Intact internodal cells of Chara are known to maintain their osmotic pressures at constant levels in artificial pond water at room temperature. Cell fragments with osmotic pressures higher and those cell fragments with osmotic pressures lower than the original, both of which are prepared from intact internodal cells using transcellular osmosis and ligation with threads, can also return their osmotic pressures to the original level within a week in artificial pond water. These regulatory phenomena are realized mainly by extrusion of K+ and Cl in the cytoplasm and/or vacuole or by absorption of K+ and Cl from the external solution. According to the electrochemical potential difference calculated for K+ between the vacuole and the external solution, the cells should be able to maintain these regulatory functions even in 50–100 m M KCl+ 1 m M CaCl2 solutions. However, novel phenomena were observed when they were immersed in such concentrated KCl solutions. To maintain electroneutrality, their osmotic pressures increased up to ca l MPa in 2 days due to absorption of K+ and Cl and many gradually died over time. Ionic and osmotic reguratory functions of Chara cells were lost when they were immersed in 50–100 m M K-salt solutions containing 1 m M Ca2+.  相似文献

Water and ion fluxes of intact root systems of Pyrus communis L. cv. Old Home × Farmingdale 97 immersed in a nutrient solution were determined at various pressures and temperatures. Water flux (Jv was normalized on the basis of initial flow rates of a root system after 30 min at 0.50 MPa and 25°C, expressed as the ratio Qv. Qv responded linearly to pressures between 0.20 and 0.62 MPa, implying a constant root hydraulic conductivity (Lp) within this range. Similarly Qv was linearly related to temperatures between 7 and 35°C; however, large, rapid temperature changes resulted in a break of the Arrhenius plot of Qv versus the reciprocal of temperature, Abscisic acid (ABA) from 2 × 10−6 to 10−4 M , applied to intact root systems, increased Qv within 10–20 min, with the effect leveling off after 1.5 h. At a pressure of 0.50 MPa, ABA at 10-4 M enhanced Qv by 28%. The stimulation of Qv was not due to the ethanol solvent since 0.13 or 1.33% ethanol decreased Qv-, The osmotic potential of the xylem fluid was determined and was used to calculate total normalized solute flux. The results suggest that ABA-induced or ethano1-induced changes in Qv were mainly due to changes in Lp and not to changes in ion transport to the xylem.  相似文献

Abstract Hydrogen oxidation rates were measured in a neutral compost soil and an acidic sandy loam at H2 mixing ratios of 0.01 to 5000 ppmv. The kinetics were biphasic showing two different K m values for H2, one at about 10–40 nM dissolved H2, the other at about 1.2–1.4 μM H2. The low- K m activity was less sensitive to chloroform fumigation than the high- K m activity. If sterile soil was amended with Paracoccus denitrificans or a H2-oxidizing strain isolated from compost soil, it exhibited only a high- K m (0.7–0.9 μM) activity. It also failed to utilize H2 mixing ratios below a threshold of 1.6–3.0 ppmv H2 (160–300 mPa). A similar result was obtained when fresh soil samples were suspended in water, and H2 oxidation was determined from the decrease of dissolved H2. However, H2 was again utilized to mixing ratios lower than 0.05 ppmv, if the supernatant of the soil suspension or the settled soil particles were dried onto sterile soil or purified quarz sand. Obviously, soils contain two different activities for oxidation of H2: (1) a high- K m, high-threshold activity which apparently is due to aerobic H2-oxidizing bacteria, and (2) a low- K m, low-threshold activity whose origin is unknown but presumably is due to soil enzymes.  相似文献

Blood samples from healthy adult Atlantic salmon fed an optimal diet in net sea pens were collected at intervals from October to May. Haematological determinations and biochemical serum analyses were carried out on 20 fish in each of seven samples. The ranges of haemato-logical values for sample means were: haematocrit 44–49%, haemoglobin 8.9–10.4 g 100ml−1, red blood cell count 0.85–1.10 × 1012 l−1, MCV 441–553 × 10−15 1, MCH 94–106 × 10−6 g, MCHC 19.4–21.7 g 100ml−1 and leucocrit 0.43–0.96%. The ranges of enzyme activities in serum, for sample means, were: alkaline phosphatase 647–988Ul−1, aspartate aminotrans-ferase 202–351 Ul−1 and alanine aminotransferase 4–8 Ul−1. The ranges of the other parameters analyzed in serum were: total protein 41.6–56.6 gl−1, albumin 18.3–24.3 gl−1, albumin/total protein ratio 39.3–44.0%, creatinine 26–46 μmol, triglycerides 2.53–4.98 μmol and cholesterol 9.3–12.8 μmol. These values are considered to be the normal ranges in healthy fish. Variations due to seasonal changes, and the clinical significance of the selected parameters, are discussed. Data showing the reproducibility of the biochemical analyses in serum are presented.  相似文献

Abstract— Partly purified chromaffin granules were incubated in vitro with Ca2+ (with trace amounts of 45Ca2+) in concentrations ranging from 4 μm to 1 mm. After incubation the granules were washed with media containing EDTA and then subjected to density gradient centrifugation (1.3 to 2.0 m-sucrose solutions) in order to characterize the particles which had taken up 45Ca2+. By using marker enzymes and various inhibitors of Ca2+ uptake into such cell particles as mitochondria it was established that under the conditions of the experiments chromaffin granules took up Ca2+ from the incubation medium. To characterize this uptake a simplified density gradient procedure was tested and found to be suitable. The uptake of Ca2+ into chromaffin granules was strongly dependent on temperature. It was not activated by ATP. The uptake was linear up to 10 min. At high calcium concentrations (above 200 μm) the rate of uptake levelled off. The uptake at 37°C was 1 nmol Ca2+/mg protein/min at a Ca2+ concentration of 500 μm. Mg2+ had no influence on Ca2+ uptake, whereas Sr2+ (1 mm) inhibited it. The methods established in this study should prove useful for a further characterization of this Ca2+ uptake into chromaffin granules which is likely to represent a useful model for the Ca2+ uptake occurring in the intact gland.  相似文献

Abstract Vitronectin, a serum and extracellular matrix protein involved in immunological reactions, interacts with Helicobacter pylori strains. Of the 20 H. pylori strains tested three strains bound more than 50% of the vitronectin added, five strains bound 25–40%, nine strains bound 10–20% and three strains bound 5–8% vitronectin. Two strains, one with high- and one with low-binding properties, were selected for further characterization of 125I-vitronectin binding. Binding to the urea-activated 125I-labelled vitronectin was fast, saturable and reversible when an excess of unlabelled vitronectin was added to the bacteria with bound 125I-vitronectin. The binding was heat- and protease-sensitive, suggesting that the binding was mediated by bacterial cell-surface proteins. Since components such as fetuin and orosomucoid but not asialofetuin inhibited the binding, sialic-acid specific proteins, related to H. pylori sialic-acid specific haemagglutinins, were probably involved.  相似文献

Abstract Newly developed low capacity columns were used in suppressed ion chromatography for rapid and highly reproducible determination of SO42− in porewater samples from freshwater sediments without preconcentration of samples. With a 50 μl injection the detection limit for SO42− was ca. 50 pmol (= 1 μ M) with a precision of 1–3% at the 10–200 μM level and <1% at concentrations above 200 μM. SO42− could be measured in 4–5 min with the routinely used eluent (3.0 mM NaHCO3/0.8 mM Na2CO3). When the strength of the eluent was increased to 3.0 mM NaHCO3/2.0 mM Na2CO3, sulfate analysis was possible in less than 3 min, provided that samples were nitrate-free. Under these conditions S2O32− could also be sensitively determined in about 6 min. Examples of application of the method are given for measurements of sulfate reduction rates in freshwater sediment samples from Lake Constance.  相似文献

Methanogenesis and methanotrophy within a Sphagnum peatland   总被引:4,自引:0,他引:4  
Abstract: Methane production and consumption activities were examined in a Massachusetts peatland. Peat from depths of 5–35 cm incubated under anaerobic conditions, produced an average of 2 nmol CH4 g−1 h−1 with highest rates for peat fractions between 25–30 cm depth. Extracted microbial nucleic acids showed the strongest relative hybridization with a 16S rRNA oligonucleotide probe specific for Archaea with samples from the 25–30 cm depth. In aerobic laboratory incubations, the peat consumed methane with a maximum velocity of 67 nmol CH4 g−1 h−1 and a K s of 1.6 μM. Methane consumption activity was concentrated 4–9 cm below the peat surface, which corresponds to the aerobic, partially decomposed region in this peatland. Phospholipid fatty acid analysis of peat fractions demonstrated an abundance of methanotrophic bacteria within the region of methane consumption activity. Increases in temperature up to 30°C produced an increase in methane consumption rates for shallow samples, but not for samples taken from depths greater than 9 cm. Nitrogen fixation experiments were carried out using 15N2 uptake in order to avoid problems associated with inhibition of methanotrophy. These experiments demonstrated that methane in peat samples did not stimulate nitrogen fixation activity, nor could activity be correlated with the presence of methanotrophic bacteria in peat fractions.  相似文献

Availability, uptake and turnover of glycerol in hypersaline environments   总被引:4,自引:0,他引:4  
Abstract A sensitive assay for glycerol and other polyols was developed, based on periodate oxidation to formaldehyde, followed by a colorimetric assay with 3-methyl-2-benzothiazolone hydrazone. Apparent glycerol concentrations thus measured in saltern crystallizer ponds were around 20–36 μM, while in the Dead Sea, during a Dunaliella bloom, values were up to 27 μM. However, these values probably overestimate the glycerol concentrations present, as shown by labeled glycerol uptake experiments. Values of [K + Sn] (natural concentration + affinity constant) in saltern ponds were as low as 0.76–1.4 μM, with Vmax values of 193–303 nmol 1−1h−1, and turnover times between 2.6–7.2 h at 35°C. Similar measurements in the Dead Sea were: [K + Sn] 0.07–1.41 μM, Vmax values 160–426 nmol 1−1h−1, and turnover times in the range of 0.45–3.3 h.  相似文献

Chlamydomonas acidophila Negoro is a green algal species abundant in acidic waters (pH 2–3.5), in which inorganic carbon is present only as CO2. Previous studies have shown that aeration with CO2 increased its maximum growth rate, suggesting CO2 limitation under natural conditions. To unravel the underlying physiological mechanisms at high CO2 conditions that enables increased growth, several physiological characteristics from high- and low-CO2-acclimated cells were studied: maximum quantum yield, photosynthetic O2 evolution (Pmax), affinity constant for CO2 by photosynthesis (K0.5,p), a CO2-concentrating mechanism (CCM), cellular Rubisco content and the affinity constant of Rubisco for CO2 (K0.5,r). The results show that at high CO2 concentrations, C. acidophila had a higher K0.5,p, Pmax, maximum quantum yield, switched off its CCM and had a lower Rubisco content than at low CO2 conditions. In contrast, the K0.5,r was comparable under high and low CO2 conditions. It is calculated that the higher Pmax can already explain the increased growth rate in a high CO2 environment. From an ecophysiological point of view, the increased maximum growth rate at high CO2 will likely not be realised in the field because of other population regulating factors and should be seen as an acclimation to CO2 and not as proof for a CO2 limitation.  相似文献

Abstract: The amyloid protein (βA4) is found in the CNS of patients with Alzheimer's disease; however, the pathogenic role of this protein is not known. In the present study, a peptide fragment of βA4βA4 25–35; Gly-Ser-Asn-Lys-Gly-Ala-Ile-Ile-Gly-Leu-Met-NH2), which contains the conserved C-terminal sequence of substance P (X-Gly-Leu-Met-NH2), and the neuropeptide substance P (SP) were examined for their ability to modulate nicotine-evoked secretion from cultured bovine adrenal chromaffin cells. Secretion of the released endogenous catecholamines was monitored by electrochemical detection after separation by HPLC. Secretion induced by 10−5 M nicotine was inhibited by SP and βA4 25–35. The IC50 of SP and βA4 25–35 was 3 × 10−6 and 3 × 10−5 M , respectively. SP and βA4 25–35 both protected against nicotinic receptor desensitization. However, βA4 25–35 was ∼ 10-fold less effective than SP in its protective effect. The present work shows that βA4 25–35 can mimic the modulatory actions of SP on the nicotinic response of cultured bovine chromaffin cells, i.e., inhibition of the nicotinic response and protection against nicotinic desensitization. These modulatory actions may be associated with changes in nicotinic receptor levels reported to occur in Alzheimer's disease.  相似文献

Abstract: 45Ca2+ uptake by synaptosomes isolated from cerebral cortex, cerebellum, midbrain, and brain stem of male Sprague-Dawley rats was measured at 1-, 3-, 5-, 15-, 30-, and 60-s time periods. The fastest rate of depolarization-dependent calcium uptake occurred in each brain region between 0 and 1 s. Uptake rates dropped off quickly with 3–5-s rates at approximately 15–20% of those observed at 0–1 s in cerebral cortex, cerebellum, and midbrain. Uptake rates at the 1–3-s interval were maintained at a relatively high rate in these three brain regions suggesting mixed fast- and slow-phase processes. The magnitude and rate of 45Ca2+ uptake were similar in synaptosomes from cerebral cortex, cerebellum, and midbrain but were significantly less in brain stem synaptosomes. These results suggest a fast and a slow component to voltage-dependent 45Ca2+ uptake by presynaptic nerve terminals from various brain regions.  相似文献

The goal of the present study was to monitor cuticular wax accumulation during leaf development of Kalanchoe daigremontiana . Leaves expanded linearly until they were 40–60 d old. Wax coverages of leaves on the third node increased steadily during initial leaf development, from 6.5  µ g·cm−2 on day 22 to 15.3  µ g·cm−2 on day 53, and then levelled off. Triterpenoids dominated the wax mixture throughout leaf development, but decreased from 74 to 40–45% in mature leaves, while very long-chain fatty acid (VLCFA) derivatives increased from 19 to 39–44%. The major VLCFA derivatives were alkanes, accompanied by fatty acids, primary alcohols, aldehydes and alkyl esters. In all compound classes, either C34 or C33 homologs predominated during leaf development. Eight different triterpenoids were identified, with glutinol constituting 70% of the fraction, and friedelin (20%) and germanicol (10%) as further major components of the young leaf wax. The glutinol percentage decreased, while the relative amounts of epifriedelanol and glutanol increased during development. Various leaf pairs upwards from the third node showed similar growth patterns and developmental time courses of cuticular wax amounts and composition. Based on these surface chemical analyses, the relative activities of biosynthetic pathways leading to various wax components can be assessed.  相似文献

In suspension cultures of Phytolacca americana L., betacyanin accumulation per cell increased with increasing total nitrogen concentration (initial NH+4:NO3 ratio 1:2) in the range 0–40 m M and then remained almost constant in the range 40–80 m M . Increasing ammonium increased growth while betacyanin accumulation was reduced. On the other hand, betacyanin accumulation increased when nitrate was increased while growth was almost constant in the concentration range examined. A time-course study of ammonium and nitrate concentration changes in the medium showed that betacyanin accumulation was associated with nitrate uptake.  相似文献

A virus obtained from naturally infected lucerne ( Medicago sativa ) in New Zealand reacted with antiserum to an Australian isolate of lucerne transient streak virus (LTSV). Some plants infected with New Zealand isolates showed yellow flecks along lateral veins of leaves; symptoms were transient in some lucerne plants but persistent in others. A New Zealand isolate (LTSV-NZ) infected 14 of 39 plant species tested by mechanical inoculation, but was not transmitted by five aphid species. In sap of Nicotiana clevelandii , LTSV-NZ was infective after storage for 4 wk at 20 oC, diluting to 10-5, or heating for 10 min at 70 oC. Purified virus preparations contained a single electrophoretic component and a single sedimenting component (s20w= 112 S ) which formed a single buoyant density component in CsCl (1.37 g cm-3) but two density components in Cs2SO4 (1.26 and 1.32 g cm-3). LTSV-NZ particles were stable in 10 ITIM EDTA at pH 5, but not at pH 8, being degraded into two sedimenting components of 105 S and 92 S. Particles contained c. 18% RNA in the form of one single-stranded RNA molecule of mol. wt 1–4 times 106, and a polypeptide of mol. wt c. 32 400. LTSV-NZ was serologically unrelated to 24 other isometric plant viruses. However, its properties are similar to those of southern bean mosaic virus and allied viruses. Abstract Using aerobic soil slurry technique nitrification and nitrous oxide production were studied in samples from a pine site in Western Finland. The site received atmospheric ammonium deposition of 7–33 kg N ha−1 a−1 from a mink farm. The experiments with soil slurries showed that the nitrification potential in the litter layer was higher at pH 6 than at pH 4. However, the nitrification potentials in the samples from the organic and mineral horizons at pH 6 and 4 were almost equal. Also N2O was produced at a higher rate at pH 6 than at pH 4 in slurries of the litter layer samples. The reverse was true for samples from the organic and mineral horizons. The highest N2O production and nitrification rates were measured in the suspensions of litter layer samples. Nitrification activity in field-moist soil samples was lower than the activity in the slurries indicating that the availability of ammonium limited nitrification in these soils. Acetylene (2.5 kPa) retarded nitrification activity (70-–100%) and N2O production (40 – 90%) in soil slurries. Acetylene inhibited the N2O production by 40–60% during the first 3 days after its addition to field-moist samples incubated in aerobic atmosphere. After 3 days the inhibition became much lower (4–5%). The results indicate that, in soil profiles of boreal coniferous forests receiving ammonium deposition, chemolithotrophic nitrification may have importance in the N2O production, and that changes in soil pH affect differently nitrification as well as N2O production in litter and deeper soil layers.  相似文献

The schooling behaviour of Atlantic mackerel was studied in a large tank at different light intensities in the range 12.6–1.8 × 10−10μEs−1 m−2. Variable light intensity was produced by accurately controlling the current to a green light-emitting diode (LED) 3 m above the experimental tank. Under high light levels (1.8 × 10−6μEs−1 m−2) mackerel always formed a single school, whereas at lower levels (1.8 × 10−8μEs−1 m−2) they swam as individuals. At light levels down to 1.0 × 10−6μEs−1 m−2 the mean nearest neighbour distance in a school remained relatively constant (0.3–0.9 body lengths), and individual mackerel swam along a path which deviated from the position of their nearest neighbours by less than 14°. As light dropped below 1.8 × 10−7μEs−1 m−2, both nearest neighbour distance and heading angle between nearest neighbours increased, with mean values of 1–1.8 body lengths and 23–92°, respectively, at 1.8 × 10−9μEs−1 m−2. The results are discussed in terms of ambient light conditions in the sea.  相似文献

Elongation, indolyl-3-acetic acid (IAA) and abscisic acid (ABA) levels, – gas chromatography-mass spectrometry quantification –, in the elongating zone were analysed for maize ( Zea mays L., Cv. LG11) roots immersed in buffer solution with or without zeatin (Z). The effect of Z depends on the initial extension rate of roots. The slower growing roots are more strongly inhibited by Z (10−7−10−5 M ) and they show a greater increase in IAA and ABA content. When compared to the rapidly growing roots, the larger reactivity of the 'slow'ones cannot be attributed to a higher Z uptake as shown when using [14C]-Z. It is suggested that Z could regulate root elongation by acting on the IAA and/or ABA level. The comparative action of these two hormones is discussed.  相似文献

Four components of rate-reducing resistance to Cercospora leaf spot in sugar beet (infection efficiency of conidia RC1, incubation period RC2, size of necrotic spots RC3 and spore yield RC4), previously measured in single infection cycle experiments, were integrated into a model simulating the chain of infection cycles under field conditions, as influenced by weather. To integrate resistance components, variables accounting for infection frequency, incubation period, affected leaf area, and infectiousness – which are computed for a susceptible cultivar – were modified by means of coefficients which reduced (RC1, RC3, RC4) or increased (RC2) them. Outputs obtained by running the model and changing resistance components actually reduced the rate of disease progress and the area under the disease progress curve of epidemics (AUDPC), as happens at field level; therefore, the approach may be considered successful. Changes in single resistance components were closely correlated with changes in AUDPC: improvements in RC1, RC3 or RC4 reduced AUDPC by the same, over the whole range of variation in infection frequency, affected leaf area, and infectiousness; on the contrary, little improvements in RC2 were more effective than stronger ones. When components acted simultaneously, each of them reduced disease progress in proportion to its magnitude; when all components were improved by the same amount, they had about the same effectiveness in slowing the epidemic. Changing more components simultaneously reduced the disease development slightly more than additively. Advantages for plant breeders in improving their selection strategies are outlined.  相似文献

Abstract. A controlled-environment chamber constructed from a standard chest freezer was used to grow and measure the CO2 exchange of small stands of lettuce ( Lactuca sativa L. ev. Ambassador). The chamber, with horizontal air flow, provided good control of air temperature ( c. 6 to 16°C), irradiance (0–300 μmol PAR m 2S1 and CO2 (350–1000 μmol mol−1). The photosynthetic response to changes in these variables was measured using an inexpensive CO2 dosing system which recorded the input rate required to maintain a constant concentration of CO2 (to ± 2.5%). Characteristis of the growth environment and the changes in response to temperature and irradiance are described.  相似文献
