Granulomatous inflammation is a specific type of chronic inflammation in which macrophages and T-cell-mediated immunity to the inciting agent play a pivotal role. In the present study, granulomatous hepatitis was induced in rats by the administration of a single intravenous dose of porcine intestinal alkaline phosphatase. The cellular composition of the hepatic granulomas was analyzed in-situ with a number of recently developed mouse anti-rat monoclonal antibodies to cells of the monocyte-macrophage lineage and lymphocyte subsets. Well-developed granulomas consisted of aggregates of macrophages with central modification into epithelioid cells, a peripheral rim of T- and B-lymphoid cells, including considerable numbers of immunoblasts and plasma cells. In addition, the periphery of the granulomas contained many fat storing cells, a sinusoidal cell type thought to play a central role in hepatic fibrosis. Moreover, intense immunostaining for the extracellular matrix proteins fibronectin and collagen type III was observed at the periphery of the lesions. The granulomas persisted for long periods without eliciting liver cirrhosis. Alkaline phosphatase induced hepatic granulomas in the rat may help to elucidate the contribution of cells of the B-lineage to chronic granulomatous inflammation.  相似文献

The epithelioid granulomas, interstitial and intra-alveolar mononuclear inflammatory infiltrates and the cellular compartments obtained by broncho-alveolar lavage (BAL) were measured in 40 patients with pulmonary involvement of sarcoidosis. The granulomas were divided into a central (epithelioid-cellular) zone and into a peripheral (lymphocytic-fibrotic) zone, and the density of various inflammatory cells was measured in both compartments. The cases were grouped and analyzed according to the radiological and clinical stages. The results are as follows: neutrophilic granulocytes seen in the BAL probably originate from the sarcoid granulomas in patients with stage 1 of the disease and may derive from other compartments of the lung parenchyma in patients with stage 2 or stage 3. Lymphocytes seen in the BAL of patients with stage 1 or stage 2 of the disease probably derive from intra-alveolar lymphocytic agglutinations. They originate from the sarcoid granulomas only in patients with stage 3. Macrophages seen in the BAL probably derive from the sarcoid granulomas independent from the stage of sarcoidosis. No relationship was found between the morphometric parameters and the clinical outcome of the disease.  相似文献

We have observed in hepatic periovular granulomas of C3H mice infected with Schistosoma mansoni, in the chronic phase of the disease (12-19 weeks of infection), groups of early precursors and immature eosinophil granulocytes corresponding, at the ultrastructural level, to promyelocytes and myelocytes. Mitosis was also seen in eosinophil myelocytes. These eosinophil myeloid foci were observed in close contact with macrophages and epithelioid cells, and they were surrounded by an extracellular matrix, rich in collagen fibres. These morphological observations give support to the concept of a peripheral proliferation of eosinophils in chronic schistosomiasis, mediated by a factor secreted by macrophages present in granulomas.  相似文献

Summary The normal murine thymus was examined by lightand electron microscopy to determine the distribution and morphology of extracellular cystic cavities. Most cavities were confined to the cranial half of each gland, situated at the junction between cortex and medulla. They varied in size and shape, and gave rise to narrow channels that coursed to the capsular surface of the gland. Large cavities could be divided into three zones. A short cranial zone exhibited gland-like features, consisting of cells lining a clear lumen. A central zone was lined by a diverse population of cells. Some possessed secretory granules, while others exhibited an apical ciliated border. Lining cells interdigitated with each other and were joined laterally by intercellular junctions. The lumen of the central zone contained lymphocytes and macrophages in an amorphous extracellular matrix. The caudal zone of each cavity had an attenuated and incomplete cellular lining, communicating directly with the surrounding thymic parenchyma. Thymic cavities may represent the initial part of the efferent lymphatic system of the gland, beginning in the tissue spaces at the corticomedullary junction. Selected cells could then enter and interact with the luminal contents in the central zone of the cavity. Ciliated cells may then propel lymphocytes and secretions into the narrow channels radiating from the uppermost part of the chamber, leaving a cell-free lumen in this region. These cavities may function in sequestering lymphocytes, macrophages and thymic secretions before their exit from the gland.  相似文献

The role of neutrophils in tuberculosis (TB), and whether neutrophils express granzyme B (grzB), a pro‐apoptotic enzyme associated with cytotoxic T cells, is controversial. We examined neutrophils in peripheral blood (PB) and lung granulomas of Mycobacterium tuberculosis‐infected cynomolgus macaques and humans to determine whether mycobacterial products or pro‐inflammatory factors induce neutrophil grzB expression. We found large numbers of grzB‐expressing neutrophils in macaque and human granulomas and these cells contained more grzB+ granules than T cells. Higher neutrophil, but not T cell, grzB expression correlated with increased bacterial load. Although unstimulated PB neutrophils lacked grzB expression, grzB expression increased upon exposure to M. tuberculosis bacilli, M. tuberculosis culture filtrate protein or lipopolysaccharide from Escherichia coli. Perforin is required for granzyme‐mediated cytotoxicity by T cells, but was not observed in PB or granuloma neutrophils. Nonetheless, stimulated PB neutrophils secreted grzB as determined by enzyme‐linked immunospot assays. Purified grzB was not bactericidal or bacteriostatic, suggesting secreted neutrophil grzB acts on extracellular targets, potentially enhancing neutrophil migration through extracellular matrix and regulating apoptosis or activation in other cell types. These data indicate mycobacterial products and the pro‐inflammatory environment of granulomas up‐regulates neutrophil grzB expression and suggests a previously unappreciated aspect of neutrophil biology in TB.  相似文献

Summary A continuous murine cell line (GRX) was obtained from fibrotic granulomas induced in C3H/HeN mice liver by experimental infection withSchistosoma mansoni. This anchorage-dependent line produces composite connective tissue/extracellular matrix, displays morphological characteristics of myofibroblasts, and can, under appropriate conditions, accumulate fat droplets. GRX cells produce viral particles of retrovirus type. We consider GRX cell line to be representative of liver connective tissue cells, responsible for fibroplasia in liver fibrotic and granulomatous reactions. Summary The maculae utriculi and sacculi of the inner ear from the European roach (Rutilus rutilus) were investigated by transmission electron microscopy. The stereovilli of peripherally and centrally located sensory cells differ in several features that suggest a developmental gradient. The stereovilli of the peripheral sensory cells, shown to be differentiating cells by other research groups, are short and less steeply graded in height than in central hair cells. All stereovilli in both kinds of hair bundles are interconnected. In the central bundles of stereovilli basal, tip, and vertical connectors are separated by unconnected regions. In contrast, filaments and sometimes other additional structures connect the stereovilli of peripheral bundles over their entire length, but vertical connectors are usually absent. Osmiophilic material occurring inside peripheral stereovilli is interpreted to be monomeric actin. Central and peripheral hair bundles also differ in their reaction to ruthenium red and cationized ferritin. Only the stereovilli of the central cells can be fused by these polycations. Ruthenium red also discriminates between supporting and sensory cells indicating differences in amount or distribution of extracellular material. Hair bundles, intermediate in properties and position between central and peripheral sensory cells, were also found, so that it became possible to propose a scheme of developmental steps leading from microvilli or microvillus-like stereovilli to the fully differentiated hair bundle.  相似文献

Summary The cytotrophoblastic cell columns and trophoblastic shell of macaque placentas accumulate progressively greater amounts of intercellular material during early gestation. We studied the composition of this material in placentas collected from 22–34 days of gestation by using immunoperoxidase techniques directed to the extracellular matrix molecules fibronectin, type IV collagen, and laminin. These antigens co-localized within the intercellular deposits at all stages studied. At day 22 the proximal cell columns were composed of cells with narrow interstices and which lacked immunoreactivity for the 3 antigens. Distally the cells were vacuolated and the intercellular spaces increased in size and contained dense matrix deposits. The trophoblastic shell consisted of closely packed, non-vacuolated cytotrophoblast cells with only a delicate meshwork of matrix. By day 27 the matrix deposits of the distal cell columns increased markedly in size. The trophoblastic shell contained larger numbers of vacuolated cells and was occupied by accumulations of matrix. By 34 days the matrix deposits of the cell columns expanded substantially along the longitudinal axes of the columns. These deposits were often continuous with a matrix-dense, cell-deficient layer in the trophoblastic shell. This matrix-rich zone lay between a cellular layer adjacent to the intervilous space and a similar, but discontinuous, cell layer that formed the junctional zone with the endometrium.  相似文献

Cellular changes in the shoot apical meristem of Helianthus annuus L. have been investigated in relation to its progress towards flowering. During the strictly vegetative phase, lasting for 6–7 days from sowing, mitotic divisions were confined to the peripheral zone, while the central mother cells zone proper, together with the distal cells positioned above the mother zone and also the central portion of the tunica, were relatively quiescent. Mitotic activity increased in the distal cells zone on day 8 and reached the level of that in the peripheral zone by day 12. This was accompanied by an enlargement of this zone and the consequent recession of the mother zone away from the central tunica. At the same time there was a substantial increase in the amount of cytoplasm in the cells of the central tunica. Mitotic activity in the central tunica began on day 12 and reached a peak on day 16. This zone then lost its distinct entity and was replaced by a uniform dome-shaped meristematic layer that became apparent by day 16. The cells of the mother zone remained quiescent during the transition period.  相似文献

The pathogenicity of the Dermatophilus-like microorganisms from porcine tonsil and the light and electron microscopic findings were studied with adult ewes. The early lesions were abscessess and advanced ones were granulomas after the subcutaneous inoculation. The granulomas were composed of the central bacterial colonies and the layers of the neutrophils, epithelioid cells and giant cells, and peripheral connective tissues. Epithelioid cells and giant cells were identified by the large euchromatic nuclei, abundant organelles and interdigitation of the blunt pseudopods in the ultrastructure. The lesions were very similar to subcutaneous granulomas in sheep and cattle due to Dermatophilus congolensis (atypical dermatophilosis), actinomycosis and nocardiosis.  相似文献

In rodents an intravenous administration of viableCryptococcus (C.) neoformans cells frequently resulted in attachment of intravascular cryptococcal granulomas to inner walls of the large to medium-sized veins of various organs, including the lungs, liver and spleen. In order to elucidate the pathogenesis of granulomatous changes, the cells composing the intravascular granulomas were observed by electron microscopic peroxidase (PO) cytochemistry. The granuloma composing cells could be divided into the following four types according to the pattern of endogenous peroxidase activity: exudate macrophage (Mφ, type I), PO-negative Mφ (type II), resident Mφ (type III) and other inflammatory cells (type IV). In the intravenous granulomas of the lung, the percentages of composed cells were 39.0% for type I, 57.9% for type II, 0% for type III and 3.1% for type IV. By contrast, in the interstitial granulomas in the lung, type III Mφs, possibly derived from alveolar Mφs, played a significant role in granuloma formation. This may indicate that the intravascular granuloma is almost composed of macrophages derived from monocytes rather than alveolar macrophages. The expression of ICAM-1 on endothelia of the pulmonary veins was examined by immunoelectron microscopy. An immunogold labeling index was significantly augmented on the surface of endothelia in response to intravenous challenge ofC. neoformans. The intravascular granuloma demonstrates that the monocytes develop into the granuloma-composing macrophages and suppress the cryptococcal activities even hi the peripheral blood resulting in an assistance of endothelial functions.  相似文献

Transforming growth factor beta-1 (TGF-beta1) is released from the extracellular matrix of rat growth plate chondrocytes and activated by stromelysin-1 (matrix metalloproteinase 3, MMP-3), an enzyme that is stored in matrix vesicles. MMP-3 is released from these extracellular organelles by the direct action of 1alpha,25(OH)2D3 via activation of phospholipase A2 (PLA2), resulting in local production of lysophospholipids and matrix vesicle membrane destabilization. This effect of 1alpha,25(OH)2D3 is greater in matrix vesicles from growth zone chondrocyte cultures and PLA2 activity is higher in the growth zone in vivo, suggesting that it may depend on chondrocyte maturation state in the endochondral lineage. Previous studies have shown that latent TGF-beta1 can be activated by mild detergents in vitro, suggesting that lysophospholipids may act in vivo in a similar manner. To test this hypothesis, we determined if rat costochondral growth plate cartilage cells produce lysophosphatidylcholine (LPC) and lysophosphatidylethanolamine (LPE) in a maturation state-dependent manner and if LPC or LPE could release and activate latent TGF-beta1 from the extracellular matrix produced by these cells. Rat growth plate chondrocytes produced both lysophospholipids, with growth zone cells producing higher levels of LPE via PLA1, and resting zone cells producing higher levels of LPC via PLA2. LPC and LPE directly increased activation of recombinant human latent TGF-beta1 in a biphasic manner with a peak at 2 microg/ml. Phosphatidylcholine, phosphatidylethanolamine, and LPE plasmalogen (LPEP), but not choline, also activated TGF-beta1. Latent TGF-beta1 incubated with LPC or LPE, but neither lysophospholipid alone, stimulated [3H]-thymidine incorporation of resting zone cells, indicating the TGF-beta1 released was biologically active. LPC and LPE also released TGF-beta1 in a dose- and time-dependent manner when incubated with cell-free extracellular matrices produced by the cells. These results indicate that LPC and LPE have important roles as regulators of rat growth plate chondrocytes by directly and indirectly activating TGF-beta1 stored in the extracellular matrix.  相似文献

Summary This study of the ultrastructure of the auditory sensilla of the New Zealand weta, Hemideina crassidens, is the first such study on a member of the orthopteran Superfamily Gryllacridoidea. Ultrastructure of the auditory sensilla is similar in all of the tibial mechanosensory organs, here called subgenual organ, intermediate organ and crista acoustica by analogy with comparable structures in Tettigoniidae.Distal to each sensory soma is a dendrite containing multiple ciliary rootlets that fuse into a single ciliary root. This splits into nine root processes that pass around the outside of the proximal basal body and then rejoin at the level of the distal basal body, distal to which the dendrite has a modified ciliary structure with a circlet of nine peripheral paired tubes and rods as it passes through the proximal extracellular space. It is then enclosed by a zone of scolopale cell cytoplasm before expanding into a dilatation within the distal extracellular space. In some sensilla this space is partially occluded by electron dense material which is part of the scolopale cell. Distal to the dilatation the cilium shrinks and ends surrounded by the scolopale cap.Accessory cells consist of glia enwrapping the sensory neuron in the region of its soma, the scolopale cell surrounding the ciliary portion of the dendrite, and the attachment cell surrounding the scolopale cell and scolopale cap and connected to them by desmosomes. The attachment cells are filled with microtubules in differing densities and orientations. Lamellae are present in the acellular matrix surrounding the attachment cells. Banded fibres, presumably of collagen, are also present in the matrix.  相似文献

An astonishing number of extracellular matrix glycoproteins are expressed in dynamic patterns in the developing and adult nervous system. Neural stem cells, neurons, and glia express receptors that mediate interactions with specific extracellular matrix molecules. Functional studies in vitro and genetic studies in mice have provided evidence that the extracellular matrix affects virtually all aspects of nervous system development and function. Here we will summarize recent findings that have shed light on the specific functions of defined extracellular matrix molecules on such diverse processes as neural stem cell differentiation, neuronal migration, the formation of axonal tracts, and the maturation and function of synapses in the peripheral and central nervous system.  相似文献

Hepatic Schistosoma mansoni periovular granulomas undergo changes in size, cellular composition and appearance with time. This phenomenon, known as "immunological modulation", has been thought to reflect host immunological status. However, as modulation has not been observed outside the liver, participation of local factors, hitherto little considered, seems crucial. Components of the extracellular matrix of periovular granulomas of the mouse were particularly studied in three different organs (liver, lung and intestine) and during three periods of infection time (acute, intermediate and chronic) by means of histological, biochemical and immunofluorescence techniques, while quantitative data were evaluated by computerized morphometry, in order to investigate participation of local factors in granuloma modulation. Results confirmed modulation as a exclusively hepatic phenomenon, since pulmonary and intestinal granulomas, formed around mature eggs, did not change size and appearance with time. The matricial components which were investigated (Type I, III and IV collagens, fibronectin, laminin, proteoglycans and elastin) were found in all granulomas and in all organs examined. However, their presence was much more prominent in the liver. Elastin was only found in hepatic granulomas of chronic infection. The large amount of extracellular matrix components found in hepatic granulomas was the main change responsible for the morphological aspects of modulation. Therefore, the peculiar environment of the liver ultimately determines the changes identified in schistosomal granuloma as "modulation".  相似文献

【目的】研究湛江沿海硇洲岛和徐闻珊瑚礁自然保护区潮间带产胞外纤溶酶样酶和纤溶酶原激活物海洋真菌的生物多样性,为发掘新型溶栓药物奠定基础。【方法】采用马铃薯葡萄糖琼脂(PDA)和酵母膏蛋白胨葡萄糖(YPD)培养基分离培养海洋真菌,采用真菌r DNA转录间隔区1-5.8S r DNA-转录间隔区2(ITS1-5.8S-ITS2)片段的序列分析及其系统进化树构建的方法鉴定分离培养的海洋真菌,采用脱脂牛奶马铃薯葡萄糖琼脂(SM-PDA)培养基培养法筛选产胞外蛋白酶的海洋真菌,采用海水纤维蛋白马铃薯葡萄糖琼脂(FN-PDA)培养基培养法筛选产胞外纤溶酶样酶和/或纤溶酶原激活物的海洋真菌。【结果】从湛江沿海的硇洲岛和徐闻珊瑚礁自然保护区潮间带分离、培养和鉴定了海洋真菌446株,含真菌的98个种,分布于真菌域子囊菌门(Ascomycota)和担子菌门(Basidiomycota)的6个纲、18个目、46个科、65个属;其中产胞外蛋白酶的海洋真菌有265株,61个种,分布于41个属;产胞外纤溶酶样酶的海洋真菌有67株,22个种,分布于14个属;产胞外纤溶酶原激活物的海洋真菌有84株,23种,分布于13个属;优势属为曲霉属(Aspergillus),其次为青霉属(Penicillium)。【结论】湛江沿海潮间带可分离培养的产胞外纤溶酶样酶和纤溶酶原激活物的海洋真菌物种丰富多样,是发掘新型溶栓药的丰富资源。  相似文献

After peripheral nerve injury in adult mammals, reestablishment of functional connections depends on several parameters including neurotrophic factors, the extracellular matrix, and hormones. However, little is known about the contribution of hormones to peripheral nerve regeneration. Thyroid hormones, which are required for the development and maturation of the central nervous system, are also important for the development of peripheral nerves. The action of triiodothyronine (T3) on responsive cells is mediated through nuclear thyroid hormone receptors (TRs) which modulate the expression of specific genes in target cells. Thus, to study the effect of T3, it is first necessary to know whether the target tissues possess TRs. The fact that sciatic nerve cells possess functional TRs suggests that these cells can respond to T3 and, as a consequence, that thyroid hormone may be involved in peripheral nerve regeneration. The silicone nerve guide model provides an excellent system to study the action of local administration of T3. Evidence from such studies demonstrate that animals treated locally with T3 at the level of transection have more complete regeneration of sciatic nerve and better functional recovery. Among the possible regulatory mechanisms by which T3 enhances peripheral nerve regeneration is rapid action on both axotomized neurons and Schwann cells which, in turn, produce a lasting and stimulatory effect on peripheral nerve regeneration. It is probable that T3 up- or down-regulates gene expression of one or more growth factors, extracellular matrix, or cell adhesion molecules, all of which stimulate peripheral nerve regeneration. This could explain the greater effect of T3 on nerve regeneration compared with the effect of any one growth factor or adhesion molecule.  相似文献

Ischemia and denervation of EDL muscle of adult rat induce a large central zone of degeneration surrounded by a thin zone of peripheral surviving muscle fibers. Muscle regeneration is a complex phenomenon in which many agents interact, such as growth factors and heparan sulfate components of the extracellular matrix. We have shown that synthetic polymers, called RGTA (as regenerating agents), which imitate the heparan sulfates, are able to stimulate tissue repair when applied at the site of injury. In crushed muscles, RGTA were found to accelerate both regeneration and reinnervation. In vitro, RGTA act as protectors and potentiators of various heparin binding growth factors (HBGF). It was postulated that in vivo their tissue repair properties were due in part to an increase of bioavailability of endogenously released HBGF. In the present work, we show that ischemic and denervated EDL muscle treated by a unique injection of RGTA differs from the control after 1 wk in several aspects: 1) the epimysial postinflammatory reaction is inhibited and the area of fibrotic tissue among fibers is reduced; 2) the peripheral zone, as measured by the number of intact muscle fibers, was increased by more than twofold; and 3) In the central zone, RGTA enhances the regeneration of the muscle fibers as well as muscle revascularization. These results suggest that RGTA both protects muscle fibers from degeneration and preserves the differentiated state of the surviving fibers. For the first time it is demonstrated that a functionalized polymeric compound can prevent some of the damage resulting from muscle ischemia. RGTA may therefore open a new therapeutic approach for muscle fibrosis and other postischemic muscle pathologies.  相似文献

The shoot apical meristem (SAM) is functionally subdivided into zones with distinct tasks. During vegetative growth the peripheral zone of the meristem gives rise to leaf primordia that develop into dorsiventral leaves under the influence of signals from the central zone. During the floral transition the function of the SAM is altered and its peripheral zone starts to form floral structures in a specific pattern. This requires alterations in the signal networks that coordinate the activities of the peripheral and central zone of the SAM. These signal networks are partly housed in the symplasmic space of the SAM. Dye-coupling experiments demonstrate that in the superficial layer of the Sinapis alba meristem this space is radially subdivided. The cells of the central zone are coupled into a symplasmic field, which is shielded from the peripheral zone by the positional closing of plasmodesmata. In the vegetative meristems, most of these central symplasmic fields have a triangular geometry and are relatively small in size. Plants that are induced to flower by exposure to a single long day alter the geometry as well as the size of their central symplasmic field. After two subsequent days under short photoperiod the central symplasmic fields exhibit a circular form. Simultaneously, their size strongly increases both in an absolute sense and relative to the enlarging meristem. The geometric change in the fields is hypothesized to be due to recruitment of extra initial cells, required to support the increase in phyllotactic complexity. The proportional increase in field size is interpreted as an adjustment in the balance between the central and peripheral zone of the SAM, accompanying the shift from leaf production to flower formation.  相似文献
