Δ8途径是合成多不饱和脂肪酸的替代途径,Δ8-脂肪酸脱氢酶是该途径的关键酶之一。根据已报道的Δ8-脂肪酸脱氢酶基因设计引物,分别从小眼虫藻基因组DNA和cDNA中扩增得到该基因片段,序列分析表明:结构基因长1 266 bp,编码421个氨基酸;该基因没有内含子,比已经报道的Δ8-脂肪酸脱氢酶基因长6 bp,并且N末端序列也有所不同。利用酿酒酵母的载体pYES2.0构建Δ8-脂肪酸脱氢酶表达载体pYEFD,并转化到营养缺陷型酿酒酵母菌株INVSc1中,在选择培养基中筛选得到酿酒酵母转化菌株YD8。YD8在合适的培养条件下,添加外源底物二十碳二烯酸和二十碳三烯酸并诱导基因表达。脂肪酸甲酯气相色谱分析表明小眼虫藻Δ8-脂肪酸脱氢酶基因在酿酒酵母中获得了高效表达,将二十碳二烯酸和二十碳三烯酸分别转化成二高-γ-亚麻酸和二十碳四烯酸,其底物转化率分别达到了31.2%和46.3%。  相似文献

Δ^6-脂肪酸脱氢酶是一种膜整合蛋白,也是多不饱和脂肪酸合成途径中的限速酶。在前期工作中,通过RT-PCR和RACE技术,从少根根霉NK300037中克隆到一个潜在编码Δ^6-脂肪酸脱氢酶的序列,序列和功能分析结果表明该序列具有一个长度为1377bp、编码由458个氨基酸组成、大小为52kD的新的Δ^6-肪酸脱氢酶基因。把少根根霉Δ^6-脂肪酸脱氢酶基因(RAD6)亚克隆到表达载体pPIC3.5K,构建重组表达载体pPICRAD6,并转化到毕赤酵母菌株GS115进行表达。提取酵母细胞总脂肪酸和进行甲酯化,经气相色谱和气相色谱-质谱连用分析表明,目的基因的编码产物能将C16:1、C17:1、C18:1、亚油酸和α-亚麻酸在△6和7位间特异性脱氢而引入一个新的双键,生成更高不饱和的脂肪酸,该催化反应没有链长特异性,只有键位特异性。此外,按Kozak序列特点,改变目的基因转译起始密码子周边序列结构,并把改变后序列导入毕赤酵母GS115中进行功能表达分析,结果表明在毕赤酵母中这种改变同样能提高目的基因的表达水平。综合所有分析结果表明,巴斯德毕赤酵母更适合用来综合分析Δ^6-脂肪酸脱氢酶基因的功能。  相似文献

通过气相色谱法(GC)快速分析8种真菌的脂肪酸成分,发现匍枝根霉(Rhizopus stolonifer)具有较高的γ-亚麻酸含量,利用RT-PCR和RACE方法获得了全长为1475bp的匍枝根霉△6-脂肪酸脱氢酶基因的cDNA序列,其中开放阅读框为1380bp,编码459个氨基酸。生物信息学分析所克隆的基因具有△6-脂肪酸脱氢酶的典型结构:N端具有细胞色素b5结构、具有3个保守的组氨酸区序列和跨膜结构;把该基因的开放阅读框序列连接到表达载体pYES2.0上,构建重组表达载体pYRnD6D,并将其转入缺陷型酿酒酵母INVScl中进行表达。GC分析表明,该序列在酵母中获得了表达,表达产物表现出△6-脂肪酸脱氢酶的酶学活性,能将底物亚油酸转化为γ-亚麻酸。新生成的γ-亚麻酸占酵母细胞总脂肪酸的12.25%。  相似文献

根据真菌△^6 -脂肪酸脱氢酶基因保守的组氨酸Ⅱ区和Ⅲ区附近保守序列设计兼并引物进行RT-PCR,得到雅致枝霉(Thamnidium elegans)As3.2806△^6 -脂肪酸脱氢酶基因459bp部分cDNA序列,然后通过快速扩增cDNA末端技术(RACE)向两端延伸得到1504bp的△^6 -脂肪酸脱氢酶基因全长cDNA序列。序列分析表明有一个1377bp、编码459个氨基酸的开放阅读框TED6。推测的氨基酸序列与已知其他真菌的△^6 -脂肪酸脱氧酶基因的氨基酸序列比对,具有3个组氨酸保守区、2个疏水区及N末端细胞色素b5融合区。将此编码区序列亚克隆到酿酒酵母缺陷型菌株INVSel的表达载体pYES2.0中,构建表达载体pYTED6,并在酿酒酵母INVSel中异源表达。通过气相色谱(GC)和气相色谱,质谱(GC-MS)分析表明,该序列在酿酒酵母中获得表达,产生γ-亚麻酸(GLA)的含量占酵母总脂肪酸的7.5%。证明此序列编码的蛋白能将外加的亚油酸转化为γ-亚麻酸,是一个新的有功能的△^6 -脂肪酸脱氢酶基因(GenBank.AY941161)。  相似文献

Construction and sequence of cDNA for rat liver stearyl coenzyme A desaturase   总被引:23,自引:0,他引:23  
Hepatic poly(A+) RNA from rats induced for stearyl-CoA desaturase was used for primer-extension of cDNA coding for stearyl-CoA desaturase. Previously, Northern blot analysis showed that translatable desaturase mRNA is 4,900 nucleotides in length (Thiede, M. A., and Strittmatter, P. (1985) J. Biol. Chem. 260, 14459-14463). Six overlapping cDNAs, ranging from 850 to 1450 bases, were used to compile the 4,689-nucleotide sequence. The cDNA includes a 1,074-base open reading frame coding for 358 amino acids, corresponding to a molecular mass of 41,400 daltons. Positive identification of this open reading frame was accomplished by matching the amino acid sequence of both amino-terminal and cyanogen bromide peptides of the purified enzyme with regions of the sequence deduced from the cDNA. Amino acid composition data from the cDNA compares well with that from the desaturase. The protein contains 62% hydrophobic amino acids. An interesting feature of this mRNA is the 3,500-base 3' noncoding region, which has been localized on a single 3' exon by Southern blot analysis.  相似文献

The similarities between delta12- and delta5-fatty acyl desaturase sequences were used to construct degenerate primers for PCR experiments with cDNA transcribed from mRNA of developing borage seeds. Screening of a borage seed cDNA library with an amplified DNA fragment resulted in the isolation of a full-length cDNA corresponding to a deduced open-reading frame of 446 amino acids. The protein showed high similarity to plant delta8-sphingolipid desaturases as well as to the delta6-fatty acyl desaturase from Borago officinalis. The sequence is characterized by the presence of a N-terminal cytochrome b5 domain. Expression of this open-reading frame in Saccharomyces cerevisiae resulted in the formation of delta8-trans/cis-phytosphingenines not present in wild-type cells, as shown by HPLC analysis of sphingoid bases as their dinitrophenyl derivatives. GLC-MS analysis of the methylated di-O-trimethylsilyl ether derivatives confirmed the presence of delta8-stereoisomers of C18- and C20-phytosphingenine. Furthermore, Northern blotting showed that the gene encoding a stereo-unselective delta8-sphingolipid desaturase is primarily expressed in young borage leaves.  相似文献

γ-亚麻酸(GLA,C18:3△6,9,12)是由△6-脂肪酸脱氢酶以亚油酸(LA,C18:2△9,12)为底物,在C6位脱氢形成的。由于在人体中,γ-亚麻酸是花生四烯酸、前列腺素类和白三烯类等生理活性物质的前体物,而深黄被孢霉是目前用于微生物发酵生产γ-亚麻酸的主要菌株。本文根据脂肪酸脱氢酶的保守区设计引物,利用反转录聚合酶链式反应从丝状真菌深黄被孢霉中克隆了编码△6-脂肪酸脱氢酶的cDNA,全长为1374个核苷酸,编码457 个氨基酸,但与其他位点的脂肪酸脱氢酶不同的是, △6-脂肪酸脱氢酶在其序列的 N 端特有细胞色素 b5(Cytb5)区。这是国际上对深黄被孢霉△6-脂肪酸脱氢酶基因的首次报道。  相似文献

Stearoyl-CoA desaturase (EC 1.14.99.5) is a key enzyme in the biosynthesis of polyunsaturated fatty acids and the maintenance of the homeoviscous fluidity of biological membranes. The stearoyl-CoA desaturase cDNA in milkfish (Chanos chanos) was cloned by RT-PCR and RACE, and it was compared with the stearoyl-CoA desaturase in cold-tolerant teleosts, common carp and grass carp. Nucleotide sequence analysis revealed that the cDNA clone has a 972-bp open reading frame encoding 323 amino acid residues. Alignments of the deduced amino acid sequence showed that the milkfish stearoyl-CoA desaturase shares 79% and 75% identity with common carp and grass carp, and 63%–64% with other vertebrates such as sheep, hamsters, rats, mice, and humans. Like common carp and grass carp, the deduced amino acid sequence in milkfish well conserves three histidine cluster motifs (one HXXXXH and two HXXHH) that are essential for catalysis of stearoyl-CoA desaturase activity. However, RT-PCR analysis showed that stearoyl-CoA desaturase expression in milkfish is detected in the tissues of liver, muscle, kidney, brain, and gill, and more expression sites were found in milkfish than in common carp and grass carp. Phylogenic relationships among the deduced stearoyl-CoA desaturase amino acid sequence in milkfish and those in other vertebrates showed that the milkfish stearoyl-CoA desaturase amino acid sequence is phylogenetically closer to those of common carp and grass carp than to other higher vertebrates.  相似文献

克隆并在酵母中表达两个不同N段序列长度的匍枝根霉△6-脂肪酸脱氢酶重组子,其中长序列的重组子LYRnD6D是从匍枝根霉中克隆的△6-脂肪酸脱氢酶基因,编码459个氨基酸,N端序列为MSTLDRQSIFTIKELESISQRIHDG-DEEAMKFIII;短序列重组子SYRnD6D是预测的匍枝根霉△6-脂肪酸脱氢酶基因的ORF序列,N端序列为MKFIIIDKKVY,编码430个氨基酸;两个重组子均具有△6-脂肪酸脱氢酶保守的组氨酸序列和HPGG序列,长序列的N端比短序列长29个氨基酸残基(MSTLDRQSIFTIKELESISQRIHDGDE-EA)。两个重组子在缺陷型酵母中均得到了的表达,产生了γ-亚麻酸。利用酶的相对活力比较两个重组子在同一温度下的稳定性,长序列重组子的酶在15℃下反应4h后相对活力仍有74%,而短序列酶的相对活力只有43%,所以长序列重组子酶在低温下比短序列酶稳定性高,是因为长序列多出的氨基酸序列增加了酶的稳定性。  相似文献

The delta 6-desaturase gene was cloned from Rhizopus stolonifer, which could accumulate up to 49% of gamma-linolenic acid (GLA, C18:3 Δ6,9,12) to the total fatty acids. The cloned DNA contains a 1,380 bp open reading frame encoding a protein of 460 amino acids, which showed high similarity to those of fungal delta 6-desaturases with three conserved histidine-rich motifs and HPGG motif. Notably, this deduced sequence had a shorter C-terminus. Results demonstrated that the cDNA sequence exhibited delta 6-desaturase activity by accumulation of about 22.4 % of GLA to the total fatty acids in the recombinant Pichia pastoris strain GS115.  相似文献

A DNA fragment was cloned from the fungal strain, Mortierella alpina 1S-4 (which is used industrially to produce arachidonic acid), after PCR amplification with oligonucleotide primers designed based on the sequence information for delta6-desaturase genes (from borage and Caenorhabditis elegans), which are involved in the desaturation of linoleic acid (delta9, delta12-18:2) to gamma-linolenic acid (delta6, delta9, delta12-18:3). This fragment was used as a probe to isolate a cDNA clone with an open reading frame encoding 457 amino acids from a M. calpina 1S-4 library. The predicted amino-acid sequence showed similarity to those of the above delta6-desaturases, and contained a cytochrome b5-like domain at the N-terminus, being different from the yeast delta9-desaturase which has the corresponding domain at the C-terminus. The full-length cDNA clone was expressed under the control of the amyB promoter in a filamentous fungus, Aspergillus oryzae, resulting in the accumulation of gamma-linolenic acid (which was not detected in the control Aspergillus) to the level of 25.2% of the total fatty acids. These findings revealed that the recombinant product has delta6-desaturase activity. The Mortierella delta6-desaturase is the first to be reported in fungi.  相似文献

The redbanded leafroller moth, Argyrotaenia velutinana (Lepidoptera: Tortricidae) uses a 92:8 mixture of (Z)-11- and (E)-11-tetradecenyl acetate in its pheromone blend. These are produced in the abdominal pheromone gland from the corresponding acids, which are biosynthesized in the gland in a 3:2 Z/E ratio by desaturation of myristoyl CoA. The delta 11 desaturase involved in this reaction exhibits unusual substrate and stereospecificities in specifically producing Z11 and E11 isomers of tetradecenoic acid, and exhibiting no activity with C16 and C18 precursor acids. This report describes the cloning and expression of the redbanded leafroller moth delta 11 desaturase, and compares its amino-acid sequence to those of other known insect Z9, Z10, Z11, and E11 desaturases. The metabolic Z9 desaturase from fat body tissue also was cloned and expressed, and found mainly to produce Z9-16:Acid and Z9-18:Acid. The open reading frame of the delta 11 desaturase encodes a protein with 329 amino acids, whereas the open reading frame of the Z9 desaturase encodes a protein with 351 amino acids. Addition of this new delta 11 desaturase with its different substrate and regiospecificites to the databank of characterized integral-membrane desaturases will be key in efforts to determine amino-acid mutations responsible for the wide array of unsaturated fatty-acid products.  相似文献

Delta 6-fatty acid desaturase (D6DES) is used in the synthesis of polyunsaturated fatty acids (PUFAs) from microorganisms to higher animals, including arachidonic acid (ARA) and eicosapentaenoic acid (EPA). A 1,338 bp full-length cDNA encoding D6DES was cloned from Acanthopagrus schlegeli (AsD6DES) through degenerate- and RACE-PCR methods. A recombinant vector expressing AsD6DES (pYES-AsD6DES) was subsequently constructed and transformed into Saccharomyces cerevisiae to test the enzymatic activity of AsD6DES towards the production of n-6 and n-3 fatty acids. The exogenously expressed AsD6DES produced γ-linolenic acid (18:3 n-6) and stearidonic acid (18:4n-3) at 26 and 36% from exogenous linoleic acid (18:2n-6) and α-linolenic acid (18:3n-3), respectively, indicating that it is essentially a delta 6-fatty acid desaturase.  相似文献

A cDNA clone encoding delta 4-3-ketosteroid 5 beta-reductase was isolated from rat liver cDNA libraries using antibodies specific for the enzyme and oligonucleotides as probes. The cDNA contained 981-base pair open reading frame encoding 327 amino acid residues (Mr 37,376) and an unusually long 3'-untranslated region rich in AT sequence in the total length of 3189 base pairs. The predicted amino acid sequence contains the sequences similar to the putative NADPH- and steroid-binding regions.  相似文献
