The activity of trypsin-like proteinases and trypsin inhibitors was measured in fruiting bodies of various species of basidial fungi (Basidiomycetes). Fruiting bodies of all fungi contained these enzymes, with the exceptions of polypore (Coriolus versicolor (Fr.) Karst) and hedgehog fungus (Hericium erinaceus (Fr.) Quel), belonging to the families Polyporaceae and Hericiaceae, respectively, in which the enzyme activities were barely detectable. The activity of trypsin-like proteinases was the highest in fruiting bodies of Boletaceae and Agaricaceae. Fruiting bodies of all fungi contained trypsin inhibitors. The highest activity of trypsin inhibitors was detected in basidiomycetes of the families Boletaceae, Agaricaceae, and Pleurotaceae, including Boletus castanus (Fr.) Karst, orange-cap boletus (Leccinum aurantiacum (Fr.) Sing), and brown-cap boletus (Leccinum melanum (Fr.) Karst).  相似文献

A detailed characteristic of the beetle fauna associated with Fomitopsis pinicola (Sw.: Fr.) Karst. (Basidiomycetes, Aphyllophorales) in the Urals and Transurals is given. Thirty species from 14 families have been revealed, the commonest species including Eridaulus jacquemarti Mel., Ennearthron laricinum (Mel.) (Ciidae), Dorcatoma dresdensis Hbst., D. lomnickii Rtt., D. punctulata Muls. (Anobiidae), Diaperis boleti (L.) (Tenebrionidae), Scaphisoma agaricinum (L.), S. inopinatum Löbl, S. subalpinum Rtt. (Scaphidiidae), Ostoma ferruginea (L.), and Peltis grossa (L.) (Peltidae). The main trends in the ecological and trophic specialization of mycetophilous beetles are discussed, and some regularities of formation of mycetophilous complexes at different stages of the trees fungi fruit body development are revealed.  相似文献

Physicochemical properties, composition, and structure of the polysaccharides synthesized by the fungi Lentinus edodes, Ganoderma lucidum, and Crinipellis schevchenkovi in a submerged culture were studied. It has been demonstrated that the exo-and endopolysaccharides of submerged mycelia of the fungi studied are similar to the β-1,3-D-glucans of the fruiting bodies.  相似文献

The beetle complex associated with Daedaleopsis confragosa (Bolton: Fr.) Schrot. (Basidiomycetes, Aphyllophorales) in the Urals and Transurals is characterized. A total of 54 species of 16 families were found in this complex, the commonest species including Cis comptus Gyll., Sulcacis affinis (Gyll.) (Ciidae), Tritoma subbasalis (Rtt.), Dacne bipustulata Thunb. (Erotylidae), Mycetophagus multipunctatus F., M. piceus (F.) (Mycetophagidae), and Thymalus oblongus Rtt. (Trogossitidae). The main trends of the ecological and trophic specialization of the mycetophilic beetles are discussed; some features of formation of their complexes at different stages of the tree-fungi fruit bodies formation were established.  相似文献

THE polysaccharides obtained from some basidiomycetes, Pleurotus ostreatus (Jacq. ex Fr.) Quel.1, Flammulina velutipes (Curt, ex Fr.) Sing.2, Lentinus edodes (Berk.) Piptoporus betulinus is a wood-rotting basidiomycete used in medicine and biotechnology. However, to date, no indoor method for cultivation of this mushroom fruiting bodies has been developed. Here we present the first report of successful production of P. betulinus mature fruiting bodies in artificial conditions. Four P. betulinus strains were isolated from natural habitats and their mycelia were inoculated into birch sawdust substrate supplemented with organic additives. All the strains effectively colonized the medium but only one of them produced fruiting bodies. Moisture and organic supplementation of the substrate significantly determined the fruiting process. The biological efficiency of the P. betulinus PB01 strain cultivated on optimal substrate (moisture and organic substance content of 55 and 65 and 25 or 35 %, respectively) ranged from 12 to 16 %. The mature fruiting bodies reached weight in the range from 50 to 120 g. Anticancer properties of water and ethanol extracts isolated from both cultured and nature-derived fruiting bodies of P. betulinus were examined in human colon adenocarcinoma, human lung carcinoma and human breast cancer cell lines. The studies revealed antiproliferative and antimigrative properties of all the investigated extracts. Nevertheless the most pronounced effects demonstrated the ethanol extracts, obtained from fruiting bodies of cultured P. betulinus. Summarizing, our studies proved that P. betulinus can be induced to fruit in indoor artificial culture and the cultured fruiting bodies can be used as a source of potential anticancer agents. In this respect, they are at least as valuable as those sourced from nature.  相似文献

Empirical approach was adopted to examine the interaction of a little and finger millet inhibitors with digestive proteinases ofdifferent field and storage pests. Accordingly, gut proteinases of four storage and phytophagous pests were characterized andtheir inhibition by finger millet (Eluesine coracana Gaertneri) and little millet (Panicum sumatrense Roth) inhibitors has beeninvestigated. The Callosobruchus sp showed acid proteinase with pH optimum of 3.5 and Sitophilus oryzae showed pHoptimum of 4.5 apart from their alkaline proteinases with pH 8.5. The proteinases of Tribolium castaneum had wider pHoptimum from 5.5 to 8.5. However, all lepidopteran insect proteinases had pH optima ranging from 8.5 to 10.5. The optimumtemperature was found to be 30 to 40 °C. The inhibitory activities of little and finger millet inhibitors towards the insectproteinases are very low except notable level inhibition of proteinases of some insects. The gut proteinase zymogram ofdifferent insects revealed 2 – 6 isozymes and the inhibitors moderately inhibited all the isozymes of insects tested.  相似文献

A trypsin inhibitor gene (CoTI1) from Cassia obtusifolia was isolated and the deduced amino acid sequence was attributed to the Kunitz-type trypsin inhibitor. The recombined CoTI1, expressed in E. coli, exhibited strong inhibitory effect on bovine trypsin and trypsin-like proteases from Helicoverpa armigera, Spodoptera exigua, and Spodoptera litura. CoTI1 thus presents insecticidal properties that may be useful for the genetic engineering of plants. Leu84, Arg86 and Thr88 were predicted as three key residues by molecular modeling in which Arg86, inserted into the substrate pocket of trypsin, interacted directly with residue Asp189 of trypsin causing the specific inhibition against trypsin. The predicted results were confirmed by site-directed mutagenesis with L84A, R86A and T88A, respectively. The substantial changing expression level of CoTI1 under salt, drought and abscisic acid treatment suggested that CoTI1 might play important role in the resistance against abiotic stress.  相似文献

Higher Basidiomycota have been used in natural medicine throughout the world for centuries. One of such fungi is Fomitopsis betulina (formerly Piptoporus betulinus), which causes brown rot of birch wood. Annual white to brownish fruiting bodies of the species can be found on trees in the northern hemisphere but F. betulina can also be cultured as a mycelium and fruiting body. The fungus has a long tradition of being applied in folk medicine as an antimicrobial, anticancer, and anti-inflammatory agent. Probably due to the curative properties, pieces of its fruiting body were carried by Ötzi the Iceman. Modern research confirms the health-promoting benefits of F. betulina. Pharmacological studies have provided evidence supporting the antibacterial, anti-parasitic, antiviral, anti-inflammatory, anticancer, neuroprotective, and immunomodulating activities of F. betulina preparations. Biologically active compounds such as triterpenoids have been isolated. The mushroom is also a reservoir of valuable enzymes and other substances such as cell wall (1→3)-α-d-glucan which can be used for induction of microbial enzymes degrading cariogenic dental biofilm. In conclusion, F. betulina can be considered as a promising source for the development of new products for healthcare and other biotechnological uses.  相似文献

The specificity of lectins from the basidiomycete Lentinus edodes (Berk.) Sing [Lentinula edodes (Berk.) Pegler] during solid-state cultivation was found to be maximum to carbohydrates detected in the pyri-dine-soluble fraction of mycelium during the brown mycelial film stage. The carbohydrate composition of the mycelium (i.e., its content of maltose, rhamnose, mannitol, and inositol) was found to be different upon normal fruiting body formation and when the nonpigmented mycelium produced defective fruiting bodies.  相似文献

Cordyceps militaris is a model species of Cordyceps fungi, and has been traditionally used as an edible and medicinal fungus due to its richness of bioactive and pharmacological metabolites. The fruiting bodies of this fungus are widely used as healthy food and nutrition supply. In industrial production, fruiting bodies are cultivated on artificial media, but their yield and quality are usually affected by the quality of fungal strains. In this study, the effect of colony growth rate of the fungal strains, fungal age and repeated subculturing on the fungal biomass accumulation was investigated. The results indicated that the fungal biomass was positively correlated with the colony growth rate and not affected by fungal age and the repeated subculturing. The preservation conditions for stock cultures, including choice of cultures, lyophilization, temperature and protective agents were optimized based on the mycelial formation and conidia production in artificial inoculum. The development of fruiting bodies from the fungal strains stored under the optimized preservation conditions were further analyzed to determine the ideal time period of preservation. Results indicated that storing the fungus at 4 °C could maintain the fungal vitality and fruiting body producing capacity for at least 12 months. This study established practical criteria of fungal inoculum for artificial cultivation of fruiting body and provided a simple and efficient preservation method for C. militaris. The results may shed light on preservation for other Cordyceps species and other edible fungi.  相似文献

The interest in proteases secreted by mycelial fungi is due to several reasons of which one of the most important is their involvement in the initiation and development of the pathogenic process. A comparison of saprophytic and phytopathogenic mycelial fungi revealed one characteristic feature, namely, the appearance of a new trypsin-like activity in phytopathogens that is absent in saprophytes. To clear up the question of whether the degree of pathogenicity of a fungus is related to the activity of secreted trypsin-like protease, several species of Fusarium of various pathogenicity were compared. In two species, F. sporotrichioides (which causes ear fusariosis of rye) and F. heterosporum (the causative agent of root rot in wheat), a clear correlation between the activity and pathogenicity was revealed: the more pathogenetic F. sporotrichioides exhibited a higher extracellular trypsin-like activity than the less pathogenetic species F. heterosporum. Thus, the presence of trypsin-like activity in a saprotroph-pathogen pair may be an indicator of the pathogenicity of a fungus; in some cases, the value of this activity may indicate the degree of its pathogenicity. This suggests that trypsin-like proteases specific to phytopathogens are directly involved in the pathogenetic process, probably, through interaction with the “sentry” protein or the product of the resistance gene.  相似文献

Tricholoma matsutake is an ectomycorrhizal fungus usually associated with Pinus densiflora in South Korea. Fruiting bodies (mushrooms) of T. matsutake are economically important due to their attractive aroma; yet, T. matsutake is uncultivatable and its habitat is rapidly being eradicated due to global climate change. Root-associated bacteria can influence the growth of ectomycorrhizal fungi that co-exist in the host rhizosphere and distinctive bacterial communities are associated with T. matsutake. In this study, we investigated how these bacterial communities affect T. matsutake growth by isolating bacteria from the roots of P. densiflora colonized by ectomycorrhizae of T. matsutake and co-culturing rootassociated bacteria with T. matsutake isolates. Thirteen species of bacteria (27 isolates) were found in pine roots, all belonging to the orders Bacillales or Burkholderiales. Two species in the genus Paenibacillus promoted the growth of T. matsutake in glucose poor conditions, likely using soluble metabolites. In contrast, other bacteria suppressed the growth of T. matsutake using both soluble and volatile metabolites. Antifungal activity was more frequent in glucose poor conditions. In general, pine rhizospheres harbored many bacteria that had a negative impact on T. matsutake growth and the few Paenibacillus species that promoted T. matsutake growth. Paenibacillus species, therefore, may represent a promising resource toward successful cultivation of T. matsutake.  相似文献

Using monokaryotic offspring from several dikaryotic parental strains, the phenomenon of monokaryotic fruiting has been previously analysed in the commercially cultivated high-quality edible mushroom Agrocybe aegerita, revealing a variety of monokaryotic fruiting types. Here, we report a single dikaryotic A. aegerita strain, A. aegerita AAE-3, and 40 monokaryons derived from it, which exhibit a wide spectrum of monokaryotic fruiting types, including a rare, previously unknown type. Advantageously, the selected parental strain A. aegerita AAE-3 completes its life cycle within three weeks by the formation of dikaryotic fruiting bodies of typical agaric morphology on malt extract agar plates. In order to morphologically compare normal dikaryotic fruiting to monokaryotic fruiting, histology was performed from all dikaryotic fruiting body development stages and all fruiting types of monokaryotic origin. No clamp connections or dikaryotic hyphae were observed within the plectenchyma of monokaryotic fruiting stages. Among the monokaryotic fruiting types of the A. aegerita AAE-3-derived monokaryons, we also characterised the rare ‘stipe type’ here described as ‘elongated initials type’ as no differentiation into a future cap and stipe was seen. The two mating-compatible monokaryotic strains representing the extremes of the fruiting type spectrum observed, A. aegerita AAE-3-13 (‘mycelium type’) and A. aegerita AAE-3-32 (‘abortive?+?true homokaryotic fruiting fruiter type, AHF?+?THF fruiter type’), were also found to readily produce oidia (arthrospores). In order to obtain a set of mating-compatible monokaryons covering the whole observed spectrum of monokaryotic fruiting, the two monokaryons A. aegerita AAE-3-40 (‘initials type’) and A. aegerita AAE-3-37 (‘elongated initials type’) have been selected for their mating compatibility with A. aegerita AAE-3-32 and A. aegerita AAE-3-13, respectively. Together with the parental dikaryotic strain A. aegerita AAE-3, this set of standard monokaryons could prove useful for studies exploring the factors regulating monokaryotic fruiting in comparison to dikaryotic mushroom formation.  相似文献

Truffle (Tuber spp.) cultivation is based on raising mycorrhizal trees in greenhouses that have been inoculated with suspensions of ascospores. The problem with this is that pests, pathogens, and other mycorrhizal fungi can contaminate the trees. Furthermore, because ascospores are produced sexually, each plant potentially has a different genetic mycorrhizal makeup from each other so tailoring the mycorrhizal component of plants to suit a particular set of soil and climatic conditions is out of the question. Here, we report on the production of Tuber borchii-mycorrhized plants using pure cultures, establishing a truffière with these and subsequent production of its fruiting bodies. This study opens up the possibility of producing commercial numbers of Tuber-mycorrhized trees for truffle cultivation using mycelial inoculation techniques. It also poses questions about the mechanism of fertilization between the different strains which were located in different parts of the experimental truffière.  相似文献

Endophytes play an important role in the growth and development of the host. However, the study of endophytes is mostly focused on plants and animals, and reports on microorganisms associated with fungus are relatively rare. We studied the microorganisms associated with Tricholoma matsutake fruiting bodies picked from three main T. matsutake-producing areas in Sichuan, China, by both culture-dependent and culture- independent methods. Altogether 13 fungus, 15 yeast and 14 bacterial strains were isolated from the T. matsutake fruiting bodies. The most abundant cultivable fungus, yeast and bacteria isolates were assigned as Fusarium solanis, Cryptococcus sp. and Pseudomonas sp., respectively. Terminal-restriction fragment length polymorphism analysis (T-RFLP) showed that the bacteria in T. matsutake were abundant and diverse. Betaand gamma-proteobacteria, Acidobacteria, Bacteroidetes and Sphingobacterium were found in samples from all collecting sites. Among these bacteria, we may find some strains that can promote the growth of T. matsutake.  相似文献

This study evaluated pre-inoculation to induce fruiting body production in wood-decaying fungi, using two species that readily create basidiomata in the laboratory: Antrodia sinuosa and Junghuhnia luteoalba. A new method was implemented in which the substrate was modified by species-specific pre-inoculation. We found that the use of the pre-inoculated substrate reduced the length of time necessary for the production of fruiting bodies. Under these conditions, both species produced basidiomata in 1–2 weeks, whereas over 2 months was required on a non-modified substrate. Thus we conclude that the induction of fruiting body production by pre-inoculation is possible, and we discuss the effect of the substrate modification.  相似文献

Phanerochaete velutina and the closely related species P. alnea, P. alnea subsp. lubrica, and P. rhodella are identified as distinct species based on morphological criteria and molecular analyses. Besides differences in macroscopic and microscopic basidiocarp features, the taxa differ in distribution and ITS sequence. Phanerochaete alnea is widely distributed in Eurasia and North America, whereas P. alnea subsp. lubrica is limited to the Pacific Coast of North America. Phanerochaete velutina is known from Eurasia and Alaska, and P. rhodella is found in North America only. Nomenclatural problems with the name Phanerochaete P. Karst. are discussed, and type specimens for Thelephora alnea Fr., the generic type of Phanerochaete, and Peniophora karstenii Massee are selected. Phanerochaete alnea, P. alnea subsp. lubrica, and P. velutina are described and illustrated. In addition, the synonymy of Phanerochaete robusta and P. aurantiobadia is confirmed.  相似文献
