Vanadate selectively inhibited dynein ATPase, 10?7M causing 50% inhibition under favorable conditions. Actomyosin ATPase was inhibited only by up to a thousand times higher concentration. In both cases vanadate inhibition was not competitive with ATP. Reversal by catecholamines was correlated with reduction of vanadate. The motility of demembranated sea urchin or mammalian sperm was arrested by vanadate concentrations similar to those which inhibited dynein ATPase; a thousand times higher concentration was needed to paralyze live sperm. The possible utility of vanadate sensitivity as a probe for dynein involvement in non-axonemal motile systems was explored with respect to brain ATPase associated with tubulin obtained by cycles of assembly, and ATPases associated with mitotic apparatus isolated from sea urchin embryos.  相似文献

Apolipoprotein A-IV (apo A-IV), a peptide expressed by enterocytes in the mammalian small intestine and released in response to long-chain triglyceride absorption, may be involved in the regulation of gastric acid secretion and gastric motility. The specific aim of the present study was to determine the pathway involved in mediating inhibition of gastric motility produced by apo A-IV. Gastric motility was measured manometrically in response to injections of either recombinant purified apo A-IV (200 microg) or apo A-I, the structurally similar intestinal apolipoprotein not regulated by triglyceride absorption, close to the upper gastrointestinal tract in urethane-anesthetized rats. Injection of apo A-IV significantly inhibited gastric motility compared with apo A-I or vehicle injections. The response to exogenous apo A-IV injections was significantly reduced by 77 and 55%, respectively, in rats treated with the CCK(1) receptor blocker devazepide or after functional vagal deafferentation by perineural capsaicin treatment. In electrophysiological experiments, isolated proximal duodenal vagal afferent fibers were recorded in vitro in response to close-arterial injection of vehicle, apo A-IV (200 microg), or CCK (10 pmol). Apo A-IV stimulated the discharge of duodenal vagal afferent fibers, significantly increasing the discharge in 4/7 CCK-responsive units, and the response was abolished by CCK(1) receptor blockade with devazepide. These data suggest that apo A-IV released from the intestinal mucosa during lipid absorption stimulates the release of endogenous CCK that activates CCK(1) receptors on vagal afferent nerve terminals initiating feedback inhibition of gastric motility.  相似文献

Pharmacological studies of abomasum motor activity were carried out in 3 sheep with chronic fistulae in the rumen and the fundus and pyloric part of the abomasum. The contractions of these parts of the stomach were recorded by the balloon method. The obtained results showed that in the abomasum stimulation of alpha-adrenergic receptors raises the degree of motor activity while stimulation of beta-adrenergic receptors inhibits it. It has been shown also that the sympathetic system has a greater influence through the beta-adrenergic receptors on the motor activity of the pyloric part than on that of the fundus of the abomasum. The analysis of the records obtained after bilateral vagotomy demonstrated that the automatism of the pyloric part was greater than that of the fundus of the abomasum.  相似文献

Extracellular calcium at millimolar concentrations inhibits collective motility of ejaculated ram spermatozoa. In untreated cells, or when motility was made dependent upon glycolytic activity, there is very small inhibition, but when motility was made dependent upon mitochondrial respiration there is very high inhibition in motility by increasing extracellular Ca2+ concentration. Quercetin, which inhibits (Ca2+ + Mg2+)-ATPase activity in isolated plasma membranes, also inhibits motility mainly in cells that have been made dependent upon glycolytic activity, but there is also inhibition in untreated cells. When motility was made dependent upon mitochondrial activity, there is no inhibition but rather some stimulation in motility by quercetin. The inhibitory effect of quercetin is enhanced by increasing Ca2+ concentration in the medium. Quercetin also inhibits uptake of calcium into the cells, in a mechanism by which a calcium channel is involved. This inhibition is high only when the glycolysis is inhibited in the cells. The rate of glycolysis is decreased by quercetin or ouabain, but their effects on motility are quite different. Based on these data, it appears that the plasma membrane (Ca2+ + Mg2+)-ATPase or the Ca2+ pump have a functional role in the regulation of spermatozoa motility. This motility regulation is functioning through mechanisms which include glycolytic activity and maintenance of intracellular calcium concentrations.  相似文献

Reindeer have been classified as intermediate feeders and muskoxen as grazers based on differences in digestive morphology and consumption of fibrous plants. We hypothesized that the digestive morphology of young (<2 months) reindeer and muskoxen anticipates transitions in diet and determines the feeding strategy of each species at adulthood. We compared structural morphology and rates of cell division in the rumen, abomasum, duodenum and liver of reindeer and muskoxen as neonates (1 day old), during the transition from milk to forage (30–60  days old) and in adults (>7 yr). Development in utero provided the neonate with a functioning mucosa of the gastric abomasum and duodenal mucosa with high surface enlargement for digestion and absorption of concentrated milks. Transition to forage was preceded by changes in ruminal papillae structure that increased surface area and likely contributed to active fermentation by 60 days of age. The abomasum also increased in acid-secreting parietal cells during the transition to forage, which may enhance digestion of plant and microbial proteins. Rates of cell division also indicated a sustained differentiation of tissue structure during the transitional period. Young arctic ruminants expressed digestive structures that preceded full function, which indicated the ultimate feeding strategy of each species. For example, the rumen of young muskoxen had thick cornified epithelia and muscle layers that would provide ruminal mucosa with better protection from fibrous abrasion and enhance motility of bulky diets. Conversely, young reindeer had more complex papillary shapes in the rumen and more foliate villi in the duodenum, indicating a greater absorptive capacity of these structures than in muskoxen. Ontogenetic programs, therefore, play the primary role for digestive development of reindeer and muskoxen and determine the nutritional strategies of adults.  相似文献

Intestinal epithelial cells are subject to repetitive deformation during peristalsis and villous motility, whereas the mucosa atrophies during sepsis or ileus when such stimuli are abnormal. Such repetitive deformation stimulates intestinal epithelial proliferation via focal adhesion kinase (FAK) and extracellular signal-regulated kinases (ERK). However, the upstream mediators of these effects are unknown. We investigated whether Src and Rac1 mediate deformation-induced FAK and ERK phosphorylation and proliferation in human Caco-2 and rat IEC-6 intestinal epithelial cells. Cells cultured on collagen-I were subjected to an average 10% cyclic strain at 10 cycles/min. Cyclic strain activated Rac1 and induced Rac1 translocation to cell membranes. Mechanical strain also induced rapid sustained phosphorylation of c-Src at Tyr(418), Rac1 at Ser(71), FAK at Tyr(397) and Tyr(576), and ERK1/2 at Thr(202)/Tyr(204). The mitogenic effect of cyclic strain was blocked by inhibition of Src (PP2 or short interfering RNA) or Rac1 (NSC23766). Src or Rac1 inhibition also prevented strain-induced FAK phosphorylation at Tyr(576) and ERK phosphorylation but not FAK phosphorylation at Tyr(397). Reducing FAK using short interfering RNA blocked strain-induced mitogenicity and attenuated ERK phosphorylation but not Src or Rac1 phosphorylation. Src inhibition blocked strain-induced Rac1 phosphorylation, but Rac inhibition did not alter Src phosphorylation. Transfection of a two-tyrosine phosphorylation-deficient FAK mutant Y576F/Y577F prevented activation of cotransfected myc-ERK2 by cyclic strain. Repetitive deformation induced by peristalsis or villus motility may support the gut mucosa by a pathway involving Src, Rac1, FAK, and ERK. This pathway may present important targets for interventions to prevent mucosal atrophy during prolonged ileus or fasting.  相似文献

Microtransducers sensitive to changes in internal diameter were chronically implanted in the oviducts of 5 dairy cows. Motility patterns were recorded throughout 9 oestrous cycles. Cyclic variations in patterns of motility were observed and compared with circulating concentrations of plasma progesterone. Luteal-phase motility patterns were of low amplitude and frequency. The frequency and amplitude of motility increased 3-5 days before behavioural oestrus. This activity consisted primarily of longitudinal muscle contractions, with an interspacing of circular muscle activity occurring during oestrus. Patterns of activity after oestrus were similar to those before oestrus, with activity decreasing 3-5 days after oestrus. Transducers implanted bilaterally in 2 animals permitted observation of asynchronous patterns between right and left oviducts. Preliminary data suggested a higher level of activity in the oviduct ipsilateral to the active ovary. These variations may be due to a local effect, possibly mediated by the functional ovary or the ovum.  相似文献

Calcium signalling coordinates motility, cell invasion, and egress by apicomplexan parasites, yet the key mediators that transduce these signals remain largely unknown. One underlying assumption is that invasion into and egress from the host cell depend on highly similar systems to initiate motility. Using a chemical‐genetic approach to specifically inhibit select calcium‐dependent kinases (CDPKs), we instead demonstrate that these pathways are controlled by different kinases: both TgCDPK1 and TgCDPK3 were required during ionophore‐induced egress, but only TgCDPK1 was required during invasion. Similarly, microneme secretion, which is necessary for motility during both invasion and egress, universally depended on TgCDPK1, but only exhibited TgCDPK3 dependence when triggered by certain stimuli. We also demonstrate that egress likely comes under a further level of control by cyclic GMP‐dependent protein kinase and that its activation can induce egress and partially compensate for the inhibition of TgCDPK3. These results demonstrate that separate signalling pathways are integrated to regulate motility in response to the different signals that promote invasion or egress during infection by Toxoplasma gondii.  相似文献

Pressure changes inside the uterine wall were recorded by telemetry in six oestrous Awassi ewes during natural hand mating and AI, and in two oestrous ewes during fright caused by the presence of dogs.During control periods, the average frequency of primary pressure peaks was 3.7/min, their average amplitude was 29.6 cm H2O pressure, and there was an average of 2.5 secondary peaks per primary pressure peak. During AI or natural mating these parameters were 1.1–1.7 and during fright 1.1–1.3 times their control values, on the average. The experimental treatments caused practically no after effects.The results do not support the hypothesis that sperm transport in artificially inseminated ewes is impaired by inhibition of uterine motility. Infusion of 67 g ethanol over four hours in fasted, non-obese normal men (a) induced hypoglycaemia by inhibiting gluconeogenesis; (b) produced noticeable increases in blood lactate, 3-hydroxybutyrate, and free fatty acid concentrations; (c) depressed plasma growth hormone concentrations, despite hypoglycaemia; and (d) raised plasma cortisol concentrations before significant hypoglycaemia occurred. These metabolic changes were explained by the reduction of redox state which accompanies ethanol oxidation. The pronounced changes in metabolic values recorded during this study suggested that the use of parenteral feeding regimens including ethanol needs to be reconsidered.  相似文献

Human observers viewed dichoptic orthogonal sine-wave gratings and indicated when exclusive visibility occurred in either eye. Contrast was held constant in one eye and was increased or decreased in the other eye for a number of alternation cycles (continuous presentation) or for only the duration of a single period of exclusive visibility (synchronous presentation). The synchronous presentation condition allowed us to identify the differing effects of contrast during the suppressed and during the dominant periods. Mixed phases were recorded as distinct from suppressed and dominant phases, and new classifications of compound-dominant and compound-suppressed phases are defined. The results indicate that binocular rivalry responds to stimulus contrast in two ways. 1) The duty-cycle of dominance and suppression is determined by the relative image contrast between the two eyes, with dominance of the higher contrast image being favored, and 2) the overall rate of alternation is driven by monocular image contrast during the suppressed phase (increased monocular contrast increases the alternation rate) and to a lesser extent by monocular contrast during the dominant phase (increased monocular contrast decreases the rate). A model is developed to reflect these ideas. These results support a reciprocal inhibition oscillator as the underlying mechanism of binocular rivalry.  相似文献

Glucagon injection (50 micrograms kg-1) produced a biphasic response in plasma thyroxine (T4) level in both fed and fasted chickens. An initial inhibition was followed by an increase to levels above control value. Glucagon reduced plasma triidothyronine (T3) possibly as a consequence of inhibition of peripheral monodeiodination. This inhibition persisted in fasted animals despite a glucagon induced hyperglycaemia. Insulin injection (4 IU kg-1) decreased plasma T4 concomitant with a profound hypoglycaemia. These effects were more pronounced in fasted birds. Insulin induced hypoglycaemia was associated with decreased plasma T3 probably as a consequence of reduced thyroidal T4 secretion and reduced peripheral monodeiodination. Glucagon and insulin may play direct or indirect roles in the regulation of thyroid hormone secretion and metabolism in the domestic fowl.  相似文献

We reported previously that intravenously administered d-glucose acts in the central nervous system to inhibit gastric motility induced by hypoglycemia in anesthetized rats. The purpose of this study was to determine whether this effect is due to inhibition of dorsal motor nucleus of the vagus (DMV) cholinergic motoneurons, which synapse with postganglionic cholinergic neurons, or to excitation of DMV cholinergic neurons, which synapse with postganglionic nonadrenergic, noncholinergic (NANC) neurons, particularly nitrergic neurons. Three approaches were employed: 1) assessment of the efficacy of d-glucose-induced inhibition of gastric motility in hypoglycemic rats with and without inhibition of nitric oxide synthase [10 mg/kg iv nitro-l-arginine methyl ester (l-NAME)], 2) assessment of the efficacy of intravenous bethanechol (30 mug.kg(-1).min(-1)) to stimulate gastric motility in hypoglycemic rats during the time of d-glucose-induced inhibition of gastric motility, and 3) determination of c-Fos expression in DMV neurons after intravenous d-glucose was administered to normoglycemic rats. Results obtained demonstrated that l-NAME treatment had no effect on d-glucose-induced inhibition of gastric motility; there was no reduction in the efficacy of intravenous bethanechol to increase gastric motility, and c-Fos expression was not induced by d-glucose in DMV neurons that project to the stomach. These findings indicate that excitation of DMV cholinergic motoneurons that synapse with postganglionic NANC neurons is not a significant contributing component of d-glucose-induced inhibition of gastric motility.  相似文献

A series of protease inhibitors were tested on the motility of human, rat, bull, and rabbit demembranated reactivated spermatozoa. Some inhibitors, including aprotinin, boc-gln-leu-lys-H, and D-phe-pro-arg- H, could inhibit motility as well as prevent initiation of motility. In general, with the exception of aprotinin, protease inhibitors were more potent in preventing the initiation of movement than in blocking motility of demembranated spermatozoa. Protease substrates could also block sperm motility. Of the substrates tested only those with arg or lys ester bonds were active. The inhibition of motility by protease substrates was reversible, as once spermatozoa hydrolyzed the added exogenous protease substrates, motility reappeared. The importance of ester bond in the inhibitory action of protease substrates was confirmed by experiments that showed the lack of effect of pre- hydrolyzed protease substrates. The results suggest that a serine protease with lys and arg ester bond specificity is involved in the control of sperm motility. The fact that protease substrates also block motility of intact spermatozoa further emphasizes the physiological relevance of this new regulatory system.  相似文献

There has been considerable debate over whether asymptomatic neonatal hypoglycaemia results in neurological damage. In a detailed multicentre study of 661 preterm infants hypoglycaemia was found to be common. Moderate hypoglycaemia (plasma glucose concentration less than 2.6 mmol/l) occurred in 433 of the infants and in 104 was found on three to 30 separate days. There was considerable variation among the centres, implying differences in decisions to intervene. The number of days on which moderate hypoglycaemia occurred was strongly related to reduced mental and motor development scores at 18 months (corrected age), even after adjustment for a wide range of factors known to influence development. When hypoglycaemia was recorded on five or more separate days adjusted mental and motor developmental scores at 18 months (corrected age) were significantly reduced by 14 and 13 points respectively, and the incidence of neurodevelopmental impairment (cerebral palsy or developmental delay) was increased by a factor of 3.5 (95% confidence interval 1.3 to 9.4). These data suggest that, contrary to general belief, moderate hypoglycaemia may have serious neurodevelopmental consequences, and reappraisal of current management is urgently required.  相似文献

Motility of salmonid sperm is inhibited by the presence of carbon dioxide (CO2) in vitro; however, whether this occurs in response to challenges to the adult in vivo is not known. To determine whether CO2 negatively impacts sperm function in vivo, mature males were exposed to exhaustive exercise as well as to acute stress, chronic stress, tricaine anesthesia and environmental hypercapnia and sperm motility and semen CO2 tensions and pH values assessed. Semen CO2 rose and pH decreased significantly only in response to exhaustive exercise and environmental hypercapnia (13 kPa CO2). These changes in semen CO2 and pH were associated with reductions in numbers of sperm becoming motile upon water activation. Chronic and acute stress and tricaine anesthesia were without effect on sperm motility or on semen CO2 or pH. The time course of CO2 inhibition and recovery was evaluated in vitro. At least 50 min was required to note 50% of the inhibitory effect of low CO2 tensions on motility when sperm were exposed to 1.6-3.1 kPa CO2. At higher CO2 levels sperm motility displayed 50% of the inhibitory effect of these tensions within about 30 min. Sperm recovered maximal motility within 1 h of being placed in a nominally CO2-free environment. This study demonstrates sperm vulnerability to not only in vitro CO2 exposure but also in vivo exposure during exhaustive exercise and as result of environmental hypercapnia.  相似文献

The effects of experimental infection of lambs with third stage Trichostrongylus larvae are described. Preceding diarrhea, disturbances in the motility of the reticulum, abomasum and jejunum were observed by electromyography. Disappearance of the migrating myoelectric complexes occurred about 24 h before diarrhea and was accompanied by a decrease in both the frequency of reticular contractions and of contractions propagated through the antro-duodenal junction. The significance of these findings is discussed and compared with natural infestation in the field.  相似文献

The authors present two cases of chronic alcoholism in two female patients aged 41 and 52 years without diabetes mellitus, in whom hypoglycaemic coma occurred during the abstinence period. Hypoglycaemia in one patient occurred suddenly as a result of fasting within 24 hours following the last alcohol intake, whereas a severe hypoglycaemia in the second patient was developing progressively during 72 hours; patient did not eat much and the last meal took 24 hours before the onset of hypoglycaemic coma. Diagnosis of hypoglycaemic coma was suspected because as no alcohol or acetic acid smell were felt, no alcohol or methanol was detected in blood (tested only in one patient). Adrenergic reactions were not distinct (no excessive sweating, convulsions, tachycardia). The authors suggest, that a severe hypoglycaemia should be considered in patients suspected of alcoholism, and the treatment should start earlier with intravenous glucose administration.  相似文献

Flavonoid supplementation is likely to be beneficial in improving rumen fermentation and in reducing the incidence of rumen acidosis and bloat. Flavonoids are also said to increase the metabolic performance during the peripartum period. Ruminants are constantly exposed to flavonoids present in feed. However, it is not clear if these phytochemicals can affect the activity of the gut smooth muscle. Therefore, the aim of the study was to verify the effect of three flavonoids on bovine isolated abomasum smooth muscle. The study was carried out on bovine isolated circular and longitudinal abomasal smooth muscle specimens. All experiments were conducted under isometric conditions. The effect of apigenin, luteolin and quercetin (0.001 to 100 µM) was evaluated on acetylcholine-precontracted preparations. The effect of multiple, but not cumulative, treatment and single treatment with each flavonoid on abomasum strips was compared. Apigenin (0.1 to 100 µM) dose-dependently showed myorelaxation effects. Luteolin and quercetin applied in low doses increased the force of the ACh-evoked reaction. However, if used in high doses in experiments testing a wide range of concentrations, their contractile effect either declined (luteolin) or was replaced by an antispasmodic effect (quercetin). Surprisingly, the reaction induced by flavonoids after repeated exposure to the same phytochemical was not reproducible in experiments testing only single exposure of abomasum strips to the same flavonoid used in a high concentration. Taking into account the physicochemical properties of flavonoids, this data suggests the ability of flavonoids to interfere with cell membranes and, subsequently, to modify their responsiveness. Assuming ruminant supplementation with luteolin or quercetin or their presence in daily pasture, a reduction of the likelihood of abomasum dysmotility should be expected.  相似文献

The effect of stressful stimuli on uterine motility during pregnancy and parturition was studied in sheep and rabbits. The effects of epinephrine and various alpha- and beta- adrenergic blocking agents were also investigated. By comparing the results of these experiments, the authors conclude that the increase in epinephrine level (stress response of the organism) is the direct cause of the observed changes in uterine motility.Both stress and epinephrine caused either an activation or an inhibition of uterine motility. The direction of the effect depended on the ratio of sex-steroid concentrations in the plasma. Blood plasma levels of oestradiol-17β and of progesterone were determined in the sheep. Both stress and epinephrine inhibited uterine motility only when plasma levels of oestradiol-17β were very high (oestrogen-domination). The biological relevance and clinical implications are discussed.  相似文献
