The most common European gastropod species, Arion vulgaris, is one of the most troublesome pests for private garden owners and commercial agriculturists. The sticky and hard to remove secretion produced by these animals allows them to overcome most artificial and natural barriers. However, this highly adherent biopolymer has recently shown great potential for novel wound-healing applications in medicine. Nevertheless, our knowledge of the underlying gland system is still limited and few studies on the ventral gland system are available. We studied the lateral and ventral pedal glands in Arion vulgaris to determine their secretory content histochemically and through lectin assays. Using these histological and histochemical methods we differentiate five gland types with different mucus composition in the lateral pedal region of the foot of Arion vulgaris. These contain sulphated and carboxylated mucosubstances (positive Alcian blue staining) but lack hexose-containing mucosubstances (negative PAS staining). In the ventral pedal region, four gland types can be differentiated producing sulphated and carboxylated mucosubstances. Within the ventral mucus, a high affinity for the lectins PNA and WGA is observed. While the lateral glands are histochemically negative for PAS, a positive staining with the lectin JAC is observed. Arion vulgaris shows clear morphological differences from other arionid species. This raises the question whether the variation in the chemistry of the secretory material and mucus composition is the result of different functions and/or is related to the animals' different environmental conditions. A comparison of some glands of Arion vulgaris with those of the helicid species Helix pomatia and Cepaea hortensis indicates morphological similarities.  相似文献

The presence of glycosaminoglycans was examined in two mollusks (Pulmonates): the terrestrial garden snail, Helix aspersa, and the opishtobranchian sea slug, Aplysia californica and also in the leech (Hirudinea, Erpobdellidae, Nephelopsis obscura). Organs in the garden snail contained predominately chondroitin sulfate and heparan sulfate as a lesser component. The ctenidium of the sea slug contained mainly chondroitin sulfate and a compound which migrated on electrophoresis as heparin but additional data indicated that it could also represent a highly sulfated form of heparan sulfate. The foregut contained only the heparin-like polymer. No standard glycosaminoglycan could be identified in the leech although a polydispersed polysaccharide containing uronic acid, hexosamine and sulfate was shown to be present. A detailed analysis of the heparan sulfate isolated from the garden snail is also given.  相似文献

The Arion hortensis agg. (Mollusca: Pulmonata) are major slug pests of agriculture and horticulture throughout much of the temperate World. A monoclonal antibody was created that could specifically identify and quantify the remains of these slugs within the carabid predator Pterostichus melanarius Illiger, a polyphagous species found in arable crops in Europe and North America. The antibody was shown to be specific for the two species of pest slug within this species aggregate, Arion distinctus Mabille and A. hortensis Férussac, and did not cross-react with any other invertebrates tested, including other slugs in the same genus. It proved to be highly sensitive in an enzyme-linked immunosorbent assay. The mean detection period for the remains of A. distinctus in predator foregut samples was 3.28 days at 16°C. However, A. distinctus remains could be identified in male carabids for 30% longer than in females, emphasizing the importance of separate analyses. The value of the antigen half-life as a measure of the detection period was questioned because analysis of covariance showed that the regression slopes for antigen decay were not significantly different between the two sexes, giving very similar half-lives (approx 18.5 h for females and 19 h for males). Calculated quantities of soluble slug proteins in predator foreguts declined more rapidly over time than concentrations, allowing separate assessment of the importance of these two variables on detection periods. It was concluded that a valuable new antibody had been created and characterized, which can be used for the analysis of predation in the field.  相似文献

Glycosaminoglycans were isolated from mucus of patients with chronic bronchitis and mucoid impaction of the bronchus, whose contents were approximately 56 mumoles and 80 mumoles of hexosamine per g of dry weight of mucus respectively. Electrophoretic and chemical characterization and enzymatic susceptibility demonstrated that the glycosaminoglycans in mucus from both groups of the patients contained hyaluronic acid as the main constituent, with undersulphated chondroitin as a minor component. In addition, in mucus from the patient with mucoid impaction of the bronchus chondroitin sulphate and heparan sulphate or heparan sulphate-like substance were identified.  相似文献

The Iberian Peninsula contains the majority of the Paleartic land slug species of the genus Arion, which exhibits diverse taxonomic problems. The present study investigated Arion taxonomy on the basis of analyses of the mitochondrial ND1 gene and nuclear internal transcribed spacer 1 (ITS1) sequences. The Iberian endemic species were monophyletically clustered in two divergent sister clades. The topotype specimens of Arion lusitanicus and the closely related species Arion nobrei and Arion fuligineus, as well as Arion hispanicus and Arion flagellus, were grouped into an ‘Atlantic’ clade, whereas Arion baeticus, Arion gilvus, Arion anguloi, Arion wiktori and Arion paularensis were included in a ‘Continental–Mediterranean’ clade. Calibration of mutation rate in the ND1 gene suggested that the divergence of these two clades occurred around the Pliocene–Pleistocene boundary, with subsequent speciation events during the Pleistocene. A group of ancestral and divergent endemic species with distribution centred in the Pyrenean mountain range (Arion molinae, Arion lizarrusti, Arion antrhacius and Arion iratii) arose in the Pliocene and survived through the Pleistocene in geographically confined small populations. Arion lusitanicus showed up to be polyphyletic: specimens, sampled outside the geographic range of the topotype in the north‐western Iberian Peninsula, were included in a non‐monophyletic clade together with the widely distributed species Arion ater and Arion rufus. The divergent species with a wide European distribution (Arion subfuscus, Arion hortensis, Arion fagophilus and Arion intermedius) were located in basal positions in all topologies. The evolutionary history of these slug species (highly sensitive to climatic factors, with capacity for both outcrossing and selfing, and with low dispersal ability) appears to have been moulded by Pliocene–Pleistocene climate events and by the rugged topography of southern Europe, giving rise to repeated cycles of population isolation during periods of glaciation alternating with interglacial expansions limited by geographic barriers.  相似文献

Although gastropods have been crawling through the ocean and on the land for 60 million years, we still know very little about the sticky mucus produced in their foot. Most research has been focused on marine species in particular and, to a lesser extent, on the well‐known terrestrial species Arion vulgaris and Cornu aspersum. Within this study, we aim to characterize the foot anatomy of a smaller representative of the family Helicidae, the banded snail Cepaea hortensis. We are particularly interested in the microanatomy of the foot glands, their position, and the histochemical nature of their secretory content. Characterization of the dorsal foot region of Cepaea hortensis reveals four glands, differing in their size and in the granules produced. Histochemically, three of them react positively for sugars (PAS staining and lectin affinity tests for mannose, glucose and N‐acetyl‐d ‐glucosamine) and acidic proteins (positive Alcian blue and Toluidine blue staining), indicating the presence of acidic glycosaminoglycans. The fourth gland type does not react to any of these dyes. The ventral pedal region includes two different gland types, which are positive for the presence of acidic glycoproteins, with a lectin affinity for mannose only. A comparison with Helix pomatia indicates differences regarding the number of glands and their contents. In Helix, only three gland types are described in the dorsal region of the foot, which show a similar granular appearance but nevertheless differ in their chemical composition. Congruently, there are two gland types in the ventral region in both species, whereas in Helix an additional sugar moiety is found. This raises the question whether these differences between the pedal glandular systems of both helicid species are the result of protection or size‐related adaptations, as they occur in the same habitat.  相似文献

Macrophages were obtained from the mouse peritoneal cavity and culturedin vitro. The cells were exposed to35S-sulphate for 20 h, and labelled proteoglycans were recovered from both medium and cell fractions by sodium dodecylsulphate solubilization. The cell fraction contained both proteoglycans and glycosaminoglycans, whereas only intact proteoglycans could be recovered from the medium fraction. 35S-Glycosaminoglycans isolated from cell and medium fractions by papain digestion were shown to contain approximately 25% heparan sulphate and 75% galactosaminoglycans comprising 55% chondroitin sulphate and 20% dermatan sulphate. The galactosaminoglycans were shown by paper chromatography to contain more than 95% 4-sulphated units. The distribution of the sibling slug species Arion hortensiss.s. and Arion distinctus in Northern Switzerland is described.In the sampled area, both species were almost equally abundantand their distributions overlapped. Most of the sampled sitescontained both species. Arion hortensis s.s. is more commonin Switzerland than previously known, and its range extendsmore towards the east of Switzerland. Arion hortensis s.s. wasmore abundant in gardens than in the other habitats sampled.At sites where only A. distinctus occurred, the form with abipartite oviduct prevailed. At sites where both species werepresent, the form of A. distinctus with a tripartite oviductwas more common than the form with a bipartite oviduct. In captivity,one specimen of A. distinctus mated with one A. hortensis s.s.,as indicated by a spermatophore of A. distinctus that was found withinthe atrium of one A. hortensis s.s. A majority of the specimens couldby assigned to the correct species by their colouration, buta safe distinction between A. hortensis s.s. and A. distinctusis only possible using characters of the reproductive system. Experiments were conducted to determine if slug density, or food quantity and quality could cause dispersal by slugs. Arion ater dispersed quickly from overcrowded areas, where high mortality was incurred by this species. Poor food conditions, caused more dispersal than good food conditions, but food had no effect on the mortality of A. ater. Neither food nor density factors had significant effects on the dispersal of Ariolimax columbianus. However, more A. columbianus died in the high- than low-density treatments. A. columbianus mortality was unaffected by the food factor. Dispersing Arion were heavier than non-dispersers, but there was no difference between the weights of dispersing and non-dispersing Ariolimax. There were significant seasonal changes in the dispersal rate, mortality and weight of dispersing Arion, whereas only the mortality of Ariolimax changed significantly during the experiments. Since Arion ater did not markedly reduce its activity during unsuitable weather it was able to leave areas of poor food and high slug density fairly rapidly. Ariolimax columbianus' large size allowed it to remain inactive during unfavourable weather and permitted this species to be relatively unresponsive to the experimental treatments.  相似文献

Cold tolerance and metabolic responses to freezing of three slug species common in Scandinavia (Arion ater, Arion rufus and Arion lusitanicus) are reported. Autumn collected slugs were cold acclimated in the laboratory and subjected to freezing conditions simulating likely winter temperatures in their habitat. Slugs spontaneously froze at about − 4 °C when cooled under dry conditions, but freezing of body fluids was readily induced at − 1 °C when in contact with external ice crystals. All three species survived freezing for 2 days at − 1 °C, and some A. rufus and A. lusitanicus also survived freezing at − 2 °C. 1H NMR spectroscopy revealed that freezing of body fluids resulted in accumulation of lactate, succinate and glucose. Accumulation of lactate and succinate indicates that ATP production occurred via fermentative pathways, which is likely a result of oxygen depletion in frozen tissues. Glucose increased from about 6 to 22 μg/mg dry tissue upon freezing in A. rufus, but less so in A. ater and A. lusitanicus. Glucose may thus act as a cryoprotectant in these slugs, although the concentrations are not as high as reported for other freeze tolerant invertebrates.  相似文献

Acid mucopolysaccharides have been isolated from different developmental stages of Calliphora. Hyaluronic acid and a ‘larval AMPS’ were identified during all developmental stages. During the later part of the development chondroitin and a poorly-sulphated keratin sulphate-like compound were also present. Chondroitin sulphate and heparan sulphate could be detected at all development stages and during the latter part possibly keratin sulphate. The variation of acid mucopolysaccharides during development is discussed.  相似文献

Summary Post nuclear supernatants prepared from homogenates of the digestive gland of the slugArion ater were analysed by rate-dependent and density-dependent density gradient zonal centrifugation. Mannitol oxidase carrying structures exhibited distinct centrifugal behaviour sedimenting more slowly than mitochondria, lysosomes and peroxisomes but faster than microsomes, and banding sharply at a density of 1.15 g/ml in sucrose gradients. By combining rate and isopycnic centrifugations mannitol oxidase carrying structures were largely separated from contaminating organelles. Electron microscopic examination of such mannitol oxidase-enriched fractions revealed the predominant presence of distinct structures of tubular form. The effects of food supply and population density on the nocturnal behaviour of Arion ater and Ariolimax columbianus were investigated. Density did not significantly affect A. ater's level of activity or short-term movement, resting, or feeding. A. columbianus was more active and moved, rested, and fed more frequently when slug density was high. A. ater foraged and rested more often, but fed less when good food was unavailable. Ariolimax's only response to the food regime was to feed more when good food was available. Seasonal changes in the level of activity and behaviour of Arion were evident, whereas Ariolimax's activity and behavioural repertoire were not similarly affected. Arion ater's nightly activity appeared to be mainly food oriented, while Ariolimax columbianus seemed most responsive to slug density during its nocturnal activity periods.  相似文献

The acid glycosaminoglycans were extracted from the skins of young rats less than 1 day post partum. The isolated products were fractionated by a cetylpyridinium chloride-cellulose column technique and identified by chemical analysis, electrophoretic mobility and susceptibility to testicular hyaluronidase digestion. Hyaluronic acid (56%) dermatan sulphate (15.6%) and chondroitin 6-sulphate (9.1%) were the major components, but chondroitin 4-sulphate, heparan sulphate and heparin were also present, together with two further fractions tentatively suggested to be a heparan sulphate-like fraction and a dermatan sulphate fraction, both of short chain length or low degree of sulphation.  相似文献

Summary The relative assimilation efficiencies of two common British species of slug, Limax flavus (L.) and Arion hortensis (Fér.), were studied in the laboratory using a gravimetric method. Results are given for adult slugs feeding on carrot root and potato tuber at 5°, 10°, 15°, and 20° C, and for L. flavus juveniles of known age feeding on potato at 10° and 15° C. A few results are also provided for Agriolimax reticulatus (Müller) feeding on both potato and carrot at 15° C.High mean assimilation efficiencies of 76.94 and 76.87% were obtained for L. flavus feeding on carrot and potato respectively. The equivalent results for A. hortensis were 88.60 and 89.42%. For A. reticulatus at 15° C the figures were 76.1 and 71.9%. Ingestion rates and assimilation rates are given for the first two species at all temperatures. Assimilation efficiency was found to be independent of temperature for L. flavus but not for A. hortensis. Ingestion rate and assimilation rate were temperature dependent in all cases except for A. hortensis feeding on potato. A. hortensis was more efficient at consuming both carrot and potato per gram live weight. All species consumed more potato than carrot per gram live weight.Juvenile L. flavus were found to have higher assimilation efficiencies and ingestion rates than the adults per gram live weight.Linear regression equations were determined from the relationship between dry weight of food consumed, and dry weight of faeces produced from that food for L. flavus.It is concluded that food quality and age of slug may be the factors most strongly influencing assimilation efficiency in this group.  相似文献

The nematode Phasmarhabditis hermaphrodita is a commercially available biocontrol agent against slugs. This product is especially interesting for use in organic farming, where products containing metaldehyde or carbamates cannot be used for controlling pest slugs. We investigated the potential of P. hermaphrodita for the control of the pest slugs Deroceras reticulatum and Arion lusitanicus. These two species are the most harmful slug pests in Switzerland. At different times of the year, we collected slug specimens of different weight and assessed their susceptibility to P. hermaphrodita in the laboratory. Batches of five slugs were subjected to five different doses of nematodes plus an untreated control and replicated three times. During six weeks, feeding and survival of the slugs were recorded. D. reticulatum was strongly affected by increasing nematode doses, irrespective of the slugs' body weight. In small specimens of A. lusitanicus, feeding and survival were strongly affected by the nematodes, while larger specimens remained almost unaffected. Because A. lusitanicus has an asynchronous development in Switzerland, it seems difficult to control the entire population with a single nematode application. To what extent nematodes will be used in practice for slug control depends on their effectivity against the pest slugs of major importance, on the longevity of the molluscicidal effect and on the price of nematodes.  相似文献

Carabid beetles (Coleoptera: Carabidae) were fed upon slugs (Mollusca: Pulmonata) in the laboratory, and their crop contents analysed for mollusc remains, using an enzyme-linked immunosorbent assay (ELISA) with an anti-Arion ater (L.) haemolymph antiserum. Crop weight loss and antigenic recognition of prey proteins were examined as separate variables in a series of validatory experiments. Two model predators,Abax parallelepipedus Piller and Mitterpacher andPterostichus madidus F., were fed upon two species of pest slugs,Deroceras reticulatum (Müller) andArion hortensis Férussac. The fitting of regression equations to the transformed antigenic response data allowed the ‘half-life’ and detection period to be calculated for each predator-prey combination. Following a one hour feeding interval, the half-life of the antigenic response toD. reticulatum remains was almost twice as long inP. madidus as that inA. parallelepipedus, and the detection period more than 2.5 times as long. However, covariant analysis showed that there was a significant difference between predator species in the rate at which detectability declined, but not in the rate of crop weight loss. WhenA. parallelepipedus was allowed to feed uponA. hortensis for eight hours, prey remains were still detectable at the end of the experiment, 13 days after feeding. Calibration of the differential rates of antigen decay and crop weight loss could potentially be used to calculate the size of the original meal, but only if prey species, and the time since feeding, can be determined. Potential solutions to these problems are discussed.  相似文献

Five monoclonal antibodies AS17, 22, 25, 38 and 48, a single monoclonal antibody ACH55, and three monoclonal antibodies NAH33, 43, 46, that recognize acharan sulfate (IdoA2S-GlcNAc)n, acharan (IdoA-GlcNAc)n and N-acetyl-heparosan (GlcA-GlcNAc)n, respectively, were generated by immunization of mice with keyhole limpet hemocyanin-conjugated polysaccharides. Specificity tests were performed using a panel of biotinylated GAGs that included chemically modified heparins. Each antibody bound avidly to the immunized polysaccharide, but did not bind to chondroitin sulfates, keratan sulfate, chondroitin nor hyaluronic acid. AS antibodies did not bind to heparan sulfate or heparin, but bound to 6-O-desulfated, N-desulfated and re-N-acetylated heparin to varying degrees. ACH55 bound to tri-desulfated and re-N-acetylated heparin but hardly bound to other modified heparins. NAH antibodies did not bind to heparin and modified heparins but bound to heparan sulfate to varying degrees. NAH43 and NAH46 also bound to partially N-de-acetylated N-acetyl-heparosan. Immunohistochemical analysis in rat cerebella was performed with the antibodies. While NAH46 stained endothelia, where heparan sulfate is typically present, neither ACH55 nor AS25 stained endothelia. On the contrary ACH55 and AS25 stained the molecular layer of the rat cerebella. Furthermore, ACH55 specifically stained Purkinje cells. These results suggest that there is unordinary expression of IdoA2S-GlcNAc and IdoA-GlcNAc in specific parts of the nervous system. Suzuki and Yamamoto contributed equally to this study.  相似文献

Ochropsora ariae was found to host-alternate between Anemone pseudo-altaica and Aruncus dioicus var. tenuifolius and Ochropsora nambuana between Anemone flaccida and Elaeagnus multiflora var. hortensis in Japan. Both species produced a sessile, thin-walled, cylindrical probasidium (teliospore), which turned into a four-celled metabasidium by continuous apical elongation of the probasidium. Several probasidia of O. nambuana were produced from a basal basidiogenous cell in a sorus hymenium. Life cycle and probasidium/metabasidium morphology showed the taxonomic identity of Ceraceopsora elaeagni with O. nambuana. Developmental morphology of the basidium found in the two Ochropsora species raised a question against the taxonomic separation of Ochropsora and Aplopsora.  相似文献

Intact and fully O[emsp4 ]-sulfonated glycosaminoglycans (GAGs) including chondroitin sulfate, dermatan sulfate, hyaluronan, heparan sulfate and heparin were chemically de-O-sulfonated on their hexosamine C-6 position (6-O[emsp4 ]-desulfonation) using N,O[emsp4 ]-bis(trimethylsilyl) acetamide. 1H NMR spectroscopy and chemical compositional analysis showed that the chemical de-O[emsp4 ]-sulfonation at C-6 position of hexosamine residues in both intact and fully O[emsp4 ]-sulfonated GAGs was completely achieved. Since GAGs and their derivatives are often used as anticoagulant agents, their anti-amidolytic activities were determined. While most of anticoagulant activity of fully O[emsp4 ]-sulfonated GAGs (FGAGs) and heparin disappeared following chemical 6-O[emsp4 ]-desulfonation, the activity of 6-O-desulfonated fully O[emsp4 ]-sulfonated dermatan sulfate (De6FDS) remained. This observation suggests the importance of the position of O-sulfonate groups for anti-coagulant activity.  相似文献
