The effects of light quality and irradiance, and supply of organic carbon and vitamins on the growth of two forms of Ecklonia radiata in tissue culture were examined. A callus of unpigmented cells developed over the cut surface of newly excised explants of stipe. This growth was best in the dark but stopped after 10 weeks. Pigmented, mainly filamentous clumps of cells developed from explants after several weeks in culture. These required light for growth, with growth being enhanced by increasing photon flux density up to 30 μmol photon m-2 s-1, with the active spectral component being red light (> 600 nm). Habitat forming algae play an important role in the ecology of temperate reefs worldwide. Despite this, our understanding of levels of gene flow within and among populations of algae is largely limited to studies on intertidal species; we know comparatively little about important habitat-forming subtidal algae. Here, we develop eight polymorphic microsatellite markers for the characterisation of population genetic diversity and structure in the subtidal kelp, Ecklonia radiata. This large macroalga is the most abundant habitat-forming kelp on the subtidal rocky reefs of temperate Australia and New Zealand where it forms extensive forests that support an astounding diversity of associated taxa.  相似文献

The metabolic significance of Se in plants is not well documented, though the presence of many selenoenzymes in bacteria and the essentiality of Se in higher animals is established. Since germination is an active process in plant growth and metabolism, the effect of Se was investigated in germinatingVigna radiata L, a nonaccumulating Sedeficient legume. Growth and protein were enhanced in seedlings supplemented with selenium (Se) as sodium selenite in the medium up to 1 μg/mL. The pattern of uptake of75Se in the differentiating tissues and the subcellular distribution were investigated. The percentage of incorporation of75Se was greater in the mitochondria at the lowest level (0.5 μg/mL) of Se supplementation compared to higher levels of Se exposure. Proteins precipitated from the postmitochondrial supernatant fractions, when separated by means of polyacrylamide gel electrophoresis (PAGE), indicated a major selenoprotein in the seedlings germinated at 2.0 μg/mL Se. In seedlings grown with supplemented Se, enhanced respiratory control ratio and succinate dehydrogenase activity were observed in the mitochondria of tissues, indicative of a role for Se in mitochondrial membrane functions.  相似文献

Assembly rules provide a useful framework for predicting patterns of community assembly under defined environmental conditions. Habitat created by canopy-forming algae (such as kelps) provides a promising system for identifying assembly rules because canopies typically have a large and predictable influence on understorey communities. Across >1,000 km of subtidal South Australian coastline, we identified natural associations between assemblages of understorey algae and (1) monospecific canopies of Ecklonia radiata, (2) canopies comprised of E. radiata mixed with Fucales (Cystophora spp. and Sargassum spp.), and (3) gaps among canopies of algae. We were able to recreate these associations with experimental tests that quantified the assembly of understorey algae among these three habitat types. We propose the assembly rule that understorey communities on subtidal rocky coast in South Australia will be (1) monopolised by encrusting coralline algae beneath monospecific canopies of E. radiata, (2) comprised of encrusting corallines, encrusting non-corallines, and sparse covers of articulated corallines, beneath mixed E. radiata-Fucales canopies, and (3) comprised of extensive covers of articulated corallines and filamentous turfs, as well as sparse covers of foliose algae and juvenile canopy-formers, within gaps. Consistencies between natural patterns and experimental effects demonstrate how algal canopies can act as a filter to limit the subsets of species from the locally available pool that are able to assemble beneath them. Moreover, the subsets of species that assemble to subtidal rocky substrata in South Australia appear to be predictable, given knowledge of the presence and composition of canopies incorporating E. radiata.  相似文献

This experiment was designed to examine the effects of aluminium (Al) on the growth of Pinus radiata (D. Don) and Eucalyptus mannifera subsp. mannifera (Mudie) seedlings in culture solutions in a glasshouse to help explain the failure of radiata pine trees on some acid, low fertility soils in Australia on which the native eucalypts flourish. Aluminium (Al) in culture solution increased the growth of roots and shoots of seedlings of both species but while growth of the eucalypt continued to increase with increases in Al to 2.222 μM, growth of the pine was largest at 370 μM Al. In addition to total root length, specific root length (length per unit dry weight), a measure of fineness of the root, increased in the eucalypt seedlings as the substrate Al increased. Growth of the shoots and roots of the pine in the absence of any added Al was extremely poor suggesting that Al, in low concentrations, may be an essential element or ameliorate some other factors in solution culture at low pH. Root and shoot concentrations of K increased with increasing Al, whilst Ca and Mg Concentrations decreased and Mn concentrations were unaffected in both species. Tissue Ca and Mg concentrations were 2 to 3 times higher in the eucalypt seedlings than the pine at all levels of added Al due to greater uptake of these elements by the eucalypt. In contrast, at the highest concentration of Al in the medium, shoot Al concentrations were lower in the cucalypt than in the pine due to a greater proportion of Al being retained in the eucalypt roots. These differences between the seedlings in terms of root growth and tissue cation concentrations may help explain the ability of eucalypt species to maintain vigorous growth on acid soils high in Al and low in Ca and P, where growth of the pines failed.  相似文献

Air-grown cells of the marine diatom Phaeodactylum tricornutum showed only 10% of the carbonic-anhydrase activity of air-grown Chlamydomonas reinhardtii. Measurement of carbonic-anhydrase activity using intact cells and cell extracts showed all activity was intracellular in Phaeodactylum. Photosynthetic oxygen evolution at constant inorganic-carbon concentration but varying pH showed that exogenous CO2 was poorly utilized by the cells. Sodium ions increased the affinity of Phaeodactylum for HCO 3 - and even at high HCO 3 - concentrations sodium ions enhanced HCO 3 - utilization. The internal inorganic-carbon pool (HCO 3 - +CO2] was measured using a silicone-oil-layer centrifugal filtering technique. The internal [HCO 3 - +CO2] concentration never exceeded 15% of the external [HCO 3 - +CO2] concentration even at the lowest external concentrations tested. Summary Addition of marl (CaCO3) and/or manure to an acid soil in pots caused lime-induced chlorosis inPinus radiata seedlings, especially after excessive irrigation for 15 days. Chlorotic symptoms and their intensity were found to be related more to soil moisture and to the HCO3 concentrations, than to percentage of free CaCO3, in the soil mixtures.Comparative chemical analysis showed lower total Fe and Mn concentrations and higher concentrations of cations and organic anions in the needles of seedlings with chlorotic symptoms than in the needles of healthy ones.  相似文献

Summary The changes between 9 and 40 years of age of the frequency distribution of tree diameters were studied in an experiment in a plantation of Pinus radiata D. Don in South Australia in which the spacing between trees at planting varied. Empirical functions were developed to relate tree diameter to growth rate in diameter, variance of growth rate in diameter and mortality rate. These functions were used in solving a forward diffusion equation to predict future diameter distributions. The contributions from each of the terms in the diffusion equation were examined and it was found that the drift (growth rate in diameter) term made the greatest contribution to the solution, whilst the diffusion (variance in growth rate) term had neglible effect. This implied that competition between individual trees for light was the dominant competitive process operating in this plantation, rather than below ground competition for soil resources. The ramifications of this finding to forest growth modelling are discussed.  相似文献

Summary The odacid fish Odax cyanomelas feeds on the kelp Ecklonia radiata, an important component of subtidal reef habitats on the central coast of New South Wales, Australia. Herbivory by Odax has a major impact on the structure and dynamics of discrete patches within larger stands of kelp at Cape Banks. This three-year study showed that each year, between August and October, approximately the same patches of kelp were denuded by preferential feeding on the meristem and primary laminae. This coincided with a variable pulse of Ecklonia recruits to the cleared patches, thereby generating patches of a single age-class of plants. Neighbouring areas of Ecklonia forest, non cleared by Odax, consisted of larger, perennial plants, which exhibited more gradual changes in abundance. The seasonal impact of Odax appeared to be due to a change in the behaviour of female Odax during their spawning period. Observations suggested that females aggregate at traditional sites prior to spawning with territorial males in adjacent areas of kelp forest. An alternative hypothesis, that Odax preferentially attacked stands of one-year old Ecklonia plants, was rejected by a field experiment; the establishment of experimental stands of one-year old plants did not lead to increased damage due to Odax or any change in the use of space by the fish. The generality of this effect of fish herbivory is unknown, but this and other Odax species are widely distributed throughout temperate Australia, where Ecklonia is the dominant laminarian alga. The effects of pulsed herbivory by Odax is contrasted to the more continuous grazing by sea urchins in the same system. The latter herbivore has been shown to maintain areas free of Ecklonia, the long-term effects of herbivory by Odax remain unclear.  相似文献

Biomass of nonliving, dried brown marine algae Sargassum natans, Fucus vesiculosus, and Ascophyllum nodosum demonstrated high equilibrium uptake of cadmium from aqueous solutions. The metal uptake of cadmium from aqueous solutions. The metal uptake by these materials was quantitatively evaluated using sorption isotherms. Biomass of A. nodosum accumulated the highest amount of cadmium exceeding 100 mg Cd(2+)/g (at the residual concentration of 100 mg Cd/L and pH 3.5), outperforming a commercial ion exchange resin DUOLITE GT-73. A new biosorbent material based on A. nodosum biomass was obtained by reinforcing the algal biomass by formaldehyde cross-linking. The prepared sorbent possessed good mechanical properties, chemical stability of the cell wall polysaccharides and low swelling volume. Desorption of deposited cadmium with 0.1-0.5M HCI resulted in no changes of the biosorbent metal uptake capacity through five subsequent adsorption/desorption cycles. Summary Segments of living stems from 2-year-old, fast growing Pinus radiata, established on a fertile ex-pasture, were examined in terms of their bending strength parallel to the grain. Stem anatomical and structural variables were examined to identify variables that could explain variation in bending strength. Half of the seedlings were physically supported to minimise the confounding effects of compression wood production in response to environmental stresses. Differences between seedlings and cuttings were examined. Variation in microfibril angle and lignin content was sufficient to account for 81% of the variation in bending strength. A positive correlation between both of these variables and elasticity was evident. Few differences were found to be caused by the support treatment or between seedlings and cuttings. The support treatment resulted in significantly fewer branches in the top most whorl, while cuttings had less branches in this whorl than seedlings.  相似文献

This study reports the effect of salinity and inoculation on growth, ion uptake and nitrogen fixation byVigna radiata. A soil ECe level of 7.5 dS m−1 was quite detrimental causing about 60% decline in dry matter and grain yield of mungbean plants whereas a soil ECe level of 10.0 dS m−1 was almost toxic. In contrast most of the studied strains of Rhizobium were salt tolerant. Nevertheless, nodulation, nitrogen fixation and total nitrogen concentration in the plant was drastically affected at high salt concentration. A noticeable decline in acetylene reduction activity occurred when salinity level increased to 7.5 dS m−1.  相似文献

We have established a reproducible culture system for callus formation and root development from juvenile stem segments of mung bean(Vigna radiata). In particular, we have studied the influence of plant growth regulators. Induction of calli from young stem explants was very effective on MS inorganic salts supplemented with 0.5 mg/L 2,4-D and 1.0 mg/L kinetin. In regenerating adventitious roots from callus tissues, we found that a combination of 0.75 mg/L NAA, 1.5 mg/L kinetin, and MS salts resulted in 20% efficiency. Histological examination showed that callus tissues originated from out-growths of the cambium rings through de-novo meristematic activity. Those rings were localized outside the vascular cambium. Adventitious roots that developed from root primordia originated from the center of the Callus masses. These primordia produced tracheid-like cells, which then became meristemoid cells for the cambium. Newly formed adventitious roots had the typical tetrarche actinostele type.  相似文献

Leader dieback associated with B deficiency in P. radiata D. Don plantations was treated with borax applied at rates of 50, 100 and 150 kg ha−1. This initially increased B in foliage from 5 to 40, 80 and 110 μg g−1 respectively, and was followed by a rapid decline and stabilisation at around 25 μg g−1 for the duration of the study. Annual fluctuations in foliage B levels were strongly correlated with rainfall during the preceding spring and summer. Uptake of N, P and K increased as a result of applied B and comparison of the distribution of these nutrients in crowns of fertilized and unfertilised trees six years after application indicated continued uptake of these nutrients probably as a result of improved root growth due to B. Foliage concentrations of B like N, P and K, increased in young needles towards the upper crown and this, together with a decline in needle concentrations of B as foliage aged, indicated some redistribution of B from older to new foliage. A limit of 5 μg g−1 was found below which little redistribution seems to occur. Application of B prevented further leader dieback, improved apical dominance and height growth and increased volume production by 25 m3 ha−1 at age 8 years. Pinus radiata is one of the most important forestry species in the southern hemisphere. This work describes the regeneration of this plant via somatic embryogenesis from immature zygotic embryos. To improve this process, somatic embryogenic cell suspensions were established in liquid media for the generation of material for embryo maturation. Each developmental stage of these suspensions was characterized by microscopy and their growth phases quantified. An alginate-containing medium was used as an encapsulation method for the somatic embryos that were then germinated as artificial seeds in vitro. The protocols described in this work are both useful and reliable for industrial purposes.  相似文献

Manganese peroxidase (MnP) from Phlebia radiata and glucose oxidase from Aspergillus niger were co-immobilized on porous silica beads. Immobilization of both enzymes on the same carrier provided an integrated system in which H2O2 required by MnP was produced by glucose oxidase. The immobilization process resulted in a decrease of both enzymatic activities and substrate affinities. However, immobilization improved the stability of MnP against H2O2 or high pH, as well as the storage stability of this enzyme.  相似文献

Summary Ethanol was produced by a strain ofPichia stipitis adapted to an inhibitory acid wood hydrolysate ofPinus radiata. The best ethanol productivity for batch cultures was 0.21 g/l h at 0.7% ethanol. Varying culture conditions increased ethanol concentration to 0.76%, however the productivity decreased to 0.18 g/l h. A decrease in ethanol concentration in the culture fluid was noted late in the batch which suggested ethanol catabolism. Values of kinetic parameters (K m,K s, max, andV max) were evaluated for this system. The use of calcium alginate immobilized cells in a continuous-flow stirred tank reactor lead to enhanced fermentative performance, namely a maximum productivity of 0.27 g/l h and 1.13% ethanol yield. The immobilized cells in continuous flow reactors represent an attractive option for fermenting sugars released by sulphuric acid hydrolysis ofP. radiata wood.  相似文献

Summary Severe dieback observed inP. radiata D. Don plantations during dry years in Southeastern Australia was found to be due to boron deficiency. Borax applied in spring (August 1982) at rates of 50, 100 and 150 kg ha–1 resulted in a marked increase in foliar B and an alleviation of B deficiency symptoms. During the dry summer following the application of borax no further dieback was observed and height growth of fertilized trees was significantly improved. Correction of B deficiency enhanced the uptake of P by the trees.  相似文献

White zones produced on biodegraded Pinus radiata wood chips were characterized by micro-localized-FTIR (Fourier Transformed Infra Red) spectroscopy and scanning electron microscopy. Both techniques permitted assignment of the white zones to a selective lignin removal process. Although both fungi studied have degraded lignin selectively in these restricted superficial areas, chemical analysis of the wood chips indicated that Ganoderma australe removed 16% of the initial amount of glucan at the 20% weight loss level. Ceriporiopsis subvermispora did not remove glucan at weight loss values below 17%. Prolonged biodegradation resulted in reduction of white zones by G. australe, and increased white zones from C. subvermispora decayed samples. In a high-salt soluble fraction of the total protein from single seeds of Pinus radiata, up to 45 polypeptides were resolved on SDS-polyacrylamide gels. At least one-fifth of these polypeptides showed variation between seeds. In the 27,000–29,000 dalton region, two polypeptides were inherited as codominant alleles at a single locus and were shown to assort independently of another seed protein locus and three allozyme loci. A survey of 120 individuals from the five known native populations of P. radiata in California detected only the 27K and 29K alleles at the locus. In all populations, the 29K allele predominated, and the two island populations were monomorphic for the 29K allele. The 27 and 29 kdalton polypeptides were shown to have very similar amino acid sequences, and the allelic difference at this locus is most probably in the gene sequence for the polypeptide.  相似文献
