Measurements of internal gas phase CO2 concentration, stomatal resistance, and acid content were made in Crassulacean acid metabolism plants growing under natural conditions. High CO2 concentrations, sometimes in excess of 2%, were observed during the day in a range of taxonomically widely separated plants (Opuntia ficus-indica L., Opuntia basilaris Engelm. and Bigel., Agave desertii Engelm., Yucca schidigera Roezl. ex Ortiges, Ananas comosus [L.] Merr., Aloe vera L., Cattleya sp. and Phalanopsis sp.) and below ambient air concentrations were observed at night.  相似文献

The changes in the amount, rale of synthesis and the nucleotide composition of different RNA fractions in excised barley leaves floated on water or kinetin (10 mg/l) in the dark were examined. In excised leaves floated on water all nucleic acid components declined and these declines were retarded by kinetin. Barley leaves floated on water showed a stimulation of 32P incorporation into various RNA fractions within 48 hours followed by a decline after 96–144 hours. The leaves floated on kinetin, however, showed an even higher incorporation of 32P into UNA by 48 hours which remained at a comparatively higher level throughout the experiment. In spite of the above changes in RNA synthesis significant differences in the 32P sucrose gradient profiles or in the 32P nucleotide composition of UNA from water and kinetin floated leaves were not noted. The results of this study show that important changes in nucleic acid metabolism occur during the early stages of leaf senescence and that alterations in nucleic acid metabolism during senescence and during kinetin treatment may involve quantitative and only subtle qualitative changes.  相似文献

Carbohydrates stored during deacidification in the light were examined in 11 Crassulacean acid metabolism (CAM) species from widely separated taxa grown under uniform conditions. The hypothesis that NAD(P) malic enzyme CAM species store chloroplastic starch and glucans, and phosphoenolpyruvate carboxykinase species store extrachloroplastic sugars or polymers was disproved. Of the six malic enzyme species examined, Kalanchoe tubiflora, Kalanchoe pinnata, Kalanchoe daigremontiana, and Vanilla planifolia stored mainly starch. Sansevieria hahnii stored sucrose and Agave guadalajarana did not store starch, glucose, fructose, or sucrose. Of the five phosphoenolpyruvate carboxykinase species investigated, Ananus comosus stored extrachloroplastic carbohydrate, but Stapelia gigantea, Hoya carnosa, and Portea petropolitana stored starch, whereas Aloe vera stored both starch and glucose. Within families, the major decarboxylase was common for all species examined, whereas storage carbohydrate could differ both between and within genera. In the Bromeliaceae, A. comosus stored mainly fructose, but P. petropolitana stored starch. In the genus Aloe, A. vera stored starch and glucose, but A. arborescens is known to store a galactomannan polymer. We postulate that the observed variation in carbohydrate partitioning between CAM species is the result of two principal components: (a) constraints imposed by the CAM syndrome itself, and (b) diversity in biochemistry resulting from different evolutionary histories.  相似文献

The role of NAD-malic enzyme (NAD-ME) in the Crassulacean acid metabolism plant Bryophyllum (Kalanchoe) fedtschenkoi was investigated using preparations of intact and solubilized mitochondria from fully expanded leaves. Intact, coupled mitochondria isolated during the day or night did not differ in their ability to take up [14C]malic acid from the surrounding medium or to respire using malate or succinate as substrate. However, intact mitochondria isolated from plants during the day decarboxylated added malate to pyruvate significantly faster than mitochondria isolated from plants at night. NAD-ME activity in solubilized mitochondrial extracts showed hysteretic kinetics and was stimulated by a number of activators, including acetyl-coenzyme A, fructose-1,6-bisphosphate, and sulfate ions. In the absence of these effectors, reaction progress curves were nonlinear, with a pronounced acceleration phase. The lag period before a steady-state rate was reached in assays of mitochondrial extracts decreased during the photoperiod and increased slowly during the period of darkness. However, these changes in the kinetic properties of the enzyme could not account for the changes in the rate of decarboxylation of malate by intact mitochondria. Gel-filtration experiments showed that mitochondrial extracts contained three forms of NAD-ME with different molecular weights. The relative proportions of the three forms varied somewhat throughout the light/dark cycle, but this did not account for the changes in the kinetics behavior of the enzyme during the diurnal cycle.  相似文献

水分胁迫期间、盆栽桃幼树叶片中淀粉和蔗糖含量降低,山梨醇、葡萄糖和果糖则升高;淀粉酶、6-磷酸醛糖还原酶(A6PR)、酸性转化酶(AI)和蔗糖合酶(SS)涤性升高,ADPG焦磷酸化酶(ADPGPPase)、中性转化酶(NI)、山梨醇脱氢酶(SDH)和磷酸蔗糖合酶(SPS)的活性降低。胁迫解除后,淀粉和葡萄糖含量恢复,山梨醇和果糖仍高于对照;A6PR、SDH和ADPGPase活性维持在原水平上;淀粉酶活性恢复;SPS活性增加而AI活性降低。  相似文献

Evidence is presented that the growth of cultured wheat roots is enhanced in closed culture systems not only by carbon dioxide but by a further and apparently very volatile product of root metabolism. The formation of this second growth-stimulating substance is not considered to depend upon a reduction of oxygen tension in the culture system. Unlike ethanol the activity of the volatile promoter does not depend upon illumination of the cultures. The volatile promoter cannot be effectively absorbed by a number of gas absorbents. Attempts to demonstrate ethylene production by the roots have been unsuccessful.  相似文献

In this paper we report for the first time the occurrence ofan inducible weak CAM in leaves of Talinwn triangulare (Jacq.)Willd. This plant is a terrestrial perennial deciduous herbwith woody stems and succulent leaves which grows under fullexposure and in the shade in northern Venezuela. Plants grownin a greenhouse (‘sun’ plants) and a growth cabinet(‘shade’ plants) with daily irrigation showed CO2uptake only during the daytime (maximum rate, 4?0 µmolm–2 s–1) and a small acid accumulation during thenight (6?0 µmol H+g–1 FW). Twenty-four hours aftercessation of irrigation, no CO2 exchange was observed duringpart of the night. Dark fixation reached a maximum (1?0 µmolCO2 m–2 s–1, 100 µmol H+ g–1 FW) onday 9 of drought. By day 30 almost no gas exchange was observed,while acid accumulation was still 10 µmol H+ g–1FW. Rewatering reverted the pattern of CO2 exchange to thatof a C3 plant within 24 h. Daytime and night-time phosphoenolpyruvatecarboxylase activity increased up to 100% (shade) and 62% (sun)of control values after 10 and 15 d of drought, respectively.Light compensation point and saturating irradiance were similarin well-watered sun and shade plants, values being characteristicof sun plants. CAM seems to be important for the tolerance ofplants of this species to moderately prolonged (up to 2 months)periods of drought in conditions of full exposure as well asshade, and also for regaining high photosynthetic rates shortlyafter irrigation. Aeration of root culture medium with air containing 5 % or 10 % CO2 enhances growth, particularly lateral initiation and growth, compared with aeration wilh air or CO2-free air. This beneficial effect of enhanced CO2 pressure on the growth of cultured roots of Atson Elite 56 wheat ean also be demonstrated in presence of a high (40 %) and inhibiting level of oxygen. 14C labelling from a carbonate–bicarbonate solution of pH 7.0 by cultured wheat roots is enhanced by light and this labelling is not inhibited by the presence of 1 % ethyl urethane.  相似文献
