive tg allele in the D genome and most had an active Tg allele in the B genome. This is consistent with spelt being derived from free-threshing hexaploid wheat by hybridization of free-threshing wheat with hulled emmer. It is proposed that the tetraploid parent of hexaploid wheat was not hulled emmer but a free-threshing form of tetraploid wheat.  相似文献

A partial promoter region of the high-molecular weight (HMW) glutenin genes was studied in two wheat specimens, a 300 year-old spelt (Triticum spelta L.) and an approximately 250 year-old bread wheat (Triticum aestivum L.) from Switzerland. Sequences were compared to a recent Swiss landrace T. spelta ’Oberkulmer.’ The alleles from the historical bread wheat were most similar to those of modern T. aestivum cultivars, whereas in the historical and the recent spelt specific alleles were detected. Pairwise genetic distances up to 0.03 within 200 bp from the HMW Glu-A1-2, Glu-B1-1 and Glu-B1-2 alleles in spelt to the most-similar alleles from bread wheat suggest a polyphyletic origin. The spelt Glu-B1-1 allele, which was unlike the corresponding alleles in bread wheat, was closer related to an allele found in tetraploid wheat cultivars. To investigate the origin of European spelt (Triticum spelta L., genome AABBDD) and its relation to bread wheat (Triticum aestivum L., AABBDD), we analysed an approximately 1-kb sequence, including a part of the promoter and the coding region, of the high-molecular-weight (HMW) glutenin B1-1 and A1-2 subunit genes in 58 accessions of hexa- and tetraploid wheat from different geographical regions. Six Glu-B1-1 and five Glu-A1-2 alleles were identified based on 21 and 19 informative sites, respectively, which suggests a polyphyletic origin of the A- and B-genomes of hexaploid wheat. In both genes, a group of alleles clustered in a distinct, so-called beta subclade. High frequencies of alleles from the Glu-B1-1 and Glu-A1-2 beta subclades differentiated European spelt from Asian spelt and bread wheat. This indicates different origins of European and Asian spelt, and that European spelt does not derive from the hulled progenitors of bread wheat. The conjoint differentiation of alleles of the A- and B-genome in European spelt suggests the introgression of a tetraploid wheat into free-threshing hexaploid wheat as the origin of European spelt.Electronic Supplementary Material Supplementary material is available in the online version of this article at Communicated by J. Intraspecific divergence of hexaploid wheat Triticum spelta was studied by C-banding method in 41 accessions of different geographic origins. The spelt accessions did not differ in karyotype structure or heterochromatin distribution from common wheat, but showed greater intraspecific polymorphism by chromosome rearrangements (translocations, inversions) and banding patterns. On evidence of C-banding patterns, spelt was assumed to occupy an intermediate position between tetraploid and hexaploid wheat species. Accessions of the Asian spelt subspecies had more diverse banding patterns than European accessions. A relatively high frequency of chromosome rearrangements was observed in Iranian accessions. Visual analysis revealed high uniformity of chromosome banding patterns in T. spelta populations of Afghanistan, Spain, and Germany (Bavarian group), suggesting a significant role of the founder effect in their evolution.  相似文献

While many reports on genetic analysis of Fusarium head blight (FHB) resistance in bread wheat have been published during the past decade, only limited information is available on FHB resistance derived from wheat relatives. In this contribution, we report on the genetic analysis of FHB resistance derived from Triticum macha (Georgian spelt wheat). As the origin of T. macha is in the Caucasian region, it is supposed that its FHB resistance differs from other well-investigated resistance sources. To introduce valuable alleles from the landrace T. macha into a modern genetic background, we adopted an advanced backcross QTL mapping scheme. A backcross-derived recombinant-inbred line population of 321 BC2F3 lines was developed from a cross of T. macha with the Austrian winter wheat cultivar Furore. The population was evaluated for Fusarium resistance in seven field experiments during four seasons using artificial inoculations. A total of 300 lines of the population were genetically fingerprinted using SSR and AFLP markers. The resulting linkage map covered 33 linkage groups with 560 markers. Five novel FHB-resistance QTL, all descending from T. macha, were found on four chromosomes (2A, 2B, 5A, 5B). Several QTL for morphological and developmental traits were mapped in the same population, which partly overlapped with FHB-resistance QTL. Only the 2BL FHB-resistance QTL co-located with a plant height QTL. The largest-effect FHB-resistance QTL in this population mapped at the spelt-type locus on chromosome 5A and was associated with the wild-type allele q, but it is unclear whether q has a pleiotropic effect on FHB resistance or is closely linked to a nearby resistance QTL.  相似文献

Various sites in the valley of the Sant Cugat stream in Cerdanyola del Vallès (Catalonia) were subject to systematic archaeobotanical sampling to obtain an overview of the crops and agriculture of the area during the Iron Age and late antiquity. In all cases, the most numerous taxa were crop plants. Among these, cereals were clearly predominant at all sites investigated, especially Hordeum vulgare var. vulgare (hulled barley) and Triticum aestivum/durum (bread or macaroni wheat), both in numbers and frequency. Other cereals, such as Triticum dicoccum (emmer) or Setaria italica (foxtail bristle-grass), were regularly present in considerably lower numbers but in fairly high frequencies. Pulses were much less numerous, although their presence increases in terms of frequency. Among them, clearly the best represented was Lens culinaris (lentil). The results show that the agriculture in the period considered was principally based on winter cereals, with a gradual substitution of hulled barley by bread/hard wheat, accompanied by other cereals of minor importance, led by Triticum dicoccum (emmer), and pulses. The appearance of Vitis (grapevine) in the Iberian period is one of the important characteristics of agriculture in the Iberian world. The allelic compositions of high- and low-molecular-weight subunits of glutenins (HMW-GS and LMW-GS) among European spelt (Triticum spelta L.) and related hexaploid and tetraploid Triticum species were investigated by one- and two-dimensional polyacrylamide-gel electrophoresis (PAGE) and capillary electrophoresis (CE). A total of seven novel glutenin alleles (designated A1a*, B1d*, B1g*, B1f*, B1j*, D1a* at Glu-1 and A3h at the Glu-3 loci, respectively) in European spelt wheat were detected by SDS-PAGE, which were confirmed further by employing A-PAGE and CE methods. Particularly, two HMW-GS alleles, Glu-B1d* coding the subunits 6.1 and 22.1, and Glu-B1f* coding the subunits 13 and 22*, were found to occur in European spelt with frequencies of 32.34% and 5.11%, respectively. These two alleles were present in cultivated emmer (Triticum dicoccum), but they were not observed in bread wheat (Triticum aestivum L.). The allele Glu-B1g* coding for 13* and 19* subunits found in spelt wheat was also detected in club wheat (Triticum compactum L.). Additionally, two alleles coding for LMW-GS, Glu-A3h and Glu-B3d, occurred with high frequencies in spelt, club and cultivated emmer wheat, whereas these were not found or present with very low frequencies in bread wheat. Our results strongly support the secondary origin hypothesis, namely European spelt wheat originated from hybridization between cultivated emmer and club wheat. This is also confirmed experimentally by the artificial synthesis of spelt through crossing between old European emmer wheat, T. dicoccum and club wheat, T. compactum.Communicated by H.F. The total concentrations of free phenolic compounds and peroxidase were determined in spikes (collected at the flowering stage) of some spelt and common wheat cultivars differing in their response to F. culmorum infection. The antifungal activity of methanol extracts obtained from spikes was also evaluated. The tested genotypes differed significantly in their response to inoculation. The most resistant were Torka and Zebra among common wheat cultivars, and Weisser Grannenspelz among spelt cultivars. The average content of free phenolic compounds in spikes of spelt and common wheat was 1246.56 μg g−1 and 1236.58 μg g−1, respectively. The cultivars whose spikes contained the largest amounts of phenols showed the weakest response to F. culmorum infection. No significant differences were observed with regard to peroxidase content, which was 5.22 U g−1 in common wheat spikes and 5.14 U g−1 in spelt spikes. Methanol extracts from spikes of all wheat cultivars contained antifungal substances. The extracts from spelt spikes inhibited the growth of F. culmorum on PDA to a lesser degree than the extracts from common wheat spikes. This corresponds to the results of field trials, in which T. spelta generally exhibited a stronger response to F. culmorum infection than common wheat. The high correlation (r = 0.816) between mycelium growth inhibition on the medium and F. culmorum infection indicates that an evaluation of the antifungal activity of extracts from spikes may be used for the selection of breeding materials directed towards increased resistance to Fusarium head blight.  相似文献

The influence of fungi on seedling emergence of naked and hulled spelt (Triticum spelta L.) and winter wheat (Triticum aestivum L.) was investigated. Seeds were sown in flat trays and placed in a growth chamber under stress conditions (low temperature and water logging) for four weeks, followed by favourable growth conditions. At weekly intervals, 150 seeds were removed, surface sterilized, and investigated for fungi. Pythium aristosporum Vanterpool, a causal agent of damping-off, was found to be best adapted to the unfavourable conditions and to be a major cause of pre-emergence damping-off. The frequency of seed-borne fungi decreased during the stress period, whereas soil-borne fungi survived and could be isolated from seeds and glumes in the following period with more favourable growth conditions. Seedlings from hulled seeds of spelt emerged more frequently than from naked kernels of spelt and both emerged at a higher rate than from seeds of winter wheat. This is good evidence that glumes protect seeds against fungal colonization and therefore increase the fitness of spelt under unfavourable germinating conditions.  相似文献

Domestication of plants and animals is the major factor underlying human civilization and is a gigantic evolutionary experiment of adaptation and speciation, generating incipient species. Wheat is one of the most important grain crops in the world, and consists mainly of two types: the hexaploid bread wheat (Triticum aestivum) accounting for about 95% of world wheat production, and the tetraploid durum wheat (T. durum) accounting for the other 5%. In this review, we summarize and discuss research on wheat domestication, mainly focusing on recent findings in genetics and genomics studies. T. aestivum originated from a cross between domesticated emmer wheat T. dicoccum and the goat grass Aegilops tauschii, most probably in the south and west of the Caspian Sea about 9,000 years ago. Wild emmer wheat has the same genome formula as durum wheat and has contributed two genomes to bread wheat, and is central to wheat domestication. Domestication has genetically not only transformed the brittle rachis, tenacious glume and non-free threshability, but also modified yield and yield components in wheat. Wheat domestication involves a limited number of chromosome regions, or domestication syndrome factors, though many relevant quantitative trait loci have been detected. On completion of the genome sequencing of diploid wild wheat (T. urartu or Ae. tauschii), domestication syndrome factors and other relevant genes could be isolated, and effects of wheat domestication could be determined. The achievements of domestication genetics and robust research programs in Triticeae genomics are of greatly help in conservation and exploitation of wheat germplasm and genetic improvement of wheat cultivars.  相似文献

Summary The cytology of bread wheat (Triticum aestivum) suspension lines, recycled lines (selected for high division frequency) and their dividing protoplasts, have been examined. Extensive numerical and structural chromosome variation was present in all the lines. The most frequently observed chromosome numbers were around 2n=32, indicating that considerable chromosome loss from the normal wheat complement (2n=6x=42) had occurred during selection of the lines. Chromosome aberrations also indicated loss of chromosome arms and chromosome segments. The implications of this variation for studies on transformation and for the potential regeneration of whole plants from protoplasts of bread wheat are discussed.  相似文献

The leaf rust resistance gene Lr47 confers resistance to a wide spectrum of leaf rust strains. This gene was recently transferred from chromosome 7S of Triticum speltoides to chromosome 7A of hexaploid wheat Triticum aestivum. To facilitate the transfer of Lr47 to commercial varieties, the completely linked restriction fragment length polymorphism (RFLP) locus Xabc465 was converted into a PCR-based marker. Barley clone ABC465 is orthologous to the type-I wheat sucrose synthase gene and primers were designed for the conserved regions between the two sequences. These conserved primers were used to amplify, clone and sequence different alleles from T. speltoides and T. aestivum. This sequence information was used to identify the T. speltoides sequence, detect allele-specific mutations, and design specific primers. Cosegregation of the PCR product of these primers and the T. speltoides chromosome segment was confirmed in four backcross-populations. To complement this dominant marker, a cleavage amplified polymorphic sequence (CAPS) was developed for the 7A allele of Xabc465. This CAPS marker is useful to select homozygous Lr47 plants from F2 or backcross-F2 segregating populations, and in combination with the T- speltoides specific primers is expected to facilitate the deployment of Lr47 in new bread wheat varieties. A winter bread wheat accession from the Arsenal collection was genetically examined to study the results of introgression, which substantially changed the physiological and morphological traits of the original spring cultivar Rodina. Apart from its winter habit, the accession was characterized by awned speltoid spikes, suggesting introgression into chromosome 5A, which carries marker genes in the order Vrn-A1-Q-B1. Genetic analysis showed that the chromosome fragment introgressed from Aegilops speltoides recombined well with the homeologous region of bread wheat chromosome 5A in the region between the Vrn-A1 and Q genes. Recombination between the Vrn-A1 and B1 genes was not detected, and it was assumed that the order of the marker genes of chromosome 5A was inverted to produce Q-Vrn-A1-B1. When the winter introgression line was crossed with Triticum spelta L., an interaction of two dominant genes determining the spike character was for the first time detected in F1, increasing the spike length and the number of spikelets, and followed with transgression in F2.  Chromosome 5A of wheat carries major gene loci for agronomic traits including the vernalization requirement (Vrn-A1) and ear morphology (Q). To determine whether the genetic variation for ear emergence time and plant height is attributable to either of these major genes as pleiotropic effects or independent QTL, we combined a RFLP map constructed from 120 recombinant substitution lines derived from a cross between ‘Chinese Spring’ (Cappelle-Desprez 5A) and CS(Triticum spelta 5A) with data collected from field trials over 3 years. For ear emergence time the main effects on flowering time were by Vrn-A1 and QEet.ocs-5A.1, the latter a QTL in the 28.6-cM Xcdo584/Q interval linked to Q by less than 10 cM. The CS(T. spelta 5A) allele at QEet.ocs-5A.1 contributed to an earlier ear emergence time by 2.7–6.0 days, which was approximately equal to the effects of Vrn-A1. For plant height, three QTLs were identified on the long arm and linked in repulsion. The CS(T. spelta 5A) allele at Vrn-A1 or closely linked to Xfba068 contributed to a height reduction of 3.5–6.1 cm, whereas both the Q allele and Qt.ocs-5A.1 allele within the Xcdo1088/Xbcd9 interval from CS(Cappelle-Desprez 5A) produced a shorter plant. When plant height was partitioned into culm length and ear length, the Vrn-A1 allele and CS(Cappelle-Desprez 5A) allele at QCl.ocs-5A.1 within the Xcd1088/Xbcd9 interval were found to contribute to a shorter culm. CS(T. spelta 5A) allele at q was a major determinant of a long ear, together with minor effects at QEl.ocs-5A.1 within the Xcdo1088/Xbcd9 interval. Tibetan semi-wild wheat (Triticum aestivum ssp. tibetanum Shao) is a semi-wild hexaploid wheat resource that is only naturally distributed in the Qinghai-Tibet Plateau. Brittle rachis and hard threshing are two important characters of Tibetan semi-wild wheat. A whole-genome linkage map of T. aestivum ssp. tibetanum was constructed using a recombinant inbred line population (Q1028×ZM9023) with 186 lines, 564 diversity array technology markers, and 117 simple sequence repeat markers. Phenotypic data on brittle rachis and threshability, as two quantitative traits, were evaluated on the basis of the number of average spike rachis fragments per spike and percent threshability in 2012 and 2013, respectively. Quantitative trait locus (QTL) mapping performed using inclusive composite interval mapping analysis clearly identified four QTLs for brittle rachis and three QTLs for threshability. However, three loci on 2DS, 2DL, and 5AL showed pleiotropism for brittle rachis and threshability; they respectively explained 5.3%, 18.6%, and 18.6% of phenotypic variation for brittle rachis and 17.4%, 13.2%, and 35.2% of phenotypic variation for threshability. A locus on 3DS showed an independent effect on brittle rachis, which explained 38.7% of the phenotypic variation. The loci on 2DS and 3DS probably represented the effect of Tg and Br1, respectively. The locus on 5AL was in very close proximity to the Q gene, but was different from the predicted q in Tibetan semi-wild wheat. To our knowledge, the locus on 2DL has never been reported in common wheat but was prominent in T. aestivum ssp. tibetanum accession Q1028. It remarkably interacted with the locus on 5AL to affect brittle rachis. Several major loci for brittle rachis and threshability were identified in Tibetan semi-wild wheat, improving the understanding of these two characters and suggesting the occurrence of special evolution in Tibetan semi-wild wheat.  相似文献

Abstract

The feasibility to switch from line to hybrid breeding is currently a hot topic in the wheat community. One limitation seems to be the lack of divergent heterotic groups within wheat adapted to a certain region. Spelt wheat is a hexaploid wheat that can easily be crossed with bread wheat and that forms a divergent genetic group when compared to elite bread wheat. The aim of this study was to investigate the potential of Central European spelt as a heterotic group for Central European bread wheat. We performed two large experimental field studies comprising in total 43 spelt lines, 14 wheat lines, and 273 wheat–spelt hybrids, and determined yield, heading time, plant height, resistance against yellow rust, leaf rust, and powdery mildew, as well as protein content and sedimentation volume. Heterosis of yield was found to be lower than that of hybrids made between elite wheat lines. Moreover, heterosis of the quality trait sedimentation volume was negative. Consequently, spelt wheat does not appear suited to be used as heterotic group in hybrid wheat breeding. Nevertheless, high combining abilities of a few spelt lines with elite bread wheat lines make them interesting resources for pre-breeding in bread wheat. Thereby, the low correlation between line per se performance and combining ability of these spelt lines shows the potential to unravel the breeding value of genetic resources by crossing them to an elite tester.

Barley yellow dwarf is the most damaging virus-caused disease in bread wheat (Triticum aestivum L.). A resistant line, SW335.1.2-13-11-1-5 (2n = 47), derived from a cross of T. aestivum x Lophopyrum ponticum was characterized by meiotic chromosome pairing, by in situ DNA hybridization and by expression of molecular markers to determine its chromosome constitution. All progeny of this line had three pairs of L. ponticum chromosomes from homoeologous chromosome groups 3, 5, and 6 and the 2n = 47 progeny had an additional L. ponticum monosome. The pairs from groups 3 and 6 were in the added state, while the group 5 pair was substituted for wheat chromosome 5D. Several wheat-wheat translocations with respect to the parental wheat genotype occurred in this line, presumably owing to the promotion of homoeologous chromosome pairing by L. ponticum chromosomes. It was hypothesized that homoeologous recombination results in homoeologous duplication-deletions in wheat chromosomes. An aberrant 3:1 disjunction creates the potential at each meiosis for replacement of these wheat chromosomes by homoeologous L. ponticum chromosomes. Wheat chromosomes 3A and 6A appeared to be in intermediate stages of this substitution process.  相似文献

以36份云南铁壳麦为试验材料,采用SDS-PAGE法分析了Glu-1位点编码的高分子量麦谷蛋白亚基(HMW-GS)及组成。结果表明,在Glu-A1位点上检测到3种(N,2*和1)亚基类型,Glu-B1位点上共检测到5种(7、7 8、17 18、13 16和6 8)亚基类型,Glu-D1位点上只检测到1种(2 12)亚基类型。共检测到6种亚基组成类型,即:N、7、2 12,N、7 8、2 12,2*、7 8、2 12,2*、17 18、2 12,1、6 8、2 12和1、13 16、2 12。云南铁壳麦的HMW-GS为普通小麦已知变异类型的18%,3个位点的Nei's遗传变异系数顺序为Glu-B1(0.5734)>Glu-A1(0.2484)>Glu-D1(0),表明云南铁壳麦属较原始类型,Glu-D1位点未发生变异。品质评分最高分为8分(3份材料),平均为5.2分。同时86%的云南铁壳麦具有适合制作优质手工馒头的高分子量麦谷蛋白亚基(N和2 12),42%的云南铁壳麦具有亚基组成类型(1、7 8、2 12和N、7 8、2 12),这些材料可作为云南小麦馒头品质改良的材料。  相似文献
