The application of Fourier transform mid-infrared (FT-MIR) spectroscopy and Fourier transform Raman (FT-Raman) spectroscopy
for process and quality control of fermentative production of ethanol was investigated. FT-MIR and FT-Raman spectroscopy along
with multivariate techniques were used to determine simultaneously glucose, ethanol, and optical cell density of Saccharomyces cerevisiae during ethanol fermentation. Spectroscopic measurement of glucose and ethanol were compared and validated with the high-performance
liquid chromatography (HPLC) method. Spectral wave number regions were selected for partial least-squares (PLS) regression
and principal component regression (PCR) and calibration models for glucose, ethanol, and optical cell density were developed
for culture samples. Correlation coefficient (R
2) value for the prediction for glucose and ethanol was more than 0.9 using various calibration methods. The standard error
of prediction for the PLS first-derivative calibration models for glucose, ethanol, and optical cell density were 1.938 g/l,
1.150 g/l, and 0.507, respectively. Prediction errors were high with FT-Raman because the Raman scattering of the cultures
was weak. Results indicated that FT-MIR spectroscopy could be used for rapid detection of glucose, ethanol, and optical cell

The taxonomic significance of the polysaccharide structures of algal cell walls has been underscored several times over the past few decades but has never been pursued systematically. Many changes in red algal systematics and the biochemical analyses of phycocolloids have occured in recent years. The cell-wall composi-tion of representatives of 167 (24.7%) genera and 470 (11.5%) species of red algae has been documented.The method developed by Chopin and Whalen for carrageenan identification by Fourier transform infrared diffuse reflectance spectroscopy is extended to the study of phycocolloids for diverse species in many red algal orders. This paper focuses on the Gigartinales in which representatives of 28 (68.3%) families, 88 (50.6%) genera and 224 (27.9%) species have been analyzed. In light of recent molecular phylogenies, some patterns of distribution of key phycocolloid attributes, corresponding to familial and ordinal level groupings, are emerging; however, more species remain to be analyzed. The well-documented biochemical alternation of generations in the Phyllophoraceae, Petrocelidaceae and Gigartinaceae still holds (with two exceptions), but this pattern was not recorded in other families of Gigartinales. 相似文献

Circular dichroism and resolution-enhanced Fourier transform infrared reveal induction of secondary structural elements on peptidyl-prolyl cis–trans isomerase A (PpiA) from Mycobacterium tuberculosis upon binding cyclosporin A (CsA). Thermal denaturation shows aggregation of PpiA at higher temperatures (>70 °C) and CsA fails to impart stabilization in protein structure. However, CsA stabilizes PpiA structure in urea denaturation. In presence/absence of CsA, urea-induced reversible unfolding of secondary and tertiary structures follows two-state and three-state transition, respectively. The chemical unfolding results also demonstrate that loss in the tertiary structure precedes the loss in secondary structure both in presence and absence of CsA at the initial stages. Fluorescence quenching suggests presence of a positive barrier around tryptophan microenvironment of PpiA. 相似文献

In most invertebrates, multiple species-specific isoforms of tachykinin-related peptide (TRP) are common. In contrast, only a single conserved TRP isoform, APSGFLGMRamide, has been documented in decapod crustaceans, leading to the hypothesis that it is the sole TRP present in this arthropod order. Previous studies of crustacean TRPs have focused on neuronal tissue, but the recent demonstration of TRPs in midgut epithelial cells in Cancer species led us to question whether other TRPs are present in the gut, as is the case in insects. Using direct tissue matrix assisted laser desorption/ionization Fourier transform mass spectrometry, in combination with sustained off-resonance irradiation collision-induced dissociation, we found that at least one additional TRP is present in Cancer irroratus, Cancer borealis, Cancer magister, and Cancer productus. The novel TRP isoform, TPSGFLGMRamide, was present not only in the midgut, but also in the stomatogastric nervous system (STNS). In addition, we identified an unprocessed TRP precursor APSGFLGMRG, which was detected in midgut tissues only. TRP immunohistochemistry, in combination with preadsorption studies, suggests that APSGFLGMRamide and TPSGFLGMRamide are co-localized in the stomatogastric ganglion (STG), which is contained within the STNS. Exogenous application of TPSGFLGMRamide to the STG elicited a pyloric motor pattern that was identical to that elicited by APSGFLGMRamide, whereas APSGFLGMRG did not alter the pyloric motor pattern. 相似文献

Diffusion approximations are ascertained from a two-time-scale argument in the case of a group-structured diploid population with scaled viability parameters depending on the individual genotype and the group type at a single multi-allelic locus under recurrent mutation, and applied to the case of random pairwise interactions within groups. The main step consists in proving global and uniform convergence of the distribution of the group types in an infinite population in the absence of selection and mutation, using a coalescent approach. An inclusive fitness formulation with coefficient of relatedness between a focal individual J affecting the reproductive success of an individual I, defined as the expected fraction of genes in I that are identical by descent to one or more genes in J in a neutral infinite population, given that J is allozygous or autozygous, yields the correct selection drift functions. These are analogous to the selection drift functions obtained with pure viability selection in a population with inbreeding. They give the changes of the allele frequencies in an infinite population without mutation that correspond to the replicator equation with fitness matrix expressed as a linear combination of a symmetric matrix for allozygous individuals and a rank-one matrix for autozygous individuals. In the case of no inbreeding, the mean inclusive fitness is a strict Lyapunov function with respect to this deterministic dynamics. Connections are made between dispersal with exact replacement (proportional dispersal), uniform dispersal, and local extinction and recolonization. The anole fauna of the Lesser Antilles is depauperate in relation to that of the Greater Antilles, where complex communities characterized by adaptive specialization and convergent structure are present. Much of this adaptation is the result of changes in body size and shape, probably as a result of interspecific competition. Here we present data on variation in size and shape within a solitary Lesser Antillean species occupying an ecologically heterogenous island, and test the hypothesis that natural selection for varying environmental conditions is the cause of this variation. Univariate (analysis of variance), bivariate (analysis of covariance) and multivariate (multiple-group principal component analysis, canonical variate analysis) analysis showed that there is considerable geographic variation in size and shape within Anolis oculatus on Dominica. Partial Mantel tests rejected the null hypothesis of no association between size and shape and environmental variation. The possible proximate and evolutionary mechanisms responsible for these patterns are discussed. Despite these overall associations, a considerable amount of variation in shape appears to be unrelated to environmental variation. 相似文献

We investigated host-plant utilisation by the candidate biocontrol agent Paradibolia coerulea (Coleoptera: Chrysomelidae) on the target plant Spathodea campanulata Beauv. (Bignoniaceae) and a closely related non-target plant, Kigelia africana (Lam.) Benth. (Bignoniaceae). Paired-choice and sequential no-choice experiments were performed and coupled with olfactory discrimination experiments to test the insects’ responses to volatiles from both plant species as well as to cues from conspecific beetles. Although K. africana was utilised by P. coerulea, S. campanulata was preferred for both adult feeding and oviposition. Interestingly, whereas females were attracted to olfactory cues emitted by S. campanulata, males demonstrated no such olfactory discrimination. Females were also attracted to cues deposited by males, and males were deterred by cues from other males, but neither sex responded to female olfactory cues. Very few eggs were recorded on K. africana and none of the larvae that hatched on K. africana survived the first instar. Both S. campanulata and K. africana are suitable for adult feeding, but persistent utilisation of K. africana in the field is unlikely because larval development is only possible on S. campanulata and because the adult females are strongly attracted to volatiles emitted by the target plant. Nevertheless, if P. coerulea is released as a biocontrol agent, spill-over adult feeding could potentially occur on K. africana growing sympatrically with S. campanulata. Because P. coerulea cannot complete its development on K. africana, non-target damage will only occur where the target plant is present, with an intensity dependent on densities of adult beetles locally. 相似文献

Summary Seedlings of Inga oerstediana Benth. (Mimosaceae) growing in three different light environments (the understory, tree-fall gaps and full sun) were tested for differences in chemistry (nutrients and tannins), wound-induced increases in tannins, growth, and susceptibility to leaf-cutter ants, Atta cephalotes (L.) (Formicidae: Attini). I hypothesized that seedlings of I. oerstediana would contain higher concentrations of tannins when growing in high light conditions and, therefore, would be less susceptible to leaf-cutter ants.Foliar concentrations of condensed tannins were much higher in plants growing in full sun compared to those growing in the understory. The concentrations of condensed tannins did not increase following damage. Despite higher concentrations of condensed tannins in sun foliage, leaf-cutter ants found these leaves more acceptable. The preference for sun leaves was consistent with higher concentrations of foliar nutrients. I suggest that the magnitude of the increase in condensed tannins was not great enough to override the benefits of increased concentrations of foliar nutrients. Finally, based on these results and those of others, I suggest that foraging by leaf-cutter ants may be an important factor determining patterns of succession in early successional habitats. 相似文献

Current advances in genetic analysis are opening up our knowledge of the genetics of species differences, but challenges remain, particularly for out‐bred natural populations. We constructed a microsatellite‐based linkage map for two out‐bred lines of Drosophila montana derived from divergent populations by taking advantage of the Drosophila virilis genome and available cytological maps of both species. Although the placement of markers was quite consistent with cytological predictions, the map indicated large heterogeneity in recombination rates along chromosomes. We also performed a quantitative trait locus (QTL) analysis on a courtship song character (carrier frequency), which differs between populations and is subject to strong sexual selection. Linkage mapping yielded two significant QTLs, which explained 3% and 14% of the variation in carrier frequency, respectively. Interestingly, as in other recent studies of traits which can influence speciation, the strongest QTL mapped to a genomic region partly covered by an inversion polymorphism. 相似文献

The Rf3 gene restores the pollen fertility disturbed by S male sterile cytoplasm. In order to develop molecular markers tightly linked to Rf3, we used amplified fragment length polymorphism (AFLP) technique with near isogenic lines (NILs) and bulk segregant analysis (BSA). A BC1F1 population from a pair of NILs with different Rf3 locus was constructed and 528 primer combinations was screened. A linkage map was constructed around the Rf3 locus, which was mapped on the distal region of chromosome 2 long arm with the help of SSR marker UMC2184. The closest marker E7P6 was 0.9 cM away from Rf3. Marker E3P1, 2.4 cM from Rf3, and E12M7, 1.8 cM from Rf3, were converted into a codominant CAPS and a dominant SCAR marker, and designated as CAPSE3P1 and SCARE12M7, respectively. These markers are useful for marker-assisted selection and map-based cloning of the Rf3 gene. 相似文献

Knowledge of how mating success is related to body size may provide insight into the evolution of social systems. This study investigated the mating system and relevant social behavior of a temperate anuran (Rana chensinensis) at three localities in northern China. During chorusing, males aggregated and persisted in the communal spawning ponds with a density of 10–26 frogs per m2 water area and operational sex ratio of 15–28 males to 1 female. The males frequently grabbed any conspecifics they encountered, releasing the grip if the individual was a male, but holding the grip when it was a female. A significant positive relationship between male and female body lengths of pairs in amplexus was detected from all the sites during the five breeding seasons, but the average correlation coefficients of determination of 18% indicated that the size-assortative mating constituted a minority of the species’ mating system. Pairing probability decreased with increased size differences between sexes, which could have prevented a few adult animals in a population from forming pairs. The strong aggregation of males could limit the opportunities for large males to exhibit contest advantages in mating and for both sexes to choose a large mate. Infrequent occurrence of scramble competition (averaging 8% of the recorded amplectant pairs) and the lack of observed takeovers, probably because of reclusion of amplectant pairs at the bottom of spawning ponds, suggested a weak role of amplexus displacement in generating non-random mating. Our results suggest that although non-random mating with respect to body size may be expected in explosive breeders, the social environment characterized by male aggregation may also impose weak selective pressure for the evolution of non-random mating. 相似文献

Insects flying into the web of an orb-weaving spiderAraneus pinguis (Karsch) and their avoidance of (pre-hitting process) and escapes from (post-hitting process) the web were examined by direct observation under natural and semi-natural conditions. In the pre-hitting process, mobile insects such as Brachycera, Lepidoptera and Hymenoptera showed a low hitting ratio (number of insects hitting/number of insects flying within 1 m3 space around the web-site) because of active web avoidance and flying activity in layers lower or higher than those in which the webs are usually laid. In contrast, less mobile insects like Heteroptera, Coleoptera and Homoptera showed a high hitting ratio. In the post-hitting process, Brachycera, Lepidoptera and some Nematocera frequently escaped without being detained by the web. Many Orthoptera and Hymenoptera escaped without any sign of detection by the spider. Coleoptera frequently escaped during the spider's attack. Small insects from the Homoptera, Nematocera and Hymenoptera rarely escaped from the web, but were not immediately attacked. Mean escape time of insects was correlated significantly with capture success of the spider. Overall most of the escapes occurred in the early phases of the predation process. This indicates that escapes are unlikely to result in heavy loss of time and energy expenditure due to unsuccessful predation. Escape patterns of insects seem to be related to their mobility. 相似文献

In Nigeria, Mucuna pruriens seeds are locally prescribed as an oral prophylactic for snake bite and it is claimed that when two seeds are swallowed they protect the individual for a year against snake bites. In order to understand the Mucuna pruriens antisnake properties, the proteins from the acqueous extract of seeds were purified by three chromatographic steps: ConA affinity chromatography, tandem anionic-cationic exchange and gel filtration, obtaining a fraction conventionally called gpMucB. This purified fraction was analysed by SDS-PAGE obtaining 3 bands with apparent masses ranging from 20 to 24 kDa, and by MALDI-TOF which showed two main peaks of 21 and 23 kDa and another small peak of 19 kDa. On the other hand, gel filtration analysis of the native protein indicated a molecular mass of about 70 kDa suggesting that in its native form, gpMucB is most likely an oligomeric multiform protein. Infrared spectroscopy of gpMucB indicated that the protein is particularly thermostable both at neutral and acidic pHs and that it is an all beta protein.All data suggest that gpMucB belongs to the Kunitz-type trypsin inhibitor family explaining the direct anti-snake venom activity of Mucuna pruriens seeds. 相似文献

Bacillus anthracis, the causative agent of anthrax, requires surface (S)-layer proteins for the pathogenesis of infection. Previous work characterized S-layer protein binding via the surface layer homology domain to a pyruvylated carbohydrate in the envelope of vegetative forms. The molecular identity of this carbohydrate and the mechanism of its display in the bacterial envelope are still unknown. Analyzing acid-solubilized, purified carbohydrates by mass spectrometry and NMR spectroscopy, we identify secondary cell wall polysaccharide (SCWP) as the ligand of S-layer proteins. In agreement with the model that surface layer homology domains bind to pyruvylated carbohydrate, SCWP was observed to be linked to pyruvate in a manner requiring csaB, the only structural gene known to be required for S-layer assembly. B. anthracis does not elaborate wall teichoic acids; however, its genome harbors tagO and tagA, genes responsible for the synthesis of the linkage unit that tethers teichoic acids to the peptidoglycan layer. The tagO gene appears essential for B. anthracis growth and complements the tagO mutant phenotypes of staphylococci. Tunicamycin-mediated inhibition of TagO resulted in deformed, S-layer-deficient bacilli. Together, these results suggest that tagO-mediated assembly of linkage units tethers pyruvylated SCWP to the B. anthracis envelope, thereby enabling S-layer assembly and providing for the pathogenesis of anthrax infections. 相似文献
