The circadian systems of rodents respond to light pulses presented during the subjective night with phase shifts and altered cellular activity in the suprachiasmatic nuclei (SCN), including expression of immediate-early genes (IEGs) such as c-fos. A recent study showed that a nonphotic stimulus (an air disturbance generated by a fan) that does not normally induce the expression of c-fos-like immunoreactivity in the SCN of rats can be made to do so after being paired repeatedly with a light pulse in a Pavlovian conditioning paradigm. Furthermore, after conditioning (but not after noncontingent exposure to these stimuli), the fan also induced phase shifts in activity and body temperature rhythms comparable to those produced by light. The authors performed three experiments designed to replicate and extend these findings in rats. In experiment 1, rats were tested for conditioning effects of repeated pairings of a light pulse with a neutral air disturbance under a full photoperiod. In experiment 2, a modified conditioning paradigm was used in which a skeleton photoperiod served as both the entraining zeitgeber and the unconditioned stimulus. Animals in the paired and unpaired training conditions were exposed to both the light pulse and the air disturbance, but the air disturbance signaled the onset of light in the paired condition only. Phase shifts of wheel-running activity rhythms and gene expression in the SCN, intergeniculate leaflet, and paraventricular nucleus of the thalamus were assessed in animals following either of the training conditions or the control procedures. Experiment 3 assessed whether the air disturbance could entrain the circadian activity rhythms of rats with or without previous pairing with light in a classical conditioning paradigm. No evidence for classical conditioning, nor for unconditioned effects of the air disturbance on the circadian system, was found in these studies.  相似文献

Blood samples were taken from conscious, chronically-catheterized rats during parturition for measurement of oxytocin by specific radioimmunoassay. After the birth of the 3rd pup, rats were allowed to remain in their nesting cage (undisturbed rats) or were transferred for 45 min to a glass bowl (disturbed rats); at the time of transfer, rats were given an i.v. injection of the opioid antagonist naloxone or saline vehicle. Subsequent parturition was prolonged in saline-treated disturbed rats, but not in naloxone-treated disturbed rats. Parturition was significantly hastened in naloxone-treated undisturbed rats. Naloxone injections were followed by a large rise in plasma oxytocin concentrations in disturbed and undisturbed rats. We conclude, from a statistical analysis of the relationship within experimental groups between plasma oxytocin concentration and speed of parturition, that the effects of disturbance and of naloxone upon parturition may be accounted for, at least in part, by their effects upon oxytocin release. However, the effects of disturbance on parturition may not be mediated entirely by activation of opioid pathways. Naloxone did not potentiate oxytocin release in non-pregnant rats, or on Day 1 post partum, but did potentiate oxytocin release on Day 22 of pregnancy even in rats before the onset of parturition. Endogenous opioid pathways regulating oxytocin release therefore appear to be active during late pregnancy and during parturition itself.  相似文献

Previously we reported that the heart norepinephrine concentration was markedly increased in diabetic rats. To further study the relationship between a disturbance in the autonomic nervous system and catecholamine metabolism in diabetes mellitus, the plasma catecholamine response to stress and catecholamine concentration of heart and adrenals were measured. Wistar male rats were made diabetic by streptozotocin and kept for 13 weeks. A silicon catheter was placed in the superior V. cava 1 week prior to the experiment. Insulin was injected subcutaneously for 3 days once daily. After an overnight fast and without anesthesia, 1 ml of blood, a control sample, was obtained and then the animals were exsanguinated. The blood was mixed with 1 mM EGTA at a final concentration and centrifuged. The tissue was homogenized with 0.4 N perchloric acid containing 1 mM EGTA and centrifuged at 10,000 x g for 20 minutes. Catecholamines were determined by high performance liquid chromatography. Normal rats responded to blood withdrawal stress, and plasma catecholamines were markedly increased, but almost no increase or an actual decrease was observed in diabetic rats. These abnormal responses were improved by insulin treatment. Heart norepinephrine was increased significantly in the diabetic rats compared with the control rats and was reduced significantly by insulin injections. Adrenal epinephrine was also significantly increased in the diabetic rats compared with the control rats, but was not significantly reduced by insulin. These result suggest a possible disturbance of catecholamine secretion in the diabetic rats.  相似文献

The method of immunization with hippocampal tissue cytosol was employed for functional disturbance of hippocampus in newborn and mature rats. This influence resulted in behavioral disorders resembling the hippocampectomy consequences. There were ++non-decremental motor activity and rearing in "open field" test and an improvement in acquisition of active avoidance response in adult rats (250-300 g). In young animals the decreased levels of motor and exploratory activity in "open field" and difficulties in active avoidance learning (in 40 days) were the main features of hippocampal disturbance. In case of immunization with neocortical cytosol no significant alterations in behavior were revealed.  相似文献

Pronouncement of stress-induced disturbance of searching behaviour (using "open field" test) and stomach ulceration were compared for the first time with activity of the catecholamine system in hypothalamus and striatum and also with activity of the stress-limiting system of nitric oxide (NO) in the rats of two strains August and Wistar, which differ in their resistance against stress-induced cardiovascular disorders. The effect of prior adaptation to hypobaric hypoxia on these disorders was also studied. August rats appeared to be more resistant than Wistar rats against stress-induced disturbance of the searching behaviour and stomach ulceration. Results of measuring the content of catecholamines in brain structures and the content of NO stable metabolites nitrate/nitrite in plasma suggested that these differences could be due to the stress activation of the nigro-striatal dopaminergic system in August rats, which was not observed in Wistar rats, and also to the higher production of NO in August than in Wistar rats. Adaptation to hypoxia considerably restricted these stress disorders in rats of both strains. Importantly, the protective effects were associated with activation of the nigro-striatal dopaminergic system in all the animals. In the result, adapted Wistar rats, as distinct from non-adapted Wistar rats, displayed a stress activation of this system. The protective effects of adaptation were also accompanied by an increased NO synthesis. Taken together, the data suggest an important role of the responsiveness of the brain dopaminergic system and NO system in the mechanism of resistance against stress-induced disturbances.  相似文献

Alterations of the expression of some peptidases in the pituitary gland of a fatigued rat model were identified. Rats were kept in a cage filled with water to a height of 1.5 cm to disturb deep sleep. After 24-h sleep disturbance, expression of neutral endopeptidase 24.11 (neprilysin) mRNA was increased in the intermediate lobe of the pituitary gland, whereas the mRNA expression of another family member, damage-induced neuronal endopeptidase, which is normally expressed in a subgroup of anterior pituitary cells, was significantly suppressed. These alterations were demonstrated by RT-PCR, northern blotting and in situ hybridization. Other family members, such as neprilysin 2 and endothelin converting enzyme-1, did not show any change in mRNA expression. An increase of neprilysin mRNA expression was not seen in any other tissues of the sleep-disturbed rats. The enzymatic activity of neprilysin was also increased in the pituitary. The augmentation of neprilysin expression and activity was prolonged as long as the sleep disturbance continued (up to 5 days), and returned to the basal level when rats were allowed to sleep freely. These results suggest that peptide processing and degradation in the pituitary may be an influential factor in fatigued states such as sleep disturbance.  相似文献

Activities of digestive enzymes (maltase, alkaline phosphatase, aminopeptidase M, and glycyl-L-leucine dipeptidase) in small and large intestine, liver, and kidney were studied in rats of different ages kept at the period of lactation under conditions of the standard (8 individuals per litter) and low (3 individuals) number of pups per litter. The low-protein diet for 10 days at once after weaning was found to change the mass of organs and their digestive enzyme activities in all studied rat groups. The revealed changes were more prominent in rats kept under conditions of excessive breast feeding. In adult animals of this group, distribution of the alkaline phosphatase activity along the small intestine differed from that in control rats. The obtained results seem to confirm that any disturbance of the nutrition quality in early ontogenesis leads to disturbance of the «metabolic programming of enzyme systems» of digestive organs.  相似文献

The content of lipids and lipoproteins was determined in the blood plasma of Campbell rats with inherited retina degeneration and Wistar rats used as control. It was shown that in the 30-day old Campbell rats the content of cholesterol in high density lipoproteins sufficiently exceeded that of control. When the distribution of lipoproteins particles in the density gradient was studied after ultracentrifugation, some "anomalous" particles were revealed in the 30-45 day old rats with inherited retina degeneration, which with respect to the flotation rate occupy intermediate position between the low and high density lipoproteins. The obtained data show that the development of inherited retina degeneration in rats involves a disturbance, of the lipoprotein spectrum of blood plasma.  相似文献

目的研究不同性别大鼠在2型糖尿病造模过程中的成功率及模型的稳定性。方法高糖高脂饮食联合腹腔注射小剂量链脲佐菌素诱导建立雄、雌性大鼠2型糖尿病模型。成模后所有大鼠每周固定时间测血糖和体重。观察24周后,心脏穿刺取血,测定空腹血糖(FPG)、血清胰岛素(FINS)、HbA1c、甘油三脂(TG)、胆固醇(TC)、高密度脂蛋白-胆固醇(HDL-C)、低密度脂蛋白-胆固醇(LDL-C)。结果单纯高糖高脂饮食喂养,雄、雌性大鼠血糖与正常组无显著差异;STZ注射后,雄性大鼠血糖升高并逐渐平稳,而雌性需两次STZ注射,模型才比较稳定。实验结束时,雄、雌性糖尿病大鼠FPG、FINS、HOMA-IR以及TG、TC、LDL-C均显著升高,说明模型存在胰岛素抵抗和脂代谢紊乱。结论高糖高脂饲料加一次性小剂量链脲佐菌素腹腔注射,可成功建立雄性大鼠2型糖尿病模型,而同等剂量,雌性模型需两次STZ。雄、雌性糖尿病大鼠模型具有高血糖、脂质代谢紊乱和胰岛素抵抗特点。造模成功率及稳定性与性别有关,雄性大鼠较雌性大鼠成模率高,稳定性好,且耗时更短。  相似文献

A group of rats with a stereotype of conditioned reflexes was preliminarily trained to hypoxia effects during 30 days (at the "altitude" of 6000 m, time of exhibition--from 10 to 60 min, for 18 days--only 60 min). Adaptive changes in the process of training consisted in a weakening of differentiation inhibition, partial amnesia of the conditioned reaction of active avoidance and appearance of phasic states (equalization and paradoxical phases) in the cerebral cortex. The following adaptation of hypoxia "trained" rats to new natural conditions of Alpine altitude (3200 m) proceeded favourably, without disturbance of differentiation inhibition and without phasic states. Rats without preliminary training to altitude chamber hypoxia, in mountains (3200 m) were subjected to moderate tension resulting in protective inhibition, partial amnesia and transient disturbance of differentiation inhibition.  相似文献

Glycogen phosphorylase (PHO) activity was demonstrated histochemically in unfixed cryostat sections of placentae from cadmium-treated and control rats with the use of the semipermeable membrane technique. Staining of the newly synthesized glycogen was performed by lugol. A high activity was present in glycogen cells, spongiotrophoblast and visceral yolk sac from cadmium-treated and control animals. A low but distinct activity could be demonstrated in placental labyrinth from control rats in late pregnancy. Cadmium-exposed rats showed a considerably higher activity in the labyrinth during this period of pregnancy. The elevated PHO activity and concomitant higher glycogen content indicate a disturbance by exposure to cadmium of placental carbohydrate metabolism from day 18 onwards.  相似文献

The role of sialidase in the depletion of glomerular sialic acid induced by diabetes has been investigated in uninephrectomized rats. Four months after streptozotocin administration, diabetic rats showed an enhanced urinary excretion of albumin and transferrin, which was associated with a decrease of sialic acid concentration in isolated glomeruli. Despite the sialic acid depletion, the glomerular sialidase activity was unchanged. These results indicate that the decreased glomerular sialic acid concentration observed in diabetic nephropathy might be caused by a disturbance of the sialylation of glomerular structures.  相似文献

The effect of prednisolone injections during the early postnatal ontogenesis on the diurnal rhythm of corticosteroid function and stress reactivity in adult animals has been studied in rats. The injections of prednisolone to 7--9 days old rats resulted in the subsequent disturbance of diurnal rhythmicity in the suprarenal cortex functioning: the diurnal fluctuations of the content of 11-oxycorticosteroids in the blood plasma are smoothed out; the diurnal changes in the hormonal reaction to the stress stimulation disappear. This disturbance appears to be due to changes in the central mechanisms of the control of hypophysis-suprarenal system. The reactivity of this system per se suffered no changes.  相似文献

During the last one-third of their gestational period, lathyritic rats incorporate significantly lower amounts of Sr85 as revealed by whole body counting techniques. The effects of bannertail kangaroo rat (Dipodomys spectabilis) mounds and associated soil-surface disturbance on plant species composition and diversity in the Chihuahuan Desert were examined with multivariate analysis. Kangaroo rat mounds created disturbance gaps and contributed to local species diversity by creating microhabitats that supported unique plant communities. These microhabitats supported populations of species that were relatively rare in surrounding areas. The diversity observed at the whole habitat level resulted from (1) local spatial heterogeneity, because the mounds offered microenvironments with distinctive nutrient, water, and light conditions; and (2) local patterning of disturbance, because the digging and traffic of the kangaroo rats maintained high levels of soil disturbance at and near the mounds. At a finer scale, species diversity was highest in the area immediately adjacent to active and inactive mounds, and was lower on both the highly disturbed soil of the mounds and in the relatively undisturbed area between mounds. Lowest species diversity occurred on inactive mounds. Annual plant biomass was much greater on mounds than in inter-mound areas. The results support the predictions that intermediate levels of disturbance and small-scale environmental heterogeneity contribute to supporting high species diversity.  相似文献

There was studied effect of severe hypobaric hypoxia and subsequent reoxygenation on level and dynamics of lipid peroxidation in membranes of neocortex cells and in mitochondriaenriched neocortex fraction of non-preconditioned rats and of rats preconditioned thrice with a moderate hypobaric hypoxia. The threefold hypoxic preconditioning increasing brain resistance has been shown to significantly prevent disturbance of lipid peroxidation processes in neocortex—one of the most hypoxia-sensitive brain structures—and to modify development of these processes in mitochondria.  相似文献

It was shown in our physiological study that angiotensin-II induced drinking behavior was changed in previously stress exposed rats as compared to control animals. In order to determine the structural basis of the data obtained in the morphology and histochemistry (nucleic acids concentration) of the nucleus paraventricularis (n. PV), nucleus supraopticus (n. SO) and subfornical organ (SFO) neurons were studied in rats of several experimental groups. The most pronounced changes manifested the shape increase of the neurons functional activity after angiotensin-II (A-II) infusion into control and especially into previously stressed rats were discovered in n. PV. It was supposed that the disturbance of A-II-induced drinking behavior in stressed rats might be mainly due to the disfunction of n. PV neurons.  相似文献
